A variety of organic compounds have been documented to bind to the oestrogen receptor and induce oestrogenic effects in different vertebrates. The presence of these environmental oestrogens or oestrogen mimics in the aquatic environment has been suspected of disrupting the normal endocrinology of wild populations of fish. In this study, induction of vitellogenin synthesis in primary hepatocytes from Atlantic salmon (Salmo salar) was optimized and validated as an oestrogenic in vitro bioassay using a sensitive capture vitellogenin enzyme-linked immunosorbent assay. After proper optimization (cell media supplements, cell density, temperature and exposure time), this assay gave a sensitive and reproducible response to both endogenous steroids (relative potency: 17beta-oestradiol>oestriol>oestrone>17alpha-oestradiol) and a range of common oestrogen mimics (relative potency: ethynyloestradiol and diethylstilboestrol>genistein and zearalenone>bisphenol A and 4-t-octylphenol>4-n-nonylphenol and 2'-chloro,4-chloro-diphenyltrichloroethane (o,p'-DDT). However, the androgen testosterone and the putative oestrogen mimics dieldrin and toxaphene were not shown to be oestrogenic using this hepatocyte bioassay. Oestrogen-induced vitellogenin synthesis was efficiently inhibited by the anti-oestrogen ZM 189.154, suggesting that this bioassay may be used for testing both the oestrogenic and the anti-oestrogenic properties of chemicals.  相似文献

Background

Oestrogenic contaminants are widespread in the aquatic environment and have been shown to induce adverse effects in both wildlife (most notably in fish) and humans, raising international concern. Available detecting and testing systems are limited in their capacity to elucidate oestrogen signalling pathways and physiological impacts. Here we developed a transient expression assay to investigate the effects of oestrogenic chemicals in fish early life stages and to identify target organs for oestrogenic effects. To enhance the response sensitivity to oestrogen, we adopted the use of multiple tandem oestrogen responsive elements (EREc38) in a Tol2 transposon mediated Gal4ff-UAS system. The plasmid constructed (pTol2_ERE-TATA-Gal4ff), contains three copies of oestrogen response elements (3ERE) that on exposure to oestrogen induces expression of Gal4ff which this in turn binds Gal4-responsive Upstream Activated Sequence (UAS) elements, driving the expression of a second reporter gene, EGFP (Enhanced Green Fluorescent Protein).

Results

The response of our construct to oestrogen exposure in zebrafish embryos was examined using a transient expression assay. The two plasmids were injected into 1?C2 cell staged zebrafish embryos, and the embryos were exposed to various oestrogens including the natural steroid oestrogen 17?-oestradiol (E2), the synthetic oestrogen 17??- ethinyloestradiol (EE2), and the relatively weak environmental oestrogen nonylphenol (NP), and GFP expression was examined in the subsequent embryos using fluorescent microscopy. There was no GFP expression detected in unexposed embryos, but specific and mosaic expression of GFP was detected in the liver, heart, somite muscle and some other tissue cells for exposures to steroid oestrogen treatments (EE2; 10?ng/L, E2; 100?ng/L, after 72?h exposures). For the NP exposures, GFP expression was observed at 10???g NP/L after 72?h (100???g NP/L was toxic to the fish). We also demonstrate that our construct works in medaka, another model fish test species, suggesting the transient assay is applicable for testing oestrogenic chemicals in fish generally.

Conclusion

Our results indicate that the transient expression assay system can be used as a rapid integrated testing system for environmental oestrogens and to detect the oestrogenic target sites in developing fish embryos.  相似文献

The finding that a variety of chemicals display oestrogenic activity has resulted in the development of in vitro and in vivo assays to assess oestrogenic activity. One such assay, the yeast oestrogen assay (YES) makes use of recombinant yeast cells that harbour an oestrogen receptor expression cassette and a reporter construct, coding for bgalactosidase. The induction mechanism starts with the binding of oestrogenic compounds to the oestrogen receptor. This complex activates the production of β-galactosidase. The β-galactosidase activity is thus a measure of the oestrogenic activity of chemical compounds. In the YES assay, the β-galactosidase activity may be quantified with the chromogenic substrate chlorophenol red-β-d-galactopyranoside (CPRG). In the present study it is reported that CPRG or its β-galactosidase degradation product chlorophenol red act in the YES as an oestrogenic compound itself. The implications of this finding are described. It is especially argued that chlorophenol red production after prolonged incubation of the assay might be misinterpreted as an oestrogenic effect of the test compound.  相似文献

Abstract

A method to quantify induction of vitellogenin (Vtg) mRNA in adult male mosquitofish was developed. Male mosquitofish were exposed to 0, 1, 20 and 250 ng l?1 17β-oestradiol (E2) for 4 and 8 days in static exposures, and liver Vtg mRNA and 18S rRNA expression were quantified in duplex RT-PCR. Liver 18S rRNA expression was very consistent among individuals, and there was a highly significant increase in Vtg mRNA expression after exposure of mosquitofish for just 4 days at 250 ng l?1 E2. Lower doses did not induce Vtg mRNA expression even at 4 or 8 days. This method could be used as a rapid test to detect exposure of mosquitofish to oestrogenic chemicals. Further work is needed to determine if increased Vtg mRNA levels in male mosquitofish induce Vtg synthesis, and to determine the usefulness of the method in field sampling.  相似文献

Summary In the newt, Pleurodeles waltli, vitellogenin synthesis can be induced by oestrogen prior to metamorphosis and well before vitellogenin synthesis is normally observed in females. Vitellogenin indicubility has been shown to occur during normal development at stage 53 in both sexes. Immersion of young larvae in thyroxine at concentrations that did not result in early morphological metamorphosis led to a precocious acquisition of liver responsiveness to oestrogen. Treatment of larvae with thiourea greatly diminished the ability of oestradiol to induce vitellogenin synthesis, although it did not abolish the hormonal response.  相似文献

Adult female crabs (Neohelice granulata) were treated during the post-reproductive period with both 17-hydroxyprogesterone (17PG) and methyl farnesoate (MF). During the 4-week in vivo assay, animals were fed hormone-enriched (17PG or MF) pellets twice a week, at a dose of 2 nmol/g of body weight. Additionally, at the middle of the in vivo experiment, ovarian explants were incubated in vitro with each hormone at a concentration of 15 μM for 24 h. At the end of the in vivo assay, both the gonadosomatic index and the vitellogenic protein content (Vg) of crabs treated with either 17PG or MF were significantly higher (p < 0.05) than the values of the concurrent controls. In addition, the hemolymphatic vitellogenin level of both hormonally treated groups was higher. At the end of the in vitro assay, ovary explants incubated with 17PG had a significantly higher (p < 0.05) protein synthesis and content of Vg proteins than controls. Ovary explants incubated with MF had similar levels to controls (p > 0.05). The involvement of these hormones in the regulation of ovarian growth during the reproductive cycle of N. granulata is discussed.  相似文献

Steroidal oestrogens are often accumulated in urban estuarine sediments worldwide at microgram per gram levels. These aromatic steroids have been classified as endocrine disruptors and group 1 carcinogens. Microbial degradation is a naturally occurring mechanism that mineralizes oestrogens in the biosphere; however, the corresponding genes in oestrogen-degrading actinobacteria remain unidentified. In this study, we identified a gene cluster encoding several putative oestrogen-degrading genes (aed; actinobacterial oestrogen degradation) in actinobacterium Rhodococcus sp. strain B50. Among them, the aedA and aedB genes involved in oestrogenic A-ring cleavage were identified through gene-disruption experiments. We demonstrated that actinobacterial oestrone 4-hydroxylase (AedA) is a cytochrome P450-type monooxygenase. We also detected the accumulation of two extracellular oestrogenic metabolites, including pyridinestrone acid (PEA) and 3aα-H-4α(3'-propanoate)-7aβ-methylhexahydro-1,5-indanedione (HIP), in the oestrone-fed strain B50 cultures. Since actinobacterial aedB and proteobacterial edcB shared < 40% sequence identity, 4-hydroxyestrone 4,5-dioxygenase genes (namely aedB and edcB) could serve as a specific biomarker to differentiate the contribution of actinobacteria and proteobacteria in environmental oestrogen degradation. Therefore, 4-hydroxyestrone 4,5-dioxygenase genes and the extracellular metabolites PEA and HIP were used as biomarkers to investigate oestrogen biodegradation in an urban estuarine sediment. Interestingly, our data suggested that actinobacteria are active oestrogen degraders in the urban estuarine sediment.  相似文献

A series of 7-O-alkoxy-4-methylumbelliferone derivatives were prepared using a convenient one step synthesis. Additionally the bromo- and azido derivatives 7-O-(4-bromobutoxy)-, 7-O-(6-bromohexyloxy)- and 7-O-(6-azidohexyloxy)-4-methylumbelliferone derivatives were prepared. In vitro evaluation of antimycobacterial activity determined % inhibition and MIC vs M. tuberculosis H37Rv with toxicity (IC50) assessed in VERO cells. The coumarins with longer alkyl chains (nonyl and decyl) showed the optimum inhibitory activity in this series (MIC 3.13?μg/mL) and IC50>10?μg/mL.  相似文献

Vitellogenin levels were determined in pieces of either ovary or hepatopancreas taken from females of the crayfish Cherax quadricarinatus during both early pre-reproductive and reproductive periods, and exposed in vitro to 0.15, 1.5, or 15?µM of the juvenoid methyl farnesoate (MF). A significant accumulation of vitellogenin was seen in ovaries treated with 15?µM of MF during the early pre-reproductive period. Besides, protein synthesis, measured as tritiated leucine incorporation, was correspondingly increased in ovaries taken during both pre- and post-reproductive periods and exposed to 15?µM of MF. On the other hand, no stimulating effect of MF on the vitellogenin content of the hepatopancreas was seen in any period. These results, taken together, support the hypothesis of the endogenous vitellogenin synthesis stimulated by MF during the early ovarian maturation. However, this hormone was not able to stimulate vitellogenin synthesis in the ovary during the reproductive period.  相似文献

Polychlorinated biphenyls (PCBs) are widespread, persistent environmental contaminants of which some congeners can act as endocrine disrupters. Previous work has shown that 3,4,3′,4′-tetrachlorobiphenyl (PCB77) can act as an oestrogen with actions mediated through the oestrogen receptor. Here, oestrogenic actions have been assessed for two further tetrachlorobiphenyl isomers. Assays of oestrogenic action have involved (1) ligand regulation of oestrogen-sensitive gene expression; (2) ligand regulation of cell growth in oestrogen-dependent human breast cancer cell lines MCF7 McGrath and ZR-75-1; and (3) ligand activity in the immature mouse uterine weight bioassay in vivo. These results demonstrate that 3,5,3′,5′-tetrachlorobiphenyl (PCB 80) can be considered to be a weak oestrogen agonist, but the 2,5,2′,5′-congener (PCB 52) revealed no oestrogenic properties in any of these assays. Implications of these results are discussed in relation to structure-activity predictions for environmental oestogens.  相似文献

Four fatty acids (FA, palmitic, myristic, decanoic, hexanoic) were individually conjugated to the N-terminus of the α-MSH fragment analog, H-Asp5-His6-D-Phe7-Arg8-Trp9-Lys10-NH2. This resulted in enhanced potency of the conjugates (compared to the unconjugated melanotropin analog) as determined in the lizard skin bioassay and in the mouse melanoma cell tyrosinase bioassay. The shorter conjugates of hexanoic and decanoic acid were at least equipotent to α-MSH in the lizard skin bioassay, whereas the longer myristoyl and palmitoyl analogs were 100 times less active. The myristoyl and palmitoyl conjugates exhibited a “creeping” potency in the lizard skin bioassay—that is, potency of the peptides increased with time in contact with the skins. These observations may be related to the more lipid nature of these FA-conjugates. In the tyrosinase assay, the conjugates were 10–100 times more active than α-MSH or the unconjugated analog. Each of the FA-melanotropic peptide conjugates exhibited prolonged (residual) melanotropic activity in both the lizard skin and melanoma cell bioassays. In other words, after removal of the melanotropin conjugates from contact with the skins or cells, responses were still manifested for hours or days thereafter. As little as 1 hr of contact with melanoma cells resulted in enhanced enzyme activity as measured 48 hr later. Since the conjugates, but not H-[Ast5,D-Phe7,Lys10]α-MSH5-10-NH2, exhibited prolonged activity, the conversion of reversible agonists to irreversible agonists was demonstrated.  相似文献

The occurrence of vitellogenin in adult haploid drones of the honeybee, Apis mellifica, was determined by sensitive immunotechniques, i.e. two-dimensional Immunoelectrophoresis and SDS-PAGE immunoblotting using a monospecific anti-vitellogenin-serum. In drones vitellogenin is one of the minor fractions of the hemolymph proteins. Genetic and regulatory aspects of vitellogenin synthesis in male bees are discussed.  相似文献

1. Gradient elution (sodium chloride concentration gradient) on DEAE-Sephadex columns is used to separate urinary oestrogen conjugates of pregnancy urine. Changes in the shape of the gradient alter the chromatograms in a predictable manner so that the dispersion of peaks can be modified at will. 2. Suitable choice of the gradient results in the separation from each other of oestrogen sulphates, oestrogen 16(or 17)-glucosiduronates, oestrogen 3-glucosiduronates and free oestrogens. 3. There is increasing concern about man-made chemicals in the aquatic environment that mimic oestrogens because they may disrupt reproductive function. Vitellogenin, a precursor of egg-yolk in fish and other oviparous animals, may be used as a biomarker for “oestrogen” exposure. This study investigated the use of a radioimmunoassay developed to carp (Cyprinus carpio) vitellogenin to measure vitellogenin in other species of fish, especially cyprinids that would be of value for field and laboratory studies on oestrogenic xenobiotics. Of the nine families of fish studied, only vitellogenin from cyprinids (to which the carp belongs) showed good cross-reactivity in the carp vitellogenin radioimmunoassay. Vitellogenin from cyprinids native to Europe that cross reacted in the carp vitellogenin radioimmunoassay included: bream (Abramis brama), roach (Rutilus rutilus), rudd (Scardinius erythropthalmus), gudgeon (Gobio gobio) and minnow (Phoxinus phoxinus). Vitellogenin from cyprinids used widely in ecotoxicology that cross reacted in the carp vitellogenin radioimmunoassay included: fathead minnow (Pimephales promelas), zebrafish (Brachydanio rerio) and goldfish (Carassius auratus). In the cyprinids studies, the concentrations of vitellogenin in mature females were between a few hundred and a thousand microgram per millilitre. Concentrations of plasma vitellogenin in immature females were always greater than 200 ng·m-1, whereas in males (with the exception of the fathead minnow) plasma vitellogenin concentrations were less than 20 ng·ml-1 (and generally, much lower). The results suggest that the structure of vitellogenin is highly conserved within the cyprinid family and that the carp vitellogenin radioimmunoassay may be used to measure the concentrations of vitellogenin in plasma from a wide variety of cyprinids.  相似文献

Parabens (4-hydroxybenzoic acid esters) have been recently reported to have oestrogenic activity in yeast cells and animal models. Since the human population is exposed to parabens through their widespread use as preservatives in foods, pharmaceuticals and cosmetics, we have investigated here whether oestrogenic activity of these compounds can also be detected in oestrogen-sensitive human cells. We report on the oestrogenic effects of four parabens (methylparaben, ethylparaben, n-propylparaben, n-butylparaben) in oestrogen-dependent MCF7 human breast cancer cells. Competitive inhibition of [3H]oestradiol binding to MCF7 cell oestrogen receptors could be detected at 1,000,000-fold molar excess of n-butylparaben (86%), n-propylparaben (77%), ethyl-paraben (54%) and methylparaben (21%). At concentrations of 10(-6)M and above, parabens were are able to increase expression of both transfected (ERE-CAT reporter gene) and endogenous (pS2) oestrogen-regulated genes in these cells. They could also increase proliferation of the cells in monolayer culture, which could be inhibited by the antiestrogen ICI 182,780, indicating that the effects were mediated through the oestrogen receptor. However, no antagonist activity of parabens could be detected on regulation of cell proliferation by 17 beta-oestradiol at 10(-10)M. Molecular modelling has indicated the mode by which paraben molecules can bind into the ligand binding pocket of the crystal structure of the ligand binding domain (LBD) of the oestrogen receptor alpha (ERalpha) in place of 17beta-oestradiol; it has furthermore shown that two paraben molecules can bind simultaneously in a mode in which their phenolic hydroxyl groups bind similarly to those of the meso-hexoestrol molecule. Future work will need to address the extent to which parabens can accumulate in hormonally sensitive tissues and also the extent to which their weak oestrogenic activity can add to the more general environmental oestrogen problem.  相似文献

A pharmacological dose of 17β-oestradiol (5 mg kg−1 per injection for a total of five injections over a period of 9 days) increased liver and serum lipid concentration and induced vitellogenin into the serum of the immature female red grouper Epinephelus akaara . The levels of electrolytes, glucose, lactate and protein in the serum, and the protein, glycogen and water content of liver and muscle were not affected. The activities of enzymes regulating gluconeogenesis, glycolysis, Krebs cycle, transamination and glycogen synthesis remained unaltered. These findings are in marked contrast to those of Ng et al . (1984) which showed a strong stimulatory effect of a ten-times smaller dose of 17β-oestradiol on these biochemical parameters. Ultrastructural examination of hepatocytes of these heavily oestrogenized fish revealed that some cells possessed organelles with an abnormally activated appearance while others had undergone necrotic changes, suggesting that some hepatocytes were overstimulated and subsequently degenerated. Results of the present investigation caution against the use of a pharmacological dose of 17β-oestradiol in fish culture.  相似文献

1. The possible mechanism of the oestrogenic inhibition of the androgen-dependent synthesis of alpha2u-globulin in rat liver was explored by a correlative study of the amounts of alpha2u-globulin, its corresponding mRNA and circulating testosterone in oestrogen-treated male rats. 2. Daily treatments of mature male rats with oestradiol-17beta (10 microgram/100g body wt.) decreased and ultimately stopped the hepatic synthesis of alpha2u-globulin as determined by both hepatic and urinary concentrations of the protein. The oestrogen-mediated decrease in the hepatic synthesis of alpha2u-globulin was correlated with a decrease in the mRNA for this protein. 3. Withdrawal of oestrogen resulted in the recovery of alpha2u-globulin synthesis and an increase in mRNA for alpha2u-globulin. 4. At higher doses of oestradiol-17beta (50 microgram/100g body wt.), synthesis of alpha2u-globulin was totally suppressed. In addition, this treatment resulted in an extended period of androgen-insensitivity during which treatment with androgens induced synthesis of neither alpha2u-globulin nor its corresponding mtrna. 5. it is concluded that the oestrogenic inhibition of alpha2u-globulin synthesis is mediated by an oestrogen-dependent decrease in the hepatic content of translatable mRNA for alpha2u-globulin.  相似文献

It has been previously demonstrated that luciferase synthesis in the luminous marine bacteria, Beneckea harveyi and Photobacterium fischeri is induced only when sufficient concentrations of metabolic products (autoinducers) of these bacteria accumulate in growth media. Thus, when cells are cultured in liquid medium there is a lag in luciferase synthesis. A quantitative bioassay for B. harveyi autoinducer was developed and it was shown that many marine bacteria produce a substance that mimics its action, but in different amounts, (20–130% of the activity produced by B. harveyi) depending on the species and strain. This is referred to as alloinduction. None of the bacteria tested produced detectable quantities of inducer for P. fischeri luciferase synthesis.
