以扬麦16和徐麦30为试验材料,利用人工气候室模拟低温逆境,研究拔节期-3 ℃和-5 ℃低温胁迫对小麦植株受冻率、叶片内源激素含量和抗氧化酶活性的影响.结果表明: 随着处理温度的降低、胁迫时间的延长,小麦植株冻害等级与冻害指数增加,-5 ℃处理72 h两品种五级冻害率均为100%.低温处理结束当天,小麦叶片中内源激素脱落酸(ABA)、玉米素核苷(ZR)含量、抗氧化酶超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性随胁迫程度加重呈先升高后降低的趋势;处理结束后3 d,ABA、ZR含量及抗氧化酶活性较处理结束当天升高;至处理结束后6 d,与自然生长的对照处理接近.低温胁迫叶片中赤霉素(GA3)含量下降,处理结束后3和6 d,扬麦16叶片中GA3含量呈上升趋势,徐麦30则表现为先升高后下降.-5 ℃ 72 h重度胁迫处理叶片中ABA、ZR、GA3含量和SOD、POD、CAT活性均较对照显著下降.相关分析表明,较高的ABA、ZR含量、SOD、POD、CAT活性以及较低的GA3含量可减缓低温胁迫对小麦植株的伤害.  相似文献

Under the same mannitol pretreatment and culture conditions, regeneration efficiency in the barley cultivar (cv.) Igri was about 10 times higher than in the cv. Digger, a difference only partially reflected by a difference in viable microspores after anther pretreatment. Therefore, a comparative study between cvs. Igri and Digger was carried out under various pretreatment conditions. For both cultivars, under water, CPW buffer and mannitol pretreatment conditions, there was a positive correlation between microspore viability and regeneration efficiency in that mannitol > CPW buffer >> water. Mannitol pretreatment of cv. Igri produced a much higher endogenous abscisic acid (ABA) level than as to Digger. Addition of ABA stimulated both percentages of viability and regeneration efficiency except in the case of mannitol pretreatment. Under CPW buffer pretreatment conditions, addition of ABA significantly stimulated regeneration efficiency and was ABA concentration dependent. However, cv. Digger was less responsive to ABA than cv. Igri. In both cultivars, under less optimal pretreatment conditions (e.g., water and CPW buffer), the effect of ABA was to stimulate increased percentages of viability and/or to reduce the number of binucleate microspores. Moreover, in cv. Igri, direct culture of anthers for 4 days without pretreatment caused an increased number of binucleate microspores compared with microspores with pretreatment for 4 days. These binucleate microspores showed DNA degradation in the nuclei. However, with mannitol pretreatment binucleate microspores and DNA fragmentation in the nuclei of microspores was rarely observed. On the basis of our observations, we suggest that the difference in regeneration efficiency in cv. Igri and cv. Digger is related to the differences in endogenous ABA production levels under mannitol pretreatment and responsiveness to ABA. One of the effects of ABA is likely due to an inhibition of cell death. 气相色谱分析的结果表明,开花后5~6天,不受精即将脱落棉铃里IAA和ABA的含量显著高于对照;GA_3处理的铃中IAA和ABA的含量则明显低于对照。IAA和GA_3含量在开花后第10天达到高峰,以后迅速下降,至第30天。又出现第二峰胚珠和纤维里尤为明显。ABA含量在开花后第15天达得高峰,以后下降,到第30天,又逐渐上升。棉纤维伸长及干重增加的最快速率与这三种激素的含量高峰密切相关。  相似文献

该试验以抗寒性不同的5年生软枣猕猴桃品种‘魁绿’、‘丰绿’和‘96-6’为试验材料,测定分析自然越冬期间枝条组织解剖结构、细胞膜透性和内源激素GA3、ABA、IAA含量水平的变化,以明确不同品种软枣猕猴桃越冬期间对低温的适应能力及其内在生理机制,为软枣猕猴桃抗寒品种的选育及鉴别提供理论依据。结果表明:(1)软枣猕猴桃枝条木质部比率大小依次为‘魁绿’>‘96-6’>‘丰绿’,皮层比率依次为‘丰绿’>96-6’>魁绿’,二者表现趋势相反,且‘魁绿’枝条的解剖结构与其他两品种差异显著。(2)在自然越冬期间,各品种软枣猕猴桃枝条相对电导率和MDA含量随温度变化均呈先升后降的趋势,在温度最低的1月份达到最高,其中‘魁绿’枝条的相对电导率和MDA含量变化幅度最小,而‘丰绿’受低温影响较大。(3)在11月份至2月份,随自然温度的变化,各品种软枣猕猴桃枝条的GA3和IAA含量均呈先降低后升高的变化趋势,ABA含量则呈先升后降的趋势,且都在1月份分别达到最低或最高值。研究发现,在自然越冬期间,低温对3个软枣猕猴桃品种的组织解剖结构、膜透性及内源激素含量均都造成较大的影响,品种‘魁绿’枝条的木质部比率最大、皮层比率最小,相对电导率和MDA含量变化幅度最小,内源激素含量GA3和ABA/GA3变化趋势较小,故其抗寒性比‘丰绿’和‘96-6’更强。  相似文献

以苗龄18~20个月的蝴蝶兰(Phalaenopsis)成熟苗为试材,通过夜温17℃/日温27℃处理诱导其成花,并研究蝴蝶兰成花过程中,叶片内源激素及多胺含量的变化动态。结果显示:低温诱导蝴蝶兰成花的临界期为第15d,芽重、内源激素和多胺含量均在第15d出现明显的消长规律;较低含量GA3、ABA,高含量ZR、Spd,高比值ZR/GA3、ZR/ABA和Spd/Put状态有利于成花;相对高含量的内源ZR是成花的关键因素;内源多胺与激素含量动态具有关联效应。  相似文献

为揭示薰衣草内源激素与抗寒能力的关系,以抗寒性相对较强的狭叶薰衣草和抗寒性较弱的宽叶薰衣草为试验材料,在田间自然条件下,于越冬前不同降温时期分别对叶片和根系取样,采用酶联免疫吸附法(ELISA)测定和分析了内源激素ABA、GA3、IAA和ZR的含量变化。结果表明,两种薰衣草的叶片和根系中的ABA、IAA和ZR的含量随气温的降低均表现为先升高后降低的趋势,但GA3表现为持续下降的趋势。抗寒性强的狭叶薰衣草叶片和根系中的ABA、IAA和ZR含量均高于抗寒性弱的宽叶薰衣草,而GA3小于宽叶薰衣草。对薰衣草越冬起重要作用的内源激素是ABA。  相似文献

There is potential to accelerate cultivar development with a doubled haploid system for breeding line production. Anther culture methodology was evaluated for U.S.A. spring barley (Hordeum vulgare L.) breeding applications. Gelrite was found to be an acceptable replacement for ficoll in the induction medium to reduce costs while maintaining embryoid and plant production levels. Beneficial effects of 28 d cold pretreatment of donor spikes for anther culture were confirmed with Pacific Northwest USA barley genotypes. A 3 d mannitol solution pretreatment of fresh anthers was shown to be less effective for green plant production compared to 28 d cold pretreatment of donor spikes. Extended donor spike cold pretreatment from 28 to 42 d did not reduce anther culture productivity. The effects of mannitol pretreatment on androgenesis of barley were systematically studied in comparison with that of cold pretreatment and control. The results showed that mannitol pretreatment could significantly increase the frequency of pollen survival reaching 19.0% on the eighth day, while in cold pretreatment and control they were 8.4% and 6.6 %, respectively. Mannitol pretreatment could also improve the quality of pollen and inhibit starch production from microspore, which were quite advantageous to microspore division and development. The developing period was shortened 2--3 days as compared with cold pretreatment and control. The major developmental pathways of androgenesis after mannitol pretreatment were the equal division (B pathway). In addition, the majority of microspore nuclei were diploids. On the contrary, the major microspores pretreated with low temperature had fewer chromosomes than with mannitol pretreatment, the microspore nuclei were haploids.  相似文献

研究了日本落叶松母株年龄、插穗内源激素含量与生根之间的关系,以及外源IBA对插穗内源激素含量的影响及其对插穗生根的促进作用。结果表明:不同株龄插穗生根性状及插穗茎和叶中激素含量差异均达极显著水平,叶中激素含量对插穗生根力没有直接影响;插穗茎中生根抑制激素(ABA)含量随株龄增长而增加,生根促进激素与抑制激素的比值(IAA+GA+ZR)/ABA却随株龄的增长而递减,与生根力随株龄的变化趋势一致,且该比值与生根性状紧密相关,因此可作为评价母株(无性系)生根力的指标;插后13~32d是插穗愈伤组织形成和不定根诱导的关键期,此期生根促进激素消耗量大,茎中含量大幅度降低,进入根伸长生长阶段,含量上升;外源IBA促进插穗生根的机制在于通过外源激素的刺激,在不定根诱导期,插穗茎中ABA含量大幅度降低,从而有利于不定根的发生和发育。  相似文献

The role of ABA in freezing tolerance and cold acclimation in barley   总被引:4,自引:0,他引:4  
The role of ABA in freezing resistance in nonacclimated and cold‐acclimated barley ( Hordeum vulgare L.) was studied. Eleven nonacclimated cultivars differed in their LT50, ranging from −10.8 to −4.8°C. Sugars, free proline, soluble proteins and ABA were analyzed in nonacclimated cultivars and during cold acclimation of one cultivar. There was an inverse correlation between LT50 and both ABA and sucrose contents. Exogenous ABA caused a decrease in the freezing point of leaf tissue in the cultivar with the lowest level of endogenous ABA, but not in the cultivar with the highest level, suggesting that ABA in the latter may be near the optimum endogenous level to induce freezing tolerance. Plants of cv. Aramir treated with ABA or allowed to acclimate to cold temperature increased their soluble sugar content to a similar level. The LT50 of leaves of cold‐acclimated cv. Aramir decreased from −5.8 to −11.4°C, with biphasic kinetics, accumulating proline and soluble sugars with similar kinetics. The biphasic profile observed during cold acclimation could be a direct consequence of cryoprotectant accumulation kinetics. ABA and soluble protein accumulation showed a single step profile, associated mainly with the second phase of the LT50 decrease. Thus, a significant increase in endogenous ABA is part of the response of barley to low temperature and may be required as a signal for the second phase of cold acclimation. Endogenous ABA contents in the nonacclimated state may determine constitutive freezing tolerance.  相似文献

为研究油茶(Camellia oleifera A)嫁接时穗条和砧木创伤后内源激素动态变化规律,解析影响砧穗嫁接面愈合的生理机制,为油茶砧穗愈合生长机理提供理论支持。以树龄6年的长林18号和53号的穗条和实生砧木为材料,按照芽苗砧嫁接方法切割穗条S0(0 min)、S10(10 min)、S40(40 min)和砧木茎段Z0(0 min)、Z10(10 min)后,利用液质联用法(HPLC-MS)测定吲哚乙酸(IAA)、脱落酸(ABA)、反式玉米素(TZR)、玉米素(Zeatin)、水杨酸(SA)和茉莉酸(JA)含量,分析不同时间段内源激素变化及品种间差异的关系。结果显示:创伤后18号的TZR、Zeatin和SA含量总体高于53号;18号IAA、SA和JA逐渐下降;TZR和Zeatin分别在S10和S0达最高值后下降;ABA在S10达最高值。53号IAA和JA爱S10达最高值后下降;TZR、Zeatin和SA在S10达低值后逐渐上升;ABA在S0达高值后逐渐下降。砧木茎段创伤前后激素含量除JA外18号高于53号;两品种Z0时激素含量下降,Z10后上升,仅53号ABA和SA含量在Z0达高值后下降。砧木茎段和根部激素含量在品种间除JA外18号高于53号,茎段的IAA、ABA高于根部,其他激素为根部高于茎段。激素比值在品种间和部位间差异明显;IAA/ABA、IAA/TZR、IAA/Zeatin和IAA/JA、ABA/TZR、ABA/Zeatin和ABA/JA比值为53号高于18号;穗条内SA/IAA为18号高于53号,SA/JA和SA/ABA为53号高于18号;砧木茎段均为18号高于53号;TZR/SA、TZR/JA比值在穗条和砧木茎段为18号高于53号。两品种创伤后IAA与JA极显著正相关,而IAA与SA,SA与JA在18号极显著正相关,53号极显著或显著负相关;53号TZR、Zeatin、SA间极显著或显著正相关,JA与TZR、Zeatin和SA极显著负相关。砧木茎段创伤后18号激素间为极显著或显著正相关;53号TZR和Zeatin与IAA、JA极显著正相关,与SA存在显著负相关,SA与JA有显著负相关。砧木茎段和根部间品种间仅在SA与各激素间相关性存在差异,其他激素间存在极显著正相关或负相关。综上所述,砧穗创伤后激素水平上18号在创伤面易于愈伤组织发育,而53号抗逆激素水平较高且与细胞分裂增殖类激素负相关,可能影响53号嫁接后愈合生长;嫁接应在创伤后10 min内较为适宜;砧穗间激素含量及比值的差异可能会影响后期嫁接部位形态重建以及穗条生长。  相似文献

低温预处理使籼稻花药在培养初期退化花粉百分率低于对照;多核花粉百分率明显高于对照。经过低温预处理的花药来见新的可溶性蛋白谱带出现;淀粉酶同工酶谱带少和弱于对照,过氧化物酶同工酶中碱性区活性初期弱而后期强于对照。低温处理使药壁组织衰退进程有所延缓,但未能制止衰退,也未出现如粳稻中所见的药壁细胞充实发育的现象。低温预处理只能稍微延缓籼稻药壁组织的衰退进程,有限度地促进籼稻花药培养效率。  相似文献

Sugars and polyols were tested at different steps of anther culture in barley (Hordeum vulgare L.) to elucidate their influence on both the overall yield of androgenesis and the structure of plastids in relation to albinism. During the pretreatment period, the osmotic regulation in the medium was beneficial to microspore embryogenesis regardless of the type and concentration of the tested osmoticum. The use of mannitol (300 mOsm/kg), sorbitol (180 mOsm/kg), PEG (240 mOsm/kg) and sucrose (180 mOsm/kg) gave the best results in terms of green plant production, although the influence of each substance differed according to the studied parameter. Similarly, during anther culture the regulation of the osmotic pressure in the medium had various effects, according to the osmoticum used. The best results were obtained using mannitol (364 mOsm/kg), providing 139.7 green plants per 100 plated anthers. Plastids were examined by electron microscopy following both pretreatment and culture. In the presence of mannitol and PEG, plastids did not accumulate starch at any stage of the protocol but they started to differentiate into chloroplasts in the microspore-derived embryos. Using sorbitol and sucrose, plastids differentiated poorly but accumulated large amounts of starch, suggesting that these sugars are metabolized by micropores and microspore derived structures. However, the accumulation of starch was not correlated with the occurrence of albinism. These results indicated that, in barley, the osmotic regulation was favourable to switch the microspore gametophytic program toward a sporophytic program regardless of the nature of the osmoticum. In addition, during the pretreatment period, mannito was found to be the most suitable osmoticum for subsequent embryo development.  相似文献

低温预处理延缓药壁中层和绒毡层的降解,促进表皮层和药室内壁层的发育,延缓花药过氧化物酶同工酶活性的增强。处理期间花药可溶性蛋白质、淀粉酶同工酶潜带发生明显变化。处理期间花药的~3H-TdR渗入和花粉的发育、分裂,表明花粉存在合成和充实活动。绒毡层和花粉间存在囊泡,表皮层和药室内壁层之间存在多泡体的穿壁运动,说明低温处理中药壁向花粉输送雄核发育所需的物质。在进入正常培养初期,经过低温处理的花药药壁仍有表皮层和药室内壁层的发育,多细胞花粉出现提早、数量增加,花粉退化延缓。而未经处理的花药药壁各层均迅速降解,花粉大量退化。  相似文献

冷锻炼处理提高了水稻(Oryza sativa L.)幼苗叶片中抗氧化剂(还原型谷胱甘肽,GSH;抗坏血酸,AsA)含量和膜保护酶(超氧化物歧化酶,SOD)的活性,同时也提高了可溶性蛋白质中热稳定蛋白的含量。CaCl2 浸种处理对上述冷锻炼的作用有加强的效果,且明显地提高了过氧化氢酶(CAT)和过氧化物酶(POD)的活性。有无CaCl2 处理的冷锻炼处理均减轻冷胁迫引起的GSH 及AsA 含量、SOD 活性及热稳定蛋白质含量的下降程度,有利于幼苗在恢复过程中GSH、AsA、CAT、SOD、POD及热稳定蛋白质水平迅速回升。结合CaCl2 处理的冷锻炼苗在冷胁迫恢复生长时增长迅速,且苗健壮浓绿,说明CaCl2浸种对冷锻炼处理提高水稻幼苗的抗冷力有明显的促进作用,这与CaCl2 浸种结合冷锻炼能更有效的提高细胞膜保护能力有关  相似文献

Drought stress at the reproductive stage causes pollen sterility and grain loss in wheat (Triticum aestivum). Drought stress induces abscisic acid (ABA) biosynthesis genes in anthers and ABA accumulation in spikes of drought-sensitive wheat varieties. In contrast, drought-tolerant wheat accumulates lower ABA levels, which correlates with lower ABA biosynthesis and higher ABA catabolic gene expression (ABA 8'-hydroxylase). Wheat TaABA8'OH1 deletion lines accumulate higher spike ABA levels and are more drought sensitive. ABA treatment of the spike mimics the effect of drought, causing high levels of sterility. ABA treatment represses the anther cell wall invertase gene TaIVR1, and drought-tolerant lines appeared to be more sensitive to the effect of ABA. Drought-induced sterility shows similarity to cold-induced sterility in rice (Oryza sativa). In cold-stressed rice, the rate of ABA accumulation was similar in cold-sensitive and cold-tolerant lines during the first 8 h of cold treatment, but in the tolerant line, ABA catabolism reduced ABA levels between 8 and 16 h of cold treatment. The ABA biosynthesis gene encoding 9-cis-epoxycarotenoid dioxygenase in anthers is mainly expressed in parenchyma cells surrounding the vascular bundle of the anther. Transgenic rice lines expressing the wheat TaABA8'OH1 gene under the control of the OsG6B tapetum-specific promoter resulted in reduced anther ABA levels under cold conditions. The transgenic lines showed that anther sink strength (OsINV4) was maintained under cold conditions and that this correlated with improved cold stress tolerance. Our data indicate that ABA and ABA 8'-hydroxylase play an important role in controlling anther ABA homeostasis and reproductive stage abiotic stress tolerance in cereals.  相似文献

沙棘(Hippophae rhamnoides)是荒漠化防治和水土保持的重要经济树种, 其果实中含有丰富的生物活性成分。内源激素生长素(IAA)、赤霉素(GA4)、脱落酸(ABA)和茉莉酸(JA)在沙棘果实发育过程中起着重要的调控作用, 但不同发育期内源激素的动态变化及其对果实生长发育的影响仍不清楚。以亲缘关系近且含油量差异明显的2个沙棘品系不同发育期果实为材料, 利用液相色谱串联质谱法检测了4种内源激素含量的动态变化。结果表明, 在果实发育过程中, 80%–85%的含水量有利于内源激素浓度的相对稳定; 果实含油量在不同发育期增速基本一致, 2个品系含油量在中后期开始出现明显差异。IAA和JA含量在果实成熟过程中呈先高后低的趋势; ABA含量在果实发育中后期始终处于较高水平; 而GA4含量一直处于较低水平。高ABA/GA4有利于果实油脂的合成与积累。研究结果为揭示不同内源激素对沙棘果实发育的调节作用提供了理论依据。  相似文献

The accumulation of abscisic acid (ABA) and the activities of antioxidative enzymes along with cell metabolic activity were monitored during androgenesis induction in triticale (×Triticosecale Wittm.). Tested cultivars ‘Mieszko’ and ‘Wanad’ were selected due to their significantly different responses to androgenic induction. Significant variation was observed in respect of superoxide dismutase activity and endogenous ABA content in anthers isolated from freshly cut tillers. For both cultivars, tillers pretreatment with low temperature decreased peroxidase activity by 36%, highly accelerated respiration rate and reduced heat production. At the same time, the level of ABA in ‘Mieszko’ was increased to the level measured in ‘Wanad’. This effect was associated with higher microspore culture viability and increased stress tolerance in ‘Mieszko’. Low temperature and metabolic starvation during 4-day anther preculture did not influence activities of antioxidative enzymes, while it resulted in slight decrease in respiration rate and heat emission. The importance of these changes for effective androgenesis induction is discussed.  相似文献

In stress conditions, microspores and young pollen grains can be switched from their normal pollen development toward an embryogenic pathway via a process called androgenesis. Androgenic embryos can produce completely homozygous, haploid or double-haploid plants. This study aimed to investigate changes in the abundance of protein species during cold pretreatment and subsequent cultivation of maize anthers on induction media using gel-based proteomics. Proteins upregulated on the third day of anther induction were identified and discussed here. Simultaneous microscopic observations revealed that the first division occurred in microspores within this period. Using 2-D electrophoresis combined with MALDI TOF/TOF MS/MS analysis 19 unique proteins were identified and classified into 8 functional groups. Proteins closely associated with metabolism, protein synthesis and cell structure were the most abundant ones. Importantly, ascorbate peroxidase, an enzyme decomposing hydrogen peroxide, was also upregulated. Isozyme analysis of peroxidases validated the proteomic data and showed increased peroxidase activities during androgenic induction. Further, the isozyme pattern of SOD revealed increased activity of the MnSOD, which could provide hydrogen peroxide as a substrate for in vivo peroxidase reactions (including ascorbate peroxidase). Together, these data reveal the role of enzymes controlling oxidative stress during induction of maize androgenesis.  相似文献
