The whitefly Bemisia tabaci is vector of plant infecting viruses and it is considered as one of the most important agricultural pests around the Mediterranean basin. At present, five biotypes of B. tabaci have been reported in the Mediterranean Basin: B, Q, S, T and M. To establish the phylogeographic relationship of these Mediterranean biotypes with others, 54 samples collected in Europe and Africa were analysed by sequencing the mitochondrial cytochrome oxidase I gene (mtCOI). The phylogeny showed that Spanish samples corresponding to the biotype S were related to the haplotype Uganda 2 of the African clade, associated with recent epidemic upsurges of cassava mosaic virus (CMD) in that country. This phylogeographic relationship gave support to a distinct subgroup revealed within the African clade. Bemisia tabaci collected from Euphorbia plants in Italy (biotype T) formed one of the three distinct subgroups existing within the Southeast/Far East Asian clade, while samples from Turkey (biotype M) clustered together with reference mitochondrial sequences from whiteflies from Pakistan and Thailand. Recent reports indicate that Bemisia populations corresponding to the biotypes S and T are distributed in areas larger than those initially delimited. Other results indicated that samples collected in Sudan grouped within the Mediterranean–North Africa clade together with reference sequences of the biotype Q corresponding to insects collected in Spain and Morocco. Mitochondrial haplotypes of B. tabaci samples collected on sweet potato in Ghana clustered with reference sequences of samples from Cameroon corresponding to one of the five Sub-Saharan subgroups already described in the African clade. These data extends the phylogenetic information of the B. tabaci species complex and present new questions to be investigated.  相似文献

Bemisia tabaci biotype B is one of three genetic groups able to induce silverleafing in squash. Several earlier studies had observed that this physiological change was induced only through the feeding of juvenile whiteflies. This study uses adult males only to show that after 11 days the adult males were able to begin to induce silverleafing. Further, the response was density-dependent with 120 adults inducing the first signs of silverleafing in 11 days, 90 adults in 13 days and 60 adults in 15 days.  相似文献

Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae) is a species complex that is one of the most devastating agricultural pests worldwide and affects a broad range of food, fiber and ornamental crops. Unfortunately, using parsimony and neighbor joining methods, global phylogenetic relationships of the major races/biotypes of B. tabaci remain unresolved. Aside from the limitations of these methods, phylogenetic analyses have been limited to only small subsets of the global collection of B. tabaci, and thus limited taxon sampling has confounded the analyses. To improve our understanding of global B. tabaci phylogenetic relationships, a Bayesian phylogenetic technique was utilized to elucidate the relationships among all COI DNA sequence data available in GenBank for B. tabaci worldwide (366 specimens). As a result, the first well-resolved phylogeny for the B. tabaci species complex was produced showing 12 major well-resolved (0.70 posterior probability or above) genetic groups: B. tabaci (Mediterranean/Asia Minor/Africa), B. tabaci (Mediterranean), B. tabaci (Indian Ocean), B. tabaci (sub-Saharan Africa silverleafing), B. tabaci (Asia I), B. tabaci (Australia), B. tabaci (China), B. tabaci (Asia II), B. tabaci (Italy), B. tabaci (New World), B. tabaci (sub-Saharan Africa non-silverleafing) and B. tabaci (Uganda sweet potato). Further analysis of this phylogeny shows a close relationship of the New World B. tabaci with Asian biotypes, and characteristics of the major sub-Saharan Africa non-silverleafing clade strongly supports an African origin of B. tabaci due to its position at the base of the global phylogeny, and the diversity of well-resolved sub-clades within this group. Bayesian re-analyses of B. tabaci ITS, COI, and a combined dataset from a previous study resulted in seven major well-resolved races with high posterior probabilities, also showing the utility of the Bayesian method. Relationships of the 12 major B. tabaci genetic groups are discussed herein.  相似文献

Abstract B biotype is a reasonably important biotype among all known biotypes in the Bemisia tabaci species complex. Local populations of B. tabaci on different host plants were collected from across the Chinese mainland, Taiwan, Pakistan and Israel. From each population of B. tabaci , an internally transcribed spacer region of the ribosomal rDNA gene was amplified, cloned and the sequence determined. Sequence homology analyses were performed and the results were similar to those based on morphology and biological characters. Based on analysis of the internally transcribed spacer 1 sequences, a B biotype-specific primer was designed. The PCR diagnosis results showed that B biotype is identifiable by a specific PCR product by using the forward diagnostic primer paired with a universal reverse primer. This diagnostic primer-based protocol can be used for preliminary analysis of mixed Bemisia populations containing B biotype, as well as other biotypes.  相似文献

Bemisia tabaci (Gennadius) biotype B, called a “superbug”, is one of the most harmful biotypes of this species complex worldwide. In this report, the invasive mechanism and management of B. tabaci bio-type B, based on our 5-year studies, are presented. Six B. tabaci biotypes, B, Q, ZHJ1, ZHJ2, ZHJ3 and FJ1, have been identified in China. Biotype B dominates the other biotypes in many regions of the country. Genetic diversity in biotype B might be induced by host plant, geographical conditions, and/or insecticidal application. The activities of CarE (carboxylesterase) and GSTs (glutathione-S-transferase) in biotype B reared on cucumber and squash were greater than on other host plants, which might have increased its resistance to insecticides. The higher activities of detoxification enzymes in biotype B might be induced by the secondary metabolites in host plants. Higher adaptive ability of biotype B adults to adverse conditions might be linked to the expression of heat shock protein genes. The in-digenous B. tabaci biotypes were displaced by the biotype B within 225 d. The asymmetric mating in-teractions and mutualism between biotype B and begomoviruses via its host plants speed up wide-spread invasion and displacement of other biotypes. B. tabaci biotype B displaced Trialeurodes vapo-rariorum (Westwood) after 4-7 generations under glasshouse conditions. Greater adaptive ability of the biotype B to adverse conditions and its rapid population increase might be the reasons of its suc-cessful displacement of T. vaporariorum. Greater ability of the biotype B to switch to different host plants may enrich its host plants, which might enable it to better compete with T. vaporariorum. Native predatory natural enemies possess greater ability to suppress B. tabaci under field conditions. The kairomones in the 3rd and 4th instars of biotype B may provide an important stimulus in host searching and location by its parasitoids. The present results provide useful information in explaining the mechanisms of genetic diversity, evolution and molecular eco-adaptation of biotype B. Furthermore, it provides a base for sustainable management of B. tabaci using biological and ecological measures.  相似文献

Following the first detection of tomato yellow leaf curl virus (TYLCV) from Reunion (700 km east of Madagascar) in 1997 and the upsurge of Bemisia tabaci (Gennadius) on vegetable crops, two genetic types of B. tabaci were distinguished using RAPD-PCR and cytochrome oxidase I (COI) gene sequence comparisons. One type was assigned to biotype B and the other was genetically dissimilar to the populations described elsewhere and was named Ms, after the Mascarenes Archipelago. This new genetic type forms a distinct group that is sister to two other groups, one to which the B biotype is a member and one to which the Q biotype belongs. The Ms biotype is thought to be indigenous to the region as it was also detected in Mauritius, the Seychelles and Madagascar. Both B and Ms populations of B. tabaci induced silverleaf symptoms on Cucurbita sp., and were able to acquire and transmit TYLCV. Taken together these results indicate that the Ms genetic type should be considered a new biotype of B. tabaci.  相似文献

The whitefly Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae) is a cryptic species complex that contains some of the most damaging pests in tropical and subtropical regions. Recent studies suggested that this complex is composed of at least 24 distinct species. We use the approach from these studies to consider the identity of B. tabaci in Argentina. Previous studies have suggested the presence of a B. tabaci presumably indigenous to the Americas and referred to as the BR biotype in Argentina. We placed the entity referred to as the BR biotype within the B. tabaci cryptic species complex using whiteflies collected in soybean and bean crops in northern and central Argentina. The whiteflies were assigned using the mitochondrial cytochrome oxidase (mtCOI) gene. Four unknown haplotypes plus two Argentina sequences from GenBank formed a cluster that was basal to the rest of the New World sequences. These sequences diverged from the consensus sequence across the range of 3.6 to 4.3%. Applying the species assignment rules of recent studies suggests that the individuals from Argentina form a separate species. A fifth unknown haplotype fell within the New World putative species and formed a distinct cluster with haplotypes from Panama. These results suggest that Argentina has two indigenous species belonging to the B. tabaci cryptic species complex. Rather than using mtCOI sequencing for all B. tabaci collected, a simple random amplified polymorphic DNA-polymerase chain reaction diagnostic was used and tested along with previously published primers designed to work specifically with the BR biotype from Brazil. These primers were either unable to distinguish between the two indigenous members of the complex in Argentina or indicated a difference when none was evident on the basis of mtCOI sequence comparison.  相似文献

烟粉虱B em isia tabaci(G ennad ius)是一种取食植物汁液的重要农业害虫,同时也是许多植物病毒最重要的传播介体之一。烟粉虱被认为是由许多具有明显遗传分化的不同种群即生物型组成的复合种,其中B型烟粉虱是一种入侵性最强的生物型,几乎在世界各地均有分布。由于不同生物型的烟粉虱在寄主范围、传毒能力、地理分布、抗药性等许多生物学方面存在差异,因此对烟粉虱生物型的鉴定及其遗传分化研究对于该害虫的可持续控制具有重要的指导意义。烟粉虱生物型的鉴定方法包括生物学鉴定、酶谱鉴定以及分子标记鉴定的方法,其中使用的分子标记包括RAPD、AFLP、rDNA-ITS1、m tDNA CO I以及SSR等标记。目前,不同生物型的烟粉虱尤其B型烟粉虱的分类地位仍然存在争议。介绍了不同生物型烟粉虱的生物学差异、鉴定方法及其遗传分化研究的最新进展,同时探讨了不同生物型烟粉虱的分类地位和研究方向。  相似文献

Wolbachia是专性的细胞内细菌,广泛存在于节肢动物生殖组织。已有的研究结果表明,节肢动物中存在A组和B组Wolbachia,而烟粉虱Bemisia tabaci中主要检测到了B组Wolbachia。本研究从福建省采集到17个不同烟粉虱地理种群,首先通过rDNA-ITS1克隆测序鉴定了不同烟粉虱地理种群的生物型,然后采用Wolbachia 16S rDNA的特异引物,并通过PCR-RFLP技术分析了不同烟粉虱地理种群中Wolbachia的感染特点。结果表明:从福建省闽侯、平潭、南平、来舟、漳平和沙县采集到的烟粉虱自然种群属于非B型,而非B型烟粉虱种群中存在广泛的超感染现象,即单个非B型烟粉虱个体中同时感染了不同型Wolbachia。相反,B型烟粉虱自然种群的个体中只感染A组Wolbachia。该研究依据密集采样的数据进一步证实了Wolbachia在烟粉虱自然种群中的分布确实与宿主的生物型密切相关,提示Wolbachia可能在烟粉虱的种群分化中发挥作用。  相似文献

Wolbachia是广泛分布于节肢动物体内一类共生细菌,它能够通过多种机制调控宿主的生殖方式。近年来的研究表明,Wolbachia与许多外来生物的成功入侵相关。本文利用长PCR的方法特异扩增了不同生物型烟粉虱(共24个种群)体内Wolbachiawsp基因,结果发现B型和Q型烟粉虱入侵种群体内均未检测到Wolbachia,而在非B/Q型的浙江种群和肯尼亚种群体内检测到了Wolbachia。对该wsp基因进行测序并和已知序列进行同源性分析发现,浙江烟粉虱种群的Wolbachia属于B组Con/Rug亚种群,而肯尼亚种群属于B组Btab1亚种群。Wolbachia的存在与否可能与烟粉虱的成功入侵有一定的关系。图2表2参23  相似文献

Bemisia tabaci has caused significant crop losses in China during the last decade. Recent research has shown that two potentially invasive variants, biotypes B and Q, have been found in several regions of China. Our objective was to determine the biotype status and the distribution of B. tabaci in Shandong province, an important agricultural region of China. Based on mitochondrial DNA markers, both biotypes B and Q were detected, with B being the predominant biotype. The results indicate that the more recently introduced biotype Q has not only been located in China but also has established and spread in some regions.  相似文献

烟粉虱Bemisia tabaci是一个复合种,它具有的生物型分化、较强的传播病毒的能力和抗药性、较快的繁殖速率等特征使其成为我国农业生产中重要害虫之一。本研究利用细胞色素线粒体氧化酶Ⅰ基因,对采集自江苏、广东和云南三省的烟粉虱样本进行了生物型鉴定,并对烟粉虱生物型与寄主植物之间的关联性开展了调查。结果表明,在广东和云南省,都存在未鉴定的土著种群与入侵的B型、Q型共存的现象;同时,在本研究中广东省尚未采集到Q型烟粉虱,而在江苏采集到的粉虱样本全部为入侵型。研究结果还表明,相对于入侵种而言,土著种群显示出更强的寄主植物趋同性;丰富的寄主植物以及本身具有的多食性特性有助于B型、Q型等生物型在世界各地的广泛入侵。  相似文献

Abstract:  Analysis of the genetic diversity among 27 different geographical populations of Bemisia tabaci and determination of biotypes of B. tabaci in China based on amplified fragment-length polymorphism (AFLP) and the mitochondrial cytochrome oxidase I (mtDNA COI) gene sequences were conducted. In AFLP assay, the use of five primer combinations selected from 64 primer combinations allowed the identification of 229 polymorphic bands (97.03%) from 60 to 500 bp, suggesting abundant genetic diversity among different geographical populations of B. tabaci. To further identify biotypes of B. tabaci in China, the mtDNA COI gene sequences of nine representative populations from China, Israel and Spain were obtained. Molecular phylogenetic tree based on AFLP and mtDNA COI gene analyses revealed the presence, in China, of at least four different genetic groups of B. tabaci. B biotype, Q biotype and two non-B/Q biotype. B biotype was distributed nationwide. Q biotype was present only in the local region of China including the YunNan province and BeiJing city. This was also the first report about the invasion of Q biotype into China. Of the other two non-B/Q biotype groups, one was found in ShanDong and HeBei provinces, and another in ZheJiang province. The non-B/Q biotype ZheJiang population showed very high similarity with another Asian population India-IW ( AF110704 ) in mtDNA COI sequences and was possibly a Chinese indigenous population. The close monitoring of the Q biotype in locales of China where commercial plants were exported or imported, is now essential to avoid the further accidental distribution of the Q biotype.  相似文献

烟粉虱是一种危害严重的世界性害虫,是一个快速进化的复合种.本文利用mtCOI分子标记方法,对2010和2011年采自我国9个省(市)的33个烟粉虱种群进行了生物型鉴定和系统发育分析.结果表明: 我国目前存在着B型、Q型、ZHJ-1型、ZHJ-3型、An型以及Nauru型等6种生物型,且不同生物型的分布是不均匀的.遗传距离及系统发育树的分析结果显示,海南省的An型和台湾省的An型聚为一支,为同一来源;中国B型与来自法国和乌干达的B型的亲缘关系较近,同源性达到99%以上;中国的Q型与来自摩洛哥和法国的Q型聚为一个分支,而来自以色列和土耳其的Q型烟粉虱单独聚为一支,说明中国的Q型烟粉虱与来自地中海西部的Q型烟粉虱亲缘关系更近,可以推断中国的Q型烟粉虱的起源地为地中海西部地区.

烟粉虱Bemisia tabaci (Gennadius) B型与Q型是烟粉虱复合种中入侵性较强、分布较广的2种生物型, 当前在许多地区混合发生。这2种生物型的快速鉴别对其种群动态调查及入侵生态学研究具有重要价值。为了验证微卫星位点BEM06与BEM23鉴别B型与Q型烟粉虱的有效性, 本研究分析了这2个微卫星位点的等位基因在国内外17个B型、4个Q型、3个非B/Q型烟粉虱种群的分布特点。结果表明: 这2个微卫星位点的联合使用可鉴别B型与Q型烟粉虱, 但是无法有效地将B型、Q型与一些非B/Q型烟粉虱某些个体区分开来。结果提示: 利用微卫星位点BEM06与BEM23鉴别B型与Q型烟粉虱具有一定的局限性, 尤其是田间烟粉虱存在其他生物型时需要慎重使用这种鉴别方法。  相似文献

The whitefly Bemisia tabaci (Hemiptera: Aleyrodidae) is a worldwide pest and a vector of numerous plant viruses. B. tabaci is composed of dozens of morphologically indistinguishable biotypes and its taxonomic status is still controversial. This phloem-feeder harbours the primary symbiont Portiera aleyrodidarum and potentially six secondary symbionts: Cardinium, Arsenophonus, Hamiltonella, Rickettsia, Wolbachia and Fritschea. In the southwest Indian Ocean, La Réunion hosts two biotypes of this species: B (invasive) and Ms (indigenous). A multiplex PCR was developed to study the symbiont community of B. tabaci on La Réunion. Symbiont community prevalence and composition, host mitochondrial and nuclear genetic diversity, as well as host plant and localization, were described on field populations of La Réunion for B and Ms B. tabaci biotypes and their hybrids. A clear association between symbiotypes and biotypes was shown. Cardinium, Arsenophonus and Rickettsia were found in the Ms biotype (73.6%, 64.2% and 3.3%, respectively). Hamiltonella (exclusively) and Rickettsia were found in the B biotype (78% and 91.2%, respectively). Hybrids harboured all symbiotypes found in Ms and B populations, but with a higher prevalence of Ms symbiotypes than expected under random hybridization. An unexpected majority was Cardinium mono-infected (65.6%), and a striking minority (9%) harboured Cardinium/Arsenophonus. In the hybrids only, genetic diversity was linked to symbiotype. Among the hybrids, significant links were found between symbiotypes and: (i) mitochondrial COI sequences, i.e. maternal origin; and (ii) alleles of nuclear microsatellite loci, specific to either Ms or B parental biotype. Taken together, our results suggest that Cardinium and/or Arsenophonus may manipulate the reproduction of indigenous (Ms) with invasive (B) biotypes of Bemisia tabaci.  相似文献

豇豆轻斑驳病毒(Cowpea mild mottle virus,CPMMV)的流行与其媒介昆虫烟粉虱Bemisia tabaci密切相关,但CPMMV感染寄主植物对B型烟粉虱寄主选择行为和生物学特性的影响尚不清楚。本研究探究了1)CPMMV感染菜豆Phaseolus vulgaris不同时间对B型烟粉虱寄主选择和产卵偏好性的影响;2)CPMMV感染菜豆对B型烟粉虱生物学特性的影响。结果发现:1)相较于健康菜豆,摩擦接种CPMMV后1周,B型烟粉虱却偏好在CPMMV感染的植株上停留和产卵;而摩擦接种后4周,B型烟粉虱却偏好在健康的植株上停留和产卵;2)无论是在摩擦接种后1周和4周的CPMMV感染植株上,B型烟粉虱在健康和CPMMV感染植株上的产卵量和存活率差异不明显。可见,CPMMV感染菜豆对于B型烟粉虱的寄主选择偏好性具有时间效应,而CPMMV感染菜豆对于B型烟粉虱的生物学特性没有显著影响。  相似文献
