Composition and seasonal patterns of the fern spore bank were compared to the surface vegetation of grasslands on dolomite rock in Hungary. Viability and potential dormancy of spores were tested through storage experiments. Although Asplenium ruta-muraria L. was the only species found at the study sites, five others, probably originating from air-borne spores from nearby areas, emerged from the soil samples. Considerable seasonal variability was detected in the number of prothallia emerging from soil samples from different sampling dates, with a peak after spore dispersal. The increased number of emerging prothallia after 1 year of storage suggests that a part of the spores stored in the soil samples were presumably dormant. Investigations on the dormancy of fern spores might be of great interest, especially in species adapted to seasonally unfavourable habitats.  相似文献

The differential light scattering characteristics of single spores of Bacillus sphaericus and Clostridium filamentosum were measured and recorded for several hundred different spores using an argon-ion laser source (λ=514.5 nm) and a single-particle light-scattering photometer. Typical recorded light-scattering patterns were interpreted analytically by adjusting the parameters of a spherically symmetrical model to the data using a least squares fitting procedure. The size fluctuations of the cores were very small which suggests that their structure may well be identical for different spores of the same species. The greater variability of the spore surface thickness seems to indicate that these components are loosely defined during sporulation and will certainly vary with growth conditions. Based on the observed lack of free water within the cores, a hypothesis is presented that dipicolinic acid (DPA) acts as a filler material in the core to prevent free water penetration.  相似文献

The effects of heat, CaCl2, and ethanol on activation of Bacillus spores were determined by monitoring the absorbance decrease during germination in inosine. Bacillus cereus T, B. subtilis A and B. megaterium QM B1551 spores were activated by heat- and CaCl2-treatments. Ethanol activated B. megaterium and B. subtilis spores yet did not activate B. cereus spores. CaCl2- and ethanol-activations were less effective than heat-activation as judged by optimal germination rates and germination extents. The presence of CaCl2 during heat-treatment inhibited heat-activation of all three Bacillus spores without affecting viability or dipicolinic acid content of the spores. The electrophoretic patterns of coat plus outer membrane proteins extracted from Bacillus spores treated with CaCl2 and heat in the presence of CaCl2 were similar to each other and were distinctively different from the patterns of proteins from unactivated spores or the spores treated with heat and/or ethanol.  相似文献

A method was developed to deposit Bacillus subtilis spores via aerosolization onto various surface materials for biological agent decontamination and detection studies. This new method uses an apparatus coupled with a metered dose inhaler to reproducibly deposit spores onto various surfaces. A metered dose inhaler was loaded with Bacillus subtilis spores, a surrogate for Bacillus anthracis. Five different material surfaces (aluminum, galvanized steel, wood, carpet, and painted wallboard paper) were tested using this spore deposition method. This aerosolization method deposited spores at a concentration of more than 10(7) CFU per coupon (18-mm diameter) with less than a 50% coefficient of variation, showing that the aerosolization method developed in this study can deposit reproducible numbers of spores onto various surface coupons. Scanning electron microscopy was used to probe the spore deposition patterns on test coupons. The deposition patterns observed following aerosol impaction were compared to those of liquid inoculation. A physical difference in the spore deposition patterns was observed to result from the two different methods. The spore deposition method developed in this study will help prepare spore coupons via aerosolization fast and reproducibly for bench top decontamination and detection studies.  相似文献

The swimming behavior of spores of the brown alga Hincksia irregularis was analyzed using computer-assisted motion analysis. We distinguished five main swimming patterns: straight paths, search circles, orientation, gyration, and wobbling. We suggest different functional values for the individual swimming patterns. Straight paths, search circles, and orientation are different but all may be important in small-scale movements in the benthic boundary layer. As such, they could enable a spore to find a suitable microenvironment for germination and growth. Gyration occurs during the initial reversible phase of adhesion that can lead to settlement. Wobbling is typical for irritated or mechanically damaged spores and does not seem to be a typical pattern associated to settlement. The dominant swimming patterns changed with spore age (10 ± 5 to 60 ± 5 min of spore age), with young spores mainly swimming in straight paths and search circles and older spores in orientation and gyration. This change in swimming patterns can be quantified by speed (decrease over time) and rate of change of direction (increase over time). Based on these results, we suggest that computer-assisted motion analysis of the swimming behavior of H. irregularis spores can be used to develop bioassays with both ecological and technological relevance.  相似文献

A transposon-based split-marker protocol was used to produce insertional mutations in the fadA ortholog of the maize anthracnose pathogen Colletotrichum graminicola. The mutants grew more slowly in culture, produced fewer oval spores, produced fusiform rather than falcate phialospores, lost their normal clockwise spiral growth pattern in culture, and were significantly reduced in their pathogenicity to maize stalks and leaves. The differential effect of the fadA mutation on oval spore versus phialospore production suggests that there are differences in the signaling pathways that regulate these two types of sporulation. It has been suggested that oval spores function in anthracnose lesion extension. In maize stalks, production of oval spores appeared to be relatively unaffected in the mutant strains, but production of vegetative hyphae and elongation of primary lesions were both reduced. This suggests that vegetative hyphae play a more important role than oval spores in primary lesion development. However, production of discontinuous secondary lesions in maize stalks infected by mutant strains did not appear to be seriously affected, and thus oval spores may play a more important role in that process.  相似文献

The humoral immune responses of grouper Epinephelus akaara to a natural infection with Glugea epinephelusis was studied by ELISA utilizing intact mature spores as the coated antigen. Results showed that a specific humoral immune response was elicited, but the intensity of infection (in terms of the number of cysts) was not related to the antibody level in naturally infected hosts. The differences in the antigenicity of intact mature spores and soluble spore proteins derived from cracked mature spores were also analyzed. Results suggested that similar antigen epitopes existed between the 2 groups. Additionally, antigen component patterns and the distribution of antigen with immunogenicity were investigated by using the western blot and the immunofluorescent antibody technique (IFAT). The new parasitic microsporidium has specific polypeptide patterns comparable to the reported fish microsporidians. The main antigenic substances are concentrated on the surface of spores, and are mostly located on the anterior and posterior end of the spore bodies. Most surface components of the G. epinephelusis spores are soluble. The potential role of the surface components in initiating infection was also discussed.  相似文献

The surface hydrophobicity of 12 strains of Bacillus spp. was examined in a hexadecane-aqueous partition system. Mature and germinated spores of Bacillus megaterium QM B1551 transferred to the hexadecane layer, while vegetative and sporulating cells did not. Wild-type spores were more hydrophobic than spores of an exosporium-deficient mutant of B. megaterium QM B1551, although the mutant spores were shown to be hydrophobic to some extent by using increased volumes of hexadecane. This result suggests that the exosporium is more hydrophobic than the spore coat and that the surface hydrophobicity of spores depends mainly on components of the exosporium. The surface hydrophobicity of spores of nine other species of Bacillus was also examined, and spores having an exosporium were more hydrophobic than those lacking an exosporium. Thus measurement of the hydrophobicity of spores by the hexadecane partition method may provide a simple and rapid preliminary means of determining the presence or absence of an exosporium.  相似文献

The fine structure of the plasma membrane in spores of the microsporidium Nosema algerae, a pathogen of mosquitoes, was examined in the resting condition and after the spores were stimulated to germinate in vitro. Slow penetration of resin caused collapse of the germinated spores. Thin sections of germinated spores showed peculiar membrane infoldings that were never found in ungerminated samples. Analogous germination-dependent configurations of the plasma membrane were observed in freeze-fractured preparations of spores either fixed and impregnated with glycerol prior to freezing, or rapidly frozen with liquid propane while in the process of germination. In every case, the replicas presented germinated spores with indentations in the protoplasmic face of the plasma membrane, and apparently complementary blunt spines on the external face, that were absent in ungerminated spores. It suggests that these alterations of the plasma membrane result from a structural adjustment to a spontaneous contraction of the spore case after germination. We discuss this interpretation with regard to conflicting views on the nature of such morphological features.  相似文献

The asci of Metschnikowia species normally contain two ascospores (never more), raising the question of whether these spores are true meiotic products. We investigated this problem by crossing genetically-marked strains of the haploid, heterothallic taxa Metschnikowia hawaiiensis, Metschnikowia continentalis var. continentalis, and M. continentalis var. borealis. Asci were dissected and the segregation patterns for various phenotypes analyzed. In all cases (n = 47) both mating types (h+ and h-) were recovered in pairs of sister spores, casting further uncertainty as to whether normal meiosis takes place. However, the segregation patterns for cycloheximide resistance and several auxotrophic markers were random, suggesting that normal meiosis indeed occurs. To explain the lack of second-division segregation of mating types, we concluded that crossing-over does not occur between the mating-type locus and the centromere, and that meiosis I is tied to spore formation, which explains why the number of spores is limited to two. The latter assumption was also supported by fluorescence microscopy. The second meiotic division takes place inside the spores and is followed by the resorption of two nuclei, one in each spore.  相似文献

The fine localization of mineral matter in spores of Bacillus megaterium and Bacillus cereus was studied by the technique of microincineration adapted for use with the electron microscope. The specimens, which included intact and thin-sectioned spores as well as shed spore coats, were burned either in the conventional way at high temperature or by a new technique using electrically excited oxygen at nearly room temperature. The ash residues were examined by bright field, dark field, and diffraction in the electron microscope and also with the phase contrast microscope. In some cases, the specimen was previewed in both microscopes before incineration. The results do not support a previous report that the mineral elements of the spore are confined to a peripheral layer, but rather indicate that the spore core as well as the coat are mineral-rich. The cortex may be deficient in minerals, but the possibility of artifact prevents a clear decision on this point. Incinerated B. megaterium spores show a highly ordered fine structure displaying 100 A periodicity in the ash of the middle layer of the coat. The nature of this structure is discussed, as is the technique which demonstrated it. The fine definition of the ash patterns, particularly those obtained with the low-temperature, excited-oxygen technique, suggests that microincineration may be generally useful in the study of fine structure.  相似文献

Unique patterns of biomarkers were reproducibly characterized by matrix-assisted laser desorption ionization (MALDI)-mass spectrometry and were used to distinguish Bacillus species members from one another. Discrimination at the strain level was demonstrated for Bacillus cereus spores. Lipophilic biomarkers were invariant in Bacillus globigii spores produced in three different media and in B. globigii spores stored for more than 30 years. The sensitivity was less than 5,000 cells deposited for analysis. Protein biomarkers were also characterized by MALDI analysis by using spores treated briefly with corona plasma discharge. Protein biomarkers were readily desorbed following this treatment. The effect of corona plasma discharge on the spores was examined.  相似文献

Pathogens and other symbiotic fungi that infect above‐ground plant parts commonly disperse as airborne spores. Here we present diel patterns of the density of airborne fungal spores in the canopy and understory of a tropical rain forest. Spores were 52‐fold more abundant in the understory than in the canopy. Additionally, spores were 5‐ to 35‐fold more abundant at night than during the day, associated with environmental conditions conducive to germination and plant infection.  相似文献

Spores of a haplosporidan infecting Teredo navalis Linnaeus have been described as morphologically indistinguishable from spores of Haplosporidium nelsoni . To test the hypothesis that these organisms are conspecific, a colloidal gold immunoassay was used to compare antigenic characteristics of the spores from both hosts. Rabbit antibody to formalin-fixed spores from T. navalis was tested against paraffin sections of Crassostrea virginica infected with spores of H. nelsoni and against paraffin sections of infected 7". navalis . Application of primary antibody was followed by addition of affinity purified goat anti-rabbit IgG coaled on 5-nm colloidal gold particles. The reaction was enhanced by precipitation of metallic silver; a positive reaction appeared as a dark brown to black signal at the site of each antigen-antibody complex. Haplosporidium nelsoni spores did not react when assayed with the antibody made to spores from T. navalis . Spores from infected T. navalis tissue reacted positively with rabbit antibody. This result indicates that the spores from the 2 hosts are antigenicaliy distinct and suggests that they are different species.  相似文献

Protein patterns from mycelial extracts of Streptomyces griseus strains No. 45H and No. 52-1 were studied by one dimensional gradient PAGE with 20 cm run. The results are reproducible, the protein patterns are the same for the same developmental stage in a given strain. There are well-defined characteristic changes of the protein patterns during the life cycle. The proteins of spores show conspicuous differences compared to protein patterns of the old (72 h) mycelia in the same culture. There is no difference between protein patterns of spores from submerged and from solid media. The protein patterns of two closely related Streptomyces griseus strains significantly differ from each other. After pulse labelling with (14C)-labelled protein hydrolyzate for 40 min, the specific activities of the proteins show considerable differences. There are characteristic bands with low specific activity and others with high incorporation. The fluorograms of the 16, 40 and 64 h cultures show that different proteins are being synthesized intensively at different ages of the culture.  相似文献

A wild-type strain of Streptomyces griseus forms spores both on solid media (aerial spores) and in liquid culture (submerged spores). Both spore types are highly resistant to sonication, but only aerial spores are resistant to lysozyme digestion. Electron micrographs suggest that lysozyme sensitivity may result from the thinner walls of the submerged spores. Studies of the life cycle indicate that neither streptomycin excretion nor extracellular protease activity is required for sporulation: the analysis of mutants, however, suggests that antibiotic production may be correlated with the ability to sporulate. A method was devised to induce the rapid sporulation of S. griseus in a submerged culture. This method, which depends on nutrient deprivation, was used to determine that either ammonia or phosphate starvation can trigger sporulation and that the enzyme glutamine synthetase may be useful as a sporulation marker after phosphate deprivation.  相似文献
