Given that the elevated serum semicarbazide-sensitive amine oxidase (SSAO) activity is associated with the severity of carotid atherosclerosis in clinic, the current study aims to investigate whether SSAO inactivation by semicarbazide is beneficial for established atherosclerotic lesions in LDLr knockout mice on a high-fat/high- cholesterol Western-type diet or after dietary lipid lowering. Despite no impact on plasma total cholesterol levels, the infiltration of circulating monocytes into peripheral tissues, and the size of atherosclerotic lesions, abrogation of SSAO activity resulted in the stabilization of established lesions as evidenced by the increased collagen contents under both conditions. Moreover, SSAO inactivation decreased Ly6Chigh monocytosis and lesion macrophage contents in hypercholesterolemic mice, while no effect was observed in mice after normalization of hypercholesterolemia by dietary lipid lowering. Strikingly, abrogation of SSAO activity significantly increased not only the absolute numbers of smooth muscle cells (SMCs), but also the percent of SMCs with a synthetic phenotype in established lesions of mice regardless of plasma cholesterol levels. Overall, our data indicate that SSAO inactivation in vivo stabilizes the established plaques mainly via inducing the switch of SMCs from a contractile to a synthetic phenotype. Targeting SSAO activity thus may represent a potential treatment for patients with atherosclerosis.  相似文献

Most types of cells in the body do not express the capability of catabolizing cholesterol, so cholesterol efflux is essential for homeostasis. For instance, macrophages possess four pathways for exporting free (unesterified) cholesterol to extracellular high density lipoprotein (HDL). The passive processes include simple diffusion via the aqueous phase and facilitated diffusion mediated by scavenger receptor class B, type 1 (SR-BI). Active pathways are mediated by the ATP-binding cassette (ABC) transporters ABCA1 and ABCG1, which are membrane lipid translocases. The efflux of cellular phospholipid and free cholesterol to apolipoprotein A-I promoted by ABCA1 is essential for HDL biogenesis. Current understanding of the molecular mechanisms involved in these four efflux pathways is presented in this minireview.  相似文献

Cigarette smoke contributes to or exacerbates airway diseases such as asthma and COPD, where airway hyperresponsiveness and airway smooth muscle (ASM) proliferation are key features. While factors such as inflammation contribute to asthma in part by enhancing agonist-induced intracellular Ca2+ ([Ca2+]i) responses of ASM, the mechanisms by which cigarette smoke affect ASM are still under investigation. In the present study, we tested the hypothesis that cigarette smoke enhances the expression and function of Ca2+ regulatory proteins leading to increased store operated Ca2+ entry (SOCE) and cell proliferation. Using isolated human ASM (hASM) cells, incubated in the presence and absence cigarette smoke extract (CSE) we determined ([Ca2+]i) responses and expression of relevant proteins as well as ASM proliferation, reactive oxidant species (ROS) and cytokine generation. CSE enhanced [Ca2+]i responses to agonist and SOCE: effects mediated by increased expression of TRPC3, CD38, STIM1, and/or Orai1, evident by attenuation of CSE effects when siRNAs against these proteins were used, particularly Orai1. CSE also increased hASM ROS generation and cytokine secretion. In addition, we found in the airways of patients with long-term smoking history, TRPC3 and CD38 expression were significantly increased compared to life-long never-smokers, supporting the role of these proteins in smoking effects. Finally, CSE enhanced hASM proliferation, an effect confirmed by upregulation of PCNA and Cyclin E. These results support a critical role for Ca2+ regulatory proteins and enhanced SOCE to alter airway structure and function in smoking-related airway disease.  相似文献

Neurodegenerative diseases, such as Alzheimer’s and Parkinson’s, are characterized by several pathological features, including selective neuronal loss, aggregation of specific proteins, and chronic inflammation. Aging is the most critical risk factor of these disorders. However, the mechanism by which aging contributes to the pathogenesis of neurodegenerative diseases is not clearly understood. Cellular senescence is a cell state or fate in response to stimuli. It is typically associated with a series of changes in cellular phenotypes such as abnormal cellular metabolism and proteostasis, reactive oxygen species (ROS) production, and increased secretion of certain molecules via senescence-associated secretory phenotype (SASP). In this review, we discuss how cellular senescence contributes to brain aging and neurodegenerative diseases, and the relationship between protein aggregation and cellular senescence. Finally, we discuss the potential of senescence modifiers and senolytics in the treatment of neurodegenerative diseases.  相似文献

The specificity of synaptic connections is directly related to the functional integrity of neural circuits. Long-range axon guidance and topographic mapping mechanisms bring axons into spatial proximity of target cells and thus limit the number of potential synaptic partners. Synaptic specificity is then achieved by extracellular short-range guidance cues and cell-surface recognition cues. Neural activity may enhance the precision and strength of specific circuit connections. Here, we focus on one of the final steps of synaptic matchmaking: the targeting of synaptic layers and the mutual recognition of axons and dendrites within these layers.Perception and behavior are critically dependent on synaptic communication between specific neurons. Understanding how neurons achieve such “synaptic specificity” is therefore one of the most fundamental issues in developmental neuroscience. Langley’s notion of “chemical relations” between synaptically connected neurons (Langley 1892) and Sperry’s “chemoaffinity” hypothesis (Sperry 1963) provided a conceptual framework for the development of precise synaptic connections in the central nervous system. Sperry postulated that molecular interactions between neurons and their extracellular environment (including between and amongst axons and dendrites) ensure that connections form only between “appropriate” synaptic partners (Sperry 1963). This hypothesis has been confirmed by experimental work over the last four decades, most importantly by the identification of molecular cues that provide synaptic specificity (see Sanes and Yamagata 2009 for a recent comprehensive review). However, within this broad framework, a number of alternate mechanisms have been shown or proposed to play roles in specific aspects of such targeting processes. Here, we focus on mechanisms that underlie the formation of synaptic layers, a prominent anatomical feature of the visual system as well as many other areas of the CNS.As reviewed previously (O''Leary 2010), the chemoaffinity principle underlies the developmental process of topographic mapping. Indeed, the precision with which neurons preserve the spatial relationships between the visual world and its representation in the brain is remarkable: Across animals ranging from flies to vertebrates, axons that bear signals from adjacent points in visual space invariably choose adjacent targets in the brain (Braitenberg 1967; Lemke and Reber 2005; Sperry 1963). Thus, position-dependent guidance of axons ensures that a visuotopic map develops. However, position in space is just one attribute of a visual stimulus; others include color, brightness, edge detection, and movement. If position in visual space is encoded by localized activation within a two-dimensional field of neurons, then these other features are encoded by local circuits that act both in series and in parallel and are reiterated many times across the field (Fig. 1). These local circuit modules are often envisioned as “columns” that lie orthogonal to the topographic map, with each column corresponding to a pixel in visual space and each level of the column representing a different, specific visual feature within that pixel, such as brightness, color, etc. (Fig. 1). How these columns acquire their laminated structure represents a developmental challenge of extraordinary scale. Although long-range axon guidance and topographic mapping no doubt contribute to restricting the astronomical number of potential synaptic partners, these mechanisms are clearly not sufficient; additional mechanisms must (and do) exist that act on a local scale to provide an additional level of positional information and cell-type-specific “chemoaffinity.”Open in a separate windowFigure 1.Laminae are a fundamental organizing unit of neural circuits. Each column corresponds to a single topographic position (e.g., location on the retina). Within each column, different cell types (shown type A: blue, and type B: red) respond to different features in the visual world, such as motion or luminance. These pixels are repeated many times over and thus cover all of visual space. A simple rule of “Cell type A connects to Layer A, etc.” ensures that functional segregation is maintained in the connections from the retina to the target (parallel processing). Each pixel P1, P2, and P3 connects to a single column (C1, C2, and C3), establishing serial processing. Within each column, there are local circuits that, too, are layer-specific. Thus, laminae ensure functional specificity of both afferent-target connections and local circuit connections.A prominent principle, which guides the formation of connections between specific cell types and is a characteristic feature of CNS architecture, is the concentration of synapses in small areas. These synapse clusters can take the form of planar layers or spherical glomeruli. Although glomeruli are a specialization that appears most prominent in the olfactory system, layers, or laminae, are an almost ubiquitous feature of central nervous system architecture. Indeed, even crude histological stains reveal that axons and dendrites often accumulate in neuropil (cell-body-free areas). Cell-type-specific or single-cell labeling has shown that, within individual neuropil layers, neurites and synapses are not distributed randomly. Rather, synaptic connections arising between neurons with the same or similar functional properties are localized to particular sublaminae that distinguish synapses with different properties (Fig. 1). The structural underpinnings of this functional principle are provided by mechanisms that ensure the lamina-specific branching of the corresponding neurites. How this enormous precision is achieved is the subject of intense investigations in the Drosophila, zebrafish, chick, and mouse visual systems. We will begin by describing three anatomical regions in these model organisms. Then, we will discuss three broad principles of layer-specific targeting in the visual system, namely cell–cell recognition, guidance by matrix cues, and activity-dependent sorting of axon terminals.  相似文献

Filamin (FLN) plays an important role in differentiation, migration, and signal transduction in vascular smooth muscle cells (VSMCs). We hypothesized that suppression of FLN expression would inhibit differentiation and migration of VSMCs, and interrupt the phenotype switch of these cells. We designed and tested different shRNA sequences to silence FLN expression. The degree of silencing was assessed at mRNA (qPCR) and protein (Western blot) levels. Two sequences, FL1 and FL2, lead to the most efficient FLN silencing. Subsequent experiments were conducted using the FL1 shRNA. Cells with silenced FLN were exposed to ox-LDL, and cell phenotypic changes (cell proliferation, cell cycle, apoptosis, and morphologic changes of cytoskeleton) were evaluated. When FLN was silenced, the phenotype switch of VSMCs exposed to ox-LDL was attenuated. Further, the injury to the cytoskeleton was less prominent in the FLN-silenced cells. To conclude, RNA silencing of FLN decreases the phenotype switch of VSMCs into a pathologic state. FLN silencing could be useful in treating atherosclerosis at genetic level.  相似文献

目的:研究去血清诱导人血管平滑肌细胞发生表型转化与microRNAs表达间的关系。方法:采取人血管平滑肌细胞克隆株HITASY,培养人血管平滑肌细胞(VSMCs)。用去血清的方法处理人血管平滑肌细胞,使VSMCs由去分化表型向分化表型转化,通过蛋白印迹法观察平滑肌细胞中分化标记物蛋白的变化,同时用实时定量PCR法检测细胞中相关microRNA的表达。结果:①去血清处理组与无去血清处理组比较,平滑肌细胞分化标记物(SM-α-actin、calponin)表达显著性增加,而通过SM-α-actin、calponin的蛋白表达量显著增加,提示血管平滑肌细胞向分化表型转化(P0.05);②同时去血清处理组与无去血清处理组比较,Mir-649、Mir-944表达量显著增加,Mir-140、Mir-361表达量显著减少(P0.05)。结论:Mir-649、Mir-944、Mir-140、Mir-361在血管平滑肌细胞表型转化中有关键性作用。  相似文献

探讨细胞代数和密度对血管平滑肌细胞(vascular smooth muscle cell,VSMC)表型重塑能力的影响及机制,观察血清饥饿诱导的不同代数和密度的VSMC骨架的组构特征及收缩反应性,检测细胞骨架中收缩蛋白的含量和比例变化。结果发现,低代数(3代)、高密度的VSMC经血清饥饿诱导后易于形成束状、极性排列的应力纤维,乙酰胆碱(Ach)刺激可产生明显的收缩反应。Western印迹显示,3代高密度VSMC中,平滑肌22α(SM22α)在F-肌动蛋白中的组成比例及其在F-/G-肌动蛋白的含量之比明显高于8代细胞。结果提示,SM22α在F-肌动蛋白中的分布比例可能决定了应力纤维的排布方式,是细胞获得收缩性的主要调节因素,在VSMC表型重塑过程中具有重要意义。  相似文献

In rat mesenteric arteries, smooth muscle cells exhibit intercellular calcium waves in response to local phenylephrine stimulation. These waves have a velocity of ∼20 cells/s and a range of ∼80 cells. We analyze these waves in a theoretical model of a population of coupled smooth muscle cells, based on the hypothesis that the wave results from cell membrane depolarization propagation. We study the underlying mechanisms and highlight the importance of voltage-operated channels, calcium-induced calcium release, and chloride channels. Our model is in agreement with experimental observations, and we demonstrate that calcium waves presenting a velocity of ∼20 cells/s can be mediated by electrical coupling. The wave velocity is limited by the time needed for calcium influx through voltage-operated calcium channels and the subsequent calcium-induced calcium release, and not by the speed of the depolarization spreading. The waves are partially regenerated, but have a spatial limit in propagation. Moreover, the model predicts that a refractory period of calcium signaling may significantly affect the wave appearance.  相似文献

MethodsConditional gene targeting generated SMC-specific Nrp1 knockout mice (Nrp1SMKO) in which Cre recombinase is driven by the smooth muscle-specific myosin heavy chain (smMHC) promoter.ResultsSMC-specific Nrp1 deficiency resulted in a significant reduction in intestinal length by 6 months, and, by 18 months, in severe constipation, and enlargement of the intestine consistent with chronic intestinal pseudo-obstruction. These effects were associated with significant thinning of the intestinal smooth muscle, and decreased intestinal contractility. Expression of contractile proteins was reduced in Nrp1SMKO mice, including the smMHC isoform, SMB, whereas we observed a significant increase in the expression of the small-conductance calcium-activated potassium channel 3 (SK3/KCa2.3), implicated in negative regulation of smooth muscle contraction.ConclusionsNrp1 deficiency in visceral SMC results in adult-onset defects in gastrointestinal contractility and motility and causes a shift to a less contractile SMC phenotype. These findings indicate a new role for Nrp1 in the maintenance of the visceral SMC contractile phenotype required for normal GI motility in aged mice.  相似文献

目的:探讨内质网应激(ERS)对肺动脉平滑肌细胞表型转化的影响。方法:采用胶原酶Ⅰ消化法培养原代大鼠肺动脉平滑肌细胞(PASMCs),用衣霉素(TM)或4-苯基丁酸(4-PBA)诱导或抑制内ERS,MTS法评价细胞增殖情况,western blot和定量RT-PCR检测蛋白和mRNA表达情况。结果:TM呈浓度依赖性诱导内质网应激标志物GRP78和XBP1 mRNA表达;较低浓度的TM促进PASMCs增殖,高浓度(5μg/mol)使细胞凋亡;TM使PASMCs表达SM22 alpha减少,分泌Ⅰ型胶原增加;4-PBA预处理可逆转TM诱导PASMCs的SM22 alpha减少和Ⅰ型胶原分泌增加。结论:内质网应激促进肺动脉平滑肌细胞表型转化,可能是内质网应激参与肺动脉高压的机制之一。  相似文献

Bindarit, a selective inhibitor of monocyte chemotactic proteins (MCPs) synthesis, reduces neointimal formation in animal models of vascular injury and recently has been shown to inhibit in-stent late loss in a placebo-controlled phase II clinical trial. However, the mechanisms underlying the efficacy of bindarit in controlling neointimal formation/restenosis have not been fully elucidated. Therefore, we investigated the effect of bindarit on human coronary smooth muscle cells activation, drawing attention to the phenotypic modulation process, focusing on contractile proteins expression as well as proliferation and migration. The expression of contractile proteins was evaluated by western blot analysis on cultured human coronary smooth muscle cells stimulated with TNF-α (30 ng/mL) or fetal bovine serum (5%). Bindarit (100–300 µM) reduced the embryonic form of smooth muscle myosin heavy chain while increased smooth muscle α-actin and calponin in both TNF-α- and fetal bovine serum-stimulated cells. These effects were associated with the inhibition of human coronary smooth muscle cell proliferation/migration and both MCP-1 and MCP-3 production. The effect of bindarit on smooth muscle cells phenotypic switching was confirmed in vivo in the rat balloon angioplasty model. Bindarit (200 mg/Kg/day) significantly reduced the expression of the embryonic form of smooth muscle myosin heavy chain, and increased smooth muscle α-actin and calponin in the rat carodid arteries subjected to endothelial denudation. Our results demonstrate that bindarit induces the differentiated state of human coronary smooth muscle cells, suggesting a novel underlying mechanisms by which this drug inhibits neointimal formation.  相似文献

Background

Patients undergoing radical prostatectomy (RP) are at high risk for erectile dysfunction (ED) due to potential cavernous nerve (CN) damage during surgery. Penile hypoxia after RP is thought to significantly contribute to ED pathogenesis.

Aim

We previously showed that corpora cavernosum smooth muscle cells (CCSMCs) undergo phenotypic modulation under hypoxic conditions in vitro. Here, we studied such changes in an in vivo post-RP ED model by investigating CCSMCs in bilateral cavernous neurectomy (BCN) rats.

Methods

Sprague-Dawley rats underwent sham (n = 12) or BCN (n = 12) surgery. After 12 weeks, they were injected with apomorphine to determine erectile function. The penile tissues were harvested and assessed for fibrosis using Masson trichrome staining and for molecular markers of phenotypic modulation using immunohistochemistry and western blotting. CCSMC morphological structure was evaluated by hematoxylin-eosin (H&E) staining and transmission electron microscopy (TEM).

Results

Erectile function was significantly lower in BCN rats than in sham rats. BCN increased hypoxia-inducible factor-1α and collagen protein expression in corpora cavernous tissue. H&E staining and TEM showed that CCSMCs in BCN rats underwent hypertrophy and showed rough endoplasmic reticulum formation. The expression of CCSMC phenotypic markers, such as smooth muscle α-actin, smooth muscle myosin heavy chain, and desmin, was markedly lower, whereas vimentin protein expression was significantly higher in BCN rats than in control rats.

Conclusions

CCSMCs undergo phenotype modulation in rats with cavernous neurectomy. The results have unveiled physiological transformations that occur at the cellular and molecular levels and have helped characterize CN injury–induced ED.  相似文献

关于肝纤维化形成的复杂的细胞和分子联系已经有了相当多的研究进展。最近的数据表明,纤维化进程的终止和纤维分解途径的恢复可以逆转晚期肝纤维化甚至肝硬化。因此,需要更好地阐明参与肝纤维化的细胞和分子机制。HSC(肝星状细胞)的激活是肝纤维化发生的中心事件,此外还有产生基质的其他细胞来源,包括肝门区的成纤维细胞,纤维细胞和骨髓来源的肌纤维母细胞。这些细胞与其邻近细胞通过多种联系聚集产生纤维疤痕并造成持续性损伤。阐明不同类型的细胞的相互作用,揭示细胞因子对这些细胞的影响,理清活化HSC基因表达的调控,将有助于我们探索新的肝纤维化治疗靶点。此外,不同的病因有不同的致病途径,弄清这一点有助于针对特异性疾病治疗方法的发现。本文概述了肝纤维化的细胞和分子机制的最新研究进展,可能为未来治疗方法带来新的突破。  相似文献

“Ecstasy” [(±)-3,4-methylenedioxymethamphetamine, MDMA, XTC, X, E] is a psychoactive recreational hallucinogenic substance and a major worldwide drug of abuse. Several reports raised the concern that MDMA has the ability to induce neurotoxic effects both in laboratory animals and humans. Despite more than two decades of research, the mechanisms by which MDMA is neurotoxic are still to be fully elucidated. MDMA induces serotonergic terminal loss in rats and also in some mice strains, but also a broader neuronal degeneration throughout several brain areas such as the cortex, hippocampus, and striatum. Meanwhile, in human “ecstasy” abusers, there are evidences for deficits in seronergic biochemical markers, which correlate with long-term impairments in memory and learning. There are several factors that contribute to MDMA-induced neurotoxicity, namely, hyperthermia, monoamine oxidase metabolism of dopamine and serotonin, dopamine oxidation, the serotonin transporter action, nitric oxide, and the formation of peroxinitrite, glutamate excitotoxicity, serotonin 2A receptor agonism, and, importantly, the formation of MDMA neurotoxic metabolites. The present review covered the following topics: history and epidemiology, pharmacological mechanisms, metabolic pathways and the influence of isoenzyme genetic polymorphisms, as well as the acute effects of MDMA in laboratory animals and humans, with a special focus on MDMA-induced neurotoxic effects at the cellular and molecular level. The main aim of this review was to contribute to the understanding of the cellular and molecular mechanisms involved in MDMA neurotoxicity, which can help in the development of therapeutic approaches to prevent or treat the long-term neuropsychiatric complications of MDMA abuse in humans.  相似文献

Metoclopramide (N-(diethylaminoethyl)-2-methoxy-4-amino-5-chlorobenzamide) has been investigated with human gastrointestinal smooth-muscle preparations in vitro. Small-intestinal and colonic circular muscle was contracted by metoclopramide. This effect was antagonized by atropine. Gastric and colonic longitudinal muscle was sensitized by metoclopramide to acetylcholine. Atropine acted here as a partial antagonist of metoclopramide.The interference of acetylcholine and metoclopramide is of a hitherto undescribed pharmacological type. The effects of metoclopramide in vivo—where its action is predominantly on the stomach and pylorus—are not fully explained by these in vitro experiments in which the effect was found to be most pronounced on the colon.  相似文献
