The ability of vacuoles prepared from V. faba leaves to convert 1-aminocyclopropane-1-carboxylic acid to C2H4 was destroyed when vacuoles were lysed by passage through a hypodermic needle, freezing and thawing, osmotic shock, treatment with ethanol or with a detergent. Ethylene synthesis in the vacuolar fraction was also inhibited by the uncouplers carbonyl cyanide m-chlorophenyl hydrazone and dinitrophenol and by the ionophores valinomycin, nigericin, and A23187. Ethylene formation increased with increasing pH of the incubation medium over the pH range of 5.0–7.5. Summary Freeze-fracture preparations of protoplasts isolated from cell suspension cultures and leaf mesophyll tissue have been examined by transmission electron microscopy. During the first 72 hours of cell wall regeneration, the 8–10nm intramembraneous particles were randomly distributed on both the protoplasmic and extracellular fracture faces of the plasma membranes of protoplasts frozen and fractured in the culture medium without glutaraldehyde fixation or cryoprotection. Incubation of living protoplasts in culture medium containing 20% v/v glycerol as cryoprotectant prior to freezing without fixation caused deformation of the plasma membrane in the form of protrusions accompanied by particle aggregation on the protoplasmic fracture face of the membrane. Intramembraneous particle aggregation was not observed in protoplasts fixed in glutaraldehyde prior to incubation in medium containing glycerol. The aggregation of particles into hexagonal close packed arrays and elongate chains is discussed in relation to a previous report in the literature of the possible involvement of intramembraneous particle complexes in microfibril formation by isolated higher plant protoplasts.  相似文献

Aim This study aimed to identify the ‘centres of endemism’ of the Chinese spermatophyte flora in order to indirectly detect the locations of past glacial refugia. The role of these areas as places for plant survival (‘plant museums’) and/or areas for plant evolution and speciation (‘plant cradles’) was also assessed. Location China. Methods Distribution patterns of 555 plant endemic taxa, taken as a representative sample of the Chinese endemic flora, were mapped on a 1° × 1° latitude/longitude grid. For each grid cell, species richness (total count of species) and weighted richness (down‐weighting each species by the inverse of its range) were calculated. Grid cells within the top 5% of highest values of weighted richness were considered centres of endemism. Based on available information, all plant taxa included in this study were classified into palaeoendemics and neoendemics, and their distributional patterns were represented separately. Results Twenty areas of endemism were identified in central and southern China, roughly corresponding to mountain ranges, including the Hengduan and Daxue Mountains, the Yungui Plateau, central China Mountains, the Nanling Mountains, eastern China Mountains, and Hainan and Taiwan. Although almost all centres of endemism contained both palaeoendemic and neoendemic taxa, considerable differences in their respective numbers were recorded, with the majority of neoendemics on the eastern fringe of the Tibetan Plateau (Hengduan Mountains sensu lato) but more palaeoendemics towards the east. Main conclusions Owing to their varied topography, the mountainous regions of central and southern China have provided long‐term stable habitats, which allowed palaeoendemics to persist and facilitated the process of speciation. Contrasting patterns between the palaeoendemics and neoendemics within refugia might be attributable to the geological and tectonic history of specific areas. The eastern fringe of the Tibetan Plateau clearly constitutes the ‘evolutionary front’ of China, probably as a result of the uninterrupted uplift of the plateau since the late Neogene. In contrast, the tectonic stability of central and southern China during the Tertiary may have facilitated the persistence of relict plant lineages.  相似文献

Spatial overlap between predators and prey is key to predicting their interaction strength and population dynamics. We constructed a spatially-explicit simulation model to explore how predator and prey behavioral traits and patterns of resource distribution influence spatial overlap between predators, prey, and prey resources. Predator and prey spatial association primarily followed the ideal free distribution. Departures from this model were intriguing, especially from the interactions of predator and prey behavior. When prey weakly avoided conspecifics, they associated more highly with resources when predators were present. Predators increased the rate of prey movement between patches, which increased their ability to sample their environment and aggregate in patches with high resources. When prey strongly avoided each other, predators decreased prey association with resources. That is, an increased rate of prey movement increased the probability that prey would interact and avoid each other without regard to the distribution of resources. More generally, a more highly clumped distribution of resources acted as a spatial anchor that generally increased prey, predator, and resource association. Prey tended to congregate with resources and predators generally congregated with prey.  相似文献

Background

We recently reported that sphingomyelin (SM) analogs substituted on the alkyl chain by various fluorophores (e.g. BODIPY) readily inserted at trace levels into the plasma membrane of living erythrocytes or CHO cells and spontaneously concentrated into micrometric domains. Despite sharing the same fluorescent ceramide backbone, BODIPY-SM domains segregated from similar domains labelled by BODIPY-D-e-lactosylceramide (D-e-LacCer) and depended on endogenous SM.

Methodology/Principal Findings

We show here that BODIPY-SM further differed from BODIPY-D-e-LacCer or -glucosylceramide (GlcCer) domains in temperature dependence, propensity to excimer formation, association with a glycosylphosphatidylinositol (GPI)-anchored fluorescent protein reporter, and lateral diffusion by FRAP, thus demonstrating different lipid phases and boundaries. Whereas BODIPY-D-e-LacCer behaved like BODIPY-GlcCer, its artificial stereoisomer, BODIPY-L-t-LacCer, behaved like BODIPY- and NBD-phosphatidylcholine (PC). Surprisingly, these two PC analogs also formed micrometric patches yet preferably at low temperature, did not show excimer, never associated with the GPI reporter and showed major restriction to lateral diffusion when photobleached in large fields. This functional comparison supported a three-phase micrometric compartmentation, of decreasing order: BODIPY-GSLs > -SM > -PC (or artificial L-t-LacCer). Co-existence of three segregated compartments was further supported by double labelling experiments and was confirmed by additive occupancy, up to ∼70% cell surface coverage. Specific alterations of BODIPY-analogs domains by manipulation of corresponding endogenous sphingolipids suggested that distinct fluorescent lipid partition might reflect differential intrinsic propensity of endogenous membrane lipids to form large assemblies.

Conclusions/Significance

We conclude that fluorescent membrane lipids spontaneously concentrate into distinct micrometric assemblies. We hypothesize that these might reflect preexisting compartmentation of endogenous PM lipids into non-overlapping domains of differential order: GSLs > SM > PC, resulting into differential self-adhesion of the two former, with exclusion of the latter.  相似文献

Empirically, α-helical membrane protein folding stability in surfactant micelles can be tuned by varying the mole fraction MFSDS of anionic (sodium dodecyl sulfate (SDS)) relative to nonionic (e.g., dodecyl maltoside (DDM)) surfactant, but we lack a satisfying physical explanation of this phenomenon. Cysteine labeling (CL) has thus far only been used to study the topology of membrane proteins, not their stability or folding behavior. Here, we use CL to investigate membrane protein folding in mixed DDM-SDS micelles. Labeling kinetics of the intramembrane protease GlpG are consistent with simple two-state unfolding-and-exchange rates for seven single-Cys GlpG variants over most of the explored MFSDS range, along with exchange from the native state at low MFSDS (which inconveniently precludes measurement of unfolding kinetics under native conditions). However, for two mutants, labeling rates decline with MFSDS at 0–0.2 MFSDS (i.e., native conditions). Thus, an increase in MFSDS seems to be a protective factor for these two positions, but not for the five others. We propose different scenarios to explain this and find the most plausible ones to involve preferential binding of SDS monomers to the site of CL (based on computational simulations) along with changes in size and shape of the mixed micelle with changing MFSDS (based on SAXS studies). These nonlinear impacts on protein stability highlights a multifaceted role for SDS in membrane protein denaturation, involving both direct interactions of monomeric SDS and changes in micelle size and shape along with the general effects on protein stability of changes in micelle composition.  相似文献

Selectivity of nuclear probes is controlled by competitive accumulation of the probe by cellular organelles as well as the high affinity for nucleic acids. Physicochemical features of probes which favor nucleic acid binding include cationic character and a planar aromatic system above a minimum size. Features of probes which permit entry into cells are low protein and lipid binding. Features which reduce accumulation in non-nuclear sites include high base strength and hydrophilicity of the cation. The overall quantitative structure–activity (QSAR) model specifying nuclear accumulation may be expressed as follows: CBN<40; 8>log P neutral species>0; AI<8; Z>0; -5<log P cation<0; pK a >10; LCF>17; LCF/CBN>0.70 (where CBN is the conjugated bond number, log P x the logarithm of the water–octanol partition coefficient of species x, AI the amphilicity index, Z the electric charge, pK a the negative logarithm of the equilibrium constant for the free base–protonated base reaction, and LCF the largest conjugated fragment). Preliminary applications of the QSAR model—to the selection of anticancer drugs, minimization of dye and drug toxicity and the designed synthesis of fluorescent probes—are outlined.  相似文献

Invasive species are a global threat to biodiversity, economy and human wellbeing. To mitigate these threats, identifying and halting the introduction of potentially invasive species is crucial. Although progress has been made in elucidating mechanisms underlying invasion success, the role of species behavioral strategies has only received scant attention. Here, we use the invasion of monk parakeets in Santa Catarina state, southern Brazil to study whether behavioral strategies such as nest thermoregulation and the ability to self-medicate against pathogens contribute to the establishment success of invading species. We relate data on monk parakeet reproductive success to ambient temperatures in- and outside nesting chambers and test the bactericidal potential of plants transported to the nest by breeding monk parakeets. Compared to breeding data from other invaded ranges and parts of the species’ native range, our results suggest both thermoregulation and the use of bactericidal plants could potentially influence monk parakeet reproductive success. Thermoregulation maintains stable temperatures of incubator chambers compared to large fluctuations (especially hotter extremes) outside the nest. At least one of the plants brought to the nest effectively inhibited growth of pathogenic bacteria. The union of these two factors could increase reproductive rates and may consequently aid the expansion of the species in new non-native environments.  相似文献

Herbivorous insects are influenced by both 'bottom-up' forces mediated through host plants and 'top-down' forces from natural enemies. Few studies have tried to evaluate the relative importance of the two forces in determining the abundance of insects. The leaf beetle Phratora vulgatissima Linnaeus sometimes occurs at high densities and severely damages the willow Salix cinerea in forest habitats. For willows growing in open agricultural landscapes (farmland S. cinerea), the leaf beetle generally occurs at low densities and plants receive little damage. The purpose of the present study was to evaluate the relative importance of host plant quality and natural enemies behind the observed difference in P. vulgatissima abundance. Female egg-laying and larval performance (growth and survival) were studied on caged willow branches in the field to investigate if plant quality differs between S. cinerea trees growing in forest and farmland habitats. The survival of eggs exposed to natural enemies was examined to see if predation could explain the low abundance of leaf beetles on farmland willows. The results indicated no difference in plant quality; female egg laying and larval performance did not differ between the forest and the farmland. However, heteropteran predators (true bugs) were more abundant, and the survival of eggs was lower, on plants in the farmland habitat than in the forest habitat. The data suggest that the low abundance of P. vulgatissima on farmland willows could not be explained by a poor quality of plants, but more likely by high predation from heteropterans.  相似文献

Class III peroxidases (Prxs) are plant enzymes capable of using H(2)O(2) to oxidize a range of plant secondary metabolites, notably phenolic compounds. These enzymes are localized in the cell wall or in the vacuole, which is a target for secondary metabolite accumulation, but very little is known about the function of vacuolar Prxs. Here, the physiological role of the main leaf vacuolar Prx of the medicinal plant Catharanthus roseus, CrPrx1, was further investigated namely by studying its capacity to oxidize co-localized phenolic substrates at the expense of H(2)O(2). LC-PAD-MS analysis of the phenols from isolated leaf vacuoles detected the presence of three caffeoylquinic acids and four flavonoids in this organelle. These phenols or similar compounds were shown to be good CrPrx1 substrates, and the CrPrx1-mediated oxidation of 5-O-caffeoylquinic acid was shown to form a co-operative regenerating cycle with ascorbic acid. Interestingly, more than 90% of total leaf Prx activity was localized in the vacuoles, associated to discrete spots of the tonoplast. Prx activity inside the vacuoles was estimated to be 1809 nkat ml(-1), which, together with the determined concentrations for the putative vacuolar phenolic substrates, indicate a very high H(2)O(2) scavenging capacity, up to 9 mM s(-1). Accordingly, high light conditions, known to increase H(2)O(2) production, induced both phenols and Prx levels. Therefore, it is proposed that the vacuolar couple Prx/secondary metabolites represent an important sink/buffer of H(2)O(2) in green plant cells.  相似文献

In their journey through the oviduct some subpopulations of sperm are preserved in a reservoir, while others are negatively selected. Sperm binding glycoprotein (SBG) is a pig oviductal epithelial cell glycoprotein that produces, under capacitating conditions, acrosome alteration, p97 tyrosine-phosphorylation and reduction of the motility of sperm. In this paper, we show that SBG is accessible at the extracellular surface of the oviductal epithelial cells, supporting a sperm interaction biological role in situ. We analyze the possible dependence of the tyrosine-phosphorylation of p97 on the PKA mechanism, finding that apparently it is not PKA dependent. Also, after SBG treatment the phosphorylated proteins locate mainly at the detached periacrosomal region and at the tail of sperm; the latter may be related to SBG's motility reduction effect. The study of the time course effect of SBG on sperm as detected by chlortetracycline (CTC) staining and of its binding to sperm by immunodetection in conjunction with CTC, shows results in agreement with the hypothesis that this glycoprotein is involved in the alteration of acrosomes in a specific sperm subpopulation. The results suggest that SBG may be part of a mechanism for negative selection of sperm.  相似文献

The annual rhythm of human reproduction was analyzed on the basis of more than 3000 years of monthly birth rates covering 166 regions of the globe. The following variables were used to characterize the annual rhythm of human conception (birth minus 9 months): amplitude, phase of maximum and minimum, phase and length of the time span when rates are above the annual mean (alpha), and the steepest upward slope (delta max) of the curve fitted to the rates. The waveform of the annual rhythm is characteristic for geographical regions (e.g., unimodal or bimodal) and persists as such for many years. In most countries, the onset of alpha coincides with delta max and lies close to the spring equinox. This phase of the rhythm is the most stable over time. In many populations, the rhythm has changed in recent years, specifically in amplitude and phase. The phase of the rhythm depends on latitude, with a 6-month difference between the Northern and Southern Hemispheres. The latitudinal distribution of amplitudes is less systematic. In spite of the many social influences on timing of conceptions, we conclude that the seasonal component in human reproduction is based on biological factors.  相似文献

Voltage-gated sodium channel alpha-subunits have been shown to be key mediators of the pathophysiology of pain. The present review considers the role of sodium channel auxiliary beta-subunits in channel modulation, channel protein expression levels, and interactions with extracellular matrix and cytoskeletal signaling molecules. Although beta-subunits have not yet been directly implicated in pain mechanisms, their intimate association with and ability to regulate alpha-subunits predicts that they may be a viable target for therapeutic intervention in the future. It is proposed that multifunctional sodium channel beta-subunits provide a critical link between extracellular and intracellular signaling molecules and thus have the ability to fine tune channel activity and electrical excitability.  相似文献

The plant plasma membrane H+-ATPase is kept at a low activity level by its C-terminal domain, the inhibitory function of which is thought to be mediated by two regions (region I and II) interacting with cytoplasmic domains essential for the catalytic cycle. The activity of the enzyme is well known to be regulated by 14-3-3 proteins, the association of which requires phosphorylation of the penultimate H+-ATPase residue, but can be abolished by phosphorylation of residues close-by. The current knowledge about H+-ATPase regulation is briefly summed up here, combined with data that query some of the above statements. Expression of various C-terminal deletion constructs of PMA2, a H+-ATPase isoform from Nicotiana plumbaginifolia, in yeast indicates that three regions, which do not correspond to regions I or II, contribute to autoinhibition. Their individual and combined action can be abolished by (mimicking) phosphorylation of three threonine residues located within or close to these regions. With respect to the wild-type PMA2, mimicking phosphorylation of two of these residues increases enzyme activity. However, constitutive activation of wild-type PMA2 requires 14-3-3 association. Altogether, the data suggest that regulation of the plant H+-ATPase occurs in progressive steps, mediated by several protein kinases and phosphatases, thus allowing gradual as well as fine-tuned adjustment of its activity. Moreover, mating-based split ubiquitin assays indicate a complex interplay between the C-terminal domain and the rest of the enzyme. Notably, their tight contact does not seem to be the cause of the inactive state of the enzyme.  相似文献

To develop technetium and rhenium-labeled imaging agents for estrogen receptor (ER) positive breast tumors, we have synthesized tridentate metal tricarbonyl chelates substituted at the 16α-position of estradiol. Their structures were characterized by IR, 1H NMR, 13C NMR, HRMS or elemental analysis. The rhenium complex 7b showed the highest ER binding affinity (RBA = 25.7) among these compounds, so ligand 6b was selected to be labeled by the precursor [99mTc(H2O)3(CO)3]+ to yield technetium(I)-99m complex 7b′ with good radiochemical yields. The lipophilicity of corresponding technetium(I)-99m complex 7b′ was appropriately reduced, which might be favorable to target tissue selectivity in vivo. The stability of complex 7b′ is excellent in 1 mM histidine, 1 mM cysteine, PBS and bovine serum within 6 h in vitro.  相似文献

Environmental stress may favour facilitative interactions among plants but whether these interactions are positive for the benefactor and how this depends on stress factors, remains to be determined. We studied the effect of beneficiary cover and biomass on reproduction of the benefactor cushion plant Laretia acaulis (Apiaceae) in the central Chilean Andes during three years. Study sites were situated along an elevational gradient at 2600, 2800, 3000 and 3150 m a.s.l. This range comprises a cold‐ and a drought‐stress gradient, with moisture increasing and temperature decreasing with elevation. We studied the effect of natural gradients in beneficiary cover and of experimental cover removal on cushion flower and fruit production. Beneficiary cover had a negative effect on flower production but not on infructescence and fruit densities or fruit weights. A positive effect of beneficiaries on the fraction of flowers converted into fruits was detected for hermaphrodite cushions. The effect of beneficiary cover on flowering was independent of elevation or cushion gender, although these latter factors explained most of the variation. Removing the aboveground parts of the beneficiaries positively affected flowering at 2800 m a.s.l. but not at the other elevations. Our results suggest negative effects of facilitation on L. acaulis flowering, but these are neutralized in fruit production. Surprisingly, this conclusion holds along the entire elevational or stress gradient. This suggests that this system of facilitation is evolutionarily stable and not very sensitive to environmental change. It remains to be tested, however, whether facilitation affects fitness via growth and long‐term survival in these slow‐growing alpine cushions.  相似文献

Concentrations of nutrients and plant secondary metabolites (PSM) vary temporally and spatially, creating a multidimensional feeding environment. Interactions between nutrients and PSM are poorly understood because research has relied largely on studying the isolated effects of nutrients or PSM on foraging behavior. Nevertheless, their interactions can influence food selection and the dynamics of plant communities. Our objective was to explore how interactions between nutrients and PSM influence food selection. For 7 d, three groups of lambs received intraruminal infusions of three different doses of a PSM (0=Control; low and high) and 2 h later they were offered two foods that contained either low (high in energy) or high (high in protein) protein/energy ratios. The foods were offered 7 d before (baseline) and 7 d after PSM infusions. We conducted five trials each with a different PSM- terpenoids, cyanogenic glycosides, sodium nitrate, quebracho tannin, and lithium chloride. Lambs consistently preferred the food high in energy to the food high in protein, but toxins modified the degree to which this preference was manifest. Terpenoids, nitrate, and lithium chloride depressed intake of the food high in energy. Cyanogenic glycosides had the opposite effect, and at higher doses they depressed intake of the food high in protein. Tannins enhanced intake of the food high in energy at lower doses and they depressed its ingestion at higher doses. Thus, PSM selectively depressed or enhanced intake depending on the macronutrient composition of the foods. These results imply that the probability of a plant being eaten will depend not only on its chemical defenses but also on the quantity and quality of nutrients in the plant and its neighbors, and that the ability of herbivores to learn associations between nutrients and PSM may have a substantial impact on the way herbivores regulate ecosystem processes.  相似文献

Familial Mediterranean fever (FMF) is an autosomal recessive disorder of unknown pathogenesis, characterized by recurrent, selflimited attacks of fever with synovitis, peritonitis, or pleurisy. Using DNAs from affected Israeli families, we have recently mapped the gene causing FMF (designated MEF) to the short arm of chromosome 16, with two-point lod scores in excess of 20. In this report we consider the possibility of a second FMF susceptibility locus. Before discovering linkage to markers on chromosome 16, we had found suggestive evidence for linkage to chromosome 17q, with the following maximal two-point lod scores: D17S74 (pCMM86), = 2.47, ( = 0.20); D17S40 (pLEW101), = 2.15( = 0.15); D17S35 (CRI-pP3-1), = 1.78 ( = 0.15); D17S46 (pLEW108), = 1.69 ( = 0.18), D17S254, = 2.30 ( = 0.20). Moreover, multipoint linkage analysis using D17S74 and D17S40 as fixed loci gave = 3.27 approximately 10 centimorgans (cM) telomeric to D17S40. Data with the chromosome 17 markers alone in our families suggested locus heterogeneity. Nevertheless, our families were not separable into complementary subsets showing linkage either to chromosome 16 or to chromosome 17. We also examined the possibility that the positive lod scores for chromosome 17 might reflect a secondary, modifying locus. By several measures of disease severity, families with positive lod scores for chromosome 17 loci had no worse disease than those with negative lod scores for these loci. We conclude that chromosome 17 does not encode a major FMF susceptibility gene for some of the families, nor does it encode a disease-modifying gene. Rather, it would appear that linkage to chromosome 17 is a false positive (type I) error. These results reemphasize the fact that a lod score of 3.0 corresponds to a posterior probability of linkage of 95%, with an attendant 1 in 20 chance of observing a false positive.  相似文献
