Bacterial Antagonists to Verticillium dahliae Kleb.   总被引:7,自引:0,他引:7  
Bacteria were isolated from the rhizosphere of Verticillium dahliae hosts and from environments. A total of 1394 bacterial isolates were screened for their ability to inhibit in vitro the growth of the phytopathogenic fungi V. dahliae ; 15% (203 of the isolates) showed antifungal effects. Seventeen isolates were selected and determined for further investigations, seven different species were identified. Several of the bacterial species listed, e.g. Erwinia herbicola, Pseudomonas chlororaphis, Pseudomonas paucimobilis and Xanthomonas maltophilia have not been reported previously as bacterial antagonists of V. dahliae. Bacillus subtilis, Pseudomonas fluorescens and Xanthomonas maltophilia are strong antagonists. We proved that lytic enzymes and siderophores are involved in the inhibition of growth. Ultrastructural and morphological changes were induced in Verticillium dahliae by the antagonistic bacteria.  相似文献

Summary Electron micrographs of the early stages in microsclerotial development in v. dahliae Kleb. showed that hyphae became swollen and vacuolate and extruded melanizing particles into the interhyphal spaces of the microsclerotium. Peripheral microsclerotial cells were killed either by a process of autoparasitisation from adjacent hyphae or by autolysis. Variations in the thickness of the melanized material surrounding cells gave the superficial appearance of variations in cell wall thickness between individual cells though actual changes in cell wall thickness were not observed.  相似文献

微丝在真菌生长发育、胞质分裂等生命过程中具有重要功能。通过农杆菌介导遗传转化方法,将荧光mCherry标记微丝的表达载体pSULPH-Lifeact-mCherry转入大丽轮枝菌(Verticillium dahliae Kleb.)野生型V592,获得稳定的微丝荧光标记菌株V592/Lifeact-mCherry,并检测了其生物学表型和孢子萌发、菌丝生长等过程中的微丝荧光动态变化。结果表明:微丝荧光标记菌株的菌落形态、生长速率、产孢量、萌发率等表型与野生型没有显著差异;且可以观察到微丝荧光信号在分生孢子和菌丝的顶端及隔膜都有清晰定位,同时对该菌株隔膜形成过程微丝动态观察发现,微丝参与胞质分裂进程中肌动球蛋白收缩环CAR (Contractile actomyosin ring)的形成。微丝荧光标记菌株可用于微丝在真菌发育中的动力学研究,这为深入研究微丝在真菌发育及致病过程中的作用机制提供理论与实践支撑。  相似文献

Thirty-four isolates of Verticillium dahliae Kleb. from nine different genera of dicotyledonous host plants and a broad range of geographic regions were analysed genotypically, Random amplified polymorphic DNA (RAPD) markers were used for the estimation of the genetic variability within the species. Using four primers for the analysis, 79 distinct fragments were obtained. The derived phenogram clustered the isolates in two main groups; one consisted almost entirely of V. dahliae isolates from oilseed rape ( Brassica napus napus ), the other group comprised isolates from a wide range of host plants. No correlation between geographic location of the isolates and the RAPD-pattern was observed.
Sequencing of the gene for the 18SrRNA and calculation of the phylogenetic tree integrated the deuteromycetous fungus V. dahliae into the sexual system of the filamentous ascomycetes.  相似文献

Summary Freeze etched replicas of hyphae of Verticillium dahliae revealed the presence of paramural bodies and elaborations of the plasmalemma. It is suggested that the presence of these structures in freeze etched preparations is indicative of their presence in living cells rather than as post-mortem artifacts following fixation.  相似文献

转基因抗虫棉的抗病性下降已成为制约我国棉花生产的重要因素之一.以转基因抗虫棉及其亲本非转基因棉花对照为材料,研究转基因抗虫棉根系分泌物对棉花黄萎病菌孢子萌发和菌丝生长的影响,并对其根系分泌物中氨基酸及糖类的组成和含量进行了测定.结果表明:与亲本非转基因棉相比,2种转基因抗虫棉对棉花黄萎病菌的抗性下降,转基因抗虫棉的根系分泌物对黄萎病菌孢子萌发和菌丝生长均具有促进作用.与亲本常规棉中23相比,转基因双价抗虫棉中41根系分泌物中多了甲硫氨酸和赖氨酸,并且天冬氨酸、谷氨酸、丝氨酸、丙氨酸、缬氨酸、亮氨酸、酪氨酸的含量显著升高.泗棉三号与单价抗虫棉GK12的根系分泌物中虽然含有相同的氨基酸组分,但GK12的根系分泌物中酪氨酸、缬氨酸、亮氨酸的含量显著下降.中41根系分泌物中检测出4种糖类,而中23中仅检测出葡萄糖.抗虫棉GK12和泗棉三号根系分泌物中都检测出4种糖类,但其含量差异明显.  相似文献

【背景】棉花黄萎病是由大丽轮枝菌(Verticillium dahliae Kleb.)引起的一种世界性病害,近年来对该病害的生物防治因具有环境友好和人畜安全的特性而倍受关注。【目的】筛选棉花黄萎病高效拮抗细菌并对其进行鉴定,为棉花黄萎病的生物防治扩充菌种资源。【方法】采用稀释涂布平板法分离细菌,并进行拮抗细菌的初筛和复筛,通过形态特征、生理生化特征和16S rRNA基因序列分析对筛选到的细菌进行鉴定,确定其分类地位。【结果】初筛分离到535株对病原菌具有拮抗作用的细菌,并选取了108株拮抗细菌进行复筛,最终筛选到了4株优势拮抗细菌。通过形态观察、生理生化特征和16SrRNA基因序列分析,将菌株BHZ-29、SHT-15、SHZ-24和SMT-24分别鉴定为贝莱斯芽孢杆菌(Bacillusvelezensis)、枯草芽孢杆菌斯皮兹仁亚种(Bacillus subtilis subsp. spizizenii)、萎缩芽孢杆菌(Bacillus atrophaeus)和香草芽孢杆菌(Bacillus vanillea)。【结论】获得了4株高效拮抗细菌,并且首次报道了香草芽孢杆菌对棉花黄萎病菌具有抑制作用。  相似文献

AIMS: A screening approach was developed to assess the potential of rhizobacterial strains to control Verticillium wilt caused by Verticillium dahliae Kleb. METHODS AND RESULTS: Sixty randomly chosen antagonistic bacterial strains originally isolated from rhizosphere of three different host plants of V. dahliae--strawberry, potato and oilseed rape--were evaluated for biocontrol and plant growth promotion by analysing in vitro antagonism towards V. dahliae and other plant pathogenic fungi, production of fungal cell wall-degrading enzymes and plant growth-promoting effects on strawberry seedlings. To test the plant growth-promoting effect, a microplate assay with strawberry seedlings was developed. Although the rhizobacterial strains were isolated from different plants they showed effects on the growth of strawberry seedlings. According to the in vitro biocontrol and plant growth-promoting activity, the three best candidates Pseudomonas putida B E2 (strawberry rhizosphere), Ps. chlororaphis K15 (potato rhizosphere) and Serratia plymuthica R12 (oilseed rape rhizosphere) were selected for greenhouse experiments to verify the in vitro screening results. Under greenhouse conditions the isolates selected according to this strategy were as effective, or more effective than commercial biocontrol agents and may therefore possibly be valuable as antagonists of V. dahliae. CONCLUSIONS: In this study, the screening strategy resulted in a selection of three interesting biocontrol candidates against Verticillium: Ps. putida B E2 (strawberry rhizosphere), Ps. chlororaphis K15 (potato rhizosphere) and Ser. plymuthica R12 (oilseed rape rhizosphere). SIGNIFICANCE AND IMPACT OF THE STUDY: A new combination of in vitro screening methods including a microplate assay with strawberry seedlings to test the plant growth promoting effect which allow to more efficiently select potential biological control agents was developed successfully.  相似文献

SUMO化修饰与植物抗病防御、信号转导和耐旱等有着直接的关系.本文以抑制差减杂交(suppression subtractive hybridization,SSH)技术获得SUMO的EST为信息探针,对棉花EST数据库进行同源搜索和电子克隆,获得了全长为396 bp的SUMO基因编码区cDNA全长,我们将该基因命名为GhSUMO,推测该基因编码95个氨基酸.分别以抗黄萎病陆地棉品种豫棉21号的cDNA和DNA为模板,对该基因进行了PCR扩增验证,测序结果表明,GhSUMO基因序列与电子克隆序列一致,且没有内含子.蛋白序列分析表明,该蛋白具有保守泛素结构域和C端双Gly的断裂/连接位点,以及保守的疏水表面和Ulp1-Smt3互作位点.系统进化分析表明,该蛋白与蓖麻的同源序列表现了最高的相似性,与其它双子叶植物同源序列次之,而与单子叶植物的相似性较低.棉苗接菌后实时定量PCR结果显示,该基因表达量在接黄萎病菌的量48 h后明显上调,96 h达到未接菌对照的5倍以上.GhSUMO基因可受黄萎病菌诱导表达,表明该基因在陆地棉抗黄萎病的机制中可能发挥重要作用.  相似文献

Root-colonizing fungi obtained from thoroughly washed root segments of symptomless young oilseed rape plants growing in field soil were examined for their inhibitory effect on the wilt fungus, Verticillium dahliae Kleb. The isolated fungi that were possible to identify belonged to the different genera Trichoderma , Gliocladium , Mortierella , Fusarium and Alternaria . Oilseed rape seedlings from seeds exposed to both sporulating and nonsporulating fungal isolates were shown to be colonized by their hyphae in gnotobiotic conditions and the emergence/root growth was significantly affected in their presence. Inoculation with a few isolates not only covered the root surface but also seemed to extensively colonize the inner root cortical tissues partly by producing characteristic structures in the roots. All of these suppressed the pathogen in in vitro assays but the mode of suppression in most cases did not seem to be due to production of antifungal substances. In nonsterile field soil, several of the isolates conferred protection against V. dahliae by delaying symptom development. None of the isolates was shown to increase wilt symptoms. In addition to protection, two isolates, yet to be identified, also showed increased shoot dry weight compared with that of healthy controls. A majority of the isolates showed proteolytic activity whereas only three produced chitinolytic enzymes.
Results for the isolates obtained by the method selected for this study could be considered as a starting point for evaluating antagonists capable of both competing for root colonization and preventing root infection, with the final aim being to achieve successful biological control of the wilt fungus.  相似文献

摘要:【目的】了解棉花内生细菌数量动态,从棉花中获得拮抗黄、枯萎病菌的内生细菌资源【方法】对棉花根、茎、叶表面灭菌,采用稀释平板法分离棉花内生细菌;通过对峙培养法体外鉴定分离的棉花内生细菌对棉花黄、枯萎病菌的拮抗作用,并对拮抗棉花黄、枯萎病菌的内生细菌16S rDNA序列进行了分析【结果】棉花根中内生细菌的数量显著高于茎、叶;根的苗期总体内生细菌数量低于开花期、吐絮期,茎、叶中的内生细菌数量在不同生育期呈现一定的波动性,但趋势性不明显;6个棉花品种根中内生细菌平均数量差异并不显著,但茎、叶中内生细菌数量不同品种间呈现一定程度差异。平板对峙鉴定显示:棉花根中具有较高比例的拮抗棉花黄、枯萎病菌的内生细菌,拮抗强致病落叶型黄萎病菌(V107)的内生细菌比例不仅低于枯萎病菌(F108),而且低于非落叶型黄萎病菌(V396)。同时拮抗棉花黄、枯萎病菌的内生细菌有44株,16S rDNA分子序列分析表明:这些拮抗内生细菌的类群包括了两个门(变形杆菌门、拟杆菌门)8个属,其中10个菌株与已报道菌株相似性<97%,可能是新的种(属),优势种群为肠杆菌属(18株)、泛菌属(15株)。【结论】棉花的品种、生育期与器官影响棉花内生细菌数量;棉花拮抗黄、枯萎病菌的内生细菌具有优势种群,且具多样性。  相似文献

Fungi with antagonistic activity toward plant pathogens play an essential role in plant growth and health. To analyze the effects of the plant species and the site on the abundance and composition of fungi with antagonistic activity toward Verticillium dahliae, fungi were isolated from oilseed rape and strawberry rhizosphere and bulk soil from three different locations in Germany over two growing seasons. A total of 4,320 microfungi screened for in vitro antagonism toward Verticillium resulted in 911 active isolates. This high proportion of fungi antagonistic toward the pathogen V. dahliae was found for bulk and rhizosphere soil at all sites. A plant- and site-dependent specificity of the composition of antagonistic morphotypes and their genotypic diversity was found. The strawberry rhizosphere was characterized by preferential occurrence of Penicillium and Paecilomyces isolates and low numbers of morphotypes (n = 31) and species (n = 13), while Monographella isolates were most frequently obtained from the rhizosphere of oilseed rape, for which higher numbers of morphotypes (n = 41) and species (n = 17) were found. Trichoderma strains displayed high diversity in all soils, but a high degree of plant specificity was shown by BOX-PCR fingerprints. The diversity of rhizosphere-associated antagonists was lower than that of antagonists in bulk soil, suggesting that some fungi were specifically enriched in each rhizosphere. A broad spectrum of new Verticillium antagonists was identified, and the implications of the data for biocontrol applications are discussed.  相似文献

利用离体抑菌圈法从茄子根际土壤筛选出8株对黄萎病菌大丽轮枝菌(Verticillium dahliae)抑菌效果明显的芽孢杆菌,其中F53菌株分泌几丁质酶和-β1,3-葡聚糖酶,对所检测的12种植物病原菌具明显的抑制作用,其发酵液热、酸碱稳定性强,用F53菌株制成的菌剂对茄子黄萎病盆栽试验防效为52.4%~75.8%,初步鉴定F53菌株为环状芽孢杆菌(Bacillus circulans)。  相似文献

Investigation of a possible association between Verticillium dahliae and H. rostochiensis (pathotype E, British notation) was based on field observations and an examination of disease development in single-stemmed potato plants grown in pots. An association was found in the distribution of the nematode and the fungus in the field, and the disease was far more severe with a combined infection than with either pathogen alone. Studies on leaf-area development and yield reduction suggest there is synergism between fungus and nematode, the reductions produced by the combined infection exceeding the sum of those produced by either pathogen alone. Fungal mycelium and the extent of host colonization by V. dahliae were greatly increased by the presence of the nematode. The possible benefits to V. dahliae in the fungus-nematode complex are discussed.  相似文献
