The contents of K, Cl and Na in the mucus containing compartments of rat small intestine epithelium were examined with X-ray microanalysis. Measurements of concentration were made on sections prepared according to techniques based on cryofixation of soft biological tissues. The concentration of potassium and chlorine within the brush border region and in the secretion product of goblet cells is 200 mM and 90 mM, respectively, while sodium concentration was lower than the detection limit of 20 mM of the method used.  相似文献

The mechanical fragility of the L form of two group A streptococcal strains was assessed by subjecting L-form suspensions to sonic oscillation (60 W, 20 kc/sec). To evaluate the effect of the sonic energy applied, the original streptococcal strains and aSerratia marcescens strain were subjected to the same treatment. The killing of the organisms by sonic oscillation followed the expression log No/Nt=–K·t1/2. The mortality rate constants K of the streptococcus,S. marcescens and the L form of the AED and GL-8 streptococcal strains were –0.19,–0.98,–1.14 and–1.52 min–1/2. respectively. The mortality rates of the L form of the two streptococcal strains differed significantly (P=0.01). From a comparison of these data, and taking into account the difference in cell envelope between the bacterial and the L form, it is concluded that the limiting envelopes of the group A streptococcal L-form elements probably possess a relatively marked stability. The factors which might be responsible for the difference in mortality rates between the L form of the streptococcal strains are discussed.The author wishes to thank Dr. W. WH3(OCH2C6H5) (PMe3)4 (1) is formed upon reaction of WH2(PMe3)5 with benzyl alcohol for 12 days at ambient temperatures. Thermolysis of 1 at 80°C in toluene solution gives the carbonyl complex, WH2(CO)(PMe3)4 (2) and benzene. The conversion is slower in the presence of H2. Reaction of 1 with D2 leads to H/D exchange in the hydride ligands and in the benzylic and ortho-phenyl positions of the benzyloxide. A mechanism for the thermolysis of 1, based on an H2 elimination, sequential C-H activations, and CO deinsertion from an acyl ligand, is proposed. Thermolysis of 1 is much faster in the presence of free benzyl alcohol and 2 is not formed. The products under these conditions are toluene, bibenzyl, WH4(PMe3)4, PMe3 and unidentified material, consistent with the intermediacy of benzyl radicals.  相似文献

The bacterial flagellar C-ring is composed of two essential proteins, FliM and FliN. The smaller protein, FliN, is similar to the C-terminus of the larger protein, FliM, both being composed of SpoA domains. While bacterial type III secretion (T3S) systems encode many proteins in common with the flagellum, they mostly have a single protein in place of FliM and FliN. This protein resembles FliM at its N-terminus and is as large as FliM but is more like FliN at its C-terminal SpoA domain. We have discovered that a FliN-sized cognate indeed exists in the Yersinia T3S system to accompany the FliM-sized cognate. The FliN-sized cognate, YscQ-C, is the product of an internal translation initiation site within the locus encoding the FliM-sized cognate YscQ. Both intact YscQ and YscQ-C were found to be required for T3S, indicating that the internal translation initiation site, which is conserved in some but not all YscQ orthologs, is crucial for function. The crystal structure of YscQ-C revealed a SpoA domain that forms a highly intertwined, domain-swapped homodimer, similar to those observed in FliN and the YscQ ortholog HrcQ(B). A single YscQ-C homodimer associated reversibly with a single molecule of intact YscQ, indicating conformational differences between the SpoA domains of intact YscQ and YscQ-C. A "snap-back" mechanism suggested by the structure can account for this. The 1:2 YscQ-YscQ-C complex is a close mimic of the 1:4 FliM-FliN complex and the likely building block of the putative Yersinia T3S system C-ring.  相似文献

Rat liver prolyl-tRNA synthetase was purified as a dimer of M(r) 60,000 subunits not associated with other aminoacyl-tRNA synthetases and as a form associated with glutamyl-tRNA synthetase. Proteolysis of the dimeric enzyme generated a less active form with M(r) 52,000 subunits and an inactive form with M(r) 40,000 subunits. A second species was isolated with polypeptides of M(r) 60,000 and 150,000. This form dissociated during gel filtration chromatography being partially resolved into the M(r) 150,000 and 60,000 components; glutamyl-tRNA synthetase was associated with the larger polypeptide and prolyl-tRNA synthetase with the smaller component. Antibodies against the M(r) 60,000 polypeptide reacted with the M(r) 60,000 and 150,000 polypeptides. Gel filtration of extracts revealed multiple forms of prolyl- and glutamyl-tRNA synthetase. Antibody against the M(r) 60,000 component detected the M(r) 60,000 and 150,000 polypeptides throughout the chromatogram; these forms could be partially separated by polyethylene glycol fractionation. The M(r) 150,000 and 60,000 polypeptides were detected by Western blot analysis of crude extracts prepared under several conditions. Antibody to prolyl-tRNA synthetase reacted with a M(r) 150,000 polypeptide of the aminoacyl-tRNA synthetase core complex identified previously as glutamyl-tRNA synthetase.  相似文献

Experimental pathogenicity of the Y form and the M form ofC. albicans separated with a high degree of purity has been evaluated. Experiments are described in which the two morphological forms ofC. albicans were separately inoculated intradermally, intraperitoneally and intravenously in mice and rabbits. In suitable conditions, higher pathogenicity was significantly provoked by the Y form ofC. albicans.

250 Candida guilliermondii strains resistant to the polyene antibiotics nystatin, levorin and amphotericin B were obtained using UV irradiation. When the mutant strains became resistant to one of the polyene antibiotics, their resistance to the other ones changed. Phenotypic analysis showed that the resistance of the strains to polyene antibiotics did not make them susceptible to a rise in osmotic pressure and to a change of the temperature of incubation. Some of the polyene-resistant strains were stained in a medium with methylene blue. Analysis of the sterol composition in the mutants by UV spectroscopy showed that the resistance to polyene antibiotics sometimes involved changes in the sterol composition. Two new UV spectrum types were recorded for the sterols of the mutant strains; they differed from the UV spectrum for the sterols of the parent sensitive strain.  相似文献
