The clam Ruditapes decussatus is distributed worldwide and due to its ecological and economical interest has been proposed as a bioindicator in areas where mussels are not available. The accumulation of several anthropogenic compounds in their tissues suggests that they possess mechanisms that allow them to cope with the toxic effects of these contaminants. Besides pollutant uptake, the use of biomarkers is pointed out in this paper since it is a promising approach to monitor the effect of these contaminants in the marine environment. Biomarkers complement the information of the direct chemical characterization of different types of contaminants. Therefore, the aim of this paper is to review the role of several biomarkers: (metallothioneins (MT), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidases (GPx) (total and selenium-dependent), lipid peroxidation (measured as MDA, one of the final products of lipid peroxidation), glutathione S-transferase (GST) and acetylcholinesterase (AChE), measured in different tissues of the clam R. decussatus, in laboratory conditions and under various environmental stresses, in two ecosystems (Ria Formosa lagoon- Portugal) and Bizerta lagoon (Tunisia) in a perspective of a multibiomarker approach to assess environmental changes. Experiment and field studies are in good agreement since MT levels, especially in the gills, the first target tissue of these contaminants, can be used as biomarker of exposure to Cd. GPx and MDA may also be determined in this respect. AChE activity is inhibited by pesticide and, to a less extent, by metal exposure in the gills and whole soft body of clams. However, the induction of GST isoforms experimentally demonstrated is not observed in the field because only global GST activity was determined. The whole set of results opens new research perspectives for the use of this species to assess the effect of mixtures of pollutants in the aquatic environment.  相似文献

Aims: To describe uptake and retention of pathogenic and nonpathogenic Vibrio parahaemolyticus in Ruditapes decussatus and Ruditapes philippinarum. Methods and Results: Ruditapes decussatus accumulated greater concentrations of pathogenic and nonpathogenic V. parahaemolyticus than R. philippinarum. These concentrations decreased earlier in R. decussatus. Nonpathogenic V. parahaemolyticus reached higher concentrations (approx. 1 log CFU g?1 in tissues of R. decussatus and more than 1 log CFU g?1 in R. philippinarum) than pathogenic V. parahaemolyticus at similar times. It also persisted longer in both species of clams (72 h in R. decussatus and 96 h in R. philippinarum), while pathogenic V. parahaemolyticus persisted 48 h in R. decussatus and 72 h in R. philippinarum. Conclusions: Ruditapes decussatus incorporated both isolates of V. parahaemolyticus faster than R. philippinarum and it eliminated both isolates earlier than R. philippinarum. Under same conditions, nonpathogenic V. parahaemolyticus might survive better than pathogenic V. parahaemolyticus within both species of clam. Significance and Impact of the Study: Pathogenic V. parahaemolyticus is an important cause of foodborne illnesses. This study shows it may be possible to use nonpathogenic Vparahaemolyticus to perform experimentation to evaluate bacterial evolution in clams, developing an in vivo model useful for risk analysis.  相似文献

This paper aims to assess the marine sediment quality along the Tunisian coast using clam Ruditapes decussatus embryos and larval bioassays tests. Elutriate samples prepared with sediment from four sites (Monastir Lagoon, Chebba, Mahres, and Zarat) were used for chemical analysis and clam bioassays tests (embryogenesis, larval growth. and metamorphosis). For embryogenesis test, five elutriates concentrations (12%, 25%, 50%, 75%, and 100%) were prepared. Metal concentrations were highest at the Mahres and Monastir Lagoon sites. Clam embryogenesis success was significantly (p < 0.05) reduced in all elutriates preparations at concentrations up to 50% except Chebba, where inhibition was evident at only the highest concentration (100%). Compared to the control (9.4 μm day?1), larval growth rate was significantly (p < 0.05) reduced only in two sites, Monastir Lagoon (6.8 μm day?1) and Zarat (5.6 μm day?1). However, larval survival was lower (42.9%) in elutriates prepared from Mahres sediment compared to other studied sites and control treatment. We did not note any significant (p > 0.05) effect of elutriate preparations on clam metamorphosis; conversely, survival in this stage was significantly affected in three sites (Monastir: 59.0 ± 6.7%; Zarat: 52.1 ± 6.3%, and Mahres: 64.1 ± 5.4%) compared to the control (83.2 ± 4.7%). This work shows that both clam embryos and larvae are sensitive to contaminants and can be used to evaluate sediment contamination and monitor pollution.  相似文献

Objective: The objective of the present study was to examine selected parameters of the blood redox system in elderly patients with hypertension.examine selected parameters of the blood redox system in elderly patients with hypertension. Methods: We analyzed differences in redox-associated molecules and enzymes among elderly hypertensive subjects (age above 65?years, n?=?49) and two groups of normotensive subjects (<65 years old – Control group I; n?=?27, and >65 – Control group II; n?=?30). Results: Decreased activity of antioxidant enzymes, increased lipid peroxidation and reduced production of nitric oxide were observed in hypertensive subjects, compared to healthy younger controls, or those of the same age. In healthy controls, an age-related decrease in the production of nitric oxide and the activities of SOD-1 and GPx-1 was also evident. The pathology of hypertension was characterised by further, significant decreases in the values of these parameters. When the subgroups of females and males were compared to their respective controls, a compromised redox balance was observed that was more evident in female hypertensives. Discussion: Hypertension in elderly patients is accompanied by changes in biomarkers of antioxidant status and lipid peroxidation status, which significantly differ from those observed in healthy ageing subjects. Our study also suggests that the relationship of gender and changes in redox balance with regard to hypertension should be further explored.  相似文献

Progress in ecotoxicological research documents that crustaceans are highly vulnerable to diverse chemicals and toxicants in the environment. In particular, pollutants affecting endocrine homeostasis in crustaceans (i.e., endocrine disruptors) are intensively studied, and serious reproductive disorders have been documented. In this review, current knowledge about the endocrine regulation of the crustacean reproduction is put together with the published ecotoxicological data with an attempt to summarize the potential of xenobiotics to affect crustacean reproduction. Following gaps and trends were identified: (1) Studies are required in the field of neurohormone (serotonin and dopamine) regulation of the reproduction and possible modulations by environmental toxicants such as antidepressant drugs. (2) Molting-related parameters (regulated by ecdysteroid hormones) are closely coordinated with the development and reproduction cycles in crustaceans (cross-links with methyl farnesoate signalling), and their susceptibility to toxicants should be studied. (3) Other biochemical targets for xenobiotics were recently discovered in crustaceans and these should be explored by further ecotoxicological studies (e.g., new information about ecdysteroid receptor molecular biology). (4) Some sex steroid hormones known from vertebrates (testosterone, progesterone) have been reported in crustaceans but knowledge about their targets (crustacean steroid receptors) and signalling is still limited. (5) Determination of the sex in developing juveniles (affecting the sex ratio in population) is a sensitive parameter to various xenobiotics (including endocrine disruptors) but its modulation by general environmental stress and non-specific toxicity should be further studied.  相似文献

Here we explore the dose-dependent response of the tropical fish tambaqui (Colossoma macropomum) to intraperitoneal injection of benzo[a]pyrene (BaP) at doses of 0 (carrier control), 1, 10, 100 and 1000 μmolar BaP Kg−1 Hepatic ethoxyresorufin-O-deethylase (EROD) activity showed a bell-shaped dose-dependent response curve, where the highest injected BaP dose caused enzyme inactivation. Activities of hepatic catalase (CAT) and superoxide dismutase (SOD) increased at the highest dose relative to the carrier control group. Lipid peroxidation (LPO), serum-sorbitol dehydrogenase (s-SDH) and DNA damage in blood cells were higher for all BaP doses when compared to the carrier control group. At high dosage, the production of BaP metabolites was paralleled by induced activity of the antioxidant enzyme SOD, and high levels of DNA damage in blood cells. In a similar way, high LPO was concomitant to elevated s-SDH in the bloodstream, suggesting that lipid peroxidation caused the loss of membrane integrity and leakage of s-SDH from hepatocytes into the bloodstream. These biomarkers were also positively co-correlated. The results demonstrate the potential use of a suite of biomarkers for tambaqui living in contaminated tropical aquatic environments. In particular, we recommend the analysis of DNA damage in blood cells, as this was highly correlated with all other biomarkers.  相似文献

Among the 19 non-native species of marine invertebrates which have invaded the Venice Lagoon and have established populations, Ruditapes philippinarum, deliberately introduced in 1983, is surely the most successful species. According to the hypothesis that alien species invasion could be favoured by an altered ecological, chemical or physical state of the system induced by anthropogenic disturbance, R. philippinarum turned out to be ‘the right species at the right moment’. By comparing historical data (1968, 1985, 1990) with 1999 data, changes in macrobenthic community, in particular bivalve molluscs, of the lagoon induced by R. philippinarum introduction and subsequent clam exploiting activity were assessed. It has been possible to describe a sharp reduction, both in terms of distribution area and density, of all other filter feeder bivalves. Moreover, by using the clearance rate of the most abundant bivalve species in 1990 and 1999 (Cerastoderma glaucum and R. philippinarum, respectively), it was possible to estimate that the filtration capacity, expressed as l h−1 m−2, has more than doubled. This has altered the functioning of the ecosystem, resulting in a stronger benthic–pelagic coupling. In this context, R. philippinarum attains control of the system. Considering all this, it is possible to state that the Venice Lagoon ecosystem has entered into a new state, probably more resistant but less resilient, with implications for future management choices.  相似文献

Hyperglycemia induces overproduction of superoxide and it is related to diabetic complications. In this study, we analyzed the antioxidant enzymatic defense and the lipid peroxidation of rat salivary glands in six different periods of diabetic condition. Ninety‐six rats were divided into 12 groups: C7/14/21/28/45/60 (non‐diabetic animals) and D7/14/21/28/45/60 (diabetic animals). Diabetes was induced by streptozotocin and the rats were euthanized after 7, 14, 21, 28, 45, or 60 days. Their parotid (PA) and submandibular (SM) glands were removed soon after the sacrifice and the total protein and malondialdehyde (MDA) concentrations, as well as, the superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT) activities were determined. Twenty‐one days after the diabetes induction, the SM glands showed an increase in SOD, CAT, and GPx activities, as well as, MDA concentration. Concerning the PA glands, an increase in the CAT activity and MDA content was observed throughout the observation period. The results suggest that diabetes can cause alterations on the salivary glands and that PA and SM glands react differently when exposed to diabetes condition. However, no impairment of antioxidant system was observed in the group whose diabetic condition had been induced 60 days earlier, herein named 60‐day group. Juvenile roach Rutilus rutilus from seven rivers of varying water quality were examined for evidence of endocrine disruption. The majority of roach from five of these rivers had female‐like reproductive ducts. The ability of aluminium to affect the oxidant status of specific areas of the brain (cerebellum, ventral midbrain, cortex, hippocampus, striatum) was investigated in rats intraperitoneally treated with aluminium chloride (10 mg Al3+/kg/day) for 10 days. The potential of aluminium to act as an etiological factor in Parkinson's disease (PD) was assessed by studying its ability to increase oxidative stress in ventral midbrain and striatum and the striatal dopaminergic neurodegeneration induced by 6-hydroxydopamine in an experimental model of PD. The results showed that aluminium caused an increase in oxidative stress (TBARS, protein carbonyl content, and protein thiol content) for most of the brain regions studied, which was accompanied by a decrease in the activity of some antioxidant enzymes (superoxide dismutase, catalase, glutathione peroxidase). However, studies in vitro confirmed the inability of aluminium to affect the activity of those enzymes. The reported effects exhibited a regional-selective behaviour for all the cerebral structures studied. Aluminium also enhanced the ability of 6-hydroxydopamine to cause oxidative stress and neurodegeneration in the dopaminergic system, which confirms its potential as a risk factor in the development of PD.  相似文献

The antioxidant activity of some compounds buffer the free radicals generated either endogenously or exogenously, thus decreasing the potential damage mediated by oxidation. Recent studies documented that raloxifene has antioxidant properties in vitro. However, there are limited animal studies available to show raloxifene's antioxidant properties. We aimed to investigate the effects of raloxifene on antioxidant enzymes such as SOD, CAT and GPX, TrxR and the levels of GSH and MDA in heart, liver and brain cortex of ovariectomized female rats. Female Sprague Dawley rats weighing 300-350 g (n=24) were divided into three groups: (I) Eight non-ovariectomized rats were used as naive controls without any treatment (non-ovariectomized group, n=8). Five weeks after ovariectomy, (II) Ovariectomized placebo group (n=8) was given physiological saline, and (III) Raloxifene group (n=8) was given raloxifene 1 mg/kg sc. daily for 12 days. Ovariectomy induced significant increases on SOD, GPX, CAT activity and MDA levels in brain, heart and liver tissues compared to non-ovariectomized rats ( p<0.05). Raloxifene treatment led to decreased levels of SOD activity in heart, GPX activity in brain and CAT activity in liver tissue when compared to ovariectomized group ( p<0.05) but there was no change in activity of TrxR in all groups. The levels of MDA in brain, heart and liver tissues increased in ovariectomized group when compared to non-ovariectomized rats ( p<0.05). Raloxifene had a significant attenuating effect on the levels of MDA in brain and heart tissues. Our results also indicate that the levels of GSH in brain, heart and liver tissue decreased when compared to non-ovariectomized rats. Raloxifene treatment was observed to significantly increase the levels of GSH in brain and heart tissues ( p<0.05). However, there were insignificant differences for the GSH levels in liver tissues of ovariectomized placebo or raloxifene groups. In conclusion, our results demonstrate that raloxifene may be more effective against oxidative stress in heart and brain than in liver tissue.  相似文献

This study aimed at investigating the extent of accumulation of selenium in the hepatopancreas of the crayfish Procambarus clarkii and its effects on the enzymatic activities of catalase, glutathione peroxidase, and glutathione reductase. Crayfish were fed for 15 days either a diet enriched in Se, ED (1.21 mg kg-1) or a standard diet, SD (0.30 mg kg-1). Results showed that Se accumulation is higher in the sample subject to ED rather than to SD. A sexual difference was found for the response of the analyzed enzymes in the ED sample only. Catalase activity and glutathione peroxidase activity decreased in females and males, respectively, whereas glutathione reductase activity showed significant reductions in both sexes. These preliminary data might provide information about prooxidant effects of Se on P. clarkii when fed a Se enriched diet.  相似文献

The present study was designed to study the seasonal variations of selenium in marine biota of different trophic levels collected from Sunderban deltaic complex, northeast India. The primary objective of this work is to provide baseline data for a future environmental quality programme. In general, the sequence of Se accumulation observed in biota is as follows: Bivalve>Zooplankton>Macroalgae>Pisces>Seagrass. An elevated level of Se was recorded during monsoon season. The Se level in zooplankton (4 μg/g) was three to four times higher than seagrass. The content of various soft tissues and shell in bivalves molluscs showed an organ-specific pattern in the following order: gill > visceral MASS = mantle > adductor muscle > podium > shell. Fish contain less Se in comparison with macroalgae, zooplankton and bivalves revealing poor or insignificant trophic transfer of Se in the marine food chain. Pelecyora trigona (Bivalve) seems to be reliable indicator of Se contamination because of its high accumulation capacity. An in-depth monitoring program is recommended in order to clarify the present trend and to establish the studied biota as indicator species.  相似文献

Soil enzyme activities have been long used as indicators of soil contamination, and their integration into numerical indexes of microbial functional diversity is a practical approach in the environmental risk assessment of soil pollutants. However, suitable numerical indexes need to be developed and standardized for monitoring deterioration of soil quality by agrochemicals. Herein, a mesocosm study was performed to examine short-term responses of selected soil enzyme activities to chlorpyrifos (Lorsban® 4E). Hydrolases (carboxylesterase, acid phosphatase, β-glucosidase, urease and protease) and oxidoreductases (dehydrogenase and catalase) were measured in Andisols 14 d after an application with two doses (4.8 and 24 kg a.i. ha−1) of chlorpyrifos. Both application rates caused a strong inhibition of carboxylesterase (62–78% of controls), acid phosphatase (56–60%) and β-glucosidase (43–58%) activities. Soil microbial activity was also reduced in pesticide-sprayed soils as indicated by the decreased dehydrogenase (47%) and catalase (38%) activities compared with control soils. However, only carboxylesterase activity showed a dose-dependent response with the chlorpyrifos application rate. An in vitro trial was further performed to provide evidence of a direct interaction between the enzyme (carboxylesterase, acid phosphatase and β-glucosidase) and the pesticide (chlorpyrifos and its main metabolites chlorpyrifos-oxon and 3,5,6-trichloro-2-pyridinol). Results of these in vitro assays showed that the activity of carboxylesterase was directly affected by chlorpyrifos-oxon and, at less extend, by chlorpyrifos, whereas variations of both acid phosphatase and β-glucosidase activities were likely dependent on changes in microbial activity. Urease and protease activities did not change in pesticide-treated soils compared with pesticide-free soils. Despite the absence of response in these two N-cycling enzyme activities, four enzymatic indexes (geometric mean, weighted mean, “treated-soil quality index” [T-SQI] and “integrated biological response” [IBRv2] index) were significantly lower in the chlorpyrifos-sprayed soils compared with controls. Moreover, there was a significant (r2 = 0.87, P < 0.0001) correlation between T-SQI and IBRv2 scores, which suggested that the IBRv2 index (an index used for assessing animal’s health inhabiting contaminated sites) may be a complementary index in soil quality assessment.  相似文献

The bacterial genus Exiguobacterium accommodates many versatile species isolated from diverse environments. Exiguobacterium was described as a genus approximately three decades ago, and now, 17 species, growing over a broad range of temperatures and pH, have been recognized. Various isolates from different niches have been explored for biotechnological and industrial purposes, including enzyme production, bioremediation and degradation of toxic substances released into the environment. Some isolates possess plant growth promoting capabilities, and they are currently being explored for increasing agricultural production. The genome sequences of various strains of this genus have shown the presence of many genes encoding products of importance to agriculture and the environment. In addition, many strains possess stress-responsive genes helping them to colonize and thrive in diverse ecological niches. This review provides a broad view of the versatile genus Exiguobacterium and its potential for applications in agriculture, the environment and industry, as well as the underlying genomic determinants that drive its diversity and adaptability to various extreme environments.  相似文献

Sediment-dwelling prokaryotes play a vital role in determining the fate and speciation of metals, yet are also susceptible to the biological effects of trace metals. In this article, optimized DNA extraction and purification techniques and species-specific primers are used to assess the genetic incidence and abundance of metal detoxification and general stress genes of Pseudomonas aeruginosa to complement chemical analysis in inferring the severity of metal-contaminated sites along the Clark Fork River, Montana. Results show the highest incidence of candidate genes related to bacterial stress at the most polluted site, while multiple regression analysis demonstrated significant correlations (P<0.05, r(2)=0.9) between in situ metal concentrations (As, Cu and Zn), total gene incidence, and the incidence of metal detoxification genes. Furthermore, principal components plotting the incidence of genes related to metal resistance show clear separation of sites giving clear clusters on the basis of contamination. Quantification of three genes (sodA, htpX and mt) from surveyed sites found significantly higher (anova, P<0.05) copy numbers at the more contaminated sites compared with reference sites. The development of rapid microbial biomarker tools represents a significant advance in the field of environmental biomonitoring and the prediction of metal bioavailability.  相似文献

Since the radiation dose tolerance of normal tissues/organs away from the site of tumor influences the success of radiation therapy of cancer, and antioxidant status is likely to be one of the factors to determine the tolerance; the radioresponse of antioxidant enzymes has been examined in the liver as a representative distant organ in the tumorbearing mice.Swiss albino male mice (7–8 weeks old) with Ehrlich solid tumor in the thigh pad were irradiated with different doses of radiation (0–9 Gy) at a dose rate of 0.0153 Gy/s and the specific activities of enzymes involved in the free radical metabolism were determined in the liver. Except GST, the activities of SOD, DTD and Gly I as well as the GSH content were found to be higher in the liver of tumorbearing mice compared to the nontumor bearing mice. The catalase activity progressively decreased with dose in both the groups of mice. However, the activity was relatively higher in the liver of tumor bearing mice than the control. Thus, the radioresponse of antioxidant enzymes seemed to be significantly different in the liver of tumorburdened mice compared to controls. The enhanced activities might be due to relatively more damage caused by radiation. The higher levels of NO· and peroxidative damage in the liver of tumorbearing mice probably suggest this possibility. These findings of the present work might have some serious implications as the increased radiationdamage of the distant normal organs (due to tumor burden) is likely to adversely affect the therapeutic gain.  相似文献

During 2006 to 2008, the water, sediment and fish in Albasini Dam, used as a reference site outside a DDT-sprayed area, had been found to be as affected as those in the nearby DDT-sprayed area were. Accordingly, the aim of the current study, conducted in 2014, was to determine the health status of fish from Albasini Dam by means of a histology-based fish health assessment protocol. High levels of nitrates, as well as the organochlorine pesticides aldrin, dieldrin, chlordane and methoxychlor, were found in the water. Fish muscle tissue contained pesticides, including methoxychlor. A semi-quantitative histological assessment on Clarias gariepinus, Coptodon rendalli and Oreochromis mossambicus showed diverse histopathology in selected target organs, with the gills being most affected. Notably, the testis tissue of one C. rendalli was filled with eosinophilic liquid, characteristic of necrosis. Microscopic intersex was found in two of the nine O. mossambicus sampled. This is the second time that intersex as microscopic testicular oocytes has been reported in O. mossambicus in South Africa. This finding perhaps indicates that reproduction may be compromised and this will be investigated further. The presence of intersex and other histopathology in organs could be early warning signs of the declining water quality in this dam.  相似文献
