We acclimated adult males of three Eremias lizards from different latitudes to 28℃,33℃ or 38℃ to examine whether temperature acclimation affects their thermal preference and tolerance and whether thermal preference and tolerance of these lizards correspond with their latitudinal distributions.Overall,selected body temperature(Tsel)and viable temperature range(VTR)were both highest in E.brenchleyi and lowest in E.multiocellata,with E.argus in between;critical thermal minimum(CTMin)was highest in E.multiocellata and lowest in E.brenchleyi,with E.argus in between;critical thermal maximum(CTMax)was lower in E.multiocellata than in other two species.Lizards acclimated to 28℃ and 38℃ overall selected lower body temperatures than those acclimated to 33℃;lizards acclimated to high temperatures were less tolerant of low temperatures,and vice versa;lizards acclimated to 28℃ were less tolerant of high temperatures but had a wider VTR range than those acclimated to 33℃ and 38℃.Lizards of three species acclimated to the three temperatures always differed from each other in CTMin,but not in Tsel,CTMax and VTR.Our results show that:temperature acclimation plays an important role in influencing thermal preference and tolerance in the three Eremias lizards,although the degrees to which acclimation temperature affects thermal preference and tolerance differ among species;thermal preference rather than tolerance of the three Eremias lizards corresponds with their latitudinal distributions. 相似文献

Objective: To investigate whether chronic administration of the long‐acting glucagon‐like peptide‐1 receptor agonist exendin‐4 can elicit sustained reductions in food intake and body weight and whether its actions require an intact leptin system. Research Methods and Procedures: Male lean and obese Zucker ( fa/fa) rats were infused intracerebroventricularly with exendin‐4 using osmotic minipumps for 8 days. Results: Exendin‐4 reduced body weight in both lean and obese Zucker rats, maximum suppression being reached on Day 5 in obese (8%) and Day 7 in lean (16%) rats. However, epididymal white adipose tissue weight was not reduced, and only in lean rats was there a reduction in plasma leptin concentration. Food intake was maximally suppressed (by 81%) on Day 3 in obese rats but was reduced by only 18% on Day 8. Similarly, in lean rats food intake was maximally reduced (by 93%) on Day 4 of treatment and by 45% on Day 8. Brown adipose tissue temperature was reduced from Days 2 to 4. Plasma corticosterone was elevated by 76% in lean but by only 28% in obese rats. Discussion: Chronic exendin‐4 treatment reduced body weight in both obese and lean Zucker rats by reducing food intake: metabolic rate was apparently suppressed. These effects did not require an intact leptin system. Neither does the absence of an intact leptin system sensitize animals to exendin‐4. Partial tolerance to the anorectic effect of exendin‐4 in lean rats may have been due to elevated plasma corticosterone and depressed plasma leptin levels, but other counter‐regulatory mechanisms seem to play a role in obese Zucker rats. 相似文献

for food preference and selection is not known. We studied 13 rhesus monkeys (2.5 to 3 years of age) for food preference in
a structured choice situation. These animals were fed two levels of dietary zinc: marginally zinc deprived (fed 4 μg zinc/g
diet from conception and throughout development; N=6), or control (fed a 100 μg zinc/g diet throughout the study; N=7). Preference for familiar vs unfamiliar food items, order of food preferences, persistence in retrieval of preferred and
nonpreferred foods, and preference under four deprivation periods were examined. Animals were offered a choice of two food
items, both containing minimal zinc levels, and food choice and latency of choice were measured. Results indicate that animals
fed the marginal zinc diets had reduced preference for unfamiliar foods relative to controls and different patterns of food
preference. These results may be relevant to maintenance of appropriate food selection in marginally zinc deprived populations. 相似文献

The objective of this work was to study the effect of early weaning on alimentary preference for the macronutrients protein, carbohydrate and fat in adult rats. Male Wistar rat pups were weaned by separation from the mother at 15 (D15) or 30 (D30) days old. Body weight and food intake were measured every 30 days until pups were 150 days old. At 110 days of age, the alimentary preference was evaluated for 1 h on 3 consecutive days. At 120 days of age, the palatable diet test was conducted during 3 consecutive 24-h periods. Body weight and food intake were not altered, but early weaning in rats induced an alimentary preference to fat and hyperphagia of a palatable diet. In conclusion, early weaning, although did not modify body weight or basal food intake, promoted an increased preference for palatable and fatty foods. This demonstrates that early weaning is not capable of promoting perceptible alterations of alimentary behavior under normal laboratory conditions. However, in the presence of a stimulating factor such as a choice of nutrients or a palatable diet, a possible latent effect on dietary preferences may become apparent. Over the long term, this preference for foods with high caloric density can lead to obesity and metabolic perturbations. 相似文献

Effects of intracerebro-ventricular infusion of leptin in obese Zucker rats. The obese Zucker rat (OZR) exhibits a missense mutation in the cDNA for the leptin receptor, producing a single amino acid substitution in the extracellular domain of the receptor. A mutation in the leptin receptor gene of the db/db mouse prevents the synthesis of the long splice variant of the receptor. The possibility that the OZR, like the db/db mouse, is refractory to the actions of murine leptin was tested by infusing the protein intracerebroventricularly via a minipump for 7 days. Lean Zucker rats (LZR) infused with leptin acted as positive controls, and other groups of OZR and LZR were infused with vehicle. In LZR, leptin reduced body-weight and food intake and increased brown adipose tissue (BAT) temperature. Plasma corticosterone increased (61%) in these rats, and plasma triglycerides fell (78%). Leptin treatment improved tolerance to an oral glucose load (16% reduction in the area under the blood glucose curve) while lowering plasma insulin. In OZR, the actions of leptin were blunted. Food intake was slightly, but not significantly, reduced. Although there was a reduction in the rate of increase in body mass, the effect of leptin was about half that seen in LZR. BAT temperature and glucose tolerance were unchanged. In contrast to the elevated plasma corticosterone seen in LZR, leptin reduced the level of this hormone (27%) in OZR. In OZR and LZR treated with leptin, the plasma leptin levels were increased 24-fold and 47-fold, respectively. The results suggest that leptin retains some efficacy in OZR, although these rats are less responsive than LZR. 相似文献

Some studies have proposed that self-reported eveningness, which reflects the preference of performing activities in the evening, may harm nutritional health and influence the eating behavior and nutritional status of individuals. However, the relationship between these variables (eveningness and nutritional status) and the consumption of breakfast, which is currently considered a marker of health, has been insufficiently explored by studies. The aim of this study was to investigate, in undergraduate students, the association between diurnal preference, being overweight, and food consumption (with special focus on breakfast). The study included 721 undergraduate students from a Brazilian public university. Dietary intake was assessed by 24-hour food recall, and the usual time for breakfast was identified. Body weight, height, and waist circumference were measured by trained researchers. Diurnal preference was determined by the Horne and Ostberg questionnaire validated for the Brazilian population, and the participants were classified into three categories: evening (coefficient: 16–41), intermediate type (coefficient: 42–58) or morning type (coefficient: 59–86). The prevalence of skipping breakfast was higher among the evening types ( p = 0.02), when compared with morning and intermediate types. A negative association between the diurnal preference coefficient and total caloric (coefficient ?0.25, p = 0.007, r2 adjusted = 0.12), carbohydrate (coefficient ?0.19, p = ?0.04, r2 adjusted = 0.04) and lipid intake (coefficient ?0.18, p = 0.04, r2 adjusted = 0.05) was also found in the breakfast skippers but not in breakfast eaters. In other words, breakfast skippers with diurnal preference values indicative of eveningness consumed more calories, carbohydrates and fats. Evening types presented significant odds of skipping breakfast (OR = 1.7, CI = 1.1–2.9, p = 0.02) when compared with morning and intermediate chronotypes. We conclude that eveningness is associated with skipping breakfast and a higher consumption of calories, carbohydrates and fats in breakfast skippers. LEBEL, CARL, AMY BOURDEAU, DAVID LAU, AND PAMELA HUNT. Biologic response to peripheral and central administration of recombinant human leptin in dogs. Obes Res. Objective: Because leptin is believed to act within the central nervous system, the objective of this study was to test that presumption by comparing the biologic responses to recombinant human leptin (rHuLeptin) when delivered either subcutaneously or intrathecally in a large animal species, the beagle dog. Methods and Procedures: Adult beagle dogs were used for all studies ( n = 3 to 14). Treatment with rHuLeptin was either as daily subcutaneous or intermittent intrathecal injections. Results: Subcutaneously administered rHuLeptin was absorbed with peak concentrations appearing at 2 to 4 hours. After intrathecal administration, cerebral spinal fluid concentrations declined in a bi-phasic manner with a terminal half-life of ?6 to 8 hours. When lean beagles were given leptin subcutaneously, at 0. 05 to 5 gkglday for up to 6 months, reductions in body weight (up to 30%) and food intake (up to 75%) were observed. Body fat loss was observed in both lean and obese dogs, and confirmed by dual energy X-ray absorptiometry and histology of adipose tissue. When rHuleptin was delivered intrathecally at 4 to 1000 μg/dose for up to 3 months, the primary effects observed were reductions in body weight and food intake. In general all findings reported in the intrathecal studies were consistent with those noted in the subcutaneous studies; however, the required intrathecal dose was substantially lower than that for subcutaneous delivery. Discussion: These studies demonstrate that both subcutaneous and intrathecal treatment of rHuLeptin was associated with effects on body weight, food intake, and body fat in dogs. These results support the concept that the central nervous system is the probable primary site of action for leptin and suggest that rHuLeptin has similar physiologic activities that influence body weight, body fat, and metabolism in large animals to those reported previously in rodents. 相似文献

Serum leptin concentrations are an important afferent signal in energy balance homeostasis. It has been speculated that the leptin responsiveness to energy restriction is affected by the functionality of the leptin receptor. The purpose of this analysis was to explore the effect of polymorphisms in the LEPR gene on the acute decline in leptin after 4 days of 65% energy restriction. Leptin concentrations of the study group ( n = 44; all men) declined by 2.3 ± 1.5 μg/L [–39.4% (95% confidence interval: ?43.6 to ?34.9)]. Leptin responses did not statistically differ between noncarriers and carriers of three mutant variants of the polymorphisms: Lys109/Lys109 (?41.4%) vs. Arg109/+ (?37.0%) ( p = 0.33); Gln223/Gln223 (?41.5%) vs. Arg223/+ (?37.8%) ( p = 0.40); Lys656/Lys656 (?39.5%) vs. Asn656/+ (?39.3%) ( p = 0.96). No effect of the assessed polymorphisms in the LEPR gene on the acute decline in leptin after energy restriction was observed. The discovery of leptin by Friedman and coll. in 1995 was a major step forward in our comprehensive view of energy homeostasis. Since the original paper, a tremendous amount of work has been performed in laboratories all over the world. Many recent reviews have described this work in details. In the present review, we focus on the role of leptin on food intake. It is accepted by most authors working in this field that the control of food intake can be divided in two closely-related system: the homeostatic system and the hedonic system. An experimental cattle population was screened for polymorphisms in the leptin gene and five SNPs were found in the regions containing the coding sequences. The association of these polymorphisms with feed intake and fat-related traits was evaluated. The results suggest an association between a polymorphism in exon 2, described here for the first time, and feed intake. Individuals with genotype A/T at this position had 19% greater mean feed intake than individuals with genotype A/A. There was also evidence for a link between leptin haplotypes and some fat-related traits. 相似文献

Synthesized and released by the adipose tissue, leptin is the widely studied 167‐amino acid hormonal protein product of the obesity gene. Originally leptin was defined in association with satiety and energy balance and claimed to be an anti‐obesity factor that functioned via a feedback effect from adipocytes to hypothalamus. There is a growing body of evidence that emphasizes the importance of leptin in the regulation of food intake and body weight in animals and humans, alike. Other research findings point out that it plays a role in the regulation of the metabolism, sexual development, reproduction, hematopoiesis, immunity, gastrointestinal functions, sympathetic activation, and angiogenesis. Purpose To investigate if chewing side preference (CSP) can be used as an indicator of hemispheric laterality in healthy adults. Materials and methods Seventy-five individuals were included. The visual analogue scale (VAS) was used to determine CSP and laterality test for preferred peripheral organs. Results Significant correlation between CSP and hand, foot, ear, and eye side preference was found ( r?=?.41, p?.001; r?=?.34, p?=?.003; r?=?.35, p?=?.03; r?=?.36, p?=?.002). Domestic ruminant selectivity induces floristic changes in pasturelands, risking sustainability and limiting the subsequent availability of susceptible plant species. Development of preferences for species of lower nutritional quality may help to overcome those problems. In this study, we tested the hypothesis that early experience of sheep with a low-quality food (LQF) in a nutritional enriched context increases preference for LQF in adulthood. We predicted a higher proportional consumption of LQF in experienced lambs (EL) than in inexperienced lambs (IL) in choice situations involving LQF and alternative foods. Additionally, we determined intake of LQF by EL and IL at different levels of high-quality food (HQF) availability. From 60 to 210 days of age, EL were fed in separated feed bunks mature oat hay (LQF) simultaneously with sunflower meal (SM) and corn grain (CG), whereas IL were fed alfalfa hay (HQF) simultaneously with SM and CG. After exposure, EL and IL were offered LQF in free choice situations involving alternative foods, and also at five levels of HQF availability (100%, 75%, 50%, 25% and 0% of ad libitum intake). Proportional consumption of LQF was lower or similar in EL than IL. Intake of LQF was also lower or similar in EL than IL at all levels of HQF availability, except when the LQF was the only food available. Our results did not support the hypothesis that early experience with a LQF in a nutritional enriched context increases preference for LQF in adulthood. On the contrary, experience with LQF diminished subsequent preference for LQF in adulthood. It is proposed that, in the conditions of our study, continuous comparison between the LQF and the high-quality supplements (CG and SM) during the early exposure period lead to devaluation of LQF by EL through a simultaneous negative contrast effect. 相似文献

Objective: To investigate the association between leptin levels, polymorphisms in the leptin receptor ( LEPR) gene, and weight gain. Research Methods and Procedures: From two large prospective cohorts in The Netherlands ( n = 17, 500), we compared the baseline leptin of 259 subjects who had gained an average of 12.6 kg (range 5.5 to 33 kg) with 277 subjects who kept stable weight (range ?2.6 to 3.1 kg) after a mean follow‐up of 6.8 years. Three polymorphisms in the LEPR gene (Lys109Arg, Gln223Arg, and Lys656Asn) were determined. Results: Weight gainers had significantly higher baseline leptin levels than those who kept stable weight (odds ratio = 1.27, 95% confidence interval 1.1 to 1.5, per SD increase in log e‐transformed leptin). Weight gainers with the Arg109 or the Arg223 alleles had higher leptin levels compared with the noncarriers of these alleles. Only among men, the association between leptin and weight gain tended to be stronger among those with an Arg223 allele compared with those without this mutation. Discussion: Relatively high leptin levels predict weight gain, suggesting that leptin resistance plays a role in the development of obesity in the general population. Higher leptin levels for those with a Lys109Arg or Gln223Arg mutation (or a linked other marker) may imply that these subjects have a modified functional leptin receptor.
