Summary Experiments on the survival ofM. phaseoli on cucurbit root pieces were carried out. The sclerotia ofM. phaseoli were almost unaffected by 10 months dry storage in the laboratory. Under dry to moderately moist soil conditions it survived for 2 months with little loss of viability. In wet soils however, the buried root pieces were colonized by other soil microorganisms which prevented the growth ofM. phaseoli.  相似文献

Germination of sclerotia of Macrophomina phaseolina was quantified by direct microscopic observation following application of experimental treatments in vitro and incubation of sclerotia in soil. To assay germination, pieces of agar containing sclerotia were macerated in dilute, liquid cornmeal agar on glass slides; thinly spread; and incubated in a saturated atmosphere for 18–22 h. Germinated sclerotia then were identified by morphological features of germ hyphae. Frequencies of germination were similar in three dilute agar media. Germination was not affected by air-drying sclerotia for 2 weeks, but it was significantly reduced after 4 weeks and greatly reduced or eliminated after 6 or 8 weeks. Survival of sclerotia for 14 days in soil was greatest at 50, 75, and 100% moisture-holding capacity, less at 0 and 25%, and least at 125% (flooded soil). Incorporation of ground poultry litter into soil at 5% by weight reduced survival of sclerotia after 13 days, and incorporation of litter at 10% nearly eliminated it. These results indicate that the direct-observation technique may be used to evaluate animal wastes and other agricultural byproducts for biocontrol activity against sclerotia of M. phaseolina in soil.  相似文献

Extracts made from Allium species were shown to exhibit marked antibiotic properties in a variety of tests. However, no evidence was obtained to suggest that intact Allium seedlings exude antibiotic compounds in sufficient quantity to cause inhibition of bacteria in the soil or in assay tests in the laboratory. Germination of sclerotia of Sclerotium cepivorum was induced by Allium extracts which were too dilute to cause antibiotic responses in laboratory tests using a number of bacteria. Synthetic methyl-methanethiolsulphinate had little effect on germination of sclerotia. The specific reversal, by Allium species and their extracts, of the mycostatic effect of unsterile soil on sclerotia of S. cepivorum does not appear to be due to antibiotic effects.  相似文献

Sclerotinia sclerotiorum survived in infected seeds of white beans as dormant mycelium in testa and cotyledons. The rate of survival averaged 85 to 89% and did not change appreciably over a 3-year period. When the infected bean seeds were sown in soil or sand, 88 to 100% failed to germinate. The seeds that failed to germinate, depending on the severity of seed infection, were rotted by S. sclerotiorum. In place of each seed, 3 to 6 sclerotia were formed. A low percentage of these sclerotia germinated carpogenically with or without preconditioning, (2.5 and 11.5% respectively). Myceliogenic germination of sclerotia with and without preconditioning was 35.5% and 70.5% on water agar and 81.0% and 93.0% on glucose agar, respectively. Both, preconditioning and nonpreconditioned sclerotia which were scattered on soil surface could germinate myceliogenically and infect bean leaves by contact. It is therefore, concluded that dormant mycelia in the infected seeds play an important role notonly in dissemination of the fungus but also in epidemiology of the disease.  相似文献

Changes in populations of microorganisms around germinating bean (Phaseolus vulgaris L.) seeds, in the rhizosphere of bean, and in a model rhizosphere were studied. Strains of Rhizobium phaseoli that were resistant to streptomycin and thiram were used, and as few as 300 R. phaseoli cells per g of soil could be enumerated with a selective medium that was devised. A direct role was not evident for bacterial competitors, lytic bacteria, antibiotic-producing microorganisms, bacteriophages, and Bdellovibrio in the suppression of R. phaseoli around germinating seeds and in the rhizosphere. Protozoa increased in numbers in the soil upon planting of the seeds. The extent of colonization of soil by R. phaseoli was inversely related to the presence of large numbers of bacteria and protozoa. Colonization of R. phaseoli was improved upon suppression of protozoa with thiram and also when the soil was amended with other protozoan inhibitors and mannitol to simulate seed and root exudation. The data support the view that the decrease in numbers of R. phaseoli is caused by an increase in protozoan predation, the protozoa increasing in number because they prey on bacteria that proliferate by using seed and root exudates as nutrients.  相似文献

Summary The germination ecology and the dynamics of the generative reproduction in populations of Digitalis purpurea L. were investigated in the field as well as in experiments. Germination of fresh seeds in the dark on moist filter paper appeared to differ between populations. These differences were eliminated when a moist natural soil functioned as germination substrate. An interaction between the spectral composition of light and the germination substrate was present. Germination in gradients of light, temperature and soil moisture revealed some clear-cut results. Germination proved to be strongly dependent on the percentage of vegetation cover. During two years of burial in litter bags, the number of buried viable seeds did not decrease. From one generation of seeds produced in a natural population, 18% was introduced into the buried seed bank, 10% germinated in autumn and 24% was present as a enforced dormant surface seed bank in late autumn.The results are discussed in relation to secondary succession. can be derived from Milton (1936), Salisbury (1942) and Thompson and Grime (1979). Soil disturbance and germination seem to be correlated in D. purpurea (Grime 1979). The purpose of this study is to analyse the dormancy and germination behaviour of D. purpurea in relation to the relevant environmental factors in order to explain the mechanisms of entry into, and the escape of D. purpurea seeds from a seed bank. Furthermore, an attempt will be made to quantify seed rain as well as the fate of different germinating and non-germinating seed rain fractions in space and time per unit area, in different stages of succession.  相似文献

Factors controlling the timing of seed germination were investigated in the small succulent winter annual Sedum pulchellum Michx. (Crassulaceae) in its natural habitat on unshaded limestone outcrops in northcentral Kentucky. At maturity in early July the dormant seeds are not dispersed but are retained in the fruits on the standing dead plants until September and October. Many, but not all, of the seeds afterripen in the fruits during summer, and at the time of dispersal some of them are dormant and some are nondormant. Germination and annual population establishment occur in September and October from seed reserves that have been in the soil for one or more years and from seeds produced in the current year. Germination of nondormant seeds may be prevented in autumn by lack of the appropriate combination of environmental factors including light, temperature and soil moisture in the seed's microsite. The effect of low winter temperatures on ungerminated seeds in the population is to induce nondormant seeds into secondary dormancy and to prevent afterripening of dormant seeds. Thus, in spring all the seeds in the population's seed reserve are dormant. During spring and summer some of these seeds afterripen, and they germinate in autumn when, and if, germination requirements are fulfilled.  相似文献

Summary The effect of different sulphur and phosphorus compounds on the growth and reproduction of three fungi causing storage rot, viz.,Fusarium solani, Botryodiplodia ananassae andMacrophomina phaseoli has been studied. Sixteen different sources of sulphur were used and out of them magnesium sulphate was found to be most favourable for the growth and reproduction of all the three fungi. Sodium sulphite and sodium bisulphite were toxic. Potassium metabisulphite prevented growth ofF. solani and M. phaseoli while it supported moderate growth ofB. ananassae. Only magnesium sulphate could induce the sporulation ofB. ananassae while sporulation and sclerotial development ofF. solani andM. phaseoli respectively varied with the type of sulphur sources used. Optimum concentration of magnesium sulphate was also determined and it was found that the growth and sporulation ofF. solani andB. ananassae were best at 0.375 g/l and 0.75 g/l.M. phaseoli tolerated higher doses of this substance as the best growth and excellent sclerotial development were recorded at 3.0 g/l (the maximum concentration used). Phosphorus was found to be essential for the present fungi as none of them could grow in complete absence of this substance. Onthophosphates and nucleic acid, were found to be favourable sources for growth and reproduction of the 3 organisms.  相似文献

Sixty-three different plants, from a wide range of families, were tested for their effects on germination of sclerotia of Sclerotium cepivorum. Germination was stimulated only by members of the genus Allium. Water extracts of Allium species also stimulated germination of sclerotia, but extracts of other plants, and a number of nutrient solutions, had little effect. There was no evidence of inhibition of sclerotial germination by plants which failed to elicit a germination response.  相似文献

Summary A study was made of the effect of soil fungistasis on the germination of conidia ofVerticillium dahliae Kleb. Conidia germinated on steam or propylene oxide sterilized soil, but not on unsterilized soil of six types tested. Peptone (0.5 per cent) nullified fungistasis. Glucose (0.5 per cent) had no effect on fungistasis. Wheat straw or lignin amended to unsterilized soil had no effect on spore germination after sterilization. Fungistasis was restored to sterilized soil by incorporating natural soil at various ratios or by lowering the soil pH. Fungistasis was also restored by incubating various micro-organisms in sterile soil. The fungistasis so obtained was nullified by incorporation of nutrients or by sterilization. The results suggest that nutrient deficiency as well as inhibitory substances of microbial origin might play a role in soil fungistasis. 5012 plant and soil  相似文献

Studies were conducted to determine the effects of soil moisture (9, 16 or 24% w/w) and temperature (5, 15, 20 or 25°C) on the control of sclerotia of Sclerotinia sclerotiorum by five fungal agents in sterile and natural field soil. All five biocontrol agents were effective in reducing the survival of sclerotia of S. sclerotiorum in sterile soil under dry (9% moisture) or wet (24% moisture) conditions at 20°C, but only Coniothyrium minitans was effective in natural soil. Coniothyrium minitans was the most effective in reducing sclerotial viability at the temperature range of 15–25°C. Trichoderma virens was effective against sclerotia of S. sclerotiorum to a lesser extent than C. minitans , and in non-autoclaved soil, it performed best at 25°C. Although Epicoccum purpurascens , Talaromyces flavus and Trichothecium roseum were effective against sclerotia of S. sclerotiorum in some instances, they were less effective than C. minitans and T. virens . Sclerotia of S. sclerotiorum conditioned for myceliogenic germination were more vulnerable to attack by the biocontrol agents than dormant sclerotia. The implications are discussed with respect to enhancement of biological control of crop diseases caused by S. sclerotiorum in different geographic regions.  相似文献

Summary Interactions of 46 isolates of fungi with an isolate ofM. phaseoli from cotton, was studied in agar culture. They were grouped into 4 different types of reactions.T. viride was found to inhibit the growth ofM. phaseoli and grow over its colony. The hyphae ofT. viride were observed to coil around the hyphae ofM. phaseoli. A majority of the isolates tested had no effect on the growth ofM. phaseoli. The response of sclerotia of Sclerotium cepivorum to a number of plant extracts and nutrient solutions was tested, aseptically, on agars, silica gels, filter paper discs, and on columns of soil. There was no specific response of sclerotia to extracts of Allium species in any of these experiments, whereas in unsterile soil consistently high levels of germination were obtained only with Allium extracts. It was shown that sclerotia are subject to the mycostatic influence of unsterile soil. With S. cepivorum soil mycostasis appears to be specifically reversed by Allium species or by water extracts made from them.  相似文献

The nutrition of developing fruits of Ricinus communis was studiednear Perth, Western Australia, where the species grows as aweed on poor sandy soil. Fruits required 60 days to mature anddehydration of the capsule began 20 days before the seeds ripened.Mature seeds accumulated 49 per cent of the fruit dry matterand over 80 per cent of its P, Zn and Cu, 50–80 per centof its Mg, N, Fe and Mn, 41–46 per cent of its S and Caand 11–21 per cent of its K and Na. Losses of nutrientsfrom capsules during fruit ripening were: Zn, 73 per cent, P,42 per cent, Cu, 23 per cent and Mn, 8 per cent. Dry matter,N, K, S, Ca, Mg, Na and Fe were not withdrawn from capsules.Apparent retranslocation from capsules could have provided from6–28 per cent of the Zn, Mn, P and Cu in mature seeds.Seeds from plants on poor sandy soil were small but had adequatelevels of nutrients when compared with those from plants growingon a fertile loam. Concentrations of all nutrients except P were higher in youngcapsules than in young seeds, but levels of N, P, Mg, Fe, Znand Cu were higher in mature seeds than in mature capsules.The intake of most nutrients by fruits was out of phase withdry matter accumulation, especially in capsules, and the elementsappeared to accumulate in fruit parts independently of eachother. Glutamine accounted for over 85 per cent of the amino-Nin phloem sap destined for fruits. Potassium made up over 90per cent of the inorganic cations in phloem exudate. Of theminor elements in the exudate, Fe was present at highest concentrationand Cu at the lowest. The results showed that retranslocation from the capsule madea very small contribution to the nutrition of seeds. It is suggestedthat R. communis would require a sustained supply of soil nutrientsto ensure maximum seed yield, partly due to the restricted retranslocationof most nutrients from capsules. Rhizoctonia tuliparum Whetzel & Arthur is a winter active soil-borne pathogen, which causes the grey bulb rot of tulips and Dutch irises. The fungus survives by means of sclerotia. It was found that the sclerotia only germinated in unsterile soil at low temperatures (0–10 °C). Germination in sterile soil occurred over a wider range of temperatures (0–25 °C) but was inhibited at 20 °C after the addition of a suitable bacterial inoculum. An unidentified pyrone antibiotic was leaked by sclerotia which had been air-dried and re-wetted. This could be detected in vitro using an agar medium seeded with a spore suspension of Bacillus subtilis Cohn emend Prazmowski. In soil, however, material leaked by sclerotia stimulated both bacteria and fungi. Undried, air-dried and oven-dried (non antibiotic) sclerotia persisted equally well in soil over 2 yr. This suggests that the resistance to lysis of sclerotial cell walls may be important in the long term survival of this organism.  相似文献

The effect of temperature on the level of dormancy of primary and secondary dormant Carex pendula and Carex remota seeds was investigated. Primary dormant and secondary dormant seeds were stratified for 4 weeks at 5, 11, 13, and 15 °C, respectively, and tested for germination at 15/5 °C in light. To obtain secondary dormant seeds, primary dormant seeds were stratified at 5 °C and afterwards at 25 °C for 4 weeks. Germination tests were carried out in water and in 25 μmol KNO3-solution to examine differences in sensitivity to nitrate between seeds relieved from primary and secondary dormancy. In both species, seeds with primary and with induced secondary dormancy showed no significant differences in germination. The two sedges showed significant differences in the effect of stratification temperatures between primary and secondary dormant seeds. Primary dormant seeds of C. pendula showed high germination (>80%) in nitrate-solution after stratification at all temperatures, while only temperatures of 5, 11, and 13 °C led to higher germination in nitrate-solution in secondary dormant seeds. Germination percentages of primary and of secondary dormant C. pendula seeds in water increased to a higher extent only after stratification at 5 and 11 °C; stratification of 11 °C was more effective in secondary than in primary dormant seeds. The only temperature that relieved primary dormancy in C. remota seeds was 5 °C where germination in water and nitrate-solution was >90%. Germination of secondary dormant seeds was increased by stratification at 11 °C independent of the test solution but higher germination after stratification at 13 °C occurred only in nitrate-solution. The results support the existence of physiological differences in the regulation of primary and secondary dormancy by temperature, and in the reaction of nitrate, at least in C. remota.  相似文献

Germination tests were done on 20 populations of Silene dioicacollected in different parts of Europe. Seeds collected fromwild plants and from their progeny growing at R.B.G. Kew, weretested using both freshly harvested and stored seeds. Responseswere compared from tests done on thermo-gradient bars and inincubators to examine germination and after-ripening processesbetween c. 2-c. 45 °C. The responses of different populations were characterized byidentifying variations due to the proportions of dormant seeds,the maximum and minimum temperatures favouring germination,and the time course of germination at particular temperaturesof freshly shed and stored seed. Germination and after-ripeningresponses of different populations displayed marked qualitativesimilarities, but differences in the degree of their expressionresulted in statistically significant quantitative differenceswhich could sometimes be correlated with features of the geographicaldistribution of the populations. The results are discussed in relation to the occurrence of thisspecies in a well-defined natural habitat in Europe, and itis concluded that they represent a situation in which fundamentallysimilar control patterns underlying the responses of every populationexamined are modulated quantitatively to produce variationsin the number of seeds germinating at particular seasons orremaining dormant within the soil.  相似文献

The method of Stanghellini and Hancock to enumerate Pythium ultimum Trow, populations in soil was modified by distributing 1 ml dilutions of soil containing the pathogen into 100–120, 2.5 mm diam. Wells made in 2% water agar in 9 × 15 mm Petri dishes. The modified method was more sensitive than the original method m which droplets or soil dilutions are placed on agar, it permitted the use of lower soil dilutions, and was rapidly performed. Sclerotia of Macrophomina phaseolina (Tassi) Goid. (=M. phaseoli [Maubl.] Ashby) were recovered by flotation using 60% sucrose, collected by aspiration onto nylon filters, and surface-sterilized. The viable Sclerotia were enumerated by counting colonies growing on a selective medium. Efficiency of recovery was 95–97% of Sclerotia added to soil. Large numbers of samples could be processed rapidly by this method which utilized readily available materials, and gave higher counts than earlier methods, which involved only sieving or flotation.  相似文献
