Summary.  Some species of Spirogyra form rosette-shaped or rod-shaped rhizoids in the terminal cell of the filaments. In the present study, we analyzed an involvement of microtubules (MTs) in rhizoid differentiation. Before rhizoid differentiation, cortical MTs were arranged transversely to the long axis of cylindrical cells, reflecting the diffuse growth. At the beginning of rhizoid differentiation, MTs were absent from the extreme tip of the terminal cell. In the other area of the cell, however, MTs were arranged transversely to the long axis of the cell. In the fully differentiated rosette-shaped rhizoid, MTs were randomly organized. However, at a younger stage of rosette-shaped rhizoids, MTs were sometimes arranged almost transversely in the lobes of the rosette. In the rod-shaped rhizoid, MTs were arranged almost transversely. MT-destabilizing drugs (oryzalin and propyzamide) induced swelling of rhizoids, and neither rosette-shaped nor rod-shaped rhizoids were formed. The role of MTs in rhizoid differentiation was discussed. 报道了贵州金盏苣苔属Isometrum一新种,即万山金盏苣苔I.wanshanenseS.Z.He。该种以花序梗、花梗被褐色长柔毛至近无毛,花冠细筒状,紫色,喉部不缢缩,上唇稍长于下唇,雄蕊及雌蕊均无毛而与柔毛金盏苣苔I.villosumK.Y.Pan相近缘,但叶具柄,柄长0.4–1.4cm,叶片较小,倒披针形,长1.5–4×0.5–1.6cm,聚伞花序具1–4朵花,花萼裂片外面近无毛或近顶端疏被褐色长柔毛,花柱与子房近等长而不同。  相似文献

By methylation analysis, it was found that the cell walls of Spirogyra contained 4,6-linked glucose, 4-linked glucose and terminal xylose, which could be components of xyloglucan. Immunocytochemical analysis was carried out using an anti-serum against xyloglucan. After removal of pectic substances, the cell walls of both rhizoid cells and inner cells were stained. Crude protein extract from Spirogyra had a hydrolase activity for xyloglucans. In addition, the exogenously applied xyloglucan prevented the detachment of the cell wall of the severed cell. Involvement of xyloglucan-like polysaccharide in cell–cell attachment was discussed.  相似文献

In recent years green nanotechnology gained significant importance to synthesize nanoparticles due to their cost effectiveness and biosafety. In the current study, silver nanoparticles were synthesized by using extract of Spirogyra hyalina as a capping and reducing agent. The synthesized nanoparticles were characterized by UV–Visible spectroscopy, Fourier transform infrared spectroscopy, Scanning electron microscopy, energy dispersive X-ray spectroscopy, and X-ray diffractive analysis. Silver nanoparticles give a characteristic Surface Plasmon Resonance peak of 451 nm at 2.21 a.u (arbitrary unit). SEM micrograph revealed the spherical morphology and average grain size of 52.7 nm. Furthermore, antibacterial, antifungal, insecticidal, antioxidant and membrane damage activities were determined. The maximum antibacterial and antifungal activity was observed for Pseudomonas aeruginosa (18 ± 1.2 mm) and Fusarium solani (14.3 ± 0.6 mm), respectively. In membrane damage assay, Pseudomonas aeruginosa absorbed A260 wavelength and gave maximum peak values of 0.286, 0.434 and 0.629 at 25, 35 and 45 µg/mL of silver nanoparticles. The membrane damage assay confirmed that nanoparticles are involved in bacterial cell membrane damage. At 500 ppm silver nanoparticles showed 30% mortality against Tribolium castaneum (a common grain pest). The silver nanoparticles also showed potent antioxidant activity and successfully scavenged the DPPH free radicals upto 53.43 ± 0.17, 43.26 ± 0.97, 31.39 ± 0.33, 24.62 ± 0.85, and 14.13 ± 0.12% at a concentration of 400, 200, 100, 50, and 25 µg/mL of nanoparticles, respectively. It is concluded that silver nanoparticles can easily be synthesized by using green algae Spirogyra hyalina as a capping and reducing agent. Silver nanoparticles showed potent biomedical activities and thus can be used for therapeutic applications invitro and invivo.  相似文献

报道了贵州省务川县发现的悬钩子属Rubus(蔷薇科Rosaceae)一新种——务川悬钩子R.wuchuanensis S.Z.He。该种在体态上与锯叶悬钩子R.wuzhianus L.T.Lu&Boufford相近,区别在于其叶片卵状披针形,边缘疏生小锯齿;叶柄较短,长0.7-1cm;花序为顶生稀疏总状花序,具花8-10朵:花梗长3.55cm;花瓣先端具骤突尖头。  相似文献

采用土壤平板法和稀释平板法,从采自贵州省的20份土壤样品中分离得到14个暗色丝孢菌分离物,隶属于11属12种。其中,浅色腐殖霉Humicola pallescens为一新种,绿色暗梗单孢霉厚垣变种Chloridium virescens var.chlamy dosporum和印度粘束霉Graphiumindicum为中国新记录。对新种及中国新记录作了较详细的描述并绘图。其余9个国内已报道种亦作了生境和分布地点的引证。所有研究菌株的干制培养物标本与活菌种均存放在山东农业大学植物病理学标本室(HSAUP)。  相似文献

The presence of algae can greatly reduce the amount of light that reaches submerged macrophytes, but few experimental studies have been conducted to examine the effects of algae on biomass and structure of submerged macrophyte communities. We constructed communities with four submerged macrophytes (Hydrilla verticillata, Egeria densa, Ceratophyllum demersum, and Chara vulgaris) in three environments in which 0 (control), 50 and 100% of the water surface was covered by Spirogyra arcta. Compared to the control treatment, the 100% spirogyra treatment decreased biomass of the submerged macrophyte communities and of all the four macrophytes except C. demersum. Compared to the control and 50% treatments, the 100% treatment significantly increased relative abundance of C. demersum and decreased that of E. densa. Therefore, the presence of S. arcta can greatly affect the productivity and alter the structure of submerged macrophyte communities. To restore submerged macrophyte communities in conditions with abundant algae, assembling communities consisting of C. demersum or similar species may be a good practice.  相似文献

为研究六价铬(Cr^6+)对水绵(Spirogyrasp.)的毒性效应和水绵对六价铬的毒性响应,实验设置5个暴露组(4、6、8、10和12mg/L)和1个对照组。结果表明:96h后各暴露组对水绵的生长均有抑制作用,铬浓度越高,藻细胞叶绿素a越低,显示出较明显的剂量一效应关系;六价铬对水绵的96h的Ec。值为7.25mg/L。细胞浸出液电导率在低浓度(4~6mg/L)组上升较缓慢,在高浓度组(8~12mg/L)上升显著;丙二醛(MDA)累积含量在Cr6+≥4mg/L时,累计含量上升较高(P〈0.01),当Cr^6+≥8mg/L时,累积含量上升较少。水绵细胞浸出液电导率与MDA存在显著正相关(P〈0.05,r=0.891)。水绵细胞浸出液电导率和MDA含量与六价铬浓度分别存在显著(P〈0.05,r=0.951)和极显著(P〈0.01,r=0.977)的非线性的毒性响应关系。  相似文献

Electron microscope observation revealed the presence of many fine filaments within the cytoplasm surrounding the leading edge of the septum in telophase cells of Spirogyra verruculosa Jao. These filaments, about 7 nm each in diameter, ran parallel to one another along the leading edge of the septum and, sometimes, they appeared to be gathered into at least two bundles. These filament distribution patterns coincided well with those of the fluorescence of rhodamine-labeled phalloidin in the vicinity of the septum in telophase cells. The present results suggest that the fine filaments observed within the cytoplasm surrounding the leading edge of the septum may be actin filaments.  相似文献

The contribution of microtubules and microfilaments to the cytomechanics of transverse nuclear centering were investigated in the charophycean green alga Spirogyracrassa (Zygnematales). Cytoplasmic strands of enhanced rigidity and fasciate appearance radiate from the rim of the lenticular nucleus through the vacuole, frequently split once or twice and are attached to the helical chloroplast bands in the peripheral cytoplasm. The nucleus is encased in tubulin and a web of F-actin. Bundles of microtubules, emerging from the nuclear rim, are organized into dividing fascicles within the strands and reach to the inner surface of the chloroplast envelope. Organelles are translocated in both directions along similarly arranged fascicles of microfilament bundles which extend from the nucleus to the peripheral actin cytoskeleton. Application of microtubule- and/or microfilament-depolymerizing drugs affected the position of the nucleus only slowly, but in distinct ways. The differential effects suggest that nuclear centering depends on the tensional integrity of the perinuclear scaffold, with microfilaments conveying tension along stabilized microtubules and the actin cytoskeleton integrating the translocation forces generated within the scaffold. 贵州疣螈(Tylototriton kweichowensis)为中国特有种,国家Ⅱ级重点保护野生动物,对生境变化具有重要指示意义。2018年9月至2019年10月,在贵州省毕节市撒拉溪石漠化综合治理示范区,对贵州疣螈栖息地、形态特征、繁殖行为进行了野外观测。调查显示,贵州疣螈栖息于山塘、山泉、蓄水池、临时性积水坑(塘)等水域,个体全长、尾长和体重雌螈均显著高于雄螈。贵州疣螈于4月18日雷雨天气后破眠外出活动,繁殖始于4月29日,最晚见于8月8日,高峰期为5至6月;繁殖期贵州疣螈的性比总体上偏雄,但在产卵期性比偏雌;雌、雄螈抱对时间几分钟到40min不等;抱对结束后开始排精、纳精;产卵活动在纳精后的1~2 d进行,卵产于繁殖场水底、草或石头上;卵的孵化率为55%,平均孵化期8d,幼体完成变态发育需130d。研究表明,贵州疣螈的繁殖与发育受降雨、水量、温度变化的影响较大,且繁殖场所相对较为固定,容易受到人类活动的干扰。因此,在石漠化治理和生态修复时注重贵州疣螈的栖息地保护,必要时应人工新建稳固繁殖场,保障其生态繁衍。  相似文献

Spirogyra is found in a wide range of habitats, including small stagnant water bodies, rivers, and streams. Spirogyra ellipsospora is common in northern Thailand. Species identification of the Spirogyra species based only on morphological characteristics can be difficult. A reliable and accurate method is required to evaluate genetic variations. This study aims to apply molecular approaches for the identification of S. ellipsospora using microsatellites and rbcL markers. Based on DNA sequencing, the rbcL gene was sequenced and the data was analyzed using the BLAST (Basic Local Alignment Search Tool) program in the NCBI (National Center for Biotechnology Information) database. The sequence of S. ellipsospora from this study revealed definitive identity matches in the range of 99% for the consensus sequences of S. ellipsospora. The 10 primers of ISSR could be amplified by 92 amplification fragments. The DNA fragments and the rbcL sequence data grouped the Spirogyra specimens into two distinct clusters.  相似文献
