Using flow cytometry, we measured the nuclear DNA contents of all known taxa in Hepatica. Nuclear DNA content of Hepatica falconeri (diploid, crenate leaf lobes) was significantly lower than that of diploid entire species. Among the tetraploid species, crenate species had lower DNA contents than the entire taxon H. nobilis var. pubescens. The DNA content of the tetraploid species was more than double that of the diploid species among the same leaf-type groups. 相似文献

Cells adapt to drastic changes in genome quantity during evolution and cell division by adjusting the nuclear size to exert genomic functions. However, the mechanism by which DNA content within the nucleus contributes to controlling the nuclear size remains unclear. Here, we experimentally evaluated the effects of DNA content by utilizing cell-free Xenopus egg extracts and imaging of in vivo embryos. Upon manipulation of DNA content while maintaining cytoplasmic effects constant, both plateau size and expansion speed of the nucleus correlated highly with DNA content. We also found that nuclear expansion dynamics was altered when chromatin interaction with the nuclear envelope or chromatin condensation was manipulated while maintaining DNA content constant. Furthermore, excess membrane accumulated on the nuclear surface when the DNA content was low. These results clearly demonstrate that nuclear expansion is determined not only by cytoplasmic membrane supply but also by the physical properties of chromatin, including DNA quantity and chromatin structure within the nucleus, rather than the coding sequences themselves. In controlling the dynamics of nuclear expansion, we propose that chromatin interaction with the nuclear envelope plays a role in transmitting chromatin repulsion forces to the nuclear membrane. 相似文献

The oytogenetic analysis of the genus Xenopus shows that X. laevis laevis, X. laevis petersi, X. laevis victorianus, X. (laevis) borealis, X. gilli, X. muelleri, and X. fraseri have chromosome numbers 2n=36; X. tropicalis has 20 (2n), X. (laevis) bunyoniensis 72 and X. ruwenzoriensis 108. This heterogeneity of the chromosome numbers is interesting as it represents new examples of polyploidy among Anurans. There are no big morphological differences among the karyotypes of the divers species, only the chromosomes with secondary constrictions vary considerably. 相似文献

DNA content was estimated by flow cytometry in seventeen taxa from the Dilatata, Quadrifaria and Paniculata groups of Paspalum and five synthetic hybrids. Results were compared to known genome constitutions and phylogenetic relationships. DNA 2C-values
ranged from 1.24 pg in diploid P. juergensii to 3.79 pg in a hexaploid biotype of P. dilatatum. The I genome of three Quadrifaria diploids is 1.2 to 1.5-fold larger than the J genome of P. juergensii (Paniculata). The 2C-values of the IIJJ tetraploids of the Dilatata group are lower than expected based on putative genome
donors. Reduction of genome sizes could have occurred after the formation of the allopolyploids of the Dilatata group. The
DNA content of all synthetic hybrids is in accordance with the sum of parental C-values. The interactions driving genome downsizing

The morphogenesis of sperm nuclei was investigated in six different species or subspecies of the genus Xenopus (Pipidae, Anura). The sequence of nuclear morphogenesis was similar in each species used in this study. Electrophoretic comparison of the basic chromatin proteins from late spermatids and sperm of each species demonstrated that the complements of histones and spermatid-sperm-specific basic proteins were extremely diverse suggesting that shape was not determined by specific basic proteins or mechanisms of histone removal. This conclusion was reinforced by the observation that Xenopus sperm DNA decondensed by 2.0 M NaCl remained contrained in residual structures which resembled intact sperm nuclei. These observations suggested that morphogenesis of sperm nuclei is directed by proteins or RNA molecules which are not directly responsible for chromatin condensation. 相似文献

The 2C nuclear DNA content has been estimated by flow cytometry in 18 species and botanical forms of the genus Lupinus (family Fabaceae), using propidium iodide as a fluorescent dye. They represented distinct infrageneric taxonomic groups and differed in somatic chromosome numbers. Estimated 2C DNA values ranged from 0.97 pg in L. princei to 2.44 pg in L. luteus, which gives a more than 2.5-fold variation. Statistical analysis of the data obtained resulted in a grouping that supports the generally accepted taxonomic classification of the Old World lupins. The rough-seeded L. princei turned out to be an interesting exception, getting closer to smooth-seeded species. Results of DNA content analyses are discussed with regards to the phylogenetic relationships among the Old World lupins and some aspects of the evolution of the genus. 相似文献

The genus Brucella has been described as consisting of six species, three of them including several biovars, which display a high degree of DNA homology by DNA-DNA hybridization. However, DNA polymorphism able to differentiate the six Brucella species and some of their biovars has been shown to exist. This work reviews the DNA variability in the genus Brucella and discusses the relationships between its members according to this genetic variability and a proposal for their evolution based on genetic diversity of the omp2 locus. 相似文献

Mitochondrial DNA evolution in the genus Equus 总被引:7,自引:0,他引:7
Employing mitochondrial DNA (mtDNA) restriction-endonuclease maps as the
basis of comparison, we have investigated the evolutionary affinities of
the seven species generally recognized as the genus Equus. Individual
species' cleavage maps contained an average of 60 cleavage sites for 16
enzymes, of which 29 were invariant for all species. Based on an average
divergence rate of 2%/Myr, the variation between species supports a
divergence of extant lineages from a common ancestor approximately 3.9 Myr
before the present. Comparisons of cleavage maps between Equus przewalskii
(Mongolian wild horse) and E. caballus (domestic horse) yielded estimates
of nucleotide sequence divergence ranging from 0.27% to 0.41%. This range
was due to intraspecific variation, which was noted only for E. caballus.
For pairwise comparisons within this family, estimates of sequence
divergence ranged from 0% (E. hemionus onager vs. E. h. kulan) to 7.8% (E.
przewalskii vs. E. h. onager). Trees constructed according to the parsimony
principle, on the basis of 31 phylogenetically informative restriction
sites, indicate that the three extant zebra species represent a
monophyletic group with E. grevyi and E. burchelli antiquorum diverging
most recently. The phylogenetic relationships of E. africanus and E.
hemionus remain enigmatic on the basis of the mtDNA analysis, although a
recent divergence is unsupported.

Microgel cell electrophoresis has been used with various species to measure breakage of DNA and DNA repair following exposure to the radiomimetic antibiotic, bleomycin. With humans, a high degree of DNA damage is considered to be predictive of cancer susceptibility. Non-isogeneic Xenopus laevis, the South African clawed toad, rarely develop spontaneous or induced cancers. Here, we investigate bleomycin-induced DNA damage and repair in splenic lymphocytes of this species to test consistency with cancer predictability. As X. laevis is pseudotetraploid in nature, while Xenopus tropicalis is diploid, we additionally explore the effect of polyploidy on DNA damage and repair in these vertebrates. The results show that higher doses of bleomycin are required to induce comparable levels of DNA damage in both Xenopus species, than in humans. X. tropicalis, the diploid, is more bleomycin-sensitive than is X. laevis. Additionally, repair rates of damaged DNA of X. laevis lymphocytes are more rapid than those of X. tropicalis, although both are hours slower than human leukocytes. While no data exist on cancer susceptibility in X. tropicalis, the results suggest greater susceptibility to cancer than X. laevis, but less than in humans. Thus, polyploidy serves as a protection against DNA damage and allows more rapid repair. 相似文献

Estimates of the 4C DNA amount per nucleus in 16 taxa of the genus Secale made by Feulgen microdensitometry ranged from 28.85 picograms (pg) in S. silvestre PBI R52 to 34.58 pg in S. vavilovii UM 2D49, compared with 33.14 pg in S. cereale cv. Petkus Spring which was used as a standard. Giemsa C-banding patterns showed considerable interspecific and intraspecific variation and several instances of polymorphism for large telomeric C-bands. The proportion of telomeric heterochromatin in the genome ranged from about 6% in S. silvestre and S. africanum to about 12% in cultivated rye. A detailed comparison of nine taxa showed no overall relationship between 4C DNA amount and the proportion of telomeric heterochromatin in the genome. However, evidence is presented which strongly supports the notion that the major evolutionary change in chromosome structure in Secale has involved the addition of heterochromatin at, or close to, the telomeres. It is suggested that saltatory amplification events at telomeres were initially responsible for each large increase in DNA amount. Subsequently unequal crossing over between homologues may have played an important secondary role by extending the range of variation in the amount of heterochromatin at a given telomere, while crossing over between non-homologues may have provided a useful mechanism allowing an increase in the DNA amount at one telomere to be distributed between chromosomes. 相似文献

A method for preparing polysomes from all embryonic stages of Xenopus laevis is described. In the oocyte only about 1–2% of the total ribosomes are present in polysomes, the remainder being a developmental reserve. Upon conversion to an egg the polysome content rises by up to 3-fold, and by about a further 2-fold after fertilization. There is only a small further increase during cleavage, but by the tailbud stage, when organogenesis begins, there is a more rapid rise. Most of the ribosomes are incorporated into polysomes by stage 42, shortly before feeding begins.At very early stages, the changes in polysome content seem to mirror the changes in protein synthesis. At later stages the polysome contents reported here provide the only available guide to changes in the rate of protein synthesis. Judged by polysome content, the stage 42 tadpole seems to make protein about 20 times faster than the unfertilized egg, though it contains very few more ribosomes. The relationship between polysome content and the synthesis of various types of RNA is discussed. 相似文献

Nucleolar expression in reciprocal hybrids between African frogs of the species Xenopus laevis and X. mulleri has been examined throughout ontogeny. Evidence is presented for the stagespecific regulation of nucleolar expression in these hybrids, and for a maternal effect that operates during the early development. Advantage has been taken of the nucleolar organizer deletion in X. laevis to demonstrate that the uninucleolate conditions of hybrid nuclei at certain developmental stages is the result of an allelic repression of the mulleri nucleolar organizer region by the laevis genome. Differences in nucleolar expression between the reciprocal hybrids and the parental species have been put on a quantitative basis for several selected tissues. 相似文献

Representatives of two major repetitive DNA sequence families from the diploid Brassica species B. campestris and B. oleracea were isolated, sequenced and localized to chromosomes by in situ hybridization. Both sequences were located near the centromeres of many chromosome pairs in both diploid species, but major sites of the two probes were all on different chromosome pairs. Such chromosome specificity is unusual for plant paracentromeric repetitive DNA. Reduction of stringency of hybridization gave centromeric hybridization sites on more chromosomes, indicating that there are divergent sequences present on other chromosomes. In tetraploid species derived from the diploids, the number of hybridization sites was different from the sum of the diploid ancestors, and some chromosomes had both sequences, indicating relatively rapid homogenization and copy number evolution since the origin of the tetraploid species. 相似文献

Comparison of electrophoretic globin phenotypes of 18 different Xenopus
taxa reveals four different basic types of banding patterns. Each type
includes species that also are similar in their morphological,
cytogenetical, and biochemical characteristics. Three of these patterns
reflect distinct evolutionary lines, while the fourth may be interpreted as
the intersection of two of these lines. The composition of the basic
pattern of the highly polyploid species is consistent with an allopolyploid
origin of most of these species. The number of distinct globin
polypeptides--four in the only extant diploid species, X. tropicalis, and
five or more in most of the tetraploid species, including X.
laevis--suggests that primordial globin genes had undergone duplication
either before or after the tetraploidization event. Finally, the individual
globin phenotypes are excellent molecular markers that are of great help in
identifying the various species but not the subspecies.

Summary DNAs have been isolated from various Oryza species and studied using physical techniques. The percent of guanine plus cytosine has been determined by thermal denaturation. While the base composition varied between the species, no heterogeneity in the base pair distribution was observed. Renaturation kinetics data of DNAs from different species show that the proportion of repeated DNA sequences vary considerably depending on the DNA content per cell, whereas the nonrepetitive DNA component remains relatively constant. These results suggest that in addition to a small range of DNA variation between the species, changes in the base composition and proportion of repeated sequences have accompanied divergence of the species within the genus. 相似文献
