Populations of the dinoflagellate Dinophysis norvegica in the Baltic Sea and in the adjacent North Sea are infected by the endoparasite Amoebophrya sp. The high diversity recently unveiled within the genus Amoebophrya brings uncertainty about their identities. We applied molecular biology techniques--18S rDNA sequencing and fluorescent in situ hybridization (FISH)--to compare this host-parasite system from both environments. The North Sea Amoebophrya sp. 18S rDNA sequence was 89% identical to the previously described Baltic Sea Amoebophrya sp. sequence, suggesting they are different species. In spite of that, a phylogenetical analysis placed the North Sea parasite sequence in a well-supported cluster with other Amoebophrya sp. sequences. The D. norvegica 18S rDNA sequences from both environments were 100% identical, indicating that the hosts have not evolved independently. A DNA probe designed for the Baltic Sea Amoebophrya sp. 18S rRNA was used in FISH assays on infected D. norvegica populations from both environments. The probe stained all infected cells from the Baltic sample, whereas none from the North Sea were stained. The results indicate that D. norvegica is released from one parasite when entering the Baltic Sea, and become less infected by an alternative parasite species.  相似文献

1. Surface sediment biofilm samples from 82 Pyrenean lakes were analysed for marker pigment composition using high performance liquid chromatography (HPLC). 2. Variability in the pigment composition among lakes was investigated by multivariate statistical analyses using a large data set of factors describing lake chemical, physical, morphological and catchment characteristics. 3. Due to the widely varying light penetration in the lakes, the most significant gradient of pigment composition extended from a benthic to a planktonic signal. The most important pigments in the gradient were alloxanthin (cryptophytes marker pigment, planktonic signal) and diatoxanthin (diatoms marker pigment, benthic signal). The molar ratio between these two marker pigments was positively correlated with lake depth. 4. Chlorophyll‐a preservation was found to be positively related to light penetration and the development of an autothrophic biofilm on the surface sediment and negatively related to decreasing pH and the percentage of alpine meadows in the lake catchments. 5. Zooplankton marker pigments in the surface sediment, including grazing by‐products (e.g. phaeophorbides) and carotenoids (astaxanthin, canthaxanthin, echinenone) incorporated into their tissues, were correlated with the areal abundance of zooplankton. 6. Marker pigments for photosynthetic bacteria, BChl‐e and okenone, were found mainly in relatively shallow lakes with large catchments that are forested, probably because of their higher loading of allochthonous organic matter. 7. The evaluation of a preservation index (Chl‐a expressed as a percentage of a‐phorbins) and the alloxanthin/diatoxanthin ratios throughout the sediment record of mountain lakes can provide evidence of historical changes in the relative importance of planktonic versus benthic primary production and might ultimately be interpreted in terms of climatic or environmental changes.  相似文献

The rhabdoms of Euphausia superba contain one digitonin-extractable rhodopsin, lambda max 485 nm. The rhodopsin undergoes unusual pH- dependent spectral changes: above neutrality, the absorbance decreases progressively at 485 nm and rises near 370 nm. This change is reversible and appears to reflect an equilibrium between a protonated and an unprotonated form of the rhodopsin Schiff-base linkage. Near neutral pH and at 10 degrees C, the rhodopsin is partiaLly converted by 420-nm light to a stable 493-nm metarhodopsin. The metarhodopsin is partially photoconverted to rhodopsin by long-wavelength light in the absence of NH2OH; in the presence of NH2OH, it is slowly converted to retinal oxime and opsin. The rhodopsin of Meganyctiphanes norvegica measured in fresh rhabdoms by microspectrophotometry has properties very similar to those of the extracted rhodopsin of E. superba. Its lambda max is 488 nm and it is partially photoconverted by short wavelength irradiation to a stable photoconvertible metarhodopsin similar to that of E. superba. In the presence of light and NH2OH, the M. norvegica metarhodopsin is converted to retinal oxime and opsin. Our results indicate that previous determinations of euphausiid rhodopsin absorbance spectra were incorrect because of accessory pigment contamination.  相似文献

Food selection by the marine cladoceran Penilia avirostris was studied in the field by HPLC analysis of phytoplankton marker pigments and in the laboratory by microscopic measurement of cell removal. Comparison between pigment composition in natural phytoplankton and in P. avirostris showed that P. avirostris preferred diatoms, cryptophytes and chlorophytes, and ignored prymnesiophytes and dinoflagellates. Peridinin, the marker pigment for dinoflagellates was found in P. avirostris only when the dinoflagellate populations were dominated by Prorocentrum. Pigment degradation rates ranged from 13.73% for alloxanthin to 36.62% for chlorophyll a. Clearance rates measured in the laboratory provided further evidence of strong preference for diatoms and cryptophytes, and avoidance of dinoflagellates. Microscopic counts suggested that P. avirostris was feeding on prymnesiophytes, although ingestion of prymnesiophytes could not be confirmed by pigment analysis.  相似文献

Kleptoplasty is the retention of plastids obtained from ingested algal prey, which can remain temporarily functional and be used for photosynthesis by the predator. With a new approach based on cell cycle analysis, we have addressed the question of whether the toxic, bloom-forming dinoflagellate Dinophysis norvegica practice kleptoplasty or if they replicate their own plastid DNA. Dividing (G2) and non-dividing (G1) D. norvegica cells from a natural population were physically separated with a flow cytometer based on their DNA content. Average numbers of nuclear and plastid rDNA copies were quantified with real-time PCR both in the G1 and G2 group. Cells from the G1 group contained 5800 ± 340 copies of nuclear rDNA and 1300 ± 200 copies of plastid rDNA; cells from the G2 group contained 9700 ± 58 copies of nuclear rDNA and 1400 ± 220 copies of plastid rDNA (mean ± SD, n  = 3). The ratio G2/G1 in average rDNA copies per cell was 1.67 for nuclear DNA and 1.07 for plastid DNA. These ratios show that plastid acquisition in D. norvegica is either uncoupled with the cell cycle, or plastids accumulate rapidly in the beginning of the cell cycle owing to feeding, as would be expected in a protist with kleptoplastic behaviour but not in a protist with own plastid replication. In addition, flow cytometry measurements on cells from the same population used for real-time PCR showed that when kept without plastidic prey, live Dinophysis cells lost on average 36% of their plastid phycoerythrin fluorescence in 24 h. Together these findings strongly suggest that D. norvegica does not possess the ability for plastid replication.  相似文献

The ecological importance and diversity of pico/nanoplanktonic algae remains poorly studied in marine waters, in part because many are tiny and without distinctive morphological features. Amongst green algae, Mamiellophyceae such as Micromonas or Bathycoccus are dominant in coastal waters while prasinophytes clade VII, yet not formerly described, appear to be major players in open oceanic waters. The pigment composition of 14 strains representative of different subclades of clade VII was analyzed using a method that improves the separation of loroxanthin and neoxanthin. All the prasinophytes clade VII analyzed here showed a pigment composition similar to that previously reported for RCC287 corresponding to pigment group prasino‐2A. However, we detected in addition astaxanthin for which it is the first report in prasinophytes. Among the strains analyzed, the pigment signature is qualitatively similar within subclades A and B. By contrast, RCC3402 from subclade C (Picocystis) lacks loroxanthin, astaxanthin, and antheraxanthin but contains alloxanthin, diatoxanthin, and monadoxanthin that are usually found in diatoms or cryptophytes. For subclades A and B, loroxanthin was lowest at highest light irradiance suggesting a light‐harvesting role of this pigment in clade VII as in Tetraselmis.  相似文献

In order to evaluate the validity of the gut pigment methodto assess grazing and diet in two freshwater zooplankters, experimentswere carried out to check chlorophyll a and xanthophyll conservationduring feeding. For both animals, two sets of experiments wereconducted by incubating animals in the laboratory, either isolatedfrom a reservoir (the calanoid copepod, Eudiaptomus gracilis)or cultured under high-food conditions (the cladoceran, Daphniagaleata). For both animals, gut pigments and clearance rateson different types of algae were determined from the same incubations.Chlorophyll a and derivatives, as well as major algal carotenoids,were analysed by High Performance Liquid Chromatography (HPLC).In copepods, the pigment profiles from the gut extracts reflectedthe diet of the animals poorly. The animal extracts containedalmost exclusively alloxanthin (or an alloxanthin-like pigment)in large amounts, whereas the other pigments were lost in highproportions (>70% for lutein and fucoxanthin; 57 and 78%for a-phorbins). The cladocerans fed on the main types of algaeabundant in the suspensions, with a preference, however, forsmall cells. Although the main xanthophylls from these algaewere detected in the Daphnia extracts, some destruction of luteinand fucoxanthin may have occurred (18.7 and 30%). The loss ratefor alloxanthin seemed more variable (0 and 68%), possibly dependingon food concentration. As for the transformation of a-phorbins,E.gracilis and D.galeata behaved quite differently. The HPLCprofiles of copepod extracts always showed a very small chlorophylla peak, along with phaeophytin a and pyrophaeophytin a. Thosefrom the cladoceran exhibited a large phaeophorbide a peak,along with some chlorophyll a and phaeophytin a. In fact, D.galeatadid not destroy a-phorbins under our experimental conditionsbut converted chlorophyll a mainly into phaeophorbide. Froma comparison of our results with data from other studies, itseems that in these two zooplankters, use of gut pigment datafor quantitative grazing assessment should be considered withcaution.  相似文献

The octoploid Draba cacuminum (Brassicaceae) is one of the few endemic species in Scandinavia and has frequently been considered an example supporting the glacial survival theory. Two subspecies, ssp. cacuminum in southern Norway and ssp. angusticarpa in northern Scandinavia, have been described and suggested to be geographically diverged remnants of a more continuously distributed ancestor. To test an alternative hypothesis of independent origins of similar octoploids in different areas, we investigated populations of D. cacuminum and its possible progenitors using enzyme electrophoresis and morphological analysis. Electrophoretic analysis of progeny of D. cacuminum revealed high levels of fixed heterozygosity, suggesting that the species is a genetic alloploid. Fixed electrophoretic differences among the populations and additivity of electrophoretic phenotypes indicate that the octoploid D. cacuminum has originated at least three times by alloploidizations involving different populations of the hexaploid D. norvegica and a diploid species, possibly D. fladnizensis. Electrophoretic and morphometric data suggest that populations of D. cacuminum with broad siliculae have originated from populations of D. norvegica that had broad siliculae, and that populations of D. cacuminum with narrow siliculae have originated from populations of D. norvegica that had narrow siliculae. However, the electrophoretic and morphometric variation within D. cacuminum did not correspond to the geographic origin of the populations, and the previously described subspecies could not be recognized. Draba cacuminum gives no relevant information on the glacial survival theory; the polyploidizations may have occurred in postglacial time as well as in pre-Weichselian periods.  相似文献

Aquatic habitats are usually structured by light attenuation with depth resulting in different microalgal communities, each one adapted to a certain light regime by their specific pigment composition. Several taxa contain pigments restricted to one phylogenetic group, making them useful as marker pigments in phytoplankton community studies. The nuisance and invasive freshwater microalga Gonyostomum semen (Raphidophyceae) is mainly found in brown water lakes with sharp vertical gradients in light intensity and color. However, its pigment composition and potential photoadaptations have not been comprehensively studied. We analyzed the photopigment composition of 12 genetically different strains of G. semen by high performance liquid chromatography after acclimation to different light conditions. We confirmed the pigments chl a, chl c1c2, diadinoxanthin, trans‐neoxanthin, cis‐neoxanthin, α and β carotene, which have already been reported for G. semen. In addition, we identified, for the first time, the pigments violaxan‐thin, zeaxanthin, and alloxanthin in this species. Alloxanthin has never been observed in raphidophytes before, suggesting differences in evolutionary plastid acquisition between freshwater lineages and the well‐described marine species. The amount of total chl a per cell generally decreased with increasing light intensity. In contrast, the increasing ratios of the prominent pigments diadinoxanthin and alloxanthin per chl a with light intensity suggest photoprotective functions. In addition, we found significant variation in cell‐specific pigment concentration among strains, grouped by lake of origin, which might correspond to genetic differences between strains and populations.  相似文献

The benthos of a perennially ice-covered Antarctic lake, Lake Hoare, contained three distinct signatures of lipophilic pigments. Cyanobacterial mats found in the moat at the periphery of the lake were dominated by the carotenoid myxoxanthophyll; carotenoids: chlorophyll a ratios in this high light environment ranged from 3 to 6.8. Chlorophyll c and fucoxanthin, pigments typical of golden-brown algae, were found at 10 to 20 m depths where the benthos is aerobic. Anaerobic benthic sediments at 20 to 30 m depths were characterized by a third pigment signature dominated by a carotenoid, tentatively identified as alloxanthin from planktonic cryptomonads, and by phaeophytin b from senescent green algae. Pigments were not found associated with alternating organic and sediment layers. As microzooplankton grazers are absent from this closed system and transformation rates are reduced at low temperatures, the benthos beneath the lake ice appears to contain a record of past phytoplankton blooms undergoing decay.  相似文献

SYNOPSIS. A new carotenoid, manixanthin , was isolated from a bleached, autolysed culture of the marine cryptomonad Chroomonas salina grown on glycerol in light, after the culture showed disappearance of the chlorophylls and carotenoids normally produced. Manixanthin lacked carbonyl, ester, and epoxy groups in chemical tests, whilst its spectroscopic properties indicated a conjugated nonaene chromophore. In its chromatographic mobility, manixanthin closely resembled alloxanthin, the major normal xanthophyll of the cryptomonad. The iodine-catalysed photostereomutation of manixanthin produced the same equilibrium mixture of cis -isomers as that given by similar treatment of alloxanthin. This mixture of isomers completely lacked the all-trans alloxanthin and contained ∼60% manixanthin. It was concluded that manixanthin is an unusually stable cis -isomer of alloxanthin, and is presumably produced in the culture medium by the action of light on alloxanthin released by autolysed cells of the cryptomonad. The unusual stability of manixanthin relative to alloxanthin was investigated by effecting partial stereoisomerizations under conditions using either of heat, light, or iodine, and evidence was obtained for a series of at least 10 isomers with a trend of stability increase from alloxanthin to manixanthin. It was inferred that manixanthin may be 9,9'-di cis -alloxanthin.  相似文献

In the Nervion River estuary surface samples were taken from March to September 2003 at six sites covering most of the salinity range with the aim to know the biomass and taxonomic composition of phytoplankton assemblages in the different segments. Nine groups of algae including cyanobacteria, diatoms, dinoflagellates, chlorophytes, prasinophytes, euglenophytes, chrysophytes, haptophytes, raphidophytes and cryptophytes were identified by means of a combination of pigment analysis by high-performance liquid chromatography (HPLC) and microscopic observations of live and preserved cells. Diatoms, chlorophytes and cryptophytes were the most abundant algae in terms of cells number, whereas fucoxanthin, peridinin, chlorophyll b (Chl b) and alloxanthin were the most abundant auxiliary pigments. Based on multiple regression analysis, in the outer estuary (stations 0, 1, 2 and 3) about 93% of the chlorophyll a (Chl a) could be explained by algae containing fucoxanthin and by algae containing Chl b, whereas in the rest of the estuary most of the Chl a (about 98%) was accounted for by fucoxanthin, Chl b and alloxanthin containing algae. The study period coincided with that of most active phytoplankton growth in the estuary and fucoxanthin was by far the dominant among those signature pigments. Several diatoms, chrysophytes, haptophytes and raphydophytes were responsible for fucoxanthin among identified species. Besides, dinoflagellates with a pigment pattern corresponding to chrysophytes and type 4 haptophytes were identified among fucoxanthin-bearing algae. Cryptophytes were the most abundant species among those containing alloxanthin. The maximum of Chl b registered at the seaward end in April coincided with a bloom of the prasinophytes Cymbomonas tetramitiformis, whereas the Chl b maxima in late spring and summer were accounted for by prasinophytes in the middle and outer estuary and by several species of chlorophytes in the middle and inner estuary. Other Chl b containing algae were euglenophytes and the dinoflagellate Peridinium chlorophorum. Dinoflagellates constituted generally a minor component of the phytoplankton.  相似文献

During most of the growing season of 1994, pigment content,as determined by HPLC analysis of algal sample extracts, wasfollowed in the River Meuse (Belgium) potamoplankton. The concentrationof some algal pigments (chlorophylls a and b, fucoxanthin, lutein,echinenone and alloxanthin) was related to biomass estimatesof total phytoplankton and of major taxonomic components (diatoms,green algae, cyanobacteria and cryptomonads). Highly significantlinear regressions were obtained for chlorophyll a-total biomass,fucoxanthin-diatoms, lutein-green algae, chlorophyll b-greenalgae. However, no relationship was found for cyanobacteriaor cryptomonads and their specific pigments, which may be attributedto poor accuracy of biomass estimates for these non-dominantalgae. In conclusion, the good relationship found for dominantalgae and their specific pigments confirms the value of pigmentsas quantitative markers of phytoplankton, as detected in othermarine and freshwater environments.  相似文献

Tethya norvegica Bowerbank, 1872, a northern species generally synonymized with T. aurantium . has been studied from Scotland, The Faroes, Norway, Iceland, Spitzbergen and Bear Island. The species is redescribed and its North East Atlantic distribution delimited Tethya norvegica is compared, on the basis of morphometric and electrophoretic features, with T. aurantium and F.citrina . Morphological, morphometric and electrophoretic data suggest that I norvegica is a distinct biological species.  相似文献

The correlation was studied between the elevated resistance of Pseudomonas methanolica, a facultative methylotroph, to the bactericidal action of UV (254 nm) and the content of a pink pigment in this organism. Absorption spectra of cell extracts containing the pigment were recorded. The ethanol and acetone extracts had two absorption maxima at 420-430 and 500-510 nm, respectively, which was typical of carotenoids. The culture with the pigment and its variant without the pigment had nearly identical survival rates. Therefore, the elevated resistance of P. methanolica cells seems to stem from the activity of DNA reparation cellular systems rather than from the presence of carotenoid pigments in the cells.  相似文献

Despite the potential importance of zooplankton in degradationof marine snow, the association of colonising zooplankton withdiscarded appendicularian houses has not been investigated innorthern areas. We sampled the vertical distributions of appendicularians,houses and potential zooplankton colonisers at two stationsin the central North Sea during late summer. In addition, grazingexperiments were performed with the copepod Microsetella norvegica,which was assumed to be the main contributor to house degradation.The results were used in (i) inverse modelling, to estimatethe factors which were significant in shaping the vertical distributionof houses and (ii) calculations to estimate potential housedegradation rates. M. Norvegica was able to feed on appendicularianhouses, with feeding rates up to 0.42 g C (g C)–1 day–1(0.14 µg C ind.–1 day–1). The model resultssuggested that the vertical distribution of houses was shapedby sinking of houses, bacterial degradation and feeding of M.norvegica and invertebrate larvae. The estimated community degradationrate by M. norvegica was low, whereas invertebrate larvae haddegradation rates close to bacterial degradation. We concludethat at the typical concentrations of M. norvegica in the NorthSea (104 ind. m–2), its role in marine snow degradationis likely to be small. Degradation by other zooplankton groups,such as invertebrate larvae, can, however, be substantial.  相似文献

The ocellar potential (OP) of planaria was recorded using microelectrode techniques. The action spectrum and spectral sensitivity of the OP are described. Maximum OP sensitivity was found with 508 nm light. A moderate increase in sensitivity to blue light was observed. This is typical of many invertebrate photoreceptors and was shown, by selective chromatic adaptation, not to indicate the presence of a second pigment.  相似文献

Microsetella norvegica is a widely distributed marine planktonicharpacticoid copepod, which is poorly known from the biologicalpoint of view. We investigated the seasonal population dynamicsandproduction of M. norvegica in the central part of the InlandSea of Japan. It occurred throughout the year, whilst its reproductionwas confined to the warm season between May and November. Theproportion of ovigerous females, which carry a single egg sac,was low (mean: 23.1%) in August and September, and high (53.6%)in October. Their brood size attained a maximum (mean: 15.8eggs per sac) in July and August and declined gradually to 6.2eggs in November. Duration time from egg laying to moultingto adulthood was temperature-dependent; it was 31.9 and 14.3days at 20 and 27°C, respectively, under excess food conditionsin the laboratory. An enormously high population abundance (7.32x 104 individuals m-3), which accounted for 86.5% of the totalcopepods, and biomass (69.6 mg C m-3) gave an annual maximumproduction rate of 4.90 mg C m-3 day-1) in October. Naupliiand copepodites disappeared in December, and the overwinteringpopulation was represented by adults, mainly large females.Associations of M. norvegica with marine snow aggregates, whichhave often been found in oligotrophic waters, were not observedin the food-rich environment of the Inland Sea of Japan.  相似文献

Abstract Natural phyto- and bacterioplankton populations of Lake Vechten (the Netherlands) were subjected to darkness under oxic and anoxic conditions at in situ temperatures in order to test the stability in time of their photosynthetic pigments. Furthermore, sedimentary fluxes and concentrations of pigments were estimated in (respectively) sediment trap catches and at the sediment-water interphase in order to measure the pigment breakdown upon burial into the sediment. The chlorophylls and most of the xanthophylls showed substantial losses of 20% to 60% in the incubation experiments as well as in the surficial sediment. β-Carotene, okenone and echinenone were most stable (2–10% losses); fucoxanthin and peridinin were degraded extensively; alloxanthin and zeaxanthin held an intermediate position as did the bacteriochlorophylls. Trends in sediment profiles of pigments were compared with limnological data obtained during the enhanced eutrophication of the lake. Evidence was provided that the β-carotene profile closely followed the increase of phytoplankton biomass. Although susceptible to substantial degradation, several profiles of pigments and of pigment ratios could be related in a qualitative way to biomass and to shifts in species composition which occurred as a result of the changing ecological conditions in the lake.  相似文献

A morphological analysis of the Scandinavian mountain endemic Draba cacuminum Elis. Ekman is presented, based on Scandinavian herbarium collections and a population sample from the Finse area, S Norway. It is compared with supposedly close relatives, especially D. norvegica Gunnerus, and is found to be a distinct species with infraspecies variation at two levels. The populations of southern and northern Scandinavia have been separated as subspecies, ssp. cacuminum and ssp. angusticarpa Elven ssp. nov., respectively, and there is also variation within the southern subspecies. A lectotype has been chosen for D. cacuminum , a revised distribution map is presented, and the ecology and phytogeography of the species is discussed. It seems to be a weak competitor, and its present very discontinuous area is assumed to be a remnant of one continuous or more probably two areas (in S Norway and N Scandinavia) in the late Weichselian.  相似文献
