Summary Daily fluctuations of nucleoli and nucleolar fibrillar centres in neurosecretory cells from the supraoptic nucleus (SON) were investigated in rats artificially synchronized for 3 weeks to a set 12 h light/12 h dark cycle with free access to food and water. Groups of 3 animals were sacrificed by intracardiac perfusion every 4 h for a 24-h period and every 2 h between 22.00 h and 07.00 h. The SON of each animal was removed, and the mean nucleolar volume and the mean volume of the nucleolar fibrillar centres were estimated by a stereological analysis. The quantitative data showed that the fluctuations in the nucleolar volume of SON neurons depend on the time of sacrifice. A peak value was found in animals sacrificed at 03.00 h which was 1.5 times the value found in animals sacrificed at 19.00 h. The volume of fibrillar centres underwent small, but not significant changes over the 24-h period. None of the large fibrillary centres that can be observed in the superior cervical ganglion were found in the SON. Our results demonstrate that in these neurons the size of the nucleolus undergoes daily fluctuations. These results are discussed in the light of previous studies conducted at our laboratory on the circadian rhythm of nucleolar volume and of nucleolar components in neurons of the superior cervical ganglion.  相似文献

The ultrastructural organization of neurosecretory cells (NSC) belonging to the hypothalamic supraoptic nucleus was investigated in young rats following periods of painful stress differing in duration (of 2 and 20 min). Intact young rats of similar age and others at late stages of development (24–27 months old) served as controls. Short- and longer-acting painful stress was found to intensify and inhibit the functional activity of NSC respectively. Complex ultrastructural changes in the NSC of young rats following protracted painful stress is compared with the ultrastructural organizational pattern in several NSC of aging rats. Findings would imply that aging occurs in rats following prolonged painful stress, resulting from functional hypersecretion and depletion.A. A. The effects of subiculum stimulation were investigated in 80 antidromically identified hypothalamic supraoptic neurons in lactating rats. Inhibition manifesting as suppression of antidromic action potentials (or of their somatodendritic component) was revealed in 26% of cells, induced by applying conditioned and test stimuli to the subiculum and neurohypophysial stalk. In some instances inhibition arose following a latency of 5–25 msec after each subicular stimulus and lasted only briefly; it set in gradually in other cases, leading to stable long-term changes in the excitability of neurosecretory cells. No activation was produced by this stimulation. It is deduced that subicular inhibitory inputs follow different patterns, thus reflecting morphological organizational aspects of synaptic inhibitory inputs to neurosecretory cells.A. A. The effects of water deprivation were investigated in the pattern of response produced by subiculum stimulation in antidromically identified hypothalamic supraoptic neurosecretory cells of lactating rats. In dehydrated animals as compared with the controls, the percentage of neurons responding to subiculum stimulation with an inhibitory action (blockade of antidromic action potential) remained unchanged, although the proportion of differing inhibitory response did alter: numbers of cells with gradually developing inhibitory response increased significantly and fewer cells showed transitory development of inhibition. Inhibitory response emerging as depression of background spike activity showed a quantitative increase, moreover. Plasticity was found to be one distinguishing feature of afferent input from the subiculum to supraoptic nucleus neurosecretory cells and, in particular, a capacity for reorganization under water deprivation.A. Humori-positive neurosecretory cells of the supraoptic nucleus of the hypothalamus have been investigated during various time of parathyroprival hypocalcemia after extirpation of the parathyroid glands. Contents of total and ionized calcium, phosphorus in blood serum have been estimated. Volume of nuclei and nucleoli has been measured. In 5 days functional activity of the supraoptic nucleus increases (lightly stained cells predominate, volume of the nuclei and nucleoli increases). In subsequent 15-30 days its activity decreases (increase in amount of dark-stained cells, nucleolar volume decreases). In 60 days there is a tendency to restoration of neurosecretion.  相似文献

The ultrastructure of the pars anterior (adenohypophysis), treated with diazepam (2.5 mg/Kg) has been studied by thin sections and electron microscopy. The structure of Thyrotrophs, Corticotrophs, Gonadotrophs, and Mammotrophs cells were described. Ultrastructural modifications were not observed. It is concluded that the chronic treatment with diazepam did not produce any visible ultrastructural effects in the secretory cells of the adeno hypophysis; maybe because the diazepam has a short life time and due to the small effect of the diazepam metabolites in this species.  相似文献

For 6 months the rats were kept on 20 degrees ethanol. Then, the rats could choose whether to drink alcohol (A) or water. The rats formed 2 groups: those preferring A and those preferring water. The control rats were kept on water. The function of hypothalamus monoamine- and peptidergic systems were disturbed following chronic A treatment. Alcohol-preferring, unlike water-preferring rats, have revealed higher hypothalamus levels of serotonin and lower levels of dopamine and noradrenaline, which correlated with changes in fluorescence intensity of hypothalamus noradrenaline structures and were accompanied by remarkable disturbances in nonapeptide neurohormone transport in the paraventricular nucleus region.  相似文献

In the squirrel monkey (Saimiri sciureus) two types of cytoplasmic inclusion bodies have been observed sporadically in neurons of both the dorsal horn (Rexed's laminae I-III in the lumbosacral region) and the supraoptic nucleus. One of these, designated here the "vesicular body", is a round inclusion which measures up to 1.4 mu in diameter. It occurs only in perikarya and is composed of vesicular-like chambers 300-400 A in diameter. We have not found previous references to this structure in the literature, but its 50 A substructural particles are similar in size to those described in nematosomes. The other inclusion, a "filamentous whorl", is found in nerve cell bodies and dendrites and it is structurally similar to the Hirano body. The structure measures up to 2.2 mu in diameter and is composed of circularly arrayed filaments which vary in configuration and size depending on the plane of section. There are no indications that the vesicular body and the filamentous whorl are in any way related to each other; and usually both are not found in the same cell profiles.  相似文献

Summary Processes of magnocellular neurosecretory cells (MNCs) are easily identifiable on the basis of their content in neurosecretory granules in the neuropil of the rostral division of the paraventricular nucleus (PVN) of the domestic fowl. In specimens sacrificed during the winter the synaptic organization of the neuropil and the pattern of synapses ending on neurosecretory processes were studied at the ultrastructural level. Synapses in the rostral part of the PVN neuropil may be divided into three main categories on the basis of their morphology and their content of clear and dense-core synaptic vesicles. These different types of terminals can be attributed to aminergic, peptidergic or other types of synapses. The percent distribution of synapses within these categories differs when all synapses observed in the neuropil or only those ending on MNC processes are compared. Present ultrastructural data obtained in birds support two physiological hypotheses already suggested for mammals, i.e., the probable existence of a recurrent pathway to MNCs via an interneuron, and the importance of aminergic and peptidergic input in regulating the electrical activity of MNCs.This work was partly supported by a CNR grant (n. 81.00377.04)  相似文献

The morphometrical and electron microscopic analysis of secretory granules in the perikaryons of neurosecretory cells of the supraoptic and paraventricular nuclei in male rats and mice has shown than in the cells of these nuclei in both species of animals there occur secretory granules of the same kind and size. Therefore this method fails to determine which of them contain oxytocin and which of them contain vasorpressin. The neurosecretory granules located in the Golgi apparatus zone are of a less size and have more osmiophilic cnetral material than the granules localized on the periphery which mainly have granular central material and are of a greater size. The distinctions in the size and type of secretory granules are associated with certain stages of their "maturation". Granular particles appear to be "swallen", more active forms of storing neurohormones. The presence of larger granular particles in the supraoptic nucleus of mice allows to suggest greater reactivity of this nucleus than in rats which is likely to be associated with a higher ability of mice, as compared with rats, to adaptate to disturbances in water-salt metabolism.  相似文献
