Under laboratory conditions, the development of larvae of the sand dollar Scaphechinus mirabilis (Agassiz) took 28.5–29 days from fertilization to settling and the end of metamorphosis at a temperature 20°C and salinity 32.2–32.6‰ The cleavage divisions were completed in 12 hours after fertilization (AF) by the release of free swimming ciliary blastula from the egg membrane. The larvae attained pluteus I stage with one pair of arms at an age of 40 hours. In 4.5–5 days the pluteus II stage with three pairs of arms, and in 9 days the pluteus III stage with four pairs of arms were formed. On the 20–21st day AF the larvae developed a vestibule, in which the adult skeleton rudiments, spicules, plates and pedicellariae were formed on the 26–27th day AF. The size of the larvae at an age of 22.5 days was 1064.3 ± 44.7 μm. The settling of larvae was recorded on the 28–29th day of development. Most of the larvae completed their metamorphosis in 4.5–5 hours after settling.  相似文献

The reproductive biology of the sand dollar Peronella lesueuri was studied between 2009 and 2011 in Cockburn Sound, a large coastal embayment in south-western Australia. Individuals of P. lesueuri did not display sexual size-dimorphism, and the population was found to have a sex ratio of 1:1. Maturity occurred over the range of 80–115 mm with all sand dollars larger than 115 mm having distinct gonads. Monthly histological analysis of gonads and changes in oocyte proportions over time indicated that P. lesueuri has an annual reproductive cycle; gametogenesis occurs in spring and spawning in summer. Differences in the rate of gametogenesis between 2009/2010 and 2010/2011 may have been influenced by higher temperatures experienced in 2010/2011. P. lesueuri have large ova (mean?=?210 μm), which suggests the species has lecithotrophic larval development.  相似文献

The Pax6 genes eyeless (ey) and twin of eyeless (toy) are upstream regulators in the retinal determination gene network (RDGN), which instructs the formation of the adult eye primordium in Drosophila. Most animals possess a singleton Pax6 ortholog, but the dependence of eye development on Pax6 is widely conserved. A rare exception is given by the larval eyes of Drosophila, which develop independently of ey and toy. To obtain insight into the origin of differential larval and adult eye regulation, we studied the function of toy and ey in the red flour beetle Tribolium castaneum. We find that single and combinatorial knockdown of toy and ey affect larval eye development strongly but adult eye development only mildly in this primitive hemimetabolous species. Compound eye-loss, however, was provoked when ey and toy were RNAi-silenced in combination with the early retinal gene dachshund (dac). We propose that these data reflect a role of Pax6 during regional specification in the developing head and that the subsequent maintenance and growth of the adult eye primordium is regulated partly by redundant and partly by specific functions of toy, ey and dac in Tribolium. The results from embryonic knockdown and comparative protein sequence analysis lead us further to conclude that Tribolium represents an ancestral state of redundant control by ey and toy.  相似文献

We investigated the inductive signals originating from the vegetal blastomeres of embryos of the sand dollar Peronella japonica, which is the only direct developing echinoid species that forms micromeres. To investigate the inductive signals, three different kinds of experimental embryos were produced: micromere-less embryos, in which all micromeres were removed at the 16-cell stage; chimeric embryos produced by an animal cap (eight mesomeres) recombined with a micromere quartet isolated from a 16-cell stage embryo; and chimeric embryos produced by an animal cap recombined with a macromere-derived layer, the veg1 or veg2 layer, isolated from a 64-cell stage embryo. Novel findings obtained from this study of the development of these embryos are as follows. Micromeres lack signals for endomesoderm specification, but are the origin of a signal establishing the oral–aboral (O–Ab) axis. Some non-micromere blastomeres, as well as micromeres, have the potential to form larval skeletons. Macromere descendants have endomesoderm-inducing potential. Based on these results, we propose the following scenario for the first step in the evolution of direct development in echinoids: micromeres lost the ability to send a signal endomesoderm induction so that the archenteron was formed autonomously by macromere descendants. Recent DNA sequence analyses indicated that Nigrosabulum globosum is a cleistothecial representative of the Bionectriaceae in the Hypocreales, but morphological characters supporting this relationship are unknown. Using light and electron microscopy we followed the development of the ascomata of this species, from the formation of gametangia through to the development of mature ascospores, and observed a series of characters that confirmed its hypocrealean affinities. These included the formation of a gel-filled centrum during early stages of ascoma development, the subsequent appearance of hyaline peridial tissue enclosed within a layer we interpret as representing a melanized uniloculate stroma, apically derived paraphyses, and an ascogenous system that gives rise to asci that were both cylindrical to clavate and globose. Ascospores, previously reported to be smooth, were ornamented with a honeycomb-like reticulum and were able to germinate within the ascoma. The carbonaceous outer (stromatic) walls of the mature, grit-like cleistothecia indicate possible resistance to UV radiation and desiccation. Furthermore, the complement of germinated ascospores would enable mature ascomata to function as propagules that could quickly initiate new growth when transferred to fresh substrate. Our reexamination of N. globosum also provides data that support the hypothesized close relationship with other bionectriaceous, cleistothecial coprophiles, i.e., species of Hapsidospora, and Bulbithecium in particular.  相似文献

Golovinomyces is a strictly herb-parasitic genus in the Erysiphaceae. Host–parasite co-speciation was reported recently between the genus Golovinomyces and Asteraceae from molecular phylogenetic analyses. The Asteraceae originated in South America and latterly expanded their geographic distribution into the Northern Hemisphere. If the co-speciation between Golovinomyces and Asteraceae originated in South America, the geographic origin of Golovinomyces could be assumed to be South America. To address this question, Golovinomyces species from hosts of the tribe Mutisieae, an asteraceous tribe endemic to South America, were collected and the ITS and 28S rDNA regions sequenced. Results indicate that Oidium mutisiae and Golovinomyces leuceriae isolated from the Mutisieae do not belong at the base of the Golovinomyces tree. Instead, they are situated separately within two different clades of Golovinomyces isolates from the Northern Hemisphere. Therefore, the tribe Mutisieae is not the most early host of Golovinomyces. Present results suggest that Golovinomyces originated in the Northern Hemisphere, and not in South America. The new species Oidium reginae for the previous O. mutisiae on Mutisia decurrens is proposed.  相似文献

The sand dollar Dendraster excentricus is a facultative suspension feeder, which is relatively rare within the Order Clypeasteroida. Field studies of regulation of its feeding mode have been mostly conducted in exposed coastal habitats, where they are typically observed in an inclined position and oriented parallel to each other during suspension feeding. Physical (current speed and direction, reduction of drag and lift) and biological factors (increased efficiency of capture of particulates, and density) have been associated with regulation of its feeding mode in exposed coastal systems. We simultaneously measured the role of physical and biological factors in regulating the selection mode for feeding under varying tidal conditions in a shallow estuary in Baja California, Mexico. We used photographic records and direct sampling in fixed plots to determine the relationship between feeding behavior and environmental conditions. Current direction and speed, tidal level, density and content of organic matter in the water column and sediments were measured with respect to feeding mode (prone or inclined) and orientation relative to prevailing currents during spring and neap tides. Multiple regression analysis indicated that the percentage of inclined sand dollars was strongly and positively correlated only with tidal level at the densities found in the estuary (mean < 180 individuals m− 2); there was no relationship with current velocity, density, and organic matter content of the water. The prone position, indicative of deposit feeding, was largely limited to low tidal levels. We used circular statistics to determine whether the orientation of inclined sand dollars was correlated with current direction and speed. Sand dollars were only oriented parallel to the prevailing currents during the strongest currents of spring tides (> 20 cm s− 1). We did not observe the predominant oral:aboral configuration found in exposed coastal systems, which may be attributed to the relatively low densities of sand dollars in the exposed coastal environment (30-180 individuals m− 2), compared to protected coastal habitats (up to 1000 individuals m− 2). Our results suggest that regulation of the feeding mode of sand dollars in shallow and hydrodynamically complex estuarine systems differs from the feeding mode found in exposed coastal environments.  相似文献

(R)-Mandelonitrile was successfully synthesized by an enzymatic transcyanation reaction of benzaldehyde and acetone cyanohydrin catalyzed by a hydroxynitrile lyase from Eriobotrya japonica (EjHNL) in an aqueous-organic biphasic system. The effects of pH, temperature, organic solvent, substrate concentration and enzyme concentration on the initial activity and enantioselectivity of the enzyme were studied. Both pH and temperature had a large effect on the initial velocity and enantiomeric excess (e.e.) of the product, (R)-mandelonitrile. High enantiomeric purity of the product was observed at low pH and temperature because the non-enzymatic reaction producing racemates of mandelonitrile was almost suppressed. The optimum pH and temperature to obtain high e.e. were pH 4.0 and 10 °C, respectively. Surprisingly, the organic solvents had a significant influence on the initial velocity of the reaction but less influence on the enantiomeric purity of product. The EjHNL was very stable in ethyl acetate, diethyl ether, methyl-t-butyl ether, diisopropyl ether, dibutyl ether and hexane for 12 h. The best solvent for the highest initial velocity and e.e. was diethyl ether with an optimum aqueous phase content of 50% (v/v). The initial reaction rate increase as the aqueous phase content rose, but when the content was more than 50%, a reduction of e.e. was observed. Increasing the concentration of the substrates accelerated the initial velocity, but caused a slight decrease in the e.e. of the product. Under the optimized conditions, the conversion and e.e. of (R)-mandelonitrile for 3 h were 40 and 99%, respectively. The aqueous phase containing the enzyme also showed considerably efficient reusability for 4 batch reactions.  相似文献

The purple sea urchin, Strongylocentrotus purpuratus, possesses a non-adaptive immune system including elements homologous to C3 and factor B (Bf) of the vertebrate complement system. SpBf is composed of motifs typical of the Bf/C2 protein family. Expression of Sp152 (encodes SpBf) was identified in the phagocyte type of coelomocyte in addition to gut, pharynx and esophagus, which may have been due to the presence of these coelomocytes in and on all tissues of the animal. Sp152 expression in coelomocytes was constitutive and non-inducible based on comparisons between pre- and post-injection with lipopolysaccharide or sterile seawater. The pattern of five short consensus repeats (SCRs) in SpBf has been considered ancestral compared to other deuterostome Bf/C2 proteins that contain either three or four SCRs. Three alternatively spliced messages were identified for Sp152 and designated Sp1521, Sp1524, and Sp1521+4, based on which of the five SCRs were deleted. Sp1524 had an in-frame deletion of SCR4, which would encode a putative SpBf4 protein with four SCRs rather than five. On the other hand, both Sp1521 and Sp1521+4 had a frame-shift that introduced a stop codon six amino acids downstream of the splice site for SCR1, and would encode putative proteins composed only of the leader. Comparisons between the full-length SpBf and its several splice variants with other Bf/C2 proteins suggested that the early evolution of this gene family may have involved a combination of gene duplications and deletions of exons encoding SCRs.  相似文献

Results of molecular studies regarding the phylogenetic placement of the order Ostropales and related taxa within Lecanoromycetes were thus far inconclusive. Some analyses placed the order as sister to the rest of Lecanoromycetes, while others inferred a position nested within Lecanoromycetes. We assembled a data set of 101 species including sequences from nuLSU rDNA, mtSSU rDNA, and the nuclear protein-coding RPB1 for each species to examine the cause of incongruencies in previously published phylogenies. MP, minimum evolution, and Bayesian analyses were performed using the combined three-region data set and the single-gene data sets. The position of Ostropales nested in Lecanoromycetes is confirmed in all single-gene and concatenated analyses, and a placement as sister to the rest of Lecanoromycetes is significantly rejected using two independent methods of alternative topology testing. Acarosporales and related taxa (Acarosporaceae group) are basal in Lecanoromycetes. However, if the these basal taxa are excluded from the analyses, Ostropales appear to be sister to the rest of Lecanoromycetes, suggesting different ingroup rooting as the cause for deviating topologies in previously published phylogenies.  相似文献

采用光镜、透射电镜和细胞化学技术,对紫萁孢子囊发育过程中孢壁的超微结构和孢子囊内多糖和脂滴的分布及其动态变化进行研究,以探讨紫萁孢子囊发育过程中多糖和脂滴的代谢特征,为蕨类孢子发生的研究提供基础资料。结果表明:(1)紫萁孢子囊由1层囊壁细胞、2层绒毡层和产孢组织构成。(2)紫萁孢子壁由发达而分2层的外壁(外壁内层和外壁外层)和薄的不连续的周壁构成,由外壁形成棒状纹饰的轮廓;孢子外壁内层由多糖类物质构成,外壁外层和周壁均含有脂类物质。(3)在紫萁孢原细胞中观察到少量脂滴;随着紫萁孢壁的形成,囊壁细胞中淀粉粒的大小逐渐变小、数目先增加后减少,它们转运到内层绒毡层原生质团并转化为孢粉素前体物质,再穿过原生质团内膜表面进入囊腔,成为孢粉素团块或以小球形式填加到孢子表面形成孢壁。(4)紫萁孢子囊将多糖类营养物质转化为脂类,以脂滴的形式储藏在孢子中。  相似文献

Kenaf(Hibiscus cannabinus)is a fast growing annual with tremendous potential as a source of fiber for ropes, textiles and paper. Kenaf is an environmentally friendly crop; however, commercial production of kenaf is hindered by weed competition at the seedling stage. Herbicide resistant kenaf cultivars would reduce seedling weed competion and make growing kenaf more profitable. Factors that are important in establishing a transformation system for kenaf were examined. The influence of Agrobacterium strain, temperature, host tissue wounding, acetosyringone, virG/virE genes and host cell division on T-DNA expression in the kenaf shoot apex were investigated. Three Agrobacterium strains were tested, and A. tumefaciens LBA4404 significantly (α=0.05) yielded a high number of shoots surviving on selection medium; no shoots survived with EHA101S or Z707S. There was no significant difference (α=0.05) in transient T-DNA expression between 28 °C and 25 °C; however, shoots did not survive 16 °C or 19 °C co-cultivation temperatures. Shoot apex survival was increased significantly (α=0.05) when virulence genes and a cytokinin, TDZ, were combined. Sonicated shoots showed an increase in transient expression and shoot survival. Optimal conditions for shoot apex T-DNA transfer and expression were sonication for 5 s, co-cultivation with LBA4404 containingvirG/virEat room temperature, and 200 μmol/L acetosyringone.  相似文献

We investigated inter- and intraspecific phylogenetic relationships in the ectomycorrhizal fungal genus Leccinum section Scabra. Species of this section are exclusively associated with Betula and occur throughout the Northern Hemisphere. We compared the phylogenetic relationships of arctic, alpine, boreal and temperate accessions of section Scabra based on DNA sequences of the single-copy nuclear gene Gapdh and the multiple-copy nuclear region 5.8S-ITS2. Exclusively arctic lineages were not detected in species that occur both in arctic-alpine or boreal regions, except in L. rotundifoliae that was restricted to cold climates. L. scabrum and L. holopus showed an intercontinental phylogeographic pattern, and L. variicolor showed a pattern unrelated to geographical distribution. Molecular clock estimates indicated that L. rotundifoliae is as old as other species in section Scabra. Individual gene trees suggest that interspecific hybridisation occurred several times in the evolution of section Scabra.  相似文献

We compared the virulence of the entomopathogenic nematodes Steinernema scarabaei, Heterorhabditis zealandica, and Heterorhabditis bacteriophora (GPS11 and TF strains) against third instars of the Japanese beetle, Popillia japonica, the oriental beetle, Anomala (=Exomala) orientalis, the northern masked chafer, Cyclocephala borealis, the European chafer, Rhizotrogus majalis, and the Asiatic garden beetle, Maladera castanea, in laboratory and greenhouse experiments. The virulence of the nematode species relative to each other differed greatly among white grub species. H. bacteriophora and H. zealandica had similar modest virulence to P. japonica, A. orientalis, C. borealis, and M. castanea. But against R. majalis, H. zealandica showed low virulence with a clear concentration response whereas H. bacteriophora caused only erratic and very low mortality. In contrast, S. scarabaei had modest virulence against C. borealis, but was highly virulent against R. majalis, P. japonica, A. orientalis, and M. castanea with R. majalis being the most susceptible and M. castanea the least susceptible.  相似文献

为探讨淡水红藻的叶绿体基因及其适应性进化特征,选取弯枝藻属(Compsopogon)及相近外类群的rbc L基因共17条,利用PAML 4.9软件,对弯枝藻属rbc L基因编码蛋白进行生物信息学分析,并分别采用分支模型、位点模型以及分支-位点模型对基因的选择位点进行检测。结果表明,弯枝藻属rbc L基因编码蛋白的二级结构主要由α螺旋和β折叠构成,结构稳定。采用最大似然法构建的系统发育树表明,内类群为单一物种,分为3个小分支,具有一定地理分布规律。在3种进化模型中均未检测到统计上显著的正选择位点,表明绝大多数位点处于负选择压力下。因此,弯枝藻属rbc L基因未发生适应性进化。  相似文献

The sand goby Pomatoschistus minutus is a major component of marine shelf and estuarine food webs and an important study organism in behavioural research. Yet, despite the sand goby’s significance, its past and present patterns of migration and gene flow are poorly understood. Here we use the mtDNA control region and parts of the flanking tRNA genes of 63 fish from six localities in the Adriatic (Eastern Mediterranean), Western Mediterranean, Atlantic, and North Sea to investigate the phylogeography of this gobiid. Phylogenetic analyses and population genetics statistics reveal the existence of an Evolutionarily Significant Unit, sensu Moritz (1994), in the Adriatic and another in the Western Mediterranean, Atlantic, and North Sea. A possible biogeographical scenario for the separation of the ancestral population is that sand gobies in the Adriatic and Western Mediterranean split between 10,000 and 5000 years ago when due to the rise in sea temperature they migrated northwards and were bisected by the Italian peninsula. A testable prediction of this scenario is that sand gobies from the Western Mediterranean, Adriatic, and Aegean form three reciprocally monophyletic groups which are the descendants of a three-way diversification event.  相似文献

The Brazilian genus Diplusodon is the second largest genus within Lythraceae. Their 85 species occupy diverse habitats within the ‘cerrado’ vegetation, and range from shrubs and treelets to dwarf, xylopodium-bearing subshrubs. A comparative-morphological survey of their inflorescence structures using Trollian typology is here presented, as well as some evolutionary considerations drawn from mapping inflorescence characters onto a preliminary phylogeny. The inflorescences of Diplusodon are mostly polytelic, ranging from single racemes to more or less complex double-, triple-, and multiple-racemes. Frondose, compound racemes are plesiomorphic within the genus. Nevertheless, an array of derived features has been found among their species, including production of lateral cymes, proliferation of the main axis, diverse patterns of internode elongation, reduction of subtending leaves to bracts, development of accessory branches, paedomorphic flowering, and, in three species, reversion to monotely.  相似文献
