Previous work led to the proposal that the precision feeding of a high-concentrate diet may represent a potential method with which to enhance feed efficiency (FE) when rearing dairy heifers. However, the physiological and metabolic mechanisms underlying this approach remain unclear. This study used metabolomics analysis to investigate the changes in plasma metabolites of heifers precision-fed diets containing a wide range of forage to concentrate ratios. Twenty-four half-sib Holstein heifers, with a similar body condition, were randomly assigned into four groups and precision fed with diets containing different proportions of concentrate (20%, 40%, 60% and 80% based on DM). After 28 days of feeding, blood samples were collected 6 h after morning feeding and gas chromatography time-of-flight/MS was used to analyze the plasma samples. Parameters of oxidative status were also determined in the plasma. The FE (after being corrected for gut fill) increased linearly (P < 0.01) with increasing level of dietary concentrate. Significant changes were identified for 38 different metabolites in the plasma of heifers fed different dietary forage to concentrate ratios. The main pathways showing alterations were clustered into those relating to carbohydrate and amino acid metabolism; all of which have been previously associated with FE changes in ruminants. Heifers fed with a high-concentrate diet had higher (P < 0.01) plasma total antioxidant capacity and superoxide dismutase but lower (P ≤ 0.02) hydroxyl radical and hydrogen peroxide than heifers fed with a low-concentrate diet, which might indicate a lower plasma oxidative status in the heifers fed a high-concentrate diet. Thus, heifers fed with a high-concentrate diet had higher FE and antioxidant capacity but a lower plasma oxidative status as well as changed carbohydrate and amino acid metabolism. Our findings provide a better understanding of how forage to concentrate ratios affect FE and metabolism in the precision-fed growing heifers.  相似文献

The objective of the study was to generate a database of in situ ruminal degradability of crude protein (CP) from different samples of a range of concentrate feedstuffs comprising protein, energy or protein+energy feeds commonly offered to ruminant animals in European countries. The in situ disappearance of CP was calculated using the in situ nylon bag technique where the test feedstuffs were subject to ruminal incubation in four Friesian steers offered grass silage and concentrate. Disappearance of CP from the test feeds from the rumen was measured at 0, 2, 4, 8, 14, 24 and 48 h. The exponential model of Ørskov and McDonald (1979) was used to measure degradation kinetics of CP. Test protein feeds were sunflower meal (SUN), rapeseed meal (RAP), soyabean meal (SBM) and cottonseed meal (CSM). Test energy feeds were palm kernel meal (PK), pollard (PO), barley (BA) and beet pulp (BP). Test protein+energy feeds were maize distillers grains (MDG), maize gluten feed (MGF), copra meal (CO) and malt combings (MC). The effective degradability of CP (EDP) in the protein feed SBM, where outflow rate (k)=0.02 was not influenced (P>0.05) by the sample of feed used. For the energy feeds, the EDP in PO for k=0.05 h−1, 0.06 h−1 and 0.08 h−1 and EDP in BA for k=0.08 h−1 were not significantly affected (P>0.05) by the sample of feed used. The data has shown that for the majority of feeds examined in this study, the different samples of any one feed are not equal in nutritive value and it is necessary to screen feeds for nutritive value before using them in ration formulation systems.  相似文献

Several reports have suggested that a treatment before in vitro maturation might improve oocyte competence and increase its developmental potential. Therefore, the objectives of the present study were to establish the kinetics of IVM in Zebu oocytes, to assess the effect of 6-dimethylaminopurine (6-DMAP), a phosphorylation inhibitor, on meiotic resumption, and to verify the developmental potential of the blocked oocytes after removal of the inhibitory conditions. To establish the kinetics of in vitro maturation 1422 oocytes were obtained from Nellore cows ovaries and matured in presence and absence of gonadotropins. Samples of oocytes were taken from culture at 0, 6, 9, 12, 15, 18, 21 and 24h, and the oocytes were fixed, stained and evaluated for nuclear morphology. Germinal vesicle break down (GVBD) occurred between 6 and 12h of culture in both groups. By 21h the majority of the oocytes had reached metaphase II in presence (71%) and absence (62%) of gonadotropins. In order to examine the inhibitory effect of 6-DMAP, 585 oocytes were cultured for 12, 18 and 24h in the presence or absence of 2mM of 6-DMAP. At each time point the oocytes were evaluated for nuclear morphology. To test the reversibility of meiotic inhibition 366 oocytes were incubated for 0, 12, 18 and 24h in the presence of 6-DMAP and then were transferred to the maturation medium and cultured for further 24h. A total of 429 oocytes were used to evaluate the developmental potential after meiotic inhibition. The oocytes were cultured in the presence of 6-DMAP for 0, 12, 18 and 24h, and then were matured, fertilized and cultured in vitro. Culture of bovine oocytes in the presence of 6-DMAP up to 24h completely blocked GVBD with more than 90% of the oocytes at GV stage. The inhibitory effect of 6-DMAP was fully reversible since maturation rates were similar (P>0.05) among all treatment groups. The evaluation of embryo development after various periods of meiotic blockage showed that inhibition, regardless the time period, had no effect (P>0.05) on penetration and cleavage rates. However, the proportion of embryos at blastocyst stage was reduced after inhibition for 12 (20.2%), 18 (20.1%) and 24h (19.0%) compared with the control group (35.6%). 6-DMAP has a reversible effect on maintenance of meiotic arrest, but reduced further embryo development.  相似文献

The effective degradability of dry matter (DM), crude protein (CP) and amino acids (AA), and the intestinal effective digestibility (IED) of DM and CP of a green forage corn (GC) and its silage (EC) were determined on freeze-dried samples using three rumen and duodenum cannulated wethers. Two rumen incubations with duplicate bags were performed for each feed. Rumen degradation was determined on one series of bags from each incubation. The other series was freeze-dried and used to determine IED using mobile nylon bags. Microbial contamination of rumen incubated residues (determined with 15N techniques) fitted exponential functions, which showed a greater microbial contribution in EC than in GC in the undegradable DM (18.6% vs. 13.5%) and CP (81.7% vs. 69.4%). Degradability was calculated considering the particle rumen outflow rate (kp : 0.056/h) of the EC (EDp) or additionally the rate of comminution and mixing (kc : 0.130/h) of these particles (EDcp). Ensiling increased EDp (9.33%, p < 0.01) or EDcp (5.30%, p = 0.062) of DM and was associated with losses of nitrogen and with large changes in the AA profile. It is necessary to correct the microbial contamination, because it represents 32.0% (GC) and 42.5% (EC) of the undegraded CP when using kp and kc . Ensiling caused higher degradabilities for some AA as well as large differences in the changes due to the rumen fermentation on the AA profile. However, it had only limited effects on the undegraded protein profile. Ensiling also reduced the IED of DM (23.3% vs. 14.6%; p = 0.057). In conclusion, results do not show losses of nutritive value by ensiling corn cut at vitreous grain stage.  相似文献

The effect of dietary protein content on the uncoupling proteins (UCP) 1, 2 and 3 expression in a number of tissues of Zucker lean and obese rats was studied. Thirty-day-old male Zucker lean (Fa/?) and obese (fa/fa) rats were fed on hyperproteic (HP, 30% protein), standard (RD, 17% protein) or hypoproteic (LP, 9% protein) diets ad libitum for 30 days. Although dietary protein intake affected the weights of individual muscles in lean and obese animals, these weights were similar. In contrast, huge differences were observed in brown adipose tissue (BAT) and liver weights. Lean rats fed on the LP diet generally increased UCP expression, whereas the HP group had lower values. Obese animals, HP and LP groups showed higher UCP expression in muscles, with slight differences in BAT and lower values for UCP3 in subcutaneous adipose tissue. The mean values of UCP expression in BAT of obese rats were lower than in their lean counterpart, whereas the expression in skeletal muscle was increased. Thus, expression of UCPs can be modified by dietary protein content, in lean and obese rats. A possible thermogenic function of UCP3 in muscle and WAT in obese rats must be taken into account.  相似文献

The objective of this study was to evaluate the effect of a fresh sugarcane-based diet and different roughage-to-concentrate ratios (70:30, 60:40, 40:60 and 20:80) on the rumen microbiota associated with rumen fermentation parameters and the intake and apparent digestibility of nutrients in Nellore steers. Eight rumen-cannulated Nellore steers (331 ± 8 kg BW) were distributed in a double 4 × 4 Latin square design balanced for the control of the residual effect. The ruminal pH decreased (p < 0.01) and the concentrations of N–NH3, isovaleric and valeric acids increased linearly (p < 0.05) with an increase dietary concentrate level. Furthermore, an increased concentrate proportion reduced the population of Fibrobacter succinogenes and Ruminococus flavefaciens (p < 0.01) and increased the population of Selenomonas ruminantium and Megasphaera elsdenii (p < 0.01). The protozoa count revealed a predominance of the genus Entodinium. The synthesis of microbial N [g/d] and the efficiency of microbial synthesis [g of microbial N/kg of organic matter apparently digested in the rumen] increased as the proportion of concentrate was increased (p < 0.05). Therefore, it can be concluded that an increasing proportion of concentrate in sugarcane-containing diets enhances the synthesis of microbial protein and does not alter the fibre digestibility, although the population of fibre fermenting bacteria was reduced.  相似文献

Aims: To determine the effects of the removal of forage in high‐concentrate diets on rumen fermentation conditions and rumen bacterial populations using culture‐independent methods. Methods and Results: Detectable bacteria and fermentation parameters were measured in the solid and liquid fractions of digesta from cattle fed two dietary treatments, high concentrate (HC) and high concentrate without forage (HCNF). Comparison of rumen fermentation conditions showed that duration of time spent below pH 5·2 and rumen osmolality were higher in the HCNF treatment. Simpson’s index of 16S PCR‐DGGE images showed a greater diversity of dominant species in the HCNF treatment. Real‐time qPCR showed populations of Fibrobacter succinogenes (P = 0·01) were lower in HCNF than HC diets. Ruminococcus spp., F. succinogenes and Selenomonas ruminantium were at higher (P 0·05) concentrations in the solid vs the liquid fraction of digesta regardless of diet. Conclusions: The detectable bacterial community structure in the rumen is highly diverse. Reducing diet complexity by removing forage increased bacterial diversity despite the associated reduction in ruminal pH being less conducive for fibrolytic bacterial populations. Quantitative PCR showed that removal of forage from the diet resulted in a decline in the density of some, but not all fibrolytic bacterial species examined. Significance and Impact of the Study: Molecular techniques such as DGGE and qPCR provide an increased understanding of the impacts of dietary changes on the nature of rumen bacterial populations, and conclusions derived using these techniques may not match those previously derived using traditional laboratory culturing techniques.  相似文献

Groups of albino rats were fed low and high protein diets and the former group was fed with different supplements. The supplements used were niacin, pyridoxine and glutamic acid. A low protein diet was found to decrease the activities of l -glutamate-NAD-oxidoreductase and l -glutamate-1-carboxy-lyase in the cerebrum; this confirmed the results of previous work in this laboratory. Addition of glutamic acid to a low protein diet was found to reverse the effects of a low protein diet to some extent. Groups of animals were fed kodri (Paspalum scorbiculatum), a millet deficient in lysine, with or without lysine itself or foodstuffs rich in it, e.g. moth bean (Phaseolus aconitifolius Jacq.), peas (Pisum sativuni) and skim milk powder. When combined, the groups fed on the lysine or lysine-source supplemented diets were found to perform better in a water maze on the basis of the relative ease with which they could reverse a previous learned pattern. They also had higher activities of l -glutamate-NAD-oxidoreductase and l -glutamate-1-carboxy-lyase than groups with no lysine or lysine source added. There was no significant difference between the groups receiving the different lysine supplements.  相似文献

Cattle are often fed high concentrate (HC) diets to increase productivity, although HC diets cause changes in ruminal environment such as pH reduction. Despite those well-documented changes in cattle fed HC diets, there is currently a paucity of data describing the molecular events regulating the ruminal environment. Our objective was to gain an understanding of which genes are differentially expressed in ruminal tissue from Holstein cows fed a HC comparing to low concentrate (LC) diet using microarray analysis using a bovine 24 k microarray. A total of 5,200 differentially expressed genes (DEG) were detected for cows fed HC relative to LC. The DEG were firstly annotated with gene ontology (GO) and Kyoto encyclopedia of Genes and Genomes (KEGG), indicating that the DEG were associated with catalytic activity and MAPK pathway, respectively. Further characterization using GeneCodis identified patterns of interrelated annotations for the DEG to elucidate the relationships among annotation groups revealed that a cAMP-dependent protein kinase A catalytic subunit beta (PRKACB), may be associated with ruminal tissue maintenance. The results contributed to understanding of the regulatory mechanisms at the mRNA level for Holstein cows fed at different concentrate ratio diets.  相似文献

Biotin is involved in many vital metabolic pathways and must be provided for an efficient fermentation in the rumen, as well as for the intermediary metabolism of the host animal. Factors influencing ruminal biotin metabolism and output are widely unknown at present. Therefore, dairy cows fitted with permanent cannulas in the dorsal rumen and in the proximal duodenum were fed differently composed diets, and the biotin flow at the proximal duodenum was measured. The diets (on DM basis) consisted of 8.9 kg grass hay (Diet 1), 8.9 kg corn silage plus 2.0 kg concentrate (Diet 2), or 7.3 and 7.4 kg grass silage plus 10.0kg concentrate (Diets 3 and 4). The concentrate in Diets 3 and 4 contained 87% wheat and corn grain, respectively. The cows were pre-fed the rations for 21 days. Thereafter duodenal digesta was sampled every two h for 5 days. Cr2O3 served as a flow marker and the microbial proportion of total nitrogen at the duodenum was estimated by near infrared spectroscopy (NIRS). The duodenal flow of biotin was not related to biotin intake, but to the amount of fermented organic matter (FOM) and the amount of microbial protein (Biotin [mg/d] = 0.518 kg FOM - 0.300; r=0.85 and biotin [mg/d] = 0.012 x g microbial protein + 1.478; r = 0.84), irrespective of the composition of the diet fed. Mean daily biotin flow was 0.48 +/- 0.11 mg/kg FOM without any systematic effect of diet composition. The ruminal biotin balance, calculated as the difference between biotin flow at the duodenum and biotin intake, was positive (1.4 - 2.0 mg/d) in cows fed the mixed roughage/concentrate diets and negative (-0.71 mg/d) when the pure hay diet was fed.  相似文献

Labeo rohita, commonly called rohu is one of the most important fish species for aquaculture in India. Digestive enzyme response and metabolic profile of fingerling L. rohita to different dietary crude protein (CP) levels (viz. 25, 30, 35 and 40%) were studied in an attempt to optimize a practical diet formulation for this species. After 45 days of feeding, activity of digestive enzymes and metabolite concentrations were assayed. Amylase, lipase and alkaline phosphatase (ALP) activities were not influenced by the dietary protein, but proteolytic and acid phosphatase (ACP) activities varied (P<0.05) between the treatments. Proteolytic activity showed a second order polynomial relationship with dietary crude protein (CP) as Y = 0.0734X(2) + 4.937X - 68.37, r(2)=0.97. A positive correlation was observed between dietary CP and amylase (r(2)=0.78). All the metabolites except muscle glucose showed significant change corresponding to the dietary protein levels. Glucose and glycogen levels corresponded to the dietary carbohydrate levels. Muscle and plasma pyruvic acid increased as the crude protein in the diet increased, whereas liver pyruvic acid showed the opposite trend. Muscle protein content was not affected by dietary CP. Protein fractions in plasma (total protein, albumin and globulin) showed maximum values in 30% CP fed group. It is concluded that proteolytic activity and ACP are the major digestive enzymes responsive to dietary CP in L. rohita fingerlings. Considering the cost effectiveness of the diet, and based on liver and plasma free amino acid levels and plasma protein fractions, 30% crude protein is recommended as the optimal dietary protein for L. rohita fingerlings.  相似文献

The applicability of the plant enzyme ficin (EC 3.4.22.3) for the prediction of protein degradability of ruminal feeds was examined. Optimum pH and the effect of enzyme concentration, incubation time and amylase pretreatment for high-starch feeds on protein solubility with ficin (FS) were determined. The predictive ability of ficin assay in the conditions specified for particular groups of feeds was estimated using 61 feeds including protein meals (7), cereal grains (2), silages (9), sugar beet pulp, dried forages (29) and compound feeds (13) of known in situ effective protein degradability (ED) determined with cows.Optimum incubation time was 2–3 h, enzyme amount (per 100 mg of crude protein in feed sample) equivalent to 9 U for compound feeds and concentrate ingredients not treated with amylase, 18 U for forages. Close linear relationships between the in vitro FS (X, %) and in situ estimates of ED (Y, %) were found. The regression equation for all samples (n = 61) was Y = 1.018 X + 1.331, R2 = 0.92 (P < 0.001), SEE 4.17. After splitting to forages (FOR) and compound feeds and ingredients (CON) SEEs were 4.25 and 3.56%, respectively. FS values accounted for 85, 92 and 95% of the variation in ED measured in situ for FOR, FOR + CON and CON samples, respectively.  相似文献

Experimental data on phytate phosphorus utilisation by ruminants are scarce. The aim of this study was to estimate the phytase activity of rumen micro-organisms when phytate phosphorus supply is high. A semi-continuous culture system fermentor (RUSITEC) was used. The inoculum was obtained from eight goats fed on either high or low forage level diets. Experimental buffers only differed by the nature of phosphorus monosodium phosphate vs. corn sodium phytate. The nylon bags containing 15 g DM of substrate were removed after a 48-hour incubation period. The system was maintained for 15 days: 5 days for adaptation, in order to obtain a steady state, and 10 days for sampling and recording. No significant differences were observed for DM digestibility, gas production, pH, N-NH3, and SCFA for the different treatments. Bacterial efficiency of phytate phosphorus utilisation was significantly higher (p < 0.001) with organic P, but remained lower than the data usually reported in the literature. These results may be explained by the relative saturation of bacterial phytase activity when the buffer contains a high level of phytate phosphorus.  相似文献

The juvenile development and survival, and demographic parameters of the predatory mite Neoseiulus cucumeris (Oudemans) (Acari: Phytoseiidae) fed on pollen of castor bean, tulip, apple, Christmas cactus, horse-chestnut, maize, and birch were assessed under laboratory conditions. Deprivation of food and pollen of castor bean plants resulted in 100 % juvenile mite mortality. Feeding mites with tulip and horse-chestnut pollen resulted in the shortest development and the highest total fecundity. Adult mites fed on birch, tulip, maize, and apple pollen lived significantly longer compared with those fed on pollen of horse-chestnut and Christmas cactus. The intrinsic rate of natural increase ranged between 0.1013 ♀♀//day for maize and 0.1806 ♀♀//day for horse-chestnut pollen as food. Net reproductive rate was the lowest when fed with maize pollen and highest when fed with horse-chestnut pollen. Population doubling time was highest on maize pollen and shortest on horse-chestnut pollen. Our study revealed that birch, tulip, horse-chestnut, apple, and maize pollen can be used by N. cucumeris from early spring to late summer as a suitable alternative food in periods when prey in the field are scarce or absent.  相似文献
