Abstract

In this study, the seasonality of the biofouling behavior of pilot-scale membrane bioreactors (MBRs) run in parallel with vacant sheets and quorum quenching (QQ) sheets using real municipal wastewater was investigated. QQ media delayed fouling, but low temperatures caused severe biofouling. The greater amount of extracellular polymeric substances (EPSs) produced in cold weather was responsible for the faster biofouling of a membrane, even with QQ media. There were significant negative relationships between EPS levels and water temperature. Cold weather was detrimental to the degradation of quorum sensing signal molecules by QQ sheets, whose activity was restored with a higher dose of QQ bacteria. The QQ bacteria in the sheets experienced a slight loss in activity during the early stage of the field test, but survived in the pilot-scale MBR fed with real wastewater. There were no significant discrepancies in treatment efficiency among conventional, vacant, and QQ MBRs.  相似文献

【背景】近年来,群体感应淬灭(Quorum Quenching,QQ)技术在膜生物污堵防控中的应用研究受到了广泛关注。然而,目前已成功分离纯化的高效QQ菌有限,更多高效QQ菌资源亟待挖掘。【目的】从实际运行的膜生物反应器(MembraneBioreactor,MBR)活性污泥中采样,分离并富集高效QQ菌。【方法】以根瘤农杆菌(Agrobacterium tumefaciens) A136为报告菌株,使用指示琼脂平板法测定各菌株的N-辛酰基高丝氨酸内酯(N-Octanoyl-DL-Homoserine Lactone,C8-HSL)降解能力。以紫色色杆菌(Chromobacterium violaceum) VIR24为报告菌株,定量测定所得QQ菌降解N-己酰高丝氨酸内酯(N-Hexanoyl-DL-Homoserine Lactone,C6-HSL)信号分子的能力。通过微生物形态、生理生化及16SrRNA基因序列测定、构建系统发育树、扫描电子显微镜形态观测等方法对菌株进行分类学鉴定。用共培养法分析QQ菌对生物膜形成的抑制能力,通过聚乙烯醇和海藻酸钠包埋固定化QQ菌。【结果】筛选出了6株高效QQ菌,其中对C8-HSL分解能力最强的为杆状、革兰氏阴性戴尔福特菌属(Delftia sp.) JL5。定量分析结果表明菌株JL5能在10 h内完全降解C6-HSL。菌株JL5显著抑制铜绿假单胞菌(Pseudomonas aeruginosa) PAO1和菠萝泛菌(Pantoea ananatis) SK-1生物膜的形成。固定化后的JL5微球仍具有高效的C6-HSL和C8-HSL信号分子分解能力,而且分解速度较被广泛报道的红球菌(Rhodococcussp.)BH4更快。【结论】研究分离得到了高效的QQ菌,能够有效抑制N-酰基高丝氨酸内酯(N-Acyl-HomoserineLactones,AHL)型群体感应菌生物膜的形成,固定化后仍然具有强QQ活性,具备广泛的应用前景,为后续QQ膜生物污堵防控技术的实践应用奠定了基础。  相似文献

Lab-scale membrane bioreactors (MBRs) were investigated at 12, 18, and 25?°C to identify the correlation between quorum sensing (QS) and biofouling at different temperatures. The lower the reactor temperature, the more severe the membrane biofouling measured in terms of the transmembrane pressure (TMP) during filtration. More extracellular polymeric substances (EPSs) that cause biofouling were produced at 18?°C than at 25?°C, particularly polysaccharides, closely associated with QS via the production of N-acyl homoserine lactone (AHL). However, at 12?°C, AHL production decreased, but the release of EPSs due to deflocculation increased the soluble EPS concentration. To confirm the temperature effect related to QS, bacteria producing AHL were isolated from MBR sludge and identified as Aeromonas sp., Leclercia sp., and Enterobacter sp. through a 16S rDNA sequencing analysis. Batch assays at 18 and 25?°C showed that there was a positive correlation between QS through AHL and biofilm formation in that temperature range.  相似文献

It has been reported that an indigenous quorum quenching bacterium, Rhodococcus sp. BH4, which was isolated from a real plant of membrane bioreactor (MBR) has promising potential to control biofouling in MBR. However, little is known about quorum quenching mechanisms by the strain BH4. In this study, various characteristics of strain BH4 were investigated to elucidate its behavior in more detail in the mixed liquor of MBR. The N-acyl homoserine lactone hydrolase (AHL–lactonase) gene of strain BH4 showed a high degree of identity to qsdA in Rhodococcus erythropolis W2. The LC-ESI-MS analysis of the degradation product by strain BH4 confirmed that it inactivated AHL activity by hydrolyzing the lactone bond of AHL. It degraded a wide range of N-acyl homoserine lactones (AHLs), but there was a large difference in the degradation rate of each AHL compared to other reported AHL–lactonase-producing strains belonging to Rhodococcus genus. Its quorum quenching activity was confirmed not only in the Luria-Bertani medium, but also in the synthetic wastewater. Furthermore, the amount of strain BH4 encapsulated in the vessel as well as the material of the vessel substantially affected the quorum quenching activity of strain BH4, which provides useful information, particularly for the biofouling control in a real MBR plant from an engineering point of view.  相似文献

Quorum sensing gives rise to biofilm formation on the membrane surface, which in turn causes a loss of water permeability in membrane bioreactors (MBRs) for wastewater treatment. Enzymatic quorum quenching was reported to successfully inhibit the formation of biofilm in MBRs through the decomposition of signal molecules, N-acyl homoserine lactones (AHLs). The aim of this study was to elucidate the mechanisms of quorum quenching in more detail in terms of microbial population dynamics and proteomics. Microbial communities in MBRs with and without a quorum quenching enzyme (acylase) were analyzed using pyrosequencing and compared with each other. In the quorum quenching MBR, the rate of transmembrane pressure (TMP) rise-up was delayed substantially, and the proportion of quorum sensing bacteria with AHL-like autoinducers (such as Enterobacter, Pseudomonas, and Acinetobacter) also decreased in the entire microbial community of mature biofilm in comparison to that in the control MBR. These factors were attributed to the lower production of extracellular polymeric substances (EPS), which are known to play a key role in the formation of biofilm. Proteomic analysis using the Enterobacter cancerogenus strain ATCC 35316 demonstrates the possible depression of protein expression related to microbial attachments to solid surfaces (outer membrane protein, flagellin) and the agglomeration of microorganisms (ATP synthase beta subunit) with the enzymatic quorum quenching. It has been argued that changes in the microbial population, EPS and proteins via enzymatic quorum quenching could inhibit the formation of biofilm, resulting in less biofouling in the quorum quenching MBR.  相似文献

Objective

A modified method was used for cell entrapped beads (CEBs) preparation and two aeration intensities (low and high aeration intensity) was supplied as factors to investigate the change of quorum quenching performance for membrane biofouling in membrane bioreactor (MBR).

Results

Dehydrogenase activity and growth trend of activated sludge were improved at high aeration intensity. Compared with C-MBR (with vacant beads), QQ-MBR (with CEBs) had more stable quorum quenching activity and longer application time at high aeration intensity, in which the proteins and polysaccharides were reduced by 15 and 20%, respectively. The difference of EPS concentration in mixed liquor was attributed to the protein concentration controlled by quorum quenching bacteria, meanwhile sufficient organics was necessary to maintain the process.

Conclusions

The better settleability, greater stability and relatively lower hydrophobicity of activated sludge properties was achieved with quorum quenching. The scouring effect of CEBs was promoted at high aeration intensity, further controlling the membrane biofouling.

The biofilm thickness in membrane biofilm reactors (MBfRs) is an important factor affecting system performance because excessive biofilm formation on the membrane surface inhibits gas diffusion to the interior of the biofilm, resulting in a significant reduction in the performance of contaminant removal. This study provides innovative insights into the control of biofilm thickness in O2-based MBfRs by using the quorum quenching (QQ) method. The study was carried out in MBfRs operated at different gas pressures and hydraulic retention times (HRTs) using QQ beads containing Rhodococcus sp. BH4 at different amounts. The highest performance was observed in reactors operated with 0.21 ml QQ bead/cm2 membrane surface area, 12 HRTs and 1.40 atm. Over this period, the performance increase in chemical oxygen demand (COD) removal was 25%, while the biofilm thickness on the membrane surface was determined to be 250 μm. Moreover, acetate and equivalent oxygen flux results reached 6080 and 10 640 mg·m−2·d−1 maximum values, respectively. The extracellular polymeric substances of the biofilm decreased significantly with the increase of gas pressure and QQ beads amount. Polymerase chain reaction denaturing gradient gel electrophoresis results showed that the microbial community in the MBfR system changed depending on operating conditions and bead amount. The results showed that the QQ method was an effective method to control the biofilm thickness in MBfR and provide insights for future research.  相似文献

Quorum quenching (QQ) is an effective approach for the prevention of bacterial infections involving biofilms. This study reports the QQ and anti-biofilm activities of a rhizospheric bacterium identified as Stenotrophomonas maltophilia BJ01. The QQ activity was demonstrated using Chromobacterium violaceum CV026 as a biosensor. A maximum of 95% reduction in violacein production, a quorum sensing-regulated behavior, was observed. Gas chromatography–mass spectroscopy of the extract showed that the active compound was cis-9-octadecenoic acid, which was confirmed by electronspray ionization–mass spectroscopy data. The extract also inhibited biofilm formation of Pseudomonas aeruginosa ATCC 9027 without affecting its growth. Scanning electron and atomic force microscopy showed architectural disruption of the biofilm when treated with the extract. This is the first report of the QQ and anti-biofilm activities of cis-9-octadecenoic acid isolated from any bacterium. It may have the potential to combat detrimental infections with P. aeruginosa. Further validation is required for any possible medical application.  相似文献

【背景】群体感应抑制剂(quorum sensing inhibitor,QSI)作为抗生素潜在替代品,可有效降低致病菌传染性和毒性。沙漠土壤蕴藏着丰富的放线菌资源,是挖掘群体感应抑制剂的重要来源。【目的】解析库木塔格沙漠土壤细菌群落多样性,筛选并挖掘群体感应抑制活性放线菌资源。【方法】采用Illumina Nova Seq高通量测序技术揭示库木塔格沙漠土壤细菌群落组成,利用可培养方法进行土壤放线菌分离和鉴定;选用紫色杆菌CV026模型筛选群体感应抑制活性放线菌,并对其功能特性进行初步评价。【结果】Illumina Nova Seq高通量测序结果显示,样品土壤细菌涉及23门96目150属,优势菌门为变形菌门(Proteobacteria,61%)、放线菌门(Actinobacteria,28%),其中分枝杆菌属(Mycobacterium)为放线菌门最优势菌属(87.3%),其次为红球菌属(Rhodococcus,6.8%)和丙酸杆菌属(Cutibacterium,0.9%)。可培养方法共分离到108株放线菌,归属9科10属,其中优势菌属为链霉菌属(Streptomyces),占65....  相似文献

The aim of the present study was to isolate a variety of quorum quenching bacteria (QB) from the rhizosphere and phyllosphere of three agricultural plants using minimal medium (MM)- and non-minimal medium (NM)-based methods. The members of the Pseudomonas genus constituted the most abundant QB genus, particularly in the rhizospheres of all plant samples and showed the highest quorum quenching (QQ) activity according to a screening assay using a biosensor and 3-oxo-C6-HSL (as an important quorum sensing signal in many phytopathogenic bacteria). In addition, QQ-Pseudomonas were recognised as versatile biocontrol agents against non-bacterial and bacterial plant pathogens, such as Pectobacterium carotovorum subsp. carotovorum (Pcc). Three types of quenching activities, including intracellular and extracellular enzymatic and non-enzymatic activities, were observed in QQ-Pseudomonas. Pseudomonas strains, particularly NM-isolated strains with extracellular activity, are the strongest QQ-based biocontrol agents.  相似文献

Aims: This study aimed to search for a novel quorum‐sensing inhibitor from some fungi and analyse its inhibitory activity. Methods and Results: Chromobacterium violaceum CV026, a double mini‐Tn5 mutant, was used as an indicator to monitor quorum‐sensing inhibition. Auricularia auricular pigments from fruiting bodies were extracted using hydrochloric acid as an infusion, dissolved in alkaline dimethylsulfoxide (DMSO), sterilized by filtration through a 0·22‐μm membrane filter and added to C. violaceum CV026 cultures. Inhibitory activity was measured by quantifying violacein production using a microplate reader. The results have revealed that the alkaline DMSO‐soluble pigments significantly reduced violacein production in a concentration‐dependent manner, a quorum‐sensing‐regulated behaviour in C. violaceum. Conclusions: Auricularia auricular pigments can inhibit bacterial quorum sensing. Significance and Impact of the Study: The results suggest the bioactive constituents from edible and medicinal fungi could interfere with bacterial quorum‐sensing system, regulate its associate functions and prevent bacterial pathogenesis. Further studies were in process in our laboratory to isolate specific compounds from A. auricular pigments, evaluate them as quorum‐sensing inhibitors and analyse the exact mechanism of action.  相似文献

Floccules are another major form of microbial aggregates in aerobic granular sludge systems. Previous studies mainly attributed the persistence of floccules to their relatively faster nutrient uptake and higher growth rate over aerobic granules; however, they failed to unravel the underlying mechanism of the long-term coexistence of these two aggregates. In this work, the existence and function of the floccules in an aerobic granule-dominated sequencing batch reactor were investigated from the view of quorum sensing (QS) and quorum quenching (QQ). The results showed that though the floccules were closely associated with the granules in terms of similar community structures (including the QS- and QQ-related ones), they exhibited a relatively higher QQ-related activity but a lower QS-related activity. A compatible proportion of floccules might be helpful to maintain the QS-related activity and keep the granules stable. In addition, the structure difference was demonstrated to diversify the QS- and QQ-related activities of the floccules and the aerobic granules. These findings could broaden our understanding of the interactions between the coexistent floccules and granules in aerobic granule-dominated systems and would be instructive for the development of the aerobic granular sludge process.  相似文献

The main objective of this work was to determine the effectiveness of various biofouling reducers (BFRs) to operational condition in hybrid membrane bioreactor (MBR) of palm oil mill effluent (POME). A series of tests involving three bench scale (100 L) hybrid MBR were operated at sludge retention times (SRTs) of 30 days with biofouling reducer (BFR). Three different biofouling reducers (BFRs) were powdered actived carbon (PAC), zeolite (Ze), and Moringa oleifera (Mo) with doses of 4, 8 and 12 g L−1 respectively were used. Short-term filtration trials and critical flux tests were conducted. Results showed that, all BFRs successfully removed soluble microbial products (SMP), for PAC, Ze, and Mo at 58%, 42%, and 48%, respectively. At their optimum dosages, PAC provided above 70% reductions and 85% in fouling rates during the short-term filtration and critical flux tests.  相似文献

In the present survey, quorum quenching activity was examined from a biocontrol point of view. Acyl-homoserine lactone (AHL) degrading bacteria were isolated from tomato rhizosphere using two standard bioreporter strains and different synthetic AHLs and then identified according to 16S rDNA sequences. Five isolates capable of inactivating both short and long 3oxo-substituted AHLs showed high similarity with the genera Bacillus, Microbacterium and Arthrobacter, and thereby Bacillus cereus U92 was determined as the most efficient quorum quencher strain. In the quantitative experiments, this strain remarkably inactivated all synthetic AHLs up to 80%. In the laboratory co-cultures, B. cereus U92 efficiently quenched QS-regulated phenotypes in Agrobacterium tumefaciens, Pseudomonas aeruginosa, Pseudomonas chlororaphis and Chromobacterium violaceum. The strain successfully reduced the frequency of Ti-plasmid conjugal transfer in A. tumefaciens by about 99% in the binary cultures. Meanwhile, in a more natural environment, this strain acted as a biocontrol agent, efficient in alleviating QS-regulated crown gall incidence on tomato roots (up to 90%) as well as attenuating Pectobacterium soft rot on potato tubers (up to 60%). On the other hand, reducing phenazine production in P. chlororaphis operated as a suppressor of its QS-regulated biocontrol activity and also inhibited pyocyanin production in P. aeruginosa, a plant growth-promoting bacterium, by 75%. In general, B. cereus U92 seems very promising in the biological control of pathogenic bacteria; however, its broad AHL-degrading activity has a detrimental role on beneficial microbes which should not be neglected.  相似文献

Abstract

Pseudomonas aeruginosa and Serratia marcescens are prominent members belonging to the group of ESKAPE pathogens responsible for Urinary Tract Infections (UTI) and nosocomial infections. Both the pathogens regulate several virulence factors, including biofilm formation through quorum sensing (QS), an intercellular communication mechanism. The present study describes the anti-biofilm and QS quenching effect of thiazolinyl-picolinamide based palladium(II) complexes against P. aeruginosa and S. marcescens. Palladium(II) complexes showed quorum sensing inhibitory potential in inhibiting swarming motility behaviour, pyocyanin production and other QS mediated virulence factors in both P. aeruginosa and S. marcescens. In addition, the establishment of biofilms was prevented on palladium (II) coated catheters. Overall, the present study demonstrates that thiazolinyl-picolinamide based palladium (II) complexes will be a promising strategy to combat device-mediated UTI infections.  相似文献

Aims: For Ophiostoma (Ceratocystis) ulmi, the ability to undergo morphological change is a crucial factor for its virulence. To gain an understanding of quorum‐sensing activity in O. ulmi as it relates to yeast‐mycelium dimorphism control, this study examines the effects of branched‐chain amino acids as well as their fusel alcohols and fusel acids as quorum sensing molecules. Methods and Results: In a defined medium containing glucose, proline and salts, O. ulmi grew as yeasts when the culture was inoculated with a high density of spores (2 × 107 CFU ml?1) and as mycelia when inoculated with a low spore density (4 × 105 CFU ml?1). The cultures displaying yeast morphology secreted a quorum‐sensing factor that shifted the morphology from mycelia to yeast. This quorum‐sensing molecule was lipophilic and extractable by organic solvents from the spent medium. Using GC/MS analysis, it was determined that the major compound in the extract was 2‐methyl‐1‐butanol. A similar effect was observed when the branched‐chain amino acids (fusel alcohol precursors) were used as the nitrogen source. E, E‐farnesol had no effect on the morphology of O. ulmi. Conclusions: Addition of the branched‐chain amino acids or one of the compounds detected in the spent medium, 2‐methyl‐1‐butanol or 4‐hydroxyphenylacetic acid, or methylvaleric acid, decreased germ tube formation by more than 50%, thus demonstrating a quorum sensing molecule behaviour in O. ulmi cultures. Significance and impact of the study: This study presents advances in the investigation of dimorphism in O. ulmi, complementing the existing scientific basis, for studying, understanding and controlling this phenomenon.  相似文献

【背景】传统抑菌剂的大量使用导致细菌产生多重耐药性与抗性,而基于细菌群体感应靶点调控的新型抑菌剂可缓解细菌耐药性与抗性,是未来抑菌剂的发展方向之一。【目的】研究连翘(Forsythiasuspensa)提取物对嗜水气单胞菌(Aeromonashydrophila)群体感应系统的影响及可能的作用机制,为新型抑菌剂的开发提供理论依据。【方法】以紫色杆菌(Chromobacterium violaceum)CV026为报告菌株,以嗜水气单胞菌为供试菌株,采用倍比稀释法测定连翘提取物对2种菌的最小抑菌浓度(minimal inhibitory concentration,MIC),通过微量法测定提取物对嗜水气单胞菌生长、群集运动及蛋白酶活性的影响,利用高效液相色谱串联质谱法分析提取物中的主要成分,采用分子对接模拟探究提取物对嗜水气单胞菌群体感应系统的作用机制。【结果】连翘提取物对紫色杆菌CV026和嗜水气单胞菌的MIC均为16.00mg/mL。在亚抑菌浓度下,连翘提取物处理显著抑制了CV026紫色菌素的产生,最大抑制率高达56.30%。经8.00mg/mL连翘提取物处理后,嗜水气单胞菌的群集运...  相似文献
