Activity changes in alanine aminotransferase (glutamate-pyruvatetransaminase, GPT) in exapanded fifth leaves of Lolium lemulentumL. were studied withn plants exposed to 8-h and 16-h photopepriodsat 20°C. Two phases were detected. The first was daylengthindependent and characterized by a rising activity either ona fresh-weight or protein basis, whilst the second phase consisitedof a rapid fall to a much lower activity during the course ofa few days. This effect was promoted by 16-h/20°C conditions.Using variable exposure to long days and light-break treatmentsit was shown that the GPT deporessions did not relate directlyto floral induction but was it response to increased incidentlight energy. Comparison with asparate aminostransferase, acidphosphatase, malate dehydrogenase, and peroxidase suggestedthat GPT activity could be used as a sensitive indicator senescence.  相似文献

Initial studies have indicated that Tagetes minuta achenes haveboth a temperature and a light requirement for germination.Temperatures tested were 10, 20, 25, 30 and 35 °C. Germinationwas optimal at 25 °C under white light conditions. Underthese conditions 100 per cent of achenes germinated within 7days of imbibition. There was no germination at 10 or 35 °Ceither in the light or in the dark. Achenes imbibed and incubatedat 35 °C for 4 days showed no visible signs of germinationbut on transfer to 25 °C, 100 per cent of these achenesgerminated within 24 h. Furthermore, achenes given this hightemperature (35 °C) treatment could be dried at 25 °C,re-imbibed at 25 °C and again 100 per cent of achenes germinatedwithin 24 h of re-imbibition. This rapid germination responsefollowing removal from the high temperature regime could alsobe induced by transfer to temperatures of 20 °C or 20 °C(16 h) alternating with 10 °C (8 h). The effects of single, combined and sequential application ofgibberellic acid (GA), chlormequat and cytokinins on the formationof anlagen, tendrils and inflorescences were studied in grapevines(cv. Muscat of Alexandria) grown with natural illumination athigh temperature (30 °C day to 25 °C night) and at lowtemperature (21 °C day to 16°C night or 18 °C dayto 13 °C night). GA promoted the formation of anlagen andgrowth of tendrils regardless of temperature, but inhibitedinflorescence production. Chlormequat had the opposite effecton anlagen formation and tendril growth and promoted inflorescenceformation from pre-formed anlagen or from tendril initials.While low temperature is normally unfavourable for inflorescenceformation, this was induced by chlormequat even at low temperature,but only with summer light conditions. Cytokinin application to plants pre-treated with chiormequatcaused tendrils to grow into inflorescences regardless of temperatureregimes. Moreover, shoot primordia were also formed in place8oftendrils in cytokinin treated plants even without chlormequatpre-treatment. The effects of photoperiod, light quality and a single applicationof gibberellic acid (GA3) on the development of the main-stemapex in Clipper barley are reported. In 16 and 24 h days spikeletinitiation was rapid but extended over a short period whereasin 8 h photoperiods both spikelet initiation and developmentwere slower but occurred for a much longer time. Initiationalways stopped when the anther primordia were clearly visiblein the most advanced spikelet. Daylength extensions with lowintensity incandescent light were most effective when they followedrather than preceeded the 8 h period of high light intensity.Plants grown in 8 h high intensity followed by 8 h low intensitylight initiated spikelets almost as rapidly as those grown in16 h high intensity light. Thus, the effects of daylength onspikelet production were primarily mediated through photoperiodicallycontrolled processes rather than through photosynthesis andassimilate supply. The effects of applied GA3 were long livedand greatest in short days where the rates of both spikeletinitiation and development were promoted. Flower buds of Viscaria candida, excised at 1–2 mm inlength, have been grown to maturity in sterile culture, oftenproducing normal flower parts. The calyx and the corolla developedunder a wide range of conditions provided gibberellic acid waspresent. The development of the ovary and of the ovules tendedto be irregular and no consistent effects of treatment couldbe obtained. Pollen production was promoted by a temperatureof 15 °C as opposed to one of 25 °C, and by the additionof gibberellic acid to the medium; it was inhibited by the additionof kinetin or hydrolysed casein to the medium, and by growthin continuous darkness as compared with that in 8 or 16 h oflight daily.  相似文献

Potato (Solanum tuberosum L.) plants were grown under long days(LD) of 18 h before a subset of the plants was transferred to10-h photosynthetic periods with either a dark night (SD) oran 8-h dim photoperiod extension with incandescent lamps (DE).Temperature was constant at 21 °C. Leaves were sampled atthe beginning and end of the high density light period for starchanalyses. Potato leaves accumulated starch more rapidly underSD than under LD; and this difference continued after a secondmajor sink, the tuber, began to develop. Starch accumulationover 10 h in SD leaves was three times higher than in LD leaves,even after 17 d of treatment. By this time SD gave higher wholeplant relative growth rates than LD, and the tuber mass of SDplants exceeded 30% of their total plant biomass. The DE treatmentresulted in starch accumulation intermediate to the LD and SDtreatments. Genotypes likewise differed: the earlier genotype,more strongly induced to tuberize, had higher leaf starch accumulationthan the later genotype. The effects of photoperiod and genotypewere also present when potatoes were grown at 27 °C, a temperatureunfavourable for tuberization under LD. The effects of day-length and temperature on flowering and dormancyinduction were studied in Anemone coronaria L., with plantsraised either from corms or achenes. An Israeli hybrid sourcewas used (de Caen cv. Hollandia x Israeli wild type). Dormancy onset is characterized by the cessation of foliageleaf production, the appearance of leaf scales protecting theperennating bud, and leaf senescence. Dormancy was induced byhigh temperature and long days but increasing temperatures (from17/12 °C to 32/12 °C) induced earlier dormancy thanprolonging the photoperiod (range 8–16 h). A significant(P = 0.01) interaction was found between these factors, withsmaller photoperiodic effects the higher the temperature. At22/17 °C the critical day-length for dormancy inductionwas between 11 and 12 h. The transition from the vegetative to the reproductive stageappears to be an autonomous process that occurs with developmentin plants raised from either corms or achenes and does not requireenvironmental induction. Photo- and thermoperiodic effects onflowering were indirect, being mediated through their influenceon dormancy induction. The response of the germination of seeds of Barbarea vema (Mill.)Aschers, Brassica chinensis L., Brassica juncea (L.) Czern.& Coss., Brassica oleracea L. var. gongylodes L., Camelinasaliva (L.) Crantz, Eruca saliva Mill., Lepidium sativum L.,Nasturtium officinale R. Br., and Rorippa palustris (L.) Besserto white fluorescent light of different photon flux densitiesapplied for different daily durations in a diurnal alternatingtemperature regime of 20 °C/30 °C (16 h/8 h) was quantifiedby linear relations between probit percentage germination andthe logarithm of photon dose, the product of photon flux densityand duration. The low energy reaction, in which increasing dosepromotes germination, was detected in all the seed populationsbut in Barbarea vema and Brassica Juncea the lowest photon doseapplied (10–5–2 and 10–5 7 mol m–2 d–1,respectively) was sufficient to saturate the response. Comparisons,where possible, between photoperiods demonstrated reciprocity,i.e. germination was proportional to photon dose irrespectiveof photoperiod, for the low energy reaction in Brassica oleracea(1 min d–1 to 1 h d–1), Camelina saliva (1 min d–1to 8 h d–1), Eruca saliva (1 min d–1 to 24 h d–1),Lepidium sativum (I min d–1 to 8 h d–1) and Rorippapalustris (1 min d–1 to 8 h d–1), but not in Brassicachinensis and Nasturtium officinale. The high irradiance reaction,in which increasing dose inhibits germination, was detectedin Barbarea vema, Brassica chinensis, Brassica juncea, Brassicaoleracea, and Camelina saliva. The minimum dose at which inhibitionwas detected was lO–0–3 mol m–2 d–1.These results are discussed in the context of devising optimallight regimes for laboratory tests intended to maximize germination The response of germination to photon dose was also quantifiedwith 3 x 10–4 M GA2, co-applied (Brassica chinensis, Camelinasaliva, and Lepidium sativum) and with 2 x 10–2 M potassiumnitrate co-applied (Brassica chinensis). In the latter casepotassium nitrate had no effect in the dark and inhibited germinationin the light, but GA2, promoted germination substantially inall three species. Variation amongst seeds in the minimum photondose required to stimulate germination was not affected by co-applicationof GA2, in Brassica chinensis and Camelina saliva, whereas seedsof Lepidium salivum showed a narrower distribution of sensitivitiesto the low energy reaction in the presence of GA2 Barbarea vema (Mill.) This study was carried out to elucidate the carbon budget inyoung tomato plants in photosynthetic light periods of 8, 12and 16 h after being acclimated to an 8-h light period. Thephotosynthetic rate was nearly constant for 16 h: thereforethe amount of 14C fixed was proportional to the light period(13·77 mg C per 8 h, 20·2 mg C per 12 h, 30·5mg C per 16 h). The amounts exported, lost by respiration and accumulated whenexpressed as percentages of the carbon fixed in the day differedlittle between the light periods. The leaf continued to exportcarbon at a nearly constant rate during the light periods and,for example, exported approximately twice as much during a 16-hperiod as in an 8-h light period, even though it was not acclimatedto the long light period. The amount of starch breakdown affected the amount of carbonexported and carbon lost by respiration at night, although itwas not sufficient to account for these losses altogether. Thepossible roles of carbon accumulation and respiration are alsodiscussed. Effectsof pre-treatment temperature on germination responses to far-redlight and on dark germination in the red light-requiring upperseeds.—J. exp. Bot. 36: 1465-1477. Red light (R) responsiveness in R-requiring upper cocklebur(Xanthium pennsylvanicum Wallr.) seeds changed in differentpatterns during a soaking period at different temperatures.At temperatures above 23°C, the responsiveness increasedand then decreased. At lower temperatures (3–18°C),however, it continued to increase throughout an experimentalperiod. The lower temperatures caused germination in the subsequentdark at 33°C, regained the R responsiveness and acquiredthe dark germinability when subsequently exposed to 8°C,to an extent proportional to the duration of the chilling. Far-red (FR) was inhibitory to germination in an earlier soakingperiod at lower temperatures, but its effect gradually decresed,and finally turned promotive. The negative FR response was repeatedlycontrolled by the following R irradiation. However, the positiveFR response was enhanced by an immediate R irradiation, andFR/R reversibility occurred after the second FR. In contrastto the R responsiveness and dark germinability, the positivegermination response to FR was not induced by soaking at 3°C,in which the growth of the axial tissue as a photoreceptivesite did not occur at all. Similarly, it was not manifestedwhen the seeds soaked at 33°C were subsequently subjectedto 8°C. The effects of photoperiod, nitrogen nutrition and temperatureon inflorescence initiation and development in onion cv. Rijnsburgerand cv. Senshyu Semi-globe Yellow were studied in controlledenvironments. Rates of inflorescence initiation were estimatedusing the data for leaf numbers formed prior to flower formationand the rates of leaf initiation. At 9 °C inflorescenceinitiation was accelerated by long photoperiods particularlyfor cv. Rijnsburger where the average time for initiation was86 days in 8 h and 38 days in 20 h photoperiods. Initiationwas as rapid at 12 °C as at 9 °C but was slower at 6°C. A reduction in the nitrate concentration in the nutrientsolution from 0.012 to 0.0018 M greatly accelerated inflorescenceinitiation particularly in photoperiods and temperatures notconducive to rapid initiation. Cv. Senshyu initiated more slowlythan cv. Rijnsburger and was less sensitive to photoperiod andnitrogen level. The development rate of inflorescences afterinitiation was accelerated by long photoperiods and increasesin temperature from 6 to 12 °C but was retarded by the lowernitrogen level. Mesophyll resistance to photosynthetic carboxylation (r'm) wasused as a criterion for leaf integrity. It was measured, at25 °C, in the light, before and after periods of high temperature(3 h at 38 °C) in the dark. During the high temperatureperiods, respiration (RD) of attached leaves of Xanthium strumariumwas suppressed from 27%-36% by either low [O2] (1.04% or 0.21%v.v.) or high [CO2] (840 µl 1–1) in the ambientair. Neither treatment affected rates of RD or photo-respirationduring the second period at 25 °C. There was no significant increase of r'm when RD was not suppressedduring the high temperature treatment. When RD was suppressedat high temperatures, r'm increased from about 3s cm–1before, to about 26 s cm–1 after the high temperaturetreatment. The increase depended upon the degree of suppression. It is concluded that increased RD at high temperature in Xanthiumleaves is partly the result of an increase of energy demandingmaintenance. The subsequent rate of carbon dioxide fixationis reduced when this increase of maintenance-induced respirationis inhibited.  相似文献

Seeds of barley (Hordeum vulgare L.) and mung bean (Vigna radiata(L.) Wilczek), with orthodox seed storage behaviour, were imbibedfor between 8 h and 96 h at 15 °C and 25 °C, respectively,while barley seeds were also maintained in moist aerated storageat 15 °C for 14 d. These seeds and seedlings, together withcontrols, were then dried to various moisture contents between3% and 16% (wet basis) and hermetically stored for six monthsat —20°C, 0°C or 15°C. In both species, neitherdesiccation nor subsequent hermetic storage of the control lotsresulted in loss in viability. The results for barley seedsimbibed for 24 h were similar to the control, but desiccationsensitivity increased progressively with duration of imbibitionbeyond 24 h in barley or 8 h in mung bean; these treatmentsalso reduced the longevity of the surviving seeds in air-drystorage. Loss in viability in barley imbibed for 48 h was mostrapid at the two extreme seed storage moisture contents of 3·6%and 14·3%, and in both these cases was more rapid at15 °C than at cooler temperatures. Similarly, for mung beanimbibed for 8 h, loss in viability was most rapid at the lowest(4·3%) moisture content, but in this case it was morerapid at –20 °C than at warmer temperatures. Flowering requirements of four Norwegian populations of Phleumalpinum were studied in controlled environments. A dual inductionrequirement was demonstrated in all populations. Inflorescenceinitiation had an obligatory requirement for short days (SD)and/or low temperature, while culm elongation and heading wereenhanced by long days (LD) and higher temperatures. At 3 and6 °C primary induction was almost independent of photoperiod,whereas SD was more effective than LD at higher temperatures.The critical temperature for primary induction was about 15°C in SD and 12 °C in LD. Saturation of induction required12 weeks of exposure to inductive conditions, although someheading and flowering took place with 6 weeks exposure to optimalconditions (9 °C/SD). Inflorescence development also tookplace in 8 h SD although it was delayed and culm elongationwas strongly inhibited compared with LD conditions. To examine flower opening and closing of aPortulacahybrid, flowerbuds were placed in darkness for 12 h (2030–0830 h) at20 °C and then exposed to various light-temperature conditions.Flower buds exposed to light at 25, 30 or 35 °C opened within1 h, and wilted 10–14 h later. Flower buds exposed tolight at 20 °C started to open after 4 h but opened slowlyand not completely. Flower buds subjected to 25, 30 or 35 °Cin darkness also opened rapidly, but did not reach full opening.Flowers opened at 30 °C in light, and partially closed andopened repeatedly in response to cycles of a 2-h exposure to20 °C and a 2-h exposure to 30 °C at any time between1000 to 1600 h. Similar phenomena were observed when the flowersopened at 30 °C in light and then were subjected to darknessand light alternately at 30 °C, although the effect of lightwas less obvious than that of alternating temperature. Floweropening and closing were not affected by relative humidity.These results indicate that a rise in temperature is requiredfor rapid flower opening in the buds kept at 20 °C, andthat light intensifies the effect of high temperature. Exposureto light without a temperature change delayed and slowed floweropening which was never complete. The involvement of an endogenousrhythm in flower opening byPortulacais indicated. One cultivar and one land-race of faba bean were subjected to18 potentially vernalizing pre-treatments (constant temperaturesof 1, 5 or 9 °C factorially combined with photoperiods of8 or 16 h d–1 for 10, 30 or 60 d), and then transferredinto four different growing regimes (‘day’/‘night’temperatures of 18/5 °C or 24/13 °C factorially combinedwith photoperiods of 11 or 16 h d–1). Control plants weregrown entirely in the latter four regimes. The times from sowingto appearance of first open flowers were recorded for all plants.Control plants of the land-race Zeidab Local flowered soonerin long days and in the warmer regime. Pre-treatment reducedthe subsequent time to flower in the four growing-on regimesbut most of the variation in the total time to first flowerfor the pre-treated plants was accounted for by differencesin the combined photothermal time accumulated in the two successiveenvironments - which was predicted by a simple photothermalmodel. Thus, there was neither a specific low-temperature nora short-day vernalization response in this accession. Similarly,no true low-temperature or short-day vernalization responsewas detected in the cv. Maris Bead. However, this UK cultivarflowered later than predicted in the 24/13 °C regime, indicatingthat the 24 °C ‘day’ temperature was supraoptimal.Delays to flowering at 24/13 °C were, however, less evidentwhen plants were grown in long days or following prolonged (30–60d) pre-treatments at cool temperatures. A comparison was made of effects of ‘low’ and ‘high’soil temperature (LST and HST, about 9 and 21·5 °Crespectively) on shoot growth of Norwegian and Portuguese populationsof Dactylis glomerata. In experiments lasting 8 short days (SD,8 h photoperiods) LST decreased leaf extension more markedlyin the Portuguese population. No differential effect of LSTon leaf growth was recorded in experiments lasting 20 or 21SD or in experiments of 8 d duration in long days (LD, 16 hphotoperiods). Since the meristem and region of cell extensionis close to the soil surface LST could directly influence bothroot and shoot growth. The application of gibberellic acid enhanced leaf extension,particularly in plants grown at HST. 6-Benzylaminopurine tendedto decrease leaf length. Extractable gibberellin levels wererelatively low after 8 SD in shoots of both races grown at LST.Cytokinin levels increased at LST, more so in shoots of Portugueseplants which sustained the greater reduction in leaf extension. The data suggest that LST may reduce the production of endogenousgibberellins important for leaf growth in Dactylis glomerata.Cytokinins are probably necessary for growth processes but theirlevel may reflect, rather than direct, the rate of leaf growth.  相似文献

Germination responses to light were studied in the upper andlower seeds of cocklebur (Xanthium pennsylvanicum Wallr.). Thelower seed was dark-germinating and negatively photoblastic;the upper one had a red-light (R) requirement and was positivelyphotoblastic. Germination of the lower seeds was inhibited bya prolonged single irradiation with R, blue (B) or far-red (FR)light applied during imbibition. The maximal inhibitory effectof a single irradiation occurred 9 h and 13 h after the startof soaking at 33 °C and 23 °C, respectively. However,the inhibitory effect of R differed from that of B and FR, byonly delaying germination. A single exposure to B or FR lightcould be replaced by intermittent B or FR irradiation, and theireffects were repeatedly reversible by the following R irradiation.If the upper seeds were not exposed to R during imbibition,they failed to germinate even at 33 °C which was optimalfor germination, and the promotive effect of R increased withdelay of its application time. The photoperceptive locus incocklebur seeds was the axial tissue for all B, R and FR. Lightreceived by the cotyledonary tissue had little effect. Germinationdimorphism in response to light is discussed with respect tothe phytochrome content and the ageing of axial tissues. Three climatic races of Achillea from central California (20,1400 and 3100 m elevation) were grown under controlled environmentalconditions at 14 combinations of temperature (0–27°C)and photoperiod (8 or 16 hr). Periodic measurements were madeon height and flowering. Dry weights of roots and shoots ofmature plants were determined. The greatest growth in heightand weight for each race occurred at 11°C on long days (16hr). The coastal race (20 m) grew well over a wide range oftemperatures (7–27°C) but the higher elevation races(1400 and 3100 m) grew well only at low temperatures (7–15°C).In contrast to the vigorous elongation and flowering of thecoastal race in short days at nearly all temperatures, racesfrom the higher elevations remained as rosettes. The sub-alpinerace (3100 m) exhibited the highest root-shoot ratios. Floweringpreceded stem elongation in the sub-alpine race but followedstem elongation in the coastal race. Exogenous applicationsof gibberellic acid (500 mg/liter) to rosette high-elevationraces promoted stem elongation and some flowering. Steroid applicationshad little or no effects.
