In 33 patients with chronic lymphatic leukaemia (CLL) the lipids were examined quantitatively and qualitatively in the serum and in the lymphocytes. Besides the 5 main categories of lipids, lysolecithin, sphingomyelin, lecithin and cephalin as well as cholesterol ester subfractions were determined according to their degree of saturation of their fatty acids. Patients suffering from CLL were found to have a moderately raise of serum triglycerides and an increase of cholesterol esters with simple unsaturated fatty acids. Simultaneously the fraction of ester with poly-unsaturated fatty acids is lowered. Leukaemic lymphocytes reveal in general a lowered total lipid content. It mainly consists of phosphatides and free cholesterol. Moreover, the leukaemic lymphocytes contrary to normal ones reveal a lowered content of cholesterol esters, diglycerides and triglycerides.  相似文献

Studies have been made on the lipid composition of total lipids, triglycerides and their fatty acids, cholesterol and phospholipids in the vertebral column of young and adult rabbits. It was shown that the content of total lipids and triglycerides increases, whereas that of cholesterol and phospholipids decreases with age. The content of total lipids in the vertebral column is 10 times higher as compared to that in the bones of the extremities. Mid-thoracic part of the vertebral column exhibits higher lipid content than other thoracic parts of the column. Lipid content of the vertebral processes is lower than that of the vertebral bodies. These data indicate lipid specificity and heterogeneity of bone tissue of the vertebral column. The main fatty acids of vertebral triglycerides are presented by those with 14-18 carbon atoms (90%), no acids with 22 atoms were found. Higher content of the linoleic acid (19%) and higher total unsaturation of triglycerides were found in the bone tissue of rabbits in comparison with those of man.  相似文献

Anorexia nervosa is a model of simple starvation accompanied by secondary hyperlipoproteinemia. The pattern of plasma fatty acids influences the levels of plasma lipids and lipoproteins. The concentration of plasma lathosterol is a surrogate marker of cholesterol synthesis de novo, concentrations of campesterol and beta-sitosterol reflect resorption of exogenous cholesterol. The aim of the study was to evaluate fatty acids in plasma lipid classes and their relationship to plasma lipids, lipoproteins, cholesterol precursors and plant sterols. We examined 16 women with anorexia nervosa and 25 healthy ones. Patients with anorexia nervosa revealed increased concentrations of total cholesterol, triglycerides, HDL-cholesterol, campesterol and beta-sitosterol. Moreover, a decreased content of n-6 polyunsaturated fatty acids was found in all lipid classes. These changes were compensated by an increased content of monounsaturated fatty acids in cholesteryl esters, saturated fatty acids in triglycerides and both monounsaturated and saturated fatty acids in phosphatidylcholine. The most consistent finding in the fatty acid pattern concerned a decreased content of linoleic acid and a raised content of palmitoleic acid in all lipid classes. The changes of plasma lipids and lipoproteins in anorexia nervosa are the result of complex mechanisms including decreased catabolism of triglyceride-rich lipoproteins, normal rate of cholesterol synthesis and increased resorption of exogenous cholesterol.  相似文献

Low density lipoprotein (LDL) isolated from the blood of healthy donors was partially desialylated by incubating the lipoprotein with sialidase (neuraminidase). The addition of LDL treated with neuraminidase to cultured human aortic intimal cells of smooth muscle origin caused a substantial increase in intracellular cholesteryl esters, free cholesterol and triglycerides. Cultured cells took up and degraded desialylated LDL much more effectively than untreated (native) LDL. LDL were also isolated from an atherogenic blood plasma of patients with coronary artery disease, i.e. the plasma capable of inducing the accumulation of lipids in cultured cells. Patients' LDL, similarly to the mother plasma, were atherogenic, i.e. stimulated the accumulation of intracellular lipids. LDL isolated from nonatherogenic plasma of healthy donors proved to be nonatherogenic. Atherogenic patients' LDL had a 2- to 5-fold lower level of sialic acid as compared with nonatherogenic LDL of healthy donors. The uptake and degradation of atherogenic patients' LDL were much more effective than in the case of nonatherogenic LDL of healthy donors. We assume that atherogenic properties of LDL obtained from patients' blood plasma are explained exactly by a low sialic acid content.  相似文献

We have previously shown that transgenic expression of catalytically inactive lipoprotein lipase (LPL) in muscle (Mck-N-LPL) enhances triglyceride hydrolysis as well as whole particle lipoprotein and selective cholesterol ester uptake. In the current study, we have examined whether these functions can be performed by inactive LPL alone or require the presence of active LPL expressed in the same tissue. To study inactive LPL in the presence of active LPL in the same tissue, the Mck-N-LPL transgene was bred onto the heterozygous LPL-deficient (LPL1) background. At 18 h of age, Mck-N-LPL reduced triglycerides by 35% and markedly increased muscle lipid droplets. In adult mice, it reduced triglycerides by 40% and increased lipoprotein particle uptake into muscle by 60% and cholesterol ester uptake by 110%. To study inactive LPL alone, the Mck-N-LPL transgene was bred onto the LPL-deficient (LPL0) background. These mice die at approximately 24 h of age. At 18 h of age, in the absence of active LPL, inactive LPL expression did not diminish triglycerides nor did it result in the accumulation of muscle lipid droplets. To study inactive LPL in the absence of active LPL in the same tissue in adult animals, the Mck-N-LPL transgene was bred onto mice that only expressed active LPL in the heart (LPL0/He-LPL). In this case, Mck-N-LPL did not reduce triglycerides or increase the uptake of lipoprotein particles but did increase muscle uptake of chylomicron and very low density lipoprotein cholesterol ester by 40%. Thus, in the presence of active LPL in the same tissue, inactive LPL augments triglyceride hydrolysis and increases whole particle triglyceride-rich lipoprotein and selective cholesterol ester uptake. In the absence of active LPL in the same tissue, inactive LPL only mediates selective cholesterol ester uptake.  相似文献

Lipids of blood plasma of lampreys and frogs are composed of phospholipids, triglycerides, free fatty acids (FA), cholesterol, cholesterol esters, and waxes. The lipids content in plasma of frogs is markedly lower as compared with that of lampreys. However, the percentage of lipid components is represented by close values. Fluidity of triglycerides and phospholipids in lampreys is determined predominantly by monoenic acids and polyenoic acids of the ω-3-type, whereas that in frogs—by monoenic acids and polyenic acids of the ω-6-type. Free FA are represented mainly by saturated and monoenic acids.  相似文献

Neutral lipids, particularly triglycerides, accounted for the major decrease in the total lipid content in Paramecium cells that occurs with culture age. Sterols, triglycerides, and steryl esters were the major classes of neutral lipids in cells and isolated cilia. Free as well as high concentrations of esterified sterols were detected in purified ciliary membrane preparations. Stigmasterol and 7-dehydrostigmasterol were the major components of both free and esterified sterols of cells and cilia; however, when cholesterol was present in the growth medium, it was desaturated to 7-dehydrocholesterol and incorporated into cellular and ciliary lipids. Free fatty acids from cells and triglycerides from cells and cilia were low in polyunsaturated fatty acids and reflected the composition of fatty acids in the culture medium. An exception was the reduced concentration of stearate in triglycerides from whole cells. Greater than 50% of triglyceride fatty acids from cilia were saturated. The fatty acid compositions of cellular triglycerides and ciliary steryl esters did not change with culture age, but those of cellular steryl esters and ciliary triglycerides did change. In comparison with phospholipids, these neutral lipid fatty acid compositional changes were smaller. The sensitivity of these stigmasterol-containing cells to polyene antibiotics indicated that they were killed by nystatin > filipin > amphotericin B. The unexpected finding of high concentrations of steryl esters in ciliary membrane preparations is discussed.  相似文献

Cells derived from Antheraea eucalypti were grown in vitro in a medium containing triglycerides, diglycerides, free fatty acids, and sterols as the main ‘neutral’ lipids. The sterol content of the medium was derived chiefly from the haemolymph component. The ‘neutral’ lipids in the cells were triglycerides, free fatty acids, and sterols. During growth over 6 days there was a quantitative balance between cholesterol and β-sitosterol gained by the cells and those sterols removed from the medium when allowance was made for losses from sterile medium. Cells metabolized more triglycerides and free fatty acids than they incorporated.  相似文献

Coconut oil is rich in medium chain fatty acids, but deficient in polyunsaturated fatty acids (PUFA). Structured lipids (SL) enriched with omega 6 PUFA were synthesized from coconut oil triglycerides by employing enzymatic acidolysis with free fatty acids obtained from safflower oil. Rats were fed a diet containing coconut oil, coconut oil-safflower oil blend (1:0.7 w/w) or structured lipid at 10% levels for a period of 60 days. The SL lowered serum cholesterol levels by 10.3 and 10.5% respectively in comparison with those fed coconut oil and blended oil. Similarly the liver cholesterol levels were also decreased by 35.9 and 26.6% respectively in animals fed structured lipids when compared to those fed on coconut oil or the blended oil. Most of the decrease observed in serum cholesterol levels of animals fed structured lipids was found in LDL fraction. The triglyceride levels in serum showed a decrease by 17.5 and 17.4% while in the liver it was reduced by 45.8 and 23.5% in the structured lipids fed animals as compared to those fed coconut oil or blended oil respectively. Differential scanning calorimetric studies indicated that structured lipids had lower melting points and solid fat content when compared to coconut oil or blended oils. These studies indicated that enrichment of coconut oil triglycerides with omega 6 fatty acids lowers its solid fat content. The omega 6 PUFA enriched structured lipids also exhibited hypolipidemic activity.  相似文献

Alteration of the rabbit serum lipids as a result of three bacterial infections was studied by quantitative thin-layer and gas-liquid chromatography. Anthrax infection slightly changed the serum lipid. Cholesterol did not change, though free fatty acids, triglycerides, and cholesteryl esters doubled, and lecithin increased threefold. Tularemia infection produced drastic changes in the serum lipid content of rabbits, increasing levels of cholesterol over 4-fold, free fatty acids 17-fold, triglycerides 11-fold, cholesteryl esters 2.5-fold, and lecithin almost 3-fold. Pneumococcus infection increased cholesterol 2.5 times, free fatty acids were more than doubled, triglycerides were increased 9.5 times, and lecithin was increased almost 4 times. Gas-liquid chromatographic analysis of the methyl esters of free fatty acids showed only quantitative changes in these acids due to infection. Some possible mechanisms of alteration of serum lipid content are discussed.  相似文献

The effect of aflatoxin B1 on lipids of liver, kidney, adipose and plasma of rats was studied. A single dose administration (6 mg/kg body weight) increased liver and kidney weights and their total lipids within 24 h. Increase in liver lipids was confined mainly to phospholipid and cholesterol, whereas triglycerides showed a significant decrease. Adipose tissue triglycerides were, however, increased. Plasma showed decreases in triglycerides, free fatty acids and cholesterol. Incorporation studies with palmitate-1-14C revealed increased incorporation in adipose tissue lipids and decreased incorporation in liver and plasma lipids, thereby indicating an increased synthesis of lipids in adipose. Their mobilization to plasma was, however, inhibited, hence the low levels of triglyceride in liver. But the adrenals showed hypo-activity upon aflatoxin B1 administration.  相似文献

Pulmonary surfactant was isolated from human lung homogenate after differential and sucrose density gradient centrifugation. Purification of the isolated material was ascertained by electron microscopy and alkaline phosphatase (AKP) specific activity. Elevated levels of phospholipid/protein ration and AKP specific activity were observed in the purified material when monitored at different stages of purification. Biochemical analysis of the isolated material showed that it consisted of 74.08% lipid, 19.06% protein and relatively smaller amounts of nucleic acids, sialic acid and hexoses. Phosphatidyl choline was the predominant phospholipid whereas triglycerides and cholesterol levels were high among neutral lipids. Gas liquid chromatography of the material showed palmitic acid (16:0) as the major saturated fatty acid. These findings indicated that a large scale isolation of surfactant might be possible and utilized for therapeutic treatment of neonatal respiratory distress syndrome.  相似文献

The effect of various doses of sodium tellurite (1/50 LD50=0.4 mg/kg, 1/25 LD50=0.8 mg/kg, and 1/10 LD50=2.0 mg/kg body weight orally) on the lipid levels (cholesterol, triglycerides, phospholipids, esterified fatty acids, gangliosides, and total lipids) in the cerebrum, cerebellum, and brainstem of male albino mice was studied after 7 and 15 d of treatment. Sodium tellurite (2.0 mg/kg body weight) for 7 d has an apparent effect on the depletion of cholesterol, triglycerides, phospholipids, esterified fatty acids, and total lipids. The cholesterol content was decreased significantly in the cerebrum, cerebellum, and brainstem after 7 d of treatment with a 2.0-mg/kg dose compared to the control. On the other hand, treatment for 15 d with doses of 0.4, 0.8, and 2.0 mg/kg body weight resulted in a significant and dose-dependent increment in cholesterol level in the cerebrum, cerebellum, and brainstem. The triglycerides content was decreased significantly in the cerebrum, cerebellum, and brainstem with the 2.0-mg/kg dose after 7 d of treatment. The doses of 0.4, 0.8, and 2.0 mg/kg orally for 15 d resulted in a significant and dose-dependent depletion of triglycerides in the cerebrum, cerebellum, and brainstem. All the doses of tellurium (0.4, 0.8, and 2.0 mg/kg) both for 7 and 15 d have depleted the level of phospholipids in varying degrees of significance in the cerebrum, cerebellum, and brainstem. However, the level of esterified fatty acids was decreased significantly with the 2.0-mg/kg dose of tellurium for 7 d but increased with the 0.4-mg/kg dose for 15 d in the cerebrum and cerebellum. The level of gangliosides was depleted in the cerebrum but elevated in the cerebellum and brainstem after receiving a 2.0-mg/kg dose of sodium tellurite for 7 d. The content of gangliosides was increased with doses of 0.4 and 0.8 mg/kg but decreased with 2.0 mg/kg for 15 d in the cerebrum, cerebellum, and brainstem. The total lipids content was depleted significantly and dose dependently after 7 and 15 d of treatment in the cerebrum, cerebellum, and brainstem. These results suggest that sodium tellurite affects the lipids content differentially in various parts of the mice brain.  相似文献

The highly optically active triglycerides (OAT) of the seed oil of Sebastiana ligustrina were isolated and characterized. As with the similar known triglycerides from Sapium sebiferum, the optically active 8-hydroxy-5,6-octadienoic acid is esterified to glycerol and to 2,4-decadienoic acid. The other two acyl moieties in the triglycerides of both these species are common fatty acids, principally 16:0, 18:1, 18:2 and 18:3 which occur in pairs ranging in degree of unsaturation from 16:0, 18:1 to 18:3, 18:3. Analyses of lipids of parts of the related species Stillingia sylvatica and S. texana revealed no allene or carbonyl conjugation, and thus by implication no OAT. A substance having IR absorptions characteristic of allenes and carbonyl conjugated dienes, but different from OAT, was found in the stems of S. texana. The seeds of S. sebiferum are known to contain OAT, but it was not found in fresh leaves.  相似文献

The fatty acid pattern of blood serum lipids was examined by gas liquid-chromatography in 30 cases with bilharzial hepatic fibrosis, 11 cases with chronic active hepatitis accompanied by jaundice, and 28 healthy individuals as a comparison group of the same socioeconomic class of patients. In addition, the fatty acid patterns of the three major serum lipid classes, namely: cholesterol ester, phospholipids and triglycerides, were also investigated in seven cases of each group by gas liquid chromatography. The most remarkable differences were: a depression of the essential fatty acid level (linoleic and arachidonic) in both groups of patients together with a concomitant elevation of oleic acid in the bilharzial group and an elevation of oleic, palmitic, palmitoleic acids in the chronic active hepatitis group. The depression of linoleic and arachidonic acids was explained by the low fat diet intake, malnutrition, and the malabsorption factors which were frequent in all the patients studied. The elevation of monoethenoid acids was attributed to the decrease in the ability of the liver to desaturate the endogenous saturated and monounsaturated acids to polyunsaturated ones.  相似文献

1. Leucocytes from the blood of adult and young donkeys (Equus asinus L.), adult horses (Equus caballus L.), adult mules (Equus asinus x Equus caballus) and adult pigs (Sus scrofa L.) were obtained in a high degree of purity (99.9%) using Na2-EDTA-dextrans mixtures. 2. Sialic acids were released, purified, identified and determined from both non-delipided and delipided leucocytes. 3. N-glycolylneuraminic was the predominant sialic acid. N-acetylneuraminic acid and N,O-diacetyl-neuraminic acid were also found in all materials. Except in pig, other unidentified sialic acid(s) were also detected. 4. The concentration of total sialic acids (microgram/mg protein) is different according to the species, and in donkey species according to the age. 5. Galactose, glucose, mannose, fucose and (in a less amount) ribose were determined. Their total content is about 2-3-fold that of hexosamines. 6. There is a higher cholesterol content in adult donkey leucocytes than in those of young ones. 7. Total lipids, cholesterol or phospholipid contents are similar among the leucocytes of the above-mentioned species. 8. The similarities are marked in the electrophoresis patterns of proteins and glycoproteins for the donkey, mule and horse samples. 9. The molecular weights for leucocytes proteins were estimated as ranging between 230,000 and 20,000; and for the main protein bands, between 120,000 and 22,000.  相似文献

Primary cultures of rabbit hepatocytes were used to investigate the effect of purified (B-100 free) chylomicron remnants (CR) on lipid and bile acid metabolism. ApoB-100-containing lipoproteins were removed from the CR-enriched plasma fraction by affinity column chromatography on Sepharose 4B conjugated with anti-apoB-100 monoclonal antibodies. CR were shown to stimulate the accumulation of neutral lipids in hepatocytes in a dose-response manner. After 24-hour preincubation of rabbit hepatocytes with 50 micrograms protein/ml CR the cellular neutral lipid content increased: 1.9-4-fold for triglycerides, 1.5-3.7-fold for free cholesterol and 1.5-2.5-fold for esterified cholesterol. This accumulation was accompanied by a decreasing incorporation of [14C] acetate into cholesterol (80-90%) and triglycerides (70-80%). At the same time the incorporation of [14]oleate into triglycerides increased by 50-65%. The inhibited biosynthesis of fatty acids might account for this effect. No effect of CR on cholesterol esterification by [14C]oleate was observed. CR increased the amount of triglycerides and free cholesterol secreted in very low density lipoproteins (VLDL). The secretion of taurocholic acid was decreased. These data confirm our hypothesis that dietary cholesterol is preferentially secreted by hepatocytes within VLDL but is not accumulated as cholesterol esters or oxidized to bile acids.  相似文献

Synopsis Preliminary bromination (with bromine water) increases the intensity of staining of tissue lipids with Sudan Black and certain other dyes. The mechanism appears to be due to the formation of sudanophilic bromo-derivatives of cholesterol and to the retention of certain other lipids, notably phosphatidyl choline and free fatty acids, during staining. The advantage of the bromine-Sudan Black method is that all tissue lipids are stained, except saturated fatty acids, saturated triglycerides and perhaps saturated cholesterol esters. In practice, such lipids rarely, if ever, occur alone, and normally are admixed with their stainable unsaturated counterparts.  相似文献

1. A review is given of the optical and architectural analogies between cholesteric liquid crystals and certain insect cuticles (Coleoptera: Scarabaeidae). Earlier observations on the optical properties (reflexion of circularly polarized light and high form optical rotation) are confirmed and extended. Both cholesteric liquid crystals and lamellate cuticle have helicoidal structure (Fig. i). Even though their chemistry and physical states are very different, we are justified in making the analogy, since their optical properties depend primarily on the pitch of their helicoidal architecture. 2. The unusual optical properties were located for the first time in the outer 5 to 20 μ of the exocuticle. This layer is transparent and has regular spacings in the range required for interference colours according to Bragg's law. Among Scarabaeid beetles which show interference colours, we distinguish two types of outer exocuticle. (i) Optically active cuticles which reflect circularly polarized interference colours; show high angles of form optical rotation in transmitted light; and anomalous form birefringence perpendicular to the cuticle surface (reversible by deproteinization). (2) Optically inactive cuticles which show none of the above properties and in which the form birefringence is parallel to the cuticle surface. In the electron microscope the ultrastructure of these two types of outer exocuticle is clearly different. 3. All of the optically active species reflect left hand circularly polarized light, irrespective of the wavelength of the reflected colour. They therefore appear dark when viewed through a right hand circular analyser. The sense of reflected circularly polarized light does not reverse at higher wavelengths as recorded by previous workers. (A simple treatment is given for combinations of various wavelengths with retardation plates of varying values, as used in circular analysers.) We confirm earlier reports that the sense of reflected circularly polarized light is of the opposite sense to the transmitted light. 4. Using monochromatic light we have measured the anomalous dispersion with wavelength of the magnitude of optical rotation for various optically active cuticles. The dispersion curves change from negative values at lower wavelengths to positive values at higher wavelengths, and cross the zero optical rotation axis at a wavelength (AQ) corresponding to the interference colour of each sample. There is reasonable agreement between A0 and the interference colour calculated from ultrastructural evidence and by comparison with interference filters of known wavelength. A dispersion curve measured for a combined sample of two cuticles with different dispersion curves showed that the resultant is an algebraic summation of the two component curves. 5. We present the first experimental verification of existing mathematical treatments of anomalous form optical rotatory dispersion curves. Although these treatments were derived for cholesteric liquid crystals, they give a reasonable fit to our measured curves for cuticle. We have confirmed from our cuticle dispersion curves that a second zero value for optical rotation occurs at a wavelength higher than A0, as predicted by the theory of Chandrasekhar and Rao (1968). This has not yet been observed in any cholesteric liquid crystal system. 6. Our evidence shows that in optically active cuticle, interference colour is determined by helicoid pitch. In Lomaptera interference coloration follows the bilateral symmetry of the insect. Hence helicoidal pitch is controlled in a bilaterally symmetrical manner. However, the sense of helicoid rotation is the same all over the beetle and is therefore bilaterally asymmetrical. This supports the view that helicoid pitch is under the local control of the epidermal cells which secrete the cuticle, whereas its sense of rotation may be determined by an extracellular self-assembly process. In view of the self-assembling properties of cholesteric liquid crystals, it is tempting to suggest that helicoidal cuticle could be formed by the stabilization of a liquid crystal. 7. We discuss in detail the differences between optically active and inactive cuticles. The constructive interference colours arising from both types are then briefly compared with other multiple layer reflecting systems in other animals. 8. A detailed comparison is made between the optics of cuticle and cholesteric liquid crystals. The optical analogy provides a two-way contact between cuticle biophysicists and liquid crystal physical chemists.  相似文献
