It is well reported that the environmental factors along with different endocrine stimulus play a crucial role in maintenance of adrenocortical activity in birds. This study is first to report a detailed seasonal activity cycle of adrenal cortex, particu-larly its secretory physiology in a tropical nocturnal bird, Indian spotted owlet Athene brama. The maximum cortical activity having highest glandular mass, glandular free cholesterol, esterified cholesterol profiles, and peak level of corticosterone in plasma coincided with the long day length, highest temperature and increasing amplitude of relative humidity and rainfall of the early summer month, May. Cortical activity declined to minimum level in August when the ecofactors also declined parallely and hence, the birds entered into partial hibernation. The cortical activity progressed slowly throughout the winter (September–March) to reach maximum level in May. Further, the electron microscopic observations of cortical cell morphology strongly supported the above seasonal activity status of the gland revealing a comparatively large number of mitochondria during May than August, along with lipid filled vacuoles during May but not in August. Besides, assessment of gonadal and pineal hormones in relation with seasonal activity of adrenal cortex presented a parallel relationship with gonad while completely inverse relationship with pineal. Therefore, the study concludes that the seasonal adrenocortical activity of this tropical nocturnal bird might be regulated by multiple factors, particularly by the environmental temperature, humidity/rainfall and photoperiod along with the internal factors at least by gonadal and pineal hormones. 相似文献

The epiphyseal neurohormone melatonin (MEL) exhibits circadian cyclicity, as noted extensively in diurnal vertebrates although very little information is available regarding nocturnal species. We have studied the MEL rhythmicity with 24-hour periodicity in a tropical nocturnal bird, Indian spotted owlet Athene brama, which possesses a well-developed pineal organ. We performed our study during two crucial reproductive phases (active and inactive), when the pineal gland activity in owlet exists in inverse states, i.e., inactive and active respectively. Independent of sex, the circadian rhythm of plasma MEL in owlets showed a two-peak cyclicity with a smaller peak at around 1400 h and the higher one at about 0200 h, while the lowest value was found at 1000 h. The night (0200 h) peak activity of plasma MEL in owlet has a resemblance with the earlier findings in diurnal birds and strongly suggests that independent of species habit the peak activity of MEL is invariably dark dependent. However, the daytime peak of MEL may be due to the daytime hiding nature of this nocturnal bird. Interestingly, it was also noted that the hours of peak activity of MEL (1400 and 0200 h) were the same during both of the reproductive phases, though the environmental day length was longer and ambient temperature was higher during the reproductively inactive phase. During daytime these birds hide in a dark burrow where, in general, the intensities of light and temperature are less, and the amplitude of variations of these factors is not prominent. Hence, the seasonal variations in these oscillatory components may not have affected the entrainment of the owlet pineal oscillator, which regulates the daily MEL rhythm in a similar pattern during both the studied phases. On the other hand, a single circadian peak (around 1000 h) circadian cyclicity of gonadal steroids (i.e., testosterone in the male and estradiol and progesterone in the female) showed an inverse relationship with plasma MEL. Biometric analysis helps in sex differentiation, understanding development and for studies of avian biology such as foraging ecology, evolutionary ecology, and survivorship. We suggest that biometry can also be a reliable, practical and inexpensive tool to determine the age of nestlings in the field by non-invasive methods. As an example we studied the biometry of wing, culmen, talon, tarsus and body mass of nestling southern Indian Spotted Owlets (Athene brama brama). Based on the growth pattern analysis using logistic growth model, discriminant analysis and CHAID (Chi-squared Automatic Interaction Detection) based decision tree, we show that biometry of nestling Spotted Owlets is an easy, reliable and inexpensive method to determine nestling age and to assess growth rate and relative nutritional status. These biometric parameters also allow us to predict their ability to initiate first flight from the nest site. This method is described here for the first time and we postulate that such charts can be devised for other avian species as well, so as to assist conservation biologists and bird rescuers. 相似文献

It has been reported that owls (Strigiformes) do not have a pineal gland. However, our light microscopy study revealed an intermediate form of tubulofollicular and solid-type large pineal gland in a tropical owlet, Athene brama. The epithelial cells forming follicles (6-8) in the distal region and the solid cluster of parenchymal cells of different diameters in the proximal region anteriorly tapered with a long cylindrical stalk and continued into commissural organs and choroid plexus. The intrapineal localization of perivascular nerve fibers and blood vessels clearly explained the sympathetic innervation as well as vascularization of this neuroendocrine gland. Further, electron microscopy revealed a developed intracellular structure of the pinealocytes with a large number of mitochondria, Golgi bodies, and granular as well as clear vesicles in the process terminals. The evidence of intrapinealocyte lipid droplets and dense bodies and a moderate amount of melatonin in plasma (ranging from 100-365 pg/mL) during different reproductive phases finally proved a defined secretory activity of the gland in this tropical, nocturnal bird. 相似文献

Unlike other temperate owls, Indian spotted owlet Athene brama possesses a well-developed pineal gland that secrets moderate amount of hydroxy- (serotonin) and methoxy- (melatonin) indoles in circulation. However, in this study, we have reported the response of this endocrine gland to exogenous L-Tryptophan (precursor of the above indoles), and also its effect on gonads of this nocturnal bird. During breeding phase or pineal inactive phase (March), oral treatment of L-Trp (0.5 mg/100 g Bwt/day) significantly increased the pineal gland wt and plasma melatonin (MEL) level, while decreased the gonadal wt and plasma sex steroids levels (estradiol and progesterone in female and testosterone in male). Interestingly, during reproductively quiescent phase or pineal active phase (August), similar amount of L-Trp significantly decreased the plasma MEL level, while increased the above sex steroid levels in plasma. Finally, the results show a clear reproductive phase-dependent inverse effect of L-Trp on pineal gland and gonads for both sexes of the spotted owlets, and suggest that the therapeutic use of this amino acid would be a great advantage for controlling the reproduction of these economically important birds. 相似文献

In a tropical nocturnal bird, the Indian spotted owlet, Athene brama, the intraperitonial injection of an identical amount (20 mg/100 g b. wt/day) of exogenous melatonin (MEL) for 15 consecutive days increased the pineal weight and plasma MEL level in sexually active birds while it decreased them in inactive birds more potently when injected in the evening (18.30-19.30 h) rather than the morning (0500-0600 h). On the other hand, more efficiently than the morning hour treatment, the evening hour MEL injection decreased the ovary weight and plasma estradiol and progesterone levels both in sexually active and inactive birds, but more potently in active than inactive birds. Thus, the exogenous MEL showed the time and reproductive phase dependent effects on the pineal gland and the ovary of this nocturnal bird. 相似文献

In this study, starting from human dental pulp cells cultured in vitro, we simulated reparative dentinogenesis using a medium supplemented with different odontogenic inductors. The differentiation of dental pulp cells in odontoblast-like cells was evaluated by means of staining, and ultramorphological, biochemical and biomolecular methods. Alizarin red staining showed mineral deposition while transmission electron microscopy revealed a synthesis of extracellular matrix fibers during the differentiation process. Biochemical assays demonstrated that the differentiated phenotype expressed odontoblast markers, such as Dentin Matrix Protein 1 (DMP1) and Dentin Sialoprotein (DSP), as well as type I collagen. Quantitative data regarding the mRNA expression of DMP1, DSP and type I collagen were obtained by Real Time PCR. Immunofluorescence data demonstrated the various localizations of DSP and DMP1 during odontoblast differentiation. Based on our results, we obtained odontoblast-like cells which simulated the reparative dentin processes in order to better investigate the mechanism of odontoblast differentiation, and dentin extracellular matrix deposition and mineralization.Key words: dental tissue, in vitro differentiation, DMP1, DSP, type I collagen 相似文献

This study investigated whether changes in illumination modify perception of day and night conditions in a diurnal species, the Indian weaver bird. Birds were initially subjected to a 12-h light:12-h dark regime (12L:12D; L?=?20 lux, D =?0.5 lux). After every 2 wks, the combinations of light illumination in L and D phases were changed as follows: 20:2 lux, 20:5 lux, 20:10 lux, 20:20 lux, 20:100 lux, and 20:200 lux. Finally, birds were released into dim constant light (0.5 lux) for 2 wks to determine the phase and period of the circadian activity rhythm. They were also laparotomized at periodic intervals to examine the effects of the light regimes on the seasonal testicular cycle. All individuals showed a consistently similar response. As evident by the activity pattern under these light regimes, both in total activity during contrasting light phases and during the 2?h in the beginning and end of first light phase, birds interpreted the period of higher light intensity as day, and the period of lower intensity as the night. During the period of similar light intensity, i.e., under LL, birds free-ran with a circadian period (~24?h). In bright LL (20 lux), the activity rhythm was less distinct, but periodogram analysis revealed the circadian period for the group as 24.46?±?0.41?h (mean?±?SE). However, in dim LL at the end of the experiment, all birds exhibited a circadian pattern with average period of 25.52?±?0.70?h. All birds also showed testicular growth and regression during the 16-wks study. It is suggested that weaver birds interpret day and night subjectively based on both the light intensity and contrast between illuminations during two phases over the 24?h. Perinatal immune challenge leads to neurodevelopmental dysfunction, permanent immune dysregulation and abnormal behaviour, which have been shown to have translational validity to findings in human neuropsychiatric disorders (e.g. schizophrenia, mood and anxiety disorders, autism, Parkinson’s disease and Alzheimer’s disease). The aim of this animal study was to elucidate the influence of early immune stimulation triggered by systemic postnatal lipopolysaccharide administration on biochemical, histopathological and morphological measures, which may be relevant to the neurobiology of human psychopathology. In the present study of adult male Wistar rats we examined the brain and plasma levels of monoamines (dopamine, serotonin), their metabolites, the levels of the main excitatory and inhibitory neurotransmitters glutamate and γ-aminobutyric acid and the levels of tryptophan and its metabolites from the kynurenine catabolic pathway. Further, we focused on histopathological and morphological markers related to pathogenesis of brain diseases - glial cell activation, neurodegeneration, hippocampal volume reduction and dopaminergic synthesis in the substantia nigra. Our results show that early immune stimulation in adult animals alters the levels of neurotransmitters and their metabolites, activates the kynurenine pathway of tryptophan metabolism and leads to astrogliosis, hippocampal volume reduction and a decrease of tyrosine hydroxylase immunoreactivity in the substantia nigra. These findings support the crucial pathophysiological role of early immune stimulation in the above mentioned neuropsychiatric disorders. 相似文献

对反交试验获得的18个月的拟鲤、粗鳞鳊和欧鳊F1代进行了鉴定.对不同于拟鲤、粗鳞鳊和欧鳊三种鱼的可量和可数形态特征分析表明,拟鲤×欧鳊和拟鲤×粗鳞鳊杂交种比粗鳞鳊×欧鳊杂交种与它们的双亲更容易区分.拟鲤杂交种间具有很多形态上的相似性,例如,上侧虹膜在拟鲤为红色,杂交种则介于双亲之间.在粗鳞鳊×欧鳊杂交种中也可以观察到居间类型.拟鲤×粗鳞鳊和拟鲤×欧鳊各自的杂交种可以通过眼睛的大小、较少的侧线鳞数和2行咽喉齿等特征相区分.粗鳞鳊×欧鳊杂交种与另外两个杂交种相比,具有更多的臀鳍分支鳍条数和一个中央具一黑线的虹斑.三个种进行杂交再获得的互交种通常不易区分. 相似文献

Biochemical and morphological studies were done on a new trembling mutant hamster CBB. The yield of myelin from the mutant was 30 and 40% of the control at 46 and 140 days of age, respectively, but myelin composition and 2',3'-cyclic nucleotide-3'-phosphohydrolase (CNPase) activity were normal. Morphologically, about 18% of the axons were myelinated in the mutant optic nerve at 46 days of age, in which the myelinated fibers were those with larger diameters (more than 0.6 micron), while the control had a peak at 0.4 micron in diameter. The ultrastructure and thickness of compact myelin lamellae in the mutant were normal. Myelination and the structure of peripheral nerve myelin appeared normal. The results indicate that the essential defect is the delay and arrest of myelination in the CNS, which is probably caused by either a decreased rate of synthesis of myelin components in oligodendrocytes or a defect in the oligodendrocyte-axon recognition in smaller axons. 相似文献

We describe a simple, rapid, and efficient method, based on separation on a Percoll centrifugation gradient, to purify glial progenitor cells from newborn rat brains. Cytofluorimetry analysis of the isolated cell population showed that 75 +/- 8 and 86 +/- 7% of the cells were A2B5- and R24-positive, respectively. Transmission electron microscopy examination of the purified cell population confirmed their homogeneity and illustrated their typical morphology, as previously described in situ. Assay of UDP-galactose-ceramide galactosyltransferase, 3'-phosphoadenosine 5'-phosphosulfate galactosylceramide sulfotransferase, and 2',3'-cyclic nucleotide 3'-phosphohydrolase activities showed that the levels of these enzymes were 446, 76, and 11 times lower, respectively, than the levels measured in mature oligodendrocytes. Low levels of mRNA coding for 2',3'-cyclic nucleotide 3'-phosphohydrolase and myelin proteolipid protein, but not for myelin basic protein, were present in the glial progenitor cells. At the time of isolation, 40% of the cells in the population were dividing, and the cells could easily be expanded in culture. After 3 weeks of culture in the presence of 1% fetal calf serum, 75% of the cells had differentiated into galactosylceramide-positive oligodendrocytes. When the culture took place in the presence of 10% fetal calf serum, only 2% of the cells expressed galactosylceramide, and 60% were glial fibrillary acidic protein-positive astrocytes; half of them were also A2B5 positive. 相似文献

Studies were conducted on three strains of Pseudomonas cepacia isolated and maintained in distilled water and on a laboratory-subcultured strain transferred to distilled water. Optimum growth rates and maximum population yields of the four strains in distilled water were obtained at 37 C, although high population levels (10(6)-10(7)/ml) were reached and maintained over extended incubation periods at temperatures from 18 C to 42 C. Two strains were able to grow in distilled water at temperatures ranging from 12 C to 48 C and to survive 48 h and 21 days at 50 C and 10 C, respectively. Cells from distilled water cultures inoculated into Trypticase soy broth showed an immediate two- to three-log drop at upper and lower temperature limits; survivors were able to initiate logarithmic growth. Results obtained in morphological, biochemical, and antibiotic tests affirmed the strain differences noted in growth studies. 相似文献

Phycovirus populations were found in 11 of the 12 waste stabilization ponds studied. These populations were comprised solely of blue-green algal (BGA) viruses. Two virus types were observed, one of which was related to the previously reported LPP-1 virus. The incidence and magnitude of the LPP group indicated that several of the ponds supported well-established BGA virus populations of this type. Counts as high as 270 plaque-forming units/ml were noted; however, marked differences in the nature and magnitude of these BGA viruses were apparent even in geographically related ponds of similar design. Of the algal strains found dominant in these ponds, none was of the type reported susceptible to the LPP viruses. 相似文献
