为确定氯化三苯基四氮唑(TTC)-脱氢酶还原法测定螺旋藻细胞活性的最优条件,首先通过单因素实验对影响藻细胞活性测定的TTC质量分数、缓冲液pH、提取剂(乙醇)质量分数、培养时间、培养温度进行分析,选定各因素的变化范围,再利用正交试验设计方法,在藻细胞活性测定的最适培养温度下,对TTC质量分数、缓冲液pH、提取剂质量分数、培养时间进行3水平优化试验。最后确定优化的TTC 脱氢酶还原法测定螺旋藻细胞活性的条件为TTC质量分数0.1%、缓冲液pH 8.0~8.5、乙醇质量分数60%、培养时间5 h、培养温度35 ℃。在此条件下所测藻细胞活性最高,为螺旋藻细胞活性的评价提供了一定参考。 相似文献

以鄂尔多斯高原碱湖特有的钝顶螺旋藻S1、引进的钝顶螺旋藻S2和极大螺旋藻S3为材料,采用Folin法和茚三酮法分别测定低温下细胞外渗液中可溶性蛋白和游离氮基酸含量。结果表明:处理温度越低,时间越长,螺旋藻细胞外渗液中可溶性蛋白和游离氨基酸的含量越高。S1膜损伤程度最轻,比引进种S2、S3有较强的抗寒能力。判断低温对螺旋藻膜损伤的程度,用可溶性蛋白较游离氨基酸更接近于实际。 相似文献

采用聚丙烯酰胺凝胶电泳法对毛乌素沙地碱湖的钝顶螺旋藻S3和鄂尔多斯螺旋藻S4与国外引进的钝顶螺旋藻S1和极大螺旋藻S2 的POD、CAT和SOD同工酶进行了比较研究。结果表明 :4个样品的 3种酶同工酶带数目不同 ,依次是S4 >S3>S1>S2 ;S3和S4 酶带数多 ,对环境适应性强 ,进化程度较高。螺旋藻不同种间的酶谱相似系数为 0 5 3~ 0 80 ,有较近的亲缘关系 ;原产地不同的钝顶螺旋藻S1和S3间酶谱相似系数最高为 0 86,表明螺旋藻种内差异 <种间差异。 相似文献

高温胁迫对烟草叶绿体NADPH脱氢酶复合体活性的促进 总被引:5,自引:0,他引:5
为探讨叶绿体NAD(P)H脱氢酶复合体(NDH)在植物抵御热胁迫中的生理意义,比较了烟草ndhJK基因缺失突变体(ΔndhJK)和野生型对50℃高温胁迫的响应.高温下,野生型中一条NBT-NADPH氧化还原酶活性带有所增加,免疫印迹分析确定了此活性染色带是NDH亚复合体,该活性带中的NDH-K表达量也在热胁迫条件下明显地增加.与ΔndhJK相比,在高温胁迫下,野生型中远红光诱导的P700 氧化速率明显地变慢,而远红光关闭后的P700暗还原速率则显著地变快,表明高温促进NDH介导的围绕光系统I的循环电子传递.根据这些结果推测,在热胁迫条件下野生型中对NADPH底物专一的NDH活性的增加可能有利于减少NADPH的积累,减轻叶绿体间质的过度还原. 相似文献

在钝顶螺旋藻的对数生长期进行硒胁迫处理,研究硒胁迫强度和硒胁迫时间对钝顶螺旋藻抗氧化酶系统与脂质过氧化作用的影响。设置4个胁迫实验组,分别从第5天到第10天分次加硒,累计加硒量均为1000 mg/L。结果表明,硒胁迫实验组中钝顶螺旋藻的抗氧化酶活性均高于对照组或与对照组相当,其中GPX的变化最大,实验组Ⅰ中的GPX活性为对照组的20.5倍;SOD的变化趋势与GPX相近,但对较低的硒胁迫的响应不敏感;POD、CAT和APX的活性均随硒胁迫强度的减小而上升;各硒胁迫实验组中只有实验组Ⅰ的MDA含量高于对照组;藻体对.OH的清除能力与藻体的MDA含量呈负相关,提示了硒对钝顶螺旋藻抗氧化系统的双重效应。 相似文献

在5℃低温胁迫条件下,对以野生黄瓜——棘瓜(Sicyos angulatus L.)和以黑籽南瓜为砧木的黄瓜嫁接苗以及自根苗的抗冷性生理生化指标的变化规律作了研究。结果表明,以棘瓜为砧木的黄瓜嫁接苗叶片的SOD、POD、CAT活性以及Pro、可溶性糖含量均高于后两者,MDA含量低于后两者。说明利用棘瓜嫁接的黄瓜苗可以忍受更长时间或更大强度的低温。 相似文献

在01%~5.0%NaCl浓度范围的培养基中培养极大螺旋藻(Spirulinamaxima),发现NaCl浓度高于2.0%时螺旋藻生长受到明显抑制。培养7天后测定超氧化物歧化酶(SOD)、抗坏血酸过氧化物酶(ASAPOD)、过氧化氢酶(CAT)活性和丙二醛(MDA)含量。结果表明:在盐胁迫下,SOD酶活性升高;抗坏血酸过氧化物酶和过氧化氢酶活性在低盐胁迫下活性升高,高盐胁迫下抗坏血酸过氧化物酶活性迅速降低,过氧化物酶则完全失活;MDA含量先随盐胁迫程度增加而降低,后随盐胁迫的进一步增强恢复至对照水平。 相似文献

采用比色法对鄂尔多斯高原碱湖的钝顶螺旋藻(S1)与国外引进的钝顶螺旋藻(S2)和极大螺旋藻(S3)的乙醇酸氧化酶(GO)进行了比较研究。结果表明:在25℃、pH 8.0条件下,S1、S2和S3的GO活性分别为70.9 U/gFW、59.6 U/gFW和80.9 U/gFW;最适温度均为30℃;在0℃~35℃(30℃)范围内比较稳定;最适pH值分别为8.6、8.2和8.4;pH值稳定范围,S1为7.6~10.0、S2为8.0~9.0;S3为8.0~8.6。S1的GO对温度和pH适应范围最宽,且在低温、高温、强酸和强碱下的活性均比引进种的高。 相似文献

将钝顶螺旋藻培养在含有不同NaCl浓度(0、0.4、0.8mol·L^-1)的培养基中,并置于室外全波段太阳辐射、阳光辐射滤除uvB以及光合有效辐射(PAR)三种辐射条件下,以探讨阳光uV辐射和盐胁迫对钝顶螺旋藻的耦合效应。结果表明,阳光uv辐射显著抑制钝顶螺旋藻的光化学效率,且随着盐浓度的提高,其受抑制程度加剧。D1蛋白含量在高水平PAR和uV辐射下都明显降低,而高盐浓度(0.8mol·L^-1NaCl)导致其含量进一步下降。此外,阳光uv辐射与盐胁迫的耦合作用使得藻丝发生明显断裂。 相似文献

The cyanobacterium Arthrospira (Spirulina) platensis was used to study the process of silver biosorption. Effects of various parameters such as contact time, dosage of biosorbent, initial pH, temperature, and initial concentration of Ag(I) were investigated for a batch adsorption system. The optimal biosorption conditions were determined as pH 5.0, biosorbent dosage of 0.4 g, and initial silver concentration of 30 mg/L. Equilibrium adsorption data were analyzed by the Langmuir and Freundlich models – however, the Freundlich model provided a better fit to the experimental data. The kinetic data fit the pseudo-second-order model well, with a correlation coefficient of 0.99. The analysis of thermodynamic parameters (ΔG°, ΔH° and ΔS°) revealed that the adsorption process of silver ion by spirulina biomass was exothermic and spontaneous (ΔG° < 0), and exothermic (ΔH° < 0) process. The biosorption capacity of biomass A. platensis serves as a basis for the development of green technology for environmental remediation. 相似文献

The physiological behaviour of Arthrospira (Spirulina) maxima during acclimation to sudden changes in irradiance from high
(HL) to low light (LL) and vice versa was studied by following parameters concerning growth rate, pigment, carbohydrate and
protein cell contents. Applying first order kinetics, the specific acclimation rates for the parameters considered were calculated.
During HL to LL shift, pigments increased to compensate for a reduction in growth irradiance in order to maintain relatively
high growth rates, whereas carbohydrates decreased at the highest rate. The synthesis of phycobiliproteins proceeded at a
rate similar or little higher than that of chlorophyll a, indicating their importance in the light harvesting at low irradiance.
During LL to HL shift, carbohydrate biosynthesis was increased, whereas pigment and protein cell contents decreased. The kinetic
analysis suggested that the pigment decrease could be accounted for both by dilution through growth and in vivo degradation.
During this transition, the initially high cell pigment content gave rise to a very heavy carbohydrate synthesis, which for
a short time, after the shift to HL conditions, overshot the final steady-state. In the same period the specific growth rate
also increased notably, overshooting the μmax. The acclimation rates of the measured parameters were faster during LL to HL
transition then during the reverse. The physiological response of A. maxima during the acclimation to sudden irradiance shifts
points out the ability of this cyanobacterium to alter light harvesting and highlights again the key role of carbohydrates
when the cells underwent an energy crisis during down-shift.

【目的】2015年7月至2015年9月,对海岸线较长、湿地类型比较多样化的渤海湿地及周边水域中螺旋藻(节旋藻)的分布进行研究,并对其与环境因子的相互关系进行分析。【方法】采用相关分析和灰关联分析对螺旋藻(节旋藻)数量与环境因子关系进行分析。【结果】螺旋藻(节旋藻)数量与pH、盐度、总氮(TN)、总磷(TP)、化学需氧量(COD)、氨氮(NH_3-N)呈显著正相关(P0.05,P0.01),与溶解氧(DO)呈显著负相关(P0.05,P0.01),与温度呈负相关,但相关不显著(P0.05);灰关联分析结果表明:各环境因素对螺旋藻数量的关联程度依次为:CODpHTN温度TPDO盐度NH_3-N。【结论】渤海海滨湿地及周边水域中螺旋藻(节旋藻)资源丰富,对螺旋藻(节旋藻)分布影响最大的环境因子是COD,最小是NH_3-N。 相似文献

Arthrospira platensis was cultivated in tubular photobioreactor in order to evaluate growth and biomass production at variable photosynthetic photon flux density (PPFD = 60, 120, and 240 μmol photons m(-2)s(-1)) and employing three different systems for cell circulation, specifically an airlift, a motor-driven pumping and a pressurized system. The influence of these two independents variables on the maximum cell concentration (X(m)), cell productivity (P(x)), nitrogen-to-cell conversion factor (Y(X/N) ), photosynthetic efficiency (PE), and biomass composition (total lipids and proteins), taken as responses, was evaluated by analysis of variance. The statistical analysis revealed that the best combination of responses' mean values (X(m) = 4,055 mg L(-1), P(x) = 406 mg L(-1)day(-1), Y(X/N) = 5.07 mg mg(-1), total lipids = 8.94%, total proteins = 30.3%, PE = 2.04%) was obtained at PPFD = 120 μmol photons m(-2)s(-1); therefore, this light intensity should be considered as the most well-suited for A. platensis cultivation in this photobioreactor configuration. The airlift system did not exert any significant positive statistical influence on the responses, which suggests that this traditional cell circulation system could successfully be substituted by the others tested in this work. 相似文献

This study dealt with the influence of both the feeding time and light intensity on the fed-batch culture of the cyanobacterium Spirulina (Arthrospira) platensis using ammonium chloride as a nitrogen source. For this purpose, a 2(2) plus star central composite experimental design combined with response surface methodology was employed, and the maximum cell concentration (X(m)), the cell productivity (P(X)), and the yield of biomass on nitrogen (Y(X/N)) were selected as the response variables. The optimum values of X(m) (1,833 mg L(-1)) and Y(X/N) (5.9 g g(-1)) estimated by the model at light intensity of 13 klux and feeding time of 17.2 days were very close to those obtained experimentally under these conditions (X(m) = 1,771 +/- 41 mg L(-1); Y(X/N) = 5.7 +/- 0.17 g g(-1)). The cell productivity was a decreasing function of the ammonium chloride feeding time and a quadratic function of the light intensity. The protein and lipid contents of dry biomass collected at the end of cultivations were shown to decrease with increasing light intensity. 相似文献
