Anal papillae from fourth instar larvae of the salt-water mosquito Aedes campestris maintained in normal (hyperosmotic) lake water are made up of a single epithelial layer. The only type of cell present in this layer shows several ultrastructural features characteristic of transporting tissues. The apical plasma membrane (facing the external medium) is infolded into a series of lamellae which bear a particulate coat on the cytoplasmic surface. The basal plasma membrane (facing the haemolymph) is also highly infolded to form a system of interconnecting channels throughout the cell. These channels may be considerably dilated. Dense mitochondria are abundant in anal papillae cells and tracheoles frequently penetrate deeply into the cells. No qualitative or statistically significant quantitative differences are observed in the ultrastructure of anal papillae taken from A. campestris larvae maintained in diluted (hyposmotic) lake water. It is suggested that in both hyperosmotic and hyposmotic external media the anal papillae are actively engaged in ion transport and that adaptive changes in transport rates which have been demonstrated physiologically may require only minor structural modifications such as permeability changes or activation of enzyme systems already present in the cell membranes.  相似文献

The genus Parostertagia is referred to the Cooperiinae with reference to a suite of shared characters of the synlophe and genital cone postulated as derived homologies for the subfamily. The genus has traditionally been relegated to the Graphidiinae or Ostertagiinae, but unique structural attributes of the synlophe and genital cone indicate close affinities with the Cooperiinae. The synlophe of Parostertagia heterospiculum is characterised by a relatively low number of ridges in the cervical zone (16–20 at the excretory pore), a frontal axis of orientation, sequential increase in ridges posteriorly, lateral addition of ridges, minuscule lateral-most ridges and hypertrophy of specific ridges in the lateral field of females. The genital cone with a tripartite “0” papilla, a pair of dorsal cloacal papillae (a character potentially homologous with lateral protuberances in some cooperiines, but reported for the first time in this study), and paired “7” papillae supporting a small accessory bursal membrane appear unique among the trichostrongylids. Additionally, Parostertagia was found to possess minuscule cervical papillae and a rudimentary (or reduced?) cephalic and cervical expansion. Inclusion of Parostertagia within the Cooperiinae is based on shared characters with this subfamily (postulated synapomorphies) rather than differences that may be utilized to separate this genus from members of other subfamilies of the Trichostrongylidae.  相似文献

The morphology, cell types, and innervation of the several small papillae (x? = 17) and two larger papillae, which together form a ring just outside the fimbriae surrounding the suctorial disc of adult Geotria australis, have been studied using various histological stains, including silver impregnation, and scanning and transmission electron microscopy. The epithelium of all papillae consists almost entirely of mucigenic cells. The multivillous and oligovillous cells, which are found elsewhere in the lamprey epidermis, were not observed, and Merkel and polyvillous cells are rare. Free nerve endings are common, however, in the basal layers of the epidermis. Unlike the small papillae, the two large papillae contain a core of skeletal muscle and a prominent layer of dermal collagen. In the submucosa of these large papillae, the nerves form a dense, compact layer that contains many large and probably sensory axons. It is suggested that the oral disc papillae of adult G. australis are encapsulated mechanosensory structures that play a role in enabling the animal to locate and attach to a suitable point on host fishes or other surfaces.  相似文献

Hoole D. and Mitchell J.B. 1981. Ultrastructural observations on the sensory papillae of juvenile and adult Gorgoderina vitelliloba (Trematoda : Gorgoderidae). International Journal for Parasitology11: 411–417. Ultrastructural observations have been made on the juvenile and adult stages of Gorgoderina vitelliloba from Rana temporaria. Four types of sensory papillae occur; button, rosette, ciliated and domed. Button papillae, which contain a ciliary-rootlet but lack a cilium, occur on the oral sucker, dorsal preacetabular surface and the lateral margins of the fluke. Scanning electron microscopical observations reveal that the tegumental protuberances of the papilla are transformed from a spiked or conical appearance in juvenile flukes to a rounded form in adult flukes. Rosette papillae, which also contain a rootlet, occur on the lip of the ventral sucker of both juvenile and adult flukes. Ciliated papillae only occur on the oral sucker of juvenile flukes. Domed papillae, which contain a large area of electron-dense material, occur on the internal surface of the ventral sucker of both juvenile and adult flukes. The functions of papillae and their possible role in the migration of the parasite are discussed.  相似文献

Background

Despite the presence of papillary structures and papillary tumors in humans, the mechanism of papillae formation is unknown. We describe herein a novel role for Niemann-Pick disease type 2C (NPC2) protein, a cholesterol binding protein in the lysosome, in papillae formation.

Methodology/Principal Finding

We examined NPC2 protein expression in surgical samples of papillary tissues by immunohistochemical stain, and all papillary tissues expressed NPC2 protein in the epithelium. To examine our hypothesis of NPC2 protein-mediated papillae formation, we carried out xenograft experiments using wild H460 cells (large cell lung carcinoma cell line) that constitutively expressed abundant NPC2 protein and NPC2 protein-depleted H460 cells by NPC2 shRNA. The xenografts of wild H460 cells and empty shRNA vector cells showed distinct papillae formation, whereas NPC2 protein-depleted H460 cells displayed markedly reduced or no papillae. Since all papillary tissues have open spaces we examined whether NPC2 protein might also contribute to the creation of open spaces. The TUNEL assay in the xenografts of wild and empty shRNA vector H460 cells showed massive cell death, and NPC2 protein-depleted cells displayed minimal cell death. Measurement of caspase 3/7 activities in cultured H460 cells supported NPC2 protein-mediated apoptotic cell death. The presence of excess NPC2 protein, however, did not always produce papillae as seen in the xenografts of CHO cells that were stably transfected with NPC2.

Conclusions/Significance

The NPC2 protein of certain cells forms papillae coupled with apoptosis that creates open space. This protein may have future applications to modulate papillae formation and papillary growth in tumor tissues.  相似文献

BACKGROUND AND AIMS: The labellar papillae and trichomes of Maxillaria Ruiz & Pav. show great diversity. Although papillae also occur upon other parts of the flower (e.g. column and anther cap), these have not yet been studied. Labellar trichomes of Maxillaria are useful in taxonomy, but hitherto the taxonomic value of floral papillae has not been assessed. The aim of this paper is to describe the range of floral papillae found in Maxillaria and to determine whether papillae are useful as taxonomic characters. METHODS: Light microscopy, histochemistry, low-vacuum scanning and transmission electron microscopy. KEY RESULTS: A total of 75 taxa were studied. Conical papillae with rounded or pointed tips were the most common. The column and anther cap usually bear conical, obpyriform or villiform papillae, whereas those around the stigmatic surface and at the base of the anther are often larger and swollen. Labellar papillae show greater diversity, and may be conical, obpyriform, villiform, fusiform or clavate. Papillae may also occur on multiseriate trichomes that perhaps function as pseudostamens. Labellar papillae contain protein but most lack lipid. The occurrence of starch, however, is more variable. Many papillae contain pigment or act as osmophores, thereby attracting insects. Rewards such as nectar or a protein-rich, wax-like, lipoidal substance may be secreted by papillae onto the labellar surface. Some papillae may have a protective role in preventing desiccation. Species of diverse vegetative morphology may have identical floral papillae, whereas others of similar vegetative morphology may not. CONCLUSIONS: Generally, floral papillae in Maxillaria have little taxonomic value. Nevertheless, the absence of papillae from members of the M. cucullata alliance, the occurrence of clavate papillae with distended apices in the M. rufescens alliance and the presence of papillose trichomes in some species may yet prove to be useful.  相似文献

The structure of the epidermis of Travisia forbesii was described using light and electron microscopy. The epidermis is a highly modified variant of the normal one-layer polychaete epithelium. It consists of basal epidermal cells and an external layer of closely sited papillae consisting of glandular and supportive epidermal cells, and extensive electron-transparent intercellular spaces. The papillae are embedded in the thick cuticle. Each papilla has a peduncle, which is formed by one cell that penetrates the inner cuticle layer to the basal epidermal cells. A fold of basement membrane forms the core of the peduncle and ends in the base of a papilla. All epidermal cells are connected to each other with apical cell junctions and to the basement membrane with hemidesmosomes, so the epithelium is continuous and uninterrupted. The epidermis has an intra-epidermal neuron plexus. The structure of the papillae is compared with papillae and tubercles of other polychaetes, and the possible functional significance and phylogenetic implications of these structures are discussed.  相似文献

The anal papillae of Drosophila melanogaster larvae showed strong developmental plasticity in media with different osmotic pressure. The size of the papillae decreased with increasing salt content of the medium, which supports the hypothesis of its absorptive function. We showed that CI-transport at the site of the anal papillae decreased with increasing salt concentration, which also supports the hypothesis, that the function of anal papillae in osmoregulation is absorption at low salt content and not excretion at high salt content. The inactive area between the anal papillae increased with increased salt concentration, and thus contributed to the decreasing size of the anal papillae. Remnants of this area in pupae were mistakenly measured as anal papillae in a study on salt adaptation by Waddington (1959).  相似文献

Summary Echinoids usually broadcast gametes, and do not generally engage in a high degree of parental care. However, when they do, juveniles are typically maintained among the spines, or in shallow, external depressions in the test itself. The brooding Antarctic holasteroids Urechinus mortenseni and Plexechinus nordenskjoldi are bizarre exceptions: females develop an elaborate brooding system in which a small number of direct developing young are protected. Ontogeny of post-natal brooding urechinids is marked by profound divergence in the growth trajectories of male and female apical systems. In females, this leads to dramatic departures from the patterns found in all other echinoids. Otherwise, coronal skeleton allometry of males and females is almost identical. Juveniles in brood pouches grow larger than the diameter of the apical aperture through which they must pass to reach the external environment. The apical plates, from which the brooding system is suspended, hinge downward to enlarge the aperture, allowing the young to emerge from the female. A possible origin for the brooding system suggests derivation by centripetal plate addition from the ocular plates in the coronal skeleton. We develop a contrasting model for the origin of the brooding system that relies on a proposed homology between genital and periproctal elements of the apical system of echinoids and the more highly developed dorsal skeleton of other echinoderm classes.  相似文献

Thaumatin I is an intensely sweet-tasting protein. It was photo-crosslinked with taste papillae of crab-eating monkey by using a conjugated photo-affinity reagent [3H]azidobenzoylthaumatin I. Serial sections of SDS-polyacrylamide gel electrophoresis of the 0.1 M sodium phosphate buffer-soluble fraction from taste papillae had a large peak of radioactivity at the Mr region of approx. 70,000; fractions from non-taste papillae did not. Excess unlabeled thaumatin I reduced the photo-crosslinking at the 70 kDa region; acetylated thaumatin I (which is not sweet) did not. The results show that taste papillae of the monkey contain a protein of Mr approx. 50,000, which binds to thaumatin I (Mr 22,209) but not to completely acetylated thaumatin I. The possibility that the thaumatin-binding protein is a sweet receptor protein is discussed.  相似文献

The mechanical papillae of the horse and cow were studied by scanning electron microscopy in order to determine their morphostructural characteristics and the differences between the two species. The horse has only thin, small and interlaced filiform papillae, while the cow shows robust and more ordered filiform papillae. Furthermore, the cow tongue presents conical and lenticular papillae surrounded by a papillary groove. A characteristic distribution of stratified scales and channeled tracts is observed in conical and lenticular papillae but not in the filiform papillae. The morphostructural features of each papilla are analyzed and compared in each species and their possible significance is discussed.  相似文献

Summary The ventral surface of the posterior sucker of Entobdella soleae, a monogenean skin parasite of the sole Solea solea, is covered with more than 800 papillae ranging in size from 2.5–19 m in diameter. The papillae are penetrated by nerves which double over themselves to form a stack-like array of lamellae running parallel to the surface of the haptor. No cilia or associated structures are present within these presumed sense organs and the papillae have no opening to the exterior. The much larger sucker papillae of a related species, E. hippoglossi, have been shown to have a similar ultrastructure. Edwards H. H., Nollen P. M. &; Nadakavukaren M. J. 1977. Scanning and transmission electron microscopy of the oral sucker papillae of Philophthalmus megalurus. International Journal for Parasitology7: 429–437. Scanning electron microscopy reveals papillae on both the inside and outside of the oral sucker of the eyefluke, Philophthalmus megalurus. In the transmission electron microscope the papillae can be divided into 3 different types with 2 of these types occurring on the outside of the oral sucker. One type of outer papilla contains a bulb cell which is terminated by a cilium having an apparent typical arrangement of microtubules. This type is a presumed sensory receptor having tango- and/or rheoreceptor function. The second type of outer papilla contains a gland cell which secretes electron-dense granules to the outside of the fluke. The third type of papilla is found only on the inside of the oral sucker and contains a bulb cell terminated by a cilium which does not have the typical 9 + 2 arrangement of microtubules. Instead, their numbers increase to over 60 randomly arranged microtubules. This inner papilla is felt to have chemoreceptor function because of its location and the presence of the modified cilium. The bulb cell of the inner papilla contains 2 newly described features. The first is granular, and associated with microtubules; it may be a possible nucleating site for microtubule synthesis. The second structure is a crystalline inclusion of unknown function and origin.  相似文献

Cephalopods (octopus, squid and cuttlefish) are known for their camouflage. Cuttlefish Sepia officinalis use chromatophores and light reflectors for color change, and papillae to change three-dimensional physical skin texture. Papillae vary in size, shape and coloration; nine distinct sets of papillae are described here. The objective was to determine whether cuttlefish use visual or tactile cues to control papillae expression. Cuttlefish were placed on natural substrates to evoke the three major camouflage body patterns: Uniform/Stipple, Mottle and Disruptive. Three versions of each substrate were presented: the actual substrate, the actual substrate covered with glass (removes tactile information) and a laminated photograph of the substrate (removes tactile and three-dimensional information because depth-of-field information is unavailable). No differences in Small dorsal papillae or Major lateral mantle papillae expression were observed among the three versions of each substrate. Thus, visual (not tactile) cues drive the expression of papillae in S. officinalis. Two sets of papillae (Major lateral mantle papillae and Major lateral eye papillae) showed irregular responses; their control requires future investigation. Finally, more Small dorsal papillae were shown in Uniform/Stipple and Mottle patterns than in Disruptive patterns, which may provide clues regarding the visual mechanisms of background matching versus disruptive coloration.  相似文献

Gunnera manicata L. glands consist of up to nine separate papillae. Surgical removal of papillae showed that more than two papillae were needed for successful infection with Nostoc. Chlamydia muridarum induction of mouse hydrosalpinx, depending on both tubal infection and inflammation, has been used for investigating Chlamydia trachomatis pathogenesis. We now report that IL-6 both inhibits C. muridarum infection and exacerbates pathogenicity in the mouse genital tract. When intravaginally inoculated with a high dose of C. muridarum, IL-6-deficient mice developed more extensive genital tract infection with severe hydrosalpinx, suggesting that IL-6 is required for controlling the high dose infection but not essential for C. muridarum-induced pathology. However, at a low dose, IL-6-deficient mice still developed more extensive infection in the genital tract but no longer with significant pathology, suggesting that IL-6 is required for both controlling the low dose infection and exacerbating the low dose infection-induced pathology. The lack of hydrosalpinx in IL-6-deficient mice correlated with significantly reduced inflammatory infiltration in the oviduct tissue and decreased spleen CD4+ and CD8+ T cells that produce TNFα. Thus, IL-6-dependent pathways are important for both limiting chlamydial colonization in the genital tract mucosal tissues regardless of the infection doses and exacerbating chlamydial pathogenicity in the upper genital tract when IL-6-independent pathogenic mechanisms are not yet activated with a low infection dose.  相似文献
