The ability of a series of structures to mimic the geometric and electronic properties of an ideal –turn has been studied. Initially, an exhaustive conformational analysis was carried out using the molecular dynamics technique at high temperature followed by minimization. Additionally, each minimum was optimized with the semi–ab initio molecular orbital method SAM1. Then, the unique minima found have been superimposed with ideal –turns, classic and inverse, using the SEAL program which takes into account steric and electronic parameters for the superpositions and finally, three molecular similarity indices were determined for each superposition. These indices consider the general steric and electronic characteristics of the structures, as well as, the position of the carbon atoms that correspond to the Ci and Ci+2 in the peptide chain.  相似文献

The aims of the present investigation were (a) to compare the lateral mobility of membrane receptors of human fibroblasts and polymorphonuclear leukocytes (PMNL) labelled with either platelet-derived growth factor (PDGF), or the lectin wheat germ agglutinin (WGA), and (b) to study effects of serum or PDGF on the mobility of these receptor molecules in human fibroblasts. Human foreskin fibroblasts (AG 1523) were grown on coverslips either under standard (10%) or under serum-free conditions yielding normal and starved cells, respectively. The receptor mobility was studied in response to exposure to PDGF, or serum, in short time or prolonged incubations. Human polymorphonuclear leukocytes (PMNL) were adhered to microscope slides by clotting drops of blood. They were stained with rhodaminated PDGF or fluoresceinated WGA. The diffusion of labelled receptors was assessed with fluorescence recovery after photobleaching (FRAP). It was found that (a) fibroblasts grown at normal serum concentration had a lower diffusion coefficient (D=3×10–10 cm2 s–1) for the PDGF-receptor and a slightly lower mobile fraction (R=60%) than starved cells (D=5×10–10 cm2s–1 and R=73%), (b) addition of serum to starved cells increased both D and R for the PDGF receptor to 12×10–10 cm2 s–1 and 96%, respectively, (c) a similar pattern was obtained for WGA-labelled glycoconjugates indicating general membrane effects of serum-induced cell stimulation, and (d) in PMNL the PDGF receptor displayed motility characteristics (D=3–4×10–10 cm2 s–1 and R=59%) similar to those in fibroblasts, possibly suggesting equivalent anchorage mechanisms in the membrane.  相似文献

Nitrification and denitrification rates were estimated simultaneously in soil-floodwater columns of a Crowley silt loam (Typic Albaqualfs) rice soil by an15N isotopic dilution technique. Labeled NO 3 was added to the floodwater of soil-water columns, half were treated with urea fertilizer. The (NO 3 +NO 2 )–N and (NO 3 +NO 2 )–N concentrations in the floodwater were measured over time and production and reduction rates for NO 3 calculated. Nitrate reduction in the urea amended columns averaged 515 mol N m–2h–1 and nitrification averaged 395 mol N m–2h–1 over the 35–153 d incubation. The nitrification rate for 4–19 d sampling period (1,560 mol N m–2h–1) in the urea amended columns was almost 9 times greater than the reduction rate (175 mol N m–2h–1) over the same period. Without the addition of urea the NO 3 production rate averaged 32 mol N m–2h–1 and reduction 101 mol N m–2h–1.  相似文献

Sixteen withanolides isolated from Iochroma gesnerioides (Kunth) Miers (Solanaceae) have been assessed for their activities as ecdysteroid agonists and antagonists. None of the compounds showed any agonistic activity, but several showed significant antagonistic activity. With a 20-hydroxyecdysone concentration of 5 × 10–8 M, the ED50 values for 2,3-dihydro-3-methoxywithaferin A, 2,3-dihydro-3-methoxywithacnistine, 2,3-dihydro-3-methoxyiochromolide and 2,3-dihydro-3-hydroxywithacnistine are 3.5 × 10-5 M, 1 × 10–5 M, 5 × 10–6 M and 2.5 × 10–6 M, respectively.  相似文献

A comparison has been made of cyclopropene (CP), 1-methylcyclopropene (1-MCP), and 3,3-dimethyl-cyclopropene (3,3-DMCP) in their ability to protect plants against ethylene. In bananas, both CP and 1-MCP are effective around 0.5 nL L–1, and 3,3-DMCP was effective at 1 L L–1. Bananas treated with CP and 1-MCP again become sensitive to ethylene at 12 days and those treated with 3,3-DMCP at 7 days. Mature green tomatoes are protected by 5–7 nL L–1 of 1-MPC for 8 days at 25°C and tomatoes treated with 3,3-DMCP at 5–10 L L–1 are protected for 5 days. Carnation flowers are protected with CP or 1-MCP after exposure to 0.5 nL L–1 for 24 hours and by 1 L L–1 of 3,3-DMCP. The display life of Campanula flowers is increased from 3.3 to 5.4 days by 10 L L–1 of 3,3-DMCP and to 9 days by 20 nL L–1 of 1-MCP. Ethylene inhibition of pea seedlings is reduced by treatment with 1-MCP at 10 L L–1 of ethylene but as ethylene is increased to 3000 L L–1 growth inhibition increases. 3,3-DMCP treatment causes very little reduction of the ethylene effect even at very low concentrations.  相似文献

The effects of eight microelements (I, BO3 3–, MoO4 2–, Co2+, Cu2+, Mn2+, Fe2+, Zn2+) on the biosynthesis of camptothecin and the growth of suspension cultures of Camptotheca acuminata were studied. The increase of I to 25 M l–1, Cu2+ to 1 M l–1, Co2+ to 2 M l–1 and MoO4 2– to 10 M l–1 in Murashige and Skoog (MS) medium resulted in 1.66, 2.84, 2.53 and 2.04 times higher of camptothecin yield than that in standard MS medium respectively. Combined treatment of I (25 M l–1), Cu2+ (1 M l–1), Co2+ (2 M l–1) and MoO4 2– (10 M l–1) lead to improve cell dry weight, camptothecin content, and camptothecin yield to 30.56 g l–1, 0.0299%, and 9.15 mg l–1, respectively, which were 20.2, 208.9 and 273.8% increment respectively when compared with those of control.  相似文献

Saccharomyces cerevisiae-based ethanol fermentations were conducted in batch culture, in a single stage continuous stirred tank reactor (CSTR), a multistage CSTR, and in a fermentor contaminated with Lactobacillus that corresponded to the first fermentor of the multistage CSTR system. Using a glucose concentration of 260 g l–1 in the medium, the highest ethanol concentration reached was in batch (116gl–1), followed by the multistage CSTR (106gl–1), and the single stage CSTR continuous production system (60gl–1). The highest ethanol productivity at this sugar concentration was achieved in the multistage CSTR system where a productivity of 12.7gl–1h–1 was seen. The other fermentation systems in comparison did not exceed an ethanol productivity of 3gl–1h–1. By performing a continuous ethanol fermentation in multiple stages (having a total equivalent working volume of the tested single stage), a 4-fold higher ethanol productivity was achieved as compared to either the single stage CSTR, or the batch fermentation.  相似文献

Candida sp. SY16 produces a glycolipid-type biosurfactant, mannosylerythritol lipid (MEL-SY16) which, at low concentrations, emulsifies hydrocarbons, vegetable oil, and crude oil. The molecular weight of MEL-SY16 was 634 Da and its hydrophilic lipophilic balance (HLB) was 8.8. It decreased the surface tension to 30 dyne cm–1 at a critical micelle concentration (CMC) of 15.8 M, and the minimum interfacial tension against kerosene (a mixture of petroleum hydrocarbons) was 0.1 dyne cm–1. MEL-SY16 was stable from pH 4 to pH 10, up to 90 °C for 1 h, and against NaCl up to 100 mM. It was readily degraded by microorganisms in activated sludge within a short time, and exhibited very low toxicity to mouse fibroblast cells. These properties confirm that MEL-SY16 is suitable for a variety of environmental applications.  相似文献

Molecular diffusion of solutes, like sucrose in the xanthan gum fermentation, is important in order to understand the complex behavior of mass transfer mechanisms during the process. This work was focused to determine the diffusion coefficient of sucrose, a carbon source for xanthan production, using similar sucrose and xanthan concentrations to those occurring in a typical fermentation. The diaphragm cell method was used in experimental determinations. The data showed that diffusion coefficient of sucrose significantly decreases when xanthan gum concentration increases. Theoretical and semiempirical models were used to predict sucrose diffusivity in xanthan solutions. Molecular properties and rheological behavior of the system were considered in the modeling. The models tested fitted well the behavior of experimental data and that reported for oxygen in the same system.List of Symbols A constant in eq. Isolated embryos ofKarwinskia humboldtiana were cultured in vitro. The growth of embryos and development to plantlets on woody plant medium supplemented with indole-3-acetic acid 6.10-2 mol l–1, gibberellic acid (GA3) 3.10-2 mol l–1, and 6-benzylaminopurine (BA) 2 mol l–1 was obtained. Multiplication of shoots and rooting of excised shoots has been achieved. Three-phase dose responses of biological systems of different levels of organization are often called paradoxical because the biological effects are clearly manifested under low- and high-intensity treatments, but are absent during moderate-strength treatments. In this work, we found anomalous changes in the cell number of a green alga Scenedesmus quadricauda (Turp.) Breb. grown in the presence of the fungicide imazalil sulfate. At low imazalil concentrations (2.5 × 10–9–2.5 × 10–6 M), the slow increase in the cell number as compared to an untreated culture was not related to cell death. As seen by the dynamics of the population structure and cell functional characteristics (photosynthesis, thermal stability of photosynthetic membranes, etc.), the decrease in the growth rate at low concentrations of imazalil (2–10 × 10–9 M) was due to a long-term arrest of cell division in a fraction of the cell population rather than to a decrease in the rate of division. The absence of a toxic effect or even a slight stimulation of culture growth at moderate concentrations (0.05–1.25 × 10–6 M) was due to the resumption of cell division after a temporal cessation. At these concentrations, imazalil induced cell stress and adaptive elevation of cell tolerance to the fungicide (acclimation). Cell death was observed only at a high fungicide content in the medium (6.25 × 10–6 and higher). Thus, the three-phase (bimodal) dose response corresponds to two regimes (steady-states) of cell functioning which differ in cell sensitivity to external stimuli. The low-sensitivity state, which is characteristic of cells that have experienced stress, is likely to be the state known as hormesis.  相似文献

Uptake kinetics of nitrogen derived from sewage–seawater mixtures (2.5–20% v/v effluent) were determined in the laboratory for Ulva rigida (Chlorophyceae) native from Bahía Nueva (Golfo Nuevo, Patagonia, Argentine). In terms of nitrogen concentration, experimental enrichment levels varied between 53.7 and 362.3M of ammonium and between 0.77 and 6.21M of nitrate+nitrite. Uptake rates were fitted to the Michaelis–Menten equation, with the following kinetic parameters: ammonium: Vmax = 591.2molg–1h–1, K s=262.3M, nitrate+nitrite: V max=12.9molg–1h–1, K s=3.5M). Both nutrients were taken up simultaneously, but ammonium incorporation was faster in all cases. The results show a high capability of Ulva rigida to remove sewage-derived nitrogen from culture media. In the field, most of the nitrogen provided by the effluent would be tied up in algal biomass, supporting low nitrogen levels found at a short distance away from the source.  相似文献

The Orbetello lagoon is now highly eutrophic and has experienced increasing incidence of anoxia causing serious economic damage. A multidisciplinary study was commenced in March 1987 to investigate the part played by the lagoon sediments and interstitial water in recycling nutrients and contributing to the observed anoxia. Eleven undisturbed cores were collected and sub-sampled at 2 cm intervals. Interstitial water was obtained by centrifugation and analyzed for pH, Eh, nutrients, major and some minor elements. Differential fluxes from the sediments of 0.2–10 g cm–2 day–1 of ammonia and 0.02–0.7 g cm–2 day–1 of orthophosphate were obtained depending on the season, temperature of the sediments and the sampling location. A highly significant linear correlation (r 2 = 0.86) was found between bicarbonate and ammonia concentrations in the interstitial waters, due to the release of these compounds during the degradation organic matter. A diagenetic model was developed to predict the alkalinity of interstitial water from the theoretical reactions involved in the decomposition of organic matter. The predicted values for bicarbonate and ammonia agreed well with the experimental results.  相似文献

Synaptoneurosomes isolated from cerebral cortices of male Sprague-Dawley rats were used for studying GABAA receptor-regulated chloride influx. The in vitro effects of GABA antagonists, SR 95531 (a pyridazinyl GABA derivative) and bicuculline, on pentobarbital-stimulated, muscimol-stimulated or flunitrazepam-enhanced, muscimol-stimulated chloride uptake were studied. The chloride uptake was determined at 30°C, for 5 sec. Pentobarbital and muscimol produced a maximal stimulation of chloride uptake in cortical synaptoneurosomes at 500 M and 50M, respectively. SR 95531 as well as bicuculline had no effect on the basal uptake of chloride. Whereas, SR 95531 (0.3–30 M) and bicuculline (0.1–100 M), when added 5 min before muscimol (50 M), produced a significant concentration-dependent inhibition of muscimol (50 M)-stimulated chloride uptake (IC50 s of 0.89±0.11 M and 13.45±2.10M, respectively). In studies of the inhibitory effects of SR 95531 and bicuculline on pentobarbital (500 M)-stimulated chloride uptake, the IC50 s were 0.81±0.12 M and 3.86±1.14 M, respectively. SR 95531 exhibited a more potent inhibitory effect than bicuculline on flunitrazepam-enhanced, muscimol-stimulated chloride uptake. The results revealed that SR 95531 has a more potent antagonistic effect than bicuculline on GABAA-regulated chloride flux.  相似文献

Adventitious shoot regeneration was observed using leaf-petiole explants from shoot-proliferating cultures of Comet red raspberry (Rubus idaeus L.). A maximum regeneration rate of 70% (3.7 shoots/explant) was obtained using 4.5–9.1 M (1–2 mg l–1) N-phenyl-N-1,2,3-thiadiazol-5-ylurea (thidiazuron or TDZ) with 2.5–4.9 M (0.5–1 mg l–1) 1H-indole-3-butanoic acid (IBA) or 2.3 M (0.5 mg l–1) TDZ with 4.9 M (1 mg l–1) IBA in modified Murashige-Skoog medium. TDZ was more effective than N-(phenylmethyl)-1H-purin-6-amine (BA) at promoting regeneration in combinations tested with IBA (maximum 50% regeneration rate; 1.8 shoots/explant). Variation in the agar concentration or incubation temperature, orientation or scoring of the leaf-petiole explants and use of separate leaf or petiole explants had no effect on shoot regeneration. Incubation in the dark for 1, 2 or 3 weeks prior to growth in the light did not influence the percent regeneration rate but depressed the number of adventitious shoots. Explant source, from micropropagated shoots or greenhouse-grown plants, had an effect on shoot regeneration that was genotype dependent. Only 8 of 22 (36%) raspberry cultivars were capable of regeneration from leaf explants derived from greenhouse-grown plants.  相似文献

Nitrate reduction in a non-polluted, coastal marine sediment was measured with an open flow-through system. The recorded rates depended upon nitrate concentration but were largely independent of the weight of sediment (14–35 g) and the dilution rate (0.7–5 h–1). Rate of nitrate uptake followed classical Michaelis-Menten kinetics, and km and Vmax values were equal to 78M and 0.168m mol g–1 hour–1, respectively. These values are in good agreement with those found by the other authors for the same biotope but by different methods. This technique of the open flow-through system is fast, simple, and inexpensive and involves small quantities of sediment (10 g).  相似文献

A tissue culture procedure was developed for the establishment and propagation of a colchi-autotetraploid of Rauvolfia serpentina for possible commercial exploitation. Multiplication of autotetraploid shoots was obtained either through axillary bud elongation on Murashige and Skoog [1] medium (MS) containing 2.65 M (0.5 mgl–1) -naphthaleneacetic acid and 0.33 M (0.05 mgl–1) kinetin, or via multiple shoot formation on MS medium supplemented with 4.44 M (1.0 mgl–1) 6-benzylaminopurine and 0.53 M (0.1 mgl–1) -naphthaleneacetic acid. Rooting could be induced by transferring the shoots to MS medium containing 7.95 M (1.5 mgl–1) -naphthaleneacetic acid alone. The plantlets, thus formed, were tetraploid in nature by cytological observations of the root tips. They exhibited 80–90% success in establishment under glass house and field conditions.  相似文献

The interaction of various hormones and regucalcin on (Ca2+–Mg2+)-ATPase activity in rat liver plasma membranes was investigated. The presence of epinephrine (10–6–10–4 M), and insulin (10–8–10 M) in the reaction mixture produced a significant increase in (Ca2+–Mg2+)-ATPase activity, while the enzyme activity was decreased significantly by calcitonin, (3×10–8–3×10–6 M). These hormonal effects, except for calcitonin, were clearly inhibited by the presence of vanadate (10–4 M) which can inhibit the Ca2+-dependent phosphorylation of enzyme. Meanwhile, regucalcin (0.25 and 0.50 M), isolated from rat liver cytosol, elevated significantly (Ca2+–Mg2+)-ATPase activity in the plasma membranes, although this elevation was not inhibited by vanadate (10–4 M). the epinephrine (10–5 M) or phenylephrine (10–4 M)-induced increase in (Ca2+–Mg2+)-ATPase activity was disappeared in the presence of regucalcin; in this case the effect of regucalcin was also weakened. However, the inhibitory effect of calcitonin (3×10–6 M) was not weakened by the presence of regucalcin (0.5 M). Moreover, GTP (10–5 and 10–4 M)-induced increase in (Ca2+–Mg2+)-ATPase activity was not seen in the presence of regucalcin (0.25 M). The present finding suggests that the activating mechanism of regucalcin on (Ca2+–Mg2+)-ATPase is not involved on GTP-binding protein which modulates the receptor-mediated hormonal effect in rat liver plasma membranes.  相似文献

E. coliwas used as a biocatalyst to compare electricity production, substrate consumption and growth in biofuel cells. With the native electrode 1.44 mV cm–2-electrode and 1.41 A cm–2-electrode electricity were produced and 21 mM acetate was consumed. With the modified electrode with Neutral Red, 3.12 mV cm–2-electrode and 3.1 A cm–2-electrode electricity were produced and 39 mM acetate was consumed.  相似文献

Antimicrobial activity of toxin produced by a freshwater bloom-forming cyanobacterium Microcystis aeruginosa has been studied. When tested against certain green algae, cyanobacteria, heterotrophic bacteria and fungi, the toxin inhibited growth of only green algae and cyanobacteria. The toxin has been partially purified employing Thin layer chromatography (TLC) and High-performance liquid chromatography (HPLC) techniques and appears to be microcystin-LR (leucine–arginine). Both crude and purified toxins showed toxicity to mice, the clinical symptoms in test mice being similar to those produced by hepatotoxin. Purified toxin at a concentration of 50 g ml–1 caused complete inhibition of growth followed by cell lysis in Nostoc muscorum and Anabaena BT1 after 6 days of toxin addition. Addition of toxin (25 g ml–1) to the culture suspensions of the Nostoc and Anabaena strains caused instant and drastic loss of O2 evolution. Furthermore a marked reduction (about 87%) in the 14CO2 uptake was also observed at a concentration of 50 g ml–1. Besides its inhibitory effects on photosynthetic processes, M. aeruginosa toxin (50 g ml–1) also caused 90% loss of nitrogenase activity after 8 h of its addition. Experiments performed with 14C-labelled toxin indicate that the toxin uptake by cyanobacterial cells occurs both in light and dark. These results demonstrate that the toxin is strongly algicidal and point to the possibility that it may have an important role in establishment and maintenance of toxic blooms of M. aeruginosa in freshwater ecosystems. The relative significance of the hepatotoxic effect and the algicidal effect of the toxin is discussed with reference both to survival and dominance of M. aeruginosa in nature.  相似文献
