Bioactive natural compounds from garlic and onions have been the focus of researches for decades, firstly due to their pharmacological effects, and secondly due to their defence properties against plant diseases. In fact, garlic and onion, belonging to Allium genus, are among the oldest food plants known since ancient times and used as ingredient of many recipes and for therapeutic properties. These plants are well known to produce bioactive apolar sulphur compounds but less is known about their polar natural compounds, such as phenols, sapogenins and saponins, that are more stable to cooking, So, we continued our work on the discovery of polar bioactive metabolites from Allium with the isolation of a number of sapogenins and saponins from the wild onion species Allium elburzense, Allium hirtifolium, Allium atroviolaceum, and Allium minutiflorum, and, more recently, from the cultivated white onion, Allium cepa, and garlic, Allium sativum. In particular, the sapogenins and saponins isolated from A. elburzense and A. hirtifolium, named elburzensosides and hirtifoliosides respectively, exhibited significant antispasmodic properties. In addition, the saponins named minutosides isolated from A. minutiflorum showed promising antimicrobial activity. More recently the phytochemical analysis of A. cepa and A. sativum has been undertaken and afforded the characterization of saponins, phenols and N-cynnamic amides which showed significant antifungal activity.  相似文献

The genotoxicity of wastewater samples from sewage, and industrial effluent from the Amritsar, India, area were investigated using the Allium micronucleus and anaphase aberration assays. Raw sewage samples and acetone extracts of the dehydrated sewage were use for treatment of the Allium roots. Industrial effluents were collected and stored in the form of sludge (semi-dried matter). The acetone extracts of the sludge samples were also used for treatment of the Allium roots. From the Allium root micronuclei tests on the sewage extracts, no significant increase in the number of micronuclei was found in comparison with negative controls. All the other extracts from industrial effluent showed positive responses both in the micronucleus and anaphase aberration assays.  相似文献

A novel antifungal protein, designated allivin, was isolated from bulbs of the round-cloved garlic Allium sativum var. round clove with a procedure involving ion exchange chromatography on DEAE-cellulose, affinity chromatography on Affi-gel blue gel, ion exchange chromatography on CM-Sepharose and FPLC-gel filtration on Superdex 75. Allivin possessed an N-terminal sequence demonstrating very little similarity to sequences of Allium sativum chitinases and ribosome inactivating proteins. Allivin exhibited a molecular weight of 13 kDa in gel filtration and SDS-polyacrylamide gel electrophoresis. It displayed antifungal activity against Botrytis cinerea, Mycosphaerella arachidicola and Physalospora piricola. It inhibited translation in a cell-free rabbit reticulocyte system with an IC50 of 1.6 microM.  相似文献

Streptomyces sp. strain Wigar10 was isolated from a surface-sterilized garlic bulb (Allium sativum var. Purple Stripe). Its genome encodes several novel secondary metabolite biosynthetic gene clusters and provides a genetic basis for further investigation of this strain's chemical biology and potential for interaction with its garlic host.  相似文献

Brassinosteroids are of universal occurrence in plants. They have been reported to affect plant growth and development through a spectrum of physiological responses. Recently they are reported to confer resistance in plants against a number of biotic and abiotic stresses. In the present study, a brassinosteroid was isolated from Aegle marmelos Correa. (Rutaceae) which was characterized to be 24-epibrassinolide (EBL) using various spectroscopic techniques (TLC and ESI-MS analysis). It was evaluated for the antigenotoxicity against maleic hydrazide (MH) induced genotoxicity in Allium cepa chromosomal aberration assay. It was shown that the percentage of chromosomal aberrations induced by maleic hydrazide (0.01%) declined significantly with 24-epibrassinolide treatment. EBL (10−7 M) proved to be the most effective concentration with 91.8% inhibition. This is the first report on the isolation of 24-epibrassinolide from Aegle marmelos and its antigenotoxic effects against MH employing Allium cepa chromosomal aberration assay.  相似文献

Phylogenetic relationships between Allium and the monotypic Himalayan genus Milula were analyzed using sequences of the nuclear ribosomal DNA internal transcribed spacer (ITS) region and of the intergenic spacers from the chloroplast trnD(GUC)-trnT(GGU) region. Both marker systems unambiguously placed Milula spicata within Allium subgenus Rhizirideum, close to A. cyathophorum. Morphologically, the main difference between Allium and Milula is the conspicuous spicate inflorescence of Milula vs the mostly capitate or umbellate inflorescences in Allium. Anatomical investigations of leaf characters support a close relationship of Milula with A. cyathophorum and A. mairei, whereas root characters are distinctive from other species of section Cyathophora. To maintain Allium as monophyletic, Milula has been included as A. spicatum in Allium subgenus Rhizirideum.  相似文献

In Allium wakegi, which is an allodiploid species between Allium cepa and Allium fistulosum, each genome can be clearly distinguished using genomic in situ hybridization (GISH). Genomic DNA of A. cepa and A. fistulosum is differentiated both qualitatively and quantitatively. We wanted to isolate nucleotide sequences that give genome-specific signals on A. cepa chromosomes in GISH experiments in A. wakegi. We isolated 23 clones that show GISH-like signal patterns in fluorescence in situ hybridization (FISH) and analyzed their distribution in the A. cepa- and A. fistulosum-derived genomes of A. wakegi. There was considerable variation in the abundance and distribution of these cloned sequences on the chromosomes of the two species. The degree of A. cepa specificity varied among the clones. Twenty-two of the clones showed an even distribution over most chromosome arms with some clustering in the pericentromeric regions, but one clone showed very distinct terminal signals on some chromosomes. Whereas these sequences are not specific for A. cepa, changes in bases in nucleotide sequences and in their amount result in genome-specific characteristics in GISH experiments.  相似文献

The aqueous extract of Allium sativum bulbs showed an antifungal effect against the fungal skin pathogen, Trichophyton rubrum, isolated from infected patients. For a given concentration (200 mg of bulbs/1 ml), the volume of the aqueous garlic extract loaded on to the discs, and the diameter of the inhibitory zone formed around the disc on the fungal lawn showed positive correlation. Extract-included microbial assay confirmed the antifungal effect of Allium sativum. The extract was not heat stable, it lost its antifungal property above 60 °C. A fully normal meiosis was found inAllium sativum L. by means of PMC analysis. Quadrivalents were formed only in several cultivars. The cultivation of inflorescence stem (after the exstirpation of bulblets) in a tetracycline solution of 900 i.u. induced a formation of fertile flowers. The first pollen mitosis was studied and the pollen parameters compared with those ofAllium cepa L. Evidence for the pollen sterility was given by crossings on sterile types. Allium L. (Alliaceae), a genus of major economic importance, exhibits a great diversity in various morphological characters and particularly in life form, with bulbs and rhizomes. Allium species show variation in several cytogenetic characters such as basic chromosome number, ploidy level, and genome size. The purpose of the present investigation was to study the evolution of nuclear DNA amount, GC content, and life form. A phylogenetic approach was used on a sample of 30 Allium species, including major vegetable crops and their wild allies, belonging to the 3 major subgenera Allium, Amerallium, and Rhizirideum and 14 sections. A phylogeny was constructed using internal transcribed spacer (ITS) sequences of 43 accessions representing 30 species, and the nuclear DNA amount and the GC content of 24 Allium species were investigated by flow cytometry. For the first time, the nuclear DNA content of Allium cyaneum and Allium vavilovii was measured, and the GC content of 16 species was measured. We addressed the following questions: (i) Is the variation in nuclear DNA amount and GC content linked to the evolutionary history of these edible Allium species and their wild relatives? (ii) How did life form (rhizome or bulb) evolve in edible Allium? Our results revealed significant interspecific variation in the nuclear DNA amount as well as in the GC content. No correlation was found between the GC content and the nuclear DNA amount. The reconstruction of nuclear DNA amount on the phylogeny showed a tendency towards a decrease in genome size within the genus. The reconstruction of life form history showed that rhizomes evolved in the subgenus Rhizirideum from an ancestral bulbous life form and were subsequently lost at least twice independently in this subgenus.  相似文献

Allium macrostemon is an important medicinal and edible plant. Its systematic position and taxonmical classification remain controversial to date. To explore this issue, we reconstructed phylogenetic relationships among Amacrostemon and other related taxa using nrDNA ITS and cpDNA trnL F markers. The phylogenetic trees derived from Bayesian inference and maximum parsimony analysis showed that Amacrostemon is monophyletic, and has a close relationship with some species of polyphyletic sections Caerulea and Pallasia instead of sections Codonoprasum and Allium. The including of Amacrostemon within section Allium was not supported by both molecular data and morphological characters of spathe, filaments and ovary. Allium macrostemon should be included in a new section, however further studies using additional samples (especially those from Central Asia) is necessary. In addition, we also provided a discussion on the phylogenetic relationships among four original plants (Amacrostemon, Achinense, Acaeruleum and Aneriniflorum) of Allii Macrostemonis Bulbus and systematic position of partial species of section Pallasia.  相似文献

为了揭示内蒙古葱属野生种表型遗传多样性,采用方差分析、主成分分析、聚类分析等方法,对收集的78份野生种的种质资源14个表型性状进行了遗传多样性评价。结果表明:内蒙古葱属野生资源存在丰富的遗传多样性。(1)14个表型性状平均变异系数为43.0%,叶片宽的变异系数最大,为94.7%;种子厚的变异系数最小,为14.6%。14个表型性状在居群间的差异除叶片宽达到显著(P0.05)水平外,其余性状均表现差异不显著;(2)14个表型性状可归成为6个主成分因子,累计贡献率达到80.77%,最大程度上反映了所有78份葱属种质资源的表型特征,在前6个主成分包括的14个农艺性状中的株丛直径、叶片长、叶片宽、单株叶片数、花序长度、花序宽度和种子宽等性状是造成葱属种质资源表型差异的主要因素;(3)14个表型性状间存在显著或极显著的相关性;(4)采用欧氏距离系统聚类法将供试材料分为7大类,78份葱属种质资源基本上按种区分开,并和地理条件有一定的关系,种间和材料间表型差异很明显。本研究为内蒙古葱属资源的利用提供了科学依据。  相似文献

对不同倍性及其非整倍体韭Allium tuberosum和野韭Allium ramosum核糖体DNA的内部转录区(ITS)进行了PCR扩增和克隆,并测定10个ITS1和ITS2的序列,用ITS1 ITS2的序列探讨Allium tuberosum和Allium ramosum的系统发育关系。研究表明,Allium tuberosum与Alli-um ramosum种间的平均Kimura遗传距离仅为0.0146,二者是一对近缘种。虽然韭经历了长期的人工选择和自然选择,但与野韭的分化仍然较小。韭不同倍性及其非整倍体之间,其ITS序列亦无明显差异。  相似文献

A bioregulator that has physicochemical and biological properties similar to a group of bioregulators isolated from various animal tissues has been found in the bulb onion (Allium cepa L.). It was determined that the biological action of the plant bioregulator is determined by a peptide with molecular weight of 4036 +/- 2 Da whose 18-C-terminal amino acid sequence consisted of 18 residues. On models of seed germination of some vegetable cultures, the ability of the bioregulator isolated from supernatant of onion extract in ultralow doses (10(-13) mg of protein/ml) to inhibit growth and development was demonstrated.  相似文献

The bioactive fractions of Tectona grandis have yielded seven apocarotenoids, two of which have been isolated for the first time as natural products (tectoionols A and B). The chemical structures were determined through 1D and 2D nuclear magnetic resonance experiments. The absolute configuration of tectoionol A was determined using a modified Mosher methodology. Some NMR assignments for the compounds 9(S)-4-oxo-7,8-dihydro-beta-ionol and 3beta-hydroxy-7,8-dihydro-beta-ionone have been corrected on the basis of g-HSQC and g-HMBC experiments. The general bioactivities of isolated compounds have been studied using etiolated wheat coleoptiles. Those compounds that presented higher levels of activity were assayed on standard target species (Lactuca sativa, Lycopersicum esculentum, Lepidium sativum and Allium cepa).  相似文献

沙葱是一种具有抗旱抗寒、抗病性和适应性强等生理特性的荒漠植物.为开发利用其固有的遗传资源,本研究利用细胞工程技术建立了沙葱(Allium mongolicum Regel)叶基愈伤组织原生质体的分离、培养和植株再生实验体系.研究结果表明,酶法分离原生质体的产率和分裂频率明显取决于用于制各原生质体的愈伤组织的状态.转代培养7~10d的松软愈伤组织可分离出大量有活性的.在附加2.0mg/L2,4-D、0.2mg/L激动素、500 mg/L水解乳蛋白、0.4 mol/L甘露醇和2%蔗糖的MS培养基中进行液体浅层培养,4~5 d后出现第一次原生质体分裂;7~10d出现第二次分裂.结果显示原生质体的分裂频率大约为5%;4周后,可见到小愈伤组织.当将原生质体分裂形成的愈伤组织转移到附加2.0mg/L6-苄氨基嘌呤(或激动素)和0.4mg/L萘乙酸(NAA)的MS固体培养基上,并在低光照条件下培养后,从愈伤组织上分化出了不定芽,进而发展成小植株,并移栽成活.本研究对沙葱抗逆遗传品质用于经济植物遗传改良的研究奠定了可行的实验基础.  相似文献

Allium cepa and garlic Allium sativa plants were used to evaluate their potential synthesis of silver nanoparticles and their antibacterial effect on Streptococcus pneumoniae and Pseudomonas aeruginosa. Transmission electron microscopy (SEM) was used to distinguish the morphology of the nanoparticles attained from plant extracts. Energy dispersive X-ray (EDX) spectrometer established the existence of elemental sign of the silver and homogenous allocation of silver nanoparticles. Diffraction by using X ray (XRD) analysis for the formed AgNPs revealed spherical plus cubical shapes structure with different planes ranged between 111 and 311 planes. The antibacterial action of AgNPs against vaginal pathogens, Streptococcus pneumoniae and Pseudomonas aeruginosa was recognized. Our work showed a rapid, eco-safety and suitable method for the synthesis of AgNPs from Allium cepa and garlic Allium sativa extracts and can be used in biomedical applications.  相似文献
