Formalin fixation is a mainstay of modern histopathologic analysis, yet the practice is poorly standardized and a significant potential source of preanalytical errors. Concerns of workflow and turnaround time drive interest in developing shorter fixation protocols, but rapid protocols can lead to poor histomorphology or inadequate downstream assay results. Additionally, assays such as immunohistochemistry for phosphorylated epitopes have historically been challenging in the context of formalin-fixed tissue, indicating that there may be room for improvement in this process that is fundamental to the practice of anatomic pathology. With these issues in mind, we studied basic formalin biochemistry to develop a novel formalin fixation protocol that involves a pre-incubation in subambient temperature formalin prior to a brief exposure to heated formalin. This new protocol is more rapid than standard protocols yet preserves histomorphology and yields tissue that is compatible with an expanded set of downstream clinical and research assays, including immunohistochemistry for phosphorylated epitopes.  相似文献

Scholarly achievement is the aim of a pathology institute. In a medical faculty it serves undergraduate, postgraduate teaching, practice and research. Historically, scholarly achievement in these fields has been promoted through the teaching responsibility of the university and hospital. With the increasing volume of work, knowledge and complexity of research, adequate performance in these fields is best obtained by limiting the volume of fixed duties for each individual. At the same time, depth of knowledge is gained by encouraging specialization to complement clinical specialties. This arrangement will guarantee that the teaching and research of the department are disease-oriented, which is the central theme of pathology. With a highly specialized staff there must be a core of intradepartmental conferences common to all staff and those in training. With this form of organization dedicated to the undergraduate and postgraduate teaching, the objectives of scholarly achievement in teaching practice and research will be obtained.  相似文献

在入侵生态学研究方面 ,物种多样性与生物入侵之间的关系已成为当前研究和争论的焦点。自Elton的经典假说提出以来 ,物种丰富度高的群落比物种贫乏的群落更能抵抗外来种入侵的观点得到广泛接受。一些理论模型和多样性处理实验支持了该假说。但现在越来越多的野外观测和实验研究开始对这一经典假说提出异议 ,甚至反对。同时 ,在入侵生态学广泛受到关注的今天 ,大量的实验研究也提出了一些新的观点。本文对Elton经典假说提出以来全球有关物种多样性与生物入侵关系的主要研究及其观点进行了评述 ,以期为我国有关研究工作的开展提供参考。  相似文献

The intention and practice of conservation reserve selection are different. A major reason for systems of reserves is to sustain biological diversity. This involves protecting examples of as many natural features, e.g. species, communities or environments, as possible. In reality, however, new reserves have rarely been dedicated for their representation of features. Furthermore, the opportunism that has characterized the development of reserve systems can actually jeopardize the representation of all features in reserves through the inefficient allocation of limited resources. More systematic approaches are essential if reserves are to play their role in protecting biodiversity. Some basic principles for conservation planning are emerging from recent systematic procedures for reserve selection. These principles will help to link intention and practice.  相似文献

Medical imaging using X-rays has been one of the most popular imaging modalities ever since the discovery of X-rays 125 years ago. With unquestionable benefits, concerns about radiation risks have frequently been raised. Computed tomography (CT) and fluoroscopic guided interventional procedures have the potential to impart higher radiation exposure to patients than radiographic examinations. Despite technological advances, there have been instances of increased doses per procedure mainly because of better diagnostic information in images. However, cumulative dose from multiple procedures is creating new concerns as effective doses >100 mSv are not uncommon. There is a need for action at all levels. Manufacturers must produce equipment that can provide a quality diagnostic image at substantially lesser dose and better implementation of optimization strategies by users. There is an urgent need for the industry to develop CT scanners with sub-mSv radiation dose, a goal that has been lingering. It appears that a new monochromatic X-ray source will lead to replacement of X-ray tubes all over the world in coming years and will lead to a drastic reduction in radiation doses. This innovation will impact all X-ray imaging and will help dose reduction. For interventional procedures, the likely employment of robotic systems in practice may drastically reduce radiation exposures to operators- but patient exposure will still remain an issue. Training needs always need to be emphasized and practiced.  相似文献

A Laboratory Branch Task Force of the National Communicable Disease Center has proposed a standardized complement fixation procedure (LBCF) and an adaptation of this to microtitration techniques (MT) as uniform methods for performing complement fixation (CF) tests. A common procedure should make CF results from one laboratory more comparable to another. In addition, it would be preferable if the common procedure reproduced the titer levels of a testing procedure which is to be replaced, particularly when valid clinical interpretations have been derived from the latter. Replicated sets of sera were tested by the LBCF, MT, and the standard Smith CF procedure for coccidioidomycosis. Results with all three procedures were highly reproducible within an acceptable one-tube variation of a twofold dilution series, but the frequency of one-tube variations was greater with the MT method than with the other two. There was no statistical difference in the titers obtained with the Smith and LBCF procedures, but there was a significant difference when the MT results were compared to those with the Smith method. The LBCF method should be acceptable as a standardized and uniform CF procedure for coccidioidomycosis, subject to comparative testing between different laboratories.  相似文献

Mass spectrometry imaging (MSI) enables the direct analysis of molecules from the surface of a wide variety of samples, allowing the multiplex measurement of both abundance and distribution of small molecules, lipids, peptides and proteins. As the technology has been refined an increasing number of ionization methods and mass analyzers has been used that enable increased spatial and spectral resolution measurements to be made at an increased speed. Alongside the instrumentation improvements there has been optimization of sample preparation procedures that allow the highest quality data to be obtained, reproducibly, from an ever increasing diversity of samples. This review will consider the development and standardization of sample preparation methods applicable to MSI, describing the stages and procedures undertaken from the instance of sample collection, through storage, preparation and on through final processing prior to analysis. Recent technical advancements will be highlighted and areas where further experimentation and optimization may well be required will be described. All aspects of the sample preparation pipeline will be considered in detail, with examples from the literature used to emphasize why rigorous sample preparation for MSI is vital to achieve the most accurate, reproducible and validated MSI data possible.  相似文献

The available data demonstrated that interphase Ag-NOR evaluation may be of help in distinguishing malignant from hyperplastic or normal cells. On the other hand, there is increasing evidence that a relationship exists between the quantity of interphase Ag-NORs and the prognosis of malignant tumors: the greater the number of interphase Ag-NORs, the worse is the prognosis. This can be explained by the observation that the interphase Ag-NOR quantity is strictly related to the cell proliferation rate. The procedures used for the measurement of the interphase Ag-NOR quantity are also critically discussed.  相似文献

In a previous publication, we described new devices and results for diagnostic histopathological transmission electron microscopy on large, ultrathin cryosections from several human tumours. As reported, cryo-ultramicrotomy is gaining in significance and use as technical improvements are made. In comparison with epoxy resin procedures, cryo-preparations are more rapid, there is negligible loss of material and thus antigenicity is well preserved. This work contains some newly developed details for a new type of cryo-ultramicrotome, especially adapted for histopathological investigations on large ultrathin cryosections. It is a software-controlled ultramicrotome advance drive system with feedback regulation and various advancement sensors. Some of these developments could also be applied to existing microtome systems.  相似文献

群落的物种数目,即物种丰富度,是最古老、同时也是最基本的一个多样性概念,从对它的估计中可以得到关于物种灭绝速率方面的信息,这对生物多样性保护是非常重要的。已经提出了很多方法来估计群落中的物种数目,这些方法可以分为两大类,即基于理论抽样的方法和基于数据分析的方法。前者包括经典估计方法和贝叶斯估计方法;后者包括对数正态分布的积分方法、再抽样方法和种-面积曲线的外推方法。发现:(1)有些方法适用于动物群落,如大多数基于理论抽样的方法;有些方法则适用于植物群落,如大多数基于数据分析的方法;(2)这些方法还没有经过全面而系统地比较;(3)还没有一个普遍认为比较好的方法。因此,建议采用野外调查与模拟研究相结合的方法对各种估计方法进行系统地评价。  相似文献

One of the major breakthroughs in molecular pathology during the last decade was the successful extraction of full-length proteins from formalin-fixed and paraffin-embedded (FFPE) clinical tissues. However, only limited data are available for the protein extraction efficiency of over-fixed tissues and FFPE blocks that had been stored for more than 15 years in pathology archives. In this study we evaluated the protein extraction efficiency of FFPE tissues which had been formalin-fixed for up to 144 hours and tissue blocks that were stored for 20 years, comparing an established and a new commercial buffer system. Although there is a decrease in protein yield with increasing fixation time, the new buffer system allows a protein recovery of 66% from 144 hours fixed tissues compared to tissues that were fixed for 6 hours. Using the established extraction procedure, less than 50% protein recovery was seen. Similarly, the protein extraction efficiency decreases with longer storage times of the paraffin blocks. Comparing the two buffer systems, we found that 50% more proteins can be extracted from FFPE blocks that were stored for 20 years when the new buffer system is used. Taken together, our data show that the new buffer system is superior compared to the established one. Because tissue fixation times vary in the routine clinical setting and pathology archives contain billions of FFPE tissues blocks, our data are highly relevant for research, diagnosis, and treatment of disease.  相似文献

Most industrial injuries are treated by physicians who have a general practice in addition to their industrial work. Because of the increasing number and seriousness of industrial injuries, better preparation for treatment is required in the patient's interest and for reasonable economy. The trend toward centralization of industrial medical facilities, increasing before and during World War II, has been somewhat reversed; the general practitioner now has an opportunity to take a useful place in the care of injured employees.A physician dealing with industrial cases must be prepared to give immediate emergency treatment, to comply with the procedures of insurance carriers, and to refer cases which he is not skilled or equipped to handle. As a personal physician having the confidence of the patient he can secure greater cooperation from all parties concerned in industrial disability and can sometimes promote a more rapid recovery.  相似文献

In a review of the operative treatment of 1,271 patients with pulmonary tuberculosis in an 11-year period, it was noted that, beginning with 1947, there was a great increase, relatively, in the number of cases in which pulmonary resection was carried out. In 1943, the first year of the period of study, there were six minor operative procedures to every four major operations; in 1953 the ratio was one minor to nine major. This reversal reflects the discoveries of antibiotics for conservative therapy on the one hand and the advances in surgical techniques for major operative treatment on the other. Now that it is safer, resection will probably be used more and more-including bilateral resection in "salvage" cases. On the other hand, with specific antibiotics available, there is a tendency at present to treat conservatively for longer periods in cases in which, formerly, minor operative procedures would have been carried out early.  相似文献

Population genetics and phylogeography of the African buffalo (Syncerus caffer) are inferred from genetic diversity at mitochondrial D-loop hypervariable region I sequences and a Y-chromosomal microsatellite. Three buffalo subspecies from different parts of Africa are included. Nucleotide diversity of the subspecies Cape buffalo at hypervariable region I is high, with little differentiation between populations. A mutation rate of 13-18% substitutions/million years is estimated for hypervariable region I. The nucleotide diversity indicates an estimated female effective population size of 17 000-32 000 individuals. Both mitochondrial and Y-chromosomal diversity are considerably higher in buffalo from central and southwestern Africa than in Cape buffalo, for which several explanations are hypothesized. There are several indications that there was a late middle to late Pleistocene population expansion in Cape buffalo. This also seems to be the period in which Cape buffalo evolved as a separate subspecies, according to the net sequence divergence with the other subspecies. These two observations are in agreement with the hypothesis of a rapid evolution of Cape buffalo based on fossil data. Additionally, there appears to have been a population expansion from eastern to southern Africa, which may be related to vegetation changes. However, as alternative explanations are also possible, further analyses with autosomal loci are needed.  相似文献

Recent very rapid developments in genome and EST projects have identified an increasing number of gene products homologous to those that were previously identified by other methods. Calpain is no exception. At the time this review is written, 83 genes from 23 living organisms have been identified in the database to encode amino acid sequences showing significant similarities to the protease domain of "conventional" calpain, which was first purified as a homogeneous protein in 1978. Progress in genome/EST projects has occurred so quickly that there seems to be some confusion as to the identity of each calpain molecule. This review will attempt to clarify all calpain homologues, to describe the common and differing features of calpain homologues in terms of structure-function relationship, and to discuss the evolutionary process of calpain.  相似文献
