1. The biliproteins C-phycocyanin and allophycocyanin were purified from the blue-green alga Anabaena variabilis by ammonium sulphate fractionation and gel filtration. 2. An assay procedure that enabled the proportion of the two pigments, present as a mixture, to be determined was devised by using the data provided by spectrophotometric analysis of the purified biliproteins. 3. The degree of association and relative proportions of the two pigments were analysed by the application of this procedure to the components separated by thin-layer gel filtration. 4. The C-phycocyanin/allophycocyanin ratio remained essentially constant in algal extracts prepared at various stages throughout the growth cycle or after growth under conditions of reduced illumination. 5. The behaviour of the C-phycocyanin aggregate species from Anacystis nidulans suggested that they were of appreciably lower molecular weight than those observed in extracts of Anabaena variabilis.  相似文献

Two methods for simulation of energy migration in the C-phycocyanin fragments of PBS were developed. Both methods are based on the statistical analysis of an excitation behavior in modeling complexes with a limited number (up to hundreds) of chromophores using the Monte-Carlo approach and calculation of migration rates for the system of linear balance equations. Energy migration rates were calculated in the case of C-phycocyanin of the blue-green algae Agmenellum quadruplicatum. The main channels of energy migration were determined in a monomer, trimer, hexamer, and in the rods consisting of 2-4 hexamers. The influence of the “screw” angle between two adjoining trimers of hexamer on the rates of energy migration and on its efficiencies in 1-4 hexamers was also estimated. The analysis was made for the average (random) and real orientation of chromophores in the C-phycocyanin. For both cases the optimal angle values were determined and the one for real C-phycocyanin structure was found to be very close (Δø ≤ 5°) to the optimal angle calculated.  相似文献

Summary The Feulgen-positive and envelope-free nucleoplasm ofAnabaena andGloeocapsa contains DNA filaments. When ultracentrifuged it is displaced between the maze of photosynthetic lamellae to form a continuous, flattened layer in the centripetal half of the cell. The absence of a mesosome and lack of attachment of the nucleoplasm to the plasmalemma make the method of duplication and separation of the genome to the progeny an enigma. In the centripetal end of ultracentrifugedAnabaena, the photosynthetic lamellae are somewhat stretched and sometimes broken. This condition suggests that the cortical positioned photosynthetic lamellae are tightly connected in several places along their surfaces to an undisplaced cortical cytoplasmic layer. It is uncertain whether or not they are also directly attached to the plasmalemma. A vesicle is often observed in the centripetal end of the cell; it probably represents for the most part, the contents of distorted intralamellar spaces. In ultracentrifugedGloeocapsa, the photosynthetic lamellae show little displacement. They appear attached, probably at their ends to the plasmalemma and, likeAnabaena andNostoc, they possess phycobilisomes which are not greatly affected by ultracentrifugation. Cells usually survive a centrifugal force greater than required to separate most nonliving protein systems. When subcultured, a redistribution of the displaced components usually occurs and growth is resumed.  相似文献

Summary Cell-free extracts capable of acetylene reduction and cyanide reduction have been prepared from heterocystous (Anabaena cylindrica) and non-heterocystous (Plectonema boryanum 594) blue-green algae. Extracts from Anabaena were obtained from cultures grown in blulk under aerobic conditions, while the Plectonema cultures were grown in bulk on nitrate-nitrogen, then washed free from nitrate and sparged with A/CO2 for 40 h after which time maximum nitrogenase activity was detected. The nitrogenases of both algae are similar and resemble in many respects nitrogenases from bacteria and legumes. Activity is located primarily in a 40,000xgx15 min supernatant fraction and the rate of C2H2 reduction observed is about 10 per cent of whole cell activity. ATP and a source of reducing power (Na2S2O4) are required for efficient functioning of the enzyme. ATP-dependent hydrogen evolution occurs, the extracts are cold labile and highly sensitive to oxygen and the oxygen inhibition is irreversible.  相似文献

Summary All of the three blue-green algae, Anabaena cylindrica, Mastigocladus laminosus and Nostoc muscorum are characterized by the presence of multi-layered envelopes (sheath, wall and plasma membrane), photosynthetic lamellae and a variety of intracellular granules. Sections of heterocysts of Anabaena cylindrica showed the presence of an internal membrane system as well as lamellae. An unusual feature of the structure of Nostoc muscorum was the presence of densely stained intracellular membranes or lamellae. The results emphasize the variability in appearance of the internal structure of the blue-green algae and point to the need for detailed investigations of the influence of change in physiological environment on the anatomy of these organisms.  相似文献

通过测定藻细胞密度,研究了5种红树植物木榄、秋茄、海漆、海芒果、小花老鼠簕水提物对赤潮藻球形棕囊藻和赤潮异弯藻的化感抑制效应,探讨了高温处理对化感抑藻效应的影响.研究结果表明,红树植物干粉水提物和新鲜组织水提物对两种赤潮藻均显示出显著的化感抑制作用.第5 d,红树植物干粉水提物对2种赤潮藻的抑制效果依次为:海漆>秋茄>小花老鼠簕>木榄>海芒果.新鲜组织水提物抑藻效果最强的红树植物是木榄和秋茄.秋茄和海漆水提物中的抑藻化感物质对高温相对不稳定,木榄和小花老鼠簕水提物中的抑藻化感物质对高温相对比较稳定.  相似文献

Three chlorophyll-protein complexes have been resolved from blue-green algae using an improved procedure for membrane solubilization and electrophoretic fractionation. One complex has a red absorbance maximum of 676 nm and a molecular weight equivalency of 255 000 ± 15 000. A second complex has an absorbance maximum of 676 nm, a molecular weight equivalency of 118 000 ± 8000, and resembles the previously described P-700-chlorophylla-protein (CPI) of higher plants and algae. The third chlorophyll-protein has a red absorbance maximum of 671 nm and a molecular weight equivalency of 58 000 ± 5000. Blue-green algal membrane fractions enriched in Photosystem I and heterocyst cells do not contain this third chlorophyll-protein, whereas Photosystem II-enriched membrane fractions and vegetative cells do. A component of the same spectral characteristics and molecular weight equivalency was also observed in chlorophyll b-deficient mutants of barley and maize. It is hypothesized that this third complex is involved in some manner with Photosystem II.  相似文献

A survey of the occurrence of toxic blooms of cyanobacteria in Finnish fresh and coastal waters was made during 1985 and 1986. Toxicity of the freeze-dried water bloom samples was tested by mouse-bioassay (i.p.). Forty-four per cent (83/188) of the bloom samples were found to be lethally toxic. Hepatotoxic blooms (54) were almost twice as common as neurotoxic ones (29). Anabaena was the most frequently found genus in toxic and non-toxic blooms and it was present in all neurotoxic samples. Statistical associations were found between hepatotoxicity and incidence of Microcystis aeruginosa, M. viridis, M. wesenbergii, Anabaena flos-aquae and Anabaena spiroides. Neurotoxicity was statistically associated with Anabaena lemmermannii, Anabaena flos-aquae and Gomphosphaeria naegeliana. Isolation of strains of cyanobacteria confirmed the occurrence of hepatotoxic and neurotoxic strains of Anabaena, as well as hepatotoxic strains of Microcystis and Oscillatoria species.Toxic blooms caused cattle poisonings at three different lakes during the study period. Toxic blooms also occurred in drinking water sources. Our study shows that toxic cyanobacteria are more common in Finnish lakes than would be expected on the basis of animal poisonings. The results of this study show the existence of toxic cyanobacteria in Finnish water supplies and the need for their continued study as agents of water based disease.  相似文献

Intergeneric transfer of streptomycin-resistance marker from a unicellular blue-green algaAnacystis nidulans to a filamentous blue-green algaAnabaena doliolum was demonstrated. Mutants ofA. nidulans resistant to streptomycin, occurring spontaneously or mutagenically-induced could be isolated easily. Naturally occurring streptomycin-resistant mutants ofA. doliolum could not be detected. Attempts at isolating such mutants either in nitrogen-free medium or in nitrate containing medium were unsuccessful. However, a streptomycin-resistant strain (recombinant) ofA. doliolum could be isolated in a mixed culture of streptomycin-sensitiveA. doliolum and streptomycin-resistantA. nidulans.  相似文献

AIMS: To partially purify and characterize bioemulsifiers produced by two new marine Halomonas species, TG39 and TG67, and to compare their emulsifying activities with those of commercial emulsifiers. METHODS AND RESULTS: The production of emulsifiers HE39 and HE67 was achieved from glucose-supplemented marine broth, and recovered by cell removal, concentration by ultrafiltration, precipitation with salt and ethanol, and lyophilization. Purification and chemical analysis revealed both emulsifiers to be glycoproteins of high molecular weight with a notably high content of protein and uronic acids. Physical characterization showed both glycoproteins to effectively emulsify a wide range of food oils under both neutral and acidic pH conditions and withstand acid and high temperature treatment. CONCLUSIONS: The emulsifying activities of these two new glycoprotein emulsifiers were comparable and, under certain conditions, superior to those produced by commercial emulsifiers tested (xanthan gum, gum arabic and lecithin). They show the highest reported emulsifying activities derived from a Halomonas species. SIGNIFICANCE AND IMPACT OF THE STUDY: These strains, and the emulsifiers produced, appear to be promising candidates for further development in applications requiring emulsifiers that are natural and compatible to the existing commercial emulsifiers.  相似文献

Various historical eras in the distant as well as the recent past of amphibian embryology are briefly reviewed. The concepts which emerged from the early years matured, then were laid to rest for several decades. A resurgence, driven by key discoveries with peptide growth factors, and fueled by modern molecular biology methods, is underway. The future for several amphibian research projects should be promising since interest in basic concepts remains strong, and application of frontier methodologies is yielding novel findings.  相似文献

通过室内实验研究了不同温度条件下主要水华藻类——铜绿微囊藻(Microcystis aeruginisa)和巨颤藻(Oscillatoria princeps)的生长和种间竞争。结果表明:无论在纯培养体系还是混合培养体系中,微囊藻在25℃下生长最好,颤藻在30℃下生长最好;温度对藻类的种间竞争抑制参数能够产生明显影响,在20℃、25℃、30℃3个温度下,颤藻对微囊藻的竞争抑制参数随温度的升高而升高,30℃时颤藻对微囊藻的竞争抑制参数分别是20℃和25℃时的1.42倍和1.13倍,微囊藻对颤藻的竞争抑制参数则表现为25℃>30℃>20℃,25℃时微囊藻对颤藻的竞争抑制参数分别是20℃和30℃时的1.54倍和1.21倍,在20℃、25℃、30℃3个温度下,微囊藻对颤藻的竞争抑制参数均大于颤藻对微囊藻的竞争抑制参数,说明在各试验温度下,微囊藻对颤藻的抑制能力均大于颤藻对微囊藻的抑制能力;根据Lotka-Volterra竞争模型中的两物种竞争结果可初步判断,20℃和25℃温度下,微囊藻在竞争中取胜,30℃下,微囊藻和颤藻可以实现不稳定共存。  相似文献
