1. The time-course of changes in content of intermediates of glycolysis in rat liver and kidney cortex after severance of blood supply was investigated. 2. The decline in content of ATP was more rapid in kidney (1.7-0.5mumol/g in 30s) than in liver (2.7-1.6mumol/g in 60s). In both tissues AMP and P(i) accumulated. 3. Net formation of lactate was 1.7mumol/g during the second minute of ischaemia in liver from well-fed rats, 1.1mumol/g in liver from 48h-starved rats, and about 1.0mumol/g during the first 30s of ischaemia in kidney. Net formation of alpha-glycerophosphate was rapid, especially in liver. 4. In kidney the concentration of beta-hydroxybutyrate rose, but that of alpha-oxoglutarate and acetoacetate decreased. 5. In both organs the concentrations of fructose diphosphate and triose phosphates increased during ischaemia and those of other phosphorylated C(3) intermediates decreased. 6. The concentration of the hexose 6-phosphates rose rapidly during the first minute of ischaemia in liver, but decreased during renal ischaemia. 7. In kidney the content of glutamine fell after 2min of ischaemia, and that of ammonia and glutamate rose. 8. The redox states of the cytoplasmic and mitochondrial NAD couple in kidney cortex were similar to those in liver. 9. The regulatory role of glycogen phosphorylase, pyruvate kinase and phosphofructokinase is discussed in relation to the observed changes in the concentrations of the glycolytic intermediates.  相似文献

A mixture of testosterone-4-14C and testosterone-1,2-3H-17-glucosiduronate was intraperitoneally administered into male and female rats with bile fistulas. Biliary metabolites were separated and purififd by a combination of column chromatography, enzymic hydrolysis or solvolysis of the conjugate fractions and identification of the liberated aglycones. The injected steroids were extensively metabolized and excreted predominantly in the blue. 5β-Androstane-3α, 17β-diol was found principally in monoglucosiduronate fraction and was produced preferentially from the injected conjugate in both sexes. Very marked sex differences from the injected conjugate in both sexes. Very marked sex differences were observed in the following metabolites: Androsterone was present only in the female as monoglucosidironate, which was preferentially derived from testosterone. 5α-Androstane-3α,17β-diol was identified in both monoglucosiduronate and diconjugate fractions of the female, which was formed significanrly more from the conjugate than testosterone. These findings provide evidence that testosterone glucosiduronate could be converted directly into 5α-steroids as well as 5β-ones invivo. In marked contrast, the major portion of testosterone was metabolized to polar steroids in the male.  相似文献

The distribution of HCO3?-ATPase activity was studied in cell fractions prepared from homogenates of rat liver. The level of mitochondrial contamination in the microsomal fraction depended on the fractionation procedure and on the method of homogenization. With proper care, microsomes with undetectable mitochondrial contamination could be prepared. These microsomes had no detectable HCO3?-ATPase activity. Approximately 85 % of the total HCO3?-ATPase activity of the post 6000 x g · min supernatant was recovered in the mitochondrialfraction. The properties of this mitochondrial HCO3?-ATPase were not distinguishable from those of the various microsomal HCO3?-ATPase previously described by other investigators.  相似文献

Conjugated linoleic acid (CLA) is known to provide certain health benefits in experimental animal models. The major CLA isomer in food is c 9,t11-CLA. A primary objective of this study was to investigate the uptake of c 9,t11-CLA and its downstream metabolites into various lipid fractions in the liver of rats fed either a high or low CLA diet (containing 0.1 or 0.8 g CLA/100 g diet, respectively). As expected, the levels of all conjugated diene (CD) fatty acids (CD 18:2 + CD 18:3 + CD 20:3 + CD 20:4) were elevated about 8-fold in the high CLA diet group. However, there was no change in the distribution of CLA and CLA metabolites into various lipid fractions due to CLA intake. Unlike linoleic acid or gamma-linolenic acid, which were distributed mainly in phospholipids, CD 18:2, CD 18:3, and CD 20:3 were incorporated primarily in neutral lipid. Furthermore, the incorporation of all nonconjugated unsaturated fatty acids was not perturbed by CLA. Regardless of the level of CLA in the diet, CD 20:4 was predominantly enriched in phosphatidylserine and phosphatidylinositol. In contrast, arachidonic acid was primarily enriched in phosphatidylcholine and less so in phosphatidylethanolamine. The above findings may have potential implication regarding the role of CLA in modulating eicosanoid metabolism.  相似文献

Sedoheptulose 1,7-bisphosphate has been shown to be present in extracts of normal rat liver. Its concentration in this tissue, estimated by colorimetric and enzymatic assays, is in the range of 5–7 nmol/g tissue. The concentration of sedoheptulose 7-phosphate in these extracts was 110 nmol/g tissue. Also present were mono- and bisphosphate esters of d-glycero-d-ido-octulose and d-glycero-d-altro-octulose, in concentrations ranging from 1–10 nmol/g tissue. Sedoheptulose 1,7-bisphosphate may function as a reservoir for erythrose 4-phosphate. The possible origin of the eight-carbon sugars and their function are discussed.  相似文献

Hydroxylation of benzylic methyl carbon atoms on drugs and carcinogenic polycyclic aromatic hydrocarbons (PAHs) forms benzylic alcohols. Many carcinogenic and mutagenic PAHs bear a primary or secondary benzylic hydroxyl group attached to the meso-region of the molecule. According to the unified theory, PAHs bearing a benzylic hydroxyl group are proximate carcinogenic metabolites. This paper demonstrates that carcinogenic benz[a]anthracenes bearing a formyl group at the meso-region undergo enzymatic reductive metabolism to the corresponding carcinogenic benzylic alcohol in vitro and in vivo. The unified theory would then predict sulfuric acid esterification of such benzylic alcohols as the final common step in their metabolic activation to generate ultimate electrophilic benzylic carbocations. Finally, oxidative metabolism of 7-formylbenz[a]anthracenes gives rise to corresponding carboxylic acids and other oxygenated metabolites that are carcinogenically inert. Thus, oxidative metabolism of meso-region formyl compounds represents an avenue for the elimination of the carcinogen in a detoxified form.  相似文献

Subcellular fractionation of male rat kidney revealed that the nuclear and plasma membrane fractions isolated from the 1,000 g pellet retained a significant proportion of the aldosterone ring-A reducing activity. Improved HPLC solvent systems separated all six possible ring-A reduced metabolites of aldosterone and revealed that 80-90% of the reduced metabolites synthesized by purified nuclei and plasma membranes were 5 alpha-reduced compounds consisting of 5 alpha-DHA and 3 alpha,5 alpha-THA in ratios of 1:2 (nuclei) and 1:1 (membranes). The 105,000 g cytosol also synthesized significant quantities of reduced, hydroxylated, and conjugated metabolites of aldosterone. In contrast, the majority of the reduced metabolites of aldosterone synthesized by kidney cytosol were 5 beta-products, consisting principally of 5 beta-DHA and smaller quantities of 3 alpha,5 beta-THA and 3 beta,5 beta-THA. The synthesis of reduced aldosterone metabolites in the cytosol, nuclear, and plasma membrane fraction was inhibited by both 5 and 50 microM concentrations of the antimineralocorticoids, progesterone, K+-canrenoate, and corticosterone. Progesterone was the strongest inhibitor of the synthesis of 5 alpha-DHA and 3 alpha,5 alpha-THA in both nuclei and plasma membranes. The overall order of inhibition of the synthesis of ring-A reduced metabolites in the kidney subcellular fractions was progesterone greater than K+-canrenoate greater than corticosterone; both progesterone and K+-canrenoate inhibited 5 alpha-reduction more than 5 beta-reduction.  相似文献

Microsomal UDP-glucuronyl transferases (uridine-5′-diphosphate-glucuronate glucuronyl transferase, acceptor unspecific, EC 2.4.1.17) function in detoxification of a wide range of aglycons containing phenolic, alcoholic, carboxylic or amino acceptor groups through formation of water-soluble glucuronic acid derivatives. To localize this activity within a specific cell component, purified rough-surfaced endoplasmic reticulum and Golgi apparatus fractions from rat liver were compared with total homogenates. The UDP-glucuronyl transferase specific activity of the Golgi apparatus fractions was less than or approximately equal to that of the total homogenate whereas this activity was concentrated 3–8 fold in the rough-surfaced endoplasmic reticulum fraction depending upon substrate. We conclude that the rough-surfaced endoplasmic reticulum is the major site of glucuronide formation within the microsome fraction of normal rat livers.  相似文献

The metabolism of testosterone (T) was examined during the second half of pregnancy in the rat to determine whether utilization of T for estradiol (E2) synthesis occurs via conversion of T to androstenedione (A). On Days 11, 16, and 21 of gestation (term = Day 23), rats (n = 7-9/group) were anesthetized and a constant infusion of [3H]T was initiated. At 60 min, blood was obtained from a jugular vein and the ovaries (Days 11, 16, and 21), and placentae and uterine tissue (Day 16 only) were removed. In a second study performed in rats on Day 16 of gestation (n = 8-10/group), the ovaries and/or gravid uterus were removed 15 min after initiation of [3H]T infusion, and blood was taken from a jugular vein 60 min later. Radiolabeled T and A were purified from serum and tissues by paper chromatography. In a third group of rats (n = 6), jugular vein samples were obtained sequentially on Days 11, 16 and 21 of gestation and serum concentrations of T were measured by radioimmunoassay. The metabolic clearance rate of T was constant during the study period (overall mean = 31 1/day). In contrast, the serum concentration of T (pg/ml) on Day 16 of gestation (863 +/- 108) exceeded (p less than 0.02) that on Day 11 (445 +/- 74); the latter was similar to that measured on Day 21 (592 +/- 109).
