The preparation of ultrathin sections of high quality with a Porter-Blum microtome requires that the drive wheel be rotated at a slow, uniform rate during actual cutting. A mechanical device to control the motion of the microtome drive wheel consists of a synchronous motor connected to a drive shaft (4 rev/min), which can be intermittently coupled to the rim of the microtome wheel by a friction drive of 1/2-inch diameter. The microtome drive wheel is rotated manually at a rapid rate until the specimen is about to come into contact with the knife; then the mechanical drive is engaged for slow movement during cutting. When the section has been cut, the drive is disengaged by a lever operated with the left hand; after which, the microtome wheel is turned quickly back to the cutting position with the right hand.  相似文献

A microtome drive, which gives a low cutting speed for the specimen and a fast return to the cutting position, was constructed. To this end, a low speed and a high speed shaft, from an electric motor with a reducing gear, are alternately coupled to the final drive shaft. This coupling is obtained by means of a magnetic clutch and a specially designed pawl clutch. Alternation of low and high speed of the drive shaft depends on the action of the magnetic clutch which is governed by a specimen position contact through a relay system. The specimen position contact is actuated by the microtome itself, which leads to its fully automatic operation. The combination of a magnetic and a pawl clutch in the drive results in a vibration free two-speed cycle of the microtome.  相似文献

A simple method has been developed that allows frozen thin sections of fresh-frozen tissue to be cut on a virtually unmodified ultramicrotome kept at room temperature. A bowl-shaped Dewar flask with a knifeholder in its depths replaces the stage of the microtome; a bar extends down into the bowl from the microtome's cutting arm and bears the frozen tissue near its lower end. When the microtome is operated, the tissue passes a glass or diamond knife in the depths of the bowl as in normal cutting. The cutting temperature is maintained by flushing the bowl with cold nitrogen gas, and can be set anywhere from about -160°C up to about -30°C. The microtome is set for a cutting thickness of 540–1000 A. Sections are picked up from the dry knife edge, and are placed on membrane-coated grids, flattened with the polished end of a copper rod, and either dried in nitrogen gas or freeze-dried. Throughout the entire process the tissue is kept cold and does not come in contact with any solvent. The morphology seen in frozen thin sections of rat pancreas and liver generally resembles that in conventional preparations, although freezing damage and low contrast limit the detail that can be discerned. Among unusual findings is a frequent abundance of mitochondrial granules in material prepared by this method.  相似文献

Thermoelectric cooling units (Frigistor thermoelements) have been used to replace CO2 gas and solid CO2 for microtome stage and knife cooling. These units consist of assemblies of series-connected thermoelements, functioning by the Peltier effect. Cooling is controlled by the direct current supply to the units. Current supply is from a double power pack giving 15 amp at 4.8 v for the knife cooling units, and 15 amp at 1 v for the stage. By varying the current flow, the optimum cutting temperature can be obtained and held indefinitely. An 8-couple Frigistor unit replaces the CO2 stage of a Lipshaw freezing microtome. The stage temperature may be lowered to -36 C in 40-60 sec and at the optimum cutting temperature, 5 μ serial sections of fixed frozen tissue are obtainable. Four 12-couple units are used to cool a 160 mm Jung plane wedge microtome knife fitted to a Reichert sledge microtome, with the stage cooled by one 8-couple unit. The knife temperature can be lowered to -20 C in 5 min; the stage in 1 min. The apparatus has been used to cut a variety of unfixed rat and mouse tissues. The optimum sectioning temperature for such unfixed liver, kidney, spleen, lymph glands, heart, testes, small intestine, pancreas, skin and lung was -20 to -22 C.  相似文献

A rheological motor model that satisfies the major mechanical properties of the skeletal muscle is proposed. The model consists of two Maxwell elements and a Voigt element connected in parallel with each other and has a force generator in it. The model well explains the mechanical behavior in quick and slow recovery phases in the isometric contraction of the muscle and achieves a sufficient isotonic shortening speed. The energy liberation of the motor in isotonic contraction is calculated and a mechanism of control is proposed, which operates so as to decrease the dissipated energy by altering the weights of the elastic and viscous constants in Maxwell elements. And thereby it becomes possible for the motor to possess non-linearity in energy liberation and load-velocity relation alike in muscle. The model would be a base model to be utilized for analyzing the kinetics of human macrosystems and/or for modeling the human neuromuscular system of motion control.  相似文献

The superiority of plastic embedding for the production of high quality sections for light microscopy is well known, but the use of conventional glass knives with a cutting edge of approximately 4 mm has severely restricted the size of specimens in the past. Ralph knives provide a much longer cutting edge and adapters are available for certain models of microtomes and ultramicrotomes. A modified knife holder for use with the Sorvall “Porter Blum” MT-2 microtome was described by Gorycki and Sohm (1979); however, this is not suitable for the MT-1 model. We have therefore designed and made an adapter which enables Ralph knives to be used with this instrument. The design allows approximately 18 mm of cutting edge to be used on each knife, allowing larger specimens to be sectioned than with a conventional glass knife and reducing the frequency with which the knife needs to be changed when working with smaller blocks.  相似文献

A microtome designed for the surface preparation of entire increment cores allows cutting plane surfaces on cores up to a length of 40 cm. Compared to the common sanding procedure, the wood cells of the annual rings remain open, not filled with swarf, and the cell walls are smooth and hence clearly visible. This article aims at describing the functionality of the microtome and the procedures needed for an accurate surface preparation to achieve a good contrast for subsequent image analysis. Possible applications for a more detailed analysis of variations in the tracheid structure of conifers and vessel sizes of oak are presented, which can be included in time series analyses.  相似文献

The quality of sections obtained by microtomy depends to a large extent on the quality and characteristics of the microtome knife itself. Despite the need for improved microtomy techniques, there have been few significant developments since the introduction of glass and diamond knives in the 1950's. The manufacture of microtome knives from vitreous carbon provides new possibilities for developing both improved methods and improved equipment for specimen sectioning. Vitreous carbon has unique physical properties that lend themselves to the generation of precision cutting edges. Such an edge can be obtained either by breaking a piece of vitreous carbon or by using lapidary techniques. The resultant edge seems well adapted to both thick and thin sectioning. The introduction of vitreous carbon as a sectioning tool offers a significant alternative to metal, glass and diamond knives.  相似文献

In discussing the theories and mechanism of sectioning, it is pointed out that virtually no concrete knowledge exists on the subject. The function of different mechanisms and their role in cutting different types of tissue, and the value of microtome knives versus safety razor blades is discussed. Razor blades failed to cut precise sections; a technic for sharpening a knife to give precise sections is outlined, pointing out errors to avoid in sharpening and sectioning. Various types of knife edges are illustrated by photomicrographs. The procedure of sharpening and sectioning technics for critical results is explained. The mechanism of cutting in wood appeared to be of the crushing and tearing type, indicating the necessity of final polishing in sharpening.  相似文献

Calcium waves are propagated in five main speed ranges which cover a billion-fold range of speeds. We define the fast speed range as 3-30μm/s after correction to a standard temperature of 20°C. Only waves which are not fertilization waves are considered here. 181 such cases are listed here. These are through organisms in all major taxa from cyanobacteria through mammals including human beings except for those through other bacteria, higher plants and fungi. Nearly two-thirds of these speeds lie between 12 and 24μm/s. We argue that their common mechanism in eukaryotes is a reaction-diffusion one involving calcium-induced calcium release, in which calcium waves are propagated along the endoplasmic reticulum. We propose that the gliding movements of some cyanobacteria are driven by fast calcium waves which are propagated along their plasma membranes. Fast calcium waves may drive materials to one end of developing embryos by cellular peristalsis, help coordinate complex cell movements during development and underlie brain injury waves. Moreover, we continue to argue that such waves greatly increase the likelihood that chronic injuries will initiate tumors and cancers before genetic damage occurs. Tissues have been conserved for satisfactory enzymatic histochemical assay for up to 12 mo by low temperature storage (in a dry ice chest or at -40° C) and by measures designed to offset the deleterious effects of sublimation of H2O by using the following modifications of standard procedures (which include steps to give a mechanical support to otherwise fragmenting sections): 1. tissue blocks are coated with a polystyrene solution between each storage period; 2. modified bolts are used as tissue holders and types of bolt holders have been designed to fit on standard microtomes which permit manipulation of each tissue block independently of its mates on the same bolt holder, or the simultaneous cutting of all blocks on any one holder with each advance of the microtome feed; 3. tissues are coated with 20% polystyrene in methylene chloride prior to cutting and rubber cement painted on the slide as an adhesive. Lillie's 20% polystyrene diethylbenzene is used as a mounting medium. Other details of practical importance include the technique of freezing, control of moisture within a cryostat and on the microtome, and tests on histochemical procedures.  相似文献

The migrating motor complex (MMC) is a cyclic motor pattern with several phases enacted over the entire length of the small intestine. This motor pattern is initiated and coordinated by the enteric nervous system and modulated by extrinsic factors. Because in vitro preparations of the MMC do not exist, it has not been possible to determine the intrinsic nerve circuits that manage this motor pattern. We have used computer simulation to explore the possibility that the controlling circuit is the network of AH/Dogiel type II (AH) neurons. The basis of the model is that recurrent connections between AH neurons cause local circuits to enter a high-firing-rate state that provides the maximal motor drive observed in phase III of the MMC. This also drives adjacent segments of the network causing slow migration. Delayed negative feedback within the circuit, provided by activity-dependent synaptic depression, forces the network to return to rest after passage of phase III. The anal direction of propagation is a result of slight anal bias observed in projections of AH neurons. The model relates properties of neurons to properties of the MMC cycle: phase III migration speed is governed by neuron excitability, MMC cycle length is governed by the rate of recovery of synaptic efficacy, and phase III duration is governed by duration of slow excitatory postsynaptic potentials in AH neurons. In addition, the model makes experimental predictions that can be tested using standard techniques.  相似文献

报道了一种利用直线电机连续-步进的扫描方式进行光声显微成像的系统,该系统在运动时走弓字型路线,其中直线电机在X轴方向上连续运动,在Y轴方向上以步进的方式运动,采集卡只在X轴电机运动的过程中连续采集。该成像系统较之前振镜扫描的方式扫描的范围更大,可达到厘米尺度范围内的生物组织光声成像;较之前的步进电机逐点扫描的方式扫描速度明显提高。同时本文采用电机带动光和超声换能器一同扫描的方式,较光和超声换能器不动电机带动样品扫描的方式更灵活。另外利用包埋碳丝的模拟样品和在体小鼠耳朵血管来验证系统的成像能力。实验结果表明,这种快速光声显微成像方法及其系统可以实现在体组织的高分辨率成像,有望成为一种无创、实时的光声显微镜应用于生物医学当中。  相似文献

Long-term exercise is associated with improved performance on a variety of cognitive tasks including attention, executive function, and long-term memory. Remarkably, recent studies have shown that even a single bout of aerobic exercise can lead to immediate improvements in declarative learning and memory, but less is known about the effect of exercise on motor learning. Here we sought to determine the effect of a single bout of moderate intensity aerobic exercise on motor skill learning. In experiment 1, we investigated the effect of moderate aerobic exercise on motor acquisition. 24 young, healthy adults performed a motor learning task either immediately after 30 minutes of moderate intensity running, after running followed by a long rest period, or after slow walking. Motor skill was assessed via a speed-accuracy tradeoff function to determine how exercise might differentially affect two distinct components of motor learning performance: movement speed and accuracy. In experiment 2, we investigated both acquisition and retention of motor skill across multiple days of training. 20 additional participants performed either a bout of running or slow walking immediately before motor learning on three consecutive days, and only motor learning (no exercise) on a fourth day. We found that moderate intensity running led to an immediate improvement in motor acquisition for both a single session and on multiple sessions across subsequent days, but had no effect on between-day retention. This effect was driven by improved movement accuracy, as opposed to speed. However, the benefit of exercise was dependent upon motor learning occurring immediately after exercise–resting for a period of one hour after exercise diminished the effect. These results demonstrate that moderate intensity exercise can prime the nervous system for the acquisition of new motor skills, and suggest that similar exercise protocols may be effective in improving the outcomes of movement rehabilitation programs.  相似文献

Experiments carried out in a sealed room, where temperature and humidity could be controlled at will, showed that a high relative humidity in the atmosphere interferes with microtome cutting of tissues embedded in polyethylene glycol wax. The higher the atmospheric temperature the less became the degree of relative humidity which could be tolerated if successful sections were to be obtained.  相似文献

Modifications have been made on the Ullberg technique of taking whole-body sagittal sections of frozen mice on Scotch tape. Three improvements are described which greatly increase the ease and reliability of taking the sections. The microtome is driven by a hydraulic system for a smooth, dependable stroke. The microtome stage has been redesigned to eliminate uneven sections. The cryostat is an ordinary, commercial freezer of the frost-free design which eliminates the need for defrosting and also maintains a lower humidity.  相似文献

The belief propagation (BP) algorithm has some limitations, including ambiguous edges and textureless regions, and slow convergence speed. To address these problems, we present a novel algorithm that intrinsically improves both the accuracy and the convergence speed of BP. First, traditional BP generally consumes time due to numerous iterations. To reduce the number of iterations, inspired by the crucial importance of the initial value in nonlinear problems, a novel initial-value belief propagation (IVBP) algorithm is presented, which can greatly improve both convergence speed and accuracy. Second, .the majority of the existing research on BP concentrates on the smoothness term or other energy terms, neglecting the significance of the data term. In this study, a self-adapting dissimilarity data term (SDDT) is presented to improve the accuracy of the data term, which incorporates an additional gradient-based measure into the traditional data term, with the weight determined by the robust measure-based control function. Finally, this study explores the effective combination of local methods and global methods. The experimental results have demonstrated that our method performs well compared with the state-of-the-art BP and simultaneously holds better edge-preserving smoothing effects with fast convergence speed in the Middlebury and new 2014 Middlebury datasets.  相似文献
