A bacterial strain M6, isolated from soil and identified as Arthrobacter globiformis, produced a novel nonreducing oligosaccharide. The nonreducing oligosaccharide was produced from starch using a culture supernatant of the strain as enzyme preparation. The oligosaccharide was purified as a crystal preparation after alkaline treatment and deionization of the reaction mixture. The structure of the oligosaccharide was determined by methylation analysis, mass spectrometry, and (1)H and (13)C NMR spectroscopy, and it was demonstrated that the oligosaccharide had a cyclic structure consisting of four glucose residues joined by alternate alpha-(1-->4)- and alpha-(1-->6)-linkages. The cyclic tetrasaccharide, cyclo-{-->6)-alpha-D-Glcp(1-->4)-alpha-D-Glcp(1-->6)-alpha-D-Glcp(1-->4)-alpha-D-Glcp(1-->}, was found to be a novel oligosaccharide, and was tentatively called cyclic maltosyl-maltose (CMM). CMM was not hydrolyzed by various amylases, such as alpha-amylase, beta-amylase, glucoamylase, isoamylase, pullulanase, maltogenic alpha-amylase, and alpha-glucosidase, but hydrolyzed by isomalto-dextranase to give rise to isomaltose. This is the first report of the cyclic tetrasaccharide, which has alternate alpha-(1-->4)- and alpha-(1-->6)-glucosidic linkages.  相似文献

A glycosyltransferase, involved in the synthesis of cyclic maltosylmaltose [CMM; cyclo-{-->6)-alpha-D-Glcp(1-->4)-alpha-D-Glcp(1-->6)-alpha-D-Glcp(1-->4)-alpha-D-Glcp(1-->}] from starch, was purified to homogeneity from the culture supernatant of Arthrobacter globiformis M6. The CMM-forming enzyme had a molecular mass of 71.7 kDa and a pI of 3.6. The enzyme was most active at pH 6.0 and 50 degrees C and was stable from pH 5.0 to 9.0 and up to 30 degrees C. The addition of 1 mM Ca2+ enhanced the thermal stability of the enzyme up to 45 degrees C. The enzyme acted on maltooligosaccharides that have degrees of polymerization of > or =3, amylose, and soluble starch to produce CMM but failed to act on cyclomaltodextrins, pullulan, and dextran. The mechanism for the synthesis of CMM from maltotetraose was determined as follows: (i) maltotetraose + maltotetraose --> 6(4)-O-alpha-maltosyl-maltotetraose + maltose and (ii) 6(4)-O-alpha-maltosyl-maltotetraose --> CMM + maltose. Thus, the CMM-forming enzyme was found to be a novel maltosyltransferase (6MT) catalyzing both intermolecular and intramolecular alpha-1,6-maltosyl transfer reactions. The gene for 6MT, designated cmmA, was isolated from a genomic library of A. globiformis M6. The cmmA gene consisted of 1,872 bp encoding a signal peptide of 40 amino acids and a mature protein of 583 amino acids with a calculated molecular mass of 64,637. The deduced amino acid sequence showed similarities to alpha-amylase and cyclomaltodextrin glucanotransferase. The four conserved regions common in the alpha-amylase family enzymes were also found in 6MT, indicating that 6MT should be assigned to this family.  相似文献

Several extracellular glucans have been isolated from Phytophthora parasitica Dastur, a phytopathogenic fungus of the carnation. These polysaccharides consist of a mixture of (1-->3)(1-->6)-beta-D-glucans whose molecular masses varied from 1 x 10(4) to 5 x 10(6) Da. All of these polysaccharides have a main chain of beta-(1-->3)-linked D-glucose residues substituted with mono-, di- and oligo-saccharidic chains attached through (1-->6) linkages.  相似文献

Three polysaccharides, glucans with mean M(r)'s of 1.5 x 10(5), 3.6 x 10(4) and 2.1 x 10(4), were isolated from dried roots of Periandra mediterranea by fractionation on Sephacryl S-300 HR and Sephadex G-25. Chemical and spectroscopic studies indicated that they have a highly branched glucan type structure composed of alpha-(1-->4) linked D-glucopyranose residues with both (3-->4) and (4-->6) branching points. The polysaccharides enhance phagocytosis in vivo, and exhibit anti-inflammatory activity.  相似文献

For the culture of organs, modified B5, KU, MS, NT, 1/2 MS media were used. Callus induction and plantlet regeneration from mots, leaves, petioles, cotyledons and hypocotyls of Angelica sinensis (Oliv.) Diels have been obtained. It was found in our experiments the following results: 1. The callus induction from the rcots has arisen rapidly and increased at a high rate and with a high differentiating frequency. 2. The phytohormones play a significant role in callus induction and differentiation. 3. The suitable combination of cytokinin (6-BA) with auxins (2, 4-D, NAA) may enhance their induction and subculture, but the effect was different on the differentiation. 4. Binucleate Rhizoctonia (BNR) isolate (232-C6) is an effective biocontrol agent for protection of potato from Rhizoctonia canker, a disease caused by Rhizoctonia solani. Production of hydrolytic enzymes is one of the best known inducible defense responses following microbial infection. We isolated and characterized a cell wall alpha-glucan from BNR, which induces beta-1,3 glucanase activities in potato sprouts, the primary site of infection by R. solani. An autoclaving method, previously reported for isolation of oligosaccharide elicitors was used, and the glucan purified by chromatographic techniques. Maximal induction of beta-1,3 glucanase activity in potato sprouts was obtained with 250 microg of the alpha-glucan elicitor after 6 days from inoculation time. Both, BNR mycelium and the alpha-glucan produced a similar kinetic response of beta-1,3 glucanase. However, the alpha-glucan did not induce phytoalexin accumulation, previously correlated with the defense response. Uronic acids (approximately 10% with respect to total neutral sugars) were determined and identified as glucuronic acid by high-pH anion-exchange chromatography. Methylation analysis showed that the glucan consists of (1-->3) and (1-->4)-linked glucose units with preponderance of the first ones. Some of the (1-->4) linkages were branched at position 6. The glucan was partially degraded with amyloglucosidase. This, together with the NMR spectra data and the high optical rotation of the original (+195 degrees ) and degraded glucans (+175 degrees ) proved the alpha configuration. Further methylation of the amyloglucosidase degraded glucans indicated that they consist of (1-->3)-linked glucoses. The present study is the first report on the isolation and characterization of an alpha-glucan from Rhizoctonia, that may be important as a biocontrol factor.  相似文献

A detailed phytochemical study on the 70% aqueous ethanol extract of stems of Akebia trifoliata (Thunb.) Koidz. var. australis (Diels) Rehd led to isolation of five compounds, together with 12 known triterpenoid saponins and three known phenylethanoid glycosides. The structures of the five compounds were elucidated on the basis of analysis of spectroscopic data and physicochemical properties as: 2alpha, 3beta, 23-trihydroxy-30-norolean-12-en-28-oic acid beta-D-glucopyranosyl ester (1), 2alpha, 3beta, 23-trihydroxy-30-norolean-12-en-28-oic acid beta-D-xylopyranosyl-(1-->3)-O-alpha-D-rhamnopyranosyl-(1-->4)-O-beta-D-glucopyranosyl-(1-->6)-O-beta-D-glucopyranosyl ester (2), 2alpha, 3beta, 23-trihydroxyurs-12-en-28-oic acid beta-D-xylopyranosyl-(1-->3)-O-alpha-L-rhamnopyranosyl-(1-->4)-O-beta-D-glucopyranosyl-(1-->6)-O-beta-D-glucopyranosyl ester (3), 3-beta-[(beta-D-glucopyranosyl-(1-->3)-O-alpha-L-arabinopyranosyl)oxy]-23-hydroxy-30-norolean-12-en-28-oic acid alpha-L-rhamnopyranosyl-(1-->4)-O-beta-D-glucopyranosyl-(1-->6)-O-beta-D-glucopyranosyl ester (4) and 3-beta-[(alpha-L-xylopyranosyl-(1-->2)-O-alpha-L-arabinopyranosyl)oxy]-30-norolean-12-en-28-oic acid alpha-L-rhamnopyranosyl-(1-->4)-O-beta-D-glucopyranosyl-(1-->6)-O-beta-D-glucopyranosyl ester (5), named mutongsaponin A, B, C, D and E, respectively.  相似文献

Through a combination of anion-exchange and repeated gel chromatographies, APS-1 was isolated from fresh leaves of Aloe vera L. var. chinensis (Haw.) Berger (an edible and medicinal plant widely cultivated and consumed in China) as a principal polysaccharide composed of mannose and glucose (ca. 18:5) with its molecular weight around 2.1 x 10(5). In a dose-dependent manner, APS-1 was demonstrated to be free radical scavenging in superoxide and hydroxyl radical assays, inhibitory to the copper-mediated oxidation of human low density lipoprotein (LDL), and protective against hydrogen peroxide (H(2)O(2))-induced lesion to rat PC12 cell (pheochromocytoma cell line). The result suggested that APS-1 could be of considerable preventive and therapeutic significance to some free radical associated health problems such as coronary heart ailments, Parkinson's and Alzheimer's diseases. Furthermore, the finding shed as well fresh light helpful for a better understanding of the health-benefiting potential of the edible plant consumed by the Chinese people for a couple of centuries.  相似文献

Cordyceps sinensis is a well known tonic food or invigorant with broad-spectrum medicinal properties that is widely used in the People's Republic of China. A neutral mannoglucan 1 with a molar mass of approximately 7.7x10(3) Da was obtained from the 0.05 M acetate buffer extract of C. sinensis mycelium. It had [alpha](D)(20)+126 (c 0.2, H(2)O) and consisted of Man and Glc units in the molar ratio of 1:9. A combination of chemical analysis and NMR and IR spectroscopy together with digestion with alpha-D-amylase showed 1 to have a alpha-D-glucan backbone with (1-->4)- and (1-->3)-linkages, and the side chains of alpha-D-(1-->6)-Manp were attached to the backbone via O-6 of alpha-(1-->3)-Glcp residues. Compound 1 showed weak cytotoxicity activity against SPC-I (IC(50)=63 microg/mL) cancer line, and no obvious cytotoxicity activities against BCAP37 (IC(50)>100 microg/mL) and SW480 (IC(50)>100 microg/mL) cancer lines.  相似文献

An O-specific polysaccharide containing 2-acetamidino-2-deoxy-beta-D-glucopyranose (Glcp2Am), 2,4-diacetamido-2,4,6-trideoxy-beta-D-glucopyranose (QuipNAc4NAc, bacillosamine) and 2,4-di-(N-acetyl-L-alanylamino)-2,4,6-trideoxy-beta-D-glucopyranose (QuipNAlaAc4NAlaAc) was isolated from the phenol-soluble lipopolysaccharide fraction of the mushroom-associated bacterium Pseudomonas reactans. The glucans of lichenized fungi are an important class of polysaccharides with structural and chemotaxonomic roles. The water-insoluble glucans of the genus Parmotrema (P. austrosinense, P. delicatulum, P. mantiqueirense, P. schindleri, and P. tinctorum) and those of Rimelia (R. cetrata and R. reticulata), were investigated in order to evaluate the significance in chemotyping, with nigeran [(1-->3),(1-->4)-alpha-glucan] and lichenan [(1-->3),(1-->4)-beta-glucan] characterized using (1)H and (13)C NMR, methylation analysis, and controlled Smith degradations. Results from all species were similar, suggesting that glucan chemistry does not support separation of Rimelia from Parmotrema.  相似文献

Several structurally different glucans (alpha- and beta-) and galactomannans were characterized as components of four species of the genus Ramalina, namely R. dendriscoides, R. fraxinea, R. gracilis and R. peruviana. Freeze-thawing treatment of hot aqueous extracts furnished as precipitates (PW) linear alpha-D-glucans of the nigeran type, with regularly distributed (1-->3)- and (1-->4)-linkages in a 1:1 ratio. The supernatants (SW) contained alpha-D-glucans with (1-->3)- and (1-->4)-linkages in a molar ratio of 3:1. The lichen residues were then extracted with 2% aq. KOH, and the resulting extracts submitted to the freeze-thawing treatment, giving rise to precipitates (PK2) of a mixture of alpha-glucan (nigeran) and beta-glucan, which were suspended in aqueous 0.5% NaOH at 50 degrees C, dissolving preferentially the beta-glucan. These were linear with (1-->3)-linkages (laminaran). The mother liquor of the KOH extractions (2% and 10% aq. KOH) was treated with Fehling's solution to give precipitates (galactomannans). The galactomannans are related, having (1-->6)-linked alpha-D-mannopyranosyl main chains, substituted at O-4 and in a small proportion at O-2,4 by beta-D-galactopyranosyl units. Despite the different habitats of these lichenized fungi, all species studied in this investigation have a similar pool of polysaccharides.  相似文献

A new water-soluble intracellular polysaccharide named as PTP, with a molecular mass of 3.7x10(4) Da, was obtained from the mycelium of Polyporus albicans (Imaz.) Teng. Structure features of the purified polysaccharide were investigated by a combination of chemical and instrumental analysis. The results indicated that PTP consisted of a backbone composed of (1-->3)-linked-beta-d-mannopyranosyl, (1-->3,6)-linked-beta-d-mannopyranosyl and (1-->6)-linked-alpha-d-galactopyranosyl residues in the ratio of 3:1:1, and terminated with a single non-reducing terminal (1-->)-beta-d-mannopyranosyl residues at the C-6 position of (1-->3,6)-linked-beta-d-mannopyranosyl, along the main chain. This is the first report describing the isolation and structure elucidation of a new intracellular polysaccharide produced from the mycelium of P. albicans (Imaz.) Teng. Preliminary tests in vitro showed PTP have potent stimulating effects on murine lymphocyte proliferation induced by concanavalin A or lipopolysaccharide and its branches are extremely important for the expression of the enhancement of the immunological activity.  相似文献

Alkali extraction and methylation analyses in the 1970s revealed that the cell walls of the yeast Schizosaccharomyces pombe contain a (1-->3)-alpha-d-glucan, a (1-->3)-beta-d-glucan, a (1-->6)-beta-d-glucan, and a alpha-galactomannan. To refine the structures of these polysaccharides, cell-wall glucans of S. pombe were extracted, fractionated, and analyzed by NMR spectroscopy. S. pombe cells were treated with 3% NaOH, and alkali-soluble and insoluble fractions were prepared. The alkali-insoluble fraction was treated with 0.5M acetic acid or Zymolyase 100T to yield an alkali-insoluble, acetic acid-insoluble fraction, an alkali-insoluble, Zymolyase-insoluble fraction, and an alkali-insoluble, Zymolyase-soluble fraction. (13)C NMR and 2D-NMR spectra disclosed that the cell wall of S. pombe is composed of three types of glucans, specifically, a (1-->3)-alpha-d-glucan, a (1-->3)-beta-d-glucan, which may either be linear or slightly branched, and a highly branched (1-->6)-beta-d-glucan, in addition to alpha-galactomannan. The highly branched (1-->6)-beta-d-glucan was identified by selective periodate degradation of side-chain glucose as a highly (1-->3)-beta-branched (1-->6)-beta-d-glucan with more branches than that of Saccharomyces cerevisiae. Flexibility of these polysaccharides in the cell wall was analyzed by (13)C NMR spectra in D(2)O. The data collectively indicate that (1-->3)-alpha- and (1-->3)-beta-d-glucans are rigid and contribute to the cell shape, while the highly branched (1-->6)-beta-d-glucan and alpha-galactomannan are flexible.  相似文献
