对4月份至11月份金叶银杏‘万年金'( Ginkgo biloba ‘Wannianjin')32个半同胞子代无性系与亲本的叶色差异进行比较;比较了不同色系叶片的色素含量和比值及叶色参数(L*、a*和b*)的变化,分析了叶色参数与叶片色素含量的相关性;并观察了不同色系的叶绿体超微结构。结果表明:32个半同胞子代无性系可被分为金黄、浅黄、草绿和蓝绿4个色系。随时间推移,草绿和蓝绿色系叶片的总叶绿素( Chl)、叶绿素a( Chla)、叶绿素b( Chlb)和类胡萝卜素( Car)含量均呈“双峰型”变化趋势,Car/Chl和Car/Chla比值的变幅均较小;而金黄和浅黄色系叶片的上述色素含量呈“升高—降低—升高”变化趋势,Car/Chl和Car/Chla比值总体呈“迅速下降—相对稳定—缓慢升高”的变化趋势。各色系叶片的上述色素含量在夏季均不同程度下降,Car/Chlb比值变化差异较大,且金黄和浅黄色系的各色素含量均低于草绿和蓝绿色系。随时间推移,金黄和浅黄色系叶片的L*、a*和b*值以及草绿和蓝绿色系叶片的L*和b*值均先降低后升高,后2个色系的a*值则先升高后下降;并且,前2个色系的L*和b*值总体上显著高于后2个色系,而a*值则总体上低于后2个色系。金黄色系的Chla和Chl含量与L*和a*值显著负相关,而其Car/Chl和Car/Chla比值则与L*、a*和b*值显著或极显著正相关;浅黄色系的Chlb含量与a*值显著负相关,其Car/Chla比值与L*和b*值以及Car/Chlb比值与a*值均显著正相关;草绿色系的Chla含量与L*值显著负相关,其Car/Chla比值与L*和b*值以及Car/Chlb比值与a*值均显著正相关;这3个色系叶片的其余指标间以及蓝绿色系叶片的各指标间均无显著相关性。观察结果显示:金黄和浅黄色系的叶绿体基粒片层发育不健全,基粒片层可见但排列较疏松,且无明显垛叠,分布范围小而稀疏;蓝绿和草绿色系叶绿体的基粒类囊体垛叠层数均较多,基粒片层发达且排列紧致、整齐,分布范围大而稠密。综合分析结果表明:‘万年金'4个色系半同胞子代无性系叶片的呈色差异和叶色变化由多种因素控制,其中,Car/Chl和Car/Chla比值高且叶绿体基粒片层发育不健全是叶片呈黄色的主要原因。  相似文献

实验研究了三疣梭子蟹(Portunus trituberculatus)卵巢发育过程中(Ⅰ—V期)组织色泽、类胡萝卜素组成与含量、抗氧化和非特异性免疫指标的变化规律,进一步探讨了肝胰腺中类胡萝卜素含量变化与抗氧化指标的关系。结果表明:(1)卵巢发育期间,卵巢指数(GSI)显著增加(P<0.05),肝胰腺指数(HSI)整体呈先升后降的趋势;卵巢的红度(a*)值和黄度(b*)值、肝胰腺的亮度(L*)值和b*值均呈显著上升趋势,但卵巢中L*值呈下降趋势。(2)在卵巢发育过程中,卵巢中的总类胡萝卜素、虾青素、叶黄素和β-胡萝卜素含量均为先升后降的趋势,虾青素含量在Ⅳ期最高;肝胰腺中的虾青素含量呈上升趋势,而β-胡萝卜素含量为显著下降趋势;内表皮中的总类胡萝卜素、虾青素、叶黄素、海胆烯酮和β-胡萝卜素含量均呈现下降趋势;就不同组织中类胡萝卜素含量而言,内表皮中虾青素含量最高。卵巢和肝胰腺的L*值与总类胡萝卜素含量呈显著负相关,卵巢的a*值和b*值均与总类胡萝卜素、虾青素、叶黄素、海胆烯酮和β-胡萝卜素含量呈显著正相关。肝胰腺的a*值与各类胡萝卜素含量无显著相关性, b*值仅与虾青素含量呈显...  相似文献

ABSTRACT

Scholars of Japan like to point out that the Japanese are interested in tamed (miniature trees, ikebana, gardens) as opposed to wild nature, and that for the Japanese, culture and nature are not mutually exclusive concepts. Moreover, there hardly seems to be any direct relationship between the Japanese sensitivity to nature and Japanese environmental behaviour. Bearing these general assumptions in mind, this paper analyzes the changes in the relationship between the Japanese and animals through two recent movements in Japan: firefly protection and fly fishing.

This study is based on interviews and participant observation among several firefly protection groups in the Kansai area from 1989 to 1997, among fly-fishing fishermen, and in fishing cooperatives of Gifu prefecture since 1997. In order to ascertain tendencies regarding new shifts in attitudes toward animals, specific observations regarding each of these issues will be disregarded so as to deal only with the commonalities. The similarities between the movements are indeed striking. Traditionally caught by the thousands, and released in houses and gardens for people's enjoyment, fireflies play a role in linking humans with what the Japanese call “close nature.” The firefly image is also very conspicuous in poetry and art. But the firefly population declined in the 1950s, and they are now being protected through drastic legal and social policies, partly influenced by Western environmentalism. The issue of firefly protection is entangled in a criss-cross of interests involving environmental concerns, urban renewal policies and the revival of depopulated rural areas, for which the firefly has become a widely used symbol. At the same time, the movement for firefly protection is highly critical of the traditional ways of dealing with fireflies. Similarly, despite the existence of a traditional Japanese method of “fly-fishing” (tenkara), the recent fly fishing boom seems to be the result of environmental trends such as the “outdoor boom,” and the “catch and release” method.

The introduction of a new fishing technique and the emergence of a consciousness and a commitment to protect fauna and flora disrupted the complex traditional relationship between humans and certain species of animals in both cases. And in both cases, the new approach to nature provoked a strong “cultural resistance” to the loss of the specific Japanese way of dealing with animals. Like “woodpigeon hunting” in southern France, or the whaling issue in other cultural areas, these phenomena appear to be symptomatic of the opposition between tradition(al culture) and environmental concerns. Indeed, the objectification of nature that underscores environmental concern is only possible through a distancing from nature, in which people differentiate and disassociate themselves from other animals… a very “un-Japanese” idea.  相似文献

Small-sized vegetative cells were found to co-occur with normal-sized cells in populations of the European bloom-forming dinoflagellate Gymnodinium cf. nagasakiense Takayama et Adachi, currently known as Gyrodinium aureolum Hulburt, but not in populations of the closely related Japanese species Gymnodiniumn agasakiense. We examined how cell size differentiation may influence growth and cell cycle progression under a 12:12-h light: dark cycle in the European taxon, as compared to the Japanese one. Cell number and volume and chlorophyll red fluorescence in both species varied widely during the photocycle. These variations generally appeared to be related lo the division period, which occurred at night, as indicated by the variations of the fraction of binucleated cells (mitotic index) as well as the distribution of cellular DNA content. “Small” cells of G. cf. nagasakiense divided mainly during the first part of the dark period, although a second minor peak of dividing cells could occur shortly before light onset. In contrast, “large” cells displayed a sharp division peak that occurred 9 h after the beginning of the dark period. The lower degree of synchrony of “small” cells could be a consequence of their faster growth. Alternatively, these data may suggest that cell division is lightly controlled by an endogenous clock in “large” cells and much more loosely controlled in “small” cells. Cells of the Japanese species, which were morphologically similar to “large” cells of the European taxon, displayed an intermediate growth pattern between the two cell types of G. cf. nagasakiense, with a division period that extended to most of the dark period.  相似文献

γ-Glutamyl transpeptidase, glutathionase, and phosphate-independent glutaminase activities, and glutathione contents were measured in IMR-90 cells, human embryonic lung diploid fibroblasts, “aging” in culture, and in cultures of 3T3 L1 mouse preadipocytes differentiating to adipocytes. γ-Glutamyl transpeptidase, glutathionase, and phosphateindependent glutaminase activities in IMR-90 cells were about 8-, 8-, and 7-fold higher in “old” cells as compared to the activities in “young” cells. Total and reduced glutathione contents, expressed in relation to either cell number or DNA content, of IMR-90 cells were 3- and 12-fold higher in “young” cells (population doubling level 22) as compared to “old” cells (population doubling level 49). The ratio of reduced to oxidized glutathione was relatively constant in “young” cells over a range of population doubling levels 18–25; however, relative to the “young” cells, the ratio was decreased in “old” cells over a range of population doubling levels 49–55. γ-Glutamyl transpeptidase, glutathionase, and phosphate-independent glutaminase activities in 3T3 L1 cells were about 4-, 5-, and 5-fold higher in differentiating cells as compared to the activities in undifferentiating cells. Total glutathione contents in 3T3 L1 cells differentiating to adipocytes were similar to glutathione contents in undifferentiating cells. As the culture differentiated to the extent of 50%, almost all of the glutathione was present as oxidized glutathione. These results suggest that the several enzymatic activities and glutathione contents may be used as markers for aging or differentiation in cells. In the course of these studies, a sensitive fluorometric assay was developed, using l-γ-glutamyl-4-methoxy-2-naphthylamide as substrate.  相似文献

How many distinct molecular paths lead to the same phenotype? One approach to this question has been to examine the genetic basis of convergent traits, which likely evolved repeatedly under a shared selective pressure. We investigated the convergent phenotype of blue iris pigmentation, which has arisen independently in four primate lineages: humans, blue‐eyed black lemurs, Japanese macaques, and spider monkeys. Characterizing the phenotype across these species, we found that the variation within the blue‐eyed subsets of each species occupies strongly overlapping regions of CIE L*a*b* color space. Yet whereas Japanese macaques and humans display continuous variation, the phenotypes of blue‐eyed black lemurs and their sister species (whose irises are brown) occupy more clustered subspaces. Variation in an enhancer of OCA2 is primarily responsible for the phenotypic difference between humans with blue and brown irises. In the orthologous region, we found no variant that distinguishes the two lemur species or associates with quantitative phenotypic variation in Japanese macaques. Given the high similarity between the blue iris phenotypes in these species and that in humans, this finding implies that evolution has used different molecular paths to reach the same end. Two subpopulations differing essentially by their mean cell size were observed regularly in cultures and natural samples of the naked dinoflagellate Gymnodinium cf. nagasakiense Takayama et Adachi (currently known as Gyrodinium aureolum Hulburt), a species which frequently forms red tides in North European seas. “Large” cells represented the typical forms; they were morphologically similar to cells of the closely related Japanese species G. nagasakiense, which did not form any subpopulation of reduced size. “Small” and “large” cells of G. cf. nagasakiense had the same DNA content, but the nucleus of the former appeared to be much more condensed during interphase. Each cell type was able to divide and had its own growth dynamics; therefore, any intermediary between pure populations of “small” and of “large” cells were observed in culture. The “large” form generated a “small” cell by an atypical budding-like division, whereas the “small” form gave back a “large” form, once it ceased to divide, by simple enlargement of its cell body. Factory inducing cell size differentiation are yet unclear. Neither nitrogen nor phosphorus starvation induced a significant increase in the relative proportion of “small” and budding cells. Although cell size differentiation is associated with the formation of gametes in a variety of dinoflagellates, we demonstrated that “small” cells of G. cf. nagasakiense are able to divide asexually, in contrast to gametes of most other species. The high proliferative power of “small” cells as compared with normal cells suggests that they could play a significant role during red tides of G. cf. nagasakiense; in contrast, cells of the Japanese species G. Nagasakiense could sustain high growth rates with larger cell size because this species generally blooms in waters much warmer than those found in northern Europe.  相似文献

深入研究花生镉毒害机理对于筛选和利用抗镉污染花生种质资源具有重要的理论和实际意义.本文以14个花生品种为材料,以花期花生功能叶叶绿素含量、根系和叶片丙二醛含量、细胞膜透性和根系氧化活力等生理参数为指标,利用人工气候箱砂培试验,研究了6种Cd浓度胁迫下花生植株的生理毒害反应及其品种间遗传差异.结果表明: 在0~60 mg Cd ·L-1范围内,随着营养液添加Cd浓度的增加,花生叶绿素含量和根氧化活力极显著降低,根、叶细胞膜透性和根、叶丙二醛含量极显著增加,且品种与Cd浓度间具有显著交互作用; 花生根、叶细胞膜透性是对镉胁迫响应最为敏感的生理参数,而叶绿素含量对镉胁迫响应相对不敏感; 各生理参数与营养液Cd浓度关系的线性回归方程的斜率(b)与截距(a)之比值的绝对值︱b/a︱能够较好地描述不同花生品种对镉胁迫的敏感性.对6个生理参数的︱b/a︱值进行综合赋值及敏感性五级聚类分析得知,在供试花生品种中,“中花4号”、“湘农55号”和“湘农3010-w”等3个品种对镉反应高度敏感(Ⅰ级);“莱农29”、“湘农小果w2-7”、“丰花2号”、“莱农13”、“豫花15号”和“丰花3号”等6个品种反应敏感(Ⅱ级);“湘农312”、“祁阳小籽”和“平度01”等3个品种反应一般(Ⅲ级);“花育23 ”和“花育20”等两个品种反应钝感(Ⅳ级).  相似文献

Red (Trifolium pratense L., cv. “Start”) and white clover varieties (Trifolium repens L., cv. “Debut” and cv. “Haifa”) were waterlogged for 14 days and subsequently recovered for the period of 21 days. Physiological and biochemical responses of the clover varieties were distinctive, which suggested different sensitivity toward flooding. The comparative study of morphological and biochemical parameters such as stem length, leaflet area, dry weight, protein content, protein pattern and proteolytic degradation revealed prominent changes under waterlogging conditions. Protease activity in the stressed plants increased significantly, especially in red clover cv. “Start”, which exhibited eightfold higher azocaseinolytic activity compared to the control. Changes in the protein profiles were detected by SDS-PAGE electrophoresis. The specific response of some proteins (Rubisco, Rubisco-binding protein, Rubisco activase, ClpA and ClpP protease subunits) toward the applied stress was assessed by immunoblotting. The results characterized the red clover cultivar “Start” as the most sensitive toward waterlogging, expressing reduced levels of Rubisco large and small subunits, high content of ClpP protease subunits and increased activity of protease isoforms.  相似文献

Abstract

Japanese and American university students rated the intelligence of 56 animals relative to that of humans. In general, American students (n=104) rated animal intelligence higher than did Japanese students (n=235), and females rated intelligence higher than did males. In spite of these differences among groups, the general patterns of ratings were almost identical: The correlation coefficients of the male–female comparisons were 0.99 in both countries, and the coefficients of Japan–US comparisons in male and female groups were 0.95 and 0.96, respectively The intelligence ratings generally corresponded to the so-called “phylogenetic scale” from amoeba to chimpanzee, with several exceptions. The implications of these findings for the cross-cultural analysis of perceptions of animal mentality are discussed.  相似文献

In the marine unicellular chlorophyte, Dunaliella tertiolecta Butcher, the spectrally averaged m vivo absorption cross section, normalized to chlorophyll a (so-called a* values), vary two-fold in response to changes in growth irradiance. We used a kinetic approach to examine the specific factors which account for these changes in optical properties as cells photoadapt. Using Triton X-100 to solubilize membranes, we were able to differentiate between “package” effects and pigmentation effects. Our analyses suggest that 43–49% of the variability in a* is due to changes in pigmentation, whereas 51–57% is due to the “package” effect. Further analyses revealed that changes in cell sue did not significantly affect packaging, while thylakoid stacking and the transparency of thylakoid membranes were important factors. Our results suggest that thylakoid membrane protein/lipid ratios change during photoadaptation, and these changes influence the effective rate of light harvesting per unit chlorophyll a.  相似文献

ABSTRACT

The objective of the present study was to assess the effect of combining CBT-I with wearing blue-light blocking glasses 90 min prior to bedtime on subjective and objective sleep parameters and daily symptoms (anxiety, depression, hyperarousal). Thirty patients (mean age 48.1 ± 16.13 years, range 21–71, 15 men/15 women) completed a CBT-I group therapy program, with groups randomly assigned to either “active” (blue-light filtering glasses) condition or “placebo” (glasses without filtering properties) condition. Patients were continually monitored by wristwatch actigraphy, kept their sleep diaries and completed a standard questionnaire battery at admission and after the end of the program. Statistical analyses showed a greater reduction of BAI score in “active” (4.33 ± 4.58) versus “placebo” (?0.92 ± 3.68) groups of patients [F = 6.389, p = .019, Cohen’s d = 1.26] and significant prolongation of subjective total sleep time in “active” (?36.88 ± 48.68 min.) versus “placebo” (7.04 ± 47.50 min.) [F = 8.56, p < .01, d = 0.91] group. When pre- and post-treatment results were compared in both groups separately, using paired-samples t-tests, significant differences were observed also in the active group for BDI-II score (t = 3.66, p = .003, Cohen’s d = 0.95) and HAS score (t = 2.90, p = .012, Cohen’s d = 0.75). No significant differences were found in the placebo group. In active group, there was also a significant reduction of subjective sleep latency (t = 2.65, p = .021, d = 0.73) and an increase of subjective total sleep time (t = ?2.73, p = .018, d = ?0.76) without change in objective sleep duration which was significantly shortened in the placebo group. We provide further evidence that blocking short-wavelength light in the evening hours may be beneficial for patients suffering from insomnia.  相似文献

IntroductionNon-pharmacological interventions are often the subject of debate in the scientific field. Therefore, it is essential to know the attitudes that health professionals have towards the novel Doll Therapy in Dementia (DTD). The objective of this study was to design and validate two questionnaires to find out the attitudes and knowledge on DTD in health professionals.Material and methodsA total of 120 Spanish-speaking Geriatrics and Gerontology professionals, with a mean age of 38.3 years (DT = 10.99), participated, mostly women (88.3%). A questionnaire (TCM_DEMCO) on knowledge about the DTD and a questionnaire (TCM_DEMAC) on attitudes towards the DTD were designed and applied. The construct validity was assessed by exploratory factor analysis, and reliability was calculated using Cronbach's α.ResultsThe exploratory factor analysis of the TCM_DEMCO questionnaire retained 10 items in 5 factors (“environment and dementia”, “therapeutic application”, “activities”, “methodology” and “benefits and implications”) that explained 70.1% of the total variability, giving a Cronbach α coefficient value > 0.50. The exploratory factor analysis of the TCM_DEMAC questionnaire retained 10 items in two factors (“positive attitudes” and “negative attitudes”) that explained 61.2% of the total variability, giving a Cronbach α value > 0.85.ConclusionsBoth questionnaires are a brief tool to learn about the attitudes and knowledge of Spanish-speaking Geriatrics and Gerontology professionals in matters of DTD. The reliability and validity analyses performed showed good psychometric properties of these tools.  相似文献

ABSTRACT

Handling and training methods of horses, which specially emphasize the importance of understanding horse body language and the use of reinforcements, are often used in practice, yet their effects are not completely known. This study investigated whether the use of a sympathetic approach during the preparation for public auctions influenced the reactivity of young horses towards humans. Sixteen thoroughbred yearlings were prepared for the public auctions during one month: eight horses (“Control”) were handled according to conventional practices, while the others (“Treated”) were handled with two sessions of basic training based on body language. The reactivity of horses was assessed in the presence of an “unfamiliar person” and a “familiar person” inside the horse's box. The experimenter recorded the presence/absence of selected behaviors during seven observational moments: “approaching the box,” “opening the box door,” “entering the box,” and four consecutive observations every thirty seconds. Reactivity of horses was ranked during the first experience of “bit,” “grooming,” “shower,” and application of the “surcingle.” Heart rate was telemetrically recorded during this final test. At the end of the auction preparation, “Treated” horses exhibited more “contact” (p = 0.08) and “lick” (p < 0.05) behaviors in the presence of a person. “Control” horses showed higher (non-significant) percentages of negative (more nervous) rankings during “bit,” “grooming,” and “surcingle” tests. Two “Control” horses showed aggressive behavior during the application of the surcingle and the test was interrupted to guarantee person and animal safety. In this pilot study, horses handled with a sympathetic approach showed less reactive behaviors compared with “Control” horses. It would be interesting to enlarge the sample size and assess if the use of non-coercive handling during the whole training period influences their welfare positively and for a long time.  相似文献

Chickpea (Cicer arietinum L.) is particularly sensitive to water stress at its reproductive phase and, under conditions of water stress, will abort flowers and pods, thus reducing yield potential. There are two types of chickpea: (i) Macrocarpa (“Kabuli”), which has large, rams head‐shaped, light brown seeds; and (ii) Microcarpa (“Desi”), which has small, angular and dark‐brown seeds. Relatively speaking, “Kabuli” has been reported to be more sensitive to water stress than “Desi”. The underlying mechanisms associated with contrasting sensitivity to water stress at the metabolic level are not well understood. We hypothesized that one of the reasons for contrasting water stress sensitivity in the two types of chickpea may be a variation in oxidative injury. In the present study, plants of both types were water stressed at the reproductive stage for 14 d. As a result of the stress, the “Kabuli” type exhibited an 80% reduction in seed yield over control compared with a 64% reduction observed for the “Desi” type. The decrease in leaf water potential (Ψw) was faster in the “Kabuli” compared with the “Desi” type. At the end of the water stress period, Ψw was reduced to ?2.9 and ?3.1 MPa in the “Desi” and “Kabuli” types, respectively, without any significant difference between them. On the last day of stress, “Kabuli” experienced 20% more membrane injury than “Desi”. The chlorophyll content and photosynthetic rate were significantly greater in “Desi” compared with “Kabuli”. The malondialdehyde and H2O2 content were markedly higher at the end of the water stress in “Kabuli” compared with “Desi”, indicating greater oxidative stress in the former. Levels of anti‐oxidants, such as ascorbic acid and glutathione, were significantly higher in “Desi” than “Kabuli”. Superoxide dismutase and catalase activity did not differ significantly between the two types of chickpea, whereas on the 10th day, the activities of ascorbate peroxidase, dehydroascorbate reductase, and glutathione reductase were higher in “Desi”. These findings indicate that the greater stress tolerance in the “Desi” type may be ascribed to its superior ability to maintain better water status, which results in less oxidative damage. In addition, laboratory studies conducted by subjecting both types of chickpea to similar levels of polyethylene glycol‐induced water stress and to 10 μ.mol/L abscisic acid indicated a greater capacity of the “Desi” type to deal with oxidative stress than the “Kabuli” type. The impact of epidermal melanin on objective measurements of human skin colour   总被引:10,自引:0,他引:10  
Objective measurements of human skin colour were made with a tristimulus (L*a*b*) chromameter in a range of different ethnic skin types. These were compared with biochemical measurements of melanin content, melanin composition and melanosome size in skin biopsies obtained from the same sites. L*, a* and b* values were found to vary significantly with ethnicity. In general, constitutively dark skin types have lower L* values, higher a* values and higher b* values than constitutively light skin types. Total epidermal melanin content appears to be the primary determinant of L* values in human skin (r = -0.88; P < 0.00001), whilst melanosome size also has a significant but more subtle influence on L* values (r = -0.73; P < 0.00001). There is also a strong positive contribution to a* values from epidermal melanin (r = 0.66, P < 0.00001), which accounts for the ethnic variation in a* values observed in this study. Melanin is also a major contributor to b* values in lighter skin types (r = 0.71, P < 0.00001). However, this relationship breaks down in darker skin types where b* values actually reach a maximum and then decrease as the concentration of melanin in the skin increases. This appears to be because of optical masking of yellow light by high concentrations of melanin in the epidermis. Analysis of the relationships between L*, a* and b* values in human skin indicate that they are very closely interrelated, and suggest that the optical properties of melanin in the epidermis are very similar to those of a dye on a fabric substrate.  相似文献

Abstract

Photomorphogenic responses induced by UV-B radiation in Brassica oleracea var. capitata.—Ultraviolet radiation can induce a plethora of “damaging” and “non damaging” effects in higher plants. We analyzed two possible photomorphogenic responses to UV-B radiation, the anthocyanin accumulation and the inhibition of hypocotyl elongation by modifying the UV spectral range with specific cut-off filters, under two levels of photon fluence rate. Experimental results suggest that detrimental effects are due to shorter wavelenghts of UV-B region (less than 305 nm); in contrast some adaptative responses may be induced by longer wavelenghts of UV-B region (between 305 and 320 nm). We attempted to determine the involvment of endogenous anthocyanin content in the UV-B photoprotection. Bleomycin (BLM) is a cancer chemotherapeutic agent that cleaves cellular DNA at specific sequences. Using next-generation Illumina sequencing, the genome-wide sequence specificity of DNA cleavage by two BLM analogues, 6′-deoxy-BLM Z and zorbamycin (ZBM), was determined in human HeLa cells and compared with BLM. Over 200 million double-strand breaks were examined for each sample, and the 50,000 highest intensity cleavage sites were analysed. It was found that the DNA sequence specificity of the BLM analogues in human cells was different to BLM, especially at the cleavage site (position “0”) and the “+1” position. In human cells, the 6′-deoxy-BLM Z had a preference for 5′-GTGY*MC (where * is the cleavage site, Y is C or T, M is A or C); it was 5′-GTGY*MCA for ZBM; and 5′-GTGT*AC for BLM. With cellular DNA, the highest ranked tetranucleotides were 5′-TGC*C and 5′-TGT*A for 6′-deoxy-BLM Z; 5′-TGC*C, 5′-TGT*A and 5′-TGC*A for ZBM; and 5′-TGT*A for BLM. In purified human genomic DNA, the DNA sequence preference was 5′-TGT*A for 6′-deoxy-BLM, 5′-RTGY*AYR (where R is G or A) for ZBM, and 5′-TGT*A for BLM. Thus, the sequence specificity of the BLM analogue, 6′-deoxy-BLM Z, was similar to BLM in purified human DNA, while ZBM was different.  相似文献

Background

Residual alcohol effects on physiological and psychological symptoms are commonly experienced the morning after alcohol consumption. The purpose of this study was to assess the effects of L-ornithine on subjective feelings and salivary stress markers the morning after alcohol consumption and to investigate whether L-ornithine acutely accelerates ethanol metabolism.

Methods

This study had a randomized, placebo-controlled, double-masked crossover design. Subjects were all healthy Japanese adults with the ‘flusher’ phenotype for alcohol tolerance. In experiment 1, 11 subjects drank 0.4 g/kg body weight alcohol 1.5 h before their usual bedtime. Half an hour after drinking, they ingested either a placebo or 400 mg ornithine. The next morning on awakening, subjects completed a questionnaire containing a visual analog scale (VAS), the Oguri-Shirakawa-Azumi sleep inventory MA version (OSA-MA), and a profile of mood states (POMS) and collected a saliva sample for measurement of salivary stress markers (cortisol, secretory immunoglobulin A, and α-amylase). In experiment 2, placebo or 400 mg ornithine were administrated to 16 subjects both before and after drinking, and the feeling of drunkenness, breath ethanol concentration and one-leg standing time were repeatedly investigated until 180 min after alcohol consumption.

Results

There were significant decreases in “awareness”, “feeling of fatigue” and “lassitude” VAS scores and in “anger-hostility” and “confusion” POMS scores and a significant increase in “sleep length” in the OSA-MA test. Salivary cortisol concentrations on awakening were reduced after ornithine supplementation. There were no differences between ornithine and placebo in any of the subjective or physiological parameters of acute alcohol metabolism.

Conclusions

Taking 400 mg ornithine after alcohol consumption improved various negative feelings and decreased the salivary stress marker cortisol the next morning. These effects were not caused by an increase in acute alcohol metabolism.
