Tropical scleractinian corals are considered autotrophic as they rely mainly on photosynthesis-derived nutrients transferred from their photosymbionts. Corals are also able to capture and ingest suspended particulate organic matter, so heterotrophy can be an important supplementary trophic pathway to optimize coral fitness. The aim of this in situ study was to elucidate the trophic status of 10 coral species under contrasted environmental conditions in a French Polynesian lagoon. Carbon (δ13C) and nitrogen (δ15N) isotopic compositions of coral host tissues and photosymbionts were determined at 3 different fringing reefs during wet and dry seasons. Our results highlighted spatial variability in stable isotopic compositions of both coral host tissues and photosymbionts. Samples from the site with higher level of suspended particulate matter were 13C-depleted and 15N-enriched relative to corals and photosymbionts from less turbid sites. However, differences in both δ13C and δ15N between coral host tissues and their photosymbionts (Δhost-photosymbionts 13C and Δhost-photosymbionts 15N) were small (0.27 ± 0.76‰ and 1.40 ± 0.90‰, respectively) and similar at all sites, thus indicating no general increases in the heterotrophic pathway. Depleted δ13C and enriched δ15N values of coral host tissues measured at the most turbid site were explained by changes in isotopic composition of the inorganic nutrients taken up by photosymbionts and also by changes in rate of isotopic fractionation with environmental conditions. Our results also highlighted a lack of significant temporal variations in δ13C and δ15N values of coral host and photosymbiont tissues and in Δhost-photosymbionts 13C and Δhost-photosymbionts 15N values. This temporal stability indicated that corals remained principally autotrophic even during the wet season when photosymbiont densities were lower and the concentrations of phytoplankton were higher. Increased coral heterotrophy with higher food availability thus appears to be species-specific.  相似文献

This is second part from an experiment where the nitrogen retention of poultry by-product meal (PBM) compared to fishmeal (FM) was evaluated using traditional indices. Here a quantitative method using stable isotope ratios of nitrogen (δ15N values) as natural tracers of nitrogen incorporation into fish biomass is assessed. Juvenile rainbow trout (Oncorhynchus mykiss) were fed for 80 days on isotopically distinct diets in which 0, 33, 66 and 100% of FM as main protein source was replaced by PBM. The diets were isonitrogenous, isolipidic and similar in gross energy content. Fish in all treatments reached isotopic equilibrium by the end of the experiment. Two-source isotope mixing models that incorporated the isotopic composition of FM and PBM as well as that of formulated feeds, empirically derived trophic discrimination factors and the isotopic composition of fish that had reached isotopic equilibrium to the diets were used to obtain a quantitative estimate of the retention of each source of nitrogen. Fish fed the diets with 33 and 66% replacement of FM by PBM retained poultry by-product meal roughly in proportion to its level of inclusion in the diets, whereas no differences were detected in the protein efficiency ratio. Coupled with the similar biomass gain of fishes fed the different diets, our results support the inclusion of PBM as replacement for fishmeal in aquaculture feeds. A re-feeding experiment in which all fish were fed a diet of 100% FM for 28 days indicated isotopic turnover occurred very fast, providing further support for the potential of isotopic ratios as tracers of the retention of specific protein sources into fish tissues. Stable isotope analysis is a useful tool for studies that seek to obtain quantitative estimates of the retention of different protein sources.  相似文献

Migratory patterns of bats are not well understood and traditional methods to study this, like capture-mark-recapture, may not provide enough detail unless there are many records. Stable isotope profiles of many animal species have been used to make inferences about migration. Each year Myotis lucifugus and M. septentrionalis migrate from summering roosts to swarming caves and mines in the fall, but the pattern of movement between them is not well understood. In this study, fur δ13C and δ15N values of 305 M. lucifugus and 200 M. septentrionalis were analyzed to make inferences about migration patterns between summering areas and swarming sites in Nova Scotia, Canada. We expected that there would be greater variability in δ13C and δ15N among individuals at swarming sites because it was believed that these sites are used by individuals originating from many summering areas. There was extensive overlap in the standard ellipse area, corrected for small sample sizes (SEAc), of bats at swarming sites and much less overlap in SEAc among groups sampled at summering areas. Meaningful inference could not be made on M. septentrionalis because their low variation in SEAc may have been the result of sampling only 3 summering areas. However, for M. lucifugus, swarming sites had larger SEAc than summering areas and predictive discriminant analysis assigned swarming bats to multiple summering areas, supporting the contention that swarming bats are mixed aggregations of bats from several summering areas. Together, these data support the contention that swarming sites have catchment areas for bats from multiple summering areas and it is likely that the catchment areas for swarming sites overlap. These data suggest that δ13C and δ15N profiling of bat fur offer some potential to make inferences about regional migration in bats.  相似文献

Nitrogen stable isotopes ratios (δ15N) were determined for selected tissues (muscle, liver, blood and yolk) of pregnant females and their embryos of a placental viviparous species, the Pacific sharpnose shark (Rhizoprionodon longurio), and a yolk-sac viviparous species, the speckled guitarfish (Pseudobatos glaucostigmus). The R. longurio embryo tissues were 15N enriched compared to the same tissues in the pregnant female, using the difference in δ15N (Δδ15N) between embryo and adult. Mean Δδ15N was 2.17‰ in muscle, 4.39‰ in liver and 0.80‰ in blood. For P. glaucostigmus, embryo liver tissue was significantly 15N enriched in comparison with liver of the pregnant female (Δδ15N mean = 1.22‰), whereas embryo muscle was 15N depleted relative to the muscle of the pregnant female (Δδ15N mean = −1.22‰). Both species presented a significant positive linear relationship between Δδ15N and embryo total length (LT). The results indicated that embryos have different Δδ15N depending on their reproductive strategy, tissue type analysed and embryo LT.  相似文献

In direct sowing, desiccation of the cover crops with herbicides can affect N availability in agroecosystems, reducing N use efficiency by the crop in succession. In this sense, the objective of the study was to evaluate nitrogen losses in the soil–plant system after application of the herbicides glyphosate and glufosinate-ammonium on pearl millet [Pennisetum glaucum (L.) R. Brown, var. ADR500]. Plants were grown in a sandy soil, fertilized with 15N ammonium sulfate, at N dose of 135.7 mg kg?1. After herbicide application, the N distribution within the plant was changed, with an observed N reduction in leaves and increased N in stalks. The glyphosate and glufosinate reduced, by 25 and 30%, respectively, the amount of N in plant tissues and increased by 3.0 g m?2 the losses in the soil–plant system of the N provided from the fertilizer. This information can be useful in preparing management strategies that will provide greater nitrogen fertilization effectiveness and, consequently, smaller environmental impacts from the use of nitrogen fertilizers and herbicides.  相似文献

We explored the use of carbon and nitrogen isotope ratios (δ13C, δ15N) in sediment organic matter as proxy indicators of historical changes in the trophic state of Lake Taihu, the third largest freshwater lake in China. Stable isotope signatures in four sediment cores spanning the 20th century were compared with instrumental records of lake-water trophic state. The comparative study shows that, between ∼ ∼1950 and 1990 AD, the δ13C and δ15N of sediment organic matter throughout Lake Taihu increased along the trophic gradient from oligotrophy to eutrophy due to biological isotopic fractionation. However, in the 1990s, the trophic state of Lake Taihu diverged into two different trophic systems, a hypereutrophic western Lake Taihu dominated by blue-green algae and a mesoeutrophic eastern Lake Taihu dominated by vascular aquatic plants. During the post-1990 AD shift from mesoeutrophic to hypereutrophic state in western Lake Taihu, organic matter δ13C and δ15N decreased sharply in response to pronounced shifts in the aquatic ecosystem. The results indicate that 13C-depleted phytoplankton replaced macrophytes in western Lake Taihu. δ15N values in western Lake Taihu also decreased because of N2 fixation by cyanobacteria in this highly productive ecosystem. By contrast, in eastern Lake Taihu, organic matter δ13C and δ15N values show a post-1990 AD trend towards slightly lower values, but they remain higher than the long-term average. This recent 13C–enrichment of organic matter indicates that periods of high productivity in the restricted eastern sub-basin of Lake Taihu limited aqueous CO2 availability, causing a decrease in isotopic discrimination during photosynthesis. After ∼ ∼1990 AD, organic matter δ15N values for eastern Lake Taihu only dropped slightly, suggesting that the contribution of phytoplankton to the sediment organic matter increased slightly. Taken together, the results indicate that nitrogen-fixing cyanobacteria probably played a much smaller role in primary productivity in this part of eastern Lake Taihu, compared with western Lake Taihu. Despite the complexity of carbon and nitrogen cycles in lakes, the agreement between the stable isotope signatures and instrumental records for Lake Taihu suggests that δ13C and δ15N in sediment organic matter are capable of recording important shifts in the spatial and temporal evolution of lake-water trophic state.  相似文献

179 species of 7 families of the Cicadina were found in the lowland Trans-Volga woodlands within Nizhniy Novgorod Province, the Republic of Mari El, and Chuvashia. Thirty-tree commonest species were distinguished, and their abundance was compared. Comparison of the species composition of the Cicadina complexes in the model biotopes has shown the greatest similarity between the complexes in the birch forests and spruce forests.  相似文献

Cyanobacterial mats (CBM) are important components of wetland ecosystems in limestone-based regions of the Caribbean. During two sampling periods (July 1999 and January 2000) we measured N2-fixation in samples from 23 different marshes simultaneously with measurements of relevant environmental factors. Samples were evaluated for abundance of five groups of cyanobacteria: (1) Leptolyngbya, (2) Oscillatoria, (3) Chroococcales, (4) Nostoc-& Stigonematales, and (5) dead sheaths. Differences in nitrogen fixation, expressed as nitrogenase activity in nmol C2H4 cm–2 h–1, were best explained by the proportion of heterocyst-forming cyanobacteria. The samples were analyzed for the natural abundance of 15N. 15N values ranged from –1.99 to 11.44 and were strongly negatively correlated with N2-fixation. With all data included, 15N was also strongly correlated with nitrates in water. With the samples from Little Belize (high nitrate content marshes) excluded, the effect of nitrate became insignificant. N2-fixation predicted from 15N measured on an independent data set from September 2000 was moderately accurate (r2 = 0.68, 0.52 and 0.54 for predictions based on July 1999, January 2000 and combined data sets, respectively). When individual sample sets were divided into two groups with 15N < 2 and 15N > 2, the two groups were always highly significantly different in terms of their N2-fixation. The presented evidence suggests that 15N can be used as a reliable indicator of N2-fixation by CBM.  相似文献

Abstract: Information on spring migration routes, geographic linkages among winter, spring, and breeding locations, and potential geographic effects on arrival body condition of northern pintails (Anas acuta) are currently unknown. Through a combination of stable-isotope measurements of tissues representing different periods of dietary integration and body composition analyses, we examined these linkages for pintails breeding in Alaska, USA. We collected 77 females at 4 locations upon spring arrival. We performed carbon (δ13C), nitrogen (δ15N), and hydrogen (δD) isotope measurements on flight feathers, breast feathers, and whole blood, and we conducted body composition analyses. Inference based on stable-isotope values in pintail tissues suggests that philopatry to Alaska was strong, as most of the collected females had stable-isotope values consistent with the boreal forest of Canada or western Alaska and most spring migrating females had whole-blood values indicating use of a food web in the boreal forest before collection. These patterns highlight the importance of the boreal forest for production and staging of pintails. Breast feather isotope values grown during prealternate molt were variable and covered the currently documented distribution of wintering pintails. Our results indicate associations among specific geographic areas, habitat use, and arrival condition of female pintails settling in Alaska. Females that wintered or staged in coastal habitat (as indicated by elevated δ13C values) arrived with less body fat compared to those that we inferred to have wintered or staged on inland freshwater habitat. Those females we inferred to use coastal areas appeared to rely more heavily on agricultural fields for nutrient acquisition (as indicated by elevated δ15N but low δ13C values). Our results provide the first link between low-condition females and inferred use of specific geographic areas before arrival. Conservation on wintering grounds should focus on restoration and protection of wetland complexes that provide adequate natural food resources in proximity to coastal systems that are heavily used by wintering pintails. Transfer of algal cells of Chlorella regularis from 3% CO2 inair into ordinary air in the light increased external carbonicanhydrase (CA) activity as well as photosynthetic affinity forCO2 by several-fold within 2 h. Since no noticeable differencewas observed in CA activity between intact cells and cell homogenates,CA seemed to be mainly localized on the cell surface. Changesin CA activity and K?(CO2) of photosynthesis were not observedin the dark. CA induction was 50%-inhibited by incubation with10 µM DCMU during adaptation of high-CO2 cells to air,whereas it was considerably suppressed when high-CO2 cells preincubatedwith DCMU in the light for 6 h or without DCMU in the dark for24 h were used. The change in K?(CO2) of photosynthesis wasonly slightly affected by DCMU. Uncoupler like carbonylcyanide-m-chlorophenyl-hydrazone(CCCP) and inhibitors of mitochondrial respiration (KCN plussalicylhydroxamic acid) suppressed CA induction during adaptationof high-CO2 cells to low CO2 conditions. These results suggest that photosynthesis is not essential forCA induction in Chlorella regularis when some amounts of photosyntheticproducts are previously stored in the cells and respirationis active. A decrease in K?(CO2) of photosynthesis during adaptationfrom high to low CO2 was mostly independent on photosynthesis.However, light is essential for both phenomena. Stable-isotope analysis (SIA) provides a valuable tool to address complex questions pertaining to elasmobranch ecology. Liver, a metabolically active, high turnover tissue (~166 days for 95% turnover), has the potential to reveal novel insights into recent feeding/movement behaviours of this diverse group. To date, limited work has used this tissue, but ecological application of SIA in liver requires consideration of tissue preparation techniques given the potential for high concentrations of urea and lipid that could bias δ13C and δ15N values (i.e., result in artificially lower δ13C and δ15N values). Here we investigated the effectiveness of (a) deionized water washing (WW) for urea removal from liver tissue and (b) chloroform-methanol for extraction of lipids from this lipid rich tissue. We then (a) established C:N thresholds for deriving ecologically relevant liver isotopic values given complications of removing all lipid and (b) undertook a preliminary comparison of δ13C values between tissue pairs (muscle and liver) to test if observed isotopic differences correlated with known movement behaviour. Tests were conducted on four large shark species: the dusky (DUS, Carcharhinus obscurus), sand tiger (RAG, Carcharias taurus), scalloped hammerhead (SCA, Sphyrna lewini) and white shark (GRE, Carcharodon carcharias). There was no significant difference in δ15N values between lipid-extracted (LE) liver and lipid-extracted/water washed (WW) treatments, however, WW resulted in significant increases in %N, δ13C and %C. Following lipid extraction (repeated three times), some samples were still biased by lipids. Our species-specific “C:N thresholds” provide a method to derive ecologically viable isotope data given the complexities of this lipid rich tissue (C:N thresholds of 4.0, 3.6, 4.7 and 3.9 for DUS, RAG, SCA and GRE liverLEWW tissue, respectively). The preliminary comparison of C:N threshold corrected liver and muscle δ13C values corresponded with movement/habitat behaviours for each shark; minor differences in δ13C values were observed for known regional movements of DUS and RAG (δ13CDiffs = 0.24 ± 0.99‰ and 0.57 ± 0.38‰, respectively), while SCA and GRE showed greater differences (1.24 ± 0.63‰ and 1.08 ± 0.71‰, respectively) correlated to large-scale movements between temperate/tropical and pelagic/coastal environments. These data provide an approach for the successful application of liver δ13C and δ15N values to examine elasmobranch ecology.  相似文献

Photosystem II (PSII) of photosynthesis has the unique ability to photochemically oxidize water. Recently an engineered bacterioferritin photochemical ‘reaction centre’ (BFR-RC) using a zinc chlorin pigment (ZnCe6) in place of its native heme has been shown to photo-oxidize bound manganese ions through a tyrosine residue, thus mimicking two of the key reactions on the electron donor side of PSII. To understand the mechanism of tyrosine oxidation in BFR-RCs, and explore the possibility of water oxidation in such a system we have built an atomic-level model of the BFR-RC using ONIOM methodology. We studied the influence of axial ligands and carboxyl groups on the oxidation potential of ZnCe6 using DFT theory, and finally calculated the shift of the redox potential of ZnCe6 in the BFR-RC protein using the multi-conformational molecular mechanics–Poisson-Boltzmann approach. According to our calculations, the redox potential for the first oxidation of ZnCe6 in the BRF-RC protein is only 0.57 V, too low to oxidize tyrosine. We suggest that the observed tyrosine oxidation in BRF-RC could be driven by the ZnCe6 di-cation. In order to increase the efficiency of tyrosine oxidation, and ultimately oxidize water, the first potential of ZnCe6 would have to attain a value in excess of 0.8 V. We discuss the possibilities for modifying the BFR-RC to achieve this goal.  相似文献

Abstract

Leiopelma hochstetteri, the most widespread of New Zealand's native frogs, is recognised as threatened, and is fully protected by legislation. As a first step to characterise the diet and trophic level of L. hochstetteri within streams in the Waitakere Ranges, Auckland, stable carbon and nitrogen isotope analyses were undertaken on a variety of sympatric terrestrial and aquatic plant and animal species, including adult frogs. These results show that: (1) aquatic and terrestrial food webs are linked by terrestrial inputs into the stream; (2) invertebrate and vertebrate predators separate well into distinct trophic groups, and (3) L. hochstetteri occupies an intermediate trophic position among predators, with a diet, at least as an adult, comprising terrestrial invertebrates. Shortfin eels and banded kokopu are identified as potential predators of L. hochstetteri, but data for rats are inconclusive. These results have important implications for the conservation of New Zealand native frog species and riparian stream habitat.  相似文献

Mosses were collected from 103 locations and analyzed for N concentration and δ15N values. At each location, the samples were collected from two types of site: under the tree canopies and in the adjacent forest openings. A spatial correlation was detected, and ordinary kriging could be applied only to the N in the mosses that were collected in the openings. In contrast, a spatial correlation was not detected for N in the mosses that were collected under the canopies or for the δ15N of the mosses at either type of site. For those, the spatial interpolation was calculated as the sum of the regression predictions and the inverse distance weighted interpolation of residuals. The maps for the N at both site types (canopy/open) identified similar areas with increased N concentrations. The sole exception was the mosses collected under the canopy, for which some local emitters of NOx were also identified.  相似文献

The effects of anticoagulant EDTA and sodium heparin (SH) on stable carbon δ13C and nitrogen δ15N isotopic values of red blood cells (RBC) and blood plasma in juvenile blacktip reef sharks Carcharhinus melanopterus were analysed. Plasma preserved with anticoagulants was not isotopically distinct from plasma stored in no-additive control tubes but RBC δ15N values exhibited small enrichments when preserved with EDTA and SH. Results suggest EDTA and SH are viable anticoagulants for stable isotopic analyses of blood fractions but further studies are advised to validate results.  相似文献
