My association with the JCB began very early in my scientific career. In fact, it predated my understanding that there would even be a scientific career. In the mid-1970s while still an undergraduate, the JCB published my very first paper, a contribution noted perhaps less so for its reporting the characterization of the first known protein in plant cell walls than for a footnote that called attention to the evolutionary conservation of a relationship between “sex and slime” throughout the plant and animal kingdoms.  相似文献

A calendar of fungal spore seasons for Melbourne during 1993was established using a 7-day volumetric Burkard trap. Twenty-ninegenera and five spore groups were identified. The dominant spore typesdetected were Cladosporium (41.7%),Leptosphaeria (14.9%), Coprinus (14.6%),`Ascospore 1' (5.5%), Ganoderma (2.1%) andAlternaria (1.4%). Seasonally, spore levels ofCladosporium and Alternaria peaked in spring andsummer, Leptosphaeria and Ganoderma peaked towardsummer and autumn, `Ascospore 1' peaked in winter, whilst spore levelsof the basidiomycete Coprinus fluctuated year round. Inconclusion, a range of allergenic fungal spores were present in the airof Melbourne throughout the year.  相似文献

In this issue of Neuron, Grant et al. report a series of elegant zebrafish experiments that reveal a previously unappreciated role for glia in the regulation of hair cell proliferation and differentiation. Phosphate uptake by the blue-green alga Oscillatoria limnetica Lemmerman is stimulated by micromolar concentrations of Ca2+. The calmodulin antagonists 4-(3-[2(trifluoromethyl)phenylthiazin-10-yl]propyl)-1-piperazine ethanol-HCl and its monofluoro-analog inhibit orthophosphate uptake of Oscillatoria limnetica by over 97% implying involvement of calmodulin in this process. A calmodulin-like protein was quantitated in cell-free extracts from O. limnetica by radioimmunoassay.  相似文献

The debate over whether the medical profession should accommodate its members' conscientious objections (COs) has raged on in the bioethics literature and on legislative floors for decades. Unfortunately, participants on all sides of the debate fail to distinguish among different types of CO, a failure that obstructs the view of which cases warrant accommodation and why. In this paper, we identify one type of CO that warrants consideration for accommodation, called Nature of Medicine COs (NoMCOs). NoMCOs involve the refusal of physicians to perform actions they reasonably judge to be contrary to the nature of medicine and their professional obligations. We argue that accommodating NoMCOs can be justified based on the profession's need to preserve reformability. Importantly, this previously underdeveloped position evades some of the concerns commonly raised by opponents of CO accommodations.  相似文献

The specific inhibitor protein (calpastatin) for the calcium-dependent intracellular proteinases (calpains) is an important regulator of these enzymes. In this communication we describe a one day procedure for purifying 3 to 5 mg of calpastatin from a kilogram of bovine myocardium. This represents a substantial improvement over previously described methods, and should facilitate future studies of calpastatin structure and function. A key, novel step in the purification was dye-matrix chromatography on an Affi-Gel Blue column. Contrary to previous indications, calpastatin purified by the new method did not contain significant amounts of carbohydrate. However, the presence of covalently bound phosphate in purified bovine myocardial calpastatin was confirmed and co-migration of phosphate and calpastatin activity was demonstrated on Bio-Gel A-1.5m chromatography. Thus, it is possible that calpastatin function is regulated by phosphorylation.  相似文献

With high productivity and stress tolerance, numerous grass genera of the Andropogoneae have emerged as candidates for bioenergy production. To optimize these candidates, research examining the genetic architecture of yield, carbon partitioning, and composition is required to advance breeding objectives. Significant progress has been made developing genetic and genomic resources for Andropogoneae, and advances in comparative and computational genomics have enabled research examining the genetic basis of photosynthesis, carbon partitioning, composition, and sink strength. To provide a pivotal resource aimed at developing a comparative understanding of key bioenergy traits in the Andropogoneae, we have established and characterized an association panel of 390 racially, geographically, and phenotypically diverse Sorghum bicolor accessions with 232,303 genetic markers. Sorghum bicolor was selected because of its genomic simplicity, phenotypic diversity, significant genomic tools, and its agricultural productivity and resilience. We have demonstrated the value of sorghum as a functional model for candidate gene discovery for bioenergy Andropogoneae by performing genome-wide association analysis for two contrasting phenotypes representing key components of structural and non-structural carbohydrates. We identified potential genes, including a cellulase enzyme and a vacuolar transporter, associated with increased non-structural carbohydrates that could lead to bioenergy sorghum improvement. Although our analysis identified genes with potentially clear functions, other candidates did not have assigned functions, suggesting novel molecular mechanisms for carbon partitioning traits. These results, combined with our characterization of phenotypic and genetic diversity and the public accessibility of each accession and genomic data, demonstrate the value of this resource and provide a foundation for future improvement of sorghum and related grasses for bioenergy production.  相似文献

Brain-related researches encompass many fields of studies and usually involve worldwide collaborations. Recognizing the value of these international collaborations for efficient use of resources and improving the quality of brain research, the International Neuroinformatics Coordinating Facility (INCF) started to coordinate the effort of establishing neuroinformatics (NI) centers and portal sites among the different participating countries. These NI centers and portal sites will serve as the conduit for the interchange of information and brain-related resources among different countries. In Japan, several NI platforms under the support of NIJC (NI Japan Center) are being developed with one platform called, Visiome, already operating and publicly accessible at "http://www.platform.visiome.org". Each of these platforms requires their own set of keywords that represent important terms covering their respective fields of study. One important function of this predefined keyword list is to help contributors classify the contents of their contributions and group related resources. It is vital, therefore, that this predefined list should be properly chosen to cover the necessary areas. Currently, the process of identifying these appropriate keywords relies on the availability of human experts which does not scale well considering that different areas are rapidly evolving. This problem prompted us to develop a tool to automatically filter the most likely terms preferred by human experts. We tested the effectiveness of the proposed approach using the abstracts of the Vision Research Journal (VR) and Investigative Ophthalmology and Visual Science Journal (IOVS) as source files.  相似文献

哺乳动物发育相关的功能基因的鉴定多来源于自发或诱发突变的小鼠。小鼠白内障作为较易鉴定的性状,目前业已明确的突变多达140余个。自发突变的小鼠主要来源于大规模饲养,诱发突变主要通过X射线与ENU处理获得,基因敲除与转基因小鼠亦可获得白内障性状。已知的白内障相关基因分布于小鼠20条染色体,其中以1号染色体最多,并常于小鼠胚胎时期起始表达。小鼠白内障表型多由单基因突变引起,突变的确定主要通过F2代家系全基因组扫描、精细定位、单倍型分型与测序验证等程序。本文从小鼠白内障基因突变来源、基因分布、基因定位与基因表达等角度较为全面的阐述了小鼠先天性白内障的研究进展。  相似文献
