When C14 carboxyl indoleacetic acid (IAA) is transported through Avena coleoptile sections a fraction of the activity becomes bound. The nature of this bound IAA has been investigated. Upon extraction with solvents and chromatography a substance having the RF of IAA in 4 solvents was detected. No evidence could be found for the formation of indoleacetyl conjugates. In pea stem sections subjected to a similar experimental regime good evidence was obtained for the occurrence of conjugates. When IAA was supplied exogenously to coleoptile sections floating in solutions the occurrence of conjugates was shown to be dependent on the presence of the primary leaf. In its absence no conjugates could be detected.

On grinding coleoptile sections and subsequent centrifugation at 240 × g the radioactivity was found to be in the tissue fraction as opposed to the supernatant. The radioactivity cannot be removed from the tissue by extraction with water, buffer solution or treatment with ribonuclease. It is readily removed by 10% urea, crystalline trypsin and chymotrypsin. It is therefore concluded that IAA becomes bound to a protein. Bound IAA does not appear to be able to cause growth in Avena coleoptile sections.

We have characterized the transport of [3H]indoleacetic acid (IAA) in intact corn (Zea mays L.) coleoptiles. We have used a wide range of concentrations of added IAA (28 femtomoles to 100 picomoles taken up over 60 minutes). The shape of the transport curve varies with the concentration of added IAA, although the rate of movement of the observed front of tracer is invariant with concentration. At the lowest concentration of tracer used, the labeled IAA in the transport stream is not detectably metabolized or immobilized, curvature does not develop as a result of tracer application, and normal phototropic and gravitropic responsiveness are not affected. Therefore we believe we are observing the transport of true tracer quantities of labeled auxin at this lowest concentration.  相似文献

InHordeum vulgare cultivar “Kirin-choku No. 1”, the final length of intact coleoptiles of totally etiolated seedlings was approximately twice as long as that of those grown under continuous red light. The fluence response curve of the latter was biphasic; the low-energy effect was saturated by red light of ca. 50 J m−2 which gave rise to about 40% of the maximum inhibition by continuous irradiation with red light of 1.2 W m−2, whereas the high-energy effect was induced by irradiation for 1 hr or longer. Coleoptiles of 3-day-old seedlings were most sensitive to light causing the low-energy effect, which was repeatedly red/far-red reversible. The growth inhibition was correlated to the photometrically measured percentage of Pfr so that the maximum effect was induced by red light of 50 J m−2 which transformed 70% of phytochrome to Pfr in the coleoptile tip. Wavelength dependence of the high-energy effect showed that monochromatic light of 400, 600 and 650 nm greatly inhibited the coleoptile growth, whereas light of 700 and 750 nm promoted it instead. The effect was also induced by intermittent irradiation with red light, and the more frequently the intermittent treatment was given, the more the growth was inhibited.  相似文献

The effect of indoleacetic acid (IAA) and fusicoccin (FC) on the breakdown of phosphatidylinositol in maize (Zea mays L.) coleoptiles has been studied. Coleoptiles were able to incorporate [3H] myo-inositol into the phospholipid fraction almost linearly for 8 hours. Thin layer chromatography analysis of total phospholipids showed that [3H]myo-inositol was incorporated only into phosphatidylinositol. Prelabeled coleoptiles treated with IAA showed a loss of the radioactivity incorporated in the phospholipid fraction, whose level decreased by 34% after 1 hour. Treatment with FC, on the contrary, did not modify the content of labelled phosphatidylinositol with respect to the control. The different effects of IAA and FC and a possible mechanism of IAA action on growth are discussed.  相似文献

Indoleacetic acid at 0.017 millimolar inhibited the formation of three peroxidase isoenzymes in both soluble and wall-bound enzyme fractions of wheat coleoptile (Triticum vulgare) tissue. Hydroxyproline at 1 millimolar prevented the indoleacetic acid-induced inhibition. Indoleacetic acid oxidase activity in the soluble fraction was decreased by indoleacetic acid and was restored by hydroxyproline. Most of the indoleacetic acid oxidase activity was located in the electrophoretic zones occupied by two of the peroxidase isoenzymes influenced by indoleacetic acid and hydroxyproline. At least part of the effect of hydroxyproline on auxin-induced elongation of coleoptile tissue may be through control of auxin levels by indoleacetic acid oxidase.  相似文献

We found that the auxin-induced growth is mediated through the activation of the dictyosomes (collectively, the Golgi apparatus). Incubation of oat (Avena sativa) coleoptile segments in indoleacetic acid-sucrose-phosphate buffer changes significantly the number of dictyosomes in the expanding cells. A further indication of auxin enhancement of dictyosome activity is a decrease in dictyosomal cisternae (flattened membranous sacs) number. This decrease occurred after 6 minutes of incubation in auxin, and then was followed by a reduction in the organelle number per se. These times are in keeping with the rapid action of auxin-induced cell elongaton, and the latent period of geotropism. In the apical cells, the effect of indoleacetic acid is more subtle and complex. The periods of increased dictyosome utilization and of increased dictyosome synthesis in auxin-treated segments alter with those of the control. These observations indicate that dictyosomes not only have a function in cell elongation, but also may participate in processes such as auxin transport and stimuli perception. The expanding cells have five times as many dictyosomes as the cells in the apex. Dictyosome number within a cell appears to be directly proportional to the length of the cell. The fluctuation of dictyosome number and the effect of auxin on the rate of elongation of individual outer epidermis are discussed.  相似文献

Proton excretion induced by optimal concentrations of indoleacetic acid (IAA) and fusicoccin (FC) differs not only in maximum rate of acidification but also in the lag before onset of H+ excretion and in sensitivity to cycloheximide. Because these differences might simply be a consequence of the difference in rate of proton excretion, FC and IAA have now been compared using oat coleoptiles (cv. Victory) under conditions where the rates of acidification are more similar, i.e. suboptimal FC versus optimal IAA. As the concentration of FC is reduced, the rate of H+ excretion decreases, the final equilibrium pH increases, and the lag before detectable acidification increases up to 7-fold. This enhanced lag period is not primarily a consequence of wall buffering, inasmuch as it persists when a low concentration of FC is added to sections which were already excreting H+ in response to IAA. An extended lag also occurs, upon reduction of FC levels, in the hyperpolarization of the membrane potential, before enhancement of O2 uptake and before the increased rate of Rb+ uptake. The presence or absence of a lag is not a distinguishing feature between FC and IAA actions on H+ excretion and cannot be used to discriminate between their sites of action. In contrast, the insensitivity of FC-induced H+ excretion to cycloheximide, as compared with the nearly complete inhibition of this auxin effect by cycloheximide, persists even at dilute concentrations of FC. This seems to be a basic difference in H+ excretion by IAA and FC.  相似文献

Brief irradiation of intact etiolated seedlings of maize (Zea mays L.) with red light (R; 30 W cm-2, 10 min) reduces the amounts of diffusible and free (solvent-extractable) indole-3-acetic acid (IAA) obtainable from excised coleoptile tips. The effect is transient, the lowest level (30% of the dark control) occurring at about 3 h after irradiation. The free-IAA content of the whole coleoptile and the diffusible-IAA yield from the base of the same organ are similarly reduced, whereas the conjugated-IAA content of the coleoptile is not affected. These results support the view that R inhibits the production of IAA at the coleoptile tip. It is further shown that R inhibits biosynthesis of [3H]IAA from [3H]tryptophan supplied to the coleoptile tip. The shapes of the fluence-response curves obtained for the reduction of the diffusible-IAA yield by R and far-red light (FR) indicate the participation of two photoreactive systems. One has thresholds at 10-3 W s cm2 of R, five orders of magnitude less than the minimum required for the appearance of spectrophotometrically measurable far-red-absorbing form of phytochrome (Pfr) in vivo, and 10-1 W s cm-2 of FR; its response is linear to the logarithm of fluence exceeding five orders of magnitude. The other system is seen above 102 W s cm-2 as an increase in the slope of the fluenceresponse curve; its response is FR reversible and related to the Pfr level of total photoreversible phytochrome. Both systems inhibit biosynthesis of IAA from tryptophan. Elongation of the coleoptile is stimulated by R; the stimulation is most apparent in the apical region, and is saturated with a fluence at which bo detectable pfr is formed. Farred light can also saturate this response. The effects of red (R), far red (FR), or blue light (B) on the enhancement of nitrate reductase (NR) activity and on nitrate uptake in etiolated rice seedlings were examined. On 5-minute illumination followed by 12-hour dark, R caused marked increase of NR activity, but FR and B caused only slight increase. Illumination with 560 ergs per square centimeter per second of R for 5 minutes caused maximal increase. The effect of R was almost completely counteracted by subsequent illumination with 2,000 ergs per square centimeter per second of FR for 10 minutes, indicating that NR induction was mediated by phytochrome. Exogenous supply of inducer nitrate was not required during the 5-minute illumination and the R-FR cycles, if the seedlings were transferred to nitrate solution at the beginning of the dark incubation. NR activity in the shoots was found high when shoots were illuminated but was low when only roots were illuminated. On continuous illumination for 12 hours, B had more effect on NR increase than R.  相似文献

Indoleacetic acid oxidase in tobacco callus cultures (Nicotiana tabacum L., cv. White Gold) was composed of at least two groups of isoenzymes, which were distinctly different in electrophoretic mobilities and in responses to growth substances. Indoleacetic acid had dual effects; at low concentrations it promoted the development of two fast-migrating indoleacetic acid oxidase isoenzymes, but at high concentrations it increased the level of other indoleacetic acid oxidase isoenzymes with low and moderate electrophoretic mobilities. However, indoleacetic acid was not unique in such effects; 2,4-dichlorophenoxyacetic acid and 2,4,5-trichlorophenoxyacetic acid were effective at concentrations lower than that of indoleacetic acid.  相似文献

Activity of indoleacetic acid oxidase was shown to increase following a period of water stress. Two fractions of indoleacetic acid oxidase were extracted from plant extracts. Similarly, two protein peaks (determined by ultraviolet absorption) were isolated. One peak, associated with an indoleacetic acid oxidase peak, increased following water stress. The second peak, not associated with extractable indoleacetic acid oxidase, decreased after water stress. The results are discussed in terms of general growth effects.  相似文献

BACKGROUND AND AIMS: This work has been conducted to assist theoretical modelling of the different stages of the blue light (BL)-induced phototropic signalling pathway and ion transport activity across plant membranes. Ion fluxes (Ca(2+), H(+), K(+) and Cl(-)) in etiolated oat coleoptiles have been measured continuously before and during unilateral BL exposure. METHODS: Changes in ion fluxes at the illuminated (light) and shadowed (dark) sides of etiolated oat coleoptiles (Avena sativa) were studied using a non-invasive ion-selective microelectrode technique (MIFE). The bending response was also measured continuously, and correlations between the changes in various ion fluxes and bending response have been investigated. For each ion the difference (Delta) between the magnitudes of flux at the light and dark sides of the coleoptile was calculated. KEY RESULTS: Plants that demonstrated a phototropic bending response also demonstrated Ca(2+) influx into the light side approximately 20 min after the start of BL exposure. This is regarded as part of the perception and transduction stages of the BL-induced signal cascade. The first 10 min of bending were associated with substantial influx of H(+), K(+) and Cl(-) into the light (concave) side of the coleoptiles. CONCLUSIONS: The data suggest that Ca(2+) participates in the signalling stage of the BL-induced phototropism, whereas the phototropic bending response is linked to changes in the transport of H(+), K(+) and Cl(-).  相似文献

Effect of d-mannose treatment on different antioxidants, phenolics, protease activity, lipid peroxidation, DNA damage and cell death was investigated in coleoptiles of etiolated wheat seedlings. Modulations in these biochemical parameters were monitored up to 96 h after treatment at 24 h intervals. With accelerating effect on initial signs of cell death, i.e., appearance of long DNA fragmentation and no effect on initiation of terminal stage, i.e., internucleosomal nDNA fragmentation, mannose treatment (1 % = 56 mM) diminished the antioxidant activities in wheat coleoptiles. Mannose treatment decreased the catalase activity at all intervals, while APX and POD activities decreased at 72 h. Peroxidation of lipids increased at 72 h after mannose treatment. Levels of most of antioxidants, i.e., SOD, peroxidases and phenolics were raised during initial time period (24–48 h) of mannose treatment probably as an attempt to counter the stress effect. Protease activity gradually increased and protein content decreased with time in both treated and non-treated coleoptiles. Sharp decrease in CAT, APX and peroxidase activities and increase in lipid peroxidation at 72 h overlaps with apoptotic internucleosomal nDNA fragmentation in this organ. This coincidence points towards the importance of compromised antioxidant defense and involvement of reactive oxygen species in initiation of terminal stage of programmed cell death in wheat coleoptile. In conclusion, accelerating effect on DNA fragmentation and lipid peroxidation along with diminished antioxidant activities at the time of internucleosomal nDNA fragmentation, provide evidence for pro-apoptotic effect of d-mannose in wheat coleoptile.  相似文献

Nucleic acid was extracted by the SLS-phenol method from Phaseolusaureus hypocotyl treated with IAA-2-14C. Radioactivity in thenucleic acid fraction was found at the positions of sRNA andrRNA on an MAK column. IAA-14C was released from the radioactivecompound(s) in the sRNA fraction, by alkaline hydrolysis, butnot by ethanol extraction, or by dialysis to 2 M NaCl, 8 M urea,and 0.1 M EDTA. When the radioactive compound at the positionof sRNA on an MAK column was further re-chromatographed on aDEAE-cellulose column and on a BD-cellulose column, it was alwayslocalized only in a settled part of the fraction of each column.From this fraction IAA-14C was released by alkaline hydrolysis.Also, IAA-14C was released from the radioactive compound insRNA fraction, by RNase digestion, but not by pronase treatment.Results of these experiments suggest the existence of some kindsof sRNA binding IAA. The genesis of this sRNA binding IAA-14Cwas observed within 30 min after the supply of IAA-14C, andthe sRNA became saturated with IAA-14C about 2 hr after thebeginning of incubation. Summary The effect of exogenous KNO3, the terminal acceptor of electrons in oxygen-free medium, on mitochondrial ultrastructure and on the growth rate of 4-day-old rice coleoptiles under strictly anoxic conditions was studied. Exogenous nitrate (10 mM) did not exert any significant effect on the growth rate of coleoptiles of intact seedlings compared to their growth in KNO3-free medium. Anaerobic incubation of detached coleoptiles in KNO3-free medium for 48 h resulted in the complete destruction of mitochondrial and other cell membranes. In the presence of KNO3, no mitochondrial-membrane destruction was observed even after 48 h anoxia although the mitochondrial ultrastructure was modifed. Cristae were arranged in parallel rows and elongated dumbbell-shaped mitochondria appeared in some cells. The data obtained indicate a protective role of exogenous nitrate as electron acceptors in oxygen-free medium. The results of the present investigation are discussed and compared with reports of either markedly damaging or favorable effects of exogenous nitrate on the growth, metabolism, and energetics of rice and other plants under hypoxic and anoxic conditions.  相似文献

When 3–4 mm long coleoptiles of etiolated rice seedlings (cv. Koshijiwase) were irradiated with continuous red light their growth was seriously inhibited. If a brief exposure of red light (4×103 ergs cm−2) was given to the short coleoptiles, the growth rate dropped immediately after the irradiation, but the growth did not stop till the coleoptile reached some calculated length. If another brief red irradaition of the same order was given 24 hr after the first, the growth rate and the final length dropped further. The effect of red light was reversed by successively given far-red light, and this response was repeatedly red and far-red reversible. The escape reaction was rather slow so that photoreversibility was not lost at all by 8th hr, and 50% of the initial reversibility was lost within ca. 16 hr at 25±0.5 C. Blue light also induced the inhibition of coleoptile elongation, the effect was reversed by subsequent far-red irradiation, and this could be obtained repeatedly. Thus, the photoinhibition of the young coleoptile can be concluded to be under the control of phytochrome, and the mode of action appeared quite different from the previously reported results with longer coleoptiles.  相似文献
