Motor endplates in fast and slow skeletal muscles have different functional and morphological characteristics, and for brevity, are termed fast and slow respectively. We have examined the terminal arborization patterns of fast fibular and slow soleus axons 3-4 and 6 months after they reinnervated old preformed endplates or formed new ectopic endplates with denervated rat soleus muscles. Ectopic endplates formed by transplanted fibular and soleus nerves were fast and slow in appearance respectively. Both the fibular and the soleus nerves formed endplates of slow appearance when they reinnervated the original endplates. The fast appearance of ectopic fibular nerve endplates was unaffected by reinnervation of the original endplates by the slow soleus nerve. Dually innervated fibres had intermediate contraction speed compared to the fast fibres reinnervated only by the fibular nerve and the slow fibres reinnervated only by the soleus nerve. Continuous stimulation of the transplanted fibular nerve at 10 Hz for 3-4 months, starting just before the onset of ectopic endplate formation, prevented the increase in contraction speed seen without stimulation. The ectopic endplates of the stimulated axons were much smaller than usual and showed some signs of fast to slow transformation, but the transformation was incomplete and varied in degree between preparations. The functional recovery of skeletal muscles after peripheral nerve transection and microsurgical repair is generally incomplete. Several reinnervation abnormalities have been described even after nerve reconstruction surgery. Less is known, however, about the regenerative capacity of reinnervated muscles. Previously, we detected remarkable morphological and motor endplate alterations after inducing muscle necrosis and subsequent regeneration in the reinnervated rat soleus muscle. In the present study, we comparatively analyzed the morphometric properties of different fiber populations, as well as the expression pattern of myosin heavy chain isoforms at both immunohistochemical and mRNA levels in reinnervated versus reinnervated-regenerated muscles. A dramatic slow-to-fast fiber type transition was found in reinnervated soleus, and a further change toward the fast phenotype was observed in reinnervated-regenerated muscles. These findings suggest that the (fast) pattern of reinnervation plays a dominant role in the specification of fiber phenotype during regeneration, which can contribute to the long-lasting functional impairment of the reinnervated muscle. Moreover, because the fast II fibers (and selectively, a certain population of the fast IIB fibers) showed better recovery than did the slow type I fibers, the faster phenotype of the reinnervated-regenerated muscle seems to be actively maintained by selective yet undefined cues.  相似文献

The total content of myosin heavy chains (MHC) and their isoform pattern were studied by biochemical methods in the slow-twitch (soleus) and fast-twitch (extensor digitorum longus) muscles of adult rat during atrophy after denervation and recovery after self-reinnervation. The pattern of fibre types, in terms of ultrastructure, was studied in parallel. After denervation, total MHC content decreased sooner in the slow-twitch muscle than in the fast-twitch. The ratio of MHC-1 and the MHC-2B isoforms to the MHC-2A isoform decreased in the slow and the fast denervated muscles, respectively. After reinnervation of the slow muscle, the normal pattern of MHC recovered within 10 days and the type 1 isoform increased above the normal. In the reinnervated fast muscle, the 2B/2A isoform ratio continued to decrease. Traces of the embryonic MHC isoform, identified by immunochemistry, were found in both denervated and reinnervated slow and fast muscles. A shift in fibre types was similar to that found in the MHC isoforms. Within 2 months of recovery a tendency to normalization was observed. The results show that (a) MHC-2B isoform and the morphological characteristics of the 2B-type muscle fibres are susceptible to lack of innervation, similar to those of type 1, (b) during muscle recovery induced by reinnervation the MHC isoforms and muscle fibres shift transiently to type 1 in the soleus and to type 2A in the extensor digitorum longus muscles, and (c) the embryonic isoform of MHC may appear in the adult skeletal muscles if innervation is disturbed.  相似文献

Calcitonin gene-related peptide (CGRP) occurs only in some motoneurons. In this study, the presence of CGRP in motor endplates in relation to muscle fibre types was examined in slow (soleus muscle) and fast [tibialis anterior (TA) and extensor digitorum longus (EDL)] leg muscles of the rat. CGRP was detected by use of immunohistochemical methods, and staining for the mitochondrial-bound enzyme NADH-TR was used for demonstration of fibre types. The fibres showing low NADH-TR activity were interpreted as representing IIB fibres. All such fibres located in the superficial portion of TA were innervated by endplates displaying CGRP-like immunoreactivity (LI), whereas in the deep portion of TA some of these fibres lacked CGRP-LI at their endplates. Thirty per cent of the IIB fibres in EDL showed CGRP-LI at the endplates. All fibres in TA and EDL displaying high NADH-TR activity and interpreted as type-IIA fibres, lacked CGRP-LI in their motor innervation. One third of the fibres with intermediate NADH-TR activity in TA exhibited CGRP-LI at their endplates, whereas in EDL only few such fibres displayed CGRP-LI in the endplate formation. These fibres are likely to belong to type-IIX or type-I motor units. CGRP-LI was very rarely detected at the endplates in the soleus muscle. These observations show that distinct differences exist between the slow muscle, soleus, and the fast muscles, TA and EDL, but that there are also differences between the different types of fibres in TA and EDL with respect to presence of CGRP-LI at the endplates. As CGRP-LI was frequently detected at endplates of IIB fibres, it is likely that CGRP has a particular role related to the differentiation and maintenance of these fibres.  相似文献

Fast and slow contracting fibers in neonatal mammalian skeletal muscle are each innervated in a highly specific manner by motor neurons of the corresponding type, even at an age when polyinnervation is widespread. Chemospecific recognition is a possible mechanism by which this pattern of innervation could be established. We have investigated this possibility by studying the degree of specificity during reinnervation of rabbit soleus muscle following nerve crush on Postnatal Day 1 or 4. We assayed fiber type composition by measuring the twitch rise times of motor units within 2 days of the onset of functional reinnervation (5-6 days after nerve crush). In contrast to the broad, bimodal distribution of single motor unit twitch rise times seen in normal muscles, motor units in reinnervated muscles yielded a narrower, unimodal distribution of rise times. Rise times of reinnervated units were intermediate to those of normal fast and slow units, suggesting that reinnervated units were composed of a mixture of fast and slow contracting fibers. An alternative possibility, that specific reinnervation was masked by contractile dedifferentiation of muscle fibers, was examined by maintaining a transmission blockade induced by botulinum toxin poisoning for an equivalent interval. Twitch rise times of treated motor units exhibited the distinctly bimodal distribution characteristic of normal muscles, suggesting that muscle fibers can retain contractile diversity during a transient period of denervation. We carried out computer simulations to estimate the amount of rise time diversity induced by varying degrees of specificity during reinnervation. Based on this analysis, we conclude that there is little if any selective reinnervation of muscle fiber types at the ages studied.  相似文献

Summary The tibialis anterior, extensor digitorum longus and soleus muscles in the rat were examined with respect to the presence of calcitonin gene-related peptide-like as well as substance P-like immunoreactivity. In some of the motor endplates in these muscles, identified by staining for acetylcholinesterase activity, calcitonin gene-related peptide-like immunoreactivity was detected, but in others it was not. Calcitonin gene-related peptide-like immunoreactivity was found to coexist with substance-P-like immunoreactivity in nerve fibres located outside and inside the capsule of the muscle spindles, as well as in nerve fibres located in nerve fascicles. These fibres presumably represent sensory nerve fibres. Calcitonin gene-related peptide-like immunoreactivity, but not substance P-like immunoreactivity, was also detected, in cap-like structures located on the surface of the intrafusal muscle fibres in the polar regions of the spindles, structures which are likely to correspond to motor plate endings. The observations suggest that calcitonin gene-related peptide is heterogeneously present in the endplates of rat hind limb muscles, and gives for the first time immunohistochemical evidence for the presence of calcitonin gene-related peptide and substance P in the innervation of muscle spindles.  相似文献

A histochemical study, using myosin-adenosine triphosphatase activity at pH 9.4, was conducted in soleus and plantaris muscles of adult rats, after bilateral crushing of the sciatic nerve at the sciatic notch. The changes in fiber diameter and per cent composition of type I and type II fibers plus muscle weights were evaluated along the course of denervation-reinnervation curve at 1, 2, 3, 4 and 6 weeks postnerve crush. The study revealed that in the early denervation phase (up to 2 weeks postcrush) both the slow and fast muscles, soleus and plantaris, resepctively, atrophied similarly in muscle mass. Soleus increased in the number of type II fibers, which may be attributed to "disuse" effect. During the same period, the type I fibers of soleus atrophied as much or slightly more than the type II fibers; whereas the type II fibers of plantaris atrophied significantly more than the type I fibers, reflecting that the process of denervation, in its early stages, may affect the two fiber types differentially in the slow and fast muscles. It was deduced that the type I fibers of plantaris may be essentially different in the slow (soleus) and fast (plantaris) muscles under study. The onset of reinnervation, as determined by the increase in muscle weight and fiber diameter of the major fiber type, occurred in soleus and plantaris at 2 and 3 weeks postcrush, respectively, which confirms the earlier hypotheses that the slow muscles are reinnervated sooner than the fast muscles. It is suggested that the reinnervation of muscle after crush injury may be specific to the muscle type or its predominant fiber type.  相似文献

Miniature endplate potentials (MEPPs) are regarded as the expression of release of a single quantum of acetylcholine by motor nerve endings in the muscle. Mepp frequency is dependent on the presynaptic mechanism, but MEPP amplitudes and time courses are the result of the characteristics of pre- and postsynaptic structures and of the interaction between them. After post-traumatic reinnervation of skeletal muscles, MEPP frequency increases, reaching slowly normal values. Two groups of male, Sprague Dawley rats were used: in the first group left sciatic nerve was crushed and nerve fibres were allowed to regenerate, whereas the others were regarded as controls. MEPPs were intracellularly recorded in end plates of normal and reinnervated left extensor digitorum longus muscle. MEPPs were sampled and recorded on a personal computer, and, subsequently, amplitude, rise time and half decay time were computed. At early stage after reinnervation, MEPPs showed rise times and decay times longer than normal. Afterwards, we did not find differences between mepp time courses by normal and reinnervated end plates. The possible relationships between the results and changes in acetylcholine receptor number and type, and in acetylcholinesterase activity occurring during denervation and reinnervation are discussed.  相似文献

The goal of this study was to determine the acute effects of permanent denervation on the length density of the capillary network in rat slow soleus (SOL) and fast extensor digitorum longus (EDL) muscles and the effect of short-lasting reinnervation in slow muscle only. Denervation was performed by cutting the sciatic nerve. Both muscles were excised 2 weeks later. Reinnervation was studied 4 weeks after nerve crush in SOL muscle only. Capillaries and muscle fibres were visualised by triple immunofluorescent staining with antibodies against CD31 and laminin and with fluorescein-labelled Griffonia (Bandeira) simplicifolia lectin. A recently developed stereological approach allowing the estimation of the length of capillaries adjacent to each individual fibre (Lcap/Lfib) was employed. Three-dimensional virtual test grids were applied to stacks of optical images captured with a confocal microscope and their intersections with capillaries and muscle fibres were counted. Interrelationships among capillaries and muscle fibres were demonstrated with maximum intensity projection of the acquired stacks of optical images. The course of capillaries in EDL seemed to be parallel to the fibre axes, whereas in SOL, their preferential direction deviated from the fibre axes and formed more cross-connections among neighbouring capillaries. Lcap/Lfib was clearly reduced in denervated SOL but remained unchanged in EDL, although the muscle fibres significantly atrophied in both muscle types. When soleus muscle was reinnervated, capillary length per unit fibre length was completely restored. The physiological background for the different responses of the capillary network in slow and fast muscle is discussed. Structure of tendon organs of the rat after neonatal de-efferentation   总被引:2,自引:0,他引:2  
Summary The number, size and structure of tendon organs were examined in leg muscles of the rat 3–19 weeks after de-efferentation performed in newborn animals by removal of the lumbosacral spinal cord. After this operation, tendon organs differentiated and grew in disused muscles and were innervated by primary sensory neurons, the dorsal roots of which had been disrupted.Three weeks after de-efferentation extensor digitorum longus muscles contained 14.1±1.0 (mean±standard error) and soleus muscles had 14.2±1.6 tendon organs, which corresponds to the mean number of tendon organs in the respective control muscles. The mean size of tendon organs was, however, changed. Tendon organs became on the average by 53% longer and by 35% thinner in de-efferented extensor digitorum longus muscles that were prolonged due to immobilization, as compared with shorter and wider tendon organs in de-efferented soleus muscle that remained in the shortened position.The ultrastructural differentiation of tendon organs was completed after the operation as under normal conditions. Thus it can be concluded that elimination of muscle function during the period of postnatal development indirectly affects the mean size of these receptors, but does not otherwise interfere with their morphogenesis.  相似文献

Phosphatidylinositol phosphodiesterase activity was determined in cytosol prepared from rat slow (soleus) and fast (extensor digitorum longus) muscles. The substrate was prepared by incubation of sarcoplasmic reticulum with myo-[2-3H]inositol. The enzyme hydrolysed both membrane-bound and extracted phosphatidylinositol. The activity determined with the isolated phospholipid exhibited an optimum at pH 5.5. Ca2+ ions stimulated the activity. The enzyme specific activity was higher in cytosol prepared from soleus muscle than in that from extensor digitorum longus muscle. After section of the motor nerve, the activity of the enzyme increased in both muscles up to 36 h and then declined. A function for this enzyme in the control of acetylcholine sensitivity in muscle is discussed.  相似文献

Mammalian nerves to fast and slow muscles have the remarkable property of changing the speed of contraction of muscles following cross-reinnervation. The biochemical basis of speed transformation is the change in myosin in ATPase activity. This paper provides electrophoretic evidence for structural changes in myosin from cross-reinnervated muscles. A method is described for the separation of intact fast and slow muscle myosins by polyacrylamide gel electrophoresis. This method utilizes the fact that ATP and its analogs prevent the formation of myosin polymers in low ionic strength buffers. In this system, normal fast muscle myosin has a higher electrophoretic mobility than slow muscle myosin. Normal rat soleus myosin has a major slow and a minor fast component due to two populations of muscle fibers. The same muscle cross-reinnervated by a fast muscle nerve shows only the fast component, The normal, homogeneous fast extensor digitorum longus muscle has only the electrophoretically fast myosin, but following cross-reinnervation it shows both fast and slow components. These results suggest that mammalian motor nerves can induce or suppress the expression of genes that code for fast and slow skeletal muscle myosins.  相似文献

The regeneration of skeletal muscles is a suitable model to study the development and differentiation of contractile tissues. Neural effects are one of the key factors in the regulation of this process. In the present work, effects of different reinnervation protocols (suture or grafting) were studied upon the regenerative capacity of rat soleus muscles treated with the venom of the Australian tiger snake, notexin, which is known to induce complete necrosis and subsequent regeneration of muscles. Morphological and motor endplate analysis indicated that the regenerative capacity of denervated, and thereafter surgically reinnervated muscles remains impaired compared to that of normally innervated muscles, showing differences in the muscle size, fiber type pattern and motor endplate structure, even 35 days after the notexin injection. A lack or deficiency of secreted neural factors, deterioration of satellite cells and/or incomplete recovery of the sutured or grafted nerves may be the cause of these discrepancies in the regeneration process.  相似文献

In experiments on neuromuscular synapses of rat fast (m. Extensor digitorum longus, EDL) and slow (m. soleus) skeletal muscles, changes in the intensity of spontaneous quantal mediator secretion in response to the activation of presynaptic cholinoreceptors by the nonhydrolyzable acetylcholine analogue carbachol and to an increase in K+ concentration in the control group of animals and in animals subjected to different terms of unloading of hindlimbs have been compared. The intensity of spontaneous secretion of mediator quanta was evaluated from the mean frequency of miniature endplate potentials. In the control group of animals, the frequency of miniature endplate potentials by the action of carbachol increased by 363% in m. EDL and by 62% in m. soleus. The frequency of miniature endplate potentials in the synapses of m. EDL was more sensitive to K(+)-induced depolarization too. The bearing unloading of hindlimbs abolished the sensitivity of spontaneous secretion to carbachol in the synapses of m. EDL, whereas in m. soleus it was unchanged. However, the preservation of sensitivity of nerve endings of fast muscle to K(+)-induced depolarization allows one to assume that the hindlimb unloading leads to a decrease in the number of functioning presynaptic receptors.  相似文献

The crossed nerve anastomosis between the peripheral end of the vagus nerve, cut above the nodose ganglion, and the peripheral end of the accessory nerve has demonstrated the capacity of some vagal afférents to reinnervate, via the accessory nerve stump, certain sternocephalicus muscle fibers in the rabbit. These results add to our understanding of the capacity of these afferents to counter the post-denervational atrophying process that occurs in the reinnervated muscles and to evaluate the changes induced in these muscles during reinnervation. Our work shows that within 3 months, the vagal sensory reinnervation of previously denervated sternocephalicus muscles induces their total weight recovery. This recovery is concomitant on the one hand with the hypertrophy of the four muscle fiber types (I, IIbd, IIc and IIa) identified histochemically in the normal muscles and, on the other, with the appearance of small newly formed myofibers, which are often underlined by characteristic central nuclei. The vagal sensory neurones induce important changes in the percentages and the muscle cross-sectional distribution of the fibers in reinnervated muscles. In these muscles we see also the disappearance of the fast myosin heavy chains MHCiib and MHCiid, the upholding of the fast MHCiia percentage and an increase in the slow MHCi isoform.  相似文献

Skeletal muscle fibers can be reinnervated by motor cholinergic fibers, that is, functional connection can be achieved. However, functional connection implies not only the capacity of the nerve impulse to elicit a contractile response but also the capability of the reinnervating neurons to evoke particular modifications of the physiological and biological features of the muscles. In order to search for some of the modifications due to reinnervation by preganglionic sympathetic fibers, muscle contraction time was studied in three different preparations of adult cats: a) cricothyroid muscle reinnervated by preganglionic fibers; b) cricothyroideus reinnervated by its own nerve; and c) the corresponding normal neuromuscular preparation. The activities of malic dehydrogenase, of aldolase and pyruvic kinase were studied in these three preparations as well as in the denervated cricothyroid muscles. Reinnervation by preganglionic fibers prolonged the twich contraction time, whereas, self-reinnervation did not alter it. On the other hand, the activities of the three enzymes decreased as a result of denervation. In contrast, the muscle reinnervated with sympathetic preganglionic fibers partially recovered the normal level of malic dehydrogenase and the aldolase activities; but showed no modification in the level of pyruvic kinase activity. Conversely, in the muscle fibers reinnervated by their own nerve, the activity of the three enzymes returned to normal levels. The shortening of contraction time of the preganglionic reinnervated muscle correlates well with the features of the enzymic activities found in these muscles. It can be concluded: a) preganglionic sympathetic axons are able to achieve functional connections with striated muscles and b) considering the trophic effect, preganglionic fibers resemble the motor nerve supplying slow muscles.  相似文献

The histochemical ATPase activity and the myosin light chains of a rat fast muscle (extensor digitorum longus, EDL) and a rat slow muscle (soleus) during development have been investigated. Both muscles initially synthesize fast myosin light chains and show the intense histochemical ATPase activity characteristic of adult fast muscle fibers. After birth, the soleus begins to accumulate slow fibers with their characteristic low histochemical ATPase activity, and slow myosin light chains begin to appear. Sciatic neurectomy prevents the development of slow fibers and the synthesis of slow myosin light chains in the soleus, while the EDL is unaffected. Similarly, cordotomy of an adult rat results, in the soleus, in the appearance of fibers with more intense staining for ATPase and an increase in fast myosin light chains. The EDL is unchanged by cordotomy. As a result, we suggest that slow muscle development, but not fast muscle development, is dependent upon the functional activity of the nervous system.  相似文献

Muscle spindle development and function are dependent upon sensory innervation. During muscle regeneration, both neural and muscular components of spindles degenerate and it is not known whether reinnervation of a regenerating muscle results in reestablishment of proper neuromuscular relationships within spindles or whether sensory neurons may exert an influence upon differentiation of these spindles. Muscle spindle regeneration was studied in bupivacaine-treated grafts of rat extensor digitorum longus (EDL) muscles. Three types of EDL graft were performed in order to manipulate the extent to which regenerating spindles might be reinnervated: (1) grafts reinnervated following severance of their nerve supply (standard grafts); (2) grafts in which intact nerve sheaths appear to facilitate reinnervation (nerveintact grafts); and (3) grafts in which reinnervation was prevented (nonreinnervated grafts). Complete degeneration of muscle fibers occurred in all grafts prior to regeneration. Initial formation of spindles in regenerating EDL grafts is independent of innervation; intrafusal muscle fibers degenerate and regenerate within spindle capsules that remain intact and viable. The extent of spindle differentiation was evaluated in each type of graft using criteria that included nucleation and ATPase activity, both of which have been shown to be regulated by sensory innervation, as well as the number of muscle fibers/spindle and morphology of spindle capsules.While most spindles contained normal numbers of muscle fibers, most of these fibers were morphologically and histochemically abnormal. Alterations of ATPase activity occurred in all spindles, but were least severe in nerve-intact grafts. While fully differentiated nuclear bag and chain fibers were not observed in regenerated spindles, large, vesicular nuclei, similar to those of normal intrafusal fibers, were present in a small number of spindles in nerve-intact grafts. Sensory nerve terminations were observed only in those spindles that also contained the distinctive nuclei. This study suggests that a specific neurotrophic influence is necessary for regeneration of normal intrafusal muscle fibers and that this influence corresponds to the properly timed sensory neuron-muscle interaction which directs muscle spindle embryogenesis. However, the infrequent occurrence of characteristics unique to intrafusal muscle fibers indicates that reinnervation of regenerating muscle grafts by sensory neurons is inadequate and/or faulty.  相似文献

NADPH-diaphorase histochemistry was combined with demonstration of acetylcholinesterase and immunocytochemistry for calcitonin gene-related peptide to study esophageal innervation in the rat. Most of the myenteric neurons stained positively for NADPH-diaphorase, as did numerous varicose nerve fibers in the myenteric plexus, among striated muscle fibers, around arterial blood vessels, and in the muscularis mucosae. A majority of motor endplates (as demonstrated by acetylcholinesterase histochemistry or calcitonin gene-related peptide immunocytochemistry) were associated with fine varicose NADPH-diaphorase-positive nerve fibers. Analysis of brainstem nuclei, sensory vagal, spinal, and sympathetic ganglia in normal and neonatally capsaicin-treated rats, and comparison with anterogradely labeled vagal branchiomotor, preganglionic and sensory fibers led to the conclusion that NADPH-diaphorase-positive fibers on motor endplates originate in esophageal myenteric neurons. No association of NADPH-diaphorasepositive nerve fibers with motor endplates was found in other organs containing striated muscle. These results suggest extensive, presumably nitrergic, co-innervation of esophageal striated muscle fibers by enteric neurons. Thus, control of peristalsis in the esophagus of the rat may be more complex than hitherto assumed.  相似文献

During muscle reinnervation, a transitory phase of polyinnervation occurs. In reinnervated muscles of vitamin E deficient rats, sprouting and polyinnervation are increased with respect to reinnervated controls. In this work, polyinnervation was observed in reinnervated extensor digitorum longus (edl) muscle of rats treated with pharmacological doses of vitamin E. Sciatic nerve was crushed and edl muscle was examined electrophysiologically at 30, 40 and 60 days after denervation. The percentage of polyinnervated cells in controls peaked at 30 days and thus it decreased. In muscles of vitamin E treated rats, the time course of percentage of polyinnervated muscle cells was qualitatively the same, but it was decreased at all times.  相似文献
