过量DMSO显著降低低模板浓度PCR扩增的特异性   总被引:2,自引:0,他引:2  
DMSO通常经验性地用于提高PCR扩增的效率,但是过量的DMSO可以显降低特异序列的扩增效率,尤其是导致非特异性扩增的现象却被忽视。在6%的DMSO存在时,开始出现非特异条带,同时特异扩增产物减少。本首次报道DMSO对PCR产物特异性的影响并确定了克服上述现象产生的途径,即通过增加模板-引物的比例消除非特异条带。而通常提高复性温度只能部分减少非特异产物,不能避免非特异扩增。本结果有助于提高常规PCR,尤其是分子遗传学分析中经常使用的随机扩增多态性DNA(random amplifeid polymorphic DNA,RAPD)检测的准确度。  相似文献

Quantum dots (QDs) are of great interest due to their unique chemical and physical properties. Recently, a hot start (HS) polymerase chain reaction (PCR) amplification performance based on QDs with a high-fidelity Pfu DNA polymerase has been reported. However, whether QDs can trigger HS effects with other high-fidelity or conventional DNA polymerases is yet to be understood. In the present study, we studied the QD-triggered HS effects with four high-fidelity and three conventional DNA polymerases, and the HS effect comparisons among them were also made. It was found that QDs could trigger a distinct HS PCR amplification performance with all the four tested high,fidelity DNA polymerases, and specific target DNA could be well amplified even if the PCR mixture was preincubated for 2 h at 50℃. On the contrary, the HS effects were not prominent with all the three conventional Taq DNA polymerases. Specifically, the fidelity of Pfu is not sacrificed in the presence of QDs, even after a 1 h pre-incu- bation at 50℃ before PCR. Furthermore, the electrophoresis results preliminarily demonstrated that QDs prefer to adsorb high-fidelity polymerases rather than conventional ones, which might result in the QD-triggered HS effects on PCR performance by using high-fidelity DNA poly- merases.  相似文献

利用孕妇血浆DNA检测胎儿性别的研究   总被引:6,自引:0,他引:6  
本文探讨应用孕妇血浆中游离DNA进行无创性产前性别诊断的可行性。用柱分离法提取73例孕妇血浆中DNA,用巢式PCR技术检测其胎儿SRY基因。 结果73位孕妇血浆DNA含量为0.0062~0.3399μg/μL。巢式PCR检测胎儿SRY基因的灵敏度为97.37%(37/38),假阴性率2.86%(1/35),特异度85.71%(30/35),假阳性率13.16%(5/38),总符合率91.78%(67/73)。采用孕妇血浆胎儿DNA和巢式PCR技术可以快速简便的进行无创性产前性别诊断,诊断结果的准确率为91.8%,对性连锁遗传病的预防具有重要意义。 Abstract:To investigate the feasibility and possibility of application of fetal DNA from maternal plasma for noninvasive prenatal diagnosis of fetal sex,plasma DNAs in blood samples of 73 pregnant women at the gestational period of 26 to 41 weeks were extracted by column separation and nested polymerase chain reaction were employed to amplify the SRY gene.A comparison was made between the amplification results and the real sex of the fetus after their delivery.The concordance rate of SRY gene amplification results of plasma free DNA with real fetal sex was 91.78% (67/73),the sensitivity rate was 97.37% (37/38),and the specific rate was 85.71% (30/35).The cell-free fetal DNA in maternal blood can be one of the valuable material sources for noninvasive prenatal diagnosis and the method of nested PCR could be useful for fetal sex determination.The specific rate of the test was 91.78%.It is of significance to prevent sex-linked inheritant diseases.  相似文献

DNA is one of the most basic and essential genetic materials in the field of molecular biology.To date,isolation of sufficient and good-quality DNA is still a challenge for many plant species,though various DNA extraction methods have been published.In the present paper,a recycling DNA extraction method was proposed.The key step of this method was that a single plant tissue sample was recycled for DNA extraction for up to four times,and correspondingly four DNA precipitations(termed as the 1st,2nd,3rd and 4th DNA sample, respectively) were conducted.This recycling step was integrated into the conventional CTAB DNA extraction method to establish a recycling CTAB method.This modified CTAB method was tested in eight plant species,wheat,sorghum,barley,corn,rice,Brachypodium distachyon,Miscanthus sinensis and tung tree.The results showed that high-yield and good-quality DNA samples could be obtained by using this new method in all the eight plant species.The DNA samples were good templates for PCR amplification of both ISSR and SSR markers.The recycling method can be used in multiple plant species and can be integrated with multiple conventional DNA isolation methods,and thus is an effective and universal DNA isolation method.  相似文献

用38~66℃不同复性温度处理,比较了18条随机引物的扩增结果.发现复性温度在40~50℃之间均有数量不等的扩增产物,不同引物的最高复性温度不同,有些引物用60℃以上的复性温度仍有扩增产物.一些在35~38℃的复性温度下非特异性产物较多的引物,通过大幅度提高复性温度,能提高扩增产物的特异性,获得清晰的RAPD带型。 Abstract:To decrease nonspecific products and obtain clear RAPD patterns,18 10- mer random primers were tested at different annealing temperatures.The results indicated that all the amplification can be performed when the annealing temperature is in the range of 40~50℃.There were a few primers with which the amplification was still performed when the annealing temperature is above 60℃.By using high annealing temperatures,some primers which produce more nonspecific product at the annealing temperatures of 35~38℃ could generate reproducible and distinct bands.  相似文献

本研究尝试将触减 PCR与3’ cDNA末端快速扩增(rapid amplification of cDNA ends,RACE)技术进行结合,仅用一条特异性引物和一条通用引物,成功地实现了从3’末端cDNA库对鸡含锰超氧化物歧化酶(manganese-containing superoxide dismutase,MnSOD)全长cDNA的一步3’RACE快速构建。与常规使用的末端PCR或亚克隆方法相比,该法具有快速、省时、经济和特异性好的优点。Abstract: RACE(rapid amplification of cDNA ends) is a popular technique to rapidly obtain the full-length cDNA. After obtaining the 3’cDNA and 5’cDNA fragments with a overlapped region by 3’RACE and 5’RACE, the full-length cDNA could be generated by end-to-end PCR or subcloning. In this study, 3’RACE combined with touch-down PCR was successfully used for the rapid construction of full-length MnSOD cDNA of chickens. Compared with the conventional end-to-end PCR or subcloning, this method, called one-step 3’RACE, is fast, economical and highly specific. It especially fits the rapid construction of full-length cDNA by RACE method.  相似文献

Loop-mediated isothermal amplification(LAMP) is a novel nucleic acid diagnostic method that can amplify rapidly a target template under isothermal conditions. In this study, a LAMP assay for rapid detection of Chinese giant salamander ranavirus(CGSRV) was developed from culture isolates and clinical samples. The LAMP assay was developed by designing one set of four specific primers, targeting the major capsid protein(MCP) gene of CGSRV. Reaction time and temperature were optimal for 40 min at 62°C. The developed LAMP assay is specific and highly sensitive for CGSRV detection, the detection limit could reach about 5 copies of cloned viral genomic fragments. The sensitivity of the LAMP assay was about 1000 and 10-fold higher than that of both conventional and nested PCR, respectively. The LAMP amplification produces a typical ladder-like pattern of products on an agarose gel that can be visually inspected after addition of ethidium bromide. The LAMP assay was evaluated further with clinical samples, and the results indicated the suitability and simplicity of the test as a rapid diagnostic tool for the detection of CGSRV.  相似文献

A pair of primers created from information of PmNOBⅢ genome DNA Sal I fragment produced a 355bp band by using Penaeus chinensis non occluded baculovirus (PcNOBV),the WSBV isolate from P.hinensis in mainland China,as the DNA template.The specific PCR product was cloned,sequenced and labeled with digoxigenin (DIG)DNA labeling kit(Boehringer Mannheim).The DIG labeled fragment was tested by dot blot hybridization for sensitivity and specificity with purified PcNOBV nucleocapsid,PcNOBV infected shrimp tissues and healthy shrimp tissues.The detection limit of the DNA probe is 6.8pg of purified PcNOBV DNA.No hybridization signals were observed using DNA from healthy shrimp as template.Healthy P.chinensis,artificially infected P.chinensis and pond reared adult P.chinensis were screened for PcNOBV infection by both PCR and the hybridization assay.The results showed a good relationship between PCR and the hybridization assay.These findings demonstrate that the DIG labeled probe can be used as a sensitive,specific and cost effective reagent for detection of PcNOBV.  相似文献

应用肠杆菌基因间重复一致序列聚合酶链反应技术(ERIC-PCR)对李斯特氏菌基因组DNA进行分析,结果显示,李斯特氏菌种间DNA指纹图谱带型差异较大;单核细胞增生性李斯特氏菌株间及相同血清型不同来源的菌株,其DNA指纹图谱带型也有明显差异。在单核细胞增生性李斯特氏菌各株的DNA指纹图谱中发现1600bp的种专一性扩增带。结果表明,ERIC-PCR技术可用于李斯特氏菌种、菌株的鉴定及进一步分型研究。 Abstract:Enterobacteia repetitive intergenic consensus sequences-based PCR(ERIC-PCR) was used to generate DNA fingerprints for Listeria spp.We got the specific profiles with ERIC-PCR technique that enables to identify Listeria species and the L.monocytogenes strains of different sterotype,and the same sterotype of L.monocytogenes from different sources also could be identified.Moreover,the species-spcific 1600bp DNA fragment was obtained from the fingerprint of L.monocytogenes.The study indicates that ERIC-PCR technique can be used in the identification of Listeria species and strains and its further typing,which is simple and quickly.  相似文献

In experiments on Black Sea skates (Raja clavata), the potential of the receptor epithelium of the ampullae of Lorenzini and spike activity of single nerve fibers connected to them were investigated during electrical and temperature stimulation. Usually the potential within the canal was between 0 and –2 mV, and the input resistance of the ampulla 250–400 k. Heating of the region of the receptor epithelium was accompanied by a negative wave of potential, an increase in input resistance, and inhibition of spike activity. With worsening of the animal's condition the transepithelial potential became positive (up to +10 mV) but the input resistance of the ampulla during stimulation with a positive current was nonlinear in some cases: a regenerative spike of positive polarity appeared in the channel. During heating, the spike response was sometimes reversed in sign. It is suggested that fluctuations of the transepithelial potential and spike responses to temperature stimulation reflect changes in the potential difference on the basal membrane of the receptor cells, which is described by a relationship of the Nernst's or Goldman's equation type.I. Evolution of living organisms is closely connected with evolution of structure of the system of regulations and its mechanisms. The functional ground of regulations is chemical signalization. As early as in unicellular organisms there is a set of signal mechanisms providing their life activity and orientation in space and time. Subsequent evolution of ways of chemical signalization followed the way of development of delivery pathways of chemical signal and development of mechanisms of its regulation. The mechanism of chemical regulation of the signal interaction is discussed by the example of the specialized system of transduction of signal from neuron to neuron, of effect of hormone on the epithelial cell and modulation of this effect. These mechanisms are considered as the most important ways of the fine and precise adaptation of chemical signalization underlying functioning of physiological systems and organs of the living organism  相似文献
