Described processing for preparation of conodonts improves the well-preservation of very fragile specimens. In classical procedures freed conodonts are airy moved without any technical care of rubbing and falling. In the new exposed technique freed conodonts are moved under distiled water; the previous inconveniences are avoided and the well-preserved conodonts number is increased.  相似文献

Discontinued treatments with BudR at different periods of the cellular cycle produce various chromosome banding after staining with acridine orange. In particular, it is possible to observe R- or Q- or an intermediary banding, simply by varying the time of incorporation of BudR. This implies that the amount of AT or GC bases present locally in DNA is not directly responsible for the banding observed. Furthermore it appears that a precise correlation exists between replication and R- or Q-banding: the DNA located at each group of bands replicates either early (R-bands) or late (Q-bands). But these timings overlap towards the middle of phase S: if the treatment is given at that time, it is possible to observe aspects intermediary between Q and R.  相似文献

There was a recent large spreading of intracytoplasmic sperm injection (ICSI) to treat male infertility in most of in vitro fertilization (IVF) laboratories. The recent data confirm the efficacy of ICSI even by using testicular sperm or sperm with grossly abnormal phenotype (round head, absence of motility). Moreover it appears that ICSI could pass beyond the last events of spermatogenesis (i.e. spermiogenesis), since normal development follows fertilization with the male gamete, spermatid, recovered just after completion of meiosis. It is obvious that the natural properties of a mature spermatozoon (motility, ADN compaction, oocyte recognition and penetration) are only necessary to reach the site of fertilization (into the female tube) and to pass through the protective enveloppes around the oocyte (cumulus oophorus, zona pellucida, plasma membrane). The current view that spermatids lack genetic maturation comparing to eggs is not valid since eggs are only secondary oocytes at a meiotic stage equivalent to that of secondary spermatocytes. Moreover genetic imprinting occurs before meiosis, and cytoplasmic structures which seem necessary for embryo development are already present in spermatids. ICSI using spermatid cells is relevant to men suffering non obstructive azzospermia if spermatids are recovered from either the ejaculate or the testicular tubes. Several normal babies were born after injection of round spermatids. Since these spermatogenic cells are present in the ejaculate of most of the patients with non obstructive azoospermia (76% in our lab), one can estimate to 5–10% the proportion of sterile men potentially concerned by conception with spermatids. However certain of these men may have occasional sperm found with testicular sperm extraction and it is to early to know if such iatrogenic extraction is always preferable to ejaculate spermatid collection.  相似文献

The proposed similarity of conformation between α-lactalbumin (α-LA) and hen egg-white lysozyme was tested by the comparison of the thermodynamic parameters obtained from the temperature dependence of denaturation. For the denaturing reaction by guanidine hydrochloride, the value of ΔCP for α-LA is almost identical with that for lysozyme, which suggests that the amount of the hydrophobic side chains buried in the interior of the molecule is the same in the native state ; the value of ΔH° and ΔS° for α-LA are also close to those for lysozyme, and the small differences are explicable by the proposed molecular model of α-LA, which implies that the somewhat large difference in ΔG° observed previously between the two proteins does not originate from large conformational differences. These results support the conformational similarity between α-LA and lysozyme as represented by the molecular model. The heat-denatured state of α-LA is also characterized by the parameters and discussed.  相似文献

The presence of hyaluronidase was detected at pH 3.8 in eight out of twelve human cancer cell line culture media. Eight cell lines derived from primary tumours and four from metastases. In three culture media the enzymatic activity was lower than 0.035 pU/cell/h. In five others (in a hepatoma cell line and in four metastasis-derived cell lines) the activity was higher than 0.057 pU/cell/h. A tumour-derived fibroblast culture was negative. The optimal activity was observed at a pH comprised between 3.6 and 4. Salt inhibition of hyaluronidase was reversible. The enzyme was denaturated by a 10-min heating at 70 °C. The enzyme was not strictly specific for hyaluronan hydrolysis but also digested chondroitin sulfates. PH20, a spermatozoid protein that has homologies with the bee venom hyaluronidase, was not expressed by cell lines tested.  相似文献

The cine Phase-Contrast Magnetic Resonance (PCMR) sequence is the only noninvasive technique for the study of cerebrospinal fluid (CSF) oscillations. It can provide CSF and blood flow measurements throughout the cardiac cycle. To study cerebral hydro-hemodynamic, models have been developed; nevertheless the majority of these models did not take into account the CSF oscillations. The objective of this study was to establish reference values for cerebral hydro-hemodynamic and propose a new electrical model of the brain dynamics.Material and methodsCSF and blood flows were measured in 19 control subjects by PCMR imaging. Dynamic flow images were analyzed on dedicated software to reconstruct the flow curves during the cardiac cycle. An electrical analogue was realized. The inputs of the model were fed by PCMR arterial and venous flows to simulate CSF oscillations. The simulated CSF oscillations were compared to the measured CSF oscillations to validate the model.ResultsThe key parameters of the CSF and blood flow curves were obtained, e.g. total cerebral blood flow was 688 ± 115 mL/min, ventricular CSF oscillatory volume was 0.05 ± 0.02 mL/cardiac cycle, and the subarachnoid CSF oscillatory volume was 0.55 ± 0.15 mL/cardiac cycle. A close agreement was found between measured and simulated cerebral CSF oscillations.ConclusionThis study established the main values characterizing cerebral hydrodynamics in a control population. It provided a better understanding of the mechanisms of intracranial volumes regulation during the cardiac cycle. Our results are now used in clinical practice and the model proposed is effective to study cerebral hydro-hemodynamic.  相似文献

Monitoring the evolution of large surface of the earth is one of the main applications of remote sensing science. It is also one of the main preoccupations of many scientific, economic and political communities. The analysis presented in this paper concerns the whole African continent. It is performed on bulk composite data from the NOAA-AVHRR sensor, which needed adapted corrections and smoothing processes. The spatial distribution of the major biomes has been mapped, according to their own phenology. Seldom detected, the ecosystem's annual biological rhythms instantly give information about the spatial and temporal behaviours of the surface. Only the use of remotely sensed data allows us to avoid problems of mixing heterogeneous information (especially climatic data). The spatial and temporal data combined through a clustering model allow analysis of the phenological behaviour of the terrestrial surface at small scale.  相似文献

Abnormal proliferation of vascular smooth muscle cells (VSMCs) is a major causative factor in atherosclerosis. Prostaglandins, secreted by endothelial cells, are reported to attenuate VSMC proliferation, but the mechanism through which this response is mediated is poorly denned. Here, the effect of prostaglandin receptor-selective agonists on the activity status of ERK and PKC, both known to modulate proliferative responses, was determined. The effect of the prostacyclin mimetic, iloprost, at inducing apoptosis was also investigated. VSMCs in culture were shown to express proteins that were detected by antibodies selective  相似文献

Résumé Le mécanisme d'assimilation des hydrocarbures par une levure, Candida lipolytica est étudié au moyen de l'analyse cinétique de la croissance du microorganisme et de la disparition du substrat hydrocarboné. Les hydrocarbures utilisés sont des n-paraffines. On ajoute au milieu soit un seul hydrocarbure (n-tetradécane ou n-hexadécane), soit un mélange binaire (n-dodécane et n-heptadécane), soit un mélange complexe (du n-undécane au n-octadécane). Contrairement à d'autres auteurs, nous pensons qu'il est peu probable que l'essentiel de la réaction s'effectue par contact des gouttes de substrat et des microorganismes puisque l'on observe des retards d'assimilation de certains hydrocarbures: ceux de faible poids moléculaire (les plus solubles) sont assimilés plus rapidement. Il semble donc que l'assimilation se fasse en grande partie à partir d'hydrocarbures préablement solubilisés.
Kinetics of hydrocarbon assimilation by Candida lipolytica
Summary The mechanism of hydrocarbon uptake by a yeast, Candida lipolytica has been studied by means of the kinetic analysis of micro-organism growth and substrate assimilation. Hydrocarbons used as only source of carbon are normal alkanes either pure (n-tetradecane or n-hexadecane) or in mixture of two paraffins (n-dodecane and n-heptadecane) or eight paraffins (n-undecane to n-octadecane). In these last cases delays in n-alkanes consumption are observed. They show that the most soluble substrates (lower molecular weight) are first consumed. In opposition to other authors we think that there is little probability for main reaction occurring by direct contact between drops and micro-organisms. The evidence indicates that n-alkanes are mainly utilized in the dissolved state.

Chara vulgaris was cultivated in a natural medium containing nickel and cobalt chloride.Growth increased in a culture solution containing Ni and Co in small amounts. Tissue analysis showed that the levels of Ni and Co in the dry matter were high. IntroductionInter-ictal 18F-2-fluoro-deoxy-D-glucose-positron emission tomography (FDG-PET) plays a key role for the preoperative evaluation of patients with pharmacoresistant temporal lobe epilepsy. PET images are usually analyzed visually, a way that is reported to provide a high diagnostic value but that remains subjective, depending on the expertise and experience of the observer. By contrast, the voxel-based quantitative analyses, such as statistical parametric mapping (SPM), are objective and therefore, observer independent methods of analyses. In this study, the accuracy of the analyses of brain FDG-PET images to lateralize the temporal lobe epileptogenic zone was compared between: (1) a conventional visual method, (2) a quantitative SPM analysis, and (3) a visual analysis of inter-hemispheric asymmetry (IHA) obtained after images substraction.Materials and methodsFDG-PET scans of 31 patients presenting a severe temporal epilepsy and whom the temporal foci had been accurately lateralized (successful subsequent surgical treatment) were retrospectively analysed by (1) a consensual visual analysis from two experienced observers; (2) SPM analysis with voxel-wise comparisons of FDG-PET images of patients with those of age-matched healthy controls, using various statistical threshold (P) and cluster (k) values; and (3) visual assessment by the two same observers of images obtained for assessing the IHA. For this purpose, a flipped image was initially obtained by reversing in the left-right direction the FDG-PET images, which had been previously spacially normalized with the SPM template. Then, flipped and non-flipped images were substracted.ResultsThe temporal hypometabolic area was accurately identified: (1) by the conventional visual analysis in 87 % of patients and with a satisfactory interobserver reproducibility (interobserver Cohen's coefficient = 0.79); (2) by SPM analysis, in 90 % of patients (when using optimal thresholds of 0.01 for P value and of 50 voxels (400 mm3) for k value); and (3) with the visual analysis of IHA in 97 % of patients with an excellent interobserver reproductibility (interobserver Cohen's coefficient = 1).ConclusionIn patients presenting severe temporal epilepsy, visual assessment of FDG-PET images from IHA seems more accurate for lateralizing the epileptogenic temporal areas when compared with either conventional visual or quantitative SPM analyses. Moreover, this method is very easy to use in clinical practice, contrary to the quantitative method using SPM  相似文献
