In Japan, although greenhouse whitefly, Trialeurodes vaporariorum (Westwood), and sweet potato whitefly, Bemisia tabaci (Gennadius), co-occur on tomato plants under greenhouse conditions, the two whiteflies are distributed differently with regard to leaf position. To elucidate the factors that determine the leaf position of these whiteflies, we investigated traits for leaflets collected from three positions on tomato plants. Furthermore, we examined leaflet selection by and fertility of the two whiteflies under choice and non-choice conditions. In addition, the effect on whitefly behavior of volatile compounds released from leaflets was evaluated by use of a Y-tube olfactometer test. Nitrogen and carbon content were highest for upper leaflets. In choice tests, more T. vaporariorum and B. tabaci adults selected upper and middle leaflets, respectively. Similarly, they oviposited more eggs on upper and middle leaflets. In non-choice tests, T. vaporariorum oviposited more eggs on upper leaflets, but B. tabaci oviposited equally on each leaflet. In Y-tube olfactometer tests, more T. vaporariorum adults moved to upper leaflets whereas more B. tabaci adults moved to middle leaflets. These results suggest that different leaflet selection by adults of these two whiteflies is likely to be associated with the different volatile compounds emitted by tomato leaflets at each position.  相似文献

Development and sporogenesis of Colletotrichum gloeosporioides on castor leaf differed from that on other known host plants. C. gloeosporioides had three kinds of hyphae on castor leaf: primary infection hyphae (PIH), runner hyphae (RH) and secondary infection hyphae (SIH). The PIH originated from conidia, grew on leaf surface and entered the leaf by direct penetration of the cuticle without forming appressoria. The RH were sub-cuticular hyphae, the track of which was traceable by the bulgings on the leaf surface, and the SIH were the hyphae that emerged to leaf surface from RH through the cuticle or stomata. Conidia were initiated as small protrusions along the lengths of RH and SIH that got differentiated into distinct conidia, each born on a short stumpy conidiophore without forming any congregation. The protrusions from RH emerged to the leaf surface by piercing the cuticle, and they developed into distinct conidia on the leaf surface. The conidia developed from RH and SIH were identical in size and shape. Even though conidia were occasionally found emerged through stomata, that appeared to be random than a preferred route for the discharge of conidia. The penetration and sporogenesis of C. gloeosporioides on castor leaf differed from that reported on mulberry leaf.  相似文献

Abstract

The pre-penetration and infection process of Colletotrichum dematium on mulberry leaf was investigated by scanning electron microscope. Conidia produced on germination appressoria directly or at the end of short germ tubes. Appressoria were formed mostly over cuticle, but sometimes over stomata also. At 72 h post-inoculation, an extensive network of sub-cuticular runner hyphae (RH) was produced. The RH were traceable by the cuticular bulgings on leaf surface. The RH emerged to leaf surface through ruptured cuticle to form secondary infection hyphae (SIH). The SIH re-entered the leaf tissue by sending penetration branches through stomata. Conidia were formed singly on short conidiophores from the RH and SIH, at short intervals. The conidia developed on RH were exposed to leaf surface through ruptured cuticle. Some times conidia were released through stomata also. The RH and SIH had thick knots from which hyphal branches and conidia were developed. Definite acervuli were not developed.  相似文献

干热河谷地区水电站建设对当地植被的潜在影响是一个值得关注的生态学问题。车桑子是当地植被灌木层的主要成分,开展水库气候效应对车桑子生长、发育影响的研究具有现实价值。以车桑子的实生幼苗为材料,将土壤含水量控制为13%、7%和1.5%,空气湿度控制为50%、65%和75%,从幼苗生长、构件发育、根系发育、生物量分配等方面研究了降水减少和大气湿度增加的气候效应对车桑子的影响,通过叶绿素含量、Fv/Fm、丙二醛(MDA)含量和叶片可溶性糖含量等指标,从光合系统特性、膜质过氧化和渗透调节3个方面研究了车桑子的受损与适应机制。结果表明,土壤干旱能够抑制车桑子幼苗高生长和根系发育的各项指标,并促进生物量向根系分配;当大气湿度增加时,幼苗高生长和根长虽呈增加趋势,但生物量积累、根系发育指标及RMR却具有单峰效应,显示空气湿度过高时对其生长发育具有抑制作用。综合而言,由于大气湿度增加能够部分补偿土壤的干旱效应,干热河谷区水库建设的气候效应不会对车桑子幼苗的生长和发育产生重要影响。结果还表明,土壤干旱和大气湿度变化对叶绿素含量无影响,土壤水分胁迫和空气湿度下降导致Fv/Fm显著下降,说明光合电子传递链受损是车桑子光合抑制的主要原因;土壤水分胁迫导致MDA含量升高,说明细胞膜质过氧化是车桑子幼苗受损的重要机制;而土壤干旱导致叶片可溶性糖含量升高,说明车桑子幼苗具有较好的渗透调节机制。研究结果对评估干热河谷区水电站建设对植被的影响具有参考价值。  相似文献

Abscisic acid (ABA) is major plant hormone involved in regulating abiotic stress responses. Several studies have established that an ABA‐signalling transduction pathway—from ABA perception to response—functions in plant cells. The group A PP2Cs constitute core components of ABA signalling, and they negatively regulate ABA signalling and stress responses. Recent studies have identified and functionally analysed regulators of PP2C activity; however, the precise regulatory mechanisms remain unclear. In the present study, we used a yeast 2‐hybrid (Y2H) screening analysis to identify the DEAD‐box RNA helicase RH8, which interacted with PP2CA in the nucleus. rh8 knockout mutants exhibited ABA hyposensitivity and drought‐susceptible phenotypes characterized by high levels of transpirational water loss via reduced stomatal closure and decreased leaf temperatures. However, rh8/pp2ca double mutants showed ABA hypersensitivity and drought‐tolerant phenotypes, indicating that RH8 and PP2CA function in the same ABA‐signalling pathway in the drought stress response; moreover, RH8 functions upstream of PP2CA. In vitro phosphatase and kinase assays revealed that RH8 inhibits PP2CA phosphatase activity. Our data indicate that RH8 and its interacting partner PP2CA modulate the drought stress response via ABA‐dependent signalling.  相似文献

(1) Most ferns are restricted to moist and shady habitats, but it is not known whether soil moisture or atmospheric water status are decisive limiting factors, or if both are equally important. (2) Using the rare temperate woodland fern Polystichum braunii, we conducted a three‐factorial climate chamber experiment (soil moisture (SM) × air humidity (RH) × air temperature (T)) to test the hypotheses that: (i) atmospheric water status (RH) exerts a similarly large influence on the fern's biology as soil moisture, and (ii) both a reduction in RH and an increase in air temperature reduce vigour and growth. (3) Nine of 11 morphological, physiological and growth‐related traits were significantly influenced by an increase in RH from 65% to 95%, leading to higher leaf conductance, increased above‐ and belowground productivity, higher fertility, more epidermal trichomes and fewer leaf deformities under high air humidity. In contrast, soil moisture variation (from 66% to 70% in the moist to ca. 42% in the dry treatment) influenced only one trait (specific leaf area), and temperature variation (15 °C versus 19 °C during daytime) only three traits (leaf conductance, root/shoot ratio, specific leaf area); RH was the only factor affecting productivity. (4) This study is the first experimental proof for a soil moisture‐independent air humidity effect on the growth of terrestrial woodland ferns. P. braunii appears to be an air humidity hygrophyte that, whithin the range of realistic environmental conditions set in this study, suffers more from a reduction in RH than in soil moisture. A climate warming‐related increase in summer temperatures, however, seems not to directly threaten this endangered species.  相似文献

The behavioural response of the predatory mite Phytoseiulus persimilis to volatiles from several host plants of its prey, spider mites in the genus Tetranychus, was investigated in a Y-tube olfactometer. A positive response to volatiles from tomato leaves and Lima bean leaves was recorded, whereas no response was observed to volatiles from cucumber leaves, or leaves of Solanum luteum and Solanum dulcamara.Different results were obtained for predators that differed in rearing history. Predators that were reared on spider mites (Tetranychus urticae) on Lima bean leaves did respond to volatiles from Lima bean leaves, while predators that had been reared on the same spider mite species but with cucumber as host plant did not respond to Lima bean leaf volatiles. This effect is compared with the effect of rearing history on the response of P. persimilis to volatile allelochemicals of prey-infested plant leaves.  相似文献

The effectiveness of several leaf water models (‘string‐of‐lakes’, ‘desert river’ and the Farquhar–Gan model) are evaluated in predicting the enrichment of leaf water along a maize leaf at different humidities. Progressive enrichment of both vein xylem water and leaf water was observed along the blade. At the tip, the maximum observed enrichment for the vein water was 17.6‰ at 50% relative humidity (RH) whereas that for the leaf water was 50‰ at 34% RH and 19‰ at 75% RH. The observed leaf water maximum was a fraction (0.5–0.6) of the theoretically possible maximum. The ‘string‐of‐lakes’ and ‘desert river’ models predict well the variation of leaf water enrichment pattern with humidity but overestimate the average enrichment of bulk leaf water. However, the Farquhar–Gan model gives good prediction for these two aspects of leaf water enrichment. Using the anatomical dimensions of vein xylem overestimates the effective longitudinal Péclet number (Pl). Possible explanations for this discrepancy between the effective and the xylem‐based estimate of Pl are discussed. The need to characterize the heterogeneity of transpiration rate over the leaf surface in studies of leaf water enrichment is emphasized. The possibility that past atmospheric humidity can be predicted from the slope of the Δ18O spatial variation of leaf macrofossils found in middens is proposed.  相似文献

To control coconut leaf beetle, Brontispa longissima (Gestro), the pupal parasitoid Tetrastichus brontispae Ferrière was imported from Taiwan and its biology was studied in quarantine in Hainan, China. The parasitoid development includes an egg, three larval instars and three pupal stages. Its developmental time from egg to adult was 19.5±0.5 days under conditions of 24±2°C and 75±5% relative humidity (RH). Temperature had no effect on the sex ratio of offspring, but significantly affected the parasitism rate and reproduction. The parasitism rates were 98.07, 97.97 and 95.03% at 28, 24 and 20°C, respectively, whereas the parasitism rate was 52.18% at 18°C and 69.48% at 30°C, respectively. Furthermore, the parasitoids reared at 18 and 30°C produced fewer offspring than those at 20, 24 and 28°C, respectively. With the increase in temperature, developmental time decreased linearly from 46.19 days at 18°C to 17.10 days at 28°C. RH significantly influenced development, parasitism rate and the reproduction of T. brontispa. With the decrease of RH, developmental time increased from 22.94 days at 20% RH to 18.84 days at 95% RH. In contrast, parasitism rate and the number of offspring per female increased with the increase of RH. Though emergence rates between 50 and 95% RH were much higher than those between 20 and 35% RH, the sex ratios between 20 and 95% RH were not different. Photoperiod had no effect on parasitism, the number of offspring per female, emergence and the sex ratio of T. brontispae, but developmental time was significantly different for different photoperiods. Sucrose, honey and glucose significantly enhanced adult longevity, parasitism and the number of offspring per female of T. brontispae, but had no effect on the sex ratio and survival. Females of T. brontispae only parasitized fourth to fifth larval instars and 1–5-day-old pupae, but there was a significant difference in the number of offspring per female, development time, emergence and the sex ratio of offspring in different instars. These results showed that 1-day-old pupae, a temperature of 24–28°C and 65–95% RH were optimal for T. brontispae. These findings should be helpful in developing a production system to rear and release T. brontispae in large enough quantities to effectively control coconut leaf beetle.  相似文献

Humidity effects on leaf expansion in sugar beets (Beta vulgaris L.) were explored using linear variable differential transducers. In continuous light, an increase in relative humidity (RH) from 35 to 61 or 75% resulted in a rapid increase in leaf extension which was maintained for 10–15 min before slowing down. Increasing RH from 35 to 85% increased leaf-extension rate (LER) in light and in dark and substantially diminished the ratio of dark LER to light LER, showing that high humidity can offset the reduction in LER which occurs on illumination. Episodes of irradiance with visible or infrared radiation resulted in diminished LER, indicating that increases in transpiration may reduce the flux of water available for leaf cell expansion. We studied whether volatiles released by putative host plants affect the antipredator response of an herbivorous mite, Tetranychus urticae, when the patch was invaded by Phytoseiulus persimilis. Tetranychus urticae laid a lower number of eggs on tomato leaves than on lima bean leaves, suggesting that lima bean is a preferred host food source for T. urticae. In addition, T. urticae preferred lima bean plant volatiles to tomato plant volatiles in a Y-tube olfactometer test. To investigate the antipredator response of T. urticae, we examined the migration of T. urticae from a lima bean leaf disc to a neighbouring leaf disc (either a tomato or lima bean leaf disc) when ten predators were introduced into the original lima bean disc. A Parafilm bridge allowed for migration between the leaf discs. No migrations occurred between leaf discs when there were no predators introduced to the original leaf disc. However, when predators were introduced migrations did occur. When the neighbouring leaf disc was upwind of the original disc, the migration rate of the mite from original lima bean leaf disc to a neighbouring tomato leaf disc was significantly lower than that to a neighbouring lima bean leaf disc. By contrast, when the neighbouring leaf disc was downwind of the original leaf disc, there was no difference in the migration rates between lima bean leaf discs and tomato leaf discs. The number of T. urticae killed by P. persimilis for each treatment was not different, and this clearly shows that the danger was the same in all treatments regardless of the decision made by T. urticae. From these results, we conclude that T. urticae change their antipredator response by evaluating the difference in host plant volatiles in the patch they inhabit.  相似文献

Aims: To investigate the effect of glycine betaine (GB) on the survival of Listeria monocytogenes on leaf surfaces under low relative humidity (RH). Methods and Results: The addition of GB (≥25 mmol l?1) improved the survival of L. monocytogenes under low RH on parsley leaves, thus suggesting that GB can improve the tolerance of L. monocytogenes to desiccation. Ten times less GB was needed to improve L. monocytogenes survival under low RH on nonbiological surfaces compared with parsley leaves, suggesting that, on the leaf surface, L. monocytogenes may have to compete for the available GB with autochthonous bacteria and/or the plant itself. Wild type and mutants carrying deletions in the three GB uptake systems, BetL, Gbu and OpuC, behaved similarly with and without added GB on parsley leaves (P > 0·05). In addition, preaccumulation of GB, triggered by osmotic stress prior to inoculation, failed to improve survival under low RH compared with osmotic stress without GB accumulation. Conclusions: Exogenous GB had a protective effect on L. monocytogenes cells from desiccation during survival on parsley leaves. This effect was independent of intracellular GB accumulation by the known uptake systems. Significance and Impact of the Study: Presence of GB could improve the survival of L. monocytogenes to desiccation on leaf surfaces and nonbiological surfaces.  相似文献

Behavioral responses of Frankliniella occidentalis Pergande (Thysanoptera: Thripidae) to visual and olfactory cues were assessed in a cylindrical black box and a Y-tube olfactometer. Frankliniella occidentalis preferred circular shapes to other five geometrical patterns (rectangle, triangle, inverted triangle, diamond, and modified circle imitating flower petals) in multiple choice tests. In pair wise choice tests, the thrips preferred the yellow artificial flower shape to the geometrical patterns tested. Frankliniella occidentalis stayed on the artificial flower about four times longer than on the geometrical patterns. Higher numbers of thrips responded to a combination of p-anisaldehyde and artificial flower compared to the arm with only the olfactory or the visual cue. These results suggest that shape is an important cue for F. occidentalis.  相似文献

The host-searching behaviour of Heterorhabditis megidis strain NLH-E 87.3 in the presence of insect hosts and plant roots, offered individually and in combination, was studied using a newly developed Y-tube olfactometer filled with sand. Within a period of 24 hours infective juveniles (IJs) were significantly attracted to living G. mellonella larvae and caused 100% larval mortality. Otiorhynchus sulcatus larvae, however, did not elicit host-oriented movement of IJs and no larval mortality was observed. Roots of strawberry plants induced a negative response in IJs. The combination of strawberry roots and O. sulcatus larvae, however, strongly attracted IJs leading to 37% host mortality. It was shown that this type of Y-tube choice arena is a useful tool in studying the searching behaviour of entomopathogenic nematodes in a semi-natural habitat.  相似文献

We investigated the ability of entomopathogenic nematodes to infect diamondback moth (DBM),Plutella xylostella (L.) (Lepidoptera: Plutellidae) on a leaf surface. In a leaf disk assay, mortality of late stage DBM larvae ranged from <7% caused bySteinernema kushidai Mamiya to >95% caused byS. carpocapsae (Weiser) All strain. LC50 values forS. carpocapsae, S. riobravis Cabanillas, Poinar & Raulston, andHeterorhabditis bacteriophora Poinar NC1 strain were 14.6, 15.4, and 65.4 nematodes/larva, respectively.S. carpocapsae, S. riobravis, andH. bacteriophora caused 29%, 33%, and 14% mortality of DBM pupae, respectively. DBM mortality caused byS. carpocapsae on radish declined at low (<76%) to moderate (76–90%) RH, because nematode survival and infectivity declined at low (<76%) to moderate (76–90%) RH. However, DBM mortality caused byS. riobravis did not decline with RH.S. riobravis survival declined with RH, but infectivity did not. Overall, nematode survival and infectivity to DBM larvae were lower forS. riobravis than forS. carpocapsae. In addition, DBM mortality was higher on radish plants (pubescent leaves) than on cabbage plants (glaborous leaves).  相似文献

Abstract The leaf surface of a plant, especially its chemical components, constitutes the first line of resistance to herbivores and other pests. Our previous research indicated that ‘Valmaine’ (Val) romaine lettuce, Lactuca sativa L., was highly resistant to feeding by adult banded cucumber beetle, Diabrotica balteata LeConte, while ‘Tall Guzmaine’ (TG) was highly susceptible. We investigated the leaf surface chemistry of these two cultivars for its possible role in their resistance to D. balteata. Three solvents with different polarity (hexane, methylene chloride, and methanol) were tested to remove leaf surface chemicals, but only methylene chloride and methanol extracts were used in feeding bioassays. Adult D. balteata consumed much more of the leaf tissue of Val and TG when their surface chemicals were removed with methylene chloride, but not methanol, compared to nonextracted leaf tissue, leading us to hypothesize that methylene‐chloride extractable leaf surface chemicals may have a role in the expression of lettuce resistance. However, leaf surface chemicals extracted from Val with methylene chloride were not a deterrent to adult D. balteata when applied to palatable lima bean leaf surfaces at various concentrations in dual‐choice tests. Furthermore, the application of surface extracts from TG did not stimulate beetle feeding in similar choice tests. In a no‐choice feeding test, there was no significant difference in leaf area consumption on lima bean leaves sprayed with extracts of Val or TG. These results suggest that leaf surface chemicals in romaine lettuce do not explain the resistance of Val to adult D. balteata, and that factors inside the leaf may play a role in resistance. We discuss the possibility that the solvent may have increased the palatability of lettuce leaves to D. balteata by causing enzymatic browning and cellular damage, which is likely to have degraded internal feeding deterrents and impaired the plant's ability to emit latex.  相似文献

We studied the response of the predatory mite Amblyseius womersleyi collected in 13 different sites in Japan toward Tetranychus urticae-infested kidney bean leaf volatiles in a Y-tube olfactometer. The predatory mites were collected from eight plant species infested by one of three tetranychid mite species. The predators' responses to the infested-leaf volatiles varied from 33% to 97% among the populations. The predators collected at 10 sites showed a significant preference for infested-leaf volatiles, whereas those collected at three tea plantations did not distinguish between the infested- and uninfested-leaf volatiles. We discussed the possible factors that affected the olfactory response of A. womersleyi towards the infested leaf volatiles.  相似文献

The predator Orius sauteri (Poppius) (Heteroptera: Anthocoridae) is an effective arthropod natural enemy of thrips, especially Thrips palmi Karny (Thysanoptera: Thripidae), a serious pest of vegetables. First, we studied O. sauteri's response to the odour from thrips-infested eggplant [Solamum melongena L. (Solanaceae)] in a laboratory by two-choice experiments using a Y-tube olfactometer. When detached eggplant leaves were used as odour sources, O. sauteri preferred the volatiles from uninfested leaves to clean air. Concerning preferences among differently infested leaves, O. sauteri preferred the volatiles from plants infested with 10–100 thrips per leaf to uninfested leaves, but showed no significant preference for artificially damaged leaves over uninfested leaves. Similar results were obtained when complete plants were tested as the odour source. Second, release and recapture experiments in a greenhouse, a more realistic set of conditions, were conducted to confirm whether a significant preference for infested plants occurred at similar infestation levels as in the laboratory. Trends favouring infested plants were detected at densities of five and 500 thrips per plant; however, at the five thrips per plant, this trend was due to the large deviation seen in infested plants in only one replicate. In light of the low tolerable thrips density of eggplant, it is necessary to confirm whether artificial treatments with chemicals induce the emission of herbivore-induced plant volatiles that can attract O. sauteri and prolong its residence time on the leaf.  相似文献

In order to functionally analyze the predicted defensive role of leaf polyphenol oxidase (PPO; EC 1.10.3.1) in Populus, transgenic hybrid aspen (Populus tremula × P. alba) plants overexpressing a hybrid poplar (Populus trichocarpa × P. deltoides) PtdPPO1 gene were constructed. Regenerated transgenic plants showed high PPO enzyme activity, PtdPPO1 mRNA levels and PPO protein accumulation. In leaf disk bioassays, forest tent caterpillar (Malacosoma disstria) larvae feeding on PPO-overexpressing transgenics experienced significantly higher mortality and reduced average weight gain compared to larvae feeding on control leaves. However, this effect was observed only when older egg masses were used and the resulting larvae showed reduced growth and vigor. In choice tests, no effect of PPO overexpression was detected. Although PPO in poplar leaves is latent and requires activation with detergents or trypsin for full enzymatic activity, in caterpillar frass the enzyme was extracted in the fully activated form. This activation correlated with partial proteolytic cleavage, suggesting that PPO latency and activation during digestion could be an adaptive and defense-related feature of poplar PPO.  相似文献
