The aim of the present study was to survey the growth and astaxanthin production of E17, an astaxanthin-rich mutant of Chlorella zofingiensis, through feeding the low-cost carbon source cane molasses. In heterotrophic batch cultivation, E17 fed with pretreated molasses achieved biomass (1.79 g L?1 day?1) and astaxanthin (1.99 mg L?1 day?1) productivities comparable to those with glucose, which were about 2- and 2.8-fold of those fed with untreated molasses, respectively. Molasses-induced astaxanthin accumulation may be attributed to the elicited expression of carotenogenic genes, in particular the genes specifically responsible for the ketolation and hydroxylation of β-carotene to form astaxanthin. A two-stage fed-batch strategy was employed to grow E17 and induce astaxathin accumulation, resulting in 45.6 g L?1 biomass and 56.1 mg L?1 astaxanthin, the highest volumetric astaxanthin yield ever reported for this alga. In addition, the astaxanthin production by E17 was tested with a semi-continuous culture method, where the directly diluted raw molasses (giving 5 g L?1 sugar) was used as the carbon source. Little growth inhibition of E17 was observed in the semi-continuous culture with a biomass productivity of 1.33 g L?1 day?1 and an astaxanthin productivity of 0.83 mg L?1 day?1. The mixotrophic semi-continuous cultures enhanced the biomass and astaxanthin productivities by 29.3 % and 42.2 %, respectively. This study highlights the potential of using the industrially cheap cane molasses towards large-scale cost-saving production of the high-value ketocarotenoid astaxanthin.  相似文献

Microalgae have been used to remove nitrogen, phosphorus, and chemical oxygen demand (COD) from brewery wastewater (BWW). The microalga Scenedesmus obliquus was grown on BWW, using bubble column photobioreactors that operated under batch and continuous regimes. For the first time, the cell physiological status cell membrane integrity and enzymatic activity was monitored during the microalgae based BWW treatment, using flow cytometry. All the cultivations batch and continuous displayed a proportion of cells with intact membrane >?87%, although the continuous cultivations displayed a lower proportion of cells with enzymatic activity (20–40%) than the batch cultivations (97%). The dilution rate of 0.26 day?1 was the most favorable condition, since the microalgae cultivation attained the maximum biomass productivity (0.2 g ash-free dry weight day?1) and the total nitrogen and COD removal rates were the highest (97 and 74%, respectively), while the phosphorous removal rate was the third (23%).  相似文献

For the design of a large field of vertical flat plate photobioreactors (PBRs), the effect of four design parameters—initial biomass concentration, optical path length, spacing, and orientation of PBRs—on the biochemical composition and productivity of Chlorella zofingiensis was investigated. A two-stage batch process was assumed in which inoculum is generated under nitrogen-sufficient conditions, followed by accumulation of lipids and carbohydrates in nitrogen-deplete conditions. For nitrogen-deplete conditions, productivity was the most sensitive to initial biomass concentration, as it affects the light availability to individual cells in the culture. An initial areal cell concentration of 50 g m?2 inoculated into 3.8-cm optical path PBR resulted in the maximum production of lipids (2.42?±?0.02 g m?2 day?1) and carbohydrates (3.23?±?0.21 g m?2 day?1). Productivity was less sensitive to optical path length. Optical path lengths of 4.8 and 8.4 cm resulted in similar areal productivities (biomass, carbohydrate, and lipid) that were 20 % higher than a 2.4-cm optical path length. Under nitrogen-sufficient conditions, biomass productivity was 48 % higher in PBRs facing north–south during the winter compared to east–west, but orientation had little influence on biomass productivity during the spring and summer despite large differences in insolation. An optimal spacing could not be determined based on growth alone because a tradeoff was observed in which volumetric and PBR productivity increased as space between PBRs increased, but land productivity decreased.  相似文献

Microalgae cultivation systems can be divided broadly into open ponds and closed photobioreactors. This study investigated the growth and biomass productivity of the halophilic green alga Tetraselmis sp. MUR-233, grown outdoors in paddle wheel-driven open raceway ponds and in a tubular closed photobioreactor (Biocoil) at a salinity of 7 % NaCl (w/v) between mid-March and June 2010 (austral autumn/winter). Volumetric productivity in the Biocoil averaged 67 mg ash-free dry weight (AFDW) L?1 day?1 when the culture was grown without CO2 addition. This productivity was 86 % greater, although less stable, than that achieved in the open raceway pond (36 mg L?1 day?1) grown at the same time in the autumn period. The Tetraselmis culture in the open raceway pond could be maintained in semi-continuous culture for the whole experimental period of 3 months without an additional CO2 supply, whereas in the Biocoil, under the same conditions, reliable semi-continuous culture was only achievable for a period of 38 days. However, stable semi-continuous culture was achieved in the Biocoil by the addition of CO2 at a controlled pH of ~7.5. With CO2 addition, the volumetric biomass productivity in the Biocoil was 85 mg AFDW L?1 day?1 which was 5.5 times higher than the productivity achieved in the open raceway pond (15 mg AFDW L?1 day?1) with CO2 addition and 8 times higher compared to the productivity in the open raceway pond without CO2 addition (11 mg AFDW L?1 day?1), when cultures were grown in winter. The illuminated area productivities highlight an alternative story and showed that the open raceway pond had a three times higher productivity (3,000 mg AFDW m?2 day?1) compared to the Biocoil (850 mg AFDW m?2 day?1). Although significant differences were found between treatments and cultivation systems, the overall average lipid content for Tetraselmis sp. MUR-233 was 50 % in exponential phase during semi-continuous cultivation.  相似文献

Semi-continuous algal cultivation was completed in outdoor flat-panel photobioreactors (panels) and open raceway ponds (raceways) from February 17 to May 7, 2015 for side-by-side comparison of areal productivities at the Arizona Center for Algae Technology and Innovation in Mesa, AZ, USA. Experiments used two strains of Scenedesmus acutus (strains LB 0414 and LB 0424) to assess productivity, areal density, nutrient removal, and harvest volume across cultivation systems and algal strains. Panels showed an average biomass productivity of 19.0?±?0.6 g m?2 day?1 compared to 6.62?±?2.3 g m?2 day?1 for raceways. Photosynthetic efficiency ranged between 1.32 and 2.24 % for panels and between 0.30 and 0.68 % for raceways. Panels showed an average nitrogen consumption rate of 38.4?±?8.6 mg N L?1 day?1. Cultivation in raceways showed a consumption rate of 3.8?±?2.5 and 7.1?±?4.2 mg N L?1 day?1 for February/March and April/May, respectively, due to increase in biomass productivity. Excess nutrients were required to prevent a decrease in productivity. Daily biomass harvest volumes between 18 and 36 % from panels did not affect culture productivity, but density decreased with increased harvest volume. High cultivation temperatures above 30 °C caused strain LB 0414 to lyse and crash. Strain LB 0424 did not show any difference in biomass productivity when peak temperatures reached 34, 38, or 42 °C, but showed decreased productivity when the peak temperature during cultivation was 30 °C. Using algal strains with different temperature tolerances can generate increased annual biomass productivity.  相似文献

Different pilot-scale outdoor photobioreactors using medium recycling were operated in a greenhouse under different environmental conditions and the growth rates (0.1 to 0.5 day?1) obtained evaluated in order to compare them with traditional systems used in aquaculture. The annualized volumetric growth rate for Nannochloropsis gaditana was 0.26 g l?1 day?1 (peak 0.4 g l?1 day?1) at 0.4 day?1 in a 5-cm wide flat-panel bioreactor (FP-PBR). The biomass productivity achieved in this reactor was 10-fold higher than in traditional reactors, reaching values of 28 % and 45 % dry weight (d.w.) of lipids and proteins, respectively, with a 4.3 % (d.w.) content of eicosapentaenoic acid (EPA). A model for predicting EPA productivity from N. gaditana cultures that takes into account the existence of photolimitation and photoinhibition of growth under outdoor conditions is presented. The effect of temperature and average irradiance on EPA content is also studied. The maximum EPA productivity attained is 30 mg l?1 day?1.  相似文献

Background

Microalgae are a potential source of sustainable commodities of fuels, chemicals and food and feed additives. The current high production costs, as a result of the low areal productivities, limit the application of microalgae in industry. A first step is determining how the different production system designs relate to each other under identical climate conditions. The productivity and photosynthetic efficiency of Nannochloropsis sp. CCAP 211/78 cultivated in four different outdoor continuously operated pilot-scale photobioreactors under the same climatological conditions were compared. The optimal dilution rate was determined for each photobioreactor by operation of the different photobioreactors at different dilution rates.

Results

In vertical photobioreactors, higher areal productivities and photosynthetic efficiencies, 19–24 g m?2 day?1 and 2.4–4.2 %, respectively, were found in comparison to the horizontal systems; 12–15 g m?2 day?1 and 1.5–1.8 %. The higher ground areal productivity in the vertical systems could be explained by light dilution in combination with a higher light capture. In the raceway pond low productivities were obtained, due to the long optical path in this system. Areal productivities in all systems increased with increasing photon flux densities up to a photon flux density of 30 mol m?2 day?1. Photosynthetic efficiencies remained constant in all systems with increasing photon flux densities. The highest photosynthetic efficiencies obtained were; 4.2 % for the vertical tubular photobioreactor, 3.8 % for the flat panel reactor, 1.8 % for the horizontal tubular reactor, and 1.5 % for the open raceway pond.

Conclusions

Vertical photobioreactors resulted in higher areal productivities than horizontal photobioreactors because of the lower incident photon flux densities on the reactor surface. The flat panel photobioreactor resulted, among the vertical photobioreactors studied, in the highest average photosynthetic efficiency, areal and volumetric productivities due to the short optical path. Photobioreactor light interception should be further optimized to maximize ground areal productivity and photosynthetic efficiency.

In this study, an alga-based simultaneous process of treating swine wastewater (SWW) and producing biodiesel was explored. Chlorella vulgaris (UTEX-265) was employed as a model species, and a SWW-based medium was prepared by dilution with tap water. Chlorella vulgaris grew well in the SWW-based medium, and at optimum dilution ratios, it exceeded the conventional culture medium in terms of biomass concentration and productivity. In eightfold diluted SWW, which supported the maximum growth, biomass productivity was 0.247 g L?1 day?1, while the productivity was merely 0.165 g L?1 day?1 in standard tris-acetate-phosphorous (TAP) algal medium. In addition, fatty acid methyl ester (FAME) productivity was greater in the SWW-based medium (0.067 versus 0.058 g L?1 day?1). This enhanced productivity resulted in more than 95 % removal of both nitrogen and phosphorous. All these show that C. vulgaris cultivation is indeed possible in a nutrient-rich wastewater with appropriate dilution, and in so doing, the wastewater can effectively be treated.  相似文献

An annular internally illuminated photobioreatcor (IIPBR) configuration based on the airlift/bubble column principles was developed and validated at an 18 l prototype scale using Scenedemus sp. and Nannochloropsis salina in batch and semi-continuous modes, at constant light supply and constant gas-to-culture volume ratio, but at varying CO2-to-air ratios. Highest biomass production was recorded at CO2-to-air ratio of 4% with Scenedesmus sp. and at 1% with Nannochloropsis salina. The energetic performance of this IIPBR was quantified in terms of biomass productivity per unit energy input, P/E (g W?1 day?1), considering energy input for illumination and for pneumatic mixing and circulation. Under optimal conditions, the IIPBR evaluated in this study achieved P/E of 1.42 g W?1 day?1 for Scenedesmus sp. and P/E of 0.34 g W?1 day?1 for Nannochloropsis salina. These P/E values are better than those estimated for airlift and bubble column photobioreactor configurations reported in the literature.  相似文献

The influences of urea, nitrate and glycine with four concentration levels on attached culture of Nannochloropsis oculata were investigated. The organic nitrogen source glycine was effective on improving not only adhesion biomass productivity but also adhesion rate. The maximum adhesion biomass productivity of 15.76 ± 0.52 g m?2 day?1 with adhesion rate of 76.67 ± 0.42 % was achieved with 18 mM glycine. To increase the lipid production, three lipid enhancing strategies were conducted afterwards, including nitrogen starvation, high light, and the combination of nitrogen starvation and high light. In nitrogen starvation situation, although the lipid content was greatly increased, the adhesion biomass productivity dropped probably due to the low cell viability. Increasing light intensity was effective on enhancing both adhesion biomass productivity and lipid content. The results indicated that nitrogen starvation was effective on improving both lipid content and adhesion rate when high light was applied. The maximum lipid yield of 4.32 ± 0.14 g m?2 day?1 with adhesion biomass productivity of 21.32 ± 0.65 g m?2 day?1, adhesion rate of 86.81 ± 0.10 % and lipid content of 20.24 ± 0.06 % was achieved with the combination strategy.  相似文献

In this study, hypersaline media were used for ocean cultivation of the marine microalga Tetraselmis sp. KCTC12432BP for enhanced biomass and fatty acid (FA) productivity. Hypersaline media (55, 80, and 105 PSU) were prepared without sterilization by addition of NaCl to seawater obtained from Incheon, Korea. The highest biomass productivity was obtained at 55 PSU (0.16 g L?1 day?1) followed by 80 PSU (0.15 g L?1 day?1). Although the specific growth rate of Tetraselmis decreased at salinities higher than 55 PSU, prevention of contamination led to higher biomass productivity at 80 PSU than at 30 PSU (0.03 g L?1 day?1). FA content of algal biomass increased as salinity increased to 80 PSU, above which it declined, and FA productivity was highest at 80 PSU. Ocean cultivation of Tetraselmis was performed using 50-L tubular module photobioreactors and 2.5-kL square basic ponds, closed- and open-type ocean culture systems, respectively. Culturing microalgae in hypersaline medium (80 PSU) improved biomass productivities by 89 and 152% in closed and open cultures, respectively, compared with cultures with regular salinity. FA productivity was greatly improved by 369% in the closed cultures. The efficacy of salinity shift and N-deficiency to enhance FA productivity was also investigated. Lowering salinity to 30 PSU with N-starvation following cultivation at 80 PSU improved FA productivity by 19% in comparison with single-stage culture without N-deficiency at 30 PSU. The results show that salinity manipulation could be an effective strategy to improve biomass and FA productivity in ocean cultivation of Tetraselmis sp.  相似文献

Microalgae are a promising feedstock for biofuel production. Lipid content in microalgae could be enhanced under nutrient depletion. This work investigated the effect of the nutrient on lipid accumulation in Ankistrodesmus sp. culture. Batch cultures were carried out using fresh BG11 medium, and after the harvest, the medium was reused for the next culture; this method was repeated two times. The maximum lipid productivity of 29.75 mg L?1 day?1 was obtained from the culture with the second reuse medium. In continuous cultures, Ankistrodesmus sp. was cultured in both fresh and modified BG11 mediums. The modified BG11 medium was adjusted to resemble the content of the first reuse medium. As a comparison between batch and continuous cultures, it was proven that the productivity in the continuous culture was better than in the batch, where the achievable maximum biomass and lipid were 188.30 and 38.32 mg L?1 day?1. The maximum lipid content of 34.22% was obtained from the continuous culture at a dilution rate of 0.08 day?1, whereas the maximum saturated and unsaturated fatty acid productivities of 79.96 and 104.54 mg L?1 day?1 were obtained at a dilution rate of 0.16 day?1.  相似文献

Ultrasonic treatment was firstly found to accelerate both biomass and hydrocarbon productivities of Botryococcus braunii algal cells cultured in the shake flasks. The most effective sonication strategy was to subject the cells to three 5-min ultrasonic treatments at a 4-day interval using a fixed frequency of 40 kHz and power of 240 W, and the ultrasound-treated algal cells showed the highest biomass productivity of 0.043 g L?1 day?1 and the highest hydrocarbon productivity of 13.1 mg L?1 day?1 among all ultrasound treatments tested. The improved productivity was proved to be mainly due to the enhancement of both endogenous indole-3-acetic acid (IAA) biosynthesis and membrane permeability in the ultrasound-stimulated algal cells. The efficient ultrasonic stimulation strategy also showed good performance for the algal culture in a 2-L airlift bioreactor. Together, these results not only illustrate the immense potential for an enhanced understanding on ultrasound-stimulated algal cells but also as provide a powerful process intensification method to improve the biomass and hydrocarbon productivity of B. braunii in practice.  相似文献

The biomass productivity of Scenedesmus obliquus was investigated outdoors during all seasons in solar tracked flat panel photobioreactors (PBR) to evaluate key parameters for process optimization. CO2 was supplied by flue gas from an attached combined block heat and power plant. Waste heat from the power plant was used to heat the culture during winter. The parameters pH, CO2, and inorganic salt concentrations were automatically adjusted to nonlimiting levels. The optimum biomass concentration increased directly with the photosynthetic active radiation (PAR) from 3 to 5 g dry weight (DW)?L?1 for a low PAR of 10 mol photons m?2 day?1 and high PAR of 40–60 mol photons m?2 day?1, respectively. The annual average biomass yield (photosynthetic efficiency) was 0.4?±?0.5 g DW mol?1 photons. However, biomass yields of 1.5 g DW mol?1 photons close to the theoretical maximum were obtained at low PAR. The productivity (including the night biomass losses) ranged during all seasons from ?5 up to 30 g DW m?2 day?1 with a mean productivity of 9?±?7 g DW m?2 day?1. Low night temperatures of the culture medium and elevated day temperatures to the species-specific optimum increased the productivity. Thus, continuous regulation of the biomass concentration and the culture temperature with regard to the fluctuating weather conditions is essential for process optimization of outdoor microalgal production systems in temperate climates.  相似文献

The present work evaluated biomass productivity, carbon dioxide fixation rate, and biochemical composition of two microalgal species, Phaeodactylum tricornutum (Bacillariophyta) and Tetradesmus obliquus (Chlorophyta), cultivated indoors in high-technology photobioreactors (HT-PBR) and outdoors both in pilot ponds and low-technology photobioreactors in a greenhouse in southern Italy. Microalgae were grown in standard media, under nitrogen starvation, and in two liquid digestates obtained from anaerobic digestion of agro-zootechnical and vegetable biomass. P. tricornutum, cultivated in semi-continuous mode in indoor HT-PBRs with standard medium, showed a biomass productivity of 21.0?±?2.3 g m?2 d?1. Applying nitrogen starvation, the lipid productivity increased from 2.3 up to 4.5?±?0.5 g m?2 d?1, with a 24 % decrease of biomass productivity. For T. obliquus, a biomass productivity of 9.1?±?0.9 g m?2 d?1 in indoor HT-PBR was obtained using standard medium. Applying liquid digestates as fertilizers in open ponds, T. obliquus gave a biomass productivity (10.8?±?2.0 g m?2 d?1) not statistically different from complete medium such as P. tricornutum (6.5?±?2.2 g m?2 d?1). The biochemical data showed that the fatty acid composition of the microalgal biomass was affected by the different cultivation conditions for both microalgae. In conclusion, it was found that the microalgal productivity in standard medium was about doubled in HT-PBR compared to open ponds for P. tricornutum and was about 20 % higher for T. obliquus.  相似文献

The effect of organic and inorganic nitrogen sources on Trichoderma reesei Rut-C30 cellulase production was investigated in submerged cultivations. Stirred tank bioreactors and shake flasks, with and without pH control, respectively, were employed. The experimental design involved the addition of individual organic nitrogen sources (soy peptone, glutamate, glycine and alanine) within a basal medium containing Avicel (i.e. micro crystalline cellulose) and ammonium sulphate. It was found that in the shake flask experiments, the highest cellulase activities (~0.1 ± 0.02 FPU ml?1) were obtained with media containing soy peptone (3–6 g l?1) and glutamate (3.6 g l?1). However, these improvements in the cellulase titers in the presence of the organic nitrogen sources appeared to be related to smaller changes in the pH of the medium. This was confirmed using stirred tank bioreactors with pH control. No significant differences were observed in the highest cellulase titers and the protein pattern (according to the SDS-PAGE) of supernatants from pH controlled stirred tank bioreactor cultivations, when different nitrogen sources were used in the medium. Here the cellulase activities (~1.0 ± 0.2 FPU ml?1) were also much greater (8–150 times) than in shake flask cultivation. Consequently, the addition of ammonium sulphate as sole nitrogen source to Avicel basal medium is recommended when performing cultivations in stirred tank bioreactors with strict pH controlled conditions.  相似文献

Biomass productivity was quantified for the marine macroalga Derbesia tenuissima cultivated outdoors at seven stocking densities from 0.25 to 8 g L?1 for 5 weeks. Total lipids and fatty acid quantity and quality was measured from samples that were freeze-dried, dried by oven (75 °C), food dehydrator (60 °C), or outdoor in the sun (40 °C) or shade (38 °C). Stocking densities of 0.25 to 2 g L?1 yielded the highest biomass productivities (>20 g dry weight m?2 day?1) with no effect on total lipid quantity (11 %), or fatty acid quantity (5.3 %) or quality at any density tested. However, there was an interactive effect of stocking density and drying technique, with a decrease of up to 40 % in polyunsaturated fatty acids in sun-dried compared to freeze-dried biomass. Notably, while fatty acid and biomass productivity may be inseparable in macroalgae, cultivation conditions have a significant carryover effect in the post-harvest delivery of high-quality bio-oils.  相似文献

In these studies, butanol (acetone butanol ethanol or ABE) was produced from concentrated lactose/whey permeate containing 211 g L?1 lactose. Fermentation of such a highly concentrated lactose solution was possible due to simultaneous product removal using a pervaporation membrane. In this system, a productivity of 0.43 g L?1 h?1 was obtained which is 307 % of that achieved in a non-product removal batch reactor (0.14 g L?1 h?1) where approximately 60 g L?1 whey permeate lactose was fermented. The productivity obtained in this system is much higher than that achieved in other product removal systems (perstraction 0.21 g L?1 h?1 and gas stripping 0.32 g L?1 h?1). This membrane was also used to concentrate butanol from approximately 2.50 g L?1 in the reactor to 755 g L?1. Using this membrane, ABE selectivities and fluxes of 24.4–44.3 and 0.57–4.05 g m?2 h?1 were obtained, respectively. Pervaporation restricts removal of water from the reaction mixture thus requiring significantly less energy for product recovery when compared to gas stripping.  相似文献

Energy consumption of municipal wastewater treatment plants can be reduced by the anaerobic pre-treatment of the main wastewater stream. After this pre-treatment, nitrogen can potentially be removed by partial nitritation and anammox (PN/A). Currently, the application of PN/A is limited to nitrogen-rich streams (>500 mg L?1) and temperatures 25–35 °C. But, anaerobically pretreated municipal wastewater is characterized by much lower nitrogen concentrations (20–100 mg L?1) and lower temperatures (10–25 °C). We operated PN/A under similar conditions: total ammonium nitrogen concentration 50 mg L?1 and lab temperature (22 °C). PN/A was operated for 342 days in a 4 L moving bed biofilm reactor (MBBR). At 0.4 mg O2 L?1, nitrogen removal rate 33 g N m?3 day?1 and 80 % total nitrogen removal efficiency was achieved. The capacity of the reactor was limited by low AOB activity. We observed significant anammox activity (40 g N m?3 day?1) even at 12 °C, improving the applicability of PN/A for municipal wastewater treatment.  相似文献

This study aims to investigate the triacylglycerol (TAG) productivity of Parachlorella kessleri grown under continuous illumination and to investigate its metabolism in simulated day/night cycles in order to estimate the feasibility of a large-scale production in outdoor solar photobioreactors. The strain was chosen for its ability to accumulate large amounts of triacylglycerol during nitrogen starvation. Several protocols of nitrogen starvation were tested in continuous illumination as well as in simulated day/night cycles. Sudden and progressive nitrogen starvation conditions have enhanced the TAG concentration and productivity of P. kessleri reaching up to 48 dry wt% and 4.4 × 10?3 kg m?2 day?1, respectively. Microalgal cell metabolism was significantly affected by the day/night illumination cycles. The energy-rich compounds (TAGs and carbohydrates) were accumulated by P. kessleri during the photoperiods and partly consumed during the dark to sustain the microalgae vitality. This TAG oxidation ultimately led to a 26% decrease in TAG productivity in cultures exposed to day/night cycles compared to ones exposed to continuous illumination of equal 24-h average photon flux density. The results can dictate the optimal time for harvesting cells for recovering the largest amount of TAGs.  相似文献
