In this work, we explored the possible polypharmacological potential of the already established antimicrobials against gastrointestinal pathogens, 4‐(alkylamino)‐3‐nitrocoumarins, as antianxiety agents, using a battery of in vivo experiments. Three chosen coumarin derivatives, differing in the substituent (sec‐butylamino, hexadecylamino, or benzylamino) at position 4, at the doses of 25, 50 and 100 mg kg–1, were evaluated in light/dark, open‐field, horizontal wire and diazepam‐induced sleep models using male BALB/c mice. Depending on the applied dose, all three tested coumarins displayed a noteworthy anxiolytic‐like effect. 4‐(sec‐Butylamino)‐3‐nitro‐2H‐chromen‐2‐one and 4‐(hexadecylamino)‐3‐nitro‐2H‐chromen‐2‐one could be recognized as true anxiolytics in the lowest applied dose, based on three tests, without exerting any sedative effects. Thus, the 3‐nitrocoumarin core deserves further chemical diversity exploration in the ‘antianxiety’ direction.  相似文献

Prostaglandin E2 (PGE2) is an endogenous lipid molecule involved in normal brain development. Cyclooxygenase‐2 (COX2) is the main regulator of PGE2 synthesis. Emerging clinical and molecular research provides compelling evidence that abnormal COX2/PGE2 signaling is associated with autism spectrum disorder (ASD). We previously found that COX2 knockout mice had dysregulated expression of many ASD genes belonging to important biological pathways for neurodevelopment. The present study is the first to show the connection between irregular COX2/PGE2 signaling and autism‐related behaviors in male and female COX2‐deficient knockin, (COX)‐2?, mice at young (4‐6 weeks) or adult (8‐11 weeks) ages. Autism‐related behaviors were prominent in male (COX)‐2? mice for most behavioral tests. In the open field test, (COX)‐2? mice traveled more than controls and adult male (COX)‐2? mice spent less time in the center indicating elevated hyperactive and anxiety‐linked behaviors. (COX)‐2? mice also buried more marbles, with males burying more than females, suggesting increased anxiety and repetitive behaviors. Young male (COX)‐2? mice fell more frequently in the inverted screen test revealing motor deficits. The three‐chamber sociability test found that adult female (COX)‐2? mice spent less time in the novel mouse chamber indicative of social abnormalities. In addition, male (COX)‐2? mice showed altered expression of several autism‐linked genes: Wnt2, Glo1, Grm5 and Mmp9. Overall, our findings offer new insight into the involvement of disrupted COX2/PGE2 signaling in ASD pathology with age‐related differences and greater impact on males. We propose that (COX)‐2? mice might serve as a novel model system to study specific types of autism.  相似文献

Phosphodiesterase‐1b (Pde1b) is highly expressed in striatum, dentate gyrus, CA3 and substantia nigra. In a new Floxed Pde1b × CreCMV global knockout (KO) mouse model, we show an immobility‐resistance phenotype that recapitulates that found in constitutive Pde1b KO mice. We use this new mouse model to show that the resistance to acute stress‐induced depression‐like phenotype is not the product of changes in locomotor activity or reactivity to other stressors (learned helplessness, novelty suppressed feeding or dexamethasone suppression), and is not associated with anhedonia using the sucrose preference test. Using tamoxifen inducible Cre, we show that the immobility‐resistant phenotype depends on the age of induction. The effect is present when Pde1b is Reduced from conception, P0 or P32, but not if reduced as adults (P60). We also mapped regional brain expression of PDE1B protein and of the Cre driver. These data add to the suggestion that PDE1B may be a target for drug development with therapeutic potential in depression alone or in combination with existing antidepressants.  相似文献

Increasing locations are often accompanied by an increase in variability. In this case apparent heteroscedasticity can indicate that there are treatment effects and it is appropriate to consider an alternative involving differences in location as well as in scale. As a location‐scale test the sum of a location and a scale test statistic can be used. However, the power can be raised through weighting the sum. In order to select values for this weighting an adaptive design with an interim analysis is proposed: The data of the first stage are used to calculate the weights and with the second stage's data a weighted location‐scale test is carried out. The p‐values of the two stages are combined through Fisher's combination test. With a Lepage‐type location‐scale test it is illustrated that the resultant adaptive test can be more powerful than the ‘optimum’ test with no interim analysis. The principle to calculate weights, which cannot be reasonably chosen a priori, with the data of the first stage may be useful for other tests which utilize weighted statistics, too. Furthermore, the proposed test is illustrated with an example from experimental ecology.  相似文献

Tests for a monotonic trend between an ordered categorical exposure and disease status are routinely carried out from case‐control data using the Mantel‐extension trend test or the asymptotically equivalent Cochran‐Armitage test. In this study, we considered two alternative tests based on isotonic regression, namely an order‐restricted likelihood ratio test and an isotonic modification of the Mantel‐extension test extending the recent proposal by Mancuso, Ahn and Chen (2001) to case‐control data. Furthermore, we considered three tests based on contrasts, namely a single contrast (SC) test based on Schaafsma's coefficients, the Dosemeci and Benichou (DB) test, a multiple contrast (MC) test based on the Helmert, reverse‐Helmert and linear contrasts and we derived their case‐control versions. Using simulations, we compared the statistical properties of these five alternative tests to those of the Mantel‐extension test under various patterns including no relationship, as well as monotonic and non‐monotonic relationships between exposure and disease status. In the case of no relationship, all tests had close to nominal type I error except in situations combining a very unbalanced exposure distribution and small sample size, where the asymptotic versions of the three tests based on contrasts were highly anticonservative. The use of bootstrap instead of asymptotic versions corrected this anticonservatism. For monotonic patterns, all tests had close powers. For non monotonic patterns, the DB‐test showed the most favourable results as it was the least powerful test. The two tests based on isotonic regression were the most powerful tests and the Mantel‐extension test, the SC‐ and MC‐tests had in‐between powers. The six tests were applied to data from a case‐control study investigating the relationship between alcohol consumption and risk of laryngeal cancer in Turkey. In situations with no evidence of a monotonic relationship between exposure and disease status, the three tests based on contrasts did not conclude in favour of a significant trend whereas all the other tests did. Abstract Acacia suaveolens (Sm.) Willd is a perennial shrub that forms even‐aged stands, recruited from a soil seed‐bank following fire. It has previously been subject to demographic studies, which used a space‐for‐time substitution to investigate temporal patterns following fire. In the present study the potential for spatial variation across sites was investigated by sampling at several similarly aged populations in Ku‐ring‐gai Chase National Park, northern Sydney, Australia. Significant variation in mean size and fecundity of A. suaveolens individuals was observed among sites, over a 2‐4.6‐fold range in plant size, and a sevenfold range in mean fecundity. The observed variation at 3 years after fire encapsulated most of the variation previously observed among sites 0‐17 years since fire, emphasizing the importance of spatial variation in this species. For each site a two‐stage (seed, plant) matrix model was constructed, and projected from 3 to 25 years following fire. Population growth was measured as number of seeds per 3‐year‐old plant, and found to vary 1.4‐fold across models for different sites. This site‐to‐site variation, as well as that in size, fecundity and survival, was statistically significant. Variation in projected seeds per plant could mostly be attributed to differences in fecundity rather than plant survival. Sensitivity analyses emphasized the biological significance of the variation in fecundity. Whereas previous studies have focused on temporal variation, this work demonstrates the importance of extending our understanding of a species to include the spatial component of population dynamics.  相似文献

To optimize resources, randomized clinical trials with multiple arms can be an attractive option to simultaneously test various treatment regimens in pharmaceutical drug development. The motivation for this work was the successful conduct and positive final outcome of a three‐arm randomized clinical trial primarily assessing whether obinutuzumab plus chlorambucil in patients with chronic lympocytic lymphoma and coexisting conditions is superior to chlorambucil alone based on a time‐to‐event endpoint. The inference strategy of this trial was based on a closed testing procedure. We compare this strategy to three potential alternatives to run a three‐arm clinical trial with a time‐to‐event endpoint. The primary goal is to quantify the differences between these strategies in terms of the time it takes until the first analysis and thus potential approval of a new drug, number of required events, and power. Operational aspects of implementing the various strategies are discussed. In conclusion, using a closed testing procedure results in the shortest time to the first analysis with a minimal loss in power. Therefore, closed testing procedures should be part of the statistician's standard clinical trials toolbox when planning multiarm clinical trials.  相似文献

Ecotoxicological tests (or bioassays) are controlled, reproducible tests where ecological responses are determined quantitatively. Due to numerous difficulties arising when airborne emissions are sampled, relatively few ecotoxicological assays have been applied. Aerosol particles are generally collected on a filter, which limits the quantity of the sample, thus also limiting the range of available test organisms. Bacterial bioassays require low sample quantity, and make a good choice when eco‐ or genotoxicity of the sample are to be determined. Of bacterial assays, the bioluminescence inhibition test has been proven applicable for assessing toxicity of airborne contaminants. Algorithm 1 in Guilbaud (2012, p. 327) in Biometrical Journal (DOI: 10.1002/bimj.201100123 ) reproduced a recently detected index error in a theorem concerning a shortcut for rejection decisions for certain multiple‐testing procedures as it was stated in Bernhard et al. (2004, p. 8) in Statistical Papers (DOI: 10.1007/BF02778266 ). This short article provides: (i) the correction to be made to Algorithm 1 and (ii) a brief discussion of the consequences. Although the theoretical developments in Guilbaud (2012) are not affected, the numerical illustrations in Section 7 are affected. Designs incorporating more than one endpoint have become popular in drug development. One of such designs allows for incorporation of short‐term information in an interim analysis if the long‐term primary endpoint has not been yet observed for some of the patients. At first we consider a two‐stage design with binary endpoints allowing for futility stopping only based on conditional power under both fixed and observed effects. Design characteristics of three estimators: using primary long‐term endpoint only, short‐term endpoint only, and combining data from both are compared. For each approach, equivalent cut‐off point values for fixed and observed effect conditional power calculations can be derived resulting in the same overall power. While in trials stopping for futility the type I error rate cannot get inflated (it usually decreases), there is loss of power. In this study, we consider different scenarios, including different thresholds for conditional power, different amount of information available at the interim, different correlations and probabilities of success. We further extend the methods to adaptive designs with unblinded sample size reassessments based on conditional power with inverse normal method as the combination function. Two different futility stopping rules are considered: one based on the conditional power, and one from P‐values based on Z‐statistics of the estimators. Average sample size, probability to stop for futility and overall power of the trial are compared and the influence of the choice of weights is investigated.  相似文献

Diagnostic or screening tests are widely used in medical fields to classify patients according to their disease status. Several statistical models for meta‐analysis of diagnostic test accuracy studies have been developed to synthesize test sensitivity and specificity of a diagnostic test of interest. Because of the correlation between test sensitivity and specificity, modeling the two measures using a bivariate model is recommended. In this paper, we extend the current standard bivariate linear mixed model (LMM) by proposing two variance‐stabilizing transformations: the arcsine square root and the Freeman–Tukey double arcsine transformation. We compared the performance of the proposed methods with the standard method through simulations using several performance measures. The simulation results showed that our proposed methods performed better than the standard LMM in terms of bias, root mean square error, and coverage probability in most of the scenarios, even when data were generated assuming the standard LMM. We also illustrated the methods using two real data sets.  相似文献

The Cochran–Armitage (CA) linear trend test for proportions is often used for genotype‐based analysis of candidate gene association. Depending on the underlying genetic mode of inheritance, the use of model‐specific scores maximises the power. Commonly, the underlying genetic model, i.e. additive, dominant or recessive mode of inheritance, is a priori unknown. Association studies are commonly analysed using permutation tests, where both inference and identification of the underlying mode of inheritance are important. Especially interesting are tests for case–control studies, defined by a maximum over a series of standardised CA tests, because such a procedure has power under all three genetic models. We reformulate the test problem and propose a conditional maximum test of scores‐specific linear‐by‐linear association tests. For maximum‐type, sum and quadratic test statistics the asymptotic expectation and covariance can be derived in a closed form and the limiting distribution is known. Both the limiting distribution and approximations of the exact conditional distribution can easily be computed using standard software packages. In addition to these technical advances, we extend the area of application to stratified designs, studies involving more than two groups and the simultaneous analysis of multiple loci by means of multiplicity‐adjusted p‐values for the underlying multiple CA trend tests. The new test is applied to reanalyse a study investigating genetic components of different subtypes of psoriasis. A new and flexible inference tool for association studies is available both theoretically as well as practically since already available software packages can be easily used to implement the suggested test procedures.  相似文献

New Delhi metallo‐β‐lactamase‐1 (NDM‐1), one of the metallo‐β‐lactamases (MBLs), has been identified from clinical isolates worldwide. Rapid detection of NDM‐1 producers is necessary to prevent their dissemination. Seven types of EDTA complexes were evaluated as MBL inhibitors in double‐disk synergy tests (DDSTs), resulting in detection of the first isolate of NDM‐1‐producing Escherichia coli (NDM‐1 Dok01) in Japan. NDM‐1 Dok01 was detected when EDTA magnesium disodium salt tetrahydrate (Mg‐EDTA), EDTA calcium disodium salt dihydrate, EDTA cobalt disodium salt tetrahydrate and EDTA copper disodium salt tetrahydrate were used as MBL inhibitors. The sensitivity and specificity of DDSTs using Mg‐EDTA for 75 MBL producers and 25 non‐MBL producers were 96.0% and 100%, respectively. These findings indicate that the DDST method using Mg‐EDTA can detect MBL‐producing strains, including NDM‐1 producers.  相似文献

Summary Clinical trials data often come in the form of low‐dimensional tables of small counts. Standard approximate tests such as score and likelihood ratio tests are imperfect in several respects. First, they can give quite different answers from the same data. Second, the actual type‐1 error can differ significantly from nominal, even for quite large sample sizes. Third, exact inferences based on these can be strongly nonmonotonic functions of the null parameter and lead to confidence sets that are discontiguous. There are two modern approaches to small sample inference. One is to use so‐called higher order asymptotics ( Reid, 2003 , Annal of Statistics 31 , 1695–1731) to provide an explicit adjustment to the likelihood ratio statistic. The theory for this is complex but the statistic is quick to compute. The second approach is to perform an exact calculation of significance assuming the nuisance parameters equal their null estimate ( Lee and Young, 2005 , Statistic and Probability Letters 71 , 143–153), which is a kind of parametric bootstrap. The purpose of this article is to explain and evaluate these two methods, for testing whether a difference in probabilities p2? p1 exceeds a prechosen noninferiority margin δ0 . On the basis of an extensive numerical study, we recommend bootstrap P‐values as superior to all other alternatives. First, they produce practically identical answers regardless of the basic test statistic chosen. Second, they have excellent size accuracy and higher power. A simple, sensitive and rapid HPLC method with fluorescence detection for the determination of dimethyl‐4,4′‐dimethoxy‐5,6,5′,6′‐dimethylene dioxybiphenyl‐2,2′‐dicarboxylate (DDB) in the raw material and pill form was developed. Liquid chromatography was performed on a C18 column (250 × 4.6 mm i.d., 5 µm particle size), the mobile phase consisted of methanol and 0.05 M sodium dihydrogen phosphate buffer (80 : 20, v/v), and the apparent pH of the mobile phase was adjusted to 3. The fluorescence detector was operated at excitation/emission wavelengths of 275/400 nm. The proposed method allows the determination of DDB within concentration range 0.1–1.5 µg/mL with a limit of detection of 0.032 µg/mL, a limit of quantification of 0.097 µg/mL and a correlation coefficient of 0.9997. The proposed method has been successfully applied for the analysis of DDB in its pills with a percentage recovery of 98.45 ± 0.32. The method was fully validated according to ICH guidelines. Moreover, the high sensitivity of the method permits its use in an in vitro dissolution test for DDB under simulated intestinal conditions.
