The taxonomy of Lepisorus (J. Sm.) Ching sect. Lepisorus in China was revised based on herbarium specimen examinations, field observations, and microscopic study of rhizome scales, soral paraphyses, leaf epidermis, and spores. As a result, nine species were recognized: Lepisorus macrosphaerus (Baker) Ching, Lepisorus asterolepis (Baker) Ching, Lepisorus marginatus Ching, Lepisorus kuchenensis (Y.C. Wu) Ching, Lepisorus megasorus (C. Chr.) Ching, Lepisorus kawakamii (Hayata) Tagawa, Lepisorus subsessilis Ching & Y.X. Lin, Lepisorus affinis Ching, and Lepisorus nudus (Hook.) Ching. Lepisorus kawakamii (Hayata) Tagawa was reinstated; Lepisorus gyirongensis Ching & S.K. Wu and Lepisorus longus Ching were reduced to synonyms ofL. nudus and L. affinis, respectively. The subdivision ofLepisorus macrosphaerus was not accepted. Rhizome scales and paraphyses are the most useful characters for species delimitation as well as for infrageneric classification. Characteristics of the leaf epidermis and spore ornamentation are usually stable and thus of great significance in understanding the relationships among groups within the genus.  相似文献

Traditional Chinese Medicine (TCM) has been widely and successfully used in treating illnesses ranging from inflammation to cancer, and compounds from medicinal herbs and minerals are playing more and more important roles in taming various kinds of diseases [1 ], exemplified by artemisinin and arsenic trioxide (ATO). Artemisinin (or Qinghaosu in Chinese) is isolated from a plant called sweet wormwood (Artemisia annua; or Qinghao in Chi- nese) which has been used as an antipyretic remedy for more than 1500 years in China. Artemisinin has impressive parasiticidal properties in vitro and in vivo, and is now one of the most important class of antimalarial agents [2, 3]. ATO is a common, naturally occurring substance which had been used in China for a long time as a therapeutic agent for some severe diseases with the ancient philosophy of 'treating an evil with a toxic' [4]. In 1990s, ATO was shown to be able to cause partial differentiation at low dose and apoptosis at high concentration of acute promyelocytic leukemia (APL) cells [5], and induce complete remission in 90% of patients with relapsed or refractory APL [6-8]. These paradigms suggest that TCM is the treasure house not only for the Chinese people, but also for the whole human beings. It is our responsibility to develop evidence-based therapeutic approaches from TCM for diseases with poor prognosis.  相似文献

Nutritional Composition and Assessment of Gracilaria lemaneiformis Bory   总被引:1,自引:0,他引:1  
The chemical composition, mineral elements, vitamins, free fatty acids and amino acid content of the edible red alga Gracilaria lemaneiformis Bory, grown in the sea near Nan'ao island, Guangdong Province, were analyzed in the present study. Gracilaria lemaneiformis Bory showed a total sugar content of 14.65%. The protein content was 21%, of which approximately 41% was determined to be essential amino acids (EAA). The major amino acid components were glutamic acid, leucine, arginine, and alanine. Of the EAA assayed, methionine and cysteine appeared to be the most limiting amino acids compared with the EAA pattern provided by Food and Agricultural Organization of the United Nations. The total lipids content was 0.87% and comprised a high composition of unsaturated fatty acids (61%), mainly as linoleic acid and oleic acid, and a little amount of polyunsaturated fatty acid; palmitic acid was the main component (39%) of saturated acids. Relatively high levels of vitamin C, iodine, phosphorus, and zinc were also present in G. lemaneiformis. The nutritional composition between G. lemaneiformis and Nostoc flagelliforme, a rare alga that is widely eaten in Chinese society, was compared. The results suggest that N. flagelliforme can be substituted for by G. lemaneiformis, not only because of their similar shape, but also because of their approximate nutritional composition. Gracilaria lemaneiformis may possibly serve as a potential healthy food in human diets in the future.  相似文献

In order to amplify the complete genome of enJSRV from the strain of Inner Mongolia (enJSRV-NM), we used enJSRV-specific and JSRV-specific DNA probes in dot blot hybridization. Seven pairs of primers were designed based on Genbank sequences. Seven fragments were obtained by PCR and were cloned into the PMD19-T vectors. The recombinant plasmids were sequenced and analyzed. The results showed that the genome was 7 942 bp in length and contained four overlapping open reading frames corresponding to the gag, pro, pol and env genes as well as an additional open reading frame (orf-x) that overlaps the 3' end of the pol gene. The nucleotide acid sequences of the enJSRV-NM loci were compared with the sequences of South Africa enJS56A1 strain (Accession No.AF153615) and USA JSRV21 strain (Accession No. AF105220). The nucleotide acid identities were 99.2% and 92.3% respectively. Two zinc fingers were found in the NC region in the predicted amino acid sequence. However, the YXXM motif, which is a reliable molecular marker for the infectious exogenous virus, was not found in the TM region. It was found that the enJSRV-NM region was 90%-98% identical at the amino acid level to its exogenous infectious counterparts in most of the retroviral genome. This is the first nucleotide sequence of enJSRV reported in P.R China. The resource work has provided a wide range of information useful not only for expression genomics and annotation of genomic DNA sequence, but also for further research on the clinical diagnosis of OPA.  相似文献

The seasonal changes of insecticide resistance and stability in hymenopteran Cotesia plutellae, collected from Jianxin, Fuzhou-City, and Shangjie, Minhou-County, Fujian, China, were assessed by using a dry residual film method. The resistance to two insecticides in the field populations of C. plutellae was not stable under insecticide-free conditions in the insectarium. Compared with susceptible F11 progeny of C. plutellae in the insectarium, the resistance ratios (RR) in F0 parents were 18.4 for fenvalerate and 11.4 for cypermethrin based on LC50 at 9 hours, and 32.8 for fenvalerate and 28.5 for cypermethrin based on LC50 at 24 hours when the parasitoids were left in contact with the insecticides for 1 hour and mortalities were recorded at 9 and 24 hours, respectively. However, the RR in a field population of C. plutellae were 9.2 for fenvalerate and 12.7 for cypermethrin, if the parasitoids were left in contact with the insecticides for 24 hours. The resistances to the two pyrethroids in other field populations collected from Jianxin and Shangjie from November 2000 and July 2004 were also determined. Significant seasonal variations of resistance to the two insecticides in the field populations of C. plutellae were found. The RR were 3.0-18.4 for fenvalerate and 4.8-20.6 for cypermethrin in Jianxin populations from November 2000 to April 2002 based on LC50 at 9 h, and 2.3-13.6 for fenvalerate and 3.6-16.0 for cypermethrin in Shangjie populations from May 2002 to July 2004 based on LC50 at 24 hours. The resistance levels were high in spring and autumn and decreased sharply in summer. In addition, significant recovery from the knocked-down caused by the insecticides was found in the F0 and field populations of C. plutellae which were resistant to fenvalerate and cypermethrin if the parasitoids were left in contact with the pyrethroids for 1 hour. In this study, an attempt has been made to predict the major functions of gramnegative bacterial proteins from their amino acid sequences. The dataset used for training and testing consists of 670 non-redundant gram-negative bacterial proteins (255 of cellular process, 60 of information molecules, 285 of metabolism, and 70 of virulence factors). First we developed an SVM-based method using amino acid and dipeptide composition and achieved the overall accuracy of 52.39% and 47.01%, respectively. We introduced a new concept for the classification of proteins based on tetrapeptides, in which we identified the unique tetrapeptides significantly found in a class of proteins. These tetrapeptides were used as the input feature for predicting the function of a protein and achieved the overall accuracy of 68.66%. We also developed a hybrid method in which the tetrapeptide information was used with amino acid composition and achieved the overall accuracy of 70.75%. A five-fold cross validation was used to evaluate the performance of these methods. Traditional Chinese medicine(TCM) preparations are widely used for healthcare and clinical practice. So far, the methods commonly used for quality evaluation of TCM preparations mainly focused on chemical ingredients. The biological ingredient analysis of TCM preparations is also important because TCM preparations usually contain both plant and animal ingredients,which often include some mis-identified herbal materials, adulterants or even some biological contaminants.For biological ingredient analysis, the efficiency of DNA extraction is an important factor which might affect the accuracy and reliability of identification. The component complexity in TCM preparations is high, and DNA might be destroyed or degraded in different degrees after a series of processing procedures. Therefore, it is necessary to establish an effective protocol for DNA extraction from TCM preparations. In this study, we chose a classical TCM preparation,Liuwei Dihuang Wan(LDW), as an example to develop a TCM-specific DNA extraction method.An optimized cetyl trimethyl ammonium bromide(CTAB) method(TCM-CTAB) and three commonlyused extraction kits were tested for extraction of DNA from LDW samples. Experimental results indicated that DNA with the highest purity and concentration was obtained by using TCM-CTAB. To further evaluate the different extraction methods, amplification of the second internal transcribed spacer(ITS2) and the chloroplast genome trnL intron was carried out.The results have shown that PCR amplification was successful only with template of DNA extracted by using TCM-CTAB. Moreover, we performed high-throughput 454 sequencing using DNA extracted by TCM-CTAB. The seed cotyledons of a euhalophyte, Suaeda physophora, were found to be dark green. The pigment extracted from the cotyledons was proved to be chlorophyll for the absorption spectra curve of extracts the same as that for leaves. Photosynthetic oxygen-exchanging could be detected after the seeds were moistened for 6 h. Microstructure of organelles in cotyledons of ungerminated seeds was detected by transmission electron microscopy. Histochemical pigmentation was used to investigate the degree of damage on the membrane of radicles. A xerophyte, Haloxylon persicum, whose cotyledons of desiccated seeds also contain chlorophyll was used as a comparative species. The results showed that S. physophora maintained the ultra-structure of chloroplasts, the integrity of plasma membranes of radicles kept much better than that of H. persicum, which showed the great adaptability to salinity of the euhalophyte even at the seed-germination stage. Seeds were incubated in 0 and 700 mmol/L NaCl for 10 days in darkness at 20 ℃ , then ungerminated seeds in NaCl solution were transferred to deionised water and reincubated for another 8 days and was recorded the germination recovery. Dry seeds moistened with deionised water germinated gradually in the dark and germination was maximal after 3 or 4 d. Seeds at 700 mmol/L NaCl for 10 d and transferred back to deionised water germinated abruptly, most seeds germinated in one day and photosynthesis was also detected. It is concluded that recovery germination of S. physophora ensured the seeds could germinate rapidly after salinity declines, for example under the mild but transitory favorable edaphic condition in early spring. Photosynthesis function in seeds might also promoted radicle growth and seedling establishment.  相似文献

Selecting oocytes that are most likely to develop is crucial for in vitro fertilization and animal cloning. Brilliant cresyl blue (BCB) staining has been used for oocyte selection in large animals, but its wider utility needs further evaluation. Mouse oocytes were divided into those stained (BCB+) and those unstained (BCB-) according to their ooplasm BCB coloration. Chromatin configurations, cumulus cell apoptosis, cytoplasmic maturity and developmental competence were compared between the BCB+ and BCB- oocytes. The effects of oocyte diameter, sexual maturity and gonadotropin stimulation on the competence of BCB+ oocytes were also analyzed. In the large- and medium-size groups, BCB+ oocytes were larger and showed more surrounded nucleoli (SN) chromatin configurations and higher frequencies of early atresia, and they also gained better cytoplasmic maturity (determined as the intracellular GSH level and pattern of mitochondrial distribution) and higher developmental potential after in vitro maturation (IVM) than the BCB-oocytes. Adult mice produced more BCB+ oocytes with higher competence than the prepubertal mice when not primed with PMSG. PMSG priming increased both proportion and developmental potency of BCB+ oocytes. The BCB+ oocytes in the large-size group showed more SN chromatin configurations, better cytoplasmic maturity and higher developmental potential than their counterparts in the medium-size group. It is concluded that BCB staining can be used as an efficient method for oocyte selection, but that the competence of the BCB+ oocytes may vary with oocyte diameter, animal sexual maturity and gonadotropin stimulation. Taken together, the series of criteria described here would allow for better choices in selecting oocytes for better development.  相似文献

Aims: Variations and potential trade-offs of leaf hydraulic and photosynthetic traits are essential for assessing and predicting the effect of climate change on tree survival, growth and distribution. Our aims were to examine variations and interrelationships of leaf hydraulic and photosynthetic traits in response to changes in site conditions for Dahurian larch (Larix gmelinii)-a dominant tree species in Chinese boreal forests. Methods: This study was conducted at the Maoershan Forest Ecosystem Research Station. A transect of 27 year-old Dahurian larch plantation was established that consisted of five plots extending from the valley to the ridge of a slope. The predawn leaf water potential (ψpre), area- and mass-based leaf hydraulic conductance (Karea and Kmass, respectively), resistance to embolism capacity (P50), leaf mass per area (LMA), net photosynthetic rate (A), and leaf nitrogen content (N) were measured in August 2016. Important findings: The ψpre, Karea, Kmass, P50, A, LMA, and N all varied significantly among the plots (p < 0.05), indicating significant intra-specific variations in these traits in response to the changes in site conditions. The P50 was significantly (p < 0.05) correlated with ψpre, Karea or Kmass, suggesting that a trade-off between hydraulic efficiency and safety exist within the species to some degree. There were significant (p < 0.05) pairwise correlations between A, LMA, and N. Nevertheless, there was no significant (p < 0.05) correlation between the measured photosynthetic traits and hydraulic traits. We concluded that the intra-specific variations and multiple interrelationships of the leaf hydraulic and photosynthetic traits for the larch reflect the plasticity of its leaf traits and strategies of its survival and growth as a result of its acclimation to diverse site conditions.  相似文献

Pine wilt disease (PWD) is a devastating disease to pine trees, which was caused by the pine wood nematode (PWN), Bursaphelenchus xylophilus, and causes major losses in coniferous forests in many countries and regions in the world. In order to screen actinomycete strains with high nematicidal activity against PWN, marine actinomycetes were isolated from the marine environment and their nematicidal activity were tested.The marine actinomycetes were isolated from the samples collected from the subtidal zones near Qingdao coast using the dilution and streak plate method and their culture supernatant were assayed in vitro for nematicidal activity against PWN using immersion test. The strain with high nematicidal activity was identified on the basis of morphology, cultural characteristics and 16S rDNA sequence analysis, and its optimal culture conditions for the production of nematicidal substances were investigated through the single-factor experiments.A total of 28 marine actinomycete strains were isolated from the samples. One of these strains, designated as strain HT-8 and isolated from sea sand, exhibited stronger nematicidal activity with a 88.30% corrected mortality of PWN treated with the culture supernatant for 30 h. The strain HT-8 was identified as Streptomyces termitum on the basis of morphology, cultural characteristics, 16S rDNA sequence and phylogenetic analysis. The results of the single factor experiment demonstrated that the optimal cultivation conditions of HT-8 for the production of nematicidal substances were inoculum age was 48 h, inoculum concentration was 6%, concentration of seawater was 100%, initial pH was 7.5 and incubating at 25 ℃ for 7 days.This study provides a theoretical basis for the development of marine microorganism resources and the utilization of natural nematicidal substances.  相似文献

Mouse early blastocysts were exposed to temporatures of 39℃ and 41℃ for 2 h,respectively,to determine their developmental competence and uhrastructural changes. The results showed that heat stress at 41 ℃ for 2 h,significantly reduced the percentages of expanded and hatched blastocysts,but not at 39℃ for 2 h. The average cell numbers in expanded blastocysts,which developed from early blastocysts heat-stressed at temperatures of 39℃ and 41 ℃,were significantly reduced. The average cell numbers in hatched blastocysts subjected to heat stress were no different from those in the control group cultured at 37℃ . The mitochondria of the early blastocysts heat-stressed at 39T℃ for 2 h,were slightly swollen,but they had recovered after culturing at 37℃ for 2 h. However,the mitochondria in the blastocysts heat stressed at 41 ℃ for 2 h were severely swollen,and their number increased. The ribosomes shed from the rough endoplasmic reticulum,and the number of secondary lysosomes in the plasma increased. The integrity of desmosomes was disrupted. The space between the nuclear envelope and the perivitelline membrane enlarged. The fibre fraction and the particulate fraction of nucleoli were separated. The heterochromatin in nucleoli was also increased in its quantity. There were some lamellar-shape structures and heterogeneous dense materials exhibiting in the cytoplasm. The ultrastructural changes induced by heat shock at 41 ℃ for 2 h were not reversible. In conclusion,the damage of heat stress to mitochondria,lysosomes,ribosomes and cell nucleus,may be one of the most important factors that inhibit the normal development of mouse early blastocysts.  相似文献

Aims Alpine shrub-meadows and steppe-meadows are the two dominant vegetation types on the Qinghai-Xizang Plateau, and plays an important role in regional carbon cycling. However, little is known about the temporal- spatial patterns and drivers of CO2 fluxes in these two ecosystem types. Methods Based on five years of consecutive eddy covariance measurements (2004-2008) in an eastern alpine shrub-meadow at Haibei and a hinterland alpine steppe-meadow at Damxung, we investigated the seasonal and annual variation of net ecosystem productivity (NEP) and its components, i.e. gross primary productivity (GPP) and ecosystem respiration (Re). Important findings The CO2 fluxes (NEP, GPP and Re) were larger in the shrub-meadow than in the steppe-meadow during the study period. The shrub-meadow functioned as a carbon sink through the five years, with the mean annual NEP of 70 g C·m-2·a-1. However, the steppe-meadow acted as a carbon neutral, with mean annual NEP of -5 g C·m-2·a-1. The CO2 fluxes of steppe-meadow exhibited large variability due to the inter-Annual and seasonal variations in precipitation, ranging from a carbon sink (54 g C·m-2·a-1) in 2008 to a carbon source (-88 g C·m-2·a-1) in 2006. The differences in carbon budget between the two alpine ecosystems were firstly attributed to the discrepancy of normalized difference vegetation index (NDVI) because NDVI was the direct factor regulating the seasonal and inter-Annual NEP. Secondly, the shrub-meadow had higher carbon use efficiency (CUE), which was substantially determined by annual precipitation (PPT) and NDVI. Our results also indicated that the environmental drivers of CO2 fluxes were also different between these two alpine ecosystems. The structure equation model analyses showed that air temperature (Ta) determined the seasonal variations of CO2 fluxes in the shrub-meadow, with NEP and GPP being positively correlated with Ta. By contrast, the seasonal CO22 fluxes in the steppe-meadow were primarily co-regulated by soil water content (SWC) and Ta, and increased with the increase of SWC and Ta. In addition, the changes of Re during the growing season in two ecosystems were directly affected by GPP and soil temperature at 5 cm depth (Ts), while Re during non-growing season were determined by Ts. These results demonstrate that the synergy of soil water and temperature played crucial roles in determining NEP and GPP of the two alpine meadows on the Qinghai-Xizang Plateau. © 2018 Editorial Office of Chinese Journal of Plant Ecology. Aims Global nitrogen (N) deposition not only alters soil N and phosphorus (P) availability, but also changes their ratio. The levels and ratios of N and P supply and their interaction may simultaneously influence plant seed traits. However, so far there has been no experiments to distinguish these complex impacts on plant seed traits in the field. Methods A pot experiment with a factorial design of three levels and ratios of N and P supply was conducted in the Nei Mongol grassland to explore the effects of levels and ratios of N and P supply and their interaction on seed traits of Chenopodium glaucum. Important findings We found that the relative contribution (15%–24%) of N and P supply levels in affecting the N concentrations, P concentrations and germination rates of seeds was larger than that (3%–7%) of N:P supply ratios, whereas seed size was only significantly influenced by N:P. Simultaneously, seed N and P concentrations were impacted by the interaction of N and P supply levels and ratios. At the same N:P, decrease in nutrient supply levels increased seed N concentrations, P concentrations and germination rates. N:P supply ratios only had a significant effect on seed size and germination rates under low nutrient levels. Overall, these results indicate that different seed traits of C. glaucum show different sensitivities to N or P limitations, leading to adaptive and passive responses under different nutrient limitations. This study presents the the first field experiment to distinguish the effects of nutrient supply levels, ratios and their interactions on plant seed traits, which provides a new case study on the influences of global N deposition on future dynamics of plant population and community. © Chinese Journal of Plant Ecology.  相似文献

Aims Our objective was to estimate the carbon storage in the forest tree layer in Qinghai Province, China. Methods Based on forest resource inventory data and field investigation data, we estimated the carbon storage, sequestration rate and potentials in the forest tree layer in the Qinghai Province. Important findings The carbon density and total carbon storage of forest tree layer in Qinghai Province was 76.54 Mg·hm-2 and 27.38 Tg, respectively, of which four forest types (Picea spp. forest, Cupressus funebris forest, Betula spp. forest and Populus spp. forest) accounted for 86.67% while their areas were 96.23% of total forest areas in Qinghai. The carbon density and carbon storage of Picea spp. forest was 106.93 Mg·hm-2 and 14.78 Tg, respectively, which was the largest among all forest types. The carbon storage of the forest tree layer at different stand ages followed the sequence of over-mature forest > middle-aged forest > mature forest > near-mature forest > young forest. In addition, the carbon storage of forest tree layer in the province increased from 23.30 Tg in 2003 to 27.38 Tg in 2011. The average annual growth of carbon and carbon sequestration rate were 0.51 Tg and 1.06 Mg·hm-2·a-1, respectively. The maximum and minimum of carbon sequestration rate were respectively found in Cupressus funebris forest (0.44 Mg·hm-2·a-1) and Betula spp. forest (-1.06 Mg·hm-2·a-1). The mean carbon sequestration potential reached 8.50 Tg in 2011, with the highest value found in Picea spp. forest (3.40 Tg). These findings suggested high carbon sequestration potential of forest tree layer in Qinghai Province. The thirty-three 5′ flanking conserved sequences of the known low-molecular-weight subunit (LMW-GS) genes have been divided into eight clusters, which was in agreement with the classification based on the deduced N-terminal protein sequences. The DNA polymorphism between the eight clusters was obtained by sequence alignment, and a total of 34 polymorphic positions were observed in the approximately 200 bp regions, among which 18 polymorphic positions were candidate SNPs. Seven cluster-specific primer sets were designed for seven out of eight clusters containing cluster-specific bases, with which the genomic DNA of the ditelosomic lines of group 1 chromosomes of a wheat variety ‘Chinese Spring’ was employed to carry out chromosome assignment. The subsequent cloning and DNA sequencing of PCR fragments validated the sequences specificity of the 5′ flanking conserved sequences between LMW-GS gene groups in different genomes. These results suggested that the coding and 5′ flanking regions of LMW-GS genes are likely to have evolved in a concerted fashion. The seven primer sets developed in this study could be used to isolate the complete ORFs of seven groups of LMW-GS genes, respectively, and therefore possess great value for further research in the contributions of a single LMW-GS gene to wheat quality in the complex genetic background and the efficient selections of quality-related components in breeding programs.  相似文献
