The clam Ruditapes decussatus is distributed worldwide and due to its ecological and economical interest has been proposed as a bioindicator in areas where mussels are not available. The accumulation of several anthropogenic compounds in their tissues suggests that they possess mechanisms that allow them to cope with the toxic effects of these contaminants. Besides pollutant uptake, the use of biomarkers is pointed out in this paper since it is a promising approach to monitor the effect of these contaminants in the marine environment. Biomarkers complement the information of the direct chemical characterization of different types of contaminants. Therefore, the aim of this paper is to review the role of several biomarkers: (metallothioneins (MT), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidases (GPx) (total and selenium-dependent), lipid peroxidation (measured as MDA, one of the final products of lipid peroxidation), glutathione S-transferase (GST) and acetylcholinesterase (AChE), measured in different tissues of the clam R. decussatus, in laboratory conditions and under various environmental stresses, in two ecosystems (Ria Formosa lagoon- Portugal) and Bizerta lagoon (Tunisia) in a perspective of a multibiomarker approach to assess environmental changes. Experiment and field studies are in good agreement since MT levels, especially in the gills, the first target tissue of these contaminants, can be used as biomarker of exposure to Cd. GPx and MDA may also be determined in this respect. AChE activity is inhibited by pesticide and, to a less extent, by metal exposure in the gills and whole soft body of clams. However, the induction of GST isoforms experimentally demonstrated is not observed in the field because only global GST activity was determined. The whole set of results opens new research perspectives for the use of this species to assess the effect of mixtures of pollutants in the aquatic environment.  相似文献

Ammonium concentrations were determined in near-bottom water and intertidal surface sediments collected in February and July 1993 at five stations of Ria Formosa, a shallow meso-tidal coastal lagoon in southern Portugal. At each station, samples were taken a few minutes before tidal inundation, and subsequently 2, 10, 15 and 20 minutes thereafter. Ammonium concentration in near-bottom waters increased dramatically in the first 2 minutes followed by a decrease during the 18 minutes of flooding (maximum range 10.3-2.2 M). The highest levels in the flooding water were concomitant with a decrease of extractable ammonium recorded in the upper sediment layer (2 cm). Laboratory experiments indicated that ammonium is easily extracted from the sediment solids by physical perturbation, as one would expect when tidal water flushes over the intertidal area. This perturbation results in the export of ammonium from the sediment, by pulse mechanisms of short time intervals. On a daily scale this amount is two orders of magnitude higher than transport resulting from molecular diffusion.  相似文献

The effect of nest aggregation in courtship behaviour was tested experimentally in an ecologically constrained, sex-role reversed population of the peacock blenny Salaria pavo . Mixed sex groups of eight males and eight females were tested in experimental tanks, containing eight potential nests either aggregated or dispersed. In the aggregated treatment, males spent more time inside their nests and monopolized other potential nests, causing a female-biased operational sex ratio (OSR). In the aggregated treatment, females also expressed more courtship behaviour. The results in general support the prediction that the aggregation of nests promotes male monopolization of potential nests, resulting in fewer nest-holding males and therefore a female-biased OSR that leads to the reversal of sex roles.  相似文献

Bivalves are excellent candidates for geographically based studies of the morphological variation in individuals of different populations based on the analysis of their shape profiles. In this study, we quantified the overall shell shape differences in individuals of different populations of Ruditapes decussatus and Ruditapes philippinarum in relation to their geographical and genetic distances. A total of 395 and 124 individuals of R. decussatus (nine populations) and R. philippinarum (four populations), respectively, were sampled in different Mediterranean and Atlantic coastal locations. Pictures of the left valve were taken from all individuals. Their profiles were analysed using elliptic Fourier analysis. Mean outlines were computed. In order to classify different individuals for species, the coefficients of harmonic equations were analysed by partial least square discriminant analysis and soft independent modelling of class analogy. The results showed a high percentage of correct classification (99%) between the two species in the independent test. We found that the morphological distance between R. philippinarum and R. decussatus is higher than the morphological distance among populations of the same species. The absence of correspondence between the geographical location and the pattern of morphological and genetic variation indicates the occurrence of a reaction norm in the morphological adaptation of shell shapes to different local environmental conditions.  相似文献

The purple dye murex, Bolinus brandaris (Linnaeus, 1758), is a muricid gastropod common throughout the Mediterranean and along the Moroccan and Portuguese Atlantic coasts. In the present study, we confirmed the diploid chromosome number of 2 n  = 70 for this species, and established for the first time the karyotype, which comprised 12 metacentric, 15 submetacentric and eight subtelocentric chromosome pairs. To facilitate cytotaxonomic comparisons, we carried out a comparative karyological analysis through multidimensional scaling between B. brandaris and three other 2 n  = 70 muricid species ( Hexaplex trunculus , Ocenebra erinaceus , and Stramonita haemastoma ) for which chromosomal measurements have been previously published. The interpretation of the ideograms and the statistical analysis highlighted the closest similarity of B. brandaris and H. trunculus compared to S. haemastoma and O. erinaceus . Indeed, B. brandaris and H. trunculus showed the smallest dissimilarities both for relative length and arm ratio, with O. erinaceus presenting intermediate values, whereas the highest dissimilarities were found between H. trunculus and S. haemastoma for both data. The karyotypes of B. brandaris and H. trunculus (subfamily Muricinae) presented the highest proportions of metacentric chromosomes compared to the other two muricids analysed, suggesting that those karyotypes could be considered primitive within the 2 n  = 70 Muricidae studied so far.  © 2009 The Linnean Society of London, Biological Journal of the Linnean Society , 2009, 96 , 185–193.  相似文献

Submarine Groundwater Discharge (SGD) has been frequently ignored as a nutrient source to marine ecosystems because it is difficult to identify and quantify. However, recent studies show its ubiquity and ecological importance to the coastal zone, particularly when associated with contaminated continental aquifers. The Ria Formosa is a coastal lagoon located in the south of Portugal and surrounded by an intensely farmed area. Following a 12-month field study using seepage meters, we identified groundwater discharge in the intertidal zone of the lagoon. The seeping fluid was a mixture of two water types: one with low salinity and high nitrate concentration and another similar to local seawater. Based on the integration of monthly seepage rate measurements throughout the year, we estimate the mean discharge of submarine groundwater into the lagoon to be 3.6 mday−1 per linear meter of coastline with freshwater contributions (per volume) ranging from 10% to 50%. The results of this study suggest a continental origin for the freshwater component, thus linking the biogeochemical cycles in the lagoon to anthropogenic activities taking place in the neighboring coastal plain. We further identify SGD as an important nutrient source to the Ria Formosa, estimating annual loads of 36.2 mol (0.507 kg) of Nitrogen, 1.1 mol (0.034 kg) of Phosphorus and 18.6 mol (0.522 kg) of Silicon per meter of coastline. Based on these results, we suggest that SGD is a potential contributor to the observed nutrification status of the Ria Formosa lagoon. Most amphibians breed in water, including the terrestrial species, and may therefore be exposed to water-borne pharmaceuticals during critical phases of the reproductive cycle, i.e. sex differentiation and gamete maturation. The objectives of this paper were to (i) review available literature regarding adverse effects of hormonally active pharmaceuticals on amphibians, with special reference to environmentally relevant exposure levels and (ii) expand the knowledge on toxicity of progestagens in amphibians by determining effects of norethindrone (NET) and progesterone (P) exposure to 0, 1, 10 or 100 ng l−1 (nominal) on oogenesis in the test species Xenopus tropicalis. Very little information was found on toxicity of environmentally relevant concentrations of pharmaceuticals on amphibians. Research has shown that environmental concentrations (1.8 ng l−1) of the pharmaceutical oestrogen ethinylestradiol (EE2) cause developmental reproductive toxicity involving impaired spermatogenesis in frogs. Recently, it was found that the progestagen levonorgestrel (LNG) inhibited oogenesis in frogs by interrupting the formation of vitellogenic oocytes at an environmentally relevant concentration (1.3 ng l−1). Results from the present study revealed that 1 ng NET l−1 and 10 ng P l−1 caused reduced proportions of vitellogenic oocytes and increased proportions of previtellogenic oocytes compared with the controls, thereby indicating inhibited vitellogenesis. Hence, the available literature shows that the oestrogen EE2 and the progestagens LNG, NET and P impair reproductive functions in amphibians at environmentally relevant exposure concentrations. The progestagens are of particular concern given their prevalence, the range of compounds and that several of them (LNG, NET and P) share the same target (oogenesis) at environmental exposure concentrations, indicating a risk for adverse effects on fertility in exposed wild amphibians.  相似文献

Despite worldwide proliferation of the genus Caulerpa and subsequent effects on benthic communities, little is known about the nutritional physiology of the Caulerpales. Here, we investigated the uptake rates of ammonium, nitrate, amino acids, and phosphate through the fronds and rhizoids + stolon, the internal translocation of nitrogen, and developed a nitrogen budget for the rapidly spreading Caulerpa prolifera in Ria Formosa lagoon, southern Portugal. Caulerpa prolifera acquired nutrients by both aboveground and belowground parts at similar rates, except nitrate, for which fronds showed 2-fold higher uptake rates. Ammonium was the preferential nitrogen source (81% of the total nitrogen acquisition), and amino acids, which accounted for a significant fraction of total N acquisition (19%), were taken up at faster rates than nitrate. Basipetal translocation of 15N incorporated as ammonium was nearly 3-fold higher than acropetal translocation, whereas 15N translocation as nitrate and amino acids was smaller but equal in either direction. The estimated total nitrogen acquisition by C. prolifera was 689 μmol · m−2 · h−1, whereas the total nitrogen requirement for growth was 672 μmol · m−2 · h−1. The uptake of ammonium and amino acids by belowground parts accounted for the larger fraction of the total nitrogen acquisition of C. prolifera and is sufficient to satisfy the species nitrogen requirements for growth. This may be one reason explaining the fast spreading of the seaweed in the bare sediments of Ria Formosa where it does not have any macrophyte competitors and the concentration of nutrients is high.  相似文献

In this study, the variations of glycogen concentrations in Ruditapes decussatus from Sfax coasts (Tunisia) were described in relation to the reproductive cycle. Separate analyses were made of gonad, adductor muscle and ‘remainder’. The timing of gametogenic development and spawning was evaluated using qualitative histology associated with image analysis including (1) the estimation of the gonadal occupation index (GOI), (2) surface area occupied by reserve tissues and (3) variation in oocyte diameter. The reproductive cycle of R. decussatus exhibited partial asynchronization between sexes, the major difference being observed in the duration of the spawning period. Contrary to previous studies, we observed continuous partial spawning (e.g. 50% of ripe oocytes still subsisted at the partial spawning stage). During the gametogenic cycle, GOI varied significantly in males and in females. Most oocytes were ripe and ready to spawn when their diameter reached or exceeded 45?µm. The expansion of gonad was inversely proportional to the development of the foot tissue. Glycogen concentration showed significant temporal variations between tissues, indicating the clear differentiation in energy storage between the clam organs.  相似文献

黄体酮(P4)是一种类固醇激素。为了探究P4的内分泌干扰效应, 选择成年斑马鱼(Danio rerio)作为受试生物, 研究了P4对斑马鱼下丘脑-垂体-性腺轴(HPG轴)相关基因转录表达影响。成年斑马鱼在不同浓度P4(2、11和16 ng•L–1)下处理21 d。结果显示: 暴露于高浓度组的P4能够抑制雌鱼大脑中促性腺激素释放激素2(gnrh2)、促性腺激素释放激素3(gnrh3), 卵泡刺激素(fshb)、雌激素受体1(esr1)基因的转录表达; 然而诱导了雄鱼大脑中fshb、黄体生成素(lhb)、雄激素受体(ar)基因的转录表达, 这些转录变化暗示了P4对成年斑马鱼有潜在的弱雄激素效应。此外, P4暴露对雌鱼卵巢和雄鱼精巢类固醇合成途径中固醇激素合成急性调节蛋白(star)、细胞色素p450介导侧链裂解酶(cyp11a1)、17α羟化酶(cyp17)、卵巢细胞色素P450芳香化酶(cyp19a1a)、11β羟化酶(cyp11b)、羟基类固醇3β脱氢酶(hsd3b)、羟基类固醇20β脱氢酶(hsd20b)、羟基类固醇17β脱氢酶3(hsd17b3)、羟基类固醇11β脱氢酶2(hsd11b2)以及受体信号途径中孕激素受体(pgr)、esr1、ar基因的转录表达没有显著影响。可见, 在P4暴露下, 斑马鱼大脑比性腺更加敏感。总而言之, P4能够改变斑马鱼大脑中HPG轴相关基因的转录表达水平, 进而对斑马鱼的内分泌系统具有潜在的危险。  相似文献

Purification and characterization of microsomal glutathione S-transferase produced by Aspergillus ochraceus TS are reported. The isozymes are located in microsomes and were active against 1-chloro-2,4-dinitrobenzene, ethacrynic acid, 1,2-dichloro-4-nitrobenzene, trans-4- phenyl-3-buten-2-one,p-nitrobenzyl chloride and bromosulphophthalein. They were inhibited by N-ethylmaleimide and bromosulphophthalein. The GST isozymes produced by Aspergillus ochraceus TS are indistinguishable in respect of their molecular mass both in native and denatured state. The subunit of the purified protein had an apparent Mr of 11 kDa while molecular mass of the native protein is around 56 kDa. The substrate specificity and pl values of the isozymes were different. The GSTs produced by Aspergillus ochraceus TS fairly share functional properties with mammalian cytosolic isozymes.  相似文献

In the Ria Formosa lagoon (S. Portugal), Carcinus maenas larvae have been observed only during the cold part of the year (October–May), when the water temperature stays below 23 °C. Larvae suffer high mortality during day time (about 80% during 6 h). Hence, they will only develop outside the lagoon, as proved by the observation of zoea I stages exclusively. Main recruitment starts in April, i.e. much later than the first larvae appear. Small crabs prefer Zostera noltii patches and colonize other sites when the carapace width exceeds about 5 mm. A reserve of some small crabs is found in Zostera noltii patches nearly all year round. It is postulated that one of the key factors for successful propagation of the species is a match between physiological reactions (here in particular temperature effect on reproductive cycle and larval release), and the ecological conditions for larval survival and recruitment, such as predator impact, food availability or currents.  相似文献

Maximum nitrate reductase (NR) activity was measured in two intertidal morphotypes of Zostera noltii (Hornem.) in Ria Formosa tidal lagoon, southern Portugal. The two morphotypes develop in the upper and lower limits of the intertidal meadows. The NR activity was measured using an in vivo method, without cell disruption. NR activity was 30-40 fold higher in leaves than in roots, which indicates that nitrate reduction is essentially made through the aerial part of the plant. The effects of assay temperature (5 °C steps, from 5 to 45 °C), pH (7, 8 and 9) and elevation (upper and lower intertidal) on leaf NR activity were tested in a factorial design (n=5). Both elevation and assay temperature had a significant effect on NR activity, but not pH. NR activity was always higher in the upper intertidal plants, at all temperatures. Activity peaks for upper and lower plants were, respectively, 6.12 μmol NO2 g−1 DW 0.5 h−1 at 25 °C, and 3.30 μmol NO2 g−1 DW 0.5h−1 at 35 °C. Further investigation on environmental factors concerning the intertidal environment must be developed, as they are probably responsible for the significant differences found between the values of NR activity in the upper and lower morphotype.  相似文献

Among the 19 non-native species of marine invertebrates which have invaded the Venice Lagoon and have established populations, Ruditapes philippinarum, deliberately introduced in 1983, is surely the most successful species. According to the hypothesis that alien species invasion could be favoured by an altered ecological, chemical or physical state of the system induced by anthropogenic disturbance, R. philippinarum turned out to be ‘the right species at the right moment’. By comparing historical data (1968, 1985, 1990) with 1999 data, changes in macrobenthic community, in particular bivalve molluscs, of the lagoon induced by R. philippinarum introduction and subsequent clam exploiting activity were assessed. It has been possible to describe a sharp reduction, both in terms of distribution area and density, of all other filter feeder bivalves. Moreover, by using the clearance rate of the most abundant bivalve species in 1990 and 1999 (Cerastoderma glaucum and R. philippinarum, respectively), it was possible to estimate that the filtration capacity, expressed as l h−1 m−2, has more than doubled. This has altered the functioning of the ecosystem, resulting in a stronger benthic–pelagic coupling. In this context, R. philippinarum attains control of the system. Considering all this, it is possible to state that the Venice Lagoon ecosystem has entered into a new state, probably more resistant but less resilient, with implications for future management choices.  相似文献

Objective: The objective of the present study was to examine selected parameters of the blood redox system in elderly patients with hypertension.examine selected parameters of the blood redox system in elderly patients with hypertension. Methods: We analyzed differences in redox-associated molecules and enzymes among elderly hypertensive subjects (age above 65?years, n?=?49) and two groups of normotensive subjects (<65 years old – Control group I; n?=?27, and >65 – Control group II; n?=?30). Results: Decreased activity of antioxidant enzymes, increased lipid peroxidation and reduced production of nitric oxide were observed in hypertensive subjects, compared to healthy younger controls, or those of the same age. In healthy controls, an age-related decrease in the production of nitric oxide and the activities of SOD-1 and GPx-1 was also evident. The pathology of hypertension was characterised by further, significant decreases in the values of these parameters. When the subgroups of females and males were compared to their respective controls, a compromised redox balance was observed that was more evident in female hypertensives. Discussion: Hypertension in elderly patients is accompanied by changes in biomarkers of antioxidant status and lipid peroxidation status, which significantly differ from those observed in healthy ageing subjects. Our study also suggests that the relationship of gender and changes in redox balance with regard to hypertension should be further explored.  相似文献

The effect of 17β‐oestradiol (E2) on the growth of the thymus and its regionalization into cortex and medulla was investigated in juvenile European sea bass Dicentrarchus labrax as they find themselves close to sources of oestrogenic pollution whilst residing in their estuarine nursery areas. While the exposure to 2, 20 and 200 ng l?1 in 60 days post‐hatch (dph) fish tended to cause a non‐monotonous dose–response curve with a significant difference of the cortex size between lowest and highest exposures, the exposure to 20 ng l?1 E2 from 90 dph onwards resulted in a distinct enlargement of the cortex. It is probable that the alteration of the cortex size also affects the T‐cell differentiation and proliferation.  相似文献

Our awareness of the causes of stress‐induced developmental dysfunction has increased dramatically over the past decade, and it is becoming increasingly clear that a number of factors can have considerable impacts on the developing fetus. Although there is a tendency in investigations of developmental teratogens to attribute specific causes to adverse fetal outcomes, it is important we recognize that for most developmental dysfunctions it is unlikely a single cause, but yet a series of environmental insults combined with genetic predisposition that ultimately leads to a disease state. Nonetheless, a number of developmental teratogens, such as maternal psychological stress and chemical exposures, have been shown to increase the likelihood of developmental defects. These defects can manifest during development, leading to observable birth defects, or could become evident long after birth, even into adulthood. In addition, epigenetic mutations in the germline can alter the phenotype of successive generations through transgenerational inheritance, and in this way environmental factors can alter the developmental outcomes and disease predispositions of future generations. Understanding this complexity is essential to interpretations of causality in the studies of stress‐induced developmental dysfunction and needs to be fully considered to more effectively interpret potential outcomes. The sub-chronic (28–56 days) effects of exposure to low concentrations of cadmium (Cd; 0·05, 0·25, 0·50 and 2·50 μg l−1) shortly following fertilization on embryos, larvae and juvenile rainbow trout Oncorhynchus mykiss were examined. Premature hatching occurred at lower concentrations (0·05 and 0·25 μg l−1 Cd), however, delayed hatching was seen in the 2·50 μg l−1 Cd group, with >90% of hatching occurring on the last day of the hatching period. Larval growth was negatively affected by Cd exposure in a concentration-dependent manner. Larvae exposed to 2·50 μg l−1 Cd were 13·9 ± 0·8% shorter in total length ( L T) and weighed 22·4 ± 3·5% (mean ± s . e .) less than controls at the end of the exposure period. Plasma sex steroid concentrations (oestradiol in juvenile females and 11-ketotestosterone in juvenile males) were elevated (four- to 10-fold over controls) in exposed fish in both males and females, following 28 days of exposure to 0·05, 0·25 and 0·50 μg l−1 Cd, respectively. These results suggest that environmentally realistic concentrations (in the μg l−1 range) of Cd can affect the development of O. mykiss impacting embryos, larvae and juvenile fish.  相似文献

The influence of altered levels of endogenous catecholamines following adrenalectomy or 6-hydroxydopamine (6-OH) treatment (alone or in combination) on enzymatic (glutathione reductase, catalase, glutathione peroxidase and Cu,Zn superoxide dismutase) and non-enzymatic (glutathione) antioxidant components of heart, liver, kidney, lung and erythrocytes in male Wistar rats was investigated. Functional antioxidant status was assessed in terms of susceptibility to t-butylhydroperoxide-induced sulfhydryl group oxidation (an indirect measure of glutathione depletion) and lipid peroxidation, as measured by thiobarbituric acid-reactive substance (TBARS) formation. Reduced levels of adrenaline and noradrenaline resulted from adrenalectomy and 6-OH treatment, respectively, while a combination of these treatments led to a reduction in the levels of both catecholamines. Adrenalectomy was associated with alterations in glutathione reductase activity in the heart and liver (increased). 6-OH treatment alone produced an elevation in glutathione reductase activity only in the heart. In adrenalectomized animals, 6-OH treatment produced no further increases in glutathione reductase activities of heart or liver. In lung, however, the combination of adrenalectomy and 6-OH treatment caused an elevation in both glutathione peroxidase and glutathione reductase activities. Glutathione levels of liver alone were elevated following adrenalectomy, while those of erythrocytes and liver (but not other tissues investigated) were increased by the combination of adrenalectomy and 6-OH treatment. The kidney was relatively resistant to the effects of sympathectomy and showed no changes in any of the antioxidant components measured. Adrenalectomy alone or in combination with 6-OH produced an increase in susceptibility to peroxide-induced sulfhydryl group oxidation only in the heart. 6-OH treatment caused a reduction in peroxide-induced TBARS formation only in the kidney. Both adrenalectomy and the combination of adrenalectomy and 6-OH treatment were associated with reduced TBARS formation in the liver, lung and kidney, but not heart. Results from this study demonstrate that the effects of sympathectomy on antioxidant status vary among tissues. Differences between adrenalectomy and 6-OH treatment on antioxidant components are suggestive of differential actions of adrenaline and noradrenaline on tissue antioxidant status which may have important implications under conditions associated with elevations in levels of these catecholamines including chronic stress and myocardial infarction.  相似文献

Hyperglycemia induces overproduction of superoxide and it is related to diabetic complications. In this study, we analyzed the antioxidant enzymatic defense and the lipid peroxidation of rat salivary glands in six different periods of diabetic condition. Ninety‐six rats were divided into 12 groups: C7/14/21/28/45/60 (non‐diabetic animals) and D7/14/21/28/45/60 (diabetic animals). Diabetes was induced by streptozotocin and the rats were euthanized after 7, 14, 21, 28, 45, or 60 days. Their parotid (PA) and submandibular (SM) glands were removed soon after the sacrifice and the total protein and malondialdehyde (MDA) concentrations, as well as, the superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT) activities were determined. Twenty‐one days after the diabetes induction, the SM glands showed an increase in SOD, CAT, and GPx activities, as well as, MDA concentration. Concerning the PA glands, an increase in the CAT activity and MDA content was observed throughout the observation period. The results suggest that diabetes can cause alterations on the salivary glands and that PA and SM glands react differently when exposed to diabetes condition. However, no impairment of antioxidant system was observed in the group whose diabetic condition had been induced 60 days earlier, herein named 60‐day group.
