Effects of bird ingestion on seed germination of Sorbus commixta   总被引:1,自引:0,他引:1  
To determine the effects of ingestion by birds on seed germination, we performed germination experiments in the field and laboratory with Sorbus commixta. The germination of four groups of seeds was compared: ingested seeds, seeds defecated in feces after feeding of fruits to birds; extracted seeds, seeds deliberately extracted from the fruit pulp; juiced seeds, seeds plus the juice of the pulp after seeds had been deliberately extracted from the pulp; intact seeds, seeds in untreated intact fruits. In the laboratory, intact and juiced seeds hardly germinated, but ingested and extracted seeds germinated. Thus, the pulp and its juice appeared to inhibit germination, but seeds could germinate without ingestion by birds once the seeds had been manually extracted from the pulp. In the field, intact fruits did not germinate in the first spring, because the seed was still covered with pulp. The pulp of intact seeds decomposed during the first summer, and thus, the seeds had the potential to germinate during the second spring. In fact, most intact seeds do not germinate during the second spring either, since they lose their viability during the first summer. Thus, under natural conditions, most seeds of Sorbus commixta cannot germinate without bird ingestion. Received: 5 July 1997 / Accepted: 7 November 1997  相似文献

Summary Amino acid, polyamine and protein concentrations in seeds and their evolution during seed germination of two dipterocarp species, Hopea odorata and Dipterocarpus alatus, were determined with the help of a multianalytical system. Glutamic acid and glutamine were the major amino compounds present. Hopea seeds also contain high levels of aspartic acid/asparagine, serine, threonine, arginine and alanine, while those of Dipterocarpus contain high levels of alanine, arginine and threonine. These species were quite different in their germination behavior and thus in their protein and amine metabolism rates. In Hopea, polyamines increased during the first 3 days of germination and reached a maximum by the 3rd day, 1 day before maximum germination rate. In Dipterocarpus polyamines reached their maximum at the 6th day while maximum germination rate is observed by the 7th day. This suggests that polyamine compounds could play a role in the early part of the germination process in Hopea and Dipterocarpus seeds. The possibility that control of polyamine biosynthesis could be used for the establishment of biochemical methods to improve seed storage and to control germination of these recalcitrant seeds is discussed.  相似文献

Cytisus scoparius and Genista florida regenerate after fire by stump-sprouting but also by seed. Seeds of these species were heated to a range of temperatures similar to those registered on the surface soil during natural fires (from 50 to 150 °C) and a range of exposure times (from 1 to 15 min). No germination was observed at high temperatures, 130 °C, when the exposure time was 5 min or more. However, moderate heat treatments (at 70 and 100 °C) significantly increased the rate of germination relative to controls. Cytisus scoparius is more favoured by fire action than Genista florida, with germination rates slightly greater following 100 °C for 5 min and 130 °C for 1 min than after mechanical scarification.  相似文献

Summary The effects of temperature, photoperiod, phytochrome photoreversion and the response to a R/FR ratio gradient were investigated in seeds of four species from two contrasting tropical habitats; two species from a rain forest (Cecropia obtusifolia and Piper umbellatum) and two from a high altitude lava field covered by low vegetation (Buddleja cordata and Chenopodium ambrosioides). In the rain forest seed species the photoblastic response seems to be adapted to light quality changes due to canopy destruction, on the other hand, the lava field seed species seem to be adapted to instantaneous light stimulus such as would be produced by the sudden exposure of a buried seed to the soil surface light environment.  相似文献

Soaking seeds of cucumber and pumpkin with an extract of Westiellopsis prolifica, an N2-fixing cyanobacterium, promoted germination and their subsequent growth and development. Characterisation of novel S-alleles from cherry (Prunus avium L.)   总被引:1,自引:0,他引:1  
In plant populations exhibiting gametophytic self-incompatibility, individuals harbouring rare S alleles are likely to have a reproductive advantage over individuals having more common alleles. Consequently, determination of the self-incompatibility haplotype of individuals is essential for genetic studies and the development of informed management strategies. This study characterises six new S alleles identified in wild cherry (Prunus avium L.). Investigations to determine the S genotype of individuals in recently planted woodland through length polymorphisms of introns associated with the stylar S-RNase gene and the pollen SFB gene revealed six S intron profiles which did not correspond to those of known S alleles. These are now attributed to S 27 to S 32 . Consensus primers, annealing in the S-RNase sequence coding for the signal peptide and C5 regions, were used to isolate the S-RNase alleles associated with the novel S intron profiles. The proteins corresponding to the new alleles were separated by isoelectric focusing from stylar extracts and their pI values determined. Similarities between the deduced amino acid sequence for the new alleles isolated and other cherry S-RNase sequences available on the databases ranged from 40% to 86%. Amplification products for SFB introns ranged from 172 to 208bp. New sequence regions exposed to positive selection were identified and the significance of the PS3 region reinforced. A phylogenetic relationship between P. avium S-RNases for S 10 and S 13 and between corresponding SFB alleles may indicate co-evolution of allele specificities of these two genes. We combined laboratory and nursery experiments to analyse the effectiveness of sheep as endozoochorous seed dispersers of six native shrubby Cistaceae species collected in SE Spain (Helianthemum apenninum (L.) Mill., H. violaceum (Cav.) Pers., Fumana ericoides (Cav.) Grand., F. thymifolia (L.) Spach, Cistus monspeliensis L. and C. laurifolius L.), considering the main stages after seed ingestion, i.e. seed recovery, seed germination, seedling emergence and early seedling establishment. Seed recovery after gut passage was high (around 40%) for all the species, except F. thymifolia (12%). Most seeds (ca. 90%) were recovered within 48 h after ingestion for all the species, although seeds were still recovered up to 96 h after ingestion. Gut passage increased germination up to seven-fold compared to non-ingested seeds. Furthermore, seedling emergence from seeds contained in pellets was overall similar (intact pellets) to or higher (crumbled pellets) than emergence from seeds without dung. Survival of emerged seedlings and mass of seedlings after 20 days were not reduced by dung. Sheep act therefore as effective dispersers of these Cistaceae species by scattering seeds and promoting germination, while faeces do not hamper seedling establishment. We conclude that the interaction between herbivorous ungulates and these dry-fruited species may be considered a mutualism qualitatively similar to the mutualism between frugivorous vertebrates and fleshy-fruited plants.  相似文献

This study examined seasonal variations in the content of water-soluble phenolics extracted from litter and Ah horizon of forest soil under Pinus laricio Poiret trees. After having identified and quantified different phenolics, the effects of seasonal phenolic extracts, single phenolic acids and synthetic phenol mixtures on seed germination of Pinus laricio Poiret, Pinus pinaster Aiton and Pinus halepensis Mill. were evaluated, and the activity of phenolics on the main enzymes involved in the glyoxylate cycle was tested. The results indicated a seasonal variation of phenol content in soil, with largest concentrations of water-soluble phenolic acids in autumn and smallest concentrations in summer. Addition of phenol extracts to germination medium reduced seed germination showing phytotoxic effects which differed, depending on the species and the fractions tested. Phenols extracted from litter and Ah horizon in autumn and winter, vanillic acid, and synthetic mixtures reproducing phenols extracted from soil in winter inhibited seed germination of Pinus laricio, pinaster and halepensis more than other treatments. A greater inhibitory effect was observed on seed germination of Pinus pinaster and halepensis compared to Pinus laricio. Summary Maximum germination response and root + shoot yields of Acacias were obtained at daily irrigation. Any deviation from daily irrigation resulted in variations of performance (germination and growth yield) between the six Acacia species studied. Species of relatively dry habitse.g. A. nubica andA. seyal were more sensitive to water supply compared with those of relatively wet habitatse.g. A. nilotica andA. albida. This was reflected at the establishment phase by the greater production of root + shoot yields of species of drier zones than by those of wetter ones. Antiserum to a protein fraction of an extract of mature styles of P. avium cv. Lambert (S3S4) was raised in rabbits. Two major antigenic components of the style extracts were detected by immunoelectrophoresis and immunodiffusion. The presence of one antigen (S-antigen) correlated with a particular S-genotype (S3S4). This antigen is restricted to mature styles of P. avium. The second antigen (P-antigen) was detected in styles of all Prunus species examined, but not in styles of other species of the Rosaceae. The S-antigen is positively charged and the P-antigen negatively charged at pH 8.8.  相似文献

We report Agrobacterium tumefaciens-mediated transformation of two Prunus salicina varieties, 'Angeleno' and 'Larry Anne', using a modification of the hypocotyl slice technique previously described for P. domestica. Regeneration rates on thidiazuron (TDZ) and indole-3-butyric acid (IBA) supplemented Murashige and Skoog (MS) media reached 11% for 'Angeleno' and 19% for 'Larry Anne' hypocotyl slices. Transformation using Agrobacterium tumefaciens GV3101 harboring a plasmid with the neomycin phosphotransferase II (nptII) and the green fluorescent protein (gfp) genes produced ten independent lines, six from 'Angeleno' and four from 'Larry Anne', representing transformation efficiencies of 0.8 and 0.3%, respectively, relative to the initial number of hypocotyl slices. Plants of six lines were found to produce the transgene encoded mRNAs. DNA blotting demonstrated the presence of transgene sequences in trees from five lines after 18 months of growth in the greenhouse.  相似文献

In vitro cultures of three Prunus clones (d. 1869, GF 677 and CAB 11E) were successfully stored at +8°, +4° and-3°C following the proliferation phase.Survival of cultures was dependent upon interactions of storage temperature, light, and age of subculture. Up to 100% of the cultures survived at the end of the trials after 170 (at +4°C) and 200 (at-3°C) days storage. Complete dardness appeared more suitable than 16-h (hour) photoperiod for successful storage at-3°C for up to 10 months. One or two weeks in normal growth room vefore storage at-3°C for up to 10 months. One or two weeks in normal growth room before storage enhanced the survival S-1. The dynamics of mutualistic interactions involving more than a single pair of species depend on the relative costs and benefits of interaction among alternative partners. The neotropical myrmecophytes Cordia nodosa and Duroia hirsuta associate with several species of obligately symbiotic ants. I compared the ant partners of Cordia and Duroia with respect to two benefits known to be important in ant-myrmecophyte interactions: protection against herbivores provided by ants, and protection against encroaching vegetation provided by ants. Azteca spp., Myrmelachista schumanni, and Allomerus octoarticulatus demerarae ants all provide the leaves of Cordia and Duroia some protection against herbivores. However, Azteca and Allomerus provide more protection than does Myrmelachista to the leaves of their host plants. Although Allomerus protects the leaves of its hosts, plants occupied by Allomerus suffer more attacks by herbivores to their stems than do plants occupied by other ants. Relative to Azteca or Allomerus, Myrmelachista ants provide better protection against encroaching vegetation, increasing canopy openness over their host plants. These differences in benefits among the ant partners of Cordia and Duroia are reflected in the effect of each ant species on host plant size, growth rate, and reproduction. The results of this study show how mutualistic ant partners can differ with respect to both the magnitude and type of benefits they provide to the same species of myrmecophytic host.  相似文献

A protocol for Agrobacterium-mediated stable transformation for scored, whole leaf explants of the apricot (Prunus armeniaca) cultivar Helena was developed. Regenerated shoots were selected using a two-step increased concentrations of paromomycin sulphate. Different factors affecting survival of transformed buds, including possible toxicity of green fluorescent protein (GFP) and time of exposure to high cytokine concentration in the regeneration medium, were examined. Transformation efficiency, based on PCR analysis of individual putative transformed shoots from independent lines was 5.6%, when optimal conditions for bud survival were provided. Southern blot analysis on four randomly chosen PCR-positive shoots confirmed the presence of the nptII transgene. This is the first time that stable transformation of an apricot cultivar is reported and constitutes also one of the few reports on the transformation of Prunus cultivars.  相似文献

A genetic linkage map of Prunus has been constructed using an interspecific F2 population generated from self-pollinating a single F1 plant from a cross between a dwarf peach selection (54P455) and an almond cultivar Padre. Mendelian segregations were observed for 118 markers including 1 morphological (dw), 6 isozymes, 12 plum genomic, 14 almond genomic and 75 peach mesocarp specific cDNA markers. One hundred and seven markers were mapped to 9 different linkage groups covering about 800 cM map distance, and 11 markers remained unlinked. Three loci identified by three cDNA clones, PC8, PC5 and PC68.1, were tightly linked to the dw locus in linkage group 5. Segregation distortion was observed for approximately one-third of the markers, perhaps due to the interspecific nature and the reproductive (i.e. self-incompatibility) differences between peach and almond. This map will be used for adding other markers and genes controlling important traits, identifying the genomic locations and genetic characterizing of the economically important genes in the genus Prunus, as well as for markerassisted selection in breeding populations. Of particular interest are the genes controlling tree growth and form, and fruit ripening and mesocarp development in peach and almond.  相似文献

The dynamics of N uptake and N partitioning in peach (Prunus persica, Batsch) trees of a very early (cv. Flordastar) and a very late (cv. Tudia) fruit ripening varieties grown under a mediterranean climate was assessed during one season. Labelled N was applied to two-year old potted trees which were destructively harvested at regular intervals during the vegetative and reproductive cycle. Tree phenology as well as vegetative and reproductive growth of the two genotypes strongly differed: bud burst started in late January in Flordastar and late March in Tudia. Leaf senescence in Flordastar was almost complete by mid October, while Tudia still retained a significant fraction of leaves at the December harvest. Fruit yield averaged 4.0 and 6.9 kg tree–1 (fresh weight) in cv. Flordastar and Tudia, respectively, and fruit size was within commercial standards for the two genotypes. After growth resumption, shoot and fruit growth mainly relied on N remobilised from reserves, which accounted for 72–80% of total N in new growth. Nitrogen uptake by both genotypes was relatively low in the first month after bud burst, then was more rapid until the end of the season. Total labelled N uptake did not differ between the two genotypes and accounted on average for 65–70% of total N supplied. The kinetics of labelled N uptake were similar in the two varieties despite the great difference in the timing of their fruit ripening. Leaves were the main sink for N during much of the experimental period. The fruits, when present, also used a significant fraction of the absorbed N, which was almost constant until fruit ripening in Flordastar. Nitrogen partitioning to leaves declined progressively after summer, when a greater fraction of the absorbed N was recovered in the twigs, the trunk, the fine roots and especially in the coarse roots. The data provide evidence for guiding the kinetics of N supply to peach orchards under a Mediterranean climate.  相似文献

The effects of nickel were studied in two serpentine species with different metal tolerance strategies:Silene italica L., which limits nickel uptake and translocation, andAlyssum bertolonii Desv., a serpentine endemic, which accumulates nickel mostly in the leaves. InS. italica, nickel 7.5 μM inhibited root growth and depressed mitotic activity in root tips. Peroxidase activity and phenol concentration both in roots and shoots were increased; under the same conditions nickel did not produce any relevant effect onA. bertolonii. InS. italica an adequate calcium concentration (25 mM) was able to reverse the effects of nickel on root growth and metabolism. InA. bertolonii the same calcium concentration reduced root growth, confirming this species adaptation also to low calcium concentrations, typical of serpentines.  相似文献

A collection of bacterial strains obtained from a wide-range origin was screened for ability to promote growth in two types of Prunus rootstocks in a commercial nursery. Only few strains promoted growth significantly and consistently, and a strong specificity for the rootstock cultivar was observed. Irrigation of plants with Pseudomonas fluorescens EPS282 and Pantoea agglomerans EPS427 significantly increased plant height and root weight of the plum Marianna 2624 and the peach–almond hybrid GF-677, respectively. Plant height showed a higher rate of growth in early stages of development (2.6–3.5 times the non-treated controls), but the effect decreased with plant age. However, in aged plants growth promotion was more significant on root weight (1.9 times the non-treated controls) than on plant height. The efficacy of growth promotion and the persistence of strains in the root environment were dependent on the bacterial inoculum concentration applied. Increases in root development were maximum at inoculum concentrations of up to 8 log10 CFU ml–1 (ca 10 log10 CFU L–1 of potting mix). Population levels at the optimum inoculum concentration were around 7 log10 CFU g f.w.–1 root material at early stages of development and decreased to 4 log10 CFU g f.w.–1 after several months of development. The best plant growth-promoting strains were very diverse in secondary metabolite production and antagonistic ability against several plant pathogens.  相似文献

The pollen tube of Prunus avium (cherry) consists of a growth zone of vesicles at the tip and an assemblage of organelles typical of an actively metabolizing cell. Electron opaque globules are closely associated with the plasma membrane and fibrillar cell wall layer at the tip. Acid phosphatase (EC 3.1.3.2) activity is localized in the membranes of 120 nm vesicles and ER system, the lumen of 50 nm vesicles, the plasma membrane and the tube nucleus.  相似文献
