Recombinant Saccharomyces cerevisiae YKU 131 (capable of expressing glucoamylase) was used to produce ethanol from sago starch. The optimum C/N ratio for ethanol production by the recombinant yeast was 7.9, where 4.7 and 10.1 g/l ethanol was produced from 20 and 40 g/l sago starch, respectively. At sago starch concentration higher than 40 g/l and C/N ratio higher than 10.4, glucoamylase production and rate of starch hydrolysis were reduced, which in turn, reduced ethanol production significantly. The theoretical yield of ethanol based on sago starch consumed in fermentation using 40 g/l was 72.6%. This yield was slightly lower than those obtained in fermentation using soluble starch such as potato and corn starch, which ranged from 80–90% as reported in the literature. However, S. cerevisiae YKU 131 could only utilize 62% of the total amount of starch added to a medium.  相似文献

Production of carotenoid by Rhodotorula gracilis was highest at 26 mg/g dry weight with a carbon/nitrogen (C/N) ratio in the medium of 10:1. This was 15 times higher than when the C/N ratio was 160:1. With this high C/N ratio, the yeast produced up to 55% lipid compared only 20% lipid with the low C/N ratio. The fatty acid composition of the alga Chlorella saccharophila was investigated under different growth conditions. Using glucose as the sole carbon source, heterotrophically-grown Chlorella saccharophila produced a greater proportion of the polyunsaturated fatty acids (C18: 2 and C18: 3) than photosynthetic cultures, with linoleic acid (C18: 2) predominating. An unexpected discovery was the observation that at the lowest glucose concentration (2.5 gl–1) the lipid content of the algae increased to between 36–47% of the cell weight, depending on the temperature. At glucose concentrations of 5 g l–1 or more, the lipid content fell to 10–12% of the cell, although total fatty acid yield was higher due to higher biomass concentrations. Aeration of heterotrophic cultures promoted the production of unsaturated fatty acids compared to non-aerated cultures.  相似文献

Porphyridium cruentum Näg. (clone 161) was found to grow best in medium containing between 0.45 M and 0.8 M NaCl. From studies done on growing cultures, the palmitic acid content of the cells decreased with increasing NaCl concentration of the medium. Conversely, when the culture was transferred from a 0.8 M NaCl medium to 0.2 M NaCl, the amount of palmitic acid in thePorphyridium cells increased with time of incubation and it contributed up to 64.5% of the total fatty acid content. There appears to be a negative correlation between the cellular content of palmitic acid and the growth lag. The oleic acid content varied only marginally with increasing NaCl concentration. The poly-unsaturated acid content (linolenic and arachidonic acids) decreased initially and then increased with NaCl concentration up to and beyond ca. 0.8 M NaCl respectively. At 1.5 M NaCl, the poly-unsaturated fatty acids amounted to 78.2% of the total fatty acids in the cell. For stationary phaseP. cruentum cultures, a similar relationship existed between fatty acids and NaCl concentration. However, palmitic acid was accumulated up to three-fold more when compared to the exponential culture grown in low salinity. The nitrate transporter from Chlorella sorokiniana (accession number AY026523) has been cloned by screening a cDNA library based on mRNA isolated after 30 min treatment of Chlorella with 5 mM nitrate and with a RT-PCR product (730 bp) as a probe. The Chlorella sequence has similarity to known nitrate transporters of the NRT2 family (high-affinity nitrate transporters). The cDNA clone was used for functional expression in Xenopus oocytes and a nitrate-dependent current was measured at pH 5.5 but not at pH 7.4. A second algal gene or a second gene product was not needed for functional expression in Xenopus. Inhibitor studies in Chlorella indicated that protein phosphorylation/dephosphorylation is involved in nitrate induction of ChNRT2.1. In addition to nitrate, ChNRT2.1 expression is induced by nitroprusside, a NO donor, and is affected by glucose.  相似文献

In Mycobacterium phlei, fatty acid unsaturation increased with decreasing temperature. The 10-hexadecenoic acid content increased as the temperature was reduced from 35°C to 26–20°C. At lower temperatures tuberculostearic acid decreased while oleic and linoleic acids increased, the latter being found in M. phlei for the first time. Concomitantly palmitic acid content decreased, and the 6- and 9-hexadecenoic acids increased slightly on reducing the temperature from 35 to 10°C. Thus, down to 26–20°C palmitic acid was mainly replaced by 10-hexadecenoic acid. From this range down to 10°C, palmitic and tuberculostearic acids were replaced by oleic and linoleic acids. Consequently, fatty acid branching decreased and mean chain length increased, as the temperature was reduced. The effects of changes in fatty acid composition of the cell membrane on different biological functions ofSalmonella typhimurium have been studied with the help of a temperature sensitive fatty acid auxotroph which cannot synthesise unsaturated fatty acids at high temperature. On being shifted to nonpermissive temperature the cells continue growing for another one and half to two generations. The rates of protein and DNA syntheses run parallel to the growth rate but the rate of RNA synthesis is reduced. Further, there is a gradual reduction in the rate of transport of exogenous uridine and thymidine into the soluble pool. The transport process can be restored by supplementing the growth medium with cis-unsaturated fatty acids but not trans-unsaturated ones although the growth of the cells is resumed by supplementation with eithercis or trans-unsaturated fatty acids. However, supplementation withtrans, trans-unsaturated fatty acids leads to only partial recovery of the transport process. The rate of oxygen uptake is also affected in cells grown in the presence of thetrans-unsaturated fatty acids, elaidic acid and palmitelaidic acid. Analysis of cells grown under different fatty acid supplementation indicate that fatty acid composition of the cell membrane, especially the ratio of unsaturated to saturated fatty acids varies with temperature shift and supplementation of the growth media with fatty acids.  相似文献

The polar lipids and fatty acid composition ofThermus aquaticus YT-1 and YS 041,T. filiformis Wai33 A1 and eighteen isolates from New Zealand, several of which are attributed toT. filiformis, were compared to complement the taxonomy of these organisms. The polar lipid patterns were essentially similar in all strains and consisted of one major phospholipid and one major glycolipid. The fatty acid analysis produced three basic groups corresponding toT. filiformis Wai33 A1,T. aquaticus and the third to the other New Zealand strains. 采用f/2培养基,在3000lx,5000lx,8000lx光照强度时对杆状裂丝藻(Stichococcus bacillaris,MACC(中国海洋大学微藻种质库)/C19)和3株小球藻(Chlorellasp.MACC/C95,MACC/C97和MACC/C102)等4株海洋绿藻的总脂含量及脂肪酸组成进行了研究。单因子方差分析结果表明,光照强度对4株海洋绿藻的相对生长率、EPA(二十碳五烯酸,20∶5(n-3))含量及PUFA(多不饱和脂肪酸)总量,C19、C97和C102的脂肪含量和SFA(饱和脂肪酸)总量,C19、C95和C102的MUFA(单不饱和脂肪酸)总量均有显著影响(P0.05)。C19和C95的总脂含量随光照强度的增加总体呈降低趋势,C97和C102的总脂含量则随光照强度的升高而明显升高。C19的EPA含量随光照强度的增加呈现低-高-低趋势,C95在中低光照强度下EPA含量较高,C97和C102的EPA含量则随光照强度的增加而明显降低,4株绿藻的AA(花生四烯酸,20∶4(n-6))含量和PUFA总量均随光照强度的增加而降低。多重比较结果表明:C19在高光照强度(8000lx)时总脂含量显著低于其它各组,低光照强度(3000lx)和中光照强度(5000lx)处理间没有显著性差异(12.9%—12.7%),C97和C102则分别在高光照强度时总脂含量显著高于其它各处理(分别为40.6%和33.3%)。C19的EPA含量在中光照强度水平显著高于其它处理(12.7%),C97和C102的EPA含量分别在低光照强度水平显著高于其它处理(分别为12.0%和10.5%),C95的EPA含量在高光照强度水平显著低于其它处理,低光照强度和中光照强度处理间没有显著性差异(14.0%—14.7%);C19和C95的PUFA总量均在高光照强度水平显著低于其它处理,在低光照强度和中光照强度处理间没有显著性差异(分别为19.1%—19.4%和21.3%—21.3%),C97和C102的PUFA总量均在低光照强度水平显著高于其它处理(分别为19.2%和18.9%)。4株绿藻的主要脂肪酸成份为14∶0、16∶0、16∶1(n-7)、16∶4(n-3)、18∶1(n-9)、18∶3(n-3)、20∶4(n-6)和EPA。  相似文献

【目的】探究了以单酯磷D-葡萄糖-6-磷酸二钠(D-glucose-6-disodium phosphate,GP)为唯一磷源时含砷(As5+)水体中不同氮磷质量比对铜绿微囊藻(Microcystis aeruginosa)生长及砷代谢和微囊藻毒素(microcystins,MCs)释出的影响。【方法】将氮磷饥饿状态的藻细胞于含砷水体中不同氮磷比条件下进行实验,通过测定藻细胞密度(OD680)、叶绿素a(chlorophyll a,Chla)、实际光合产率(Yield)、超氧化物歧化酶(superoxide dismutase,SOD)、砷的存在形态以及微囊藻毒素含量,分析该藻在砷胁迫下的生理响应以及砷代谢机制。【结果】氮磷饥饿状态藻细胞对较高GP水平(0.1 mg/L)下的低氮磷比有更好的适应性,较低GP水平(0.02 mg/L)下的高氮磷比能显著促进培养初期藻细胞的OD680、Chla和Yield;SOD在培养初期与末期受氮磷比影响显著。GP环境下铜绿微囊藻经8 d培养后氮磷比为10:0.1介质中的砷表现为以亚砷酸盐(As^(3+))为主,占水体总砷(total arsenic,TAs)含量的78.8%,其余氮磷比环境中仍以As^(5+)为主,藻体砷形态则均以As^(5+)为主,氮磷比为1:0.1时有机砷占藻体TAs比例最高。藻细胞砷代谢受GP水平影响显著,较高GP环境(0.1 mg/L)下砷的代谢总量也更高,氮磷比为10:0.1时砷代谢以As^(5+)的还原和As^(3+)释出为主,低GP环境下(0.02 mg/L)砷代谢的甲基化水平提高。介质中MCs的含量与GP水平有关,较高GP的低氮磷比水体中MCs含量最低。【结论】研究结果对全面了解有机磷源含砷水体中藻华暴发及砷生态风险的科学管控具有重要意义。  相似文献

To clarify whether nutrients limit the growth of Microcystis aeruginosa (Kütz.) Kütz during the growing season in Lake Yogo, we examined the cellular ratios of carbon (C), nitrogen (N), and phosphorus (P) in the populations of M. aeruginosa from August to December 2001. We also measured cellular C, N, and P ratios of M. aeruginosa under batch culture conditions. The cellular levels of N and P of M. aeruginosa in natural population changed more than twofold. The atomic N: C ratio of natural populations of Microcystis fluctuated from 0.11 to 0.26. The atomic P: C ratio fluctuated from 0.0080 to 0.024. The N: C, P: C, and N: P ratios of exponentially growing M. aeruginosa in N-and P-rich medium were 0.19, 0.013, and 15 on average. The growth of M. aeruginosa was suppressed below the N: C ratio of 0.13 under the N-free condition and below the P: C ratio of 0.0026 in the P-free condition. In the natural population, the N: C ratio was low on August 1-2 (0.11) and the P: C ratio was low (less than 0.011) until September. The Microcystis population on August 1-2 was N limited, judging from the results of the culture experiment. In other periods, the population seemed to be supplied with a sufficient amount of N. Although the P: C ratio was low (approximately 0.01) during August and September, it was several times larger than the value of the reduction of growth rate that occurred in culture. P limitation did not occur during the study period. N became more of a limiting factor than P for the formation of blooms of Microcystis. No blooms were observed in August and September, in spite of the increase of cellular levels of N. The formation of Microcystis blooms in Lake Yogo seems to be affected by artificial manipulations such as pumping from Lake Biwa and outflow.  相似文献

The influence of growth temperature, media composition and cell age on the chemical composition of Bacillus stearothermophilus strain AN 002 has been determined. The total cellular protein decreased and the free amino acid content increased with growth temperature, in both exponential and stationary growth phase. The protein and free amino acid contents of cells were higher in the stationary phase than in the exponential phase, irrespective of growth temperature and media composition. The RNA content was only reduced in cells grown at 55° C. No significant variations were observed in the DNA and carbohydrate contents with respect to growth temperature and cell age. The total lipid and fatty acid compositions on the other hand varied as a function of growth temperature, cell age and media composition. Differences in the relative concentrations of even, odd and branched chain fatty acids were noticed. Novariation was observed in the antiiso and unsaturated fatty acids with respect to growth temperature. The unique variations in the fatty acid composition and total lipids at the growth temperature of 50° C and their variations in the stationary growth phase seem to be characteristic for B. stearothermophilus AN 002.  相似文献

Supplementing dairy cow diets with oilseed preparations has been shown to replace milk saturated fatty acids (SFA) with mono- and/or polyunsaturated fatty acids (MUFA, PUFA), which may reduce risk factors associated with cardio-metabolic diseases in humans consuming milk and dairy products. Previous studies demonstrating this are largely detailed, highly controlled experiments involving small numbers of animals, but in order to transfer this feeding strategy to commercial situations further studies are required involving whole herds varying in management practices. In experiment 1, three oilseed supplements (extruded linseed (EL), calcium salts of palm and linseed oil (CPLO) and milled rapeseed (MR)) were included in grass silage-based diets formulated to provide cows with ~350 g oil/day, and compared with a negative control (Control) diet containing no supplemental fat, and a positive control diet containing 350 g/cow per day oil as calcium salt of palm oil distillate (CPO). Diets were fed for 28-day periods in a 5×4 Latin Square design, and milk production, composition and fatty acid (FA) profile were analysed at the end of each period. Compared with Control, all lipid supplemented diets decreased milk fat SFA concentration by an average of 3.5 g/100 g FA, by replacement with both cis- and trans-MUFA/PUFA. Compared with CPO, only CPLO and MR resulted in lower milk SFA concentrations. In experiment 2, 24 commercial dairy farms (average herd size±SEM 191±19.3) from the south west of the United Kingdom were recruited and for a 1 month period asked to supplement their herd diets with either CPO, EL, CPLO or MR at the same inclusion level as the first study. Bulk tank milk was analysed weekly to determine FA concentration by Fourier Transform mid-IR spectroscopy prediction. After 4 weeks, EL, CPLO and MR all decreased herd milk SFA and increased MUFA to a similar extent (average −3.4 and +2.4 g/100 g FA, respectively) when compared with CPO. Differing responses observed between experiments 1 and 2 may be due in part to variations in farm management conditions (including basal diet) in experiment 2. This study demonstrates the importance of applying experimental research into commercial practice where variations in background conditions can augment different effects to those obtained under controlled conditions.  相似文献

The influence of salinity, nutrient level and soil aeration on the transpiration coefficient, defined as amount of water transpired/unit biomass produced (transpiration/biomass ratio) of carrots was investigated under non-limiting conditions with respect to water supply.Under optimum conditions and favorable nutrient supply, the transpiration coefficient amounted to 280–310 g H2O g–1 storage root dry weight (RDW). The transpiration coefficient did not change significantly up to salt concentration of 16 mS cm–1 in the soil solution under otherwise optimum conditions. Higher salt concentrations or low nutrient levels increased the transpiration coefficient to values of 390–540 g H2O g–1 RDW. It is suggested that the transpiration coefficient is not affected by salinity as long as toxic effects and nutrient imbalances do not occur. The transpiration coefficient was not increased by impeded soil aeration. Biomass production was more negatively influenced by adverse soil conditions (salinity, low nutrient level, impeded soil aeration) than was the transpiration coefficient.  相似文献

为探究不同C/N比对杉木凋落叶分解特性的影响,通过添加外源氮,采用分解袋法,分析林下植被保留和林下植被去除2种林分中不同C/N比值的杉木凋落叶分解300 d时细菌群落结构差异。研究显示:一定范围内,初始C/N比降低有利于杉木凋落叶分解和细菌群落多样性提升,过低的C/N比则具有抑制作用;相比于林下植被去除,林下植被保留管理措施更有利于杉木凋落叶的分解,且细菌群落多样性更高,但当初始C/N比值为20.3时,杉木凋落叶分解所受的抑制作用更为明显;在门水平上,变形菌门、放线菌门和浮霉菌门为杉木凋落叶中主要优势种群;在属水平上,慢生根瘤菌属、嗜酸栖热菌属和Singulisphaera属在杉木凋落叶中相对丰度较高;不同处理间的杉木凋落叶细菌群落结构具有显著差异,变形菌门、放线菌门和酸杆菌门等细菌门中的多类细菌相对丰度差异显著;在门分类水平上,杉木凋落叶C/N比值与主要细菌种群相对丰度的相关性最高,C/P比值与杉木凋落叶细菌群落结构变异的相关性最高;在属水平上,杉木凋落叶全碳含量、C/N比值和全钾含量与主要细菌种群相对丰度的相关性最高,全钾含量与杉木凋落叶细菌群落结构变异的相关性最高。结果表明初始C/N比的降低改变了杉木凋落叶细菌群落结构,进而作用于杉木凋落叶的分解。  相似文献

Biochemical assessment of liver damage during ethanol-induced stress was done by measuring the activities of serum enzymes, viz., aspartate transaminase (AST) and alkaline phosphatase (ALP), which were significantly elevated in rats fed ethanol. Ethanol administration for a period of 60 days modifies the fatty acid composition, and the analysis of fatty acids showed that there was a significant increase in the concentrations of palmitic acid (16:0), stearic acid (18:0), and oleic acid (18:1) in liver, kidney, and brain, whereas the concentrations of palmitoleic (16:1) and arachidonic acid (20:4) were significantly decreased. The breakdown products of arachidonic acids (20:4), prostaglandins, were elevated. The antioxidants curcumin and N-acetylcysteine (NAC) decreased the activities of serum AST and ALP. Curcumin and NAC decreased the concentrations of fatty acids, viz., palmitic, stearic, and oleic acid, whereas arachidonic acid and palmitoleic acid were elevated. The prostaglandin concentrations were also decreased after curcumin and N-acetylcysteine treatment. Thus the present investigation shows that curcumin and N-acetylcysteine prevent the fatty acid changes produced by ethanol and also reduce the inflammatory response of ethanol by reducing the level of prostaglandins.
