The content variation dynamics of glucose and free amino acids in blood plasma was followed for lampreys and frogs from autumn till spring, when the exogenous feeding is switched off. In October, the glucose level is rather high (8-10 mM) in blood plasma of both lampreys and frogs. It falls by 50% during winter and falls to a critically low level (1-2 mM) during spring. The lamprey plasma amino acid levels increase by 74% from November to April and thus reach the lower limit known for mammals. The amount of free amino acids in frog plasma decreases by 40% by spring in comparison with the values in autumn. More intensive proteolysis in lamprey tissues compared with that in frog tissues has been confirmed by quantitatively determining leucine, isoleucine, and valine in the blood of these animals. Besides these three amino acids, alanine, glycine, lysine, threonine and, in certain periods, tyrosine have been found to be quantitatively significant in the plasma of both animals.  相似文献

The work studies the content and fatty acid composition of phospholipids as well as the absorption spectra of lipid extracts from red blood cells of poikilothermal and homoiothermal animals at different evolutionary levels. The objects of study include two poikilothermal species, the river lamprey (Lampetra fluviatilis) that uses oxygen dissolved in water, and the common frog (Rana temporaria) that consumes oxygen both from water and from air. A homoithermal animal is the white rat (Rattus rattus) that inhabits the terrestrial-aerial environment. The animals are studied in winter and spring. The phospholipid content in lamprey blood plasma is found to be twice higher than that in its erythrocytes. In the frog and the rat, the ratio is reverse. Determination of the fatty acid lipid composition of red blood cell phospholipids suggests that membranes in the lamprey are denser than in the frog. As for the fatty acids in the erythrocyte fraction of rat blood, they appear to be less diverse, with a double prevalence of saturated acids over unsaturated ones and devoid of long chain (C22) ω3 fatty acids. All of this results in a lower degree of unsaturation and a denser packing of fatty acids in the membrane structures of rat erythrocytes. The mechanism of reversible binding of O2 molecules to hemoglobin in erythrocytes is discussed. Presumably, the mechanism of interaction between molecules of O2 and molecules of water prevents the exchange interaction of electrons of the hemoglobin iron atoms with an oxygen molecule. This is confirmed by our obtained absorption spectra, which show that in the lipid extract almost totally devoid of water the heme isolated from erythrocytes is converted to hemin.  相似文献

The messages for two small but abundant apolipoproteins found in lamprey blood plasma were cloned with the aid of oligonucleotide probes based on amino-terminal sequences. In both cases, numerous clones were identified in a lamprey liver cDNA library, consistent with the great abundance of these proteins in lamprey blood. One of the cDNAs (LAL1) has a coding region of 105 amino acids that corresponds to a 21-residue signal peptide, a putative 8-residue propeptide, and the 76-residue mature protein found in blood. The other cDNA (LAL2) codes for a total of 191 residues, the first 23 of which constitute a signal peptide. The two proteins, which occur in the "high-density lipoprotein fraction" of ultracentrifuged plasma, have amino acid compositions similar to those of apolipoproteins found in mammalian blood; computer analysis indicates that the sequences are largely helix-permissive. When the sequences were searched against an amino acid sequence data base, rat apolipoprotein IV was the best matching candidate in both cases. Although a reasonable alignment can be made with that sequence and LAL1, definitive assignment of the two lamprey proteins to typical mammalian classes cannot be made at this point.  相似文献

Dietary proteins are digested and amino acids from these are utilized by animals to meet body needs for maintenance and production. The form in which these amino acids are absorbed and how they may be transported in the blood seem less certain than once thought. Free amino acids are absorbed and transported in the plasma as well as possibly in the blood cells. These two pools may serve unique functions and therefore deserve individual attention. Peptides are absorbed by the enterocytes. Amino acids in the peptide form appear to be absorbed more readily than free amino acids. The question of whether these peptides are hydrolyzed in the cytosol of the enterocyte or whether they can pass intact into the circulation needs more attention. Evidence suggests that the portal appearance of peptides ranges from negligible contributions to as much as 70% of the amino acids appearing in plasma.  相似文献

A procedure involving the aminoacylation of tRNA for the measurement of l-amino acids is described. The procedure has been applied successfully to the measurements of amino acids in cold acid extracts from blood, plasma, as well as animal tissues. The slight reaction in the absence of tRNA was shown to be due probably to the tRNA content of the enzyme preparation. The requirements for aminoacylation (ionic strength, pH) of tRNA vary with individual amino acids. Nevertheless, the inclusion of a standard curve with each experiment allows the accurate measurement of amino acids under non ideal or even suboptimal conditions. The procedure has been found particularly useful for the study of serial changes in the concentrations of certain amino acids during organ perfusion.  相似文献

The available spawning habitat for the anadromous sea lamprey, Petromyzon marinus L., population that enters the River Mondego has been drastically reduced in the last 20 years. The installation of a fish passage in the first impassable dam, the Açude-Ponte, would enable sea lamprey to recolonise the 34.6-km river stretch between the Açude-Ponte and Raiva dams. In order to assess the suitability of the upstream river stretches for this species, 10 radio-tagged sea lamprey were released upstream of the Açude-Ponte dam and tracked continuously throughout the entire migratory path. Lamprey were unable to pass over intact weirs that had been built for recreational purposes. Sea lamprey movements were more frequent during dusk and night than other periods. Increased river discharge at night, resulting from the operation of the Raiva power station, stimulated lamprey movements but reduced ground speeds recorded. After reaching a certain location, some of the animals maintained their position for several weeks, before undergoing a new movement. Two of the main tributaries of this river stretch were used by some sea lamprey, indicating that the animals were able to find these historical spawning grounds.  相似文献

The effects of starvation and force-feeding on certain tissue and blood constituents were studied in the Northern pike, Esox lucius L. Starvation resulted in a reduction of liver and muscle glycogen and liver lipid. Blood glucose concentration and haematocrit were reduced, total plasma cholesterol levels were increased, while the levels of plasma free fatty acids (FFA), amio acid nitrogen and protein remained unaltered. No significant changes were observed in either muscle protein, muscle water or the response to amino acid loading during the starvation period.
The force-feeding of pike starved for 3 months resulted in liver lipid and muscle glycogen being increased to levels higher than those observed in freshly-captured fish. Liver glycogen, however, increased to values only slightly higher than those of starved animals. Furthermore, while force-feeding had little effect on plasma FFA or protein concentrations, blood glucose, plasma cholesterol and haematocrit returned to the levels found in freshlycaptured fish and those of amino acid nitrogen were higher.
The results indicate that pike are well adapted for periods of prolonged starvation and that hepatic and extra-hepatic lipid and glycogen stores serve for metabolic needs during food shortage, while body protein is conserved. The endocrine basis for these changes in the tissue and blood constituents is discussed.  相似文献

There are many complexities and theoretical aspects to consider for studies of amino acid absorption and metabolism by the gut, liver, and peripheral tissues. The experimental approach must vary depending on the amino acid. Whether to sample whole blood or plasma has to be considered carefully. Also, a specific blood vessel has to be chosen for taking samples. A jugular vein can be the poorest sampling site for many studies. The amounts of individual amino acids appearing in portal blood are different from amounts disappearing from the gut lumen. Some are absorbed in amounts equal to that disappearing but most are absorbed in lesser quantities because of intestinal metabolism. Further, the liver removes absorbed amino acids and synthesizes plasma proteins, urea, and glucose. Peripheral tissues, of course, exchange amino acids with protein for normal turnover but also use amino acids for oxidation and transamination. Alanine, glutamine, glycine, and arginine are important in transporting nitrogen out of peripheral tissues in a nontoxic form. Branched-chain amino acids are removed by both liver and peripheral tissues mainly for plasma protein and ketoacid formation, respectively. During fasting, however, muscle releases branched-chain amino acids while removal by liver is maintained.  相似文献

Mechanisms have been examined by which hyperosmotic blood plasma might elevate the levels of aspartate and glutamate in the brain of the toadBufo boreas. CO2 fixation was assessed by two in vivo methods using [2-14C]glucose injected intracisternally. Thirty minutes after injection, the14C labeling of glutamate and aspartate was more than 100 times greater in brain than in liver. In brain tissues, 40+% of14C atoms appeared to be incorporated into aspartate via the pyruvate carboxylase pathway. Brain tissues of control toads and toads adapting or adapted to hyperosmotic plasma osmolality revealed no differences in the rate of CO2 fixation as related to glucose utilization or tissue pool sizes of glutamate and aspartate. Elevated levels of these amino acids in blood plasma preceded increases in brain tissues. Carbon atoms required during hyperosmotic adaptation for expansion of amino acid pools in brain tissues may, in part, originate from amino acids in blood but apparently not from CO2 fixation in brain.  相似文献

The effect of certain amino acids on transformation of glucoseoxidase as an antigen in different tissues of the animals immunized with it showed that the used glucoseoxidase of the fungus Penicillium vitale Pidopl. et Bilaj possesses antigenic properties peculiar to this enzyme isolated from other sources. 1.5 minutes after a single administration of the antigen to nonimmunized rabbits it is determined in them in descending amounts in such an order: blood, lungs, liver, kidneys, lymphatic nodes. The same order is for immunized animals, but the enzyme quantity 1.5 minutes after the last injection is lower considerably in the kidneys, spleen, lymphatic nodes and in blood serum and changes slightly in the lungs and liver. The preliminary loading of animals under experiment immunized with amino acids accelerates essentially disappearance of the antigen under determination (glucoseoxidase in the given case) in the studied tissues. The data obtained testify to the fact that acceleration of antigen disappearance may be not related to interaction with antibodies.  相似文献

The amino acid concentrations in plasma and blood cells of 5-day old domestic fowl hatchlings that received either standard feeding, protein-feeding or were starved have been determined. The effects of 5-day starvation or protein feeding did not alter significantly the combined amino acid concentration of blood plasma, but decreased blood cell levels. The patterns of individual amino acid changes observed in starvation or protein-feeding were similar in both groups when compared with those of controls. However, starvation-induced effects were actually more marked than those observed in protein-fed animals. The patterns of change with starvation of individual amino acids in the hatchling blood compartments were very different from those observed in mammals subjected to short or medium-term starvation. The mechanisms controlling circulating amino acid concentrations act in both situations studied to maintain the plasma amino acid concentrations despite marked changes in the availability of 2-amino nitrogen energy to the animal; changes in blood amino acid compartmentation buffering plasma amino acid availability.  相似文献

Unlike other amino acids, the branched-chain amino acids (BCAAs) largely bypass first-pass liver degradation due to a lack of hepatocyte expression of the mitochondrial branched-chain aminotransferase (BCATm). This sets up interorgan shuttling of BCAAs and liver–skeletal muscle cooperation in BCAA catabolism. To explore whether complete liver catabolism of BCAAs may impact BCAA shuttling in peripheral tissues, the BCATm gene was stably introduced into mouse liver. Two transgenic mouse lines with low and high hepatocyte expression of the BCATm transgene (LivTg-LE and LivTg-HE) were created and used to measure liver and plasma amino acid concentrations and determine whether the first two BCAA enzymatic steps in liver, skeletal muscle, heart and kidney were impacted. Expression of the hepatic BCATm transgene lowered the concentrations of hepatic BCAAs while enhancing the concentrations of some nonessential amino acids. Extrahepatic BCAA metabolic enzymes and plasma amino acids were largely unaffected, and no growth rate or body composition differences were observed in the transgenic animals as compared to wild-type mice. Feeding the transgenic animals a high-fat diet did not reverse the effect of the BCATm transgene on the hepatic BCAA catabolism, nor did the high-fat diet cause elevation in plasma BCAAs. However, the high-fat-diet-fed BCATm transgenic animals experienced attenuation in the mammalian target of rapamycin (mTOR) pathway in the liver and had impaired blood glucose tolerance. These results suggest that complete liver BCAA metabolism influences the regulation of glucose utilization during diet-induced obesity.  相似文献

Summary Amino acids in exudates of uninoculated pea roots were compared quantitatively and qualitatively with exudates of roots inoculated with Gliocladium catenulatum. This fungus has the potential of causing severe root necrosis. Twenty-one amino acids were found in exudates of healthy roots and apparently some of these were utilized by the fungus. A relatively high concentration of ammonia was detected in exudates of inoculated pea roots, indicating an intense deamination by the fungus. No other imbalance in amino acids was found which could be related to known toxic effects of amino acids on plant tissues.  相似文献

The fatty acid composition of thymus cells, liver, blood plasma, muscle tissue, and tumor focus has been studied in mice with solid Ehrlich carcinoma. The tumor growth in the mice was associated with an increase in the total content of monounsaturated fatty acids in all organs and tissues studied and with a decrease in the total amount of polyunsaturated fatty acids in all tissues except blood plasma. The tumor tissue was characterized by increased levels of monounsaturated fatty acids in comparison with their levels in organs and tissues of intact animals. In the thymus of tumor-bearing mice, the contents of myristic and palmitic saturated fatty acids, which are associated with activation of the T-cell immunity, were increased. The most expressed and considerable changes in the fatty acid composition during tumor growth were observed in the muscle tissue of the animals. A possible role of changes in the fatty acid composition in the investigated organs and tissues of tumor-bearing mice in the organism’s response to tumor growth is discussed.  相似文献

A Xenopus laevis egg cortical granule, calcium-dependent, galactosyl-specific lectin participates in forming the fertilization layer of the egg envelope and functions in establishing a block to polyspermy. We report the cDNA cloning of the lectin, expression of the cortical granule lectin gene during oogenesis and early development, and identification of a new family of lectins. The translated cDNA for the cortical granule lectin had a signal peptide, a structural sequence of 298 amino acids, a molecular weight of 32.7 K, contained consensus sequence sites for N-glycosylation and a fibrinogen domain. The lectin cDNA was expressed during early stages of oogenesis. Lectin glycoprotein levels were constant during development with 2/3 of the lectin associated with the extracellular perivitelline space and the egg/embryo fertilization envelope. Lectin mRNA levels were from 100- to 1000-fold greater in ovary than in other adult tissues. The lectin had no sequence homology to the previously identified lectin families. The lectin had 41-88% amino acid identity with nine translated cDNA sequences from an ascidian, lamprey, frog, mouse, and human. Based on the conserved carbohydrate binding and structural properties of these glycoproteins, we propose a new family of lectins, the eglectin family.  相似文献

The present study was undertaken to determine whether the alteration in amino acid distribution between the plasma and cellular compartment of the blood, previously described in dietary-obese rats, also occurs in genetically obese mice. The blood concentration of individual amino acids and its distribution between plasma and cells of lean and genetically obese mice (ob/ob) have been measured. The results demonstrated that genetically obese mice showed a decrease (55%, P = 0.0489) of free amino acids in the blood cells. Most amino acids were affected and among the most noteworthy characteristics was the observation that the reduction in concentration was more pronounced for the total concentration of the essential amino acids which was reduced by 76% (P = 0.0112) compared to cells of lean mice. These results suggest that an altered amino acid distribution between plasma and blood cells is a consequence of both diet-induced and genetic obesities.  相似文献

The purified preparations of glyceraldehyde-3-phosphate dehydrogenase isolated from frog and pike skeletal muscles were found homogenous under polyacrylamide gel electrophoresis. Their amino acid composition is similar to that of glyceraldehyde-3-phosphate dehydrogenase from other animal species. The interaction kinetics for frog and pike glyceraldehyde-3-phosphate dehydrogenase SH-groups with 5,5'-dithio-bis-(2-nitrobenzoate) (DTNB) were studied. A negative correlation between the thermal stability of the enzyme preparations from pig, pike, lamprey and frog muscles and the reactivity of their SH-groups with respect to DTNB was observed. NAD at saturating concentrations was found to protect the enzyme from lower vertebrates muscles against thermal inactivation in a lesser degree than does the pig muscle enzyme. The weaker protective effect of NAD was observed for lamprey and frog enzyme preparations, which are characterized by a low SH-group reaction ability. Frog and pike apoenzymes are considerably more resistant to trypsin proteolysis than the pig apoenzyme.  相似文献
