A study has been made of the RNA and protein synthesising systems of wheat embryos isolated from seed lots having high viability but differing in vigour. The rate of RNA and protein synthesis in wheat embryos during the early hours of germination is related to the vigour of the seed lot. The imposition of a stress factor, in the nature of a sub-optimal germination temperature, during germination of isolated wheat embryos magnifies the differences in rates of protein and RNA synthesis between high and low vigour seed. Using cell-free protein synthesising systems it has been demonstrated that an important difference between high and low vigour embryos lies in the relative levels of messenger RNA in the embryo. High vigour embryos contain relatively higher levels of poly A+-RNA (i.e. potential mRNA species) than lower vigour embryos and furthermore the level of poly A+-RNA in high vigour embryos increases during early germination whilst in lower vigour embryos the level decreases. The laboratory germination (criterion radicle emergence) of seven seed lots of winter wheat cv. Slejpner was similar. However, they differed in vigour as demonstrated by differences in germination after controlled deterioration carried out at a range of seed moisture contents, at two temperatures and for different times. A vigour assessment for each lot was quantified by calculating a value for the seed lot constant Ki, of the viability equation. Germination in lower water potentials reduced the uptake of water, radicle and coleoptile emergence and radicle and coleoptile extension. There was no difference in the water uptake of seed lots differing in vigour. However, seed lots of lower vigour showed less radicle emergence, less coleoptile emergence and shorter radicles than higher vigour seed lots in low water potentials. Similarly, controlled deterioration resulted in reduced radicle and coleoptile emergence and growth compared to unaged seed, and also to a greater sensitivity to low water potentials. The implications for field establishment are discussed.  相似文献

Five muskmelon (Cucumis melo L.) cultivar seedlots from a commercialsource and freshly produced seeds of two cultivars, when artificiallyaged, were found to differ in their viability and vigour asdetermined by germination tests. Furthermore, the various commercialseedlots without ageing also exhibit a range of deteriorationlevels. Low vigour seeds had higher respiratory quotient valuesthan the high vigour seeds as a result of a higher level ofCO2 production. This high level of CO2 evolution in low vigourseeds may have been due to anaerobic respiration. Levels of acetaldehyde and ethanol produced by imbibing seedswere negatively associated with seed viability and vigour. After6 h of imbibition low vigour seeds produced significantly moreethanol and acetaldehyde than high vigour seeds. After 24 hof imbibition, ethanol continued to accumulate in the commercialseedlots up to 10-fold the amount produced after 6 h of imbibition,whereas, acetaldehyde levels increased less. However, in thefreshly produced, artificially aged seeds (except the most extremeageing), ethanol levels were reduced and no acetaldehyde productioncould be detected, indicating re-utilization of ethanol. It is suggested that ethanol production in the first hours ofimbibition can be used as a reliable index to predict germinationin muskmelon seedlots. Both low and high vigour seed lots were more than 90% viablewhen germinated at 25 °C. The tissue weights and the contentsof mineral ions and glycinebetaine in the ungerminated embryoswere the same for each lot. The release of K+ and phosphateions from isolated embryos into aqueous media were also thesame. These similarities remained for at least 36 h of germination.At 48 h the coleoptiles of the low vigour embryos were about20% shorter and their fresh weight was about 30% less than thoseof the high vigour embryos. These differences were reduced againto about 10% at 72 h. In contrast to these differences, H+ efflux and 36Rb+ uptakeby isolated embryos were up to 50% slower in low vigour embryoscompared with the high vigour ones. The difference was maximalat 24 h of germination and appeared to depend upon the anionpresented and the pH of the incubation medium. The H+/36Rb+exchange ratio was not, however, changed in low vigour embryoscompared with high vigour ones.  相似文献

Thermotolerance is developmentally dependent in germinating wheat seed   总被引:1,自引:1,他引:0  
During the initial 9 to 12 hours of imbibition, the imbibing wheat (Triticum aestivum L.) seed was found to exhibit substantial tolerance to high temperature relative to later times of imbibition. Tolerance was assessed by seed viability and seedling growth. This initial high temperature tolerance gradually declines with increasing time of seed imbibition. A range of 2 hour heat pretreatments (38-42°C) prior to imposition of a 2 hour heat shock (51-53°C) during this same 9 to 12 hour interval was unable to increase survival or seedling growth over that of seed that did not receive a pretreatment. However, after 9 to 12 hours of imbibition the pretreatment provided both increased survival and increased seedling growth, measured 120 hours later, i.e., classical thermotolerance could be acquired. This response is called a `thermotolerance transition.' Isolated embryos responded in a similar manner using a 2,3,5-triphenyltetrazolium chloride assay for viability determination following heat treatments. The high temperature tolerance during early imbibition indicates that the thermotolerance transition involves the loss of an existing thermotolerance coincident with acquiring the ability to become thermotolerant following heat pretreatment. Despite the inability to acquire thermotolerance, heat shock protein synthesis was induced by heat shock immediately upon imbibition of wheat seed or isolated embryos. Developmentally regulated heat shock proteins of 58 to 60, 46, 40, and 14 kilodaltons were detected at 1.5 hours of imbibition following heat shock, but were absent or greatly reduced by 12 hours. Constitutive synthesis of 70 and 90 kilodalton hsp groups appeared to be greater at 1.5 hours of imbibition than at 12 hours of imbibition.  相似文献

Differences in seed vigour of zero- and high-tannin faba beans were investigated using 25 seed lots of 12 cultivars following earlier reports of poor emergence in the zero-tannin types. Field emergence ranged from 54–96% indicating differences in seed vigour between cultivars all having high laboratory germination (>91%). Seed from zero-tannin accessions with poor emergence had a higher incidence of testa and cotyledon cracking, a smaller percentage of hard seeds, more rapid water uptake, a lower percentage of vital staining of cotyledons and a greater leaching of solutes than high-tannin types. Nevertheless, variation in these characteristics existed between cultivars and lines of both types. Seeds with more cracks in the seed coat and fewer hard seeds imbibed water more rapidly and consequently showed lower levels of vital staining and more cracks in the abaxial surface of the cotyledons. Slower imbibition in polyethylene glycol lessened the incidence of these deleterious characteristics and may provide a practical resolution to the problem of poor field emergence in zero-tannin lines of faba bean with low seed vigour.  相似文献

Seeds frequently face a hostile environment during early germination. In order to determine whether seeds have evolved unique mechanisms to deal with such environments, a survey of the heat shock response in isolated embryos of wheat (Triticum aestivum L.) was undertaken. Embryos simultaneously heat shocked and labeled following several different periods of prior imbibition up to 12 hours synthesized many groups of heat shock proteins (hsps) typical of other plant and animal systems. Also, five developmentally dependent hsps, present only in treatments imbibed less than 6 hours prior to heat shock, were detected. These proteins have relative molecular masses of 14, 40, 46, 58, and 60 kilodaltons. One of the developmentally dependent hsps is among the most highly labeled hsps found in early imbibed embryos. The possibility that this protein is the Em protein is discussed. The hypothesis that the capacity for hsp synthesis is affected by seed vigor was also tested. The heat shock responses of embryos from two high and two low vigor seed lots were compared using one- and two-dimensional electrophoresis of labelled protein extracts. The results indicate that both of the low vigor lots tested had weaker heat shock responses than their high vigor counterparts overall. Not all hsps were relatively less abundant in low vigor embryos. The developmentally dependent hsps showed little relationship to vigor. Some of the developmentally dependent hsps were actually made in greater amounts, relative to other proteins, in the low vigor seed lots. The results presented here demonstrate that imbibing embryos are capable of expressing an enhanced heat shock response, and that this response is related to seed vigor.  相似文献

Determination of seed vigor was attempted by comparing ATP-levels of deteriorating seed to germination percentage and production of dry matter. Immediately after imbibition of any seed lot investigated, a production of ATP took place. This ATP-accumulation invariably reached a plateau after 6 h of imbibition. Two well germinating seed lots of rape, one of cauliflower and one of sugar beet, were artificially aged by means of elevated storage temperature and humidity. Every second week through 16 weeks of deterioration the levels of ATP, ADP and AMP after 7 h of imbibition were compared with the germination percentage. While ADP- and AMP-contents of germinating seed displayed no change (when imbibed 7 h) during the period of artificial aging, seed deterioration was reflected in the ATP-levels long before loss of viability could be detected by the conventional germination test.
When ATP-levels per seed were related to germination percentage throughout the aging, all four seed lots displayed similar patterns although the absolute figures differed. In contrast to the conventional "per seed' basis, however, ATP per gram seed not only displayed similar deterioration patterns, but the absolute values were also of the same magnitude.  相似文献

Using seed priming and accelerated ageing techniques, a singlelot of leek (Allium porrum) seeds was manipulated to producefour lots of seeds with different germination performance. Changesin content of the major nucleic acid species in whole seedsand embryos of two of these lots (primed and unprimed), weredetermined over the early stages of germination. The major effectof priming was an increased level of RNA species in the seedsand embryos, and this difference was maintained during germination.Comparison of nucleic acid levels in the dry seeds of thesetwo lots and two others (aged and aged then primed) indicatedthat there was no correlation with germination performance.Similar comparisons of the nucleic acid levels in the embryosof seeds imbibed for 1 d showed only a limited correlation betweenrRNA levels and germination performance. Analysis of these datasuggests that accelerated ageing has an adverse effect uponendosperm cells, which results in the degradation of their nucleicacids during priming. Furthermore, the viability of these agedseeds also falls during priming. The data also indicate thatratios of rRNA to DNA correlate with germination performanceof the four lots of seeds studied. It is proposed that sucha relationship is indicative of the efficiency of a primingtreatment, and may be useful in comparisons of naturally varyingseed lots. Electron microscope observation revealed that mitochondria in plumular root cells of soy bean seed were invisible at the beginning of seed germination. They were sequentially formed re-differentiated and developed from proplastids during the process of germination. The proplastids in the control plumular root markedly expanded at low temperature imbibition process and did not develop into mitochondria when normal temperature was restored. Even the structure of reticulata membrane was further destroyed and vacuolized. In contrast, mitochondria of soybean seed pretreated with polyvinyl alcohol (PVA) and polythylene glycol-6000 (PEG) continused to develop well under the same conditions, as characterized by a clear structure of bilayered membrane and tube cristae. The results suggest that continuous re-differentiation and development of proplastids into mitochondria at low temperature imbibition process play an important role in the increase of soybean seed vigour and its resistance to chilling injury.  相似文献

A simple method is presented for selecting individual seedsfor growth and vigour prior to germination. The selection isbased on the measurement of an electric current originatingduring the initial stage of seed imbibition. After selecting,seeds may be returned to the quiescent state without affectingviability. Results of both laboratory germination and fieldexperiments demonstrate that high germination rates, more extensivegrowth, higher yields, and fewer defective plants are associatedwith small currents. Seeds of low viability and high currentsmay be removed to upgrade the germination and vigour. Resultsare presented from several varieties of Phaseolus vulgaris andfrom three other plant species. Experiments are discussed whichindicate that seeds graded by current levels possess markedlydifferent respiration rates.  相似文献

The germination of viable seeds is the basis for new plant growth and development. Seeds lose viability during storage, but the biochemical mechanisms of seed death are not fully understood. This study aimed to investigate degradation patterns of nucleic acids during seed ageing and subsequent water uptake. Seeds of Pisum sativum L. were artificially aged at 50°C and 12% seed water content (WC). Nucleic acids degradation was studied during ageing and during imbibition of four seed lots with differential viability from highly viable to dead. As seeds lost viability during ageing, DNA was gradually degraded into internucleosomal fragments, resulting in ‘DNA laddering’, in conjunction with disintegration of 18S and 28S rRNA bands. During imbibition, non-aged controls had high levels of DNA and RNA integrity through to radicle protrusion. In an aged seed lot with 85% total germination (TG) DNA fragmentation decreased upon imbibition probably due to nucleosome degradation, while rRNA integrity did not improve. In an aged seed lot with 44% TG, neither DNA nor rRNA integrity improved upon imbibition. Dead seeds showed DNA degradation as laddering throughout imbibition along with extensive degradation of rRNA. We present a model in which interlinked programmed and non-programmed events contribute to seed ageing, and suggest that protection of nucleic acids during ageing is key to seed longevity.  相似文献

电镜观察发现,大豆种子在刚开始萌发时胚根细胞中未能见到线粒体,线粒体是在种子萌发过程中逐渐出现的,由原质体再分化发育而成。对照胚根细胞内原质体在低温吸张过程中明显膨胀,在回温后胚根细胞中原质体仍不能发育成线粒体,甚至网状膜结构破坏,呈空泡化;经聚乙烯醇(PVA)和聚乙二醇(PEG 6000)预处理的大豆种子在同样条件下线粒体能继续发育,在回温后预处理胚根细胞中线粒体发育良好,具有明显的双层膜和管状嵴的结构。这些结果表明,在低温吸胀过程中原质体能够继续再分化发育成线粒体是提高大豆种子活力和抗冷力的重要原因。  相似文献

On germination of low viability embryos of rye, var. LovaszPatonai, the rate of protein synthesis increases during theearly hours of imbibition and high rates of DNA replicationcommence around the 9th hour. If embryos are imbibed for 3 or6 h then dehydrated back to their original weight, their rateof protein synthesis when next imbibed closely corresponds tothat of embryos germinated for a period equal to that of thehydration pre-treatment plus the duration of the second imbibition.Pre-treatment also enhances subsequent RNA synthesis and embryoshydrated for 9 h then dehydrated start major DNA synthesis atonce as water is again supplied. Many changes occurring duringthese periods of hydration pre-treatment must therefore be stableto subsequent dehydration. Damage occurs to areas that are firstactive in protein and RNA synthesis if pre-treatments extendbeyond 9 h and subsequent germination of the embryo is thenimpaired. The implications of these results are discussed inrelation to the effects of hydration pre-treatments upon enhancedgermination and the stability to dehydration of the productsof early RNA and protein synthesis.  相似文献
