Nodal explants of in vivo plants and in vitro seedlings of Wattakaka volubilis were cultured on Murashige and Skoog medium fortified with various concentrations of cytokinins — BA (0.5–5 mg l?1), KN (0.5–10 mg l?1),TDZ (0.05–1 mg l?1) either singly or in combination with NAA (0.1 mg l?1). KN proved best for inducing healthy shoots in both in vitro and in vivo derived explants. Maximum number of shoots (14.1±0.84) with 80% regeneration frequency was obtained from nodal explants of seedlings cultured on 5 mg 1?1 KN + 0.1 mg l?1 NAA. In vivo nodal explants produced a maximum of 4.2 shoots on MS medium fortified with 2 mg l?1 BA+0.1 mg l?1 NAA. The differentiated shoots from both could be rooted with 85% frequency on 1/2 strength MS medium (1% sucrose) with 0.6% agar + 1 mg l?1 IBA + 0.2 mg l?1 KN. Rooted shoots were transplanted to vermiculite-soil (3:1) mixture in polyethylene covered pots with 45% transplantation success. Peroxidase isozymes (native PAGE) analysis helped to verify the variation in regenerated plants.  相似文献

Callus cultures were established from seedling explants of Pergularia daemia (Forsk) Chiov on Murashige and Skoog (MS) medium supplemented with different concentrations of auxins. Optimal callus developed from leaf explants on MS medium supplemented with 2,4-D (2 mg l?1) + 2iP (0.1 mg l?1), was used for morphogenesis. Adventitious shoots were regenerated (70%) from the calli on MS medium supplemented with NAA (0.1 mg l?1)+ BAP (2 mg l?1). Individual shoots were rooted on half strength MS medium supplemented with 0.1 mg l?1 IBA. Plantlets with well developed roots were successfully transferred to soil and 50% of the transferred plants survived.  相似文献

We describe here the development of a micropropagation protocol for mass multiplication of Zantedeschia aethiopica by using root tubers as explant. The surface sterilized root tubers produced five to six shoot-buds on semi-solid Murashige and Skoog’s (MS) medium with 10.0 mg l?1 of 6-benzylaminopurine (BAP) and additives (50.0 mg l?1 of ascorbic acid; 25.0 mg l?1 each of adenine sulphate, L-arginine and citric acid). The cultures were multiplied by sub-culture of individual shoot bud produced in vitro and clumps of shoot buds generated in vitro in cultures on MS medium containing 3.0 mg l?1 of BAP and additives. Further multiplication of propagules was achieved through tuber formation along with amplifying shoots on MS medium with 5.0 mg l?1 of BAP. The micropropagated shoots were rooted both in vitro as well as ex vitro. Cent percent of the cloned shoots rooted in vitro within 15–18 days on hormone-free 1/2 strength MS salts with 200.0 mg l?1 of activated charcoal. Alternatively 95–100% shoots rooted ex vitro under greenhouse conditions on soilrite after pulse-treatment with 500.0 mg l?1 of Indole-3-butyric acid (IBA) or β-naphthoxyacetic acid (NOA) for 300 sec. The cloned plants were hardened in the greenhouse. The hardened plants were transplanted to soil for further acclimatization.  相似文献

Paedaria foetida L, which has great medicinal value is facing danger of extinction. For its conservation, in vitro multiplication may prove one of the best techniques. Micropropagation of P. foetida has been achieved through the culture of nodal explants. The explants produced shoots on MS medium with 0.8% agar. This medium, however, caused high degree of browning of the explants and death of sprouted shoots. Agar free liquid MS medium with polyvinylpyrrolidone (PVP) proved better. Maximum shoot proliferation, free from callus and vitrification but with poor rooting could be obtained in liquid MS medium with PVP (0.8%), NAA (0.5 mg l?1) and BA (2.0 mg l?1). The best rooting occurred on semisolid MS medium containing 0.8% agar and 0.5 mg l?1 IBA.  相似文献

Thidiazuron (TDZ) stimulated shoot proliferation from different seedling explants (i.e., shoot, basal node, node and apical segment) of bamboo (Dendrocalamus strictus) when incorporated in half-strength Murashige and Skoog (MS) medium having 2% (w/v) sucrose. All the concentrations of TDZ (0.01 to 1.0 mg l?1) tried were effective in shoot proliferation. Maximum shoots (14.8 ± 1.0) were obtained from the shoot explants cultured in 0.5 mg l?1 TDZ supplemented halfstrength MS liquid medium for 21 days and subsequently transferred to the same medium devoid of TDZ. The longer culture period (i.e. 28 and 35 days) in TDZ medium caused reduction in shoot proliferation. The shoots regenerated with lower concentrations of TDZ treatment (i.e. 0.01 to 0.1 mg l?1) rooted in half-strength MS liquid medium. The shoots formed with 0.5 mg l?1 TDZ treatment did not root in basal medium and required auxin supplementation in the medium for rooting and about 55% shoots produced roots in 1.0 mg l?1 IBA supplemented medium. The shoots formed with 1.0 mg l?1 TDZ did not root even after auxin treatment. The well rooted shoots transplanted to plastic pots filled with sand and garden soil (1:1) mixture showed 98% establishment.  相似文献

Mungbean (Vigna radiata (L) Wilczek cv ML — 267) is a recalcitrant grain legume species. Direct multiple shoots were developed from the cotyledonary node explants of 2-day-old in vitro grown seedlings of mungbean. Maximum number of shoots (an average 12.1 shoots per explant) was obtained on a medium containing MS salts, B5 vitamins and 5.0 mg l?1 BAP. A medium with lower BAP concentration appeared suitable for rapid shoot elongation. The elongated shoots were rooted on 0.2 mg l?1 NAA. The rooted plants were acclimatized under field conditions. The survival of the plants in the greenhouse was 90 %. Plants flowered and set seed normally.  相似文献

Regeneration of adventitious shoots from leaf and petiole pieces of Gerbera jamesonii has been obtained on Murashige and Skoog (MS) medium supplemented with different concentrations of auxins and cytokinins. About 75’77 per cent of the calli from both types of the explants produced 12’15 shoots per callus with 3 mg l?1 SAP. Auxins and kinetin, separately failed to produce shoots. The shoots regenerated on the callus induction medium (elM). The regenerated shoots multiplied with 1 mg l?1 SAP, were rooted on MS medium containing 1mg l-1 BAP + 0.1 mg l-1 IAA. The plants obtained were transferred to pots and acclimatized with 60’70 per cent success.  相似文献

A successful report on the in vitro propagation of Homalomena aromatica via rhizome axillary bud multiplication is presented. Rhizome bud explants were cultured on Murashige and Skoog medium supplemented with various concentrations of cytokinins to induce multiple shoot formation for micropropagation. The highest number of shoots was achieved in MS medium supplemented with 2.0 mg?l?1 6-benzylaminopurine. The regenerated shoots rooted most efficiently on half-strength MS medium supplemented with 0.5 mg?l?1 α-naphthalene acetic acid. The regenerated plantlets showed no morphological differences from the parent plant. This protocol takes approximately 6 months to reach the acclimatization stage from the initiation stage and facilitates commercial and rapid propagation of H. aromatica.  相似文献

The axillary bud-break and multiple bud induction were obtained from the nodal explants of field-grown culms of Bambusa tulda in liquid Murashige and Skoog’s (MS) basal medium supplemented with 2.0 mg l?1 6-benzylaminopurine (BAP), 1.0 mg l?1 kinetin (Kn) and 8% coconut water. Multiple shoots regenerated and proliferated in the liquid MS medium fortified with 3.0 mg l?1 indolebutyric acid (IBA). While, in B. balcooa, MS medium supplemented with 2.5 mg l?1 BAP and 1.0 mg l?1 Kn induced axillary bud-break, bud multiplication and subsequently shoot elongation was obtained after three passages in the same medium. A clump with at least three shoots of both these bamboo species was used as propagule for successful root induction in half-strength MS liquid basal medium supplemented with 0.2 mg l?1 IBA. Sympodial type of microrhizomes developed in B. tulda and the regenerants acclimatized in the soil easily. Explants collected in the month of October produced best in vitro regeneration response in these two bamboo species. Endogenous phenol content proved detrimental for efficient shoot regeneration. The clonal fidelity of the regenerants was established by RAPD analysis advocating clonal propagation through axillary meristem culture of B. balcooa and B. tulda is reliable for commercial exploitation.  相似文献

Axillary buds obtained from 5-month-old in vitro growing plants of Dendrobium lituiflorum Lindl were cultured in Murashige and Skoog (MS) medium supplemented with different concentrations of 2,4-D. Fastest initiation (13.3 days) of protocormlike bodies (PLBs) was observed in cultures containing MS medium supplemented with 0.5 mg l?1 2,4-D. Maximum explant response of 83% was also observed in the same medium. PLBs obtained in MS medium containing 0.5 mg l?1 2,4-D showed maximum regeneration potential of seedlings (19 explant?1) when subcultured in MS medium. Well developed shoots and roots of the seedlings were obtained in the medium containing 0.5 mg l?1 each of NAA and BAP, in combination. Encapsulated PLBs of D. lituiflorum could be stored at 8°C for 90 days with 80% regeneration. However, it was observed that regeneration potential of encapsulated PLBs reduced with further storage. Seventy seven per cent hardened plants survived and bloomed after 2.5 years of hardening.  相似文献

In vitro propagation methods using seeds and nodal segments of a 21-year old Couroupita guianensis - a medicinally important but threatened tree have been developed. Hundred percent of the seeds germinated on half strength Murashige and Skoog (MS) medium with 2.0 mg l?1 indole-3 butyric acid (IBA). Nodal segments were found most suitable for the establishment of cultures. About 90 % explants responded and 4.1 ± 0.23 shoots per node were induced after five weeks of inoculation on MS medium +4.0 mg l?1 6-benzylaminopurine (BAP). Further shoot multiplication was achieved by repeated transfer of mother explants and subculturing of in vitro produced shoots on fresh medium. Maximum number (8.2 ± 0.17) of shoots were regenerated on MS medium with 1.0 mg l?1 each of BAP and Kinetin (Kin) + 0.5 mg l?1 α-naphthalene acetic acid (NAA) with additives (50 mg l?1 of ascorbic acid and 25 mg l?1 each of adenine sulphate, L-arginine and citric acid). The multiplied shoots rooted (4.3 ± 0.26 roots/shoot) on half strength MS medium with 2.5 mg l?1 IBA. All the shoots were rooted ex vitro when pulse treated with 400 mg l?1 of IBA for five min with an average of 7.3 ± 0.23 roots per shoot. Nearly 86 % of these plantlets were acclimatized within 7–8 weeks and successfully transferred in the field. Biologically significant developmental changes were observed during acclimation particularly in leaf micromorphology in terms of changes in stomata, veins and vein-islets, and trichomes. This study helps in understanding the response by the plants towards outer environmental conditions during acclimatization. This is the first report on micropropagation of C. guianensis, which could be used for the large-scale multiplication, restoration and conservation of germplasm of this threatened and medicinally important tree.  相似文献

A protocol for large-scale propagation of Piper betle cvs Desawari and Desi Bangla was developed through axillary shoot proliferation. Due to systemic infection as well as high phenol content the crop was very difficult to establish in aseptic condition. But a mixture of 5 mg l?1 each of chloramphenicol and oxytetracyclin and 100 mg l?1 each of citric acid and ascorbic acid used in MS medium for 2 days helped in establishment. After 48 h, the explants were transferred to the antibiotic free medium having PVP and ascorbic acid (100 mg l?1 each), citric acid (50 mg l?1), and glutathione (20 mg l?1). Regular subculturing of the explants into liquid medium, use of antioxidants and incubation of the cultures in the dark for initial 7–10 days played a crucial role for keeping them fresh and green. Maximum numbers of axillary shoots were obtained with 2 mg l?1 BA and 0.2 mg l?1 NAA as growth supplements. The plants were rooted in 0.25 mg l?1 IBA and hardened in the soil. Phenolic compound analysis showed almost the same results in tissue-raised and in vivo grown plants in Desawari.  相似文献

Nodal explants, excised from young shoots from mature trees of Syzygium cuminii, were cultured on MS medium supplemented with different concentrations of BA or kinetin. Among these, BA (0.5 or 1.0 mg l–1) induced greening and opening of the incipient shoot buds, which however did not elongate. Elongation of the shoot buds was facilitated on MS medium with 1.0 mg l–1 BA supplemented with casein hydrolysate (1.5 g l–1) or glutamine (200 mg l–1). Nodal explants (microcuttings), taken from shoots developed in vitro, also developed multiple shoots when cultured on MS with1 mg l–1 BA. These explants did not require an additional supply of reduced nitrogen, for further normal development. Shoots developed from explants from mature trees and microcuttings were rooted by sub-culturing them on Knop's medium supplemented with 2% sucrose and 1 mg l–1 IAA. The plants that developed in vitro were planted in soil and were transferred to the field after an acclimatization period of 7–8 months. These plants have been thriving well for more than three years and have no apparent phenotypic aberrations.  相似文献

A protocol for regular in vitro regeneration of Carthamus tinctorius var HUS-305 is described. The morphogenic response of seed explants and explants from seedlings of different ages were studied on Murashige and Skoog’s medium (MS) supplemented with different growth regulators. The protocol finally standardized involves culture of cotyledonary explants from 2 cm long, 2- to 6-day-old seedlings on MS supplemented with 1.0 mg l?1 BAP and 0.1 mg l?1 kinetin. Various other adjuvants viz., adenine sulphate, glutamine and casein hydrolysate were also tested. None of these promoted the caulogenic response significantly. The microshoots could be rooted on medium supplemented with different auxins. The highest rhizogenic response was on 1/2 MS supplemented with 0.2 mg l?1 NAA.  相似文献

In this communication we report shoot organogenesis from hypocotyl explants in cumin (Cuminum cyminum) genotype RZ-19 by the use of thidiazuron (TDZ). Various levels of TDZ were incorporated in MS basal medium to induce regeneration. Regeneration was achieved with a frequency up to 30% on 0.5 and 0.1 mg l?1 concentration of TDZ. Shoots once produced could be multiplied on 0.5 mg l?1 kinetin (KN) at the rate of approximately 8 shoots per regenerated shoot. These multiplied shoots could go through 3–4 multiplication cycles after which they root on 1.0 mg l?1 IAA.  相似文献

Embryogenic avocado cultures derived from ‘Hass’ protoplasts were genetically transformed with the plant defensin gene (pdf1.2) driven by the CaMV 35S promoter in pGPTV with uidA as a reporter gene and bar, the gene for resistance to phosphinothricin, the active ingredient of the herbicide Finale® (Basta) (Bayer Environmental Science, Research Triangle Park, Durham, NC ). Transformation was mediated by Agrobacterium tumefaciens strain EHA105. Transformed cultures were selected in the presence of 3.0 mg l?1 phosphinothricin in liquid maintenance medium for 3–4 mo. Liquid maintenance medium consisted of modified MS medium containing (per liter) 12 mg NH4NO3 and 30.3 mg KNO3 and supplemented with 0.1 mg l?1 thiamine HCl, 100 mg l?1 myo-inositol, 30 g l?1 sucrose, 3.0 mg l?1 phosphinothricin, and 0.41 μM picloram. Somatic embryo development from transformed cultures was initiated on MS medium supplemented with 45 g l?1 sucrose, 4 mg l?1 thiamine HCl, 100 mg l?1 myo-inositol, 10% (v/v) filter-sterilized coconut water, 3.0 mg l?1 phosphinothricin, and 6.0 g l?1 gellan gum. Limited plant recovery occurred from somatic embryos on semi-solid MS medium supplemented with 3.0 mg l?1 phosphinothricin, 4.44 μM 6-benzylaminopurine (BA), and 2.89 μM GA3; transformed shoots were micrografted on in vitro-grown seedling rootstocks. Approximately 1 yr after acclimatization in the greenhouse, transformed shoots were air-layered to recover transformed roots. Genetic transformation of embryogenic cultures, somatic embryos, and regenerated plants was confirmed by polymerase chain reaction (PCR), Southern blot hybridization, the XGLUC reaction for uidA, and application of the herbicide Finale® to regenerated plants.  相似文献

Cichorium pumilum, a member of Asteraceae, is widely used as a traditional medicinal herb. An efficient protocol for callus formation and whole plant propagation has been developed. Callus cultures were induced from leaf explants on Murashige and Skoog (MS) medium supplemented with 1.5?mg?l?1 6-Benzyladenine (BA) and 0.5?mg?l?1 Naphthalene acetic acid (NAA). Maximum numbers of shoots were obtained from calli transferred to shoot regeneration medium containing MS basal medium with 1.5?mg?l?1 BA or Kinetin (Kin). The shoots were effectively rooted on MS medium supplemented with different concentrations of Indole-3-butyric acid. In the present study, the antibacterial activity of C. pumilum extracts was assayed in vitro by agar disc diffusion and agar well diffusion methods against 10 different bacterial species. The results showed effect on the growth of 50 and 70% of the tested bacterial species using methanol and ethanol extracts respectively. Klebsiella pneumoniae was susceptible to the ethanolic and methanolic extracts of wild plants and in vitro tissues, whereas Enterococcus faecalis was resistant to all the extracts. This study verified that the methanol extracts have strong antioxidant activities with high levels of phenolic compounds. The antioxidant activity and total phenol content of callus cultures and in vitro plantlets were lower than those of the wild plants. The results obtained confirm the therapeutic potency of Cichorium used in the traditional medicine, in addition, the efficient in vitro production system developed in this study provide sterile and consistent tissues for the investigation of phytochemical and pharmacological effects and germplasm conservation of C. pumilum.  相似文献

An efficient micropropagation protocol has been developed for Withania coagulans, a highly endangered medicinal herb and an important natural source of withanolides. Prolific multiplication of axillary buds occurred from the nodal segments taken from adult plant, and cultured on MS medium enriched with BA (0.5 mg l?1), Kn (0.5 mg l?1) and PG (0.5 mg l?1). Nodal segments and shoot tips of elongated microshoots also behaved the same way in cultures and formed multiple shoots through axillary bud multiplication. Addition of PG (0.5 mg l?1) in the regeneration medium significantly improved induction and elongation of shoot buds. Elongated shoots were placed on filter paper bridges soaked in MS medium with CC (10 mg l?1) and PG (0.5 mg l?1) for the initial 7 days’ pulse treatment and thereafter, they were transferred to rooting medium containing IBA (0.25 mg l?1) + PAA (0.5 mg l?1) + CC (2 mg l?1). This protocol has the capacity of producing 1000 plants from one nodal segment after 4 subcultures of 2 weeks each.  相似文献

Callus cultures were established in three commercial sugarcane varieties viz., CoJ 64, CoJ 83 and CoJ 86 from spindle explants on MS + 2,4-D (4 mg l?1) + BAP (0.5 mg l?1) medium. Shoots were regenerated from two-month-old calli on MS + BAP (0.5 mg l?1) medium. Callus and callus derived shoots were treated with gamma (γ) radiation at 20, 40, 60 and 80 Gray (Gy). Per cent shoot regeneration from y-irradiated calli in the three varieties ranged from 90 to 93.8 at 20 Gy, 83.3 to 87.5 at 40 Gy, 30 to 36.4 at 60 Gy and 0 at 80 Gy. Upon irradiating shoots, subsequent shoot proliferation in the three varieties ranged from 90.9 to 93.1% at 20 Gy, 82.6 to 84.0% at 40 Gy and 27 to 32.3% at 60 Gy, whereas 80 Gy dose was 100% lethal. Thus, 60 Gy dose of y-radiation was found to be optimum for carrying out mutagenesis of both callus and callus derived shoots. In the field, different irradiated clones of the same variety exhibited huge variability with respect to number of canes, cane girth, cane height and sucrose content.  相似文献

Protocols have been developed for the in vitro regeneration of plants from calli derived from internode explants of chickpea (Cicer arietinum L) cv Pusa-372. Callusing was induced on both B5 and MS media supplemented with different combinations and concentrations of auxins and cytokinins, but shoot regeneration was achieved only in B5 medium supplemented with 4.0 mg l?1 IAA and 0.5 mg l?1 BAP after serial subculture of callus on media with increasing concentration of IAA and constant concentration of BAP. Rooting could not be achieved in in vitro regenerated shoots on any one of the media tried. Complete plantlets were, therefore, developed through grafting of the in vitro regenerated shoot on established root stock. The grafting methodology was found to be highly efficient and reproducible. The somaclones developed produced viable seeds which showed variability in terms of seed colour and seed weight. Thus, the protocols developed in this study remove one important bottleneck in the development of transgenic chickpea.  相似文献
