The reactions of the alkylsulfonated phosphines LM=Ph2P(CH2)nSO3Na/K (n=2, 3, 4) with K2PtCl4 and K2PdCl4 have been studied in homogeneous aqueous solution as a function of pH. In homogeneous acidic solution the protonated phosphines react to give cis- and trans-PtCl2(LH)2. The biphasic reaction between 1,5-cyclooctadiene platinum(II) chloride in dichloromethane and acidified aqueous LNa/K gives a higher proportion of the cis isomer. In neutral solution the initial reaction to give [PtCl(LNa/K)3]+Cl is followed by slow formation of cis-PtCl2(LNa/K)2. K2PdCl4 reacts more rapidly to give PdCl2(LNa/K)2. In homogeneous alkaline solution rapid oxidation of the phosphine occurs with only small amounts of platinum complex being observable. The biphasic reaction yields phosphine oxide in the aqueous layer and a small amount of the chelate complexes PtL2 in the organic. Representative complexes have been isolated and characterised and the mechanisms for the reactions discussed. The electrospray mass spectra of solutions of the isolated complexes have been recorded in both positive and negative ionisation modes. The positive ionisation spectra are complicated, but platinum and palladium containing ions derived from loss of chloride, H+ and HCl are observed in the negative ionisation spectra.  相似文献

Two dinuclear palladium(II) complexes, [{Pd(en)Cl}2(μ-pz)](NO3)2 and [{Pd(en)Cl}2(μ-pydz)](NO3)2, have been synthesized and characterized by elemental microanalysis and spectroscopic (1H and 13C NMR, IR and UV–vis) techniques (en is ethylenediamine; pz is pyrazine and pydz is pyridazine). The square planar geometry of palladium(II) metal centers in these complexes has been predicted by DFT calculations. The chlorido complexes were converted into the corresponding aqua complexes, [{Pd(en)(H2O)}2(μ-pz)]4+ and [{Pd(en)(H2O)}2(μ-pydz)]4+, and their reactions with N-acetylated l-histidylglycine (Ac–l–His–Gly) and l-methionylglycine (Ac–l–Met–Gly) were studied by 1H NMR spectroscopy. The palladium(II)-aqua complexes and dipeptides were reacted in 1:1 M ratio, and all reactions performed in the pH range 2.0 < pH < 2.5 in D2O solvent and at 37 °C. In the reactions of these complexes with Ac–l–His–Gly and Ac–l–Met–Gly dipeptides, the hydrolysis of the amide bonds involving the carboxylic group of both histidine and methionine amino acids occurs. The catalytic activities of the palladium(II)-aqua complexes were compared with those previously reported in the literature for the analogues platinum(II)-aqua complexes, [{Pt(en)(H2O)}2(μ-pz)]4+ and [{Pt(en)(H2O)}2(μ-pydz)]4+.  相似文献

One of the equatorially coordinated terminal phosphorus atoms of tris(2-(diphenylphosphino)ethyl)phosphine (pp3) ligand on the five-coordinate trigonal-bipyramidal palladium(II) complex, [Pd(4-Cltp)(pp3)](BF4) (4-Cltp = 4-chlorothiophenolate), was selectively oxidized by photolysis to form the four-coordinate square-planar complex. Further selective oxidation of another coordinated terminal phosphorus atom proceeded quantitatively by the substitution reaction with 4-chlorothiophenolate. The solid state structures of these stepwise-oxidized square-planar complexes were determined by X-ray crystal structure analyses, and the structures of the starting trigonal-bipyramidal and the oxidized complexes in solution have been characterized by 31P NMR spectroscopy.  相似文献

As a complement to our previous studies on the development of a class of chiral phosphahelicenes, this article discloses the synthesis, spectroscopic, and structural characterizations of a new phosphahelicene transition metal complex. It demonstrates the ability of these hindered chiral ligands to coordinate Pd (II) in trans‐complexes Cl2Pd(L*)2. In the solid state, the complex adopts a C2‐symmetric arrangement with two ligands facing each other on the same face of the coordination plane. X‐Ray data highlight bending of the Pd (II) unit from the expected planar coordination geometry that might be due to a significant π‐π stacking effect between the central rings of two helical units.  相似文献

Dinaphthylmethylarsine complexes of palladium(II) and platinum(II) with the formulae [MX2L2] (M = Pd, Pt; L = di(1-naphthyl)methylarsine = Nap2AsMe and X = Cl, Br, I), [M2Cl2(μ-Cl)2L2], [PdCl(S2CNEt2)L], [Pd2Cl2(μ-OAc)2L2] and [MCl2(PR3)L] (PR3 = PEt3, PPr3, PBu3, PMePh2) have been prepared. These complexes have been characterized by elemental analyses, IR, Raman, NMR (1H, 13C, 31P) and UV-vis spectroscopy. The stereochemistry of the complexes has been deduced from the spectroscopic data. The crystal structures of trans-[PdCl2(PEt3)(Nap2AsMe)] and of [Pd(S2CNEt2)2], a follow-up product, were determined. The UV-vis spectra of [MX2L2] complexes show a red shift on going from X = Cl to X = I. The complexes [PdX2L2] and [PtX2L2] are strongly luminescent in fluid solution and in the solid at ambient temperature.  相似文献

Abstract

The side effects and resistance of metal-based anticancer drugs prompted us to synthesis a novel series of five Pd(II) complexes of the type [Pd(8-QO)(AA)]; where 8-QO?=?anion of 8-hydroxyquinoline and AA?=?anions of amino acids having nonpolar aliphatic side chain such as glycine (–H), alanine (–CH3), valine (–CH(CH3)2), leucine (–CH2–CH(CH3)2) and isoleucine (–CH(CH3)CH2–CH3). The complexes have been characterized with the help of FT-IR, UV–Vis, one and two-dimensional 1H-NMR, elemental analysis and conductivity measurements. On the basis of these characterization data, a four coordinated square planar geometry for all of these complexes have been proposed. The compounds were screened for their in vitro activities against human cancer cell line, MOLT-4 and their 50% inhibition concentration were ascertained by means of MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay. Since four out of the five newly synthesized compounds were found to be more active than the standard anticancer drug, cisplatin, their detailed interaction with calf thymus DNA (as a target) and bovine serum albumin (BSA) (as a carrier) were also carried out by utilizing absorption spectra, fluorescence spectra and ethidium bromide displacement studies. In these experiments, several binding and thermodynamic parameters were also calculated. These results suggested that hydrogen binding and van der Waals forces play a major role in the interaction between metal complexes with CT-DNA and BSA.

The interaction of [PdII(mida)(Cl)] (1) (mida2− = N-methyliminodiacetate) and [PdII(pydc)(Cl)] (2) (pydc2− = pyridyl-2,6-dicarboxylate) with adenosine-5′-monophosphate (AMP), inosine-5′-monophosphate (IMP) and glutathione (GSH) was studied kinetically as a function of [L] (L = AMP, IMP, GSH) and [Cl] and temperatures (10-35 °C) at pH 4.0. The kinetic results suggest that the reaction of 1 and 2 with the 5′-nucleotides (AMP, IMP) is characterized by the hydrolysis of chloro-complexes followed by the aquo-substitution with purine based 5′-nucleotides through its N7 atom. The reaction of 1 and 2 with GSH takes place through the direct chloride replacement with GSH. Kinetic data and activation parameters are interpreted in terms of an associative mechanism and discussed in reference to the data reported earlier. The [PtII(mida)(Cl)] (3) and [PtII(pydc)(Cl)] (4) complexes were prepared and allowed to interact with AMP and IMP and their reaction products were characterized by 1H NMR studies. The antitumor activity of 3 and 4 was examined against MCF-7 (breast cancer), NCI-H460 (lung cancer) and SF-268 (CNS) cell lines.  相似文献

The new pyrazole ligand 5-(2-hydroxyphenyl)-3-methyl-1-(2-pyridylo)-1H-pyrazole-4-carboxylic acid methyl ester (2) and the corresponding Pt(II), Pd(II) and Cu(II) complexes 3-5 have been synthesized as potential anticancer compounds, and characterized using IR, and (1)H NMR as well as mass spectrometry. The 3-D structures of the Cu(II) complexes were determined by quantum mechanic calculation DFT methodology (density functional theory). The cytotoxicity assay of the ligand and complexes has been performed on leukemia cell lines. In general, the complexes showed lower cytotoxicity than cisplatin, and the Pt(II) and Cu(II) complexes were found to be more efficient in the induction of leukemia cell death than the Pd(II) complex. Our investigations indicate that the antiproliferating activity of the Pt(II) and Cu(II) complexes was partly due to the modulation of cellular differentiation.  相似文献

The reaction of [Rh2(acam)4(H2O)2]ClO4 (1) (Hacam = acetamide) with K2PtCl4 in aqueous solution gave crystals of [Rh2(acam)4(H2O)2][Rh2(acam)4{(μ-Cl)2PtCl2}] · 2H2O (2). The reaction of 1 with K2PdCl4 produced the palladium analog [Rh2(acam)4(H2O)2][Rh2(acam)4{(μ-Cl)2PdCl2}] · 2H2O (3) and a small amount of an aquated palladium complex [Rh2(acam)4{(μ-Cl)2PdCl(H2O)}] · H2O (4). Complexes 2 and 3 have anionic chains of [Rh2(acam)4{(μ-Cl)2MCl2}] (M = Pt, Pd), while 4 includes neutral chains of [Rh2(acam)4{(μ-Cl)2PdCl(H2O)}]. Although all of the structures include infinite chains of (-Rh-Rh-Cl-M-Cl-)n (M = Pt, Pd), the chain structures are different; zigzag for 2 and 3 and helical for 4. In the structures of 2 and 3, the counter cation [Rh2(acam)4(H2O)2]+ made a hydrogen-bonded chain with the crystallization water molecules. The cationic chains and the anionic chains are connected with hydrogen bonds. In the structure of 4, the chains are also linked together by direct hydrogen bonds between the chains and those with the crystallization water molecules. ESR spectra of the powdered samples of 2 and 3 at 77 K were consistent with a rhombic structure: for 2, g1 = 2.111, g2 = 2.054, g3 = 2.004; for 3, g1 = 2.115, g2 = 2.057, g3 = 2.007. These results indicate that there is a spin flip-flop exchange between the cations, [Rh2(acam)4(H2O)2]+, and the units in the anionic chains. The electrical conductivities of 2 and 3 were in the order of 10−7 S cm−1 at room temperature.  相似文献

The synthesis of diethyl (pyridin-2-, -3-, -4-ylmethyl)phosphate (2-pmOpe, 3-pmOpe, 4-pmOpe) ligands and their palladium (II) complexes of general formula trans-[PdCl2L2] (L = 2-pmOpe, 3-pmOpe,4-pmOpe) has been described. Pyridine phosphate derivatives were synthesized via the condensation of phosphorochloridic acid diethyl ester with an appropriate pyridinylmethanol in the presence of triethylamine. The compounds have been identified and characterized by IR, far-IR, 1H NMR, 31P NMR, 31P CP-MAS NMR and elemental analyses. The crystal and molecular structures of palladium (II) complexes, i.e., [PdCl2(2-pmOpe)2] and [PdCl2(4-pmOpe)2] determined by the X-ray diffraction method, are presented. In both structures, Pd(II) ions are four-coordinated by two chlorine atoms and two pyridine nitrogen atoms. The geometry of complexes is square-planar and adopt a trans configuration, which is consistent with preparation method.  相似文献

The preparation of new palladium(II) and platinum(II) complexes derived from alpha-diphenyl ethanedione bis(thiosemicarbazone), 1, and alpha-diphenyl ethanedione bis(4-ethylthiosemicarbazone), 2, is described. The palladium complexes 3 and 4 and platinum complexes 5 and 6 have been characterized by elemental analyses, fast atom bombardment mass spectrometry (FAB(+)) and spectroscopic studies (IR, (1)HNMR). The crystal and molecular structures of the dimeric cyclopalladated compound 4 and the mononuclear platinum complex 6 have been determined by single crystal X-ray diffraction. The cytotoxic activity of the free ligands and palladium and platinum complexes against human A2780 and A2780cisR (acquired resistance to cisplatin) epithelial ovarian carcinoma cells lines is also reported. The IC(50) values for compounds 1, 5 and 6 were found to be higher than that of cisplatin but the maximum antiproliferative activity was similar. Furthermore, the compounds largely retain their activity in the A2780cisR cell line, having a much better resistance factor than cisplatin in the pair of cell lines tested.  相似文献

Reaction of benzisothiazolinone (Bit), a well-known biocide, with the Pd(II) and Pt(II) am(m)ine precursors cis-[Pd(en)(H2O)2](NO3)2 and cis-[Pt(NH3)2(H2O)2](NO3)2 yielded cis-Pd(en)(Bit−1H)2 and cis-Pt(NH3)2(Bit−1H)2, respectively. Bit is bound to the metal centres in both cases through the deprotonated isothiazolinone N. The crystal structures of a Bit/BitO co-crystal and cis-Pd(en)(Bit−1H)2·H2O are also described.  相似文献

Reaction of the ligands 3-phenyl-5-(2-pyridyl)pyrazole (HL1), 3,5-bis(2-pyridyl)pyrazole (HL2), 3-methyl-5-(2-pyridyl)pyrazole (HL3) and 3-methyl-5-phenylpyrazole (HL4) with [MCl2(CH3CN)2] (M = Pd(II), Pt(II)) or [PdCl2(cod)] gives complexes with stoichiometry [PdCl2(HL)2] (HL = HL1, HL2, HL3), [Pt(L)2] (L = L1, L2, L3) and [MCl2(HL4)2] (M = Pd(II), Pt(II)). The new complexes were characterised by elemental analyses, conductivity measurements, infrared and 1H NMR spectroscopies. The crystal and molecular structure of [PdCl2(HL1)] was resolved by X-ray diffraction, and consists of monomeric cis-[PdCl2(HL1)] molecules. The palladium centre has a typical square planar geometry, with a slight tetrahedral distortion. The tetra-coordinated metal atom is bonded to one pyridine nitrogen, one pyrazolic nitrogen and two chloro ligands in a cis disposition. The ligand HL1 is not completely planar.  相似文献

Abstract

It has been suggested that Vibrio vulnificus attaches to plankton and algae and is found in large numbers in the environment. Factors affecting attachment, biofilm formation and morphology of V. vulnificus have not been thoroughly investigated. This study evaluated the role of quorum sensing (QS) and environmental conditions on biofilm development of V. vulnificus. It was found that biofilm development by V. vulnificus was affected by nutrient and glucose concentration, but not by NaCl concentration or temperature under the conditions used here. Moreover, biofilm development of a QS mutant strain proceeded rapidly and sloughing occurred earlier than for the isogenic parent strain. There was a significant loss of viability for the QS mutant biofilm early in development. Hence, it is hypothesised that factors regulated by the QS system play a role in proper biofilm development and maintenance of V. vulnificus. Furthermore, it is shown that biofilm development varied among isolates.  相似文献

In this study, the effects of four novel mononuclear palladium(II) and platinum(II) complexes on the activity of human serum paraoxanase1 were examined. First, four novel mononuclear palladium(II) and platinum(II) complexes were synthesized with a nitrogen donor ligand 1‐(2‐aminoethyl)pyrrolidine and nonsteroidal anti‐inflammatory drugs diclofenac, mefenamic acid. These complexes were characterized by spectroscopic, thermal, and elemental analyses. The crystal structures of complex [Pd(2‐amepyr)2](dicl)2 1 and [Pd(2‐amepyr)2](mef)2 3 were determined by X‐ray crystallography. Then, paraoxonase1 enzyme was purified from human serum. The effects of these complexes on enzyme were evaluated in vitro. The complexes consist of the cationic unit and the counterions. The diclofenac and mefenamic acid acted as a counterion in the complexes. It was observed that all the complexes were stable up to high temperatures. These complexes, even at low doses, inhibited the activity of the enzyme with different inhibition mechanisms.  相似文献

Copper(II), nickel(II) and cobalt(II) complexes of the aspirin metabolite salicylglycine (H2L), of stoichiometry M(HL)2·solvate, have been prepared and characterised. In these complexes salicylglycinate is coordinated to the metal via its carboxylato group and possibly also its amide oxygen in the copper(II) complex. Under basic conditions copper(II) forms the complex Cu(LH−1)·2H2O·MeOH, in which the ligand is coordinated to the metal via its carboxylate and phenolate oxygen atoms and the deprotonated peptide nitrogen atom.  相似文献

We studied the cytotoxic effect and the uptake of Pd(II) complexes of doxycycline (Dox), [Pd(Dox)Cl2], and tetracycline (Tc), [Pd(Tc)Cl2], in chronic myelogenous leukemia cells. The effect of the compounds on macrophage viability was also investigated. Compound 1 is more effective than compound 2 in inhibiting the growth of K562 cells with the IC(50) values of 14.44 and 34.54 microM, respectively. There is a good correlation between cell-growth inhibition and intracellular metal concentrations, determined by inductively coupled plasma atomic emission spectroscopy (ICP-AES). Incubation of the cells with equitoxic concentrations of both compounds yields approximately the same intracellular Pd concentration. At the IC(50) doses, intracellular concentration is ca. 33 x 10(-16) mol/cell for both compounds 1 and 2. This suggests that more [Pd(Tc)Cl2] is needed to produce a cytotoxic effect, because it enters cells more slowly. Both compounds up to 16 microM did not affect the viability of mouse peritoneal macrophages after a 48-h incubation. After 72 h of incubation, the IC(50) values are 22 for [Pd(Dox)Cl2] and 40 microM for [Pd(Tc)Cl(2)]. Therefore, the cytotoxic effect in cancer cells exhibited by both compounds is higher than their effect in macrophages.  相似文献

Some amino acid derivatives, such as R-glycine, have been synthesized together with their full spectroscopic characterization. The sodium salts of these bidentate amino acid ligands have been interacted with [M(bpy)(H2O)2](NO3)2 giving the corresponding some new complexes with formula [M(bpy)(R-gly)]NO3 (where M is Pt(II) or Pd(II), bpy is 2,2′-bipyridine and R-gly is butyl-, hexyl- and octyl-glycine). Due to less solubility of octyl derivatives, the biological activities of butyl and hexyl derivatives have been tested against chronic myelogenous leukemia cell line, K562. The interaction of these complexes with highly polymerized calf thymus DNA has been extensively studied by means of electronic absorption, fluorescence and other measurements. The experimental results suggest that these complexes positive cooperatively bind to DNA presumably via groove binding. Molecular dynamic results show that the DNA structure is largely maintained its native structure in hexylglycine derivative–water mixtures and at lower temperatures. The simulation data indicates that the more destabilizing effect of butylglycine is induced by preferential accumulation of these molecules around the DNA and due to their more negative free energy of binding via groove binding.  相似文献
