Survival of infective juveniles of Steinernema carpocapsae and Steinernema glaseri gradually declined during 16 weeks of observation as the tested soil pH decreased from pH 8 to pH 4. Survival of both species of Steinernema dropped sharply after 1 week at pH 10. Survival or S. carpocapsae and S. glaseri was similar at pH 4, 6, and 8 during the first 4 weeks, but S. carpocapsae survival was significantly greater than S. glaseri at pH 10 through 16 weeks. Steinernema carpocapsae and S. glaseri that had been stored at pH 4, 6, and 8 for 16 weeks, and at pH 10 for 1 or more weeks were not infective to Galleria mellonella larvae. Steinernema carpocapsae survival was significantly greater than that of S. glaseri at oxygen:nitrogen ratios of 1:99, 5:95, and 10:90 during the first 2 weeks, and survival of both nematode species declined sharply to less than 20% after 4 weeks. Survival of both nematode species significantly decreased after 8 weeks as the tested oxygen concentrations decreased from 20 to 1%, and no nematode survival was recorded after 16 weeks. Steinernema carpocapsae pathogenicity was significantly greater than that of S. glaseri during the first 2 weeks. No nematode pathogenicity was recorded at oxygen concentrations of 1, 5, and 10% after 2 weeks and at 20% after 16 weeks.  相似文献

On agar media supplemented with NaCl (5% = 856 mM), seven Trichoderma species aggregates and an isolate intermediate between T. harzianum and T. viride (T. 410) grew more slowly, sporulated poorly, and there was no characteristic pigmentation of the colonies. NaCl or KCl concentrations inducing 'albinization' differed with different isolates. CaCl2 (90-270 mM) reversed the anti-sporulating effect of NaCl (856 mM) on T. 410, and stimulated conidiation in media with no NaCl added. Gliocladium virens but not G. roseum, reacted like Trichoderma to NaCl. Fourteen other fungal genera offered various reactions. Since 856 mM NaCl decreases osmotic potential (eta) of the media used from-1.2 to -41 bars, the influence of eta on T. 410 was examined. After 3 to 4 days, maximal growth was observed between-1.2 and -10 bars. Growth was reduced to 50% at about -30 bars. No growth occurred at about -90 bars. No pigmentation was observed at -32 bars (NaCl) and -41 bars (KCl). Abundant sporulation and pigmentation occurred at -31 bars with CaCl2 as sole electrolyte added. Na+ was toxic at high concentration. Results are discussed in view of possible use of Trichoderma in biological control.  相似文献

Streptomyces thermoviolaceus was grown in a chemostat under conditions of glutamate limitation. The effects of growth rate on production of the antibiotic granaticin, extracellular protein and protease activity as components of secondary metabolism were studied at 37, 45 and 50 degrees C. The amount of each secondary metabolite synthesized was highly dependent on growth rate and temperature. Granaticin yields were highest at growth rates of 0.1 to 0.15 h-1 at 37 degrees C, 0.175 h-1 at 45 degrees C and 0.045 h-1 at 50 degrees C. Protease activity of culture supernatants responded to low nutrient concentration and/or low growth rate. Measurements of extracellular protein revealed complex changes in amount which were dependent on growth rate and temperature. At 45 degrees C and a growth rate of 0.15 h-1, biomass yield was highest between pH 5.5 to 6.5 whereas granaticin synthesis was low at pH 5.5 and rose to highest values at between pH 6.5 and 7.5.  相似文献

Amino acids, salts, and vitamins were combined with dextrose to test their effect on growth and sporulation of Entomophthora virulenta in liquid shake culture. The addition of a vitamin solution to the tested media did not enhance growth or sporulation. MgSO4·7H2O was the only salt individually tested that allowed for good growth and sporulation. MgSO4·7H2O concentrations exceeding 250 mg/liter in media lacking other salts inhibited sporulation. A simple medium of l-arginine, l-leucine, glycine, and mineral salts allowed high growth and sporulation.  相似文献

Cellulose is a major component of the extracellular coat that surrounds the terminally-differentiated spore of Dictyostelium. It is sandwiched between two layers of proteins that derive from prespore vesicles by exocytosis. Strains unable to synthesize cellulose due to null mutations in the gene encoding the catalytic subunit of cellulose synthase (dcsA) failed to make detergent-resistant spores but produced small, highly refractile, round spore-like cells up to a day late. Although these cells resembled spores in appearance, they were unstable, only transiently ellipsoid in shape, and sensitive to hypo-osmotic shock, drying, or detergents. Differentiation of these pseudo-spores was induced in the normal time frame by activation of the cAMP-dependent protein kinase or co-development with wild type cells, and coat proteins were secreted by the dcsA-null cells at the same time as wild type cells. A substantial fraction of secreted coat proteins was loosely associated with the surface of the mutant cells, resembling the precoat posited to form early during normal sporulation. Transmission electron microscopy revealed that the precoat had little ultrastructural organization in the absence of cellulose. Thus, cellulose in the coat appears to be required for the organization of the pre-coat precursors as well as the stability, dormancy, and shape of the spore.  相似文献

The ability of 13 strains of Salmonella , representing 12 serotypes, to grow in a tryptone-yeast extract-glucose medium, acidified with HC1 to pH values between 3.80 and 5.60 at intervals of 0.20 units, has been investigated. During incubation at 30°C, growth occurred at minimum pH values of 3.8–4.0 in 1–3 d. At 20°C, growth occurred at minimum pH values of 3.8–4.2 in 3–5 d. In tests incubated at 10°C, growth occurred at minimum pH values of 4.4–4.8 in between 10 and 19 d.  相似文献

Most media in which the growth of shaken submerged cultures of Streptomyces akiyoshiensis was examined did not support the formation of well-dispersed mycelial suspensions. Investigation of the culture conditions promoting dispersed growth showed the pH of the culture medium to be of critical importance; an initial value of 5.5 minimized aggregation of the mycelium while supporting adequate biomass production. In cultures started at this pH, spore inocula gave better mycelial dispersal than did vegetative inocula; with spore inocula, growth morphology was also less affected by inoculum size. The composition of the nutrient solution influenced the extent of mycelial dispersal; slow growth was often associated with clumping but no clear correlation was observed between pellet formation and the ability of carbon or nitrogen sources to support rapid growth. Increasing the phosphate concentration from 0.5 to 15 mM caused a modest decrease in mycelial aggregation. Conditions promoting a well-dispersed mycelium suitable for studying the physiological control of secondary metabolism also supported the formation of 5-hydroxy-4-oxonorvaline by S. akiyoshiensis.  相似文献

Twenty streptomycete strains were isolated from marine sediment samples collected from Nabq area, Sharm El-Sheikh, Red Sea Coast, Egypt. Four of them produce exopolysaccharides (EPS) showing marked in vitro antitumor activities. Morphological and cultural characteristics of the most significant strain (No. 3) were shown. Moreover, the sequence of this strain showed similarity with Streptomyces carpaticus. The results reveal that EPS produced by Streptomyces carpaticus No. 3 had high cytotoxicity reaching 51.7% and 59.1% against human tumor cells of breast and colon lines respectively. A chemical analysis of EPS indicated that the composing monosaccharides were galactouronic acid, glucose, xylose, galactose, mannose, and fructose with relative ratio of 3:1:1:2:2:1 respectively, with an average molecular weight (Mw) 1.180 × 105?g/mol and of a number average molecular weight (Mn) 1.052 × 105?g/mol. Also the EPS contained uronic acid (0.5072%) and monosaccharide sulphates (21.753%).  相似文献

Distinct morphological changes in the ultrastructure of Sarcina ventriculi were observed when cells were grown in medium of constant composition at pH extremes of 3.0 and 8.0. Transmission electron microscopy revealed that at low pH (less than or equal to 3.0) the cells formed regular packets and cell division was uniform. When the pH was increased (to greater than or equal to 7.0), the cells became larger and cell division resulted in irregular cells that varied in shape and size. Sporulation occurred at high pH (i.e., greater than or equal to 8.0). The sporulation cycle followed the conventional sequence of development for refractile endospores, with the appearance of a cortex and multiple wall layers. The spores were resistant to oxygen, lysozyme, or heating at 90 degrees C for 15 min. Spores germinated within the pH range of 4.6 to 7.0.  相似文献

The individual and synergistic potentials of two Streptomyces strains and compost for growth promotion and biocontrol against dumping off caused by Rhizoctonia solani on pepper were evaluated. Diflubenzuron, an insect growth regulator that blocks chitin deposition in insect cuticles, was tested for its effects on morphogenesis of Streptomyces spp. Use of diflubenzuron resulted in reduced dominance of spore hairs, reduced the width of the outer wall, and prevented formation of the inner spore wall in S. bambergiensis. In S. coelicolor, diflubenzuron altered the structure of the fibrillar pattern of spore envelopes. Exposure to diflubenzuron resulted in small increases in exported protein and in a ca. 20% increase in chitinase in both Streptomyces spp.  相似文献

Relationships between the growth of certain fungi isolated from city waste and pH and temperature were examined by two methods. The tested isolates showed their maximum growth and sporulation at different pHs while temperature requirements were the same (28°C), except forHumicola grisea (43°C).Cladosporium herbarum andH. grisea showed double pH optima. The ranges of pH and temperature for sporulation were more limited than those for the vegetative growth. Although all the tested isolates showed wide tolerances to pH and temperature, the degree of tolerance varied with the isolates. Caulerpa taxifolia, an invasive species elsewhere in the world, is native to Moreton Bay where its distribution has been increasing in recent years. In Australia, dense beds of C. taxifolia are predominantly found in areas of low light and high nutrients (low water quality). Monitoring data from Moreton Bay suggests that native C. taxifolia is not directly replacing seagrass, but that there is a successional trend of seagrass loss and subsequent C. taxifolia colonization. The current study examined responses of C. taxifolia in relation to changes in environmental conditions using ambient water quality and a light/nutrient manipulative experiment. In the ambient water quality experiment we found that C. taxifolia grew significantly faster in areas with higher light (lower turbidity). The manipulative experiment demonstrated that nutrients stimulate C. taxifolia growth, however, light availability and seasonality appear to influence the response of C. taxifolia growth to nutrients in Moreton Bay. These findings suggest that C. taxifolia is unlikely to colonize seagrass beds in areas with high light and low nutrients; however, in areas with moderate light and moderate to high nutrients C. taxifolia and seagrass are likely to coexist.  相似文献

Summary Differences in maternal investment and initial offspring size can have important consequences for offspring growth and development. To examine the effects of initial size variability in the frogBombina orientalis, we reared larvae (N=360) in one of two treatments representing different levels of environmental quality. We used snout-vent length at the feeding stage (stage 25, Gosner 1960) as a measure of maternal investment. In a “low quality” treatment, larvae were reared with two conspecific tadpoles and food was limited, whereas in a “high quality” treatment, larvae were reared individually and were fed ad libitum. Among tadpoles reared in the low quality treatment, individuals that were initially small had smaller body sizes through metamorphosis and longer larval periods than individuals that were initially large. Among tadpoles reared in the high quality treatment, initial size had only a weak influence on later larval size, and did not significantly affect metamorphic size of the duration of the larval period. This interaction between maternal investment and rearing conditions suggests that production of initially small offspring could be advantageous if these offspring develop in relatively benign environments, but disadvantageous if environments are more severe. These findings are discussed in light of previous studies that have demonstrated such interactions in organisms with complex life cycles.  相似文献

A number of studies show that predators can depress prey growth rates by inducing reductions in foraging activity, but the size of this non-lethal effect is quite variable. Here I investigate how prey density and resource productivity may alter the extent to which predators depress the growth rates of their prey. Theory predicts that when resources are overgrazed, an increase in predation risk will have little net effect on individual food intake because the decline in foraging effort will be offset by an increase in resource level. Thus, the non-lethal effects of predators on prey growth rates should depend upon prey density and resource productivity in a predictable manner, with the growth penalty imposed by predators being strongest when resources are undergrazed and weakest when resources are overgrazed. I tested this hypothesis by manipulating predation risk, prey density, and nutrient additions in a mesocosm experiment with the pulmonate snail Helisoma trivolvis . Refuge use by snails was 45% higher in the presence of caged crayfish than in their absence. Snail growth rates were reduced, on average, by 24% in the presence of caged crayfish. However, the magnitude of the growth penalty exacted by crayfish depended on snail density and nutrient additions. When snails were stocked at high density and nutrient additions were low, growth suppression was just 2.6%. At the other extreme, when snails were at low density and nutrient additions were high, growth suppression was 44.6%. Thus, the non-lethal effects of predators on prey growth depend on environmental context, illustrating an important link between individual traits and system-level properties.  相似文献

The dependence of Escherichia coli membrane H+ conductance (Gm H+) with a steady-state pH in the presence and absence of an external source of energy (glucose) was studied, when cells were grown under anaerobic and aerobic conditions, with an assay pH of 7.0. Energy-dependent H+ efflux by intact cells growing at pH of 4.5-7.5 was also measured. The elevated H+ conductance and lowered H+ flux were shown for cells growing in acidic pH and under anaerobic conditions, when bacteria were fermenting glucose. The atp mutant, which is deprived of the F0F1- adenosine triphosphatase, had less Gm H+ independent of growth conditions. In contrast with wild-type or precursor strain, a remarkable difference in Gm H+ for atp mutant was observed between aerobic and anaerobic conditions; such a difference was significant at pH 4.5. These results could indicate distinguishing pathways determining Gm H+ under anaerobic conditions after the fermentation of glucose at different pH and an input of the F0F1-adenosine triphosphatase in Gm H+. In addition, the effect of osmotic stress was demonstrated with grown cells. Gm H+ and H+ efflux both were increased after hyperosmotic stress at pH 7.5, and these changes were inhibited by N,N\'-dicyclohexylcarbodiimide, whereas these changes were lower in atp mutant. A role of the F0F1-adenosine triphosphatase in osmo-sensitivity of bacteria was confirmed under fermentative conditions.  相似文献
