Leaflets of 65 species ofCaesalpinia s.l. and seven species ofHoffmannseggia were studied in clearings supplemented by resin sections and scanning electron microscopy. Three types of secretory structure occurred among 46 species; in 43 species they were distributed mutually exclusively (external glands: 8 species; internal cavities: 5 species; idioblastic cells: 30 species); three other species each had two types. Species with secretory structures conform mostly to proposed subgenera and informal groups. Other unusual features were external glands with internal spaces, thickened walls or conspicuous localized wall thickenings in epidermal cells or mesophyll cells of certain species, and differentially stained epidermal cells surrounding stomata. Prismatic crystals predominate but druse crystals also occur.  相似文献

为探讨香樟(Cinnamomum camphora)叶肉含晶细胞超微结构的季节变化,阐明香樟叶肉中草酸钙晶体在春夏秋冬的变化规律。该研究以多年生香樟(C. camphora)叶片为材料,分别于春夏秋冬四个季节露地取样,制作超薄切片,用透射电子显微镜(TEM)观察叶肉含晶细胞超微结构的变化。结果表明:春季时香樟叶肉中只有少数细胞有草酸钙晶体,数量较少,晶体结构多为柱状晶、方晶; 夏季时香樟叶肉细胞中随机分布于液泡的草酸钙晶体明显比春季的数量多、体积大、形态丰富,晶体多为柱状晶、方晶、针晶、簇晶; 秋季时香樟叶肉细胞草酸钙晶体和夏季的类似,数量较多,形态多样,以方晶和柱状晶针晶为主,伴有晶簇; 冬季时香樟叶肉含晶细胞晶体形态为柱状晶、方晶、针晶,数量比夏季和秋季的数量略有减少。该研究结果表明在一年四季中香樟叶肉细胞液泡中均有草酸钙晶体结构存在。  相似文献

Calcium oxalate crystal types and distribution within leaves ofPrunus sensu lato (Rosaceae; Prunoideae) were surveyed from mostly herbarium specimens (196 specimens of 131 species of all five subgenera usually recognized). Rehydrated samples were bleached, mounted unstained, and viewed microscopically between crossed polarizers. Six patterns were recognized based on crystal type and relative distribution around veins and in mesophyll. Druses predominate in four subgenera, but prismatics are most common in subgenus Padus. Prunophora and Amygdalus, considered to be the most advanced subgenera, have virtually only druses, which are almost always associated with veins. Cerasus and Laurocerasus, intermediate subgenera, have the greatest diversity of patterns, but few species with prismatics. A trend is evident from mostly mesophyll prismatics in Padus to fewer prismatics and more druses of mixed distribution in Laurocerasus and Cerasus, to mostly druses restricted to veins in Amygdalus and Prunophora.  相似文献

We tested the hypothesis that avocado idioblast oil cells play a defensive role against herbivorous insects. Toxicities of the intact avocado idioblast oil cells and the extracted idioblast oil were compared for three insect herbivores. Spodoptera exigua (Hübner) larvae are generalists that do not feed on avocados. By contrast, Sabulodes aegrotata (Guenée) and Pseudoplusia includens (Walker) larvae are generalist herbivores that readily feed on avocados. All bioassays were performed at a naturally occurring concentration of idioblast oil cells (2% w/w). Choice experiments showed that S. exigua larvae avoided diet treated with avocado idioblast oil cells and consume more control than treated diet. In contrast, idioblast oil cells had no significant antifeedant effects on the adapted S. aegrotata and P. includens larvae. Subsequent experiments designed to assess resistance mechanisms separated pre-ingestive (behavioral) and post-ingestive (physiological) effects of the avocado idioblast oil cells, and the extracted idioblast oil, on the two adapted herbivores. Post-ingestive adaptation was the mechanism that allows feeding. Because the impact of the avocado idioblast oil cells was greatest on the performance of non-adapted S. exigua, additional experiments determined that larvae fed diet containing the oil cells had higher mortality and reduced larval growth compared to controls. Developmental times were significantly prolonged for the survivors. Thus, increased mortality, reduced developmental rates, and antifeedant activity in the non-adapted insect indicate that defense against non-adapted herbivores may be an important function of idioblast cells in avocados.  相似文献

Several anatomical characters in leaves were described, and their distribution determined, for 153 species ofPolygonum, mostly from herbarium specimens. Structures surveyed were epidermal (glandular and nonglandular trichomes, nodules, specialized parenchyma, stomatal apparatus) and internal (cavities, crystals, laticifer-like cells, nodules, subepidermal fibers). Cleared leaves were supplemented by resin-embedded sections and SEM preparations of selected species. No feature defines any taxonomic section, but some features occur only within one section. Laticifer-like cells, epidermal and internal nodules, resin cups, and unique epidermal and subepidermal cavities seem to be unknown elsewhere; other features (invaginated epidermal cells; enlarged crystal cells confined to paraveinal layer) are rare among angiosperms.  相似文献

The epidermal structures of the foliage leaves, prophylls (perigynia) and nuts of 26 species and one variety ofKobresia (24 species and one variety from the Himalaya and two species from outside Himalaya) were studied by a scanning electron microscope (SEM) with an aim of making clear the relationship among these species. Most of the epidermal structures of the leaf and prophyll showed generic or familial relationships. The epidermal structures of the nuts were found useful for showing inter-specific relationships among morphologically closely related species though these structures were not found to be consistent with the infrageneric classification ofKobresia recognized by Clarke (1894, 1908) and Kükenthal (1909). On the basis of the epidermal structures of the nuts revealed by SEM the species ofKobresia could be put into four groups.  相似文献

Summary Crystal idioblasts are cells which are specialized for accumulation of Ca2+ as a physiologically inactive, crystalline salt of oxalic acid. Using microautoradiographic, immunological, and ultrastructural techniques, the process of raphide crystal growth, and how crystal growth is coordinated with cell growth, was studied in idioblasts ofPistia stratiotes. Incorporation of45Ca2+ directly demonstrated that, relative to surrounding mesophyll cells, crystal idioblasts act as high-capacity Ca2+ sinks, accumulating large amounts of Ca2+ within the vacuole as crystals. The pattern of addition of Ca2+ during crystal growth indicates a highly regulated process with bidirectional crystal growth. In very young idioblasts,45Ca2+ is incorporated along the entire length of the needle-shaped raphide crystals, but as they mature incorporation only occurs at crystal tips in a bidirectional mode. At full maturity, the idioblast stops Ca2+ uptake, although the cells are still alive, demonstrating an ability to strictly regulate Ca transport processes at the plasma membrane. In situ hybridization for ribosomal RNA shows young idioblasts are extremely active cells, are more active than older idioblasts, and have higher general activity than surrounding mesophyll cells. Polarizing and scanning electron microscopy demonstrate that the crystal morphology changes as crystals develop and includes morphological polarity and an apparent nucleation point from which crystals grow bidirectionally. These results indicate a carefully regulated process of biomineralization in the vacuole. Finally, we show that the cytoskeleton is important in controlling the idioblast cell shape, but the regulation of crystal growth and morphology is under a different control mechanism.Abbreviation SEM scanning electron microscopy  相似文献

Comparative development of the narrow pinnules of rheophyticOsmunda lancea and of the broad pinnules of a related dryland species,O. japonica, was examined and the origin of rheophytic stenophylly was discussed. The mature leaves and their various parts ofO. lancea are smaller and narrower than those ofO. japonica. The young pinnules ofO. lancea at the initiation of cell expansion are smaller than those ofO. japonica. The growth pattern of the pinnules is fundamentally the same in the two species, but pinnule growth period is shorter inO. lancea than inO. japonica. While the largest growth rate in pinnule length is quite similar, inO. lancea the pinnules are less elongated and much less broadened during ontogeny. Cell expansion in the mesophyll and epidermis proceeds acropetally and toward the margin along the axes of costules and veins. Although the numbers of mesophyll and epidermal cells between two adjacent veinlets are almost the same inO. lancea andO. japonica, during the subsequent growth period inO. lancea, the cells expand to a smaller extent and the veinlets become more narrowly oblique to the costule. This oblique distortion of laminar segments framed by veins causes stenophylly, an allometric modification. The stenophylly ofO. lancea is believed to have arisen by heterochronic evolution, in particular, progenesis.  相似文献

Paraveinal mesophyll (PVM) is a distinctive anatomical feature of the leaf mesophyll of some plant taxa that may represent a specialized physiological compartment. A comprehensive review of the 42 published references that mention PVM or similar cell layers and a survey of 121 of the 272 species of all nine genera of thePhaseoleae subtribeErythrininae demonstrate that PVM is nearly exclusively found inLeguminosae. InLeguminosae, PVM is either rare or absent in subfamilyCaesalpinioideae, uncommon inMimosoideae, and extensively distributed amongPapilionoideae. In subtribeErythrininae, PVM is ubiquitous inErythrina, and occurs in four other genera. ThreeErythrininae genera (Apios, Mucuna, andCochlianthus) lack PVM. Unique chloroplast-poor, enlarged conical cells (pellucid palisade idioblasts) occur in 80 species ofErythrina but not in any other genus ofErythrininae.  相似文献

RFLPs of cpDNA were examined for 18 species ofAster, six species ofKalimeris, two species ofMiyamayomena and one species and one variety ofHeteropappus from Japan, using 16 restriction endonucleases. Approximately 275 restriction sites were surveyed, and a total of 74 restriction site mutations was detected, and 31 of these were phylogenetically informative. Sixteen most parsimonious trees constructed from Wagner parsimony analysis indicated the polyphyly ofKalimeris andMiyamayomena sensu Kitamura;K. miqueliana belongs to a different clade from the remaining species ofKalimeris, and two species ofMiyamayomena did not make a single clade. This result suggests that the shortening or loss of pappus have happened parallelly in different evolutionary lineages. We must be careful to assess the pappus character in taxonomy and phylogeny, and it is desirable to examine their phylogenetic relationships using a molecular data.  相似文献

This study investigated the cellular and subcellular compartmentation of Ni in the Eurasian serpentine species Alyssum murale, Alyssum bracteatum and Cleome heratensis and a non-serpentine population of A. murale (as a control) grown in hydroponic culture. Plant growth responses and Ni uptake clearly revealed the higher Ni tolerance of serpentine plants than the non-serpentine plants. Serpentine A. murale and A. bracteatum grew better at elevated (0.01 mM) Ni in the nutrient solution, supporting the view that the Ni hyperaccumulators have a higher requirement for Ni than normal plants. Low shoot Ni content of C. heratensis in response to the high Ni treatments indicated that this species employs an avoidance strategy for Ni tolerance. Energy-dispersive X-ray microanalysis showed that Ni was highly concentrated in the cell walls and cell lumen, most likely the vacuoles, of leaf epidermis of A. murale and A. bracteatum rather than in the mesophyll cells. EDX spectra from leaves of the non-serpentine A. murale suggested that Ni accumulated in both epidermal and mesophyll cells but not in the epidermal cell walls. Growth reduction and Ni toxicity in plants of the non-serpentine A. murale could be due to accumulation of Ni in the lumen of leaf mesophyll cells. Our data suggest that cellular and subcellular compartmentation are both possible mechanisms for Ni tolerance employed by the serpentine A. murale and A. bracteatum.  相似文献

Species in the subfamily Monotropoideae (family Ericaceae) are achlorophyllous and myco-heterotrophic. They have become highly specialized in that each plant species is associated with a limited number of fungal species which in turn are linked to autotrophic plants. This study provides an updated and comprehensive examination of the anatomical features of two species that have recently received attention with respect to their host-fungal specificity. Root systems of Monotropa uniflora and Pterospora andromedea collected from the field were characterized by light microscopy and scanning electron microscopy. All roots of both species were associated with fungi, each root having a well-developed mantle, paraepidermal Hartig net, and intracellular fungal pegs within epidermal cells. The mantle of M. uniflora was multi-layered and numerous outer mantle hyphae developed into cystidia of two distinct morphologies. Large calcium oxalate crystals were present, primarily on the mantle surface. The outer mantle of P. andromedea was more loosely organized, lacked cystidia, and had smaller plate-like as well as cylindrical crystals on the surface and between outer mantle hyphae. Fungal pegs in M. uniflora originated from inner mantle hyphae that penetrated the outer tangential wall of epidermal cells; in P. andromedea, these structures were initiated either from inner mantle hyphae or Hartig net hyphae and penetrated radial walls of epidermal cells. With respect to function, fungal pegs occurred frequently in both host species and, although presumed to be the sites of active nutrient exchange, no direct evidence exists to support this. Differences between these two monotropoid hosts, resulting from the mycorrhizal fungi with which each associates, are discussed.  相似文献

Summary The epidermal cell layer of the apical end of the ceras was investigated in two species of aeolid nudibranchs. Based on cellular inclusions, mostly two cell types were found: mucoid and ellipsoid-vacuolate cells. Mucoid cells ofCoryphella rufibranchialis have large heterogeneous and fibrillar secretory granules whereas inAeolidia papillosa, the granules are homogeneous, but vary in electron density from one cell to another. Ellipsoid-vacuolate cells contained large quantities of small vacuoles with an included ellipsoidal structure. Both species contained very numerous ellipsoid-vacuolate cells. Secretory granules and ellipsoid-vacuoles appear to arise from the Golgi apparatus and these contents stain with PAS, suggesting a polysaccharide composition. Mucoid cells contained both secretory granules and ellipsoid-vacuoles which may arise from the same Golgi apparatus.  相似文献

Restriction site mutations in the chloroplast (cpDNA) and ribosomal DNA (rDNA) were examined in 41 populations representing five of the seven recognized species of the genusRobinsonia, which is endemic to the Juan Fernandez Islands. No intraspecific variation was detected for cpDNA but one population of one of the species (R. evenia) had a restriction site mutation in rDNA not detected elsewhere. No restriction site mutations were unique to all species ofRobinsonia relative to the species ofSenecio used as outgroups. All 13 mutations (eight from cpDNA and five from rDNA) are restricted to single species, and thus provide no cladistically useful information within the genus. The distribution of mutations is concordant with the hypothesis of a rapid adaptive radiation ofRobinsonia subsequent to the dispersal of its ancestor to Masatierra.  相似文献

The karyotypes ofP. juncea (Elymus junceus) andP. huashanica (both outbreeders) were investigated by Feulgen-staining and by C-, N-, and Agbanding, based on a single plant in cach case. Both species have 2n=2x=14 and large chromosomes, possibly a generic character. The karyotype ofP. juncea has 8 metacentrics and 6 SAT-chromosomes with minute, heterochromatic satellites while that ofP. huashanica has 9 metacentrics and 5 SAT-chromosomes only, 2 of which with small, heterochromatic satellites. The C-banding patterns ofP. juncea chromosomes comprise from one to five, mostly small, bands at distal, and terminal positions, while those ofP. huashanica chromosomes are characterized by large telomeric bands in most arms. Banding patterns and chromosome morphology allow identification of the homologues of the seven chromosome pairs inP. juncea, but of two pairs inP. huashanica only. The patterns of both taxa are polymorphic, supporting that both taxa are outbreeders. The karyotypic characters suggest thatP. juncea is more closely related toP. fragilis than either is toP. huashanica. N-banding stains weakly. Silver nitrate staining demonstrates that nucleolus organizers of both species have different nucleolus forming capacities. The presence of micronucleoli suggests that both species have an extra unidentified chromosome with nucleolus forming capacity.  相似文献

Leaf protoplasts of two wild species, Solanum nigrum var. gigantea (S. ngr gig) and S. bulbocastanum Dun. (S. blb), were electrofused with leaf protoplasts of two diploid potato clones, H-8105 and ZEL-1136, respectively, in order to confer the late blight-resistance from the wild species to the cultivated potato. The S. ngr gig mesophyll (+) H-8105 mesophyll combination resulted in regenerants of mostly normal ngr phenotype. Two regenerants from this combination were proved to be true hybrids by RAPD analysis but they rooted poorely in vitro and did not survive the transfer to soil. The S. ngr gig (+) H-8105 fusion combination was also performed with H-8105 cell suspension derived protoplasts enabling an easy identification of interspecific fusants on basis of their intermediate morphology. From the S. ngr gig mesophyll (+) H-8105 cultured cell combination, many abnormal shoots were regenerated. The two lines which survived had normal ngr phenotype but the presence of tuberosum (tbr) genome in those regenerants was not confirmed by RAPD analysis. No plants with tbr phenotype were obtained from both of S. ngr gig (+) H-8105 combinations. On the contrary, when S. blb mesophyll protoplasts were electrofused with ZEL-1136 mesophyll protoplasts, all regenerated plants had tbr phenotype, indicating much lower morphogenetic potential of S. bulbocastanum in comparison with that of S. nigrum var. gigantea. However, the hybridity of those regenerants has not been confirmed by RAPD analysis with two different primers. The efficiency of the applied fusion procedure and analysis of the regenerants is discussed.  相似文献

Twenty-five samples of maize kernels collected at harvest time from geographically different corn fields in Peru, were examined for the occurrence of toxigenicFusarium species. The most frequently recovered species wereF. subglutinans (48%),F. moniliforme (46%), andF. equiseti (5%). OtherFusarium species isolated (up to 1%) includedF. graminearum, F. acuminatum, F. solani, F. oxysporum, andF. culmorum. Assays ofFusarium culture extracts usingArtemia salina larvae, showedF. subglutinans as one of the most toxigenic species, and its toxicity was mostly correlated to the capability to produce beauvericin (BEA). All eight tested isolates ofF. subglutinans grown on autoclaved corn kernels produced BEA (from 50 to 250 mg/Kg) as well as moniliformin (M) (from 70 to 270 mg/Kg). This is the first report on BEA and M production by maize isolates ofF. subglutinans from South America.  相似文献

Cross-pollination was tested between different genotypes ofLycopersicon pimpinellifolium in an experimental plot situated in the southerly range of the species. Rates of cross-pollination were found to be highly correlated with flower size and were related to a lesser extent with degree of stigma exsertion. Comparisons of fertility suggest that exsertion of 1.5 mm or more from the anther tube orifice interferes with automatic self-pollination, thereby increasing rates of cross-pollination. A single species ofExomalopsis (Anthophoridae), observed to visit all tested genotypes ofL. pimpinellifolium, was responsible for most of the pollination. Other species of solitary bees and other bee families participated in cross-pollination.  相似文献

When electrophoretic profiles of native proteins from vegetative mycelia ofPhytophthora palmivora, Phytophthora capsici and Phytophthora citrophthora causing black pod disease of cocoa in India were compared on a single Polyacrylamide gel, the isolates of same species were readily distinguished both qualitatively by visual similarity in banding patterns and quantitatively by calculating similarity coefficients. Similarity coefficients were generally much higher between isolates within a species than between isolates of different species. The dendrograms obtained after unweighted pair grouping with arithmetic averaging cluster analysis, revealed that all the isolates ofPhytophthora capsici were highly homogenous and formed a single cluster. The isolates ofPhytophthora citrophthora were resolved into two electrophoretic types which were clustered into two distinct sub groups.Phytophthora palmivora formed a separate group. Thus, the results reveal that polyacrylamide gel electrophoresis can be used successfully in distinguishing species and sub groups within a species ofPhytophthora encountered on cocoa. The ultrastructural aspects ofCyperus iria leaves showing the C4 syndrome and the typical C3 species,Carex siderosticta, in the Cyperaceae family were examined.C. iria exhibited the chlorocyperoid type, showing an unusual Kranz structure with vascular bundles completely surrounded by two bundle sheaths. The cellular components of the inner Kranz bundle sheath cells were similar to those found in the NADP-ME C4 subtype, having centrifugally arranged chloroplasts with greatly reduced grana and numerous starch grains. Their chloroplasts contained convoluted thyla-koids and a weakly-developed peripheral reticulum, although it was extensive mostly in mesophyll cell chloroplasts. The outer mestome bundle sheath layer was sclerenchymatous and generally devoid of organelles, but had unevenly thickened walls. Suberized lamellae were present on its cell walls, and they became polylamellate when traversed by plasmodesmata. Mesophyll cell chloroplasts showed well-stacked grana with small starch grains. InC. siderosticta, vascular bundles were surrounded by the inner mestome sheath and the outer parenchymatous bundle sheath with intercellular spaces. The mestome sheath cells degraded in their early development and remained in a collapsed state, although the suberized lamellae retained polylamellate features. Plastids with a crystalline structure, sometimes membrane-bounded, were found in the epidermal cells. The close interveinal distance was 35–50 μm inC. iria, whereas it was 157–218 μm inC. siderosticta. These ultrastructural characteristics were discussed in relation to their photosynthetic functions.  相似文献
