Seeds and plants of wild type Lupinus albus are bitter and contain high level of alkaloids. During domestication, at least three genes conferring low-alkaloid content were identified and incorporated into commercial varieties. Australian lupin breeders exclusively utilize one of these sweetness genes, “pauper”, in all varieties to prevent possible bitterness contamination via out-crossing. A cross was made between a sweet variety Kiev Mutant (containing pauper gene) and a bitter type landrace P27174, and the population was advanced into F8 recombinant inbred lines (RILs). Twenty-four plants representing sweetness and bitterness were subjected to DNA fingerprinting by the microsatellite-anchored fragment length polymorphism (MFLP) technique. A dominant polymorphism was discovered in an MFLP fingerprint. The MFLP marker was converted into a co-dominant, sequence-specific, simple PCR-based marker. Linkage analysis by the software program MapManager with marker score data and alkaloid phenotyping data from a segregating population containing 190 F8 RILs indicated that the marker is linked to the pauper gene at the genetic distance of 1.4 centiMorgans (cM). This marker, which is designated as “PauperM1”, is capable of distinguishing the pauper gene from the other two low-alkaloid genes exiguus and nutricius. Validation on germplasm from the Australian lupin breeding program showed that the banding pattern of the marker PauperM1 is consistent with the alkaloid genotyping on a wide range of domesticated varieties and breeding lines. The PauperM1 marker is now being implemented for marker assisted selection in the Australian albus lupin breeding program.  相似文献

Twenty compounds, including three sterols (13), three phenols (4, 14 and 15), four anthraquinones (5, 7, 8 and 16), one chromone (6), two stilbenes (9 and 10), three amides (1113), three flavonoids (1719) and one organic acid (20), were obtained by modern phytochemical isolation methods. Their structures were identified by spectroscopic methods and in comparison with the published data in the references. Among them, compound 2, 3, 11 and 13 were firstly discovered from genus Fallopia, and compounds 1, 58, 10, 14, 15, 17, 19 and 20 were obtained from F. convolvulus for the first time. The chemotaxonomic significance of these compounds was also discussed, which revealed the relationships between F. convolvulus and some other species of Polygonaceae family.  相似文献

Summary The nature and extent of heterosis for grain yield and its component characters was estimated in a seven-parent F1-diallel cross in rice under the cold stress conditions of Palampur, representing sub-temperate climate of the mid hills of Himachal Pradesh. The highest heterotic effects for grain yield were observed in Himalaya 1/Phul Patas 72 and China 988/Himdhan hybrids, which gave 60.36 and 32.48% heterosis, respectively, over mid parent, 38.76 and 26.86% heterosis, respectively, over high parent and 32.30 and 26.86% heterosis, respectively, over the best variety, Himdhan. Heterosis for grain yield in these crosses was due to an increase in tiller number, panicle length, spikelets/panicle and 1,000-grain weight. Large number of crosses exhibited significant heterosis for high spikelet sterility and the majority of them had significant heterosis for late flowering, taking more than 120 days to flower, which resulted in lack of heterosis for grain yield in such crosses due to cold stress at the reproductive stage.Part of the thesis submitted by the senior author to Himachal Pradesh Agricultural University, Palampur (H.P.) To increase the use efficiency of potassium (K) fertilizer, special attention was paid to the dynamics of soil K in the root zone and non-root zone. Difference in K dynamics between yellowish red soil and yellow cinnamon soil under rapeseed (Brassica napus L.)rice (Oryza sativa L.) rotation was studied using a rhizobox system. Results showed that soil water soluble K (Sol-K) and exchangeable K (Ex-K) in the root zone of both soils were reduced in the early stage of rapeseed growth. Along with plant growth and K uptake, soil Sol-K in the inner (0–20 mm to root zone), middle (20–40 mm) and outer (40–60 mm) compartments of the non-root zone of yellowish red soil migrated towards the root zone. As a result, soil Ex-K was transformed into Sol-K. The changes in soil Sol-K and Ex-K in the non-root zone of yellow cinnamon soil were similar to yellowish red soil, and soil non-exchangeable K (Nonex-K) in the root zone also decreased significantly. In the early stage of rice growth, waterlogging promoted diffusion of soil Sol-K from non-root zone to root zone and transformation of Ex-K into Sol-K. Along with the growth of rice and K uptake, soil Ex-K in each compartment of yellowish red soil decreased significantly. Soil Sol-K and Ex-K in the yellow cinnamon soil declined to a certain extent, and then remained unchanged, while soil Nonex-K kept on decreasing. It revealed that the plants first absorbed K in the root zone, of which K reserve was replenished by a gradual diffusion of K from the non-root zone. The closer to the root zone, the greater the contribution to K uptake by plants. Within one rotation cycle, Ex-K and Sol-K in yellowish red soil were the main forms of K available to the plants, and little Nonex-K could be absorbed. However, in the yellow cinnamon soil, Nonex-K was the main form of K available to the plants, followed by Ex-K and Sol-K.  相似文献

The effects of Fe concentrations in the pretreatment solution on the induction of plaque and the differences between genotypes on arsenate uptake by and translocation within rice seedlings grown in nutrient solution in the greenhouse were investigated. After iron plaque on rice roots was induced in solutions containing 20, 40, 60, 80, and 100 mg Fe2+ l(-1), seedlings were transplanted into nutrient solution with 0.5 mg As l(-1). The formation of iron plaque was clearly visible as a reddish coating on the root surface after 12 h induction. Fe2+ concentrations in the pretreatment solution and 0.5 mg As l(-1) in the treatment solutions did not significantly affect rice growth. There was a significant correlation between the concentrations of Fe and As in iron plaque on the root surface for the three genotypes. About 75-89% of total As was concentrated in iron plaque (DCB-extracts). There were no significant differences in As concentrations in the roots between the three genotypes; however, As concentrations in shoots differed significantly between them. Arsenic concentrations in shoots were positively correlated with iron concentrations in the shoots. The results suggest that iron plaque may act as a 'buffer' for As in the rhizosphere.  相似文献

In theory, selection for effective camouflage (i.e. dull coloration) in fish should be strongest when the conditions for visual predation are most favourable, such as in structurally simple pelagic habitats. By contrast, in more sheltered (e.g. littoral) habitats, selection may favour effective intra‐specific communication (i.e. bright coloration) (at the expense of crypsis). Poor transparency, as in highly humic waters, should constrain colour adaptations. We investigated phenotypic variation in body coloration of Eurasian perch (Perca fluviatilis L.) in littoral and pelagic habitats of four humic boreal lakes. Perch from the most transparent lake had the lightest and less coloured belly and perch were more colourful in the littoral habitats than in the pelagic areas, with the pattern being clearest in the most transparent lake. In addition, perch in the most transparent lake exhibited sexual dichromatism, with males having a more colourful belly than the females, whereas no indications of sexual dichromatism were found in more humic lakes. Moreover, in the most transparent lake, the condition of fish correlated with bright belly coloration in the littoral, but with dull belly coloration in the pelagic habitat. The results obtained in the present study suggest that selection on perch coloration may differ between lakes as a result of visual properties of the water, and within lakes as a result of divergent selection for camouflage and communication in pelagic and littoral habitats. © 2010 The Linnean Society of London, Biological Journal of the Linnean Society, 2010, 99 , 47–59.  相似文献

Summary Seedlings of Scots pine (Pinus sylvestris L.) from Russia (59°58N) and Poland (53°34N) were grown for 4 months in controlled environment chambers, simulating the photoperiod conditions of 50° and 60° N. The Russian population grown at 50° N showed earlier height growth cessation than the Polish population. Photoperiodic conditions of 60° N increased proportional allocation of dry mass to shoots and lowered allocation to roots in the Russian population, which also had greater allocation to roots than the Polish population in both treatments. Total non-structural carbohydrate concentrations in roots and secondary needles of both populations were significantly higher at the end of the 4 month growing season at 50° compared to 60° N. Net photosynthesis rates were similar for both provenances and both treatments. The rate of transpiration was higher and water-use efficiency lower for plants grown in long-day conditions of 60° N. The mean respiration rate of roots ranged between 30 and 36 nmol CO2 · g-1 dry mass · s-1 and was 2–4 times higher than values observed for needles. Root respiration rates were greater in the Polish than the Russian population. Despite this, the greater allocation to root dry mass of the Russian population resulted in greater root respiratory cost as a proportion of daily carbon gain. Overall, root respiration accounted for between 18 to 34% of the total daily net carbon assimilation of these populations. Root and total respiration as a proportion of net daily carbon assimilation were greater at 50° than 60°N. Mean net integrated CO2 gains were 2.2–2.5 mmol CO2 · day-1 for seedlings from Russia compared to 3 mmol CO2 · day-1 for Poland.  相似文献

 Transgenic white poplar plants (Populus alba L.) expressing the nptII gene and the bar gene from Streptomyces hygroscopicus have been produced using Agrobacterium tumefaciens-mediated gene transfer. Eleven kanamycin-resistant plant lines were obtained with a transformation frequency of 7%. Successful genetic transformation was confirmed by Southern and northern analyses. The level of resistance to the commercial preparation of phosphinothricin (Basta; Roussel-Hoechst Agrovet) was evaluated by in vitro and in vivo assays. Using in vitro selective conditions for phosphinothricin, only plantlets from four kanamycin-resistant independent lines remained green and continued to grow and root. After transfer to the growth chamber, all selected transgenic lines were shown to be completely resistant to the herbicide Basta with doses equivalent to 6 l ha–1 (normal field dosage) and were tolerant at concentration of 12 l ha–1. This is the first report describing the genetic transformation of a P. alba clonal cultivar of commercial interest with a gene of agronomic value. Norway maple (Acer platanoides) is a Eurasian introduced tree species which has invaded the North American range of its native congener, sugar maple (A. saccharum). One hypothesis used to explain the success of an invasive species is the enemy release hypothesis (ERH), which states that invasive species are often particularly successful in their new range because they lack the enemies of their native range. In this study, we hypothesized that Norway maple would have less insect damage than sugar maple due to such enemy release. Autumn 2005 and summer 2006 leaves of Norway and sugar maple were collected from six sites in New Jersey and Pennsylvania to compare percent leaf area loss, gall damage, fungal damage, and specific leaf area (cm2/g). Although both species had low overall mean levels of leaf damage (0.4–2.5%), in both years/seasons Norway maple had significantly less leaf damage than sugar maple. Insects were also collected to compare insect assemblies present on each tree species. The numbers of insect taxa and individuals found on each species were nearly equivalent. Overall, the results of this study are consistent with the enemy release hypothesis for Norway maple. In addition, sugar maples when surrounded by Norway maples tended to show reduced herbivory. This suggests that the spread of Norway maple in North America, by reducing amounts of insect herbivory, may have further ecosystem-wide impacts.  相似文献

Lupinus albus L. were grown in rhizoboxes containing a soil amended with sparingly available Fe–P or Al–P (100 μg P g−1 soil/resin mixture). Root halves of individual plants were supplied with nutrient solution (minus P) buffered at either pH 5.5 or 7.5, to assess whether the source of mineral-bound P and/or pH influence cluster-root growth and carboxylate exudation. The P-amended soil was mixed 3:1 (w/w) with anion-exchange resins to allow rapid fixation of carboxylates. Treatments lasted 10 weeks. Forty percent and 30% of the root mass developed as cluster roots in plants grown on Fe–P and Al–P respectively, but cluster-root growth was the same on root-halves grown at pH 5.5 or 7.5. Mineral-bound P source (Al– or Fe–P) had no influence on the types of carboxylates measured in soil associated with cluster roots—citrate (and trace amounts of malate and fumarate) was the only major carboxylate detected. The [citrate] in the rhizosphere of cluster roots decreased with increased shoot P status (suggesting a systemic effect) and also, only for plants grown on Al–P, with decreased pH in the root environment (suggesting a local effect). In a separate experiment using anion exchange resins pre-loaded with malate or citrate, we measured malate (50%) and citrate (79%) recovery after 30 days in soil. We therefore, also conclude that measurements of [citrate] and [malate] at the root surface may be underestimated and would be greater than the 40- and 1.6-μmol g−1 root DM, respectively estimated by us and others because of decomposition of carboxylates around roots prior to sampling.  相似文献

Leaf development of Quercus robur (deciduous), Q. ilex (evergreen) and of their hybrid Q. × turneri was assessed under Central European climate conditions. In all three taxa, development of maximum photosynthetic capacity was slow, although whole chain electron transport, grana development, the xanthophyll cycle and the biochemical capacity for photosynthesis were already completely present at day 7 after budbreak (DAB 7). In the course of the following 30 days, shifts in the levels of metabolites of the photosynthetic dark reactions were observed, indicating a change from ribulose bisphosphate (RubP) regeneration limitation towards RubP consumption limitation of photosynthesis. At the same time, electron transport rates had strongly increased and the capacity of the light reactions did not seem to limit photosynthesis. Sucrose levels in the leaves increased strongly, indicating sink limitation of photosynthesis, which might be responsible for the observed slow development of maximum photosynthetic capacity in all three taxa.  相似文献

Summary The physiological activity and C and N allocation of beech seedlings planted in two different beech forest soils (acid brown earth, rendzina) were studied over 80 days. One adult earthworm Octolasion lacteum (Örley) was placed in half of the containers. Transpiration, CO2, assimilation, chlorophyll synthesis and N incorporation were higher in the acid brown earth than in the rendzina. Fine root production was also higher in the acid brown earth. Production of stems and total N incorporation were higher in the O. lacteum treatments than in the controls. Burrowing activity of O. lacteum did not affect total root production but stimulated the transfer of C and N towards aboveground parts of the plants and shifted the large root to fine root ratio towards large roots. O. lacteum effects were similar in both soils. It is concluded that endogeic earthworms significantly affect resource allocation patterns of beech seedlings over a wide range of soils. This effect can be partly explained by an increased availability of N in soils worked by earth-worms.  相似文献

The emission of reduced volatile sulfur compounds from twigs of Norway spruce (Picea abies (L.) Karst.) was measured in the field by cryosampling and gaschromatographic analysis. Trees were growing in the Erzgebirge (E-Germany) at Oberbärenburg and at the Kahleberg and at a third stand in NW-Bavaria (S-Germany). Emission rates were also measured for Scotch pine (Pinus sylvestris L.) and Blue spruce (Picea pungens Engelm.) at the Kahleberg. Twigs still attached to the trees were enclosed in a flow-through gas exchange cuvette. H2S was detected as the predominant reduced sulfur compound emitted from the twigs. The mean H2S emission rate from twigs of Norway spruce varied between 0.04 pmol kg-1 dw s-1 at Würzburg and 6.21 pmol kg-1 dw s-1 at the Kahleberg. Comparing different species at the Kahleberg, the mean H2S emission rate was almost the same from twigs of Norway spruce (6.2 pmol kg-1 dw s-1) and Blue Spruce trees (5.9 pmol kg-1 dw s-1) but it was approximately 18 times higher for Scotch pine (110 pmol kg-1 dw s-1). The percentage of SO2-exclusion via H2S-emission of the tree species investigated at the Kahleberg is calculated on the basis of data on SO2 fluxes. It is very small for Norway spruce and Blue spruce. However, for Scotch pine, H2S emission contributes about 10% to the detoxification of SO2.  相似文献

The carrier-mediated transport of GABA in rat brain synaptosomes was strongly and permanently inhibited byl-2,4-diaminobutyric acid (DAB). In order to discriminate between carrier-mediated and non-carrier-mediated release of [3H]GABA, synaptosomes prelabeled with 0.5 M [3H]GABA in the presence of 100 M DAB, or with 0.2 M [3H]GABA without DAB, were superfused in conditions stimulating the release of [3H]GABA. Only the release elicited by unlabeled GABA or DAB (by homo- and heteroexchange, respectively) was strongly inhibited in DAB-pretreated synaptosomes. The spontaneous release and the release induced by 56 mM KCl in the presence of CaCl2, by the ionophore A23187, by ouabain, by lack of K+, or by purified black widow spider toxin were unaffected or only barely decreased in DAB-treated synaptosomes, and therefore do not seem to be mediated by the DAB-blocked GABA carrier.  相似文献

The nucleotide sequence and derived amino acid sequence of two different -glucosidase cDNA clones were determined. One clone (TRE104) was identified as the cyanogenic -glucosidase by homology with the N-terminal and internal peptide amino acid sequence of the purified enzyme. The biological function of the other -glycosidase (TRE361) is not known. Co-segregation of genomic restriction fragments uniquely identified by each cDNA clone shows that these two genes are linked in the white clover genome. Both TRE104 and TRE361 fragments co-segregate with cyanogenic -glucosidase activity. Extensive homology was found between the white clover -glucosidase sequences and a group of prokaryote and mammalian -glycosidases. This group of sequences has no homology with a separate set of -glucosidase genes isolated from fungi and the thermophilic bacterium Clostridium thermocellum.  相似文献

Hypotheses and models to explain female ornaments often assume that the elaborated traits are condition dependent; nevertheless, few empirical studies have addressed this topic. We studied a population of three-spined sticklebacks (Gasterosteus aculeatus) in which the females have conspicuous, carotenoid-based red coloration to their pelvic spines. The red coloration seems not to be condition dependent, as coloration is negatively associated with age and body length and not associated with condition. Furthermore, redder females did not have a lower density of leucocytes. We found a negative association between the females' red carotenoid-based coloration in the spines and the amount of carotenoids in the female's gonads. Males choosing red-coloured females will fertilize eggs with small amounts of carotenoids and appear not to gain any benefit from their mates' phenotypic quality that could result in offspring of improved quality. These results do not support the 'direct selection hypothesis' to explain the existence of the female ornaments.  相似文献

The effect of salt stress, under glasshouse conditions, was studied on plant biomass, nodulation, and activities of acid phosphatases (APase, EC 3.1.3.2) and trehalose 6-phosphate phosphatase (TPP, EC 3.1.3.12) in the symbiosis common bean (Phaseolus vulgaris L.)-rhizobia nodules. Four common bean recombinant inbred lines (147, 115, 104 and 83) were separately inoculated, with CIAT 899 or RhM11 strains and grown in hydroaeroponic culture. Two NaCl levels (0 and 25 mM NaCl plant?1 week?1 corresponding, respectively, to the control and the salt treatment) were applied and the culture was assessed during 42 days after their transplantation. The results showed that the nodulation of these lines was not affected by salinity except for the line 83 inoculated with CIAT 899, whose nodule dry weight decreased by 48.24 % compared with the corresponding controls. For the other symbiotic combinations, shoot and root biomasses were not significantly affected by salt constraint. Salinity stress generally reduced acid phosphatise and trehalose phosphate phosphatase activities in nodules that were less affected in plants inoculated with RhM11. Based on our data, it appears that nodule phosphatase activity may be involved in salinity tolerance in common beans and the levels of salt tolerance depend principally on specific combination of the rhizobial strain and the host cultivar.  相似文献
